1. Intensive Broadband Reverberation Mapping of Fairall 9 with 1.8 yr of Daily Swift Monitoring

Abstract: We present 1.8 yr of near-daily Swift monitoring of the bright, strongly variable Type 1 active galactic nucleus (AGN) Fairall 9. Totaling 575 successful visits, this is the largest such campaign reported to date. Variations within the UV/optical are well correlated, with longer wavelengths lagging shorter wavelengths in the direction predicted by thin-disk/lamppost models. The correlations are improved by “detrending,” subtracting a second-order polynomial fit to the UV/optical light curves to remove long-term trends that are not of interest to this study. Extensive testing indicates detrending with higher-order polynomials removes too much intrinsic variability signal on reverberation timescales. These data provide the clearest detection to date of interband lags within the UV, indicating that neither emission from a large disk nor diffuse continuum emission from the broad-line region (BLR) can independently explain the full observed lag spectrum. The observed X-ray flux variations are poorly correlated with those in the UV/optical. Further, subdivision of the data into four ∼160 days light curves shows that the UV/optical lag spectrum is highly stable throughout the four periods, but the X-ray to UV lags are unstable, significantly changing magnitude and even direction from one period to the next. This indicates the X-ray to UV relationship is more complex than predicted by the simple reprocessing model often adopted for AGN. A “bowl” model (lamppost irradiation and blackbody reprocessing on a disk with a steep rim) fit suggests the disk thickens at a distance (∼10 lt-day) and temperature (∼8000 K) consistent with the inner edge of the BLR.

Abstract: [Abridged] We use high-resolution $HST$ imaging and $e$-MERLIN 1.5-GHz observations of galaxy cores from the LeMMINGs survey to investigate the relation between optical structural properties and nuclear radio emission for a large sample of galaxies. We perform accurate, multi-component decompositions of new surface brightness profiles extracted from $HST$ images for 163 LeMMINGs galaxies and fit up to six galaxy components (e.g., bulges, discs, AGN, bars, rings, spiral arms, and nuclear star clusters) simultaneously with Sérsic and/or core-Sérsic models. By adding such decomposition data for 10 LeMMINGs galaxies from our past work, the final sample of 173 nearby galaxies (102 Ss, 42 S0s, 23 Es plus 6 Irr) with bulge stellar mass (typically) M_*, bulge ~ 10^6-10^12.5 M_sun, encompasses all optical spectral classes (LINER, Seyfert, ALG and H II). We show that the bulge mass can be significantly overestimated in many galaxies when components such as bars, rings and spirals are not included in the fits. We additionally implement a Monte Carlo method to determine errors on bulge, disc and other fitted structural parameters. Moving (in the opposite direction) across the Hubble sequence, i.e., from the irregular to elliptical galaxies, we confirm that bulges become larger, more prominent and round. Such bulge dominance is associated with a brighter radio core luminosity. We also find that the radio detection fraction increases with bulge mass. At M_*,bulge > 10^11 M_sun, the radio detection fraction is 77%, declining to 24% for M_bulge < 10^10 M_sun. Abstract: Very-high Energy (VHE) gamma-ray astroparticle physics is a relatively young field, and observations over the past decade have surprisingly revealed almost two hundred VHE emitters which appear to act as cosmic particle accelerators. These sources are an important component of the Universe, influencing the evolution of stars and galaxies. At the same time, they also act as a probe of physics in the most extreme environments known - such as in supernova explosions, and around or after the merging of black holes and neutron stars. However, the existing experiments have provided exciting glimpses, but often falling short of supplying the full answer. A deeper understanding of the TeV sky requires a significant improvement in sensitivity at TeV energies, a wider energy coverage from tens of GeV to hundreds of TeV and a much better angular and energy resolution with respect to the currently running facilities. The next generation gamma-ray observatory, the Cherenkov Telescope Array Observatory (CTAO), is the answer to this need. In this talk I will present this upcoming observatory from its design to the construction, and its potential science exploitation. CTAO will allow the entire astronomical community to explore a new discovery space that will likely lead to paradigm-changing breakthroughs. In particular, CTA has an unprecedented sensitivity to short (sub-minute) timescale phenomena, placing it as a key instrument in the future of multi-messenger and multi-wavelength time domain astronomy. Abstract: We have measured the wavelength-dependent lags between the X-ray, UV and optical bands in the high accretion rate ($L/L_{\rm Edd}\approx40\%$) Active Galactic Nucleus Mrk 110 during two intensive monitoring campaigns in February and September 2019. We divide the observations into three intervals with different X-ray luminosities. The first interval, already published in Vincentelli et al. (2021), has the lowest X-ray luminosity and did not exhibit the U-band excess positive lag, or the X-ray excess negative lag that is seen in most AGN. However, these excess lags are seen in the two subsequent intervals of higher X-ray luminosity. Although the data are limited, the excess lags appear to scale with X-ray luminosity. Our modelling shows that lags expected from reprocessing of X-rays by the accretion disc vary hardly at all with increasing luminosity. Therefore, as the U-band excess almost certainly arises from Balmer continuum emission from the broad line region (BLR), we attribute these lag changes to changes in the contribution from the BLR. The change is easily explained by the usual increase in the inner radius of the BLR with increasing ionising luminosity., Comment: 6 pages, 4 figures. Accepted for publication in MNRAS Letters

Abstract: Despite the plethora of deep (sub-mJy) radio surveys there remains considerable doubt as to the exact nature of the galaxies contributing to the source counts. Current evidence suggests that starformation in moderately luminous 'normal' galaxies is responsible for the bulk of the emission below 1mJy. However given the sensitivities of these surveys we would expect a fraction of these sources to be distant radio galaxies. Using deep VLA and GMRT data we have found ~20 high-z candidate radio galaxies in two fields using the classical ultra-steep radio spectrum technique (De Breuck et al., 2000) and selecting galaxies with faint (i'>25) optical counterparts. Several of these sources have X-ray detections in our deep XMM/Chandra observations and have fluxes high enough to put them in the quasar regime if they lie above redshift 3. Abstract: We present the results of a multi‐wavelength analysis of the properties of a sample of 86 X‐ray selected AGN. X‐ray flux and optical magnitude are strongly correlated in broad‐line AGN (BLAGN) as expected from the unified model. Narrow emission line galaxies (NELGs), however, display a wide range in optical brightnesses for a given X‐ray flux, which we attribute to varying amounts of obscuration and host galaxy contamination. We find that more luminous AGN and increasingly softer X‐ray AGN show redder mid infrared power law slopes in their spectra between 3.6 and 8 μm. The power law slope between 3.6 and 24 μm is however uncorrelated with X‐ray luminosity and X‐ray spectral index.

Abstract: We present radio-to-optical data taken by the WEBT, supplemented by VLBA and RXTE observations, of 3C 279. Our goal is to use this extensive database to draw inferences regarding the physics of the relativistic jet. We assemble multifrequency light curves with data from 30 ground-based observatories and the space-based instruments, along with linear polarization vs. time in the optical R band. In addition, we present a sequence of 22 images (with polarization vectors) at 43 GHz at resolution 0.15 milliarcsec, obtained with the VLBA. We analyse the light curves and polarization, as well as the spectral energy distributions at different epochs, corresponding to different brightness states. The IR-optical-UV continuum spectrum of the variable component corresponds to a power law with a constant slope of -1.6, while in the 2.4-10 keV X-ray band it varies in slope from -1.1 to -1.6. The steepest X-ray spectrum occurs at a flux minimum. During a decline in flux from maximum in late 2006, the optical and 43 GHz core polarization vectors rotate by ~300 degrees. The continuum spectrum agrees with steady injection of relativistic electrons with a power-law energy distribution of slope -3.2 that is steepened to -4.2 at high energies by radiative losses. The X-ray emission at flux minimum comes most likely from a new component that starts in an upstream section of the jet where inverse Compton scattering of seed photons from outside the jet is important. The rotation of the polarization vector implies that the jet contains a helical magnetic field that extends ~20 pc past the 43 GHz core., 12 pages, aa.cls style; accepted for publication in A&A

Abstract: Of the blazars detected by EGRET in GeV gamma rays, 3C 279 is not only the best-observed by EGRET, but also one of the best-monitored at lower frequencies. We have assembled eleven spectra, from GHz radio through GeV gamma rays, from the time intervals of EGRET observations. Although some of the data have appeared in previous publications, most are new, including data taken during the high states in early 1999 and early 2000. All of the spectra show substantial gamma-ray contribution to the total luminosity of the object; in a high state, the gamma-ray luminosity dominates over that at all other frequencies by a factor of more than 10. There is no clear pattern of time correlation; different bands do not always rise and fall together, even in the optical, X-ray, and gamma-ray bands. The spectra are modeled using a leptonic jet, with combined synchrotron self-Compton + external Compton gamma-ray production. Spectral variability of 3C 279 is consistent with variations of the bulk Lorentz factor of the jet, accompanied by changes in the spectral shape of the electron distribution. Our modeling results are consistent with the UV spectrum of 3C 279 being dominated by accretion disk radiation during times of low gamma-ray intensity., Comment: 39 pages including 13 figures; data tables not included (see ApJ web version or contact author)

Abstract: We present the results of the multi-wavelength campaigns on 3C 273 in 1993-1995. During the observations in late 1993 this quasar showed an increase of its flux for energies greater equal 100 MeV from about 2.1 x 10^-7 photons/cm^2/s to approximately 5.6 x 10^-7 photons/cm^2/s during a radio outburst at 14.5, 22 and 37 GHz. However, no one-to-one correlation of the gamma-ray radiation with any frequency could be found. The photon spectral index of the high energy spectrum changed from (3.20 +- 0.54) to (2.20 +- 0.22) in the sense that the spectrum flattened when the gamma-ray flux increased. Fits of the three most prominent models (synchrotron self-Comptonization, external inverse Comptonization and the proton initiated cascade model) for the explanation of the high gamma-ray emission of active galactic nuclei were performed to the multi-wavelength spectrum of 3C 273. All three models are able to represent the basic features of the multi-wavelength spectrum. Abstract: Four fields containing radio sources with spectral indices greater than unity at low frequencies have been observed over a wide range of radio wavelengths and at X-ray wavelengths. For most of the steep-spectrum sources no optical identifications have been found to the detection limit of the Palomar Schmidt prints, even with the positional precision of about 1 arcsec from VLA maps. This implies that the associated galaxies or clusters lie at a great distance. X-ray emission was not detected from any of the steep-spectrum radio sources. In one case, an extended source appears to be a radio halo in an uncataloged cluster of galaxies at a redshift of 0.125, and it is suggested that this object is a promising candidate for the detection of inverse Compton X-rays, since it is not a high-luminosity source of thermal X-rays.

Abstract: The RS CVn stars Capella and σ 2 CrB have been measured with EXOSAT in soft and medium X-rays for about 24 hours each and the less active late-type star Procyon for about 6.5 hours. In addition, the RS CVn star γ. And was twice observed about one month apart for a total of about 7 hours, with the ME and the LE in the photometer mode only. All three RS CVn stars were detected with the ME-detector. The star σ 2 CrB showed a flare both in LE and ME with a rise time of about twelve minutes and a decay time of three hours. The active late-type stars σ 2 CrB and Capella show in the spectral region between 90 and 140 A lines from Fe XVIII to Fe XXIII, which can be resolved with the moderate resolution ( Δγ ≈ 5 A) of the spectrometer. These lines are indicative of the presence of hot (≈ 10 MK) plasma like that in a Solar flare. In contrast, the spectrum of the cooler corona of the star Procyon does not show the hot Fe XXII and Fe XXIII lines but instead a blend at 175 A of Fe IX, X and XI lines that are formed in a typically quiet corona of a temperature around 1.5 MK. From the spectral intensities and the additional results of the simultaneous multi-color photometry coronal temperatures and emission measures are derived. There are indications in the spectra that the emission should be interpreted in terms of differential emission measure distribution models.

Abstract: The Arcminute Microkelvin Imager (AMI) is a telescope specifically designed for high sensitivity measurements of low-surface-brightness features at cm-wavelength and has unique, important capabilities. It consists of two interferometer arrays operating over 13.5-18 GHz that image structures on scales of 0.5-10 arcmin with very low systematics. The Small Array (AMI-SA; ten 3.7-m antennas) couples very well to Sunyaev-Zel'dovich features from galaxy clusters and to many Galactic features. The Large Array (AMI-LA; eight 13-m antennas) has a collecting area ten times that of the AMI-SA and longer baselines, crucially allowing the removal of the effects of confusing radio point sources from regions of low surface-brightness, extended emission. Moreover AMI provides fast, deep object surveying and allows monitoring of large numbers of objects. 19. Swift Monitoring of NGC 4151: Evidence for a Second X-Ray/UV Reprocessing.

Abstract: Swift monitoring of NGC 4151 with an ∼6 hr sampling over a total of 69 days in early 2016 is used to construct light curves covering five bands in the X-rays (0.3–50 keV) and six in the ultraviolet (UV)/optical (1900–5500 Å). The three hardest X-ray bands (>2.5 keV) are all strongly correlated with no measurable interband lag, while the two softer bands show lower variability and weaker correlations. The UV/optical bands are significantly correlated with the X-rays, lagging ∼3–4 days behind the hard X-rays. The variability within the UV/optical bands is also strongly correlated, with the UV appearing to lead the optical by ∼0.5–1 days. This combination of ≳3 day lags between the X-rays and UV and ≲1 day lags within the UV/optical appears to rule out the “lamp-post” reprocessing model in which a hot, X-ray emitting corona directly illuminates the accretion disk, which then reprocesses the energy in the UV/optical. Instead, these results appear consistent with the Gardner & Done picture in which two separate reprocessings occur: first, emission from the corona illuminates an extreme-UV-emitting toroidal component that shields the disk from the corona; this then heats the extreme-UV component, which illuminates the disk and drives its variability. Abstract: The Cherenkov Telescope Array (CTA) is a new observatory for very high-energy (VHE) gamma rays. CTA has ambitions science goals, for which it is necessary to achieve full-sky coverage, to improve the sensitivity by about an order of magnitude, to span about four decades of energy, from a few tens of GeV to above 100 TeV with enhanced angular and energy resolutions over existing VHE gamma-ray observatories. An international collaboration has formed with more than 1000 members from 27 countries in Europe, Asia, Africa and North and South America. In 2010 the CTA Consortium completed a Design Study and started a three-year Preparatory Phase which leads to production readiness of CTA in 2014. Abstract: High-time-resolution X-ray observations of compact objects provide direct access to strong-field gravity, to the equation of state of ultra-dense matter and to black hole masses and spins. A 10 m^2-class instrument in combination with good spectral resolution is required to exploit the relevant diagnostics and answer two of the fundamental questions of the European Space Agency (ESA) Cosmic Vision Theme "Matter under extreme conditions", namely: does matter orbiting close to the event horizon follow the predictions of general relativity? What is the equation of state of matter in neutron stars? The Large Observatory For X-ray Timing (LOFT), selected by ESA as one of the four Cosmic Vision M3 candidate missions to undergo an assessment phase, will revolutionise the study of collapsed objects in our galaxy and of the brightest supermassive black holes in active galactic nuclei. Thanks to an innovative design and the development of large-area monolithic Silicon Drift Detectors, the Large Area Detector (LAD) on board LOFT will achieve an effective area of ~12 m^2 (more than an order of magnitude larger than any spaceborne predecessor) in the 2-30 keV range (up to 50 keV in expanded mode), yet still fits a conventional platform and small/medium-class launcher. Abstract: Ground-based gamma-ray astronomy has had a major breakthrough with the impressive results obtained using systems of imaging atmospheric Cherenkov telescopes. Ground-based gamma-ray astronomy has a huge potential in astrophysics, particle physics and cosmology. CTA is an international initiative to build the next generation instrument, with a factor of 5-10 improvement in sensitivity in the 100 GeV-10 TeV range and the extension to energies well below 100 GeV and above 100 TeV. CTA will consist of two arrays (one in the north, one in the south) for full sky coverage and will be operated as open observatory. The design of CTA is based on currently available technology. Abstract: Background: Point-of-care (POC) Coccidioides antibody assays may provide veterinarians with rapid and accurate diagnostic information., Objectives: To determine the agreement of a POC lateral flow assay (LFA), sona Coccidioides (IMMY, Norman, Oklahoma) with the current diagnostic standard, the immunodiffusion assay (agar gel immunodiffusion [AGID]; Coccidioidomycosis Serology Laboratory, University of California, Davis, California)., Animals: Forty-eight sera specimens from 48 dogs., Methods: Sera specimens were collected from client-owned dogs that had a clinical suspicion for coccidioidomycosis. Animals were classified as Coccidioides antibody-positive (n = 36) based on a positive AGID or Coccidioides antibody-negative (n = 12) based on a negative AGID. The performance of the LFA assay was determined by comparing results to AGID results., Results: The LFA assay demonstrated agreement in 32 of 36 Coccidioides antibody-positive specimens and 12 of 12 Coccidioides antibody-negative specimens, resulting in a positive percentage agreement of 88.9% (95% confidence interval [CI], 74.7-95.6%) and negative percentage agreement of 100% (95% CI, 75.8-100%) as compared to AGID. A receiver operator characteristic curve was constructed, and the area under the curve was 0.944 (CI, 0.880-1.000)., Conclusion and Clinical Importance: This LFA is a rapid alternative to the traditional AGID. The LFA provides excellent predictive value for positive results. Positive agreement was lower in dogs with low AGID titers; therefore, confirmatory testing is recommended if a high index of suspicion exists., (© 2021 The Authors. Abstract: The COVID-19 pandemic caused by the new SARS-CoV-2 coronavirus has imposed severe challenges on laboratories in their effort to achieve sufficient diagnostic testing capability for identifying infected individuals. In this study, we report the analytical and clinical performance characteristics of a new, high-throughput, fully automated nucleic acid amplification test system for the detection of SARS-CoV-2. The assay utilizes target capture, transcription-mediated amplification, and acridinium ester-labeled probe chemistry on the automated Panther system to directly amplify and detect two separate target sequences in the open reading frame 1ab (ORF1ab) region of the SARS-CoV-2 RNA genome. The probit 95% limit of detection of the assay was determined to be 0.004 50% tissue culture infective dose (TCID 50 )/ml using inactivated virus and 25 copies/ml (c/ml) using synthetic in vitro transcript RNA targets. Analytical sensitivity (100% detection) was confirmed to be 83 to 194 c/ml using three commercially available SARS-CoV-2 nucleic acid controls. No cross-reactivity or interference was observed with testing of six related human coronaviruses, as well as 24 other viral, fungal, and bacterial pathogens, at high titers. Clinical nasopharyngeal swab specimen testing ( n = 140) showed 100%, 98.7%, and 99.3% positive, negative, and overall agreement, respectively, with a validated reverse transcription-PCR nucleic acid amplification test (NAAT) for SARS-CoV-2 RNA. 26. Coccidioidomycosis in llamas and alpacas diagnosed at the University of California, Davis (1990-2016).

Abstract: Given the predisposition of South American camelids to coccidioidomycosis, we sought to describe the disease presentation in alpacas and llamas and identify potential risk factors for these species. The records of 224 llamas and alpacas that were tested for Coccidioides infection using immunodiffusion serology at the Coccidioidomycosis Serology Laboratory of the University of California, Davis, between 1990 and 2016 were examined; of those, 46 alpacas and 42 llamas had positive test results. The remaining 99 alpacas and 37 llamas were used as control groups. We found that male llamas were at increased risk for Coccidioides infection when compared with female llamas and when compared with male alpacas. South American camelids living within California were at higher risk for infection than camelids living in other states. Alpacas were more likely than llamas to have subclinical infections. We documented five cases of abortion or neonatal mortality attributable to coccidioidomycosis in alpacas. Our study demonstrates that South American camelids are susceptible to Coccidioides infection in areas where the disease is endemic, lending support to the importance of vigilance for this disease in alpacas and llamas and suggesting a possible role for these animals as sentinel species., Lay Summary: We examined cases of Valley Fever and described the disease and risk factors for llamas and alpacas. Male llamas were at increased risk for infection as were animals living within California. 27. Posaconazole Serum Drug Levels Associated With Pseudohyperaldosteronism.

Abstract: Background: Posaconazole tablets are well tolerated and efficacious in the prophylaxis and treatment of aspergillosis, mucormycosis, and other invasive fungal infections. There have been case reports of posaconazole-induced pseudohyperaldosteronism (PIPH); however, its occurrence and association with serum posaconazole drug levels have not previously been investigated., Methods: In this single-center, retrospective, observational study, we examined the occurrence of PIPH in outpatients newly starting posaconazole and evaluated differences in serum posaconazole concentrations and clinical characteristics between those with and without this syndrome., Results: Sixty-nine patients receiving posaconazole were included, of whom 16 (23.2%) met the definition of PIPH. Patients with PIPH were significantly older (61.1 vs 44.7 years, P = .007) and more frequently had hypertension prior to starting posaconazole (68.8% vs 32.1%, P = .009). Patients with PIPH had a significantly higher median serum posaconazole level than those without PIPH (3.0 vs 1.2 µg/mL, P ≤ .0001). There was a positive correlation between serum posaconazole levels and changes in systolic blood pressure (r = .37, P = .01), a negative correlation between serum posaconazole levels and changes in serum potassium (r = -.39, P = .006), and a positive correlation between serum posaconazole levels and serum 11-deoxycortisol (r = .69, P < .0001)., Conclusions: Posaconazole is associated with secondary hypertension and hypokalemia, consistent with pseudohyperaldosteronism, and development is associated with higher serum posaconazole concentrations, older age, and baseline hypertension., (© The Author(s) 2019. 28. Emergence of stationary multimodality under two-timescaled dichotomic noise.

Abstract: We study a linear Langevin dynamics driven by an additive non-Markovian symmetrical dichotomic noise. It is shown that when the statistics of the time intervals between noise transitions is characterized by two well differentiated timescales, the stationary distribution may develop multimodality (bi- and trimodality). The underlying effects that lead to a probability concentration in different points include intermittence and also a dynamical locking of realizations. Our results are supported by numerical simulations as well as by an exact treatment obtained from a Markovian embedding of the full dynamics, which leads to a third-order differential equation for the stationary distribution.

Abstract: Intravenous immunoglobulin (IVIG) is used to treat an increasing number of conditions. The anti-inflammatory and immunomodulatory effects of IVIG can be life-saving; however, recent administration may complicate evaluation for infection. To assess the impact of IVIG therapy on a variety of common viral, bacterial, fungal, and parasitic serologies we prospectively evaluated serologic changes pre- and post-IVIG infusion in 7 participants. The number of new antibody detections ranging from 2 to 5. New detections included positivity for Epstein-Barr virus early D antigen, herpes simplex virus, West Nile virus, cytomegalovirus, and the endemic mycoses Histoplasma and Coccidioides. The greatest number of newly positive serologies was observed in subjects receiving cumulative doses of IVIG in excess of 100 g. Our results illustrate the difficulty in serologic interpretation following IVIG therapy and suggest a dose-response to new positive results. Abstract: Background: Point-of-care (POC) Cryptococcus antigen assays may provide veterinarians with a more rapid, patient-side diagnosis when compared with traditional laboratory-based latex agglutination tests., Objective: To determine the sensitivity and specificity of 2 POC lateral flow cryptococcal serum antigen tests, CrAg LFA (Immy, Norman, OK) and the CryptoPS (Biosynex, Strasbourg, France) for diagnosis of cryptococcosis in dogs and cats, using the cryptococcal antigen latex agglutination system (CALAS) as the reference standard., Animals: 102 serum samples from 51 dogs and 40 cats., Methods: Specimens were classified as CALAS-positive (n = 25) or CALAS-negative (n = 77). The sensitivity and specificity of each POC assay was calculated by comparing the results to the serologic reference standard results., Results: The CrAg LFA assay correctly classified 23/25 CALAS-positive specimens and 69/74 CALAS-negative specimens resulting in a sensitivity of 92.0% (confidence interval [CI], 75.0%-98.6%) and specificity of 93.2% (CI, 85.1%-97.1%). The CryptoPS assay correctly classified 8/10 tested CALAS-positive specimens and 56/59 tested CALAS-negative specimens resulting in a sensitivity of 80.0% (CI, 49.0%-96.5%) and specificity of 94.9% (CI, 86.1%-98.6%)., Conclusion and Clinical Importance: The POC assays appear to be a sensitive and specific alternative to the traditional CALAS assay with more rapid turnaround times, which may result in earlier diagnosis and treatment., (© 2019 The Authors. Abstract: Patients with coccidioidal meningitis require lifelong antifungal therapy. Cumulative toxicity and lack of antifungal efficacy require salvage therapy in the treatment of some patients. In a retrospective review of nine patients with coccidioidal meningitis treated with isavuconazole, successful therapy was seen in three patients and stable disease was confirmed in six patients. Abstract: Fluconazole-induced alopecia is a significant problem for patients receiving long-term therapy. We evaluated the hair cycle changes of fluconazole in a rat model and investigated potential molecular mechanisms. Plasma and tissue levels of retinoic acid were not found to be causal. Human patients with alopecia attributed to fluconazole also underwent detailed assessment and in both our murine model and human cohort fluconazole induced telogen effluvium. Abstract: Coccidioidal meningitis remains a difficult clinical problem, and despite life-long therapy with triazole antifungals, relapses of disease and medication intolerance occur necessitating salvage treatment. We report two patients with recurrent coccidioidal meningitis who improved following a 2-week course of liposomal amphotericin B monotherapy and discuss potential advantages of this treatment option.

Abstract: We describe a case of apparent mineralocorticoid excess (hypertension, hypokalemia, metabolic alkalosis and low plasma renin activity) secondary to itraconazole therapy. Inhibition of 11β-hydroxysteroid dehydrogenase 2 was demonstrated, and withholding itraconazole led to resolution of adverse effects that did not recur with voriconazole. Abstract: We describe a case of apparent mineralocorticoid excess (AME) secondary to posaconazole therapy and suggest the biochemical mechanism. Clinical and laboratory investigation confirmed 11β-hydroxysteroid dehydrogenase inhibition and withholding therapy led to a resolution of all clinical and laboratory abnormalities. Posaconazole was later restarted at a lower dose and prevented recurrence of this syndrome. Additional studies are necessary to determine the frequency of posaconazole-induced AME and whether other azole antifungals can be associated with this phenomenon., (Copyright © 2017 American Society for Microbiology.)

37. Microgeographic Proteomic Networks of the Human Colonic Mucosa and Their Association With Inflammatory Bowel Disease.

Abstract: Background & Aims: Interactions between mucosal cell types, environmental stressors, and intestinal microbiota contribute to pathogenesis in inflammatory bowel disease (IBD). Here, we applied metaproteomics of the mucosal-luminal interface to study the disease-related biology of the human colonic mucosa., Methods: We recruited a discovery cohort of 51 IBD and non-IBD subjects endoscopically sampled by mucosal lavage at 6 colonic regions, and a validation cohort of 38 no-IBD subjects. Metaproteome data sets were produced for each sample and analyzed for association with colonic site and disease state using a suite of bioinformatic approaches. Localization of select proteins was determined by immunoblot analysis and immunohistochemistry of human endoscopic biopsy samples., Results: Co-occurrence analysis of the discovery cohort metaproteome showed that proteins at the mucosal surface clustered into modules with evidence of differential functional specialization (eg, iron regulation, microbial defense) and cellular origin (eg, epithelial or hemopoietic). These modules, validated in an independent cohort, were differentially associated spatially along the gastrointestinal tract, and 7 modules were associated selectively with non-IBD, ulcerative colitis, and/or Crohn's disease states. In addition, the detailed composition of certain modules was altered in disease vs healthy states. We confirmed the predicted spatial and disease-associated localization of 28 proteins representing 4 different disease-related modules by immunoblot and immunohistochemistry visualization, with evidence for their distribution as millimeter-scale microgeographic mosaic., Conclusions: These findings suggest that the mucosal surface is a microgeographic mosaic of functional networks reflecting the local mucosal ecology, whose compositional differences in disease and healthy samples may provide a unique readout of physiologic and pathologic mucosal states.

38. On the existence of low-mass dark matter and its direct detection.

Abstract: Dark Matter (DM) is an elusive form of matter which has been postulated to explain astronomical observations through its gravitational effects on stars and galaxies, gravitational lensing of light around these, and through its imprint on the Cosmic Microwave Background (CMB). This indirect evidence implies that DM accounts for as much as 84.5% of all matter in our Universe, yet it has so far evaded all attempts at direct detection, leaving such confirmation and the consequent discovery of its nature as one of the biggest challenges in modern physics. Here we present a novel form of low-mass DM χ that would have been missed by all experiments so far. While its large interaction strength might at first seem unlikely, neither constraints from particle physics nor cosmological/astronomical observations are sufficient to rule out this type of DM, and it motivates our proposal for direct detection by optomechanics technology which should soon be within reach, namely, through the precise position measurement of a levitated mesoscopic particle which will be perturbed by elastic collisions with χ particles. We show that a recently proposed nanoparticle matter-wave interferometer, originally conceived for tests of the quantum superposition principle, is sensitive to these collisions, too.

Abstract: Fucosyltransferase 2 (FUT2) is an enzyme that is responsible for the synthesis of the H antigen in body fluids and on the intestinal mucosa. The H antigen is an oligosaccharide moiety that acts as both an attachment site and carbon source for intestinal bacteria. Non-secretors, who are homozygous for the loss-of-function alleles of FUT2 gene (sese), have increased susceptibility to Crohn's disease (CD). To characterize the effect of FUT2 polymorphism on the mucosal ecosystem, we profiled the microbiome, meta-proteome and meta-metabolome of 75 endoscopic lavage samples from the cecum and sigmoid of 39 healthy subjects (12 SeSe, 18 Sese and 9 sese). Imputed metagenomic analysis revealed perturbations of energy metabolism in the microbiome of non-secretor and heterozygote individuals, notably the enrichment of carbohydrate and lipid metabolism, cofactor and vitamin metabolism and glycan biosynthesis and metabolism-related pathways, and the depletion of amino-acid biosynthesis and metabolism. Similar changes were observed in mice bearing the FUT2(-/-) genotype. Metabolomic analysis of human specimens revealed concordant as well as novel changes in the levels of several metabolites. Human metaproteomic analysis indicated that these functional changes were accompanied by sub-clinical levels of inflammation in the local intestinal mucosa. Therefore, the colonic microbiota of non-secretors is altered at both the compositional and functional levels, affecting the host mucosal state and potentially explaining the association of FUT2 genotype and CD susceptibility.

40. Characterization of aid1, a novel gene involved in Fusobacterium nucleatum interspecies interactions.

Abstract: The oral opportunistic pathogen Fusobacterium nucleatum is known to interact with a large number of different bacterial species residing in the oral cavity. It adheres to a variety of Gram-positive bacteria, including oral streptococci via the arginine-inhibitable adhesin RadD. In this study, we describe a novel protein encoded by the predicted open reading frame FN1253 that appears to play a role in interspecies interactions of F. nucleatum, particularly with oral streptococci and related Gram-positive species. We designated FN1253 as aid1 (Adherence Inducing Determinant 1). Expression analyses demonstrated that this gene was induced in F. nucleatum single species biofilms, while the presence of representative members of the oral microbiota known to adhere to F. nucleatum triggered its suppression. Inactivation as well as overexpression of aid1 affected the ability of F. nucleatum to coaggregate with oral streptococci and the closely related Enterococcus faecalis, but not other Gram-positive oral species tested. Furthermore, overexpression of aid1 led to a drastic change in the structure of dual species biofilms of F. nucleatum with oral streptococci. Aid1 function was abolished in the presence of arginine and found to be dependent on RadD. Interestingly, differential expression of aid1 did not affect messenger RNA and protein levels of RadD. These findings indicate that RadD-mediated adhesion to oral streptococci involves more complex cellular processes than the simple interaction of adhesins on the surface of partner strains. Aid1 could potentially play an important role in facilitating RadD-mediated interaction with oral streptococci by increasing binding specificity of F. nucleatum to other microbial species.

Abstract: Exopolysaccharide (EPS) of Myxococcus xanthus is a well-regulated cell surface component. In addition to its known functions for social motility and fruiting body formation on solid surfaces, EPS has also been proposed to play a role in multi-cellular clumping in liquid medium, though this phenomenon has not been well studied. In this report, we confirmed that M. xanthus clumps formed in liquid were correlated with EPS levels and demonstrated that the EPS encased cell clumps exhibited biofilm-like structures. The clumps protected the cells at physiologically relevant EPS concentrations, while cells lacking EPS exhibited significant reduction in long-term viability and resistance to stressful conditions. However, excess EPS production was counterproductive to vegetative growth and viable cell recovery declined in extended late stationary phase as cells became trapped in the matrix of clumps. Therefore, optimal EPS production by M. xanthus is important for normal physiological functions in liquid.

Abstract: One intriguing discovery in modern microbiology is the extensive presence of extracellular DNA (eDNA) within biofilms of various bacterial species. Although several biological functions have been suggested for eDNA, including involvement in biofilm formation, the detailed mechanism of eDNA integration into biofilm architecture is still poorly understood. In the biofilms formed by Myxococcus xanthus, a Gram-negative soil bacterium with complex morphogenesis and social behaviors, DNA was found within both extracted and native extracellular matrices (ECM). Further examination revealed that these eDNA molecules formed well organized structures that were similar in appearance to the organization of exopolysaccharides (EPS) in ECM. Biochemical and image analyses confirmed that eDNA bound to and colocalized with EPS within the ECM of starvation biofilms and fruiting bodies. In addition, ECM containing eDNA exhibited greater physical strength and biological stress resistance compared to DNase I treated ECM. Taken together, these findings demonstrate that DNA interacts with EPS and strengthens biofilm structures in M. xanthus.

Abstract: The periodontal pathogen T. denticola resides in a stressful environment rife with challenges, the human oral cavity. Knowledge of the stress response capabilities of this invasive spirochete is currently very limited. Whole genome expression profiles in response to different suspected stresses including heat shock, osmotic downshift, oxygen and blood exposure were examined. Most of the genes predicted to encode conserved heat shock proteins (HSPs) were found to be induced under heat and oxygen stress. Several of these HSPs also seem to be important for survival in hypotonic solutions and blood. In addition to HSPs, differential regulation of many genes encoding metabolic proteins, hypothetical proteins, transcriptional regulators and transporters was observed in patterns that could betoken functional associations. In summary, stress responses in T. denticola exhibit many similarities to the corresponding stress responses in other organisms but also employ unique components including the induction of hypothetical proteins.

Abstract: Human gammaherpesviruses, Epstein-Barr virus, and human herpesvirus 8/Kaposi's sarcoma-associated herpesvirus are important pathogens associated with diseases, including lymphomas and other malignancies. Murine gammaherpesvirus 68 (MHV-68) is used as an experimental model system to study the host immune control of infection and explore novel vaccine strategies based on latency-deficient live viruses. We studied the properties and the potential of a recombinant MHV-68 (AC-RTA) in which the genes required for persistent infection were replaced by a constitutively expressed viral transcription activator, RTA, which dictates the virus to lytic replication. After intranasal infection of mice, replication of AC-RTA in the lung was attenuated, and no AC-RTA virus or viral DNA was detected in the isolated splenocytes, indicating a lack of latency in the spleen. Infection of the AC-RTA virus elicited both cellular immune responses and virus-specific IgG at a level comparable to that elicited by infection of the wild-type virus. Importantly, vaccination of AC-RTA was able to protect mice against subsequent challenge by the wild-type MHV-68. AC-RTA provides a vaccine strategy for preventing infection of human gammaherpesviruses. Furthermore, our results suggest that immunity to the major latent antigens is not required for protection.

Abstract: G10KHc, a specifically targeted antimicrobial peptide developed in our laboratory, has shown rapid and selective killing activity against Pseudomonas aeruginosa in culture medium. Because of the major role played by this pathogen in cystic fibrosis, we sought to evaluate the utility of G10KHc under more physiologic conditions in vitro. In the current study, we found that robust G10KHc activity could be maintained in expectorated sputum if serine protease-dependent digestion associated with this fluid was inhibited, either by chemical antagonists or by the construction of a D-amino acid enantiomer of G10KHc. Further investigations revealed that specifically targeted antimicrobial peptide activity in sputum could be further enhanced when samples were treated with a combination of peptide and recombinant human DNase. Our results illustrate the importance of investigating combination therapy to treat cystic fibrosis, especially if protease-sensitive peptide-based agents, such as G10KHc, are to be developed as alternatives to, or in conjunction with, conventional small-molecule antibiotics.

Abstract: Pseudomonas aeruginosa is a common opportunistic human pathogen that is associated with life-threatening acute infections and chronic airway colonization during cystic fibrosis. Previously, we converted the wide-spectrum antimicrobial peptide novispirin G10 into a selectively-targeted antimicrobial peptide (STAMP), G10KHc. Compared to novispirin G10, the STAMP had an enhanced ability to kill Pseudomonas mendocina. In this study, we explored the activity of G10KHc against P. aeruginosa. G10KHc was found to be highly active (as active as tobramycin) against P. aeruginosa clinical isolates. Most interestingly, we observed a synergistic-like enhancement in killing activity when biofilms and planktonic cultures of P. aeruginosa were cotreated with G10KHc and tobramycin. The data indicate that the mechanism of enhanced activity may involve increased tobramycin uptake due to G10KHc-mediated cell membrane disruption. These results suggest that G10KHc may be useful against P. aeruginosa during acute and chronic infection states, especially when it is coadministered with tobramycin.

Abstract: Nipah virus (NiV) is a deadly emerging paramyxovirus. The NiV attachment (NiV-G) and fusion (NiV-F) envelope glycoproteins mediate both syncytium formation and viral entry. Specific N-glycans on paramyxovirus fusion proteins are generally required for proper conformational integrity and biological function. However, removal of individual N-glycans on NiV-F had little negative effect on processing or fusogenicity and has even resulted in slightly increased fusogenicity. Here, we report that in both syncytium formation and viral entry assays, removal of multiple N-glycans on NiV-F resulted in marked increases in fusogenicity (>5-fold) but also resulted in increased sensitivity to neutralization by NiV-F-specific antisera. The mechanism underlying the hyperfusogenicity of these NiV-F N-glycan mutants is likely due to more-robust six-helix bundle formation, as these mutants showed increased fusion kinetics and were more resistant to neutralization by a fusion-inhibitory reagent based on the C-terminal heptad repeat region of NiV-F. Finally, we demonstrate that the fusogenicities of the NiV-F N-glycan mutants were inversely correlated with the relative avidities of NiV-F's interactions with NiV-G, providing support for the attachment protein "displacement" model of paramyxovirus fusion. Our results indicate that N-glycans on NiV-F protect NiV from antibody neutralization, suggest that this "shielding" role comes together with limiting cell-cell fusion and viral entry efficiencies, and point to the mechanisms underlying the hyperfusogenicity of these N-glycan mutants. These features underscore the varied roles that N-glycans on NiV-F play in the pathobiology of NiV entry but also shed light on the general mechanisms of paramyxovirus fusion with host cells.

Abstract: Murine gammaherpesvirus 68 (MHV-68) has been developed as a model for the human gammaherpesviruses Epstein-Barr virus and human herpesvirus 8/Kaposi's sarcoma-associated herpesvirus (HHV-8/KSHV), which are associated with several types of human diseases. Open reading frame 45 (ORF45) is conserved among the members of the Gammaherpesvirinae subfamily and has been suggested to be a virion tegument protein. The repression of ORF45 expression by small interfering RNAs inhibits MHV-68 viral replication. However, the gene product of MHV-68 ORF45 and its function have not yet been well characterized. In this report, we show that MHV-68 ORF45 is a phosphorylated nuclear protein. We constructed an ORF45-null MHV-68 mutant virus (45STOP) by the insertion of translation termination codons into the portion of the gene encoding the N terminus of ORF45. We demonstrated that the ORF45 protein is essential for viral gene expression immediately after the viral genome enters the nucleus. These defects in viral replication were rescued by providing ORF45 in trans or in an ORF45-null revertant (45STOP.R) virus. Using a transcomplementation assay, we showed that the function of ORF45 in viral replication is conserved with that of its KSHV homologue. Finally, we found that the C-terminal 23 amino acids that are highly conserved among the Gammaherpesvirinae subfamily are critical for the function of ORF45 in viral replication.
