Abstract

Artemisia absinthium L. is a highly medicinal plant with a broad range of biomedical applications. A. absinthium callus cultures were established in response to bio-fabricated single NPs (Ag and Cu) or a combination of both NPs (Ag and Cu) in different ratios (1:2, 2:1, 1:3, and 3:1) along with thidiazuron (TDZ) (4 mg/L) to elicit the biomass accumulation, production of non-enzymatic compounds, antioxidative enzymes, and antioxidant activity. Silver and copper nanoparticles (Ag and Cu NPs) were synthesized using the leaves of Moringa oleifera as reducing and capping agent and further characterized through UV-Visible spectroscopy and SEM. The 30 µg/L suspension of Ag and Cu NPs (1:2, 2:1) and 4 mg/L TDZ showed 100% biomass accumulation as compared to control (86%). TDZ in combination with Ag NPs enhanced biomass in the log phases of growth kinetics. The Cu NPs alone enhanced the superoxide dismutase activity (0.56 nM/min/mg FW) and peroxidase activity (0.31 nM/min/mg FW) in callus cultures. However, the combination of Ag and Cu NPs with TDZ induced significant total phenolic (7.31 µg/g DW) and flavonoid contents (9.27 µg/g DW). Furthermore, the antioxidant activity was highest (86%) in the Ag and Cu NPs (3:1) augmented media. The present study provides the first evidence of bio-fabricated single NPs (Ag and Cu) or a combination of both NPs (Ag and Cu) in different ratios (1:2, 2:1, 1:3, and 3:1) along with TDZ (4 mg/L) on the development of callus culture, production of endogenous enzymes, non-enzymatic components, and further antioxidant activity in callus cultures of A. absinthium.

Abstract

The present study was aimed to biosynthesize selenium nanoparticles (SeNPs) and assess their foliar applications to improve the growth of wheat plants under controlled irrigation and drought stress. Bud aqueous extract of Allium sativum L. was used as a reducing and stabilizing agent of SeNPs followed by their optical and morphological characterization by using ultraviolet-visible spectroscopy, scanning electron microscopy, Fourier-transform infrared, and energy dispersive X-ray analysis. Various concentrations of SeNPs (10, 20, 30, and 40 mg/L) were applied exogenously to drought-tolerant (V1) and drought-susceptible (V2) wheat varieties at the trifoliate stage. Under the positive control conditions, plants were irrigated with 450 mL of water/pot (100% field capacity); and under water-deficit environment, plants were irrigated with 160 mL of water/pot (35% field capacity). Remarkable increase in plant height, shoot length, shoot fresh weight, shoot dry weight, root length, root fresh weight, root dry weight, leaf area, leaf number, and leaf length has been observed when 30 mg/L concentration of SeNPs was used. However, the plant morphological parameters decreased gradually at higher concentrations (40 mg/L) in both selected wheat varieties. Therefore, 30 mg/L concentration of SeNPs was found most preferable to enhance the growth of selected wheat varieties under normal and water-deficient conditions.

Abstract

According to the United Nations FAO (Food and Agriculture Organization), the world population will expand by another 2 billion in 2050, but extra land area under cultivation will account for only 4% of the total land area at that time. In such cases, more efficient farming practices can be achieved by utilizing recent technical developments and solutions to current farming bottlenecks. Crop metrics could be constructed over hundreds of acres of cultivable land using remote sensing (RS) techniques and 3D laser scanning. AI-based crops produced a 30% increase in average crop output per ha and provided a rapid GTM (go-to-market) approach for crops. Abstract

Introduction: Antibioticshave significant importance in medicinal drugs but unfortunately bacteria can adapt various resistance mechanisms against those due to their abuse. A frequent way through which nitrate-reducing uropathogens acquire resistance mechanism against fluoroquinolones group of antibiotics is by continuously occurring mutation either in gyrA or gyrB genes through altering enzyme DNA gyrase that help in DNA replication. Microorganisms causing Urinary Tract Infection (UTI) vary in their susceptibility to antimicrobials from place to place and from time to time. Aim: The current study was performed to find the prevalence of nitrate-reducing bacteria in district Kohat and Hangu, their antimicrobial sensitivity pattern to various fluoroquinolones group of antibiotics and detection of mutation in gyrA and gyrB genes in the resistant bacterial isolates. Nitrate-reducing bacteria i.e E.coli, Klebsiella and Proteus are considered to be the frequent cause of UTI that reduce nitrate to nitrite by releasing enzyme nitrate reductase. Methodology: A Total of 252 urine specimens of suspected UTI patients were collected from Districts Head Quarter Hospitals (DHQ) and Combined Military Hospital (CMH) hospitals of districts Kohat and Hangu, Pakistan. Specimens reactive to nitrate strip reduction test were inoculated on (Cysteine Lactose Electrolyte Deficient) CLED media, and incubated at 37°C for 24 hours. Ciprofloxacin, levofloxacin, norfloxacin, ofloxacin and moxifloxacin fluoroquinolones group of antibiotics were used for to check their sensitivity patterns. Result: One hundred and seventy-five (69.44%) urine specimens were positive for nitrate reduction test, including 135 (77.14%) E. coli, 33 (18.85%) Klebsiella and 7 (4.0%) Proteus, verified through biochemical tests. Conclusion: Among the nitrate-reducing bacteria, fifty-one (37.22%) out of 135 E. coli, 9 (27.27 %) out of 33 Klebsiella and 2 (28.57%) out of 7 Proteus were resistant to most of fluoroquinolones group of antibiotics with moxifloxacin being the least resistant while ofloxacin being the most resistant antibiotic Malformed DNA gyrase producing mutated gyrA and gyrB genes were further detected in nitrate-reducing bacteria showing resistant to fluoroquinolones.
