O uso da biotecnologia no desenvolvimento de novos produtos pode trazer resultados positivos no tratamento de doenças, melhorando a qualidade de vida da população. Vendo esse cenário é possível relembrar os Objetivos de Desenvolvimento Sustentável da ONU. Pensando em formas de fortalecer e engajar novos produtos biotecnológicos e startups no mercado médico, esta pesquisa tem como objetivo estabelecer as percepções e o uso prático das inovações biotecnológicas. Em seguida, realizamos um estudo inicial exploratório qualitativo que visa construir hipóteses sobre o que está limitando o desenvolvimento da interface entre a biotecnologia e o campo médico, a partir de percepções sobre a biotecnologia e sua aplicação prática na perspectiva médica. Foi realizada uma entrevista semiestruturada em uma amostra foi intencional não probabilística, totalizando um N=6. As respostas foram registradas e analisadas pela plataforma Atlas.ti. Destacaram pontos do cotidiano atual como o desconhecimento do mercado biotecnológico nacional, enfatizando a produção internacional, bem como seu alto custo de produção e aquisição como se a inclusão dessas novas tecnologias estivesse longe da prática clínica.

O conhecimento e acesso de informações tecnológicas é atualmente considerado um dos mais importantes recursos econômicos para organizações empresariais, além de ser utilizado como vantagem comparativa estratégica para avaliação do cenário econômico. Embora o conhecimento estratégico de mercado seja útil na fase de desenvolvimento de um produto, duvidas emergidas durante o processo da atividade de pesquisa torna necessária a utilização de ferramentas como analises de anterioridades e prospecções tecnológicas a fim de se encontrar e reconhecer estágios de um produto perante ao cenário da sociedade, além de identificar lacunas que podem ser preenchidas com novos modelos e variações. O objetivo do atual trabalho foi realizar uma prospecção tecnológica de polímeros utilizados para o desenvolvimento de hidrogéis direcionados a bioimpressão. A análise foi realizada a partir dos dados publicados nos periódicos das bases de dados Web of Science e Scopus e também por meio de buscas de patentes nas bases do Instituto Nacional de Propriedade Intelectual (INPI), Derwernt Innovation Index e do European Patent Office (EPO) no período dos últimos 10 anos. Baseados nos resultados encontrados foi possível verificar que o colágeno, a gelatina e o alginato são os polímeros de maior utilização no desenvolvimento de biotintas. Além de se observar que há um aumento no interesse em se desenvolver novos materiais biopoliméricos para esta área sendo como indicativo o crescimento de artigos relacionados a este tema. Adicionalmente, nota-se uma pequena participação do Brasil no desenvolvimento de biotintas, contudo estudos apontaram que para a área de bioimpressão com outros materiais e desenvolvimento de técnicas, já estão sendo realizados, o que indica um futuro promissor para a área de aplicação e desenvolvimento de biotintas a nível nacional.

No. of bitstreams: 0 Previous issue date: 2021-01-01 Additive manufacturing (AM) is gaining considerable interest due to the inherent capacity of fast and low-cost prototyping of customized devices and parts. In this work, seventeen commercial thermoplastic polymer filaments (PLA, TPU, and ABS) and one PLA pellet for fused deposition modeling (FDM) were characterized and evaluated to develop a disposable sample holder to be used as a solid sampling platform in graphite furnace atomic absorption spectroscopy (GFAAS). For GFAAS application, the selection of polymer filaments took into account no or minimal mass residue, decomposition profile, the lowest thermal decomposition temperatures, and colorless filament (preferably). These conditions are essential for the rapid and complete elimination of polymeric matrix without generating residues inside the graphite tube atomizer and lower analytical blanks. Thus, the filaments and pellets were characterized by thermogravimetry, derivative thermogravimetry, differential thermal analysis, differential scanning calorimetry, Fourier-transform infrared spectroscopy and scanning electron microscopy/energy-dispersive X-ray spectroscopy. It is desirable that the sample holder can be entirely decomposed at the lowest possible temperature. The PLA pellet material was chosen because it presented glass transition 71.7 ∘C, melting temperature at 182.7 ∘C, and showed single decomposition step in the 245–325 ∘C range (with peak temperature at 304.4 ∘C), without generating mass residue. The printed sample holder was tested in a commercial spectrometer. As “proof-of-concept, the calibration curve for cobalt (0–9.0 ng) was built up with a correlation coefficient of 0.9987. The RSD was < 10%, and the quantification limit was 1.18 ng. Recoveries of Co added to water samples varied from 96–102%. No. of bitstreams: 0 Previous issue date: 2021-01-01 Cellophane film production generates cellulosic residues from scraps, edges, and low-quality films. In this work, cellophane was used as a raw material to produce cellulose acetate (CA). CA was produced through the homogeneous route for 6 to 24 hours producing materials with a degree of substitution (DS) between 2.6 and 2.9. The CA materials were characterized by Fourier transform infrared spectroscopy (FTIR), differential scanning calorimetry (DSC), thermogravimetric analysis (TGA), X-ray diffraction (XRD), and viscosity-average molecular weight. DS values (DS > 2.6) indicate the preferential formation of cellulose triacetate, and the reaction time leads to materials with different molecular weight from 32,000 to 121,000 g mol- 1 The TGA and DSC curves allowed obtaining information that, together with other techniques, confirm the formation of cellulose triacetate and the production of a material with a heterogeneous structure. The CA membrane was prepared with CA produced with 14 hours of the reaction intended to control the release rates of acetaminophen (ACP). A dense, seemingly pore-free membrane has been produced, as seen by SEM images, which allows the release of small amounts of ACP. The results were promising in the development of CA membranes for the controlled release of drugs from the chemical modification of cellophane residues. Poly(hydroxybutyrate-co-hydroxyvalerate) (PHBV) is a biodegradable, biocompatible, and non-toxic biopolymer. The biopolymer properties can be improved using cellulosic-based materials, often derived from agro-industrial residues, and promoting reuse/re-significance of a by-product for bone tissue engineering applications. Biocomposites of PHBV filled with bleached fibers of palm residues (BFPR) (0–10 % wt/wt) for 3D-printing were prepared. The scaffolds were obtained by additive manufacturing (fused deposition modeling (FDM)). The samples were characterized by stereomicroscopy, SEM, TGA, nanohardness, wettability, FTIR, and biocompatibility. Biocomposites filaments revealed homogeneous diameters, suitable for FDM. Composite filaments had thermal stability at 100–250 °C (processing did not degrade the material). The -OH groups of cellulose (enhanced by bleaching treatment) BFPR added to PHBV had advantages: optimal cell viability, wettability improvement, and slight nanohardness increase. PHBV/BFPR1 % scaffolds had an interconnected porous structure with a pore size of ~900 µm and 60 % filling.

So, the use of fibers from pine cone as reinforcement in acrylonitrile butadiene styrene (ABS) composites filaments as a potential for 3D pen was evaluated. The effect of the treatment chemical (alkaline and bleaching) was studied. A thermokinetic mixer processed the ABS/pine cone fibers (2 and 5% wt.) composites. After, the filaments were prepared by mini extruder and printing 3D pen. Filaments were characterized by Scanning Electron Microscopy (SEM), Thermogravimetry (TGA), and Infrared Spectroscopy (FTIR). Filaments revealed homogeneous diameters. The addition of 2 and 5% wt. fiber not significantly influenced the filament’s diameter and density. On the other hand, thermal stability and morphological analysis influenced the type of fiber (raw, treated, and bleached). The addition of bleached fibers to ABS increased composites’ thermal stability compared to other fibers (treated and raw). Also, inserting bleached fibers was perceptible a uniformly distributed and embedded throughout the wire cross-section compared to treated and raw fiber added to ABS due to good interfacial bonding. Results indicated that fibers were hydrogen-bonded to ABS chains and increased the filament’s density. So, it is possible to affirm that the addition of fibers from pine cone to ABS thermally improved and can be a low-cost feedstock for printing 3D pen applications. Despite the low concentration of natural fiber on the composites investigated in this work, the successful obtainment of ABS reinforced with biodegradable natural fiber, compromising neither its thermal properties nor its processability and printability, opens the possibility for future work investigation into a composite with larger fiber content. No. of bitstreams: 0 Previous issue date: 2021-01-01 Nowadays, flexible organic electronics are under intense investigation for environmentally friendly and biocompatible applications. One of the main components of electronic devices is the substrate, which gives support for building devices. There is great interest in the scientific community for the development of biocompatible and biodegradable substrates for the manufacture of these kinds of devices, aiming at technological and medical applications. In this work, we produced a flexible and transparent biosubstrate based on a gellan gum polymer by a solvent casting method to manufacture flexible organic light-emitting diodes (FOLEDs). The gellan substrate exhibited optical transparency of about 78% at 550 nm and 80% at 900 nm. In addition, the optimized indium tin oxide (ITO)/SiO2-coated gellan substrate exhibited a resistivity of 2.8 × 10-4 ω·cm and a sheet resistance of 11.2 ω/sq. These values are better or equal to those found in the literature for similar biosubstrates. Finally, the fabricated FOLEDs exhibited a maximum luminance of about 2327 cd/m2 and the current efficiency reached a maximum value of 2.9 cd/A. These characteristics reveal that this biosubstrate has interesting potential for applications in flexible green electronics, mainly due to its biocompatible properties and the results obtained by the developed FOLEDs. It is well known that Diabetes mellitus (DM) is classified as a metabolic disease and in consequence several dysfunctions are expected, such as difficulties in wound healing process. Objectives: Due to the excellent properties of the developed bacterial cellulose/alginate hydrogel (BC/ALG), this material was applied to treat a diabetic ulcer patient. Method: The treatment was carried out at the Ambulatory of Santa Casa de Misericordia de São Carlos, by applying the hydrogel twice a week during 30 days. Results: After the period of treatment, the wound showed 84% of closure reduction when compared to its initial size Conclusion: Bacterial cellulose/ alginate hydrogel presented a high potential for successful treatment of wound closure. Fused deposition modeling (FDM) has been a widely applied technology as one of the most practical tools of additive manufacturing in terms of industry 4.0. Biopolymer filaments obtained by extrusion can be a promising material for scaffold manufacturing by FDM 3D printers. In this work, composite filaments of polyhydroxybutyrate-cohydroxyvalerate (PHBV) reinforced with ZrO2·nH2O particles were obtained (1–10% wt/wt.) and characterized aiming the production of scaffolds by FDM process. ZrO2·nH2O particles were prepared and mixed to the PHBV in a mini-extruder. The pristine PHBV and composite filaments (PHBV/ZrO2) were characterized by stereomicroscopy, scanning electron microscopy (particle analysis), thermogravimetric analysis (TGA and DSC), X-ray diffractometry, Fourier transformed infrared spectroscopy , Vickers microhardness test (HV), and relative density. The addition of ZrO2·nH2O particles altered the behavior of the PHBV matrix: increased the number of ZrO2·nH2O particles in the composite filament surface, enhanced the amorphous phase and the relative density. The PHBV/7.5%ZrO2 sample presented higher microhardness. It was possible to print the filaments by FDM and the appearance of the scaffolds obtained was a cylindrical structure with rounded inner pores, contributing to the future application in regenerative medicine.

After obtaining the as-prepared composites, BC-Ce:CaP was submitted to a thermal treatment at 600 degrees C for 3 h, and Ce:CaP was characterized by scanning electronic microscopy (SEM), energy dispersive X-ray spectrometry (EDX), thermogravimetric analysis (TG), derivative thermogravimetric analysis (DTG) and X-ray diffraction (XRD). Ce:CaP presented hydroxyapatite, chlorapatite and buchwaldite (sodium calcium phosphate) phases and revealed a trabecular structure composed per nanowires with interconnected pores. Furthermore, BC-Ce:CaP and Ce:CaP show cell viability and has been suggested for use as a mineral scaffold. Boluses characterize materials with an electromagnetic radiation attenuation coefficient similar to biological tissue and used to restrict the penetration of high energy photons and electrons used in radiotherapy for the treatment of superficial tumors. The development of new materials, mainly from sustainable biotechnological routes, will contribute to increase efficiency and expand the use of these technologies. The objective of this research was to develop the “proof of concept” regarding the use of the bacterial nanocellulose membrane (BNCm) as a bolus. For this purpose, BNCm were produced, purified and subjected to the physical–chemical characterization. The radiological density (RD) and radiation attenuation potential (RAP) of the BNCm were established and compared to a commercial bolus (CB). The moldability of BNCm was established and compared to the virtual bolus of dosimetric planning. The physical–chemical analysis demonstrated the constitution of a pure, highly hydrated, homogeneous and nanostructured network of cellulose fibers. BNCm showed superiority in relation to RD and similar RAP values when compared to CB. Moldability analysis showed a profile identical to a virtual bolus. The results validate the concept of using BNCm as a highly efficient biotechnological device, aligned with the idea of sustainability, as a bolus for use in radiotherapy.

Microparticles, films and oral tablets based on alginate and gellan gum were developed also as a strategy to overcome the low aqueous solubility of morin. The systems were characterized in terms of morphological characteristics, mucoadhesion and in vitro drug release. Antibiofilm activity was analyzed for acidogenicity, microbial viability and the composition of the extracellular matrix of single-species biofilms. Scanning Electron Microscopy demonstrated that the microparticles were spherical, rough and compact. The film and the tablet presented smooth and continuous surface and in the inner of the tablet was porous. These systems were more mucoadhesive compared to the microparticles. The in vitro morin release profiles in artificial saliva demonstrated that the microparticles controlled the release better (39.6%), followed by the film (41.1%) and the tablet (91.4%) after 20 h of testing. The morin released from the systems reduced the acidogenicity, microbial viability, concentration of insoluble extracellular polysaccharides and dry weight of biofilms, when compared to the control group. The findings of this study showed that the morin has antibiofilm activity against cariogenic microorganisms. Biocompatibility, lack of immunogenicity, mechanical strength and crystallinity make BNC a highly applicable product for drug delivery and wound dressing. Previously, we demonstrated that diethylditiocarbamate (DETC), a Superoxide Dismutase 1 inhibitor, incorporated into BNC bio-curatives was effective for treating Cutaneous leishmaniasis (CL) lesions, a parasitic disease caused by Leishmania. We herein investigated the interactions between DETC and BNC. For this purpose, DETC was incorporated into BNC and thermal analysis, x-ray diffraction and Scanning electron microscopy were performed. Furthermore, in vitro DETC release and stability tests as well as degradation studies were also performed. Our results show that DETC is well incorporated into BNC, however it is short lived as suggested by degradation experiments. Future use of BNC DETC-based bio-curatives for the treatment of CL shall require further development in order to increase stability of DETC in the bio-curative. Commodity counterfeiting can be resolved by attaching labels made from biopolymers and down- and up-converting particles. Measurements of luminescence spectrum and SEM were made, proving the formation of particles and luminescence.

No. of bitstreams: 0 Previous issue date: 2022-01-01 This study aimed to encapsulate and characterise a potential anti-tuberculosis copper complex (CuCl2(INH)2.H2O:I1) into polymeric nanoparticles (PNs) of polymethacrylate copolymers (Eudragit®, Eu) developed by nanoprecipitation method. NE30D, S100 and, E100 polymers were tested. The physicochemical characterisations were performed by DLS, TEM, FTIR, encapsulation efficiency and, in vitro release studies. Encapsulation of I1 in PN-NE30D, PN-E100, and PN-S100 was 26.3%, 94.5%, 22.6%, respectively. The particle size and zeta potentials were 82.3 nm and −24.5 mV for PNs-NE30D, 304.4 nm and +18.7 mV for PNs-E100, and 517.9 nm and −6.9 mV for PNs-S100, respectively. All PDIs were under 0.5. The formulations showed an I1 controlled release at alkaline pH with 29.7% from PNs-NE30D, 7.9% from PNs-E100 and, 28.1% from PNs-S100 at 1 h incubation. PNs were stable for at least 3 months. Particularly, PNs-NE30D demonstrated moderate inhibition of M. tuberculosis and low cytotoxic activity. None of the PNs induced mutagenicity. Unlike the latter, cellulose biodegrades and may represent a carbon sink. Inspired by the multiscale architecture of cellulose, we report on all-cellulose composites comprising cellulose ether as a matrix and highly pure bacterial cellulose nanocrystals (BCNCs) as fillers. Optimum performance as a packaging material was achieved by engineering BCNC surface chemistry as well as the filler-in-matrix dispersion, targeting the replacement of unsustainable, fossil-derived plastics intended for single-use applications. Cost could pose a hurdle, eliminated through the valorization of underutilized scraps from industrial operations, which is also in line with the circular bioeconomy in terms of the integral use of biomass. As far as performance, the optically transparent hydroxypropyl methylcellulose (HPMC) films presented improved tensile strength (from 61 ± 6 to 86 ± 9 MPa) and Young's modulus (from 1.5 ± 0.2 to 2.7 ± 0.4 GPa) while reduced elongation at break (from 15 ± 2 to 12 ± 2%) and water vapor permeability (from 0.40 ± 0.02 to 0.31 ± 0.01 g mm h-1 m-2 kPa-1) when filled with only 5 wt % of (120 ± 31) nm long, (13 ± 3) nm wide, 88% crystalline BCNC. This dual, win-win effect of BCNCs on the mechanical and barrier properties of HPMC films was enabled by a suitable dispersion state, achieved via high-energy mixing, and quenched by casting. This study adds to the current literature on all-cellulose composites and helps pave the route for the technical and economical feasibilities of replacing non-renewable, non-biodegradable plastics in short-term applications by materials that are both renewable and biodegradable, that are also produced through green protocols and isolated from surplus biomass, and that still perform similarly or even better. In this work, filament based on ɛ-polycaprolactone (PCL) and containing the bioactive ceramics nanohydroxyapatite (nHap) and Laponite® (Lap) was prepared by the extrusion process. To obtain the material, a mass ratio of 89:10:1 (PCL:nHap:Lap) was used, and structural and morphological characterization was realized. In addition, cytotoxicity (using Allium cepa bulbs) and viability tests on L929 cells also were performed. The results showed that filament (diameter of 1.79 ± 0.17 mm) presented a good dispersion of nHap and Lap into polymeric matrices. Fourier transform infrared spectroscopy identified typical bands at 1720, 1091, and 1045 cm−1 addressed to PCL and nHAp, In addition, Lap was identified through dispersive energy system and X-ray diffraction analyses. All filaments did not exhibit cytotoxic effects.

Biopolymeric blends based on bacterial cellulose (BC) films modified with low molecular weight chitosan (Chi) were developed for controlled release of ciprofloxacin (Cip). Biophysical studies revealed a compatible and cooperative network between BC and Chi including deep structural changes in the BC matrix shown by spectroscopic and thermal analyses (SEM, roughness analysis, FTIR, XRD, TGA, mechanical properties and water vapor transmission rate). Incorporation of chitosan to BC matrix generated a thickening scaffold with high permeability to water vapor from 0.7 to 3.2 g mm/m2 h. Cip loaded onto the BC-Chi film showed a hyperbolic release profile with a 30% decrease in antibiotic release mediated by the presence of Chi. BC-Chi blend films containing Cip tested against Pseudomonas aeruginosa and Staphylococcus aureus showed a synergic effect of chitosan on Cip antimicrobial activity. Besides, in vitro studies revealed the lack of cytotoxicity of BC-Chi-Cip films in human fibroblasts.

Cellulose synthesized by bacteria has unique properties such as high water retention capacity, biocompatibility, biodegradability and flexibility. Nevertheless, modification of this biomaterial is required in order to obtain multifunctional materials, which may be applied in several high-value added products, as catalytic and cell culture platforms. The surface of bacterial cellulose (BC) can be modified by several approaches, namely: (i) physical treatment by plasma, (ii) adsorption of molecules onto BC surface, and (iii) chemical modification. In this sense, the aim of this study was to modify the BC surface by silanization reaction at room temperature using a mixture of ethanol and water, using two different protocols. Thus, BC membranes synthesized by Komagataeibacter xylinus were modified by adding the thiol (SH) functional group with (3-mercaptopropyl)trimethoxysilane under mild conditions. The produced materials were analyzed by elemental analysis, ATR-FTIR, TGA and SEM, and the successful modification was proven by elemental analysis and SEM.

The coacervation process of Graham’s Salt Na(PO3)n enabled the preparation of polyphosphate coacervates (CPPs) by adding different metallic ions (Mn+), and/or low-molecular-weight solvents at a specific solution, which resulted in a phase separation. The more viscous phase is termed as CPP and the less viscous one is the supernatant. CPPs have been extensively studied in recent years as a glass precursor at room temperature. Preparation at room temperature allows the preservation of the physicochemical properties of a wide range of active principles that may be used in controlled drug-delivery systems. Despite the present limitations of CPPs in terms of high hygroscopicity, this vulnerability to water is considered one of the great advantages of this material regarding applications in the controlled release of drugs and tissue engineering. Thus, in the presence of water, CPPs will dissolve in calcium and phosphate ions, which are the major inorganic constituents of bone and hydroxyapatite (HAP). It is believed that, in contact with body fluids, CPPs will degrade and, as this process occurs, they will be replaced by HAP. Therefore, CPPs configure a potential biomaterial that offers excellent properties with a wide range of biomedical applications that will be discussed and reviewed in this paper, including the structural properties and the recent improvements of materials based on CPPs and related devices.

Thus, we tested the hypothesis that edible onion film can positively influence the sensory properties, quality and increasing shelf life of beef burgers patties. The experiment was designed in a 4 × 2 factorial scheme, with two treatments (beef burgers patties with or without edible onion film) at an interval of four storage times (0, 3, 6 and 9 days) at 4◦ C. The uncoated burger patties (control) suffered the most intense color modifications during the storage (p < 0.05). The luminosity index was higher (p < 0.05) in the control at all storage times, except at day 6, and redness, yellowness and chrome were higher (p < 0.05) in the edible onion film patties at all storage times. The pH of the beef burger patties was lower (p < 0.05) at all storage times when the edible onion film was applied. For the texture profile, only the chewiness was affected, as the inclusion of the edible onion film improved the chewing of the beef burgers patties over the storage time (p < 0.05). Additionally, there was an inhibition of the microbial growth of mesophiles and psychrophiles with the application of the edible onion film in beef burgers patties. The use of edible onion film improved the perception of panelists for the variables texture, color, flavor, odor and overall appearance, and increased the preference of panelists. The edible onion film is recommended for preserving beef burgers patties, as it delays the proliferation of unwanted microorganisms, stabilizes and improves the color parameters and sensory attributes, and increases the overall acceptance of the consumer.
