Peanut is a plant characterised by belowground fruiting that absorbs nutrients not only through its roots but also through its pods. However, little is currently known regarding the species of bacteria that contribute to nutrient absorption and utilisation in this plant's pod and root zones. This study examined the effects of root and pod area nitrogen (N) fertiliser application on peanut rhizosphere and geocarposphere microbial communities and functions. Using two peanut cultivars [nodulated Huayu 22 (H) and non-nodulated NN-1 (B)], we applied the following four treatments: no N fertiliser (HT1, BH1); N applied to geocarposphere soil (HT2, BT2); N applied to rhizosphere soil (HT3, BT3), and N applied to both rhizosphere and geocarposphere soil (HT4, BT4). The results revealed that compared with HT1 and BT1, the HT3, HT4, BT3, and BT4 treatments promoted increases in total plant accumulated N of 11.2, 30.1, 38.5, and 9.9%, respectively. Moreover, N input contributed to an increase in the abundance of bacteria colonising the surrounding pods, which differed significantly from bacteria colonising the rhizosphere. Among the top four bacterial phyla detected, we recorded a significant increase in the relative abundances of Proteobacteria and Gemmatimonadetes in response to treatments HT2 and HT4, whereas the highest relative abundances of Acidobacteria and Actinobacteria were detected in HT3 plants. Regarding cultivar B, we detected increases in the relative abundances of Bacteroidetes and Gemmatimonadetes in response to the BT2 and BT4 treatments, and in the relative abundance of Actinobacteria in BT3 treated soil. The findings of FAPROTAX functional analysis revealed clear differences among the T2, T4, and T3 treatments of two peanut cultivars concerning the functional groups with the highest relative abundances. These findings will make a considerable contribution to enhancing our understanding of the effects of N fertilisation on soil microbial structure and function in the rhizosphere and geocarposphere of peanuts and can provide a basis for identifying beneficial bacteria for promoting N utilisation and yield enhancement.

The effects of large granular slow-controlled release fertiliser prepared by a double coating of sulfur and sodium alginate on peanut growth, nitrogen fertiliser utilisation, and soil microbial community were investigated through peanut pot experiments, with a view to providing a theoretical and practical basis for the development of large granular slow-controlled release fertiliser. The results showed that the homemade large granular fertiliser could promote the root development of peanuts, and the root volume increased by 45.10% compared with the uncoated fertiliser at the fruiting stage. At the same time, the soil NH4+-N and NO3--N content were reduced at the seedling stage and increased at the fruiting stage to achieve the fertiliser's slow and controlled release effect. A significant contribution to the net photosynthetic rate was made for growth development and yield in the middle and late stages. Pod dry weight was significantly higher at the blooming stage than uncoated fertiliser, 4.8% higher at the fruiting stage, and 22.9% higher in nitrogen use efficiency (NUE). In terms of microbial bacterial communities, the large granular slow-release fertiliser promoted the diversity of the treated bacterial communities to some extent, with little difference in the relative abundance of soil bacterial communities. These results showed that a one-time application of homemade large granular slow-release fertiliser positively affected peanuts in terms of yield increase, promotion of nitrogen uptake and improved nitrogen utilisation under nitrogen application with urea equivalent, but the overall effect on soil microbial community was small.

Diversifying cultivation management, including different crop rotation patterns and soil amendment, are effective strategies for alleviating the obstacles of continuous cropping in peanut (Arachis hypogaea L.). However, the peanut yield enhancement effect and temporal changes in soil chemical properties and microbial activities in response to differential multi-year crop rotation patterns and soil amendment remain unclear. In the present study, a multi-year localization experiment with the consecutive application of five different cultivation managements (including rotation with different crops under the presence or absence of external quicklime as soil amendment) was conducted to investigate the dynamic changes in peanut nutrient uptake and yield status, soil chemical property, microbial community composition and function. Peanut continuous cropping led to a reduction in peanut yield, while green manure-peanut rotation and wheat-maize-peanut rotation increased peanut yield by 40.59 and 81.95%, respectively. A combination of quicklime application increased yield by a further 28.76 and 24.34%. Alterations in cultivation management also strongly affected the soil pH, nutrient content, and composition and function of the microbial community. The fungal community was more sensitive than the bacterial community to cultivation pattern shift. Variation in bacterial community was mainly attributed to soil organic carbon, pH and calcium content, while variation in fungal community was more closely related to soil phosphorus content. Wheat-maize-peanut rotation combined with quicklime application effectively modifies the soil acidification environment, improves the soil fertility, reshapes the composition of beneficial and harmful microbial communities, thereby improving soil health, promoting peanut development, and alleviating peanut continuous cropping obstacles. We concluded that wheat-maize-peanut rotation in combination with quicklime application was the effective practice to improve the soil fertility and change the composition of potentially beneficial and pathogenic microbial communities in the soil, which is strongly beneficial for building a healthy soil micro-ecology, promoting the growth and development of peanut, and reducing the harm caused by continuous cropping obstacles to peanut.

The accumulation of microplastics in agricultural soil brings unexpected adverse effects on crop growth and soil quality, which is threatening the sustainability of agriculture. Biochar is an emerging soil amendment material of interest as it can remediate soil pollutants. However, the mechanisms underlying biochar alleviated the toxic effects of microplastics in crops and soil were largely unknown. Using a common economic crop, peanut as targeted species, the present study evaluated the plant physiologica and molecular response and rhizosphere microbiome when facing microplastic contamination and biochar amendment. Transcriptome and microbiome analyses were conducted on peanut root and rhizosphere soil treated with CK (no microplastic and no biochar addition), MP (1.5% polystyrene microplastic addition) and MB (1.5% polystyrene microplastic+2% peanut shell biochar addition). The results indicated that microplastics had inhibitory effects on plant root development and rhizosphere bacterial diversity and function. However, biochar application could significantly promote the expressions of key genes associated with antioxidant activities, lignin synthesis, nitrogen transport and energy metabolism to alleviate the reactive oxygen species stress, root structure damage, nutrient transport limitation, and energy metabolism inhibition induced by microplastic contamination on the root. In addition, the peanut rhizosphere microbiome results showed that biochar application could restore the diversity and richness of microbial communities inhibited by microplastic contamination and promote nutrient availability of rhizosphere soil by regulating the abundance of nitrogen cycling-related and organic matter decomposition-related microbial communities. Consequently, the application of biochar could enhance root development by promoting oxidative stress resistance, nitrogen transport and energy metabolism and benefit the rhizosphere microecological environment for root development, thereby improved the plant-soil system health of microplastic-contaminated agroecosystem.

We aimed to clarify the nutrient allocation strategies of Cunninghamia lanceolata (CL) and Schima superba (SS) and their differences in N uptake and utilization under acidic stress and heterogeneous allocation of N. One-year-old seedlings of CL and SS were cultured in sands with three pH levels (pH rate 4.5, 5.5, and 6.5) and five different N form ratios (NH4+-N to NO3−-N ratios 10:0, 7:3, 5:5, 3:7, and 0:10) in the greenhouse. The C, N, and P contents and C:N:P stoichiometry in leaves, total biomass, and NH4+-N and NO3−-N contents in the cultivation substrate were analyzed after 160 days. At the pH rate of 4.5, the N contents of CL and SS significantly decreased. However, the N contents of SS increased at the pH rate of 5.5. At the pH rate of 5.5, the N content of CL and SS increased under the cultivation substrate, with ratios of 10:0 and 7:3, and 3:7 and 0:10, respectively. The N content of CL was significantly positively correlated with its P content. The C:P ratios of CL and SS were significantly positively correlated with their C:N ratios and N:P ratios. Under the same treatment, the differences in C:N:P stoichiometry were more significant between CL and SS at the pH rate of 5.5. The NH4+-N content of the cultivation substrate in CL and SS was higher than the NO3−-N content. At the ratio of 7:3, the C:N stoichiometric ratio of CL and C:N:P stoichiometric ratio of SS were negatively correlated with total biomass. At the pH rate of 5.5, cultivation environments with high NH4+-N and NO3−-N amount respectively increased the N content of CL and SS. The CL is susceptible to N limitation, and SS is susceptible to N and P limitation.

BackgroundPokémon GO, an augmented reality game with widespread popularity, can potentially influence players’ physical activity (PA) levels and psychosocial well-being. ObjectiveThis review aims to systematically examine the scientific evidence regarding the impact of Pokémon GO on PA and psychosocial well-being in children and adolescents. MethodsUsing the PRISMA (Preferred Reporting Items for Systematic Reviews and Meta-Analyses) guidelines and the GRADE (Grading of Recommendations, Assessment, Development, and Evaluations) framework, we conducted keyword and reference searches in the PubMed, CINAHL, Web of Science, and Scopus databases. We performed title and abstract screening, full-text review, evidence synthesis, and identified research gaps. ResultsOur review included 10 studies that explored the effect of Pokémon GO on PA or psychosocial well-being among children and adolescents. These studies used diverse designs across multiple countries and regions. Pokémon GO use measures encompassed frequency, experience, adherence, and motivation. PA assessment methods ranged from self-reported questionnaires to technology-based evaluations and qualitative approaches. Psychosocial well-being measures included emotional intelligence, personal well-being, self-control, emotionality, and sociability. In general, the estimated impact of Pokémon GO on PA was positive, with gaming elements and engagement correlating with increased PA levels. However, the effect on psychosocial well-being presented mixed results, with positive associations for sociability but a complex relationship involving well-being and internet gaming disorder. The limitations of these studies comprised the absence of randomized controlled trials, heterogeneity in study designs and outcome measures, and potential confounding bias. ConclusionsOverall, Pokémon GO tends to positively affect PA levels, while the impact on psychosocial well-being remains complex and requires further investigation. Future research should investigate the mechanisms connecting Pokémon GO use with PA and psychosocial well-being and the potential risks of excessive gameplay. These findings can help inform public health interventions to harness gaming technologies for promoting PA and enhancing well-being among the younger generation. BackgroundThe bacterial communities of the root surface and rhizosphere play a crucial role in the decomposition and transformation of soil nitrogen (N) and are also affected by soil N levels and distribution, especially the composition and diversity, which are sensitive to changes in the environment with high spatial and temporal heterogeneity of ammonium N (NH4+-N) and nitrate N (NO3--N).MethodsOne-year-old seedlings of Cunninghamia lanceolata and Schima superba were subjected to N stress (0.5 mmol L-1) and normal N supply (2 mmol L-1), and five different N form ratios (NH4+-N to NO3--N ratio of 10:0, 0:10, 8:2, 2:8, and 5:5) were created. We analyze the changes in composition and diversity of bacteria in the root surface and rhizosphere of two tree species by high-throughput sequencing.ResultsDifferences in the composition of the major bacteria in the root surface and rhizosphere of C.lanceolata and S. superba under N stress and N form ratios were not significant. The dominant bacterial phyla shared by two tree species included Proteobacteria and Bacteroidota. Compared to normal N supply, the patterns of diversity in the root surface and rhizosphere of two tree species under N stress were distinct for each at five N form ratios. Under N stress, the bacterial diversity in the root surface was highest at NH4+-N to NO3--N ratio of 10:0 of C. lanceolata, whereas in the root surface, it was highest at the NH4+-N to NO3--N ratio of 0:10 of S. superba. The NH4+-N to NO3--N ratio of 5:5 reduced the bacterial diversity in the rhizosphere of two tree species, and the stability of the bacterial community in the rhizosphere was decreased in C. lanceolata. In addition, the bacterial diversity in the root surface was higher than in the rhizosphere under the N stress of two tree species.ConclusionThe bacterial compositions were relatively conserved, but abundance and diversity changed in the root surface and rhizosphere of C. lanceolata and S. superba under N stress and different N form ratios. The heterogeneity of ammonium and nitrate N addition should be considered for N-stressed environments to improve bacterial diversity in the rhizosphere of two tree species.

Soil compaction is one of the crucial factors that restrains the root respiration, energy metabolism and growth of peanut (Arachis hypogaea L.) due to hypoxia, which can be alleviated by ventilation. We therefore carried out a pot experiment with three treatments: no ventilation control (CK), (2) ventilation volumes at 1.2 (T1), and 1.5 (T2) times of the standard ventilation volume (2.02 L/pot). Compared to no-ventilation in compacted soil, ventilation T1 significantly increased total root length, root surface area, root volume and tips at the peanut anthesis stage (62 days after sowing), while T2 showed a negative impact on the above-mentioned root morphological characteristics. At the podding stage (S2, 95 days after sowing), both ventilation treatments improved root morphology, especially under T1. Compared to CK, both ventilation T1 and T2 decreased the activities of enzymes involving the anaerobic respiration, including root lactate dehydrogenase, pyruvate decarboxylase and alcohol dehydrogenase. The activities of antioxidant enzymes of root superoxide dismutase, peroxidase and catalase also decreased at S1, while superoxide dismutase and peroxidase significantly increased under T1 at S2. The ventilation of compacted soil changed soil nitrogen-fixing bacterial communities, with highest bacterial alpha diversity indices under T1. The Pearson correlation analyses indicated a positive relationship between the relative abundance of Bradyrhizobiaceae and root activity, and between unclassified_family of Rhizobiales and the root surface area, while Enterobacteriaceae had a negative impact on the root nodule number. The Pearson correlation test showed that the root surface, tips and activity positively correlated with root superoxide dismutase and peroxidase activities. These results demonstrate that soil ventilation could enhance plant root growth, the diversity and function of soil nitrogen-fixing bacterial communities. The generated results from this present study could serve as important evidence in alleviating soil hypoxia caused by compaction.

Exogenous carbon (C) not only regulates plant growth but also provides energy for microbes and improves the soil environment. We hypothesised that exogenous C could improve plant growth by affecting the soil environment. Therefore, pot experiments were conducted and peanut cvs. Huayu 22(H) and NN-1(B) were used under three different treatments (the control, single nitrogen (N), and N combined with glucose (CN)). The results showed that the abundance and diversity of N-fixing bacteria are obviously influenced by the C and N, and exogenous C can promote the restoration of microbial diversity. The relative abundances of Burkholderiales were increased under HCN and BCN to 9.8% and 9.5%, respectively, compared to the control (3.9%, 2.5%). The abundance of N fixation bacteria increased mainly due to the soil nutrient change. In comparison with the single N treatment, the addition of the C significantly decreased the soil NH4+-N and NO3--N contents by 31.0% and 13.3%, respectively. And the activities of soil urease and nitrogenase were significantly increased. Compared to the control, single N significantly limited the root development, while the addition of C played a promoting role in root growth. Plant N accumulation increased compared with the control, but there was no significant difference between N treatment and CN treatment. These results indicated that exogenous C promoted soil microorganism activity and strengthened plant growth by changing the soil environment.

Abstract Background Phosphorus (P) is one of the most essential macronutrients for crops. The growth and yield of peanut (Arachis hypogaea L.) are always limited by P deficiency. However, the transcriptional and metabolic regulatory mechanisms were less studied. In this study, valuable phenotype, transcriptome and metabolome data were analyzed to illustrate the regulatory mechanisms of peanut under P deficiency stress. Result In present study, two treatments of P level in deficiency with no P application (–P) and in sufficiency with 0.6 mM P application (+ P) were used to investigate the response of peanut on morphology, physiology, transcriptome, microRNAs (miRNAs), and metabolome characterizations. The growth and development of plants were significantly inhibited under –P treatment. A total of 6088 differentially expressed genes (DEGs) were identified including several transcription factor family genes, phosphate transporter genes, hormone metabolism related genes and antioxidant enzyme related genes that highly related to P deficiency stress. The Gene Ontology (GO) and Kyoto Encyclopedia of Genes and Genomes (KEGG) enrichment analyses indicated that 117 genes were annotated in the phenylpropanoid biosynthesis pathway under P deficiency stress. A total of 6 miRNAs have been identified significantly differential expression between + P and –P group by high-throughput sequencing of miRNAs, including two up-regulated miRNAs (ahy-miR160-5p and ahy-miR3518) and four down-regulated miRNAs (ahy-miR408-5p, ahy-miR408-3p, ahy-miR398, and ahy-miR3515). Further, the predicted 22 target genes for 6 miRNAs and cis-elements in 2000 bp promoter region of miRNA genes were analyzed. A total of 439 differentially accumulated metabolites (DAMs) showed obviously differences in two experimental conditions. Conclusions According to the result of transcripome and metabolome analyses, we can draw a conclusion that by increasing the content of lignin, amino acids, and levan combining with decreasing the content of LPC, cell reduced permeability, maintained stability, raised the antioxidant capacity, and increased the P uptake in struggling for survival under P deficiency stress.

Abstract Background Hypospadias is a congenital anomaly of the male urogenital system. Genetics factors play an important role in its pathogenesis. To search for potential causal genes/variants for hypospadias, we performed exome sequencing in a pedigree with three patients across two generations and a cohort of 49 sporadic patients with hypospadias. Results A novel BRAF variant (NM_004333.6: c.362C > A) was found to co-segregate with the hypospadias phenotype in the disease pedigree. In cells overexpressing the BRAF mutant, the phosphorylation level of p38 MAPK was significantly increased as compared with the cells overexpressing the wild-type BRAF or RASopathy-related BRAF mutant. This variant further led to a reduced transcription level of the SRY gene, which is essential for the normal development of the male reproductive system. In the cohort of sporadic patients, we identified two additional variants in p38 MAPK signaling-related genes (TRIM67 and DAB2IP) potentially associated with hypospadias. Conclusion Our study expands the phenotypic spectrum of variants affecting p38 MAPK signaling toward the involvement of hypospadias.

Peanut (Arachis hypogaea L.) is an important oil crop globally because of its high edible and economic value. However, its yield and quality are often restricted by certain soil factors, especially nitrogen (N) deficiency, and soil compaction. To explore the molecular mechanisms and metabolic basis behind the peanut response to N deficiency and soil compaction stresses, transcriptome and metabolome analyses of peanut root were carried out. The results showed that N deficiency and soil compaction stresses clearly impaired the growth and development of peanut's aboveground and underground parts, as well as its root nodulation. A total of 18645 differentially expressed genes (DEGs) and 875 known differentially accumulated metabolites (DAMs) were identified in peanut root under differing soil compaction and N conditions. The transcriptome analysis revealed that DEGs related to N deficiency were mainly enriched in “amino acid metabolism”, “starch and sucrose metabolism”, and “TCA cycle” pathways, while DEGs related to soil compaction were mainly enriched in “oxidoreductase activity”, “lipids metabolism”, and “isoflavonoid biosynthesis” pathways. The metabolome analysis also showed significant differences in the accumulation of metabolisms in these pathways under different stress conditions. Then the involvement of genes and metabolites in pathways of “amino acid metabolism”, “TCA cycle”, “lipids metabolism”, and “isoflavonoid biosynthesis” under different soil compaction and N deficiency stresses were well discussed. This integrated transcriptome and metabolome analysis study enhances our mechanistic knowledge of how peanut plants respond to N deficiency and soil compaction stresses. Moreover, it provides new leads to further investigate candidate functional genes and metabolic pathways for use in improving the adaptability of peanut to abiotic stress and accelerating its breeding process of new stress-resistant varieties.

Aging is closely related to many diseases and is a long-term challenge that humans face. The oxidative damage caused by the imbalance of free radicals is an important factor in aging. In this study, we investigate the antioxidant and antiaging activities of fermented coix seed polysaccharides (FCSPs) via in vitro and in vivo experiments. The FCSPs were extracted by fermenting coix seed with Saccharomyces cerevisiae for 48 h and utilizing water-extracted coix seed polysaccharides (WCSPs) as a control. Their antiaging activity and mechanism were evaluated based on the antiaging model organism Caenorhabditis elegans (C. elegans). The results showed that the molecular weight of the FCSPs extracted by fermentation was smaller than that of the WCSPs, making them more easily absorbed and utilized. At a concentration of 5 g/L, the FCSPs’ capacity to scavenge the DPPH·, ABTS+·, OH·, and O2−· radicals was greater than the WCSPs’ capacity by 10.09%, 14.40%, 49.93%, and 12.86%, respectively. Moreover, C. elegans treated with FCSPs exhibited higher antioxidant enzyme activities and a lower accumulation of malonaldehyde. By inhibiting the expression of the pro-aging genes daf-2 and age-1, and upregulating the expression of the antiaging genes daf-16, sod-3, skn-1, and gcs-1 in the insulin/insulin-like growth factor-1 (IIS) signaling pathway, the FCSPs could effectively enhance stress tolerance and delay C. elegans aging. The lifespan of C. elegans in the FCSPs group was 5.91% higher than that of the WCSPs group. In conclusion, FCSPs exert better antioxidant and antiaging effects than WCSPs, which can act as a potential functional ingredient or supplement in food.

Our previous work demonstrated that Piwil2 reactivated by the human papillomavirus oncoproteins E6 and E7 may reprogram somatic cells into tumor-initiating cells (TICs), which contribute to cervical neoplasia lesions. Maintaining the stemness of TICs is critical for the progression of cervical lesions. Here, we determined that canonical Wnt signaling was aberrantly activated in HaCaT cells transfected with lentivirus expressing Piwil2 and in cervical lesion specimens of low-grade squamous intraepithelial lesion, high-grade squamous intraepithelial lesion, and invasive carcinoma. Blocking the β-catenin and CREB binding protein interaction with ICG-001 significantly downregulated the reprogramming factors c-Myc, Nanog, Oct4, Sox2, and Klf4, thus leading to cell differentiation and preventing tumorigenicity in Piwil2-overexpressing HaCaT cells. Similarly, Piwil2 also critically regulated the canonical Wnt signaling pathway in cervical cancer. We further demonstrated that ICG-001 increased cisplatin sensitivity and significantly suppressed tumor growth of cervical cancer alone or in combination with cisplatin both in vitro and in vivo. The β-catenin/ CREB binding protein-mediated transcription activated by Piwil2 is essential for the maintenance of TICs, therefore contributing to the progression of cervical oncogenesis.

As a natural plat growth accelerator, seaweed fertilizer can be used as a good agricultural practice for sustainable and organic crop production. To investigate the effects of seaweed liquid fertilizer on the growth of peanut, the following experiments were carried out in 2020. Three types of seaweed liquid fertilizers were prepared by enzymatic hydrolysis with papain enzymatic and cellulase enzymatic. The peanut pot experiment was set up with five treatments (applying water, seaweed solution, papain enzymatic liquid fertilizer, cellulase enzymatic liquid fertilizer, two - mixed enzymatic liquid fertilizer). Relevant data about photosynthesis, agronomic traits and root system were measured. The results show that the overall growth of peanut fertilized with seaweed liquid fertilizer is better than that of the control group. Especially for the two-mixed enzymes enzymatic liquid fertilizer, the values of photosynthesis, leaf chlorophyll content, main stem height, lateral branch length and dry matter accumulation are all increased by 13.8%, 11.4%, 45.7%, 225.0% and 8.7% respectively, compared with the control group. Meanwhile, the promotion of peanut root morphology by spraying cellulase enzymatic liquid fertilizer was more pronounced. It increased by 156.7%, 103.0%, 58.3% and 101.9% in root length, root surface area, root volume and root tip number. This experiment provides a theoretical and experimental basis for the preparation of seaweed fertilizer. It also develops the utilization of seaweed resources.

ObjectiveTo report a novel multi-port containment (NMC) system for laparoscopic power morcellation to prevent tumoral spread and to evaluate its safety, validity, and feasibility.MethodsThis retrospective study included women who underwent laparoscopic myomectomy (LM) between January 2014 and August 2020 at a single academic institution. The NMC system was used in the study group (n = 193); the control group underwent unprotected LM (n = 1753).ResultsAfter 1:1 propensity score matching, no significant differences in the baseline characteristics were observed between 193 matched pairs. Bag damages were detected in two cases in the study group before morcellation, and the NMC systems were replaced. There were no significant differences between the two groups in terms of the complications, total operative time, estimated blood loss, or postoperative hospitalization duration. In the study group, all operations were completed and no system rupture or leakage was observed. The median follow-up times were 21 and 54 months in the study and control groups, respectively. There was no peritoneal tissue spread in the study group. However, three (3/5, 0.6%) and six (6/1,753, 0.3%) patients in the control group experienced malignant and benign peritoneal tissue spread, respectively.ConclusionThe NMC system for laparoscopic power morcellation is valid, safe, and feasible for preventing a tumor spread.

Abstract Background Genetically encoded fluorescent proteins are often used to label proteins and study protein function and localization in vivo. Traditional cloning methods mediated by restriction digestion and ligation are time-consuming and sometimes difficult due to the lack of suitable restriction sites. Invitrogen developed the Gateway cloning system based on the site-specific DNA recombination, which allows for digestion-free cloning. Most gateway destination vectors available for use in plants employ either the 35S or ubiquitin promoters, which confer high-level, ubiquitous expression. There are far fewer options for moderate, cell-type specific expression. Results Here we report on the construction of a Gateway-compatible cloning system (SWU vectors) to rapidly tag various proteins and express them in a cell-type specific manner in plants. We tested the SWU vectors using the HISTONE (H2B) coding sequence in stable transgenic plants. Conclusions The SWU vectors are a valuable tool for low cost, high efficiency functional analysis of proteins of interest in specific cell types in the Arabidopsis root.

Autism spectrum disorders (ASD) are generally defined as a development disorder typically characterized by social interaction and communication ailments and stereotyped actions due to combined genetic and environmental factors. Different critical aspects contribute to ASD, and consensus has been reached among autism researchers about its predominant genetic factors. However, the pathogenesis of ASD has not been fully revealed, and a systematic method must be developed to identify the genes related to this disease. Here, we predicted new ASD-associated genes by random walk method on the basis of prior-known ASD genes from AutDB. New genes such as RAC3, AC1 (ADCY1), PKC (PRKC) gamma, EPH receptor A5, WNT3A, calretinin, RAS-R, KLF4, and calpain 3 were found to play an irreplaceable role in ASD pathogenesis.

Background: Low-dose methylenetetrahydrofolate (LD-MTX) has been widely used for the treatment of the ectopic pregnancy (EP) for many decades, and related severe adverse toxic effects are rare. Current studies have shown that the polymorphisms of methylenetetrahydrofolate reductase (MTHFR) gene can decrease the MTX clearance, leading to the metabolite accumulation. However, there is a lack of literature report on an MTHFR gene polymorphism associated with adverse toxic effects resulting from the use of LD-MTX in an EP.Case Presentation: We report a rare case of a 38-year-old female who developed persistent fever, grade IV myelosuppression, skin lesions, mucositis, and liver injury after single dose of LDMTX to treat EP. The personalized genetic testing showed that MTHFR TT (677C>T) and MTHFR AA (1298A>C) were detected. Gradually, the symptoms improved after calcium leucovorin (CF) rescue, continuous renal replacement therapy (CRRT), promoting blood system regeneration, and multiple supportive treatments.Conclusion: This is the first report on the serious adverse toxic effects of LD-MTX on an EP patient with MTHFR mutations. We aim to alert obstetricians and gynecologists to this rare condition. The unexpected life-threatening toxicity with LD-MTX should be highly considered and recognized early. In particular, some easily overlooked gastrointestinal, skin, and mucosal symptoms occur earlier than severe myelosuppression. When toxic effects are suspected, detecting the polymorphisms of an MTHFR gene and monitoring MTX concentration in blood could assist us to formulate individualized and active treatments.

The inorganic nitrogen (N) that can be absorbed and utilized by plants is mainly ammonium N (NH4+-N) and nitrate N (NO3−-N), which may affect seedlings’ root morphology and growth through its heterogeneous distribution. Root morphology and seedling growth were investigated in a subtropical major conifer (Cunninghamia lanceolata) and a broadleaf tree species (Schima superba) under five different NH4+-N to NO3−-N ratios (10:0, 0:10, 7:3, 3:7, 5:5). Results: (1) While both species developed thinner roots under the treatment with a high NO3−-N concentration, the roots of C. lanceolata were longer than those of S. superba. In contrast, the roots of both species were thicker under the treatment with a high NH4+-N concentration, with those in S. superba being much longer than those in C. lanceolata. (2) The mixed NH4+-N and NO3−-N treatments were more conducive to the aboveground growth and biomass accumulation of both tree species and the underground growth of S. superba. N sources with high NO3−-N concentrations were more suitable for underground growth in C. lanceolata seedlings and aboveground growth in S. superba seedlings. Under the N sources with high NH4+-N concentrations, C. lanceolata tended to develop aboveground parts and S. superba tended to develop underground parts. (3) The roots of the two tree species adopted the expansion strategy of increasing the specific root length and reducing the root tissue density under the N sources with high NO3−-N concentrations but the opposite with high NH4+-N concentrations. The root-to-shoot ratio of C. lanceolata increased under high NO3−-N concentrations, while that of S. superba increased under high NO3−-N concentrations. These results indicate that the responses of root morphology to different N forms are species-specific. Furthermore, according to the soil’s N status, NH4+-N can be appropriately applied to C. lanceolata and NO3−-N to S. superba for cultivating seedlings.

Initial translation interference refers to the impact of the initial translation on other-revisers. The factor of initial translation interference on reviser trainees has not been examined systematically. The current research investigates two related questions: How does trainees’ revision performance relate to their translation performance? And how is trainees’ revision performance impacted by the initial translated text they are provided with? A mixed research design of quantitative and semi-qualitative research methods is used for a more comprehensive analysis and a higher validity of the findings. The results suggest that good translator trainees tend to be also competent in revision, and poor translator trainees tend to be also weak at revision. However, no significant correlation is found for the intermediate group. The trainees tend to find revision tasks more challenging than the translation tasks and negative interference of the initial translation is prevalent both lexically and syntactically. They are very likely to be distracted and misled by the initial translation. The methodology and findings of this study have implications for translation researchers as well as educators.

Membrane protein is an important type of proteins and has been confirmed to play essential roles in various cellular processes. Based on their intramolecular arrangements and positions in a cell, they can be categorized into several types. However, it is time- and cost-consuming to recognize the type of a given membrane protein via traditional biophysical methods. In view of this, several computational models have been proposed in recent years. Most models adopted various information of membrane proteins, such as their sequences, domain profiles, physiochemical properties, etc. to extract different features, which were fed into downstream classification algorithms. In this study, we built two novel prediction models, which incorporated novel feature extraction methods, i.e., network embedding methods. To this end, several protein networks were constructed using the protein-protein interaction information retrieved from STRING. Among these models, one model was constructed based on features obtained by applying Mashup on seven protein networks, another model was built using features yielded by Node2Vec on one comprehensive protein network. Each model adopted random forest as the classification algorithm and employed the Synthetic Minority Over-sampling Technique (SMOTE) to overcome the influence yielded by the great difference on sizes of different membrane protein types. Furthermore, two models were integrated into one model to improve the predicted quality. The test results shown that the integrated model had good performance and was superior to any individual model. Also, we compared our models with some previous models, suggesting that our models were competitive.

Background: TNF-α has a wide range of functions in processes within the ovaries, including follicular development, ovarian hormones synthesis, ovulation, granulosa cell necroptosis, and follicular atresia. Innate immune response in the ovaries is the main source of TNF-α. The purpose of this study is to explore the dose- and time-dependent interactions between TNF-α and human granulosa cells. Methods: The follicular fluids (FF) were obtained from infertile patients undergoing IVF with PCOS (n = 20) and healthy women (n = 19). Granulosa cells were cultured and treated with varying concentration of recombinant human TNF-α. Cell viability was detected by CCK-8 method, the levels of TNF-α, estradiol, and inhibin were measured with ELISA, and cell apoptosis was detected by flow cytometry. Q-PCR and western-blot were used to evaluate the expression of TNFR1/2, P450, Inhibin α/βA, and caspase 3. Results: The level of TNF-α increased gradually with the development of follicles and was remarkably higher in the FF of PCOS patients than in that of healthy women. Treatment with TNF-α as low as 0.2 ng/mL promoted the proliferation of granulosa cells, thus upregulating P450 aromatase and inhibin A. On the other hand, a high concentration of TNF-α (0.4, 0.8, 2, and 8 ng/mL) showed dose- and time-dependent inhibitory effects on cell viability and E2 and inhibin A secretion. However, no marked differences in the expression of the two types of TNFR were observed in TNF-α-treated granulosa cells. Conclusions: The data presented here verify a biphasic effect of TNF-α on human ovarian endocrine and follicular development.

Objectives: Explore the diagnostic and prognostic value of cyclooxygenase-2 and wnt3a/β-catenin pathway in endometrial cancer.Methods: A prospective cohort study of 93 women underwent hysterectomy at the China-Japan Friendship Hospital (61 patients with primary endometrial carcinoma, and 32 control patients with uterine prolapse or leiomyoma of uterus). Cox2 and β-catenin expression were determined by immunohistochemistry. The serum levels of cox2 and wnt3a were detected via ELISA assays.Results: Patients with endometrial cancer showed overexpression of cox2 and β-catenin, as well as significantly higher serum levels of cox2 and wnt3a. The serum cox2 level, which is highly significant in predicting the risk of disease progression (RR, 9.617, 95% confidence interval, 1.162–79.622, P = 0.036), showed good diagnostic and prognostic potential, with cut-off of 55 U/L, but alongside β-catenin expression in tissues, were related to poor prognosis (RR, 12.426; 95% confidence interval, 1.618–95.450; P = 0.015).Conclusion: Serum levels of cox2 and wnt3a exhibited diagnostic value for endometrial cancer. Cox2 serum levels and β-catenin expression also showed potential value of prognostic prediction. Cox2 serum levels might be a potential marker for early diagnosis and prognosis prediction in endometrial cancer.

Ovarian cancer is the leading cause of death among gynecological malignancies. Cyclooxygenase 2 is widely expressed in various cancer cells and participates in the occurrence and development of tumors by regulating a variety of downstream signaling pathways. However, the function and molecular mechanisms of cyclooxygenase 2 remain unclear in ovarian cancer. Here, we demonstrated that cyclooxygenase 2 was highly expressed in ovarian cancer and the expression level was highly correlated with ovarian tumor grades. Further, ovarian cancer cells with high expression of cyclooxygenase 2 exhibit enhanced proliferation and invasion abilities. Specifically, cyclooxygenase 2 promoted the release of prostaglandin E2 upregulated the phosphorylation levels of phospho-nuclear factor-kappa B p65. Celecoxib, AH6809, and BAY11-7082 all can inhibit the promoting effect of cyclooxygenase 2 on SKOV3 and OVCAR3 cell proliferation and invasion. Besides, celecoxib inhibited SKOV3 cell growth in the xenograft tumor model. These data suggest that high expression of cyclooxygenase 2 promotes the proliferation and invasion of ovarian cancer cells through the prostaglandin E2/nuclear factor-kappa B signaling pathway. Cyclooxygenase 2 may be a potential therapeutic target for the treatment of ovarian cancer.

Lower nitrogen recovery efficiency (NRE) and negative environmental impacts caused by excessive nitrogen (N) fertilization threaten the sustainability of agriculture. Efficient and appropriate fertilization practices are extremely important to achieve higher crop yield with minimum N loss. A field microplot experiment was conducted in a wheat-maize rotation system in Shaanxi province, at North China Plain, using the 15N isotope tracer technique to qualify the different annual N managements in terms of crop yield, NRE, N distribution in plant-soil, and N losses to optimize the N management. The experiment included four N treatments: conventional practice with 510 kg ha−1 annually in four applications (N1), and three optimized N treatments, reducing N rate to 420 kg ha−1, adjusting topdressing fertilizer times and using slow-release fertilizer (SRF) (N2, N3, N4). The results showed that the grain yield and N uptake did not differ significantly among treatments. N from fertilizer taken up (Ndff) by wheat was not affected by N management; however, in maize, Ndff performed differently. Optimized treatments significantly decreased the Ndff as compared to N1 treatment. Furthermore, NRE of wheat and annual nitrogen recovery efficiency (annual NRE) did not differ among treatments in 2016 but significantly increased in 2017 compared to N1. Annual NRE in 2017 was similar to that obtained for wheat. For maize, optimized N managements decreased the NRE in N3 and N4 treatments of two years. Potential losses in wheat were also similar amongst treatments, but in maize, N3 and N4 had lower residual N in the soil’s top 60 cm but resulted in higher potential losses than N1 and N2. Overall, our results demonstrate that applying 420 kg N ha−1 annually in three applications and combining SRF and urea are effective to sustain crop yield, improve the efficiency of N usage by maize, and reduce N losses in this region.

In order to reduce the harm caused by corrosion to the normal operation of offshore oil and gas pipelines, the effects of the synergistic effect of Cl- and SO42- on the corrosion behavior of 316L stainless steel in the simulated South China Sea marine environment were investigated through the AC impedance technique and the cyclic polarization technique in electrochemical tests, and the corrosion morphology of 316L stainless steel surface was characterized by scanning electron microscopy(SEM). Results showed that when the SO42- content was constant, an increase in Cl- content could reduce the capacitive arc radius, polarization resistance Rp and pitting potential Eb. As the Cl- content increased from 0 to 50 g/L, significant corrosion was observed on the surface of 316L stainless steel. When the Cl- content was constant, an increase in SO42- content reduced the capacitive arc radius, Rp and Eb values. At the same time, the diameter of the corrosion pit on the surface of the specimen expanded, and the depth gradually deepened.

BACKGROUND: The accumulation of microplastics in agricultural soil poses a threat to the sustainability of agriculture, impacting crop growth and soil health. Due to the geocarpy feature of peanut, geocarposphere soil environment is critical to pod development and its nutritional quality. While the effects of microplastics in the rhizosphere have been studied, their impact on peanut pod in the geocarposphere remains unknown. Biochar has emerged as a potential soil agent with the ability to remediate soil contamination. However, the mechanisms of biochar in mitigating the toxic effects of microplastics-contaminated geocarposphere soil on peanut pod development remain largely unexplored. RESULTS: We evaluated the peanut pod performance and microbiome when facing microplastics contamination and biochar amendment in geocarposphere soil. The results showed that microplastics present in geocarposphere soil could directly enter the peanut pod, cause pod developmental disorder and exert adverse effects on nutritional quality. Aberrant expression of key genes associated with amino acid metabolism, lipid synthesis, and auxin and ethylene signaling pathways were the underlying molecular mechanisms of microplastics-induced peanut pod developmental inhibition. However, these expression abnormalities could be reversed by biochar application. In addition, peanut geocarposphere microbiome results showed that biochar application could restore the diversity of microbial communities inhibited by microplastics contamination and promote the relative abundance of bacteria correlated with pathogen resistance and nitrogen cycle of geocarposphere soil, further promoting peanut pod development. CONCLUSION: This study demonstrated that biochar application is an effective strategy to mitigate the toxic effects of microplastics-contaminated geocarposphere soil on pod development and nutritional quality.
