Background: Ragweed is an invasive, highly allergenic weed predicted to expand its habitat with warming global temperatures. Several Ambrosia species have been identified in South Africa for well over a century; however, its presence remained undetected by allergists and aerobiologists until the development of an extensive aerospora monitoring system across South African urban areas since 2019. This paper presents the inventory of preliminary investigation of the Ambrosia airborne pollen and the taxonomic identification of ragweed species. Methods: Burkard volumetric spore traps for collecting pollen samples are set up in 9 South African cities (Johannesburg, Cape Town, Pretoria, Kimberley, Durban, Potchefstroom, Ermelo, Bloemfontein, and Gqeberha). Light microscopic identification was combined with environmental DNA metabarcoding analysis to confirm the species level of airborne Ambrosia at selected monitoring stations. Ragweed sensitisation was examined in Cape Town between February 2019 and February 2024, using Allergy Xplorer (ALEX2) multicomponent allergen array. Results: Ambrosia pollen was detected in 5 aerobiological monitoring stations over the sampling period (Durban, Kimberley, Pretoria, Potchefstroom, Johannesburg). Periods of 4 consistent pollination years were observed in Kimberley (min: 1; max: 16 p.g/m3) and Durban (min: 26; max: 66 p.g/m3). In Pretoria, ragweed pollen was detected for 2 years (2020–2021; 2022–2023) with average total annuals (5-17 p.g/m3). A peak flowering period between March and April was observed in Potchefstroom, and several ragweed pollen peaks were present between the end of December and the beginning of May in Durban. The highest number of Ambrosia pollen grains was recorded in Potchefstroom, with 308 grains, and a maximum peak of 47 p.g/m3. eDNA metabarcoding confirmed the presence of Ambrosia artemisiifolia and A.trifida species. The overall prevalence of Ambrosia-sensitisation amongst 673 tests (age range 7–72 years) was 8.2% (55/673), with no significant difference in sensitisation patterns between age groups. Conclusion: Our study confirms the need to monitor the spread of ragweed, and an increasing awareness of Ambrosia as an allergen of concern in Southern Africa. Extension of aerobiological networks and testing for Ambrosia sensitisation across urban and rural sites will be required.

Abstract In this work we present a keV-scale sterile-neutrino search with a low-tritium-activity data set of the KATRIN experiment, acquired in a commissioning run in 2018. KATRIN performs a spectroscopic measurement of the tritium $$\upbeta $$ β -decay spectrum with the main goal of directly determining the effective electron anti-neutrino mass. During this commissioning phase a lower tritium activity facilitated the measurement of a wider part of the tritium spectrum and thus the search for sterile neutrinos with a mass of up to $$1.6\, \textrm{keV}$$ 1.6 keV . We do not find a signal and set an exclusion limit on the sterile-to-active mixing amplitude of $$\sin ^2\theta < 5\times 10^{-4}$$ sin 2 θ < 5 × 10 - 4 ( $$95\%$$ 95 % C.L.) at a mass of 0.3 keV. This result improves current laboratory-based bounds in the sterile-neutrino mass range between 0.1 and 1.0 keV.

Abstract South Africa’s urban population is increasing, and in parallel, urban green infrastructure has shown an increase in alien tree species, e.g., mulberry (Morus sp.), oak (Quercus spp.) and plane trees (Platanus spp.) to name a few. This causes ecological problems since alien trees are often more water-demanding and competitive than indigenous trees, but they also increase the abundance of respiratory diseases often triggered by an allergic reaction towards the pollen of those alien taxa. In the current study, utilizing 7-day volumetric spore traps, we illustrate that the most abundant tree pollen in the two largest cities of South Africa, Cape Town and Johannesburg, is produced by alien trees with a high risk of allergenicity. This adds another aspect related to public health when evaluating plant species composition in urban forestry and urban ecology, which underlines the urgency of more intense monitoring. More importantly, this—for South Africa—newly emphasized risk for public health underlines the applicability of current directives [i.e., Spatial Development Frameworks (SDFs), localized Precinct Plans, Land Use Schemes (LUSs)] and implementation options in urban planning. Here, we present ideas that may be implemented in such a framework. From both a public health and an ecological perspective, it is recommended to plant indigenous trees like Combretum erythrophyllum, Vachellia and Senegalia spp. that have fewer ecosystem disservices, like a lower impact on public health due to lower allergenicity/lower pollen occurrence and providing more ecosystem services such as lower water needs.

Abstract Background Pollen monitoring has been discontinuously undertaken in South Africa, a country with high biodiversity, a seasonal rainfall gradient, and nine biomes from arid to subtropical. The South African Pollen Monitoring Network was set up in 2019 to conduct the first long‐term national aerospora monitoring across multiple biomes, providing weekly reports to allergy sufferers and healthcare providers. Methods Daily airborne pollen concentrations were measured from August 2019 to August 2021 in seven cities across South Africa. Updated pollen calendars were created for the major pollen types (>3%), the average Annual Pollen Index over 12 months was calculated, and the results were compared to available historical data. Results The main pollen types were from exotic vegetation. The most abundant taxa were Poaceae, Cupressaceae, Moraceae and Buddleja. The pollen season start, peak and end varied widely according to the biome and suite of pollen taxa. The main tree season started in the last week of August, peaked in September and ended in early December. Grass seasons followed rainfall patterns: September–January and January–April for summer and winter rainfall areas, respectively. Major urban centres, for example, Johannesburg and Pretoria in the same biome with similar rainfall, showed substantive differences in pollen taxa and abundance. Some major differences in pollen spectra were detected compared with historical data. However, we are cognisant that we are describing only 2 years of data that may be skewed by short‐term weather patterns. Conclusions Differences in pollen spectra and concentrations were noted across biomes and between geographically close urban centres. Comparison with historical data suggests pollen spectra and seasons may be changing due to anthropogenic climate change and landscaping. These data stress the importance of regional and continuous pollen monitoring for informed care of pollinosis.

Abstract The KArlsruhe TRItium Neutrino experiment (KATRIN) aims to determine the effective electron (anti)-neutrino mass with a sensitivity of 0.2eV/c $$^2$$ 2 by precisely measuring the endpoint region of the tritium $$\beta $$ β -decay spectrum. It uses a tandem of electrostatic spectrometers working as magnetic adiabatic collimation combined with an electrostatic (MAC-E) filters. In the space between the pre-spectrometer and the main spectrometer, creating a Penning trap is unavoidable when the superconducting magnet between the two spectrometers, biased at their respective nominal potentials, is energized. The electrons accumulated in this trap can lead to discharges, which create additional background electrons and endanger the spectrometer and detector section downstream. To counteract this problem, “electron catchers” were installed in the beamline inside the magnet bore between the two spectrometers. These catchers can be moved across the magnetic-flux tube and intercept on a sub-ms time scale the stored electrons along their magnetron motion paths. In this paper, we report on the design and the successful commissioning of the electron catchers and present results on their efficiency in reducing the experimental background.

Objective: Multiple endocrine neoplasia type 2A (MEN 2A) is a rare syndrome caused by RET germline mutations and has been associated with primary hyperparathyroidism (PHPT) in up to 30% of cases. Recommendations on RET screening in patients with apparently sporadic PHPT are unclear. We aimed to estimate the prevalence of cases presenting with PHPT as first manifestation among MEN 2A i ndex cases and to characterize the former cases. Design and methods: An international retrospective multicenter study of 1085 MEN 2A index cases. Experts from MEN 2 centers all over the world were invited to participate. A total of 19 centers in 17 different countries provided registr y data of index cases followed from 1974 to 2017. Results: Ten cases presented with PHPT as their first manifestation of M EN 2A, yielding a prevalence of 0.9% (95% CI: 0.4–1.6). 9/10 cases were diagnosed with medullary thyroid carcinoma (MTC) in relation to parathyroid surgery and 1/10 was diagnosed 15 years after parathyroid surgery. 7/9 cases with full TNM data were node-positive at MTC diagnosis. Conclusions: Our data suggest that the prevalence of MEN 2A index cases that present with PHPT as their first manifestation is very low. The majority of i ndex cases presenting with PHPT as first manifestation have synchronous MTC and are often n ode-positive. Thus, our observations suggest that not performing RET mutation analysis in patients with apparently sporadic PHPT would result in an extremely low false-negative rate, if no other MEN 2A component, specifically MTC, are found during work-up or resecti on of PHPT.

This work focuses on the behavior of the C and O isotopes and major and trace element chemistry of the carbonate rocks of the Tambaba Formation, Paraíba Basin, Northeastern Brazil. Thirteen carbonate samples collected from three vertical profiles located perpendicular to the bedding along the coast of the state of Paraíba were analyzed. The δ13C values ranged from 1.6 to 2.8‰ VPDB while the δ18O values ranged from -1.2 to 1.8‰ VPDB, thus suggest a restricted shallow-platform depositional environment. X-ray fluorescence analyses suggested diagenetic changes, such as the replacement of dolomite by calcite (Mn/Sr ratios from 0.6 to 28) and dolomitization, which was identified by high Mg/Ca ratios (0.5 to 0.6). The very low SiO2 and Al2 O3 content attested to the low terrigenous material influx. The carbon isotope values suggest that the carbonate rocks were deposited after the Paleocene-Eocene Thermal Maximum (PETM) event

Abstract The KATRIN experiment aims to measure the effective electron antineutrino mass $$m_{\overline{\nu }_e}$$ mν¯e with a sensitivity of $${0.2}\,{\hbox {eV}/\hbox {c}^2}$$ 0.2eV/c2 using a gaseous tritium source combined with the MAC-E filter technique. A low background rate is crucial to achieving the proposed sensitivity, and dedicated measurements have been performed to study possible sources of background electrons. In this work, we test the hypothesis that gamma radiation from external radioactive sources significantly increases the rate of background events created in the main spectrometer (MS) and observed in the focal-plane detector. Using detailed simulations of the gamma flux in the experimental hall, combined with a series of experimental tests that artificially increased or decreased the local gamma flux to the MS, we set an upper limit of $${0.006}\,{\hbox {count}/\hbox {s}}$$ 0.006count/s (90% C.L.) from this mechanism. Our results indicate the effectiveness of the electrostatic and magnetic shielding used to block secondary electrons emitted from the inner surface of the MS.

Pollen and fungal spores (aerospora) are the major atmospheric bioaerosols. Aerospora occurrence and concentration vary by geographical region and blossoming period, and with meteorological factors. Allergic respiratory diseases affect about 20 million South Africans, with pollen and fungal allergens amongst the leading triggers. Asthma triggered by aerospora can be life threatening; allergic rhinitis causes considerable morbidity and carries financial implications for individuals and health systems. Thus, knowledge about geographical variation, seasonal timing and intensity, as well as annual aerospora fluctuations in South Africa, where climate and vegetation are exceptionally diverse, is essential for effective diagnosis and treatment of allergies. Unfortunately, there is a lack of continuous aeropalynological data from South Africa. The longest annual monitoring exists for Cape Town (~20 years), with almost no data out of the Cape since the 1990s, and many parts of South Africa have never been monitored. In this brief review, we highlight the cross-disciplinary need for strengthened and expanded continuous aeropalynological study in South Africa, the history of efforts to date, and the introduction of the interdisciplinary South African Pollen Network (SAPNET). SAPNET was launched in seven major cities in August 2019 in order to monitor weekly variations of aerospora, and provide online data for allergy sufferers and health-care providers (www.pollencount.co.za) with the aim to establish regional pollen calendars. Significance: • The significance of aerobiology as the study of airborne organic particles which include pollen, spores of fungi and cryptogams, plant hair, insect remains etc. especially as agents of allergic rhinitis (commonly known as hay fever) is outlined. Cases of allergic rhinitis are globally increasing due to current climate change. • Very limited pollen monitoring data are available for South Africa; such data are a prerequisite for establishing pollen calendars to help medical practitioners and allergy sufferers. SAPNET (South African Pollen Network) addresses this lack of data. Since August 2019, seven 7-day volumetric spore traps have been operating in South Africa in major cities, and are continuously gathering data about the pollen and spore contents in a country which is highly affected by allergic rhinitis.

Abstract The KATRIN experiment aims to determine the effective electron neutrino mass with a sensitivity of $${0.2}{\hbox { eV/c}^{2}}$$ 0.2eV/c2 (%90 CL) by precision measurement of the shape of the tritium $$\upbeta $$ β -spectrum in the endpoint region. The energy analysis of the decay electrons is achieved by a MAC-E filter spectrometer. A common background source in this setup is the decay of short-lived isotopes, such as $${}^{\text {219}}\text {Rn}$$ 219Rn and $${}^{\text {220}}\text {Rn}$$ 220Rn , in the spectrometer volume. Active and passive countermeasures have been implemented and tested at the KATRIN main spectrometer. One of these is the magnetic pulse method, which employs the existing air coil system to reduce the magnetic guiding field in the spectrometer on a short timescale in order to remove low- and high-energy stored electrons. Here we describe the working principle of this method and present results from commissioning measurements at the main spectrometer. Simulations with the particle-tracking software Kassiopeia were carried out to gain a detailed understanding of the electron storage conditions and removal processes.

ABSTRACT The Ipubi Formation represents the Aptian-Albian siliciclastic-evaporitic succession of Araripe Basin, NE Brazil. This succession comprises siliciclastic rocks (bituminous shales and claystones) and evaporites (gypsum and secondary anhydrite) and represents part of the lacustrine-shallow marine post-rift phase I. This study used sequence stratigraphy concepts to define the relations between changes in the relative lake level and the formation of Ipubi deposits. Results show that the organic-rich shales of the Ipubi Formation formed during a transgressive pulse that covered large areas of the proximal domains. These deposits overlie a regional unconformity that marks the end of the deposition of the underlying Crato Formation. A High Stand stage that followed the transgression influenced the formation of evaporitic deposits. Climate conditions played a major role in influencing the triggering and stopping of evaporite deposition. Thus, a new relative lake level fall event caused the exposure of the Ipubi Formation deposits, and created another regional subaerial unconformity accompanied by widespread karstification of evaporite beds. A posterior transgression caused the deposition of siliciclastic rocks of the Romualdo Formation over the Ipubi Formation strata, and also promoted a new event of karstification of the Ipubi upper evaporite beds.

Abstract The neutrino mass experiment KATRIN requires a stability of 3 ppm for the retarding potential at − 18.6 kV of the main spectrometer. To monitor the stability, two custom-made ultra-precise high-voltage dividers were developed and built in cooperation with the German national metrology institute Physikalisch-Technische Bundesanstalt (PTB). Until now, regular absolute calibration of the voltage dividers required bringing the equipment to the specialised metrology laboratory. Here we present a new method based on measuring the energy difference of two $$^{83{\mathrm{m}}}$$ 83m Kr conversion electron lines with the KATRIN setup, which was demonstrated during KATRIN’s commissioning measurements in July 2017. The measured scale factor $$M=1972.449(10)$$ M=1972.449(10) of the high-voltage divider K35 is in agreement with the last PTB calibration 4 years ago. This result demonstrates the utility of the calibration method, as well as the long-term stability of the voltage divider.

Objective: Hyperglucagonemia is present in many forms of diabetes and contributes to hyperglycemia, and glucagon suppression can ameliorate diabetes in mice. Leptin, a glucagon suppressor, can also reverse diabetes in rodents. Lipid nanoparticle (LNP) delivery of small interfering RNA (siRNA) effectively targets the liver and is in clinical trials for the treatment of various diseases. We compared the effectiveness of glucagon receptor (Gcgr)-siRNA delivered via LNPs to leptin in two mouse models of diabetes. Methods: Gcgr siRNA encapsulated into LNPs or leptin was administered to mice with diabetes due to injection of the β-cell toxin streptozotocin (STZ) alone or combined with high fat diet (HFD/STZ). Results: In STZ-diabetic mice, a single injection of Gcgr siRNA lowered blood glucose levels for 3 weeks, improved glucose tolerance, and normalized plasma ketones levels, while leptin therapy normalized blood glucose levels, oral glucose tolerance, and plasma ketones, and suppressed lipid metabolism. In contrast, in HFD/STZ-diabetic mice, Gcgr siRNA lowered blood glucose levels for 2 months, improved oral glucose tolerance, and reduced HbA1c, while leptin had no beneficial effects. Conclusions: While leptin may be more effective than Gcgr siRNA at normalizing both glucose and lipid metabolism in STZ diabetes, Gcgr siRNA is more effective at reducing blood glucose levels in HFD/STZ diabetes.

Background: The hormone leptin is an important regulator of metabolic homeostasis, able to inhibit food intake and increase energy expenditure. Leptin can also independently lower blood glucose levels, particularly in hyperglycemic models of leptin or insulin deficiency. Despite significant efforts and relevance to diabetes, the mechanisms by which leptin acts to regulate blood glucose levels are not fully understood. Scope of review: Here we assess literature relevant to the glucose lowering effects of leptin. Leptin receptors are widely expressed in multiple cell types, and we describe both peripheral and central effects of leptin that may be involved in lowering blood glucose. In addition, we summarize the potential clinical application of leptin in regulating glucose homeostasis. Major conclusions: Leptin exerts a plethora of metabolic effects on various tissues including suppressing production of glucagon and corticosterone, increasing glucose uptake, and inhibiting hepatic glucose output. A more in-depth understanding of the mechanisms of the glucose-lowering actions of leptin may reveal new strategies to treat metabolic disorders.

Objective: It has been thought that the depletion of insulin is responsible for the catabolic consequences of diabetes; however, evidence suggests that glucagon also plays a role in diabetes pathogenesis. Glucagon suppression by glucagon receptor (Gcgr) gene deletion, glucagon immunoneutralization, or Gcgr antagonist can reverse or prevent type 1 diabetes in rodents suggesting that dysregulated glucagon is also required for development of diabetic symptoms. However, the models used in these studies were rendered diabetic by chemical- or immune-mediated β-cell destruction, in which insulin depletion is incomplete. Therefore, it is unclear whether glucagon suppression could overcome the consequence of the complete lack of insulin. Methods: To directly test this we characterized mice that lack the Gcgr and both insulin genes (GcgrKO/InsKO). Results: In both P1 pups and mice that were kept alive to young adulthood using insulin therapy, blood glucose and plasma ketones were modestly normalized; however, mice survived for only up to 6 days, similar to GcgrHet/InsKO controls. In addition, Gcgr gene deletion was unable to normalize plasma leptin levels, triglycerides, fatty acids, or hepatic cholesterol accumulation compared to GcgrHet/InsKO controls. Conclusion: Therefore, the metabolic manifestations associated with a complete lack of insulin cannot be overcome by glucagon receptor gene inactivation. The austral westerlies strongly influence precipitation and ocean circulation in the southern temperate zone, with important consequences for cultures and ecosystems. Global climate models anticipate poleward retreat of the austral westerlies with future warming, but the available paleoclimate records that might test these models have been limited to South America and New Zealand, are not fully consistent with each other and may be complicated by influences from other climatic factors. Here we present the first high-resolution diatom and sedimentological records from the winter rainfall region of South Africa, representing precipitation in the equatorward margin of the westerly wind belt during the last 1400 yr. Inferred rainfall was relatively high ∼1400–1200 cal yr BP, decreased until ∼950 cal yr BP, and rose notably through the Little Ice Age with pulses centred on ∼600, 530, 470, 330, 200, 90, and 20 cal yr BP. Synchronous fluctuations in Antarctic ice core chemistry strongly suggest that these variations were linked to changes in the westerlies. Equatorward drift of the westerlies during the wet periods may have influenced Atlantic meridional overturning circulation by restricting marine flow around the tip of Africa. Apparent inconsistencies among some aspects of records from South America, New Zealand and South Africa warn against the simplistic application of single records to the Southern Hemisphere as a whole. Nonetheless, these findings in general do support model projections of increasing aridity in the austral winter rainfall zones with future warming.

The late Cambrian - late Tremadocian La Silla Formation is a carbonate unit of the eastern Precordillera in Argentina whose facies indicate a shallow platform environment. Until this moment, there were no studies that referred to the diagenetic evolution of these rocks. The present study involves the characteristics and distribution of the silicification that affects this unit and determines its different diagenetic stages through petrographic (with cathodoluminescence support) and stratigraphic analyses. An early diagenetic chert, in a stage previous to the compactation, was observed. This chert is related to silica-rich seawater in contact with permeable and porous sediments. A later pulse of chert, associated with fracturing, also occurs. The knowledge provided by the characteristics and distribution of chert in these carbonates is significant, especially when considering that the La Silla Formation in San Juan province is the most quarried unit for the elaboration of lime.A Formação La Silla (Neo Cambriano-Neo Tremadociano) é formada por depósitos carbonáticos da região da Pré-Cordilheira leste, Argentina. As faciologias encontradas em tais depósitos indicam um ambiente de deposição caracterizado por uma plataforma rasa. Até o momento, nenhum estudo tratou especificamente da evolução diagenética dessas rochas. O presente estudo envolve as características e distribuição da silicificação que afeta esta formação e apresenta os diferentes estágios diagenéticos ocorridos, através de análises petrográficas (auxiliadas por catodoluminescência) e estratigráficas. Foi identificado um pulso precoce de silicificação, anterior ao processo de compactação das rochas afetadas. Esta silicificação foi favorecida pela presença de água do mar rica em sílica em contato com sedimentos porosos e permeáveis. Também foi identificado um pulso de silicificação tardia, associado a eventos de fraturamento das rochas. O conhecimento adquirido sobre as características e a distribuição da silicificação nestes depósitos carbonáticos é importante, especialmente quando se considera que a Formação La Silla, na Província de San Juan, é a formação mais minerada para a produção de calcário.

At least 12 genes (FH, HIF2A, MAX, NF1, RET, SDHA, SDHB, SDHC, SDHD, SDHAF2, TMEM127, and VHL) have been implicated in inherited predisposition to phaeochromocytoma (PCC), paraganglioma (PGL), or head and neck paraganglioma (HNPGL) and a germline mutation may be detected in more than 30% of cases. Knowledge of somatic mutations contributing to PCC/PGL/HNPGL pathogenesis has received less attention though mutations in HRAS, HIF2A, NF1, RET, and VHL have been reported. To further elucidate the role of somatic mutation in PCC/PGL/HNPGL tumourigenesis, we employed a next generation sequencing strategy to analyse “mutation hotspots” in 50 human cancer genes. Mutations were identified for HRAS (c.37G>C; p.G13R and c.182A>G; p.Q61R) in 7.1% (6/85); for BRAF (c.1799T>A; p.V600E) in 1.2% (1/85) of tumours; and for TP53 (c.1010G>A; p.R337H) in 2.35% (2/85) of cases. Twenty-one tumours harboured mutations in inherited PCC/PGL/HNPGL genes and no HRAS, BRAF, or TP53 mutations occurred in this group. Combining our data with previous reports of HRAS mutations in PCC/PGL we find that the mean frequency of HRAS/BRAF mutations in sporadic PCC/PGL is 8.9% (24/269) and in PCC/PGL with an inherited gene mutation 0% (0/148) suggesting that HRAS/BRAF mutations and inherited PCC/PGL genes mutations might be mutually exclusive. We report the first evidence for BRAF mutations in the pathogenesis of PCC/PGL/HNPGL.
