Ultra-low-wear polyethylene (ULWPE) is a promising material for artificial joint implants, owing to its good biocompatibility and wear resistance. However, its molecular weight is almost an order of magnitude lower than that of Ultrahigh molecular weight polyethylene (UHMWPE). The mechanism between wear resistance and the structure of ULWPE is still unknown. This study analyzed their attributes from microstructure to macroscopic performance and revealed the wear resistance mechanisms of ULWPE by conducting an in-depth investigation of four types of polyethylene, including ULWPE, UHMWPE, and high-density polyethylene. Impact tests showed that ULWPE, with a molecular weight of 282 kg/mol, exhibited an impact energy of 110 kJ/m2, surpassing the 97 kJ/m2 recorded for UHMWPE (molecular weight over 5000 kg/mol). Furthermore, tribology tests indicated that the wear rate of the most anti-wear ULWPE was 4.479 ± 0.040 mm³/Mc, which was significantly lower than the 5.601 ± 0.456 mm³/Mc for UHMWPE. The excellent mechanical properties and wear resistance of ULWPE were derived from its unique structure. Dynamic rheological assessments and thermal classification techniques confirmed that ULWPE had long-branched chains. The long-branched chains markedly increased the proportion of the high entanglement and the tie molecules in the amorphous regions, endowing it with superior toughness. At the same time, the high crystallinity of ULWPE gives it sufficient hardness and strength. The combination of high crystallinity and high entanglement endowed ULWPE with a good balance between strength and toughness, leading to its exceptional mechanical properties and wear resistance. This study provided robust data for ULWPE's clinical use and theoretical insights for designing durable, implantable polymers.

Photosynthesis is one of the most important reactions for sustaining our environment. Photosystem II (PSII) is the initial site of photosynthetic electron transfer by water oxidation. Light in excess, however, causes the simultaneous production of reactive oxygen species (ROS), leading to photo-oxidative damage in PSII. To maintain photosynthetic activity, the PSII reaction center protein D1, which is the primary target of unavoidable photo-oxidative damage, is efficiently degraded by FtsH protease. In PSII subunits, photo-oxidative modifications of several amino acids such as Trp have been indeed documented, whereas the linkage between such modifications and D1 degradation remains elusive. Here, we show that an oxidative post-translational modification of Trp residue at the N-terminal tail of D1 is correlated with D1 degradation by FtsH during high-light stress. We revealed that Arabidopsis mutant lacking FtsH2 had increased levels of oxidative Trp residues in D1, among which an N-terminal Trp-14 was distinctively localized in the stromal side. Further characterization of Trp-14 using chloroplast transformation in Chlamydomonas indicated that substitution of D1 Trp-14 to Phe, mimicking Trp oxidation enhanced FtsH-mediated D1 degradation under high light, although the substitution did not affect protein stability and PSII activity. Molecular dynamics simulation of PSII implies that both Trp-14 oxidation and Phe substitution cause fluctuation of D1 N-terminal tail. Furthermore, Trp-14 to Phe modification appeared to have an additive effect in the interaction between FtsH and PSII core in vivo. Together, our results suggest that the Trp oxidation at its N-terminus of D1 may be one of the key oxidations in the PSII repair, leading to processive degradation by FtsH.

Root-knot nematodes are the major diseases in protected cultivation around the world. Bio-organic fertilizer has become a research hotspot, with a variety of microorganisms that control various vegetable soil-borne diseases. This study screened nematocidal microorganisms from fresh vermicompost, explored the inhibitory substances produced by biocontrol agents, and evaluated their potential biocontrol ability in the pot and field under greenhouse conditions. The highly effective antagonistic microbes of Meloidogyne incognita (M. incognita) were screened. Strains YL1 and YL31 were identified as Peribacillus frigoritolerans, and strain YL6 was identified as Lysinibacillus fusiformis. The three strains all produced chitinase and protease, which prevented the normal development of eggs and the second-stage juveniles (J2) by destroying their appearance. The three strains all improved potassium-dissolving ability, and the strains YL1 and YL6 also enhanced phosphorus-dissolving ability. Pot experiments showed that tomato root knots were reduced, and plant growth improved. Field tests showed that the root-knot index and nematode population were reduced significantly, and cucumber growth and yield were enhanced. Strain YL1 had the best control effect with 70.6%, and the yield increased by 14.9% compared with the control. Overall, this study showed the ability of antagonistic bacteria YL1, YL6, and YL31 to control root-knot nematodes, and these antagonistic bacteria could be developed as biocontrol agents for sustainable agriculture.

Researchers have described protection mechanisms against the photoinhibition of photosystems under strong-light stress. Cyclic Electron Flow (CEF) mitigates electron acceptor-side limitation, and thus contributes to Photosystem I (PSI) protection. Chloroplast protease removes damaged protein to assist with protein turn over, which contributes to the quality control of Photosystem II (PSII). The PGR5 protein is involved in PGR5-dependent CEF. The FTSH protein is a chloroplast protease which effectively degrades the damaged PSII reaction center subunit, D1 protein. To investigate how the PSI photoinhibition phenotype in pgr5 would be affected by adding the ftsh mutation, we generated double-mutant pgr5ftsh via crossing, and its phenotype was characterized in the green algae Chlamydomonas reinhardtii. The cells underwent high-light incubation as well as low-light incubation after high-light incubation. The time course of Fv/Fm values in pgr5ftsh showed the same phenotype with ftsh1-1. The amplitude of light-induced P700 photo-oxidation absorbance change was measured. The amplitude was maintained at a low value in the control and pgr5ftsh during high-light incubation, but was continuously decreased in pgr5. During the low-light incubation after high-light incubation, amplitude was more rapidly recovered in pgr5ftsh than pgr5. We concluded that the PSI photoinhibition by the pgr5 mutation is mitigated by an additional ftsh1-1 mutation, in which plastoquinone pool would be less reduced due to damaged PSII accumulation.

Abstract To assess the efficacy of sacral neuromodulation (SNM) for neurogenic bladder (NB), guided by intraoperative three-dimensional imaging of sacral computed tomography (CT) and mobile C-arm fluoroscopy through video-urodynamics examination. We enrolled 52 patients with NB who underwent conservative treatment with poor results between September 2019 and June 2021 and prospectively underwent SNM guided by intraoperative three-dimensional imaging of sacral CT and mobile C-arm fluoroscopy. Video-urodynamics examination, voiding diary, quality of life questionnaire, overactive bladder symptom scale (OABSS) scoring, and bowel dysfunction exam were completed and recorded at baseline, at SNM testing, and at 6-month follow-up phases. Finally, we calculated the conversion rate from period I to period II, as well as the treatment efficiency and the occurrence of adverse events during the testing and follow-up phases. The testing phase of 52 NB patients was 18–60 days, with an average of (29.3 ± 8.0) days. Overall, 38 patients underwent SNM permanent electrode implantation, whose follow-up phase was 3–25 months, with an average of (11.9 ± 6.1) months. Compared with baseline, the voiding times, daily catheterization volume, quality of life score, OABSS score, bowel dysfunction score, maximum detrusor pressure before voiding, and residual urine volume decreased significantly in the testing phase. The daily voiding volume, functional bladder capacity, maximum urine flow rate, bladder compliance, and maximum cystometric capacity increased significantly in the testing phase. Besides, the voiding times, daily catheterization volume, quality of life score, OABSS score, bowel dysfunction score, maximum detrusor pressure before voiding, and residual urine volume decreased further from the testing to follow-up phase. Daily voiding volume, functional bladder capacity, maximum urine flow rate, bladder compliance, and maximum cystometric capacity increased further from testing to follow-up. At baseline, 10 ureteral units had vesicoureteral reflux (VUR), and 9 of them improved in the testing phase. Besides, there was 1 unit that further improved to no reflux during the follow-up phase. At baseline, 10 patients had detrusor overactivity (DO), and 8 of them improved in the testing phase. Besides, 1 patient’s symptoms further improved during the follow-up phase. At baseline, there were 35 patients with detrusor-bladder neck dyssynergia (DBND); 14 (40.0%) of them disappeared during the testing phase. Among 13 cases who had DBND in the testing phase, 6 (46.2%) disappeared during the follow-up phase. Of the 47 patients with detrusor-external sphincter dyssynergia (DESD) at baseline, 8 (17.0%) disappeared during the testing phase. Among 26 cases who had DESD in the testing phase, 6 (23.1%) disappeared during the follow-up phase. The effective rate of this study was 88.5% (46/52), and the conversion rate from phase I to phase II was 73.1% (38/52). Additionally, the efficacy in a short-term follow-up was stable. SNM guided by intraoperative three-dimensional imaging of sacral CT and mobile C-arm fluoroscopy is an effective and safe treatment option for NB in short time follow-up. It would be well improved in the bladder storage function, sphincter synergetic function and emptying efficiency by video-urodynamics examination in this study. To explore the effect of vermicompost on the yield and quality of tomato cultivated in salty soil, we investigated the soil chemical properties, the yield, vitamin C, organic acid, soluble solids, and nitrate of fruit, photosynthesis, and carbohydrates of plants grown under various salt levels applied with the application of either commercial chemical fertilizers, cow manure, or vermicompost. Results showed that the tomato yield was not increased from the chemical fertilizer application, while there was an increase from the cow manure and vermicompost (increased 31.7% and 65.2%, respectively) under salt stress. Compared to no salt stress, the contents of vitamin C, organic acid, soluble solids, and nitrate increased 26.55%, 40.59%, 46.31%, and 35.08%, respectively, under salt stress (2 g NaCl·kg−1 soil). Compared with the Control, the application of chemical fertilizers failed to improve the sugar/acid ratio but increased nitrate content, while cow manure and vermicompost improved the sugar/acid ratio by 42.0% and 73.1%, respectively. Particularly, vermicompost increased vitamin C and reduced nitrate to the greatest extent among the different fertilizer treatments. The decrease in sodium (Na+) in the roots and leaves, increase in carbohydrates in fruit, and photosynthetic efficiency of leaves imply an amendment effect of vermicompost on salt stress. Moreover, vermicompost also facilitated the transit of carbohydrates from leaves to fruits by increasing the accumulation of nitrogen, phosphate, and potassium in fruits, leaves, and roots, while decreasing proline and soluble protein accumulation in leaves and roots. In conclusion, vermicompost could alleviate the adverse effect of salt stress and improve tomato yield and fruit quality by improving the photosynthetic capacity and promoting carbohydrate transport to fruit. The findings give a new perspective on the beneficial effect of vermicompost on tomato yield and quality.

ABSTRACTHuman coronavirus NL63 (HCoV-NL63) is primarily associated with common cold in children, elderly and immunocompromised individuals. Outbreaks caused by HCoV-NL63 are rare. Here we report a cluster of HCoV-NL63 cases with severe lower respiratory tract infection that arose in Guangzhou, China, in 2018. Twenty-three hospitalized children were confirmed to be HCoV-NL63 positive, and most of whom were hospitalized with severe pneumonia or acute bronchitis. Whole genomes of HCoV-NL63 were obtained using next-generation sequencing. Phylogenetic and single amino acid polymorphism analyses showed that this outbreak was associated with two subgenotypes (C3 and B) of HCoV-NL63. Half of patients were identified to be related to a new subgenotype C3. One unique amino acid mutation at I507 L in spike protein receptor binding domain (RBD) was detected, which segregated this subgenotype C3 from other known subgenotypes. Pseudotyped virus bearing the I507 L mutation in RBD showed enhanced entry into host cells as compared to the prototype virus. This study proved that HCoV-NL63 was undergoing continuous mutation and has the potential to cause severe lower respiratory disease in humans.

A greenhouse pot test was conducted to study the impacts of replacing mineral fertilizer with organic fertilizers for one full growing period on soil fertility, tomato yield and quality using soils with different tomato planting history. Four types of fertilization regimes were compared: (1) conventional fertilizer with urea, (2) chicken manure compost, (3) vermicompost, and (4) no fertilizer. The effects on plant growth, yield and fruit quality and soil properties (including microbial biomass carbon and nitrogen, NH4+-N, NO3--N, soil water-soluble organic carbon, soil pH and electrical conductivity) were investigated in samples collected from the experimental soils at different tomato growth stages. The main results showed that: (1) vermicompost and chicken manure compost more effectively promoted plant growth, including stem diameter and plant height compared with other fertilizer treatments, in all three types of soil; (2) vermicompost improved fruit quality in each type of soil, and increased the sugar/acid ratio, and decreased nitrate concentration in fresh fruit compared with the CK treatment; (3) vermicompost led to greater improvements in fruit yield (74%), vitamin C (47%), and soluble sugar (71%) in soils with no tomato planting history compared with those in soils with long tomato planting history; and (4) vermicompost led to greater improvements in soil quality than chicken manure compost, including higher pH (averaged 7.37 vs. averaged 7.23) and lower soil electrical conductivity (averaged 204.1 vs. averaged 234.6 μS/cm) at the end of experiment in each type of soil. We conclude that vermicompost can be recommended as a fertilizer to improve tomato fruit quality and yield and soil quality, particularly for soils with no tomato planting history.

Upper tract urothelial carcinomas in the proximal ureter are an uncommon disease. We present a case in which it was firstly detected by fluorescence in situ hybridization and not by endoscopy and radiologic imaging. Consequently, a radical nephroureterectomy with excision of the bladder cuff was performed as the gold standard treatment.

Soil microbial abundance and diversity change constantly in continuous cropping systems, resulting in the prevalence of soil-borne pathogens and a decline in crop yield in solar greenhouses. To investigate the effects of rice straw and biochar on soil microbial abundance and diversity in soils with a history of continuous planting, three treatments were examined: mixed rice straw and biochar addition (RC), rice straw addition (R), and biochar addition (C). The amount of C added in each treatment group was 3.78 g kg−1 soil. Soil without rice straw and biochar addition was treated as a control (CK). Results showed that RC treatment significantly increased soil pH, available nitrogen (AN), available phosphorus (AP), and potassium (AK) by 40.3%, 157.2%, and 24.2%, respectively, as compared to the CK soil. The amount of soil labile organic carbon (LOC), including readily oxidizable organic carbon (ROC), dissolved organic carbon (DOC), and light fraction organic carbon (LFOC), was significantly greater in the RC, R, and C treatment groups as compared to CK soil. LOC levels with RC treatment were higher than with the other treatments. Both rice straw and biochar addition significantly increased bacterial and total microbial abundance, whereas rice straw but not biochar addition improved soil microbial carbon metabolism and diversity. Thus, the significant effects of rice straw and biochar on soil microbial carbon metabolism and diversity were attributed to the quantity of DOC in the treatments. Therefore, our results indicated that soil microbial diversity is directly associated with DOC. Based on the results of this study, mixed rice straw and biochar addition, rather than their application individually, might be key to restoring degraded soil.

The effect of exogenous CaCl2 on photosystem I and II (PSI and PSII) activities, cyclic electron flow (CEF), and proton motive force of tomato leaves under low night temperature (LNT) was investigated. LNT stress decreased the net photosynthetic rate (Pn), effective quantum yield of PSII [Y(II)], and photochemical quenching (qP), whereas CaCl2 pretreatment improved Pn, Y(II), and qP under LNT stress. LNT stress significantly increased the non-regulatory quantum yield of energy dissipation [Y(NO)], whereas CaCl2 alleviated this increase. Exogenous Ca2+ enhanced stimulation of CEF by LNT stress. Inhibition of oxidized PQ pools caused by LNT stress was alleviated by CaCl2 pretreatment. LNT stress reduced zeaxanthin formation and ATPase activity, but CaCl2 pretreatment reversed both of these effects. LNT stress caused excess formation of a proton gradient across the thylakoid membrane, whereas CaCl2 pretreatment decreased the said factor under LNT. Thus, our results showed that photoinhibition of LNT-stressed plants could be alleviated by CaCl2 pretreatment. Our findings further revealed that this alleviation was mediated in part by improvements in carbon fixation capacity, PQ pools, linear and cyclic electron transports, xanthophyll cycles, and ATPase activity.

Comprehensive observations of hydroxyl (OH) and hydroperoxy (HO2) radicals were conducted in October 2019 at a coastal continental site in the Pearl River Delta (YMK site, 22.55°N, 114.60°E). The average daily maximum OH and HO2 concentrations were (4.7-9.5) × 106 cm-3 and (4.2-8.1) × 108 cm-3, respectively. The synchronized air mass transport from the northern cities and the South China Sea exerted a time-varying influence on atmospheric oxidation. Under a typical ocean30 atmosphere (OCM), reasonable measurement model agreement was achieved for both OH and HO2 using a 0-D chemical box model incorporating the regional atmospheric chemistry mechanism version 2-Leuven isoprene mechanism (RACM2-LIM1). Land mass (LAM) influence promoted more active photochemical processes, with daily averages of 7.1 × 106 cm-3 and 5.2 × 108 cm-3 for OH and HO2, respectively. Intensive photochemistry occurred after precursor accumulation, allowing local net ozone production comparable with surrounding suburban environments (5.52 ppb/h during the LAM period). The rapid oxidation process was accompanied by a higher diurnal nitrous acid (HONO) concentration (> 400 ppt). After a sensitivity test, HONO-related chemistry elevated the ozone production rate by 33% and 39% during the LAM and OCM periods, respectively, while the nitric acid and sulfuric acid formation rates were 52% and 35% higher, respectively. The simulated daytime HONO and ozone concentrations were reduced to a low level (~70 ppt and ~35 ppb) without the HONO constraint. This work challenges the conventional recognition of the MBL in a complex atmosphere. For coastal cities, the particularity of the HONO chemistry in the MBL tends to influence the ozone-sensitive system and eventually magnifies the background ozone. Therefore, the promotion of oxidation by elevated precursor concentrations is worth considering when formulating emission reduction policies. [ABSTRACT FROM AUTHOR]
