Virus-induced gene silencing (VIGS) showed several advantages to identify gene functions such as short experimental cycle, more broad hosts, etc. In this study, the feasibility and efficiency of employing Barley stripe mosaic virus (BSMV)-based VIGS system to evaluate Fusarium head blight (FHB) resistance were explored in wheat. With variable conditions tested, it showed that the maximal silencing efficiency 78% on spike was obtained when the recombinant BSMV was inoculated on flag leaf at flagging stage. However, the plant may reduce its own immunity to FHB when inoculated with BSMV. To induce this impact, different Fusarium graminearum strains were tested and SF06-1 strain was selected for FHB resistance evaluation. Using this system, TaAOC, TaAOS, and TaOPR3 involved in jasmonic acid (JA) signaling pathway were identified to positively regulate FHB resistance, which was underpinned by the results when silencing TaAOS in wheat by stable transgenic plants.

The mutation of the gene encoding a stearoyl-acyl carrier protein fatty acid desaturase (ssi2) has been proved to enhance pathogen resistance in several plants, while it's potential to regulate biotic and abiotic stresses in wheat is still unclear. In this study, we cloned TaSSI2 gene in wheat and provided several evidences of its involvement in multiple biological functions. By using barley stripe mosaic virus (BSMV)-induced gene silencing (VIGS) in wheat, it was found that TaSSI2 negatively regulated both powdery mildew and Fusarium head blight (FHB) resistance, which was consistent with the phenotype observed in knock-out mutants of Kronos. The expression of TaSSI2 was down-regulated by in vitro treatments of methyl jasmonate (MeJA), but positively regulated by salicylic acid (SA) and abscisic acid (ABA), implying the cross-talk between different hormone signaling pathways involved in wheat to regulate biotic stresses is still to be elucidated. Furthermore, the up-regulated expression of PR4 and PR5 indicated that TaSSI2 probably regulated FHB resistance by depressing the SA signaling pathway in wheat. In addition, the over-expression of TaSSI2 increased the content of linolenic acid (18:3) and subsequently enhanced drought tolerance of transgenic Brachypodium. This phenomenon might be associated with its subcellular localization in the whole cytosol, partly overlapping with Golgi apparatus and the secreted vesicles. As a stearoyl-acyl carrier protein fatty acid desaturase, TaSSI2 was proposed to be involved in cell lipid metabolism and carried targets out of the cell from membrane or wax synthesis, resulting in enhanced drought tolerance in plant.

Autophagy is involved in aging-related cognitive impairment. Aerobic exercise training can improve cognitive function in the elderly and this effect may be associated with autophagic mechanisms and mitochondrial respiratory function. High intensity interval training (HIIT) has beneficial effects on heart and skeletal muscles by activating autophagy and/or mitophagy, but the effects of HIIT on autophagy/mitophagy in the aging brain are unknown. This study investigated the effects of HIIT on the mitochondrial respiratory complex and autophagy/mitophagy, and its relation to brain function. Thirteen middle-aged male ICR mice underwent HIIT for 7 weeks. The exercise program reduced the spontaneous behavior and exploration activities of the mice. The phosphorylation level of cAMP response element binding protein (CREB) and the protein expression of brain-derived neurotrophic factor (BDNF) decreased after the 7-week HIIT. Exercise downregulated the protein expression of Complex Ⅰ and upregulated the protein expression of Complex Ⅲ, Complex Ⅳ and Complex Ⅴ. HIIT also decreased the expression of mitophagy-related proteins in the mitochondrial fractions of the hippocampus. However, HIIT did not change the expression of autophagy-related proteins LC3, P62, Atg5, Atg7, Beclin-1 and Lamp2 in the total lysate of the hippocampus. These data indicated that HIIT might have negative effects on the plasticity of the hippocampus in middle-aged mice. The effects may be related to the dysregulation of CREB-BDNF signaling, mitochondrial respiratory complex and mitophagy induced by HIIT.

The pleiotropic drug resistance (PDR) sub-family of adenosine triphosphate (ATP)-binding cassette (ABC) transporter had been reported to participate in diverse biological processes of plant. In this study, we cloned three novel PDR genes in Fusarium head blight (FHB) resistant wheat cultivar Ning 7840, which were located on wheat chromosomes 6A, 6B and 6D. In phylogeny, these genes were members of cluster I together with AePDR7 and BdPDR7. Subcellular localization analysis showed that TaPDR7 was expressed on the plasmalemma. The quantitative real time PCR (RT-PCR) analysis showed that this gene and its probable orthologues in chromosomes 6B and 6D were both up-regulated sharply at 48 h after infected by Fusarium graminearum and trichothecene deoxynivalenol (DON) in spike. When knocking down the transcripts of all TaPDR7 members by barely stripe mosaic virus-induced gene silencing (BSMV-VIGS) system, it could promote the F. graminearum hyphae growth and made larger pathogen inoculation points in Ning 7840, which suggested that TaPDR7 might play an important role in response to F. graminearum. Although salicylic acid (SA), methyl jasmonate (MeJA) and abscisic acid (ABA) had been reported to possibly regulate wheat FHB resistance, here, we found that the three members of TaPDR7 were negatively regulated by these three hormones but positively regulated by indoleacetic acid (IAA).

Purpose – This study aims to consider active vision in low-visibility environments to reveal the factors of optical properties which affect visibility and to explore a method of obtaining different depths of fields by multimode imaging.Bad weather affects the driver’s visual range tremendously and thus has a serious impact on transport safety. Design/methodology/approach – A new mechanism and a core algorithm for obtaining an excellent large field-depth image which can be used to aid safe driving is designed and implemented. In this mechanism, atmospheric extinction principle and field expansion system are researched as the basis, followed by image registration and fusion algorithm for the Infrared Extended Depth of Field (IR-EDOF) sensor. Findings – The experimental results show that the idea we propose can work well to expand the field depth in a low-visibility road environment as a new aided safety-driving sensor. Originality/value – The paper presents a new kind of active optical extension, as well as enhanced driving aids, which is an effective solution to the problem of weakening of visual ability. It is a practical engineering sensor scheme for safety driving in low-visibility road environments.

Plant epidermis serves important functions in shoot growth, plant defense and lipid metabolism, though mechanisms of related transcriptional regulation are largely unknown. Here, we identified cis-elements specific to shoot epidermis expression by dissecting the promoter of Triticum aestivum lipid transfer protein 1 (TaLTP1). A preliminary promoter deletion analysis revealed that a truncated fragment within 400 bp upstream from the translation start site was sufficient to confer conserved epidermis-specific expression in transgenic Brachypodium distachyon and Arabidopsis thaliana. Further, deletion or mutation of a GC(N4)GGCC motif at position &ndash, 380 bp caused a loss of expression in pavement cells. With an electrophoretic mobility shift assay (EMSA) and transgenic reporter assay, we found that a light-responsive CcATC motif at position &ndash, 268 bp was also involved in regulating pavement cell-specific expression that is evolutionary conserved. Moreover, expression specific to leaf trichome cells was found to be independently regulated by a CCaacAt motif at position -303 bp.

Eighty-one patients who had been diagnosed with hepatitis B were recruited and the stage of fibrosis was determined by biopsy. In order to successfully monitor a large-scale distributed system, it is an important issue that the monitoring function fully covers all the entities in the system. To this end, a key challenge is to efficiently transmit state information of the entities in the system. This paper solves this challenge from two aspects. First, in virtue of the idea of self-organizing networks, this paper proposes a neighborhood organization algorithm, which self-organizes the nodes into several monitoring neighborhoods based on the t distance between nodes. The second aspect focuses on message transmission. There are three common message transmission methods in network, i.e., flooding, multicast and unicast. Flooding may cause high network overhead, while unicast may pose high system delay. Based on the idea of Gossip protocol, this paper proposes a directional message dissemination algorithm (D-Gossip), which is a kind of probabilistic multicast. D-Gossip reduces message dissemination uncertainty of traditional Gossip protocols. It effectively improves the efficiency and coverage of message dissemination, while reducing redundant information in the system due to Gossip protocol. The experimental results show that the neighborhood organization algorithm and the DGossip can effectively solve the above challenge.

The preharvest application of Ca-containing foliar fertilizers can reduce the incidence of bitter pit (BP) in apples and improve fruit quality by increasing the Ca content and decreasing both the N content and the N/Ca ratio in fruits. In this study, we aimed to investigate the control efficacy of Ca-containing fertilizers on the incidence of BP and their effects on the Ca and N contents in bagged 'Fuji' apples by spraying foliar fertilizer containing calcium chloride (CaClThe incidences of BP were reduced significantly by 43.2-73.0%, and the efficacy of spraying at an early stage was significantly higher than that of spraying at a late stage. Cyanobacteria are the major producers of geosmin in natural waters. To identify a gene involved in geosmin biosynthesis in cyanobacteria, the polymerase chain reaction (PCR) was used to amplify a 2298-bp open reading frame (ORF) from the geosmin-producing cyanobacterium Lyngbya kuetzingii UTEX 1547. This ORF encoded a protein of 765 amino acids. Alignment of the deduced amino acid sequence demonstrated that geoL had high similarity to the corresponding genes of Oscillatoria sp. To analyze the physiological mechanisms underlying the increased tolerance to drought and high temperature stress combination by overproduction of glycinebetaine (GB) in wheat, a transgenic wheat line T6 and its wild-type (WT) Shi4185 were used. The transgenic line was generated by introducing a gene encoding betaine aldehyde dehydrogenase (BADH) into a wheat line Shi4185. The gene was cloned from Garden Orache (Atriplex hortensis L.). Wheat plants were exposed to drought (withholding irrigation), high temperature stress (40 °C), and their combination at the flowering stage. Analyses of oxygen-evolving activity and photosystem II (PSII) photochemistry, modulated chlorophyll fluorescence, rapid fluorescence induction kinetics, and the polyphasic fluorescence transients (OJIP) were used to evaluate PSII photochemistry in wheat plants. The results suggest that the PSII in transgenic plants showed higher resistance than that in wild-type plants under the stresses studied here, this increased tolerance was associated with an improvement in stability of the oxygen-evolving complex and the reaction center of PSII; streptomycin treatment can impair the protective effect of overaccumulated GB on PSII. The overaccumulated GB may protect the PSII complex from damage through accelerating D1 protein turnover to alleviate photodamage. The results also suggest that the PSII under combined high temperature and drought stress shows higher tolerance than under high temperature stress alone in both transgenic and wild-type plants.

In order to study the greenness of CNC machine tool, the influence greenness on fault information of CNC machine tools main shaft system was analyzed. According to the fault characteristics and green evaluation index, the failure influence degree of spindle system to the surrounding environment, people's physical and mental health was analyzed in the aspects of environment, human and economic by combining fuzzy evaluation with fault analysis. The main factors affecting its green were found out, the failure reasons were analyzed, and measurement improvements were suggested. The greenness was analyzed before and after improvement. The results show that the failure rate of the improved main shaft system reduces greatly and the green level is improved significantly. The comprehensive evaluation score of green degree is increased from 55.62 to 64.87, and the green product is obtained.

Lung adenocarcinoma (AC) is one of the most deadly malignancies. The disease has a low five-year survival rate; therefore, the identification of novel therapeutic agents is required. This study aimed to investigate the effect of small interfering RNA (siRNA) targeting hypoxia‑inducible factor 1α (HIF‑1α) on the growth of AC A549 cells. A549 cells were transfected with various concentrations of HIF‑1α or control siRNA, and the effect on HIF‑1α expression was analyzed using quantitative polymerase chain reaction and western blot analysis. The effects of HIF-1α siRNA on growth inhibition and apoptosis were then assessed using standard methods. HIF‑1α siRNA treatment significantly reduced HIF‑1α mRNA and protein expression in A549 cells. Furthermore, the downregulation of HIF-1α expression inhibited the growth of A549 cells and induced apoptosis of A549 cells by upregulating caspase-3 expression. The present in vitro study demonstrates that the downregulation of HIF‑1α is capable of suppressing AC A549 cell growth, through the induction of apoptosis. This suggests that HIF‑1α inhibition may represent a promising strategy for the treatment of AC.

Directly modified bentonite was obtained by directly adding modifier cetyl trimethyl ammonium bromide (CTMAB) to sodium bentonite.The modified bentonites adsorption performance to methyl orange and methylene blue was studied, and it was compared with modified bentonite prepared by classical method. The results show that directly modified bentonite has a greater and faster absorption for both methyl orange and methylene blue; the relationship between adsorption quantity and equilibrium concentration conforms to the Langmuir isotherm equation; absorption kinetics is in accordance with the secondary dynamics equation.

In order to achieve the application requirements of a mobile asphalt concrete mixer, a distributed computer control system of the mixer is developed with an industrial control computer and a S7-200 PLC. Through analyzing the operation principle of mixer, main functions and specifications of the system are proposed. In this paper, the hardware constitution and the software design method of the system are introduced in detail, and the working process of the asphalt concrete mixer can be controlled automatically and intelligently in real time. Moreover, the functions of batching control, process monitoring and data management has been realized. Test results show that the asphalt concrete mixer works stably and reliability.

Wheat, which is the most important food crop worldwide, is a cereal that presents considerable potential for increased yield. A new wheat (Triticum aestivum L.) mutant tasg1 with delayed leaf senescence was constructed using ethyl methane sulfonate as a mutagen. Natural senescence in tasg1 was distinctly delayed in the field, as indicated by the slower progression of chlorophyll degradation, net photosynthetic rate than its wild type. Further, the malondialdehyde and the hydrogen peroxide content was lower and antioxidative enzyme activity higher in tasg1 than those in its wild type during both natural senescence and methyl viologen-induced oxidative stress. The results suggest that tasg1 is a functional stay-green wheat mutant with the Type B (in which senescence initiates on schedule, but progresses at a rate lower than that in the respective WTs) or Type A (in which senescence initiates late but proceeds at a normal rate) and B combination and that the competence of the antioxidant defense system is one of the most important mechanisms underlying the expression of the stay-green phenotype.

The effects of positive and reverse working time, electroplating time, current density, duty cycle on the weight-gain rate were investigated. The optimized technical parameters were as follows: room temperature, 300 ms of positive working time, 30 ms of reverse working time, 40% of duty cycle, 8.20 ×10-5 A/mm2 of current density, 6 min of electroplating time. The quality of copper coating was checked with SEM. The adhesion between the coating and carbon fibers was tested by the method of psychro-thermal cycles. Additionally, comparison to pulse plating and direct current plating was made. The obtained results showed that a flat, detailed and well-crystalloid copper coating on the carbon fiber surface was obtained with the present conditions. The adhesion between the coating and carbon fibers was 450kPa.

The asynchronous induction motor has obtained more applications in electric vehicle (EV) or hybrid EV drive systems. Above all, this Paper discusses several main factors which influence harmonics elimination, and then, describes an effective online method for identifying both stator and rotor resistance. The novel method uses Suboptimal Multiple Fading Extended Kalman Filter (SMFEKF) which is a kind of strong tracking filters can accomplish Parameter Identification. An important advantage of this method is that it offers a parallel search way can overcome local optima so as to find the globally optimal solution. Together with harmonic elimination Algorithm which improved the power factor of the drive and weaken the injected low-order harmonics to the induction motor, robust speed control of induction motor can obtained easily. The simulative results confirm that the proposed method has efficiency to EV Drive Systems.

Within their natural habitat, crops are often subjected to drought and heat stress, which suppress crop growth and decrease crop production. Causing overaccumulation of glycinebetaine (GB) has been used to enhance the crop yield under stress. Here, we investigated the response of wheat (Triticum aestivum L.) photosynthesis to drought, heat stress and their combination with a transgenic wheat line (T6) overaccumulating GB and its wild-type (WT) Shi4185. Drought stress (DS) was imposed by controlling irrigation until the relative water content (RWC) of the flag leaves decreased to between 78 and 82%. Heat stress (HS) was applied by exposing wheat plants to 40°C for 4 h. A combination of drought and heat stress was applied by subjecting the drought-stressed plants to a heat stress as above. The results indicated that all stresses decreased photosynthesis, but the combination of drought and heat stress exacerbated the negative effects on photosynthesis more than exposure to drought or heat stress alone. Drought stress decreased the transpiration rate (Tr), stomatal conductance (Gs) and intercellular CO2 concentration (Ci), while heat stress increased all of these; the deprivation of water was greater under drought stress than heat stress, but heat stress decreased the antioxidant enzyme activity to a greater extent. Overaccumulated GB could alleviate the decrease of photosynthesis caused by all stresses tested. These suggest that GB induces an increase of osmotic adjustments for drought tolerance, while its improvement of the antioxidative defense system including antioxidative enzymes and antioxidants may be more important for heat tolerance.

To investigate the role of glycine betaine in photosynthesis under stress, a transgenic wheat (Triticum aestivum L.) line T6 overaccumulating glycine betaine and its wild type Shi4185 were used. Seedlings were exposed to conditions of drought (30%, PEG-6000), heat (40°C) and their combination. The results revealed ultrastructural damage to the chloroplast and thylakoid lamellae with the withered phenotype by both drought and heat stress, and the damage was exacerbated by the combination of drought and heat. The appearance of a K step in the typical O-J-I-P curve and the decrease of Hill activity indicated a reduction of oxygen evolving complex function caused by stress. The greater damage was found in wild type than T6. Overaccumulation of glycine betaine in T6 could protect lipids in the thylakoid membrane from damage and stabilize the index of unsaturated fatty acids under stress. A lower ratio of monogalactosyl diacylglycerol/digalactosyl diacylglycerol and higher phosphatidylglycerol content in the thylakoid membrane of T6 were also observed under stress. These effects can promote stability of the thylakoid membrane. Otherwise, glycine betaine overaccumulation decreased photoinhibition of PSII under stress. The results also suggest that xanthophyll cycle-dependent non-radiative energy dissipation may be involved in the GB-mediated effects on PSII function under stress conditions.

In this paper, we advance a fast approximative iterative inversion technique (FAIIT) of dual induction tool (DIT) logs. First, we establish the fast algorithm of Frechet derivatives of DIT logs with respect to bed interfaces and formation resistivities simultaneously, and give the process of the iterative inversion by use of normalization and singular value decomposition (SVD). The initial model parameters including interface and resistivities per bed must be extracted by comprehensively blocking of DIT logs for realization of the inversion of both synthetic and field logs. After the initial model is obtained, the bed resistivity and interface per bed are iteratively revised through the inversion of the medium induction log (IM) only. Then, deep induction log (ID) is only used to reconstruct other bed resistivities. Due to the difference of ID and IM at exploring depth, the former can obviously give shallower resistivities at exploring depth than the later. The inversion results of both synthetic and field logs clearly prove that the inversion results from ID and IM respectively can demonstrate the exact characteristics of formation invasion. Besides, because of ID with deeper exploring depth, inversion results of ID are largely closer to formation virgin resistivities than apparent resistivity of ID in beds of either deep or shallow invasion, so the better estimation of formation virgin resistivity is also obtained.

The bacterial diversity in the biological desulfurization reactor operated continuously for 1 year was studied by the 16S rDNA cloning and sequencing method. Forty clones were randomly selected and their partial 16S rDNA genes (ca. 1,400 bp) were sequenced and blasted. The results indicated that there were dominant bacterias in the biological desulfurization reactor, where 33 clones belonged to 3 different published phyla, while 1 clone belonged to unknown phylum. The dominant bacterial community in the system was Proteobacteria, which accounted for 85.3%. The bacterial community succession was as follows: the gamma-Proteobacteria(55.9%), beta-Proteobacteria(17.6%), Actinobacteridae (8.8%), delta-Proteobacteria (5.9%) , alpha-Proteobacteria(5.9%), and Sphingobacteria (2.9%). Halothiobacillus sp. ST15 and Thiobacillus sp. UAM-I were the major desulfurization strains.

Introduction: The esophagus squamous cell carcinoma (ESCC) is one of the most deadly malignances, and a current challenge is the development of effective therapeutic agents. Our present work addressed the effect of HIF-1α siRNA alone or in combination with cisplatin on the growth of ESCC in nude mice. Materials and Methods: Xenografts were established by inoculating ESCC TE-1 cells in nude mice, and transplanted tumors were treated with HIF-1α siRNA, cisplatin alone or together. Growth was assessed by measuring tumor volume. HIF-1α mRNA and protein expression were detected using RT-PCR and immunohistochemistry, respectively. Apoptosis of ESCC TE-1 cells was analyzed by flow cytometry. Results: In our nude mice model, HIF-1α siRNA effectively inhibited the growth of transplanted ESCC, downregulating HIF-1α mRNA and protein expression, and inducing ESCC TE-1 cell apoptosis. Notably when combinated with cisplatin, HIF-1α siRNA showed synergistic interaction in suppressing tumor growth. A laboratory study was made on the calling and mating behaviors of adult Orthaga achatina at (27 +/- 1) degrees C, (60 +/- 10)% RH, and 14 L: 10 D photoperiod. The female O. achatina did not call until the start of scotophase, with the calling percentage increased sharply at the 5th hour and reached the peak at the 6th-7th hour of scotophase. The calling percentage varied with moth age, being the highest (70%) at the age of 2-3 d. The mating behavior could be divided into two successive periods, i.e., calling period and mating period. The mating between female and male O. achatina occurred during the 5th-9th hour of scotophase, with the peak at the 6th-7th hour into scotophase, which was in accordance with the peak time of female calling behavior. Females mated only one time during their whole life. Treatment 1 female : 2 male showed a significantly higher mating percentage, but a significantly shorter mating duration time, compared with treatment 1 female : 1 male.

Recent evidence suggests that rapid eye movement (REM) sleep deprivation (REMSD) causes learning and memory deficits. However, the mechanism of REMSD-induced memory impairment remains unclear. Calcineurin (CaN) is involved in synaptic plasticity and is known as a negative constraint on learning and memory. Here we report that 72 h REMSD by the modified multiple platform method in rats resulted in spatial memory impairment in the Morris water maze and elevated hippocampal cytosolic CaN activity, both of which were reversed after 18 h sleep recovery. CaN expression in the whole-tissue homogenate of the hippocampus was not altered by REMSD. The results suggest that elevated hippocampal CaN activity is involved in REMSD-induced spatial memory impairment.

The fusion protein (F) and attachment glycoprotein (G) of Nipah virus (NiV) are important for the virus to infect cells and induce protective immunity. In this study, the NiV F1 and G gene fragments without the sequences of signal peptide and transmembrane domain were amplified by PCR, then cloned into E. coli expression vector pGEX-6P-1 and modified baculovirus vector, respectively. After induction by IPTG, NiV F1 and G proteins were efficiently expressed in E. coli when analyzed by SDS-PAGE, both showing good reactivity with the rabbit antiserum anti-NiV serum in Western blot. The expression of NiV F1 and G in baculovirus system were also detected by indirect immunofluorescent assay (IFA) of fixed Sf9 cells monolayer infected with the recombinant baculoviruses expressing F1 and G. Furthermore the anti-F1 and anti-G hyperimmune sera were prepared by immunization of rabbits respectively with purified E. coli-expressed F1 and G proteins. Western blot and IFA as well as ELISA showed that antisera against both protein had high titers with good reactivity and specificity. The present study has provided a base for development of diagnostic reagents for detection of NiV infection.

Mab35 is a monoclonal antibody against one specific immunogenic region in alpha1, alpha3, alpha5 subunits of nicotinic acetylcholine receptors (N-AChR) of a variety of species. It has previously been claimed that N-AChR-like immunoreactivity (-LI) identified by mab35 is present in vasopressin-containing magnocellular neurons. However, we show here by double immunofluorescence labelling that mab35 immunoreactivity is predominantly localized to oxytocinergic rather than vasopressinergic magnocellular neurons. We further infer that mab35 predominantly stained the alpha3 and/or alpha5 subunits in rat oxytocinergic neurons, and suggest that the unbalanced distribution of these subunits may contribute to some specific physiological properties of oxytocinergic neurons.
