Introduction: Fraser Syndrome is a rare genetic disorder characterized by urogenital defects, cutaneous syndactyly and cryptophthalmos commonly diagnosed during foetal autopsy. Vaginal atresia is one of the major diagnostic criteria of Fraser Syndrome and could be a contributing factor of endometriosis following the development of hematocolpos. Imperforate hymen, often misdiagnosed as vaginal atresia, is a rare diagnosis in patients with Müllerian anomalies. Here, we report a case of Fraser Syndrome with chronic pelvic pain as a result of delayed hymenotomy and the extensive management of endometriosis. Case Report: A 15-year-old girl with Fraser Syndrome presented with pelvic pain. Examination revealed a large hematocolpos caused by an imperforate hymen. Due to the lack of paediatric intensive care locally, she had a delayed hymenotomy. She continued to suffer from chronic pelvic non-cyclical pain post-hymenotomy. Preliminary laparoscopy and biopsy showed endometriosis. Despite excision and clearance of endometriosis, she experienced persistent pain while taking morphine and was trialled with several hormonal therapies such as leuprorelin acetate, progestogen and combined hormonal therapy. She received medical therapy for 10 years until she re-presented with erratic bleeding and pain. Laparoscopy again identified endometriosis. Conclusion: Early recognition and treatment minimize risk of endometriosis especially in premenarcheal adolescent girls with obstructive Müllerian anomalies presenting with pelvic pain. Adequate pain control and medical management permit a delay in surgical intervention facilitating further investigations and thorough counselling with the patient and family about the implications of endometriosis on fertility and quality of life. Complete inactivation of infectious Ebola virus (EBOV) is required before a sample may be removed from a Biosafety Level 4 laboratory. The United States Federal Select Agent Program regulations require that procedures used to demonstrate chemical inactivation must be validated in-house to confirm complete inactivation. The objective of this study was to develop a method for validating chemical inactivation of EBOV and then demonstrate the effectiveness of several commonly-used inactivation methods. Samples containing infectious EBOV (Zaire ebolavirus) in different matrices were treated, and the sample was diluted to limit the cytopathic effect of the inactivant. The presence of infectious virus was determined by assessing the cytopathic effect in Vero E6 cells. Crucially, this method did not result in a loss of infectivity in control samples, and we were able to detect less than five infectious units of EBOV (Zaire ebolavirus). We found that TRIzol LS reagent and RNA-Bee inactivated EBOV in serum; TRIzol LS reagent inactivated EBOV in clarified cell culture media; TRIzol reagent inactivated EBOV in tissue and infected Vero E6 cells; 10% neutral buffered formalin inactivated EBOV in tissue; and osmium tetroxide vapors inactivated EBOV on transmission electron microscopy grids. The challenges of sampling rare fauna limit efforts to understand and mitigate the factors that restrict their distribution. Camera traps have become a standard technique for sampling large mammals, but their utility for sampling small, rare species remains largely unknown. The central rock-rat ( Zyzomys pedunculatus ) is critically endangered and restricted to rugged range country in central Australia. Using Z. pedunculatus as a focal species, we sought to evaluate the effectiveness of camera trapping for sampling small mammals in this environment and to better understand the factors driving the occurrence of this species. We installed baited camera traps at 50 sites across 1795 ha of core refuge habitat for Z. pedunculatus . We recorded all six species of small mammals known previously from this area, including the highly detectable Z. pedunculatus at five sites. Occupancy modelling showed that distance to the nearest occupied site was the most important predictor of Z. pedunculatus occurrence, suggesting that this rodent occurs in discrete sub-populations within the matrix of refuge habitat. Fire history and ruggedness may also influence occupancy of Z. pedunculatus at the landscape-scale and could assist in locating additional sub-populations. At the site-scale, occupancy of Z. pedunculatus was high and there was no clear influence of any site-scale variables. Management of Z. pedunculatus will require protection and expansion of known sub-populations. We conclude that camera trapping provided useful and cost-effective insight into the factors limiting rock-rat distribution, and predict that it will become a standard tool for sampling rare small mammals in difficult-to-access environments. Patch-mosaic burning is a widely accepted practical approach to managing biodiversity, whereby spatial and temporal diversity of fire is manipulated to benefit biotic diversity. We use simulation experiments based on stochastic population viability analysis to evaluate the implications of contrasting patch-mosaic burning scenarios for the population dynamics and risk of decline of four species of small mammals in northern Australia. Our results, based on models developed from detailed mark-recapture data, suggest that fire frequency has more influence on small-mammal persistence than fire extent. Risk of extinction increased for all four species when fire frequency exceeded once every five years. Under current ambient fire regimes most Australia tropical savannas burn more frequently and therefore seem to have unfavourable consequences for this faunal group and risk precipitating severe future declines. Abstract: The recycling of iron bearing dusts produced during iron and steel manufacturing processes is vital to the sustainability of these processes; however, contamination of these dusts with zinc increases the difficulty to do this. Blast furnace dust, collected by a wet scrubber system, was sampled and characterised to investigate the removal of zinc to produce a treated residue with low zinc content suitable for recycling through the blast furnace. This paper examines a leaching process for the dust using different organic carboxylic acids, to establish if they were capable of extracting high levels of zinc and low levels of iron. Prop-2-enoic acid was found to be particularly effective, extracting high levels of zinc up to 85.7% and low levels of iron, 8.5%. The paper also discusses the mechanisms of extraction more specifically for organic carboxylic acids and found that the iron extraction was well explained by the variation in pH and the Bronsted–Lowry theory; whereas zinc extraction was well explained by substituent group effects and the Lewis acid/base theory. The novel use of a non-aqueous solvent with prop-2-enoic acid, to minimise the ion solvating ability and proton dissociation, was found to reduce the level of iron extraction from 8.5% to 0.1% without detrimental effect on zinc extraction when leaching. A range of mineral and carboxylic acids were also tested to investigate and compare the effect of pH and chemical structure on the leaching efficacy. [Copyright &y& Elsevier]

Background: Anecdotally, it is suggested that patients with preeclampsia have a shorter induction to delivery interval than patients without preeclampsia, despite there being no good objective evidence.Methods: A retrospective analysis of 136 primiparous, singleton, cephalic, nonassisted vaginal deliveries between January 1997 and January 2000. Sixty-eight women had induction of labor for preeclampsia and 68 women without preeclampsia had induction of labor for other reasons. The two groups were already case-matched for maternal and gestational age. The following details were recorded from the case notes: maternal age, gestation at delivery, initial Bishop score, total dose of prostaglandin administered, induction to delivery interval, duration of second-stage labor, birthweight, and the percentage of Apgar scores < 7 at 5 min. Statistical analysis was carried out with a Mann-Whitney U-test, with the aid of SPSS version 9, Microsoft WindowsResults: The women with preeclampsia had a statistically significant longer induction to delivery interval than those without preeclampsia. The mean induction to delivery interval of women with and without preeclampsia was 1049.5 min and 762.5 min, respectively (p < 0.001)Conclusion: Women with preeclampsia had a longer induction to delivery interval than patients without preeclampsia. A high diversity of rock-dwelling macropod species inhabit the tropics of the Northern Territory, Australia. Within this region, individual species have varied distributions, with ranges variously being widespread, restricted, or geographically disjunct with variable levels of sympatry between them. The cause of these patterns is unknown and little is known of the ecology of these rock-dwelling macropods. We hypothesized that differences in the habitat requirements is the key to understanding the biogeography and sympatry of the species. We examined this hypothesis at both regional and local scales. We analysed records of occurence of Petrogale brachyotis, P. concinna, Macropus bernardus and M. robustus and environmental correlates (such as geology type, vegetation type, distance to drainage and distance to ‘rugged’ terrain) throughout the monsoon tropics using geographic information systems and generalized linear modelling. We surveyed 80 sites across the tropics of the Northern Territory and collected presence-absence data using scats and environmental correlates to examine fine-scale habitat requirements. From the regional scale analysis, it was clear that distance to rugged terrain strongly influences presence of all four species. Responses to this variable suggest M. bernardus and P. concinna have greater requirements for rugged terrain, whereas M. robustus and P. brachyotis are found across a broader range of habitats. The local scale analysis suggested presence of M. robustus is primarily driven by vegetation type, M. bernardus by the cover of outcrops, boulders and large rocks, and P. brachyotis by the density of caves. Although these species overlap in their use of habitats, there are also differences in their habitat requirements that are likely to play a role in their niche separation and in some cases their biogeography. However, it is apparent that the distributions of the species have also been influenced by historical factors given the absence of these species from some apparently suitable sites. Background: Rectovaginal endometriosis is a severe form of pelvic endometriosis in which pharmacological treatment is relatively ineffective (Vercellini et al., Fertil Steril. 2005;84:1375-87). Laparoscopic surgical treatment is effective, but has the potential risks of bowel perforation and colostomy formation (Darai et al., Am J Obstet Gynecol. 2005;192:394-400). Transrectal ultrasound scanning can be applied as a preoperative tool to predict the presence of rectovaginal endometriosis and bowel wall involvement (Abrao et al., J Am Assoc Gynecol Laparosc. 2004;11:50-4). Methods: Thirty-two women underwent transrectal ultrasound followed by therapeutic laparoscopy. Likelihood ratios and post-test prevalences were calculated with Fagan's normogram. This was then extrapolated with the aid of a mathematical model to a low-risk population. Results: A positive likelihood ratio was found to be 10.89 (95% confidence ratio (CI): 1.62-73.15) and a negative likelihood ratio was found to be 0.24 (95% CI: 0.1-0.57). The pre-test prevalence of rectovaginal endometriosis was 56%. The positive post-test prevalence probability was 93%, and the negative post-test prevalence probability was 23%. Conclusion: Preoperative transrectal ultrasound scanning for rectovaginal endometriosis is an extremely accurate predictive test, and strongly predicts the need for extensive laparoscopic dissection and potential bowel resection. Abstract: Objective: The purpose of this prospective study was to record endothelin-1 (ET-1) concentrations in the second trimester amniotic fluid and compare these values in women who developed intrauterine growth restriction (IUGR) later in pregnancy with those with uneventful pregnancies. Method: Amniotic fluid was retrieved by amniocentesis from 125 women in the second trimester of pregnancy. The levels of ET1 were measured by a sensitive and specific radioimmunoassay. Results: From the 125 women included in the study 12 had pregnancies that later developed IUGR and 88 had uneventful pregnancies. The ET1 concentration was significantly higher (P <0.005) in women who later developed IUGR than in normal pregnancy (106pg/ml versus 64.7pg/ml). Conclusion: The amniotic fluid concentration of ET1 is elevated by the second trimester in women who later develop IUGR. [Copyright &y& Elsevier]

We have investigated a herpes simplex virus mutant that expresses low levels of thymidine kinase (TK), a phenotype associated with drug resistance and pathogenicity, despite a single-base deletion it the gene. Using a dual-reporter system, a 39-nt sequence including the mutation was shown to direct expression of the downstream reporter gene in reticulocyte lysate. Translation of the downstream reporter was not impaired when the mRNA lacked a 5' cap or had a stable stem loop 5' of the upstream reporter and was relatively resistant to edeine, an antibiotic that prevents AUG codon recognition by the 40S-elF2-GTP/Met-tRNAi complex. Twelve nucleotides were as active as the original sequence for translation of the downstream reporter. Surprisingly, this sequence lacks an AUG codon. Analysis of point mutations showed that a CUG codon in the sequence was important. However, many single-base changes had only limited effects, and introduction of AUG codons did not increase translation. A mutant virus containing both the single- base deletion and a mutation that reduced downstream translation in vitro had significantly less TK activity than a virus with the single-base deletion alone. Thus, a remarkably short internal ribosome entry site (IRES) that lacks an AUG codon resides in the viral tk gene. The IRES appears to be responsible for TK expression from a drug-resistant mutant that would otherwise express no TK, which may contribute to pathogenicity. Because we found numerous short sequences with IRES activity, there might be many hitherto unrecognized polypeptides expressed at low levels from eukaryotic mRNAs. [ABSTRACT FROM AUTHOR]

Actin has a pivotal function in hyphal morphogenesis in filamentous fungi, but it is not certain whether its function is equivalent to that of a morphogen, or if it is simply part of a mechanism that executes orders given by another regulatory entity. To address this question we selected for cytochalasin A resistance and isolated act1, the first actin mutant in Neurospora crassa. This mutant branches apically and shows an altered distribution of actin at the tip. Based on the properties of this mutant, we propose a model of tip growth and branching in which actin effects tip growth by regulating the rate of vesicle flow from proximal to distal regions of a hypha, thereby controlling the tip-high gradient of cytoplasmic calcium. The actin-controlled calcium gradient at the tip is necessary for maintenance of tip growth as well as the dominance of one polarized site at the hyphal tip. There are at least 100 loci encoding products that influence tip growth and branching in Neurospora crassa. The functional relationships between 38 of these loci were examined by an analysis of gene interaction in double mutants. A complex range of interactions was revealed. These have been grouped into full and partial epistasis, costasis, novel phenotypes, and synthetic lethality and sublethality. Epistasis was used to construct the simplest “pathway” that accommodated the results; this pathway was Y-shaped. If synthetic sublethality is interpreted to reflect mutations in the same pathway, the sublethal connections are compatible with the chart of epistasis. Aboriginal people in remote Australian communities have limited opportunity to participate in the market economy. The Aboriginal arts and craft industry offers an important option to generate cash income and the sustained supply of native plants used in production is vital. We assessed the current harvest of a rainforest tree species commonly used for sculpture, Bombax ceiba. Information from field surveys, a regional rainforest inventory and sales figures from the Maningrida region were used to estimate the population size and structure of B. ceiba, the harvest intensity of patches and the economic value of the harvest. We estimate the density of B. ceiba to be 105.10±12.32 stems ha−1 and a regional population of 76615±14063 stems. A cumulative harvest over 20 years of 6% suggests approximately 4596 stems harvested, and the harvest was worth about $200,000 over the last three years. The majority (80%) of harvested stems coppiced and were suitable for harvesting. Logistic regression suggests both distance to outstation and township influence harvesting intensity, with some localised over-harvest. Rainforests provide a valuable resource to Aboriginal artists in Maningrida and their maintenance provides important conservation benefits to the Australian community. [Copyright &y& Elsevier]

Herpes simplex virus thymidine kinase is important for reactivation of virus from its latent state and is a target for the antiviral drug acyclovir. Most acyclovir-resistant isolates have mutations in the thymidine kinase gene; however, how these mutations confer clinically relevant resistance is unclear. Reactivation from explanted mouse ganglia was previously observed with a patient-derived drug-resistant isolate carrying a single guanine insertion within a run of guanines in the thymidine kinase gene. Despite this mutation, low levels of active enzyme were synthesized following an unusual ribosomal frameshift. Here we report that a virus, generated from a pretherapy isolate from the same patient, engineered to lack thymidine kinase activity, was competent for reactivation. This suggested that the clinical isolate contains alleles of other genes that permit reactivation in the absence of thymidine kinase. Therefore, to establish whether thymidine kinase synthesized via a ribosomal frameshift was sufficient for reactivation under conditions where reactivation requires this enzyme, we introduced the mutation into the well-characterized strain KOS. This mutant virus reactivated from latency, albeit less efficiently than KOS. Plaque autoradiography revealed three phenotypes of reactivating viruses: uniformly low thymidine kinase activity, mixed high and low activity, and uniformly high activity. We generated a recombinant thymidine kinase-null virus from a reactivating virus expressing uniformly low activity. This virus did not reactivate, confirming that mutations in other genes that would influence reactivation had not arisen. A double-guanine-insertion mutation within a run of guanines in the herpes simplex virus gene encoding thymidine kinase (TK) was previously found in an acyclovir-resistant clinical isolate. This mutation was engineered into strain KOS, and stocks were generated from single plaques. Plaque autoradiography revealed that most plaques in such stocks exhibited low levels of TK activity, while ∼3% of plaques exhibited high levels of TK activity, indicating a remarkably high frequency of phenotypic reversion. This virus was able to reactivate from latency in mouse ganglia; a fraction of the reactivating virus expressed a high level of TK activity due to an additional G insertion, suggesting that the observed genetic instability contributed to pathogenicity. The Carpentarian rock-rat (Zyzomys palatalis) is a critically endangered endemic rodent known from only four sandstone gorges in the southeast Gulf of Carpentaria, Northern Territory, Australia. These gorges harbour thickets of monsoon rainforest and broadleaf woodland, surrounded by a Eucalypt savanna matrix. The long-term persistence of Z. palatalis is threatened by altered fire regimes, grazing by feral animals and stock, weed intrusion, and the stochastic hazards associated with small, fragmented populations. To assess the relative importance of these threats and develop practical management options, a population and habitat simulation model was developed, based on the best existing data. Population viability was predicted to be highly sensitive to the frequency of hot, late dry-season fires. Progressive habitat degradation (due predominantly to intense late dry-season fires) is likely to substantially reduce population size and lead to the probable extinction of the species within the next 100 years. The most effective management strategy to counteract this threat would be regular, controlled, fuel reduction burns in the vegetation around the gorge entrances during the early dry season. Establishing a new population (through translocation of captive-bred individuals) would not appreciably reduce extinction risk, but could provide valuable additional data on the impact of threats, if conducted as an adaptive management experiment. Abstract The impact of dry deposition of SO2 emissions on ant abundance, diversity and composition was investigated at Mount Isa in the semiarid tropics of northern Australia. Forty plots were sampled, stratified at two levels: sulfur deposition zones (high, medium, low, and two control zones) and habitat (Ridge and Plain). The two habitats supported distinctly different ant communities. Ants had clear responses to SO2 emissions. Ant abundance was lowest in the high and medium sulfur zones in both habitats. Species richness in high SO2 plots (up to 5 km from the source) was approximately half that of control plots in Ridge habitat, and was substantially less than controls in the Plain habitat. Ant community composition in the high sulfur zone was clearly separated from those of other zones in ordinations. Vector fitting showed soil SO4 concentration as a primary correlative factor in this separation. Ant abundance and richness were both negatively correlated with soil SO4 concentration, and positively correlated with plant species richness and distance away from the smelters. The abundance of 10 of the 21 most common species showed significant responses to emissions. Five species showed positive responses, and all belong to species-groups known to be abundant at disturbed sites throughout northern Australia. Relative abundance and richness of Eyrean (arid adapted) taxa collectively responded positively to sulfur, and Torresian (tropical) and Widespread species responded negatively. Despite large changes in species composition and abundances, there was relatively little change in the abundance of functional groups that have been widely used in studies of Australian ant communities. The group of patients who received urinary gonadotropins (n = 117) for follicular stimulation had a significantly higher incidence of high perifollicular blood flow compared with that of the group who received recombinant FSH (n = 114; 46.3% vs. 22.7%). Monoclonal antibodies (mAbs) are an important class of antiviral therapeutics. MAbs are highly selective, well tolerated, and have long in vivo half-life as well as the capacity to induce immune-mediated virus clearance. Their activities can be further enhanced by integration of their variable fragments (Fvs) into bispecific antibodies (bsAbs), affording simultaneous targeting of multiple epitopes to improve potency and breadth and/or to mitigate against viral escape by a single mutation. Here, we explore a bsAb strategy for generation of pan-ebolavirus and pan-filovirus immunotherapeutics. Filoviruses, including Ebola virus (EBOV), Sudan virus (SUDV), and Marburg virus (MARV), cause severe hemorrhagic fever. Although there are two FDA-approved mAb therapies for EBOV infection, these do not extend to other filoviruses. Here, we combine Fvs from broad ebolavirus mAbs to generate novel pan-ebolavirus bsAbs that are potently neutralizing, confer protection in mice, and are resistant to viral escape. Moreover, we combine Fvs from pan-ebolavirus mAbs with those of protective MARV mAbs to generate pan-filovirus protective bsAbs. These results provide guidelines for broad antiviral bsAb design and generate new immunotherapeutic candidates. The emergence of highly transmissible severe acute respiratory syndrome coronavirus 2 (SARS-CoV-2) variants of concern (VOCs) that are resistant to the current COVID-19 vaccines highlights the need for continued development of broadly protective vaccines for the future. Here, we developed two messenger RNA (mRNA)-lipid nanoparticle (LNP) vaccines, TU88mCSA and ALCmCSA, using the ancestral SARS-CoV-2 spike sequence, optimized 5' and 3' untranslated regions (UTRs), and LNP combinations. Our data showed that these nanocomplexes effectively activate CD4+ and CD8+ T cell responses and humoral immune response and provide complete protection against WA1/2020, Omicron BA.1 and BQ.1 infection in hamsters. Critically, in Omicron BQ.1 challenge hamster models, TU88mCSA and ALCmCSA not only induced robust control of virus load in the lungs but also enhanced protective efficacy in the upper respiratory airways. Antigen-specific immune analysis in mice revealed that the observed cross-protection is associated with superior UTRs [Carboxylesterase 1d (Ces1d)/adaptor protein-3β (AP3B1)] and LNP formulations that elicit robust lung tissue-resident memory T cells. Strong protective effects of TU88mCSA or ALCmCSA against both WA1/2020 and VOCs suggest that this mRNA-LNP combination can be a broadly protective vaccine platform in which mRNA cargo uses the ancestral antigen sequence regardless of the antigenic drift. The International Committee on Taxonomy of Viruses (ICTV) Filoviridae Study Group continues to prospectively refine the established nomenclature for taxa included in family Filoviridae in an effort to decrease confusion of genus, species, and virus names and to adhere to amended stipulations of the International Code of Virus Classification and Nomenclature (ICVCN). Recently, the genus names Ebolavirus and Marburgvirus were changed to Orthoebolavirus and Orthomarburgvirus, respectively. Additionally, all established species names in family Filoviridae now adhere to the ICTV-mandated binomial format. Virus names remain unchanged and valid. The article studies the growth of desmoid tumors with familial adenomatous polyposis during pregnancy. A 35-year-old woman who was known to have familial adenomatous polyposis was first referred to antenatal clinic at 11 weeks gestation. In addition she was noted to have essential hypertension treated with alpha methyldopa and an asymptomatic ventricular septal defect confirmed on two-dimensional echocardiography at 15 weeks of pregnancy. Her obstetric history included five previous pregnancies. To obtain a histological diagnosis it was suggested that a biopsy would be prudent. However, there was some concern about impairment of progress in labor due to tumors.

RuNPs stabilised by amino‐decorated imidazolium‐based polymer immobilized ionic liquids catalyse the dimethylamine borane mediated reduction of quinolines to 1,2‐dihydroquinoline (DHQ) and 1,2,3,4‐tetrahydroquinoline (THQ). Partial reduction of 3‐substituted quinolines to the corresponding 1,2‐dihydroquinoline was achieved with 100 % selectivity in toluene under mild conditions. This is the first report of the selective partial reduction of 3‐substituted quinolines to the corresponding 1,2‐dihydroquinolines with a heterogeneous nanoparticle‐based catalyst. A wide range of substituted quinolines have also been reduced to the corresponding 1,2,3,4‐tetrahydroquinoline with high selectivity and good yields by adjusting the reaction time. The 1,2‐dihydroquinolines readily release dihydrogen in toluene at 60 °C in the absence of catalyst with no evidence for disproportionation and as such are potential organo‐hydride reagents. The initial TOF of 610 mol quinoline converted mol Ru−1 h−1 for the reduction of quinoline is among the highest to be reported for a metal nanoparticle‐based catalyst and the conversion of 96 % obtained after 4 h at 65 °C is significantly higher than its platinum nanoparticle counterpart PtNP@NH2‐PEGPIILS as well as 5 wt/% Ru/C, which only reached 9 % and 11 % conversion, respectively, at the same time. A microfluidic sample preparation multiplexer (SPM) and assay procedure is developed to improve amplification-free detection of Ebola virus RNA from blood. While a previous prototype successfully detected viral RNA following off-chip RNA extraction from infected cells, the new device and protocol can detect Ebola virus in raw blood with clinically relevant sensitivity. The Ebola RNA is hybridized with sequence specific capture and labeling DNA probes in solution and then the complex is pulled down onto capture beads for purification and concentration. After washing, the captured RNA target is released by irradiating the photocleavable DNA capture probe with ultraviolet (UV) light- The released, labeled, and purified RNA is detected by a sensitive and compact fluorometer. Exploiting these capabilities, a detection limit of 800 attomolar (aM) is achieved without target amplification. The new SPM can run up to 80 assays in parallel using a pneumatic multiplexing architecture. Importantly, our new protocol does not require time-consuming and problematic off-chip probe conjugation and washing. Monoclonal antibody (mAb) therapies have rapidly become a powerful class of therapeutics with applications covering a diverse range of clinical indications. Though most widely used for the treatment of cancer, mAbs are also playing an increasing role in the defense of viral infections, most recently with palivizumab for prevention and treatment of severe RSV infections in neonatal and pediatric populations. In addition, during the COVID-19 pandemic, mAbs provided a bridge to the rollout of vaccines; however, their continued role as a therapeutic option for those at greatest risk of severe disease has become limited due to the emergence of neutralization resistant Omicron variants. Although there are many techniques for the identification of mAbs, including single B cell cloning and immunization of genetically engineered mice, the low cost, rapid throughput and technological simplicity of antibody phage display has led to its widespread adoption in mAb discovery efforts. Here we used our 27-billion-member naïve single-chain antibody (scFv) phage library to identify a panel of neutralizing anti-SARS-CoV-2 scFvs targeting diverse epitopes on the receptor binding domain (RBD). Although typically a routine process, we found that upon conversion to IgG, a number of our most potent clones failed to maintain their neutralization potency. Kinetic measurements confirmed similar affinity to the RBD; however, mechanistic studies provide evidence that the loss of neutralization is a result of structural limitations likely arising from initial choice of panning antigen. RuNPs stabilised by a polymer immobilised ionic liquid derived from co-polymerisation of a PEG-substituted imidazolium-based styrene monomer and diphenyl(4-vinylphenyl)phosphine oxide, RuNP@O = PPh 2 -PEGPIILS, (2) is a remarkably efficient and selective catalyst for the hydrazine hydrate-mediated partial reduction of nitroarenes to the corresponding N -arylhydoxylamine. Near quantitative conversion to N -phenylhydroxylamine with > 99 % selectivity was obtained after only 2 h when the reaction was conducted at 25 °C in ethanol under an inert atmosphere using 0.1 mol% catalyst. Under these conditions, the composition-time profile showed that the reduction occurred via the direct pathway whereas reactions in air gave a mixture of azoxy-based products due to competing condensation resulting from reversible formation of N -phenylhydroxylamine. The initial TOF of 6,100 h−1 obtained after 10 min at 40 °C with 0.1 mol% 2 is among the highest to be reported for the metal nanoparticle catalysed reduction of nitrobenzene to N -phenylhydroxylamine and a significant improvement on 5 wt% Ru/C which gave a modest conversion of 21 % (initial TOF = 240 h−1) to a mixture of N -phenylhydroxylamine and aniline. A broad range of substituted N -aryl and N -heteroaryl nitroarenes were reduced to the corresponding N -arylhydroxylamine in high yield and with excellent selectivity by adjusting the reaction times. However, reduction of electron rich amino-substituted nitroarenes was extremely slow and resulted in reduction to the aniline with no evidence for the corresponding hydroxylamine. Complete reduction of amino substituted nitroarene is proposed to be facilitated by amine-assisted elimination of hydroxide from the hydroxylamine to afford a readily reducible quinondiimine-derived iminium intermediate that reacts with a surface hydride to liberate the amine. Under optimum conditions the catalyst could be reused five times for the reduction of nitrobenzene to N -phenylhydroxylamine with no detectable change in activity and only slight decrease in selectivity. Coronavirus disease 2019 (COVID-19) has had devastating effects worldwide, with particularly high morbidity and mortality in outbreaks on residential care facilities. Amantadine, originally licensed as an antiviral agent for therapy and prophylaxis against influenza A virus, has beneficial effects on patients with Parkinson's disease and is used for treatment of Parkinson's disease, multiple sclerosis, acquired brain injury, and various other neurological disorders. Recent observational data suggest an inverse relationship between the use of amantadine and COVID-19. Adamantanes, including amantadine and rimantadine, are reported to have in vitro activity against severe acute respiratory syndrome coronavirus (SARS-CoV) and, more recently, SARS-CoV-2. We hypothesized that adamantanes have antiviral activity against SARS-CoV-2, including variant strains. To assess the activity of adamantanes against SARS-CoV-2, we used in vitro and in vivo models of infection. We established that amantadine, rimantadine, and tromantadine inhibit the growth of SARS-CoV-2 in vitro in cultured human epithelial cells. While neither rimantadine nor amantadine reduces lung viral titers in mice infected with mouse-adapted SARS-CoV-2, rimantadine significantly reduces viral titers in the lungs in golden Syrian hamsters infected with SARS-CoV-2. In summary, rimantadine has antiviral activity against SARS-CoV-2 in human alveolar epithelial cells and in the hamster model of SARS-CoV-2 lung infection. Abstract: Vermicompost was produced from a green waste compost feedstock and assessed for its potential use in a high value horticultural market. Replicated plant growth trials were undertaken with lettuce using pure worm cast (vermicompost), green waste-derived compost and mixtures of the two, i.e. 50/50 (v/v) and 20/80 (v/v) of worm casts and green waste feedstock. Results showed that plant biomass production was optimal with a 20/80 (v/v) compost blend, whilst pure worm cast and green waste compost yielded poor growth. Leaf chlorophyll content indicated that pure worm cast inhibited plant growth and depressed N content, whereas plant grown with the other treatments contained similar amounts of chlorophyll. In general, the vermicomposting process did not result in an increased availability of nutrients or potentially toxic elements, the only exception being Zn. [Copyright &y& Elsevier]

Many acyclovir-resistant herpes simplex virus isolates from patients contain insertions or deletions in homopolymeric sequences in the thymidine kinase (TK) gene (tk). Viruses that have one (G8) or two (G9) base insertions in a run of seven G's (G string) synthesize low levels of active TK (TK-low phenotype), evidently via ribosomal frameshifting. These levels of TK can suffice to permit reactivation from latently infected mouse ganglia, but in a majority of ganglia, especially with the G9 virus, reactivation of virus that has reverted to the TK-positive phenotype predominates. To help address the relative contributions of translational mechanisms and reversion in reactivation, we generated viruses with a base either inserted or deleted just downstream of the G string. Both of these viruses had a TK-low phenotype similar to that of the G8 and G9 viruses but with less reversion. Both of these viruses reactivated from latently infected trigeminal ganglia, albeit inefficiently, and most viruses that reactivated had a uniformly TK-low phenotype. We also generated viruses that have one insertion in a run of six C's or one deletion in a run of five C's. These viruses lack measurable TK activity. However, they reactivated from latently infected ganglia, albeit inefficiently, with the reactivating viruses having reverted to the wild-type TK phenotype. Therefore, for G-string mutants, levels of active TK as low as 0.25% generated by translational mechanisms can suffice for reactivation, but reversion can also contribute. MicroRNAs (miRNAs) are key regulators of gene expression in higher eukaryotes. Recently, miRNAs have been identified from viruses with double-stranded DNA genomes. To attempt to identify miRNAs encoded by herpes simplex virus 1 (HSV-1), we applied a computational method to screen the complete genome of HSV-1 for sequences that adopt an extended stem-loop structure and display a pattern of nucleotide divergence characteristic of known miRNAs. Using this method, we identified 11 HSV-1 genomic loci predicted to encode 13 miRNA precursors and 24 miRNA candidates. Eight of the HSV-1 miRNA candidates were predicted to be conserved in HSV-2. The precursor and the mature form of one HSV-1 miRNA candidate, which is encoded ∼450 bp upstream of the transcription start site of the latency-associated transcript (LAT), were detected during infection of Vero cells by Northern blot hybridization. These RNAs, which behave as late gene products, are not predicted to be conserved in HSV-2. Additionally, small RNAs, including some that are roughly the expected size of precursor miRNAs, were detected using probes for miRNA candidates derived from sequences encoding the 8.3-kilobase LAT, from sequences complementary to UL15 mRNA, and from the region between ICP4 and US1. However, no species the size of typical mature miRNAs were detected using these probes. Three of these latter miRNA candidates were predicted to be conserved in HSV-2. A collection of genomic DNA sequences of herpes simplex virus (HSV) strains has been defined and analyzed, and some information is available about genomic stability upon limited passage of viruses in culture. The nature of genomic change upon extensive laboratory passage remains to be determined. In this report we review the history of the HSV-1 KOS laboratory strain and the related KOS1.1 laboratory sub-strain, also called KOS (M), and determine the complete genomic sequence of an early passage stock of the KOS laboratory sub-strain and a laboratory stock of the KOS1.1 sub-strain. The genomes of the two sub-strains are highly similar with only five coding changes, 20 non-coding changes, and about twenty non-ORF sequence changes. The coding changes could potentially explain the KOS1.1 phenotypic properties of increased replication at high temperature and reduced neuroinvasiveness. While RuNP@NH2-PEGPIILS and PtNP@NH2-PEGPIILS catalyse the hydrolytic evolution of hydrogen from NaBH4 with comparable initial TOFs of 6,250 molesH2.molcat−1.h−1 and 5,900 molesH2.molcat−1.h−1, respectively, based on the total metal content, RuNP@NH2-PEGPIILS is a markedly more efficient catalyst for the dehydrogenation of DMAB and AB than its platinum counterpart, as RuNP@NH2-PEGPIILS gave initial TOFs of 8,300 molesH2.molcat−1.h−1 and 21,200 molesH2.molcat−1.h−1, respectively, compared with 3,050 molesH2.molcat−1.h−1 and 8,500 molesH2.molcat−1.h−1, respectively, for PtNP@NH2-PEGPIILS. Gratifyingly, for each substrate tested RuNP@NH2-PEGPIILS and PtNP@NH2-PEGPIILS were markedly more active than commercial 5wt % Ru/C and 5wt% Pt/C, respectively. Next-generation sequencing is now commonly used for a variety of applications in virology including virus discovery, investigation of quasispecies, viral evolution, metagenomics, and analyses of antiviral resistance. However, there are limitations with the current sample preparation methods used for deep sequencing of viral genomes, especially during de novo sequencing. For example, current methods are unable to capture the terminal sequences of viral genomes in an efficient and effective manner; data representing the 3′ and 5′ ends are typically insufficient. Methods such as Rapid Amplification of cDNA Ends address this issue but these methods can be time consuming, may require some prior knowledge of the viral sequence, and require multiple independent procedures. The current study outlines a sample preparation technique that overcomes some of these shortcomings. The method relied on random fragmentation with divalent cations and subsequent adapter ligation directly to RNA, rather than cDNA, to maximize the quality and quantity of terminal reads. The technique was tested on RNA samples from two different RNA viruses, Ebola virus and hepatitis C virus. This method permits rapid preparation of samples for deep sequencing while eliminating the use of sequence specific primers and captures the entire genome sequence, including the 5′ and 3′ ends. Platinum nanoparticles stabilized by imidazolium‐based phosphine‐decorated Polymer Immobilized Ionic Liquids (PPh2‐PIIL) catalyze the hydrolytic evolution of hydrogen from sodium borohydride with remarkable efficiency, under mild conditions. The composition of the polymer influences efficiency with the catalyst based on a polyethylene glycol modified imidazolium monomer (PtNP@PPh2‐PEGPIILS) more active than its N‐alkylated counterpart (PtNP@PPh2‐N‐decylPIILS). The maximum initial TOF of 169 moleH2.molcat−1.min−1 obtained at 30 °C with a catalyst loading of 0.08 mol% is among the highest to be reported for the aqueous phase hydrolysis of sodium borohydride catalyzed by a PtNP‐based system. Kinetic studies revealed that the apparent activation energy (Ea) of 23.9 kJ mol−1 for the hydrolysis of NaBH4 catalyzed by PtNP@PPh2‐PEGPIILS is significantly lower than that of 35.6 kJ mol−1 for PtNP@PPh2‐N‐decylPIILS. Primary kinetic isotope effects kH/kD of 1.8 and 2.1 obtained with PtNP@PPh2‐PEGPIILS and PtNP@PPh2‐N‐decylPIILS, respectively, for the hydrolysis with D2O support a mechanism involving rate determining oxidative addition or σ‐bond metathesis of the O−H bond. Catalyst stability and reuse studies showed that PtNP@PPh2‐PEGPIILS retained 70 % of its activity across five runs; the gradual drop in conversion appears to be due to poisoning of the catalyst by the accumulated metaborate product as well as the increased viscosity of the reaction mixture. Background: Convalescent plasma has been used to treat many viral diseases including Ebola. The manufacture of a purified anti-Ebola virus (EBOV) intravenous immunoglobulin (IVIG) from pooled convalescent plasma is described in this paper.Methods: An enzyme-linked immunosorbent assay (ELISA) targeting an EBOV surface glycoprotein antigen was used to determine the immunoglobulin titer of pooled plasma and purified anti-EBOV IVIG. Anti-EBOV IVIG was also tested in neutralization assays using a vesicular stomatitis virus pseudovirion expressing EBOV glycoprotein on its surface and with live EBOV. Finally, the efficacy of the anti-EBOV IVIG was assessed in a mouse model of EBOV infection.Results: In the ELISA, the anti-EBOV IVIG was shown to have a 7-fold increase in immunoglobulin G (IgG) titer over pooled convalescent plasma. In both the pseudovirion and live virus assays, the anti-EBOV IVIG showed approximately 5- to 6-fold increased potency over pooled plasma. Anti-EBOV IVIG also significantly improved survivability in mice infected with the virus when administered concurrently or 2 days after infection.Conclusions: These data support this purified anti-EBOV IVIG merits additional investigation and clinical trials for treatment and postexposure prophylaxis of Ebola virus disease. Abstract: This paper describes the practical combustion issues encountered with biomass co-firing on a large scale trial in a 500MW down fired utility boiler at Aberthaw power station. It also investigates and discusses the effect of biomass particle size and physical properties on devolatilisation; flame stability; and slagging by using the biomass energy crop miscanthus. During large scale biomass co-firing, the air flow around the injectors plays an important part in the fuel combustion and stable boiler operation. Secondary air flow at the point of biomass injection was adjusted during the experiments to provide higher airflows around the biomass injectors, hence the mixing, and air available for biomass combustion, was improved producing less unburnt material in the ash and reducing potential filter blockages. However, higher biomass air flows disrupt the temperature distribution in the boiler causing a wider variation of superheater temperatures compared to the baseline condition firing coal only. This reduces the combustion efficiency and can lead to localised hot spots, in the superheaters, with the potential to cause equipment damage. The trials indicated during the experiments that a secondary biomass air flow of 50% of its maximum, gave an optimum balance between air/biomass mixing and variations in the superheater temperatures. Abstract: Lean premixed swirl combustion is widely used in gas turbines and many other combustion Processes due to the benefits of good flame stability and blow off limits coupled with low NO x emissions. Although flashback is not generally a problem with natural gas combustion, there are some reports of flashback damage with existing gas turbines, whilst hydrogen enriched fuel blends, especially those derived from gasification of coal and/or biomass/industrial processes such as steel making, cause concerns in this area. Thus, this paper describes a practical experimental approach to study and reduce the effect of flashback in a compact design of generic swirl burner representative of many systems. A range of different fuel blends are investigated for flashback and blow off limits; these fuel mixes include methane, methane/hydrogen blends, pure hydrogen and coke oven gas. Swirl number effects are investigated by varying the number of inlets or the configuration of the inlets. The well known Lewis and von Elbe critical boundary velocity gradient expression is used to characterise flashback and enable comparison to be made with other available data. Two flashback phenomena are encountered here. The first one at lower swirl numbers involves flashback through the outer wall boundary layer where the crucial parameter is the critical boundary velocity gradient, Gf . Values of Gf are of similar magnitude to those reported by Lewis and von Elbe for laminar flow conditions, and it is recognised that under the turbulent flow conditions pertaining here actual gradients in the thin swirl flow boundary layer are much higher than occur under laminar flow conditions. At higher swirl numbers the central recirculation zone (CRZ) becomes enlarged and extends backwards over the fuel injector to the burner baseplate and causes flashback to occur earlier at higher velocities. This extension of the CRZ is complex, being governed by swirl number, equivalence ratio and Reynolds Number. Under these conditions flashback occurs when the cylindrical flame front surrounding the CRZ rapidly accelerates outwards to the tangential inlets and beyond, especially with hydrogen containing fuel mixes. Conversely at lower swirl numbers with a modified exhaust geometry, hence restricted CRZ, flashback occurs through the outer thin boundary layer at much lower flow rates when the hydrogen content of the fuel mix does not exceed 30%. The work demonstrates that it is possible to run premixed swirl burners with a wide range of hydrogen fuel blends so as to substantially minimise flashback behaviour, thus permitting wider used of the technology to reduce NOx emissions. Plant resources are used, managed and conserved by local communities in many parts of the world. However, very few studies have examined the site-specific factors and mechanisms that affect resource extraction. We apply methodology from the social and biological sciences to examine the cultural and socio-economic factors that influence the harvest practice and resource use of indigenous wood carvers in the Maningrida region of central Arnhem Land. Woodcarvers from this region use a small number of carving timbers with two species dominant, Bombax ceiba and Brachychiton diversifolius. There were many cultural differences in harvest practice, with artists from the Kuninjku/Kunibeidji language community harvesting a greater number of tree species, larger quantities per harvest trip and smaller sized stems. Socio-economic factors also played an important role in facilitating the collection of stems as artists owning a vehicle acquired more stems than those who did not. Harvest sites closest to the township of Maningrida had higher visitation frequencies than those further away. These influences on harvest practice have significant implications for the ecological sustainability of timber harvesting in this region and we highlight the need to examine such localised factors when assessing the sustainability of indigenous wildlife harvests.
