Abstract

The African School of Fundamental Physics and Applications, also known as the African School of Physics (ASP), was initiated in 2010, as a three-week biennial event, to offer additional training in fundamental and applied physics to African students with a minimum of three-year university education. Since its inception, ASP has grown to be much more than a school. ASP has become a series of activities and events to support academic development of African students, teachers and faculties. We report on the eighth African School of Physics, ASP2024, organized in Morocco, on April 15--19 and July 7--21, 2024. 13. Working within an Aesthetic of Relationality: Theoretical Considerations of Embodiment, Imagination and Foolishness as Part of Theatre Making about Dementia

Abstract

Through this paper, I consider how we might be aesthetically and relationally accountable in creating research-informed theatre through the conceptual frame of an aesthetic of relationality, and in-depth considerations of the theoretical notions embodiment, imagination and foolishness (as vulnerability-bravery). Through the example of "Cracked: new light on dementia," of which I am the playwright and director, I consider the ways artist-researchers foolishly and imaginatively engage in exploration from the starting point their own bodies, as socially, culturally and historically situated. I additionally extend these considerations towards a vision for how we might engage with persons with dementia beyond the tragic.

14. Working at the Margins: Theatre, Social Science and Radical Political Engagement

Abstract

As artist and social researchers working in health research, we work 'against the grain' in marginalised spaces to resist the marketisation of knowledge and related neoliberal practices. Our cross-disciplinary alliance, between theatre / performance studies and critical health science, is premised on engaging the humanities, arts, and social sciences to resist the subversion of both critical qualitative research and arts-based inquiries, and to interrogate structural inequalities that foster human injustices. Here we offer reflections on tensions we experienced with our interdisciplinary collaborations, how these tensions exposed limitations of our own disciplines/fields, and also enhanced the fruitfulness of our partnership.

Abstract

Viral entry targets with therapeutic neutralizing potential are subject to multiple escape mechanisms, including antigenic drift, immune dominance of functionally irrelevant epitopes, and subtle variations in host cell mechanisms. A surprising finding of recent years is that potent neutralizing antibodies to viral epitopes independent of strain exist, but are poorly represented across the diverse human population. Identifying these antibodies and understanding the biology mediating the specific immune response is thus difficult. An effective strategy for meeting this challenge is to incorporate multiplexed antigen screening into a high throughput survey of the memory B cell repertoire from immune individuals. We used this approach to discover suites of cross-clade antibodies directed to conformational epitopes in the stalk region of the influenza A hemagglutinin (HA) protein and to select high-affinity anti-peptide antibodies to the glycoprotein B (gB) of human cytomegalovirus. In each case, our screens revealed a restricted VH and VL germline usage, including published and previously unidentified gene families. The in vivo evolution of paratope specificity with optimal neutralizing activity was understandable after correlating biological activities with kinetic binding and epitope recognition. Iterative feedback between antigen probe design based on structure and function information with high throughput multiplexed screening demonstrated a generally applicable strategy for efficient identification of safe, native, finely tuned antibodies with the potential for high genetic barriers to viral escape.

Abstract

Background and Objectives Relational caring has the capacity to reduce stigma associated with dementia by shifting the focus from dysfunction and behavior management, to attending to the interdependencies and reciprocities that underpin caring relationships, and making explicit the centrality of relationships to quality care, growth, and quality of life. Education, particularly arts-based approaches, has been identified as a key strategy to decrease stigma. Yet rarely are the arts utilized in educational initiatives, and particularly so in community care settings. With an interest in redressing this, our team evaluated the impact of a Canadian filmed research-based drama— Cracked: new light on dementia —about stigma associated with people living with dementia and their families. Research Design and Methods We conducted interviews with family carers of people living with dementia and formal care providers affiliated with community-based dementia care, and also the general public at 3 and 8 months postscreening. Results Our analysis of participants' perceptions/experiences illustrates the effectiveness of Cracked in reducing stigma by demonstrating changes in the understanding of dementia and changes in practice. Our analysis also includes attention to how the film, as a form of cultural production, deepened engagement and facilitated transformation. Discussion and Implications Our evaluation of Cracked demonstrates that it is an effective strategy for decreasing the stigma associated with dementia by promoting relational caring. 28. The Incentives of Leaders in International Organizations: Evidence from the UN General Assembly.

Abstract

Voters and selectorates tend to prioritize domestic policy, so when and why do national political leaders choose to spend their time at international organizations (IOs)? Heads of state only began to address international assemblies such as the UN in recent decades; their appearances are puzzling, considering that leaders rarely get direct political benefit from multilateral cooperation, whose gains tend to be diffuse and slow-moving. This paper presents a theory of the motivations behind leaders' appearances at IOs. We argue that because multilateralism tends to be incidental to survival in office, heads of state tend to use IOs for prestige, not policy. But prestige is a luxury: both democrats and autocrats tend to prioritize international cooperation only when they can afford to do so domestically. Autocrats and democrats alike face domestic constraints—namely, the end of a term or gridlock in democracies, or under-institutionalized regimes in autocracies—when they consider international appearance, although the presence of other leaders amplifies the prestige value of a visit to an IO. Novel data on speakers at the United Nations General Assembly support our argument on leader appearances. We show that for the majority of leaders, domestic constraints temper their ability to appear at the UN, despite the draw of other leaders in recent years. Leurs apparitions sont déconcertantes, étant donné que les dirigeants ne bénéficient que rarement sur le plan politique d'une coopération multilatérale. Cet article présente une théorie des motivations qui se cachent derrière les apparitions des dirigeants aux OI. Nous affirmons que puisque le multilatéralisme a tendance à ne pas avoir d'incidence sur un mandat, les chefs d'État utilisent les OI pour gagner en prestige, et non pour la politique. Néanmoins, la présence d'autres dirigeants décuple la valeur prestigieuse d'une visite dans une OI. Des données inédites sur les intervenants lors de l'Assemblée générale des Nations unies soutiennent notre argument quant aux apparitions de dirigeants. Abstract

Therapeutic modulation of TREM2-dependent microglial function might provide an additional strategy to slow the progression of Alzheimer's disease. Although studies in animal models suggest that TREM2 is protective against Alzheimer's pathology, its effect on tau pathology and its potential beneficial role in people with Alzheimer's disease is still unclear. Our aim was to study associations between the dynamics of soluble TREM2, as a biomarker of TREM2 signalling, and amyloid β (Aβ) deposition, tau-related pathology, neuroimaging markers, and cognitive decline, during the progression of autosomal dominant Alzheimer's disease.We did a longitudinal analysis of data from the Dominantly Inherited Alzheimer Network (DIAN) observational study, which includes families with a history of autosomal dominant Alzheimer's disease. Participants aged over 18 years who were enrolled in DIAN between Jan 1, 2009, and July 31, 2019, were categorised as either carriers of pathogenic variants in PSEN1, PSEN2, and APP genes (n=155) or non-carriers (n=93). We measured amounts of cleaved soluble TREM2 using a novel immunoassay in CSF samples obtained every 2 years from participants who were asymptomatic (Clinical Dementia Rating [CDR]=0) and annually for those who were symptomatic (CDR>0). CSF concentrations of Aβ40, Aβ42, total tau (t-tau), and tau phosphorylated on threonine 181 (p-tau) were measured by validated immunoassays. Predefined neuroimaging measurements were total cortical uptake of Pittsburgh compound B PET (PiB-PET), cortical thickness in the precuneus ascertained by MRI, and hippocampal volume determined by MRI. Cognition was measured using a validated cognitive composite (including DIAN word list test, logical memory delayed recall, digit symbol coding test [total score], and minimental status examination). We based our statistical analysis on univariate and bivariate linear mixed effects models.In carriers of pathogenic variants, a high amyloid burden at baseline, represented by low CSF Aβ42 (β=-4·28 × 10-2 [SE 0·013], p=0·0012), but not high cortical uptake in PiB-PET (β=-5·51 × 10-3 [0·011], p=0·63), was the only predictor of an augmented annual rate of subsequent increase in soluble TREM2. Augmented annual rates of increase in soluble TREM2 were associated with a diminished rate of decrease in amyloid deposition, as measured by Aβ42 in CSF (r=0·56 [0·22], p=0·011), in presymptomatic carriers of pathogenic variants, and with diminished annual rate of increase in PiB-PET (r=-0·67 [0·25], p=0·0060) in symptomatic carriers of pathogenic variants. Presymptomatic carriers of pathogenic variants with annual rates of increase in soluble TREM2 lower than the median showed a correlation between enhanced annual rates of increase in p-tau in CSF and augmented annual rates of increase in PiB-PET signal (r=0·45 [0·21], p=0·035), that was not observed in those with rates of increase in soluble TREM2 higher than the median. Furthermore, presymptomatic carriers of pathogenic variants with rates of increase in soluble TREM2 above or below the median had opposite associations between Aβ42 in CSF and PiB-PET uptake when assessed longitudinally. Augmented annual rates of increase in soluble TREM2 in presymptomatic carriers of pathogenic variants correlated with decreased cortical shrinkage in the precuneus (r=0·46 [0·22]), p=0·040) and diminished cognitive decline (r=0·67 [0·22], p=0·0020).Our findings in autosomal dominant Alzheimer's disease position the TREM2 response within the amyloid cascade immediately after the first pathological changes in Aβ aggregation and further support the role of TREM2 on Aβ plaque deposition and compaction. Furthermore, these findings underpin a beneficial effect of TREM2 on Aβ deposition, Aβ-dependent tau pathology, cortical shrinkage, and cognitive decline. Soluble TREM2 could, therefore, be a key marker for clinical trial design and interpretation. Abstract

Introduction: One goal of the Longitudinal Early Onset Alzheimer's Disease Study (LEADS) is to define the fluid biomarker characteristics of early‐onset Alzheimer's disease (EOAD). Methods: Cerebrospinal fluid (CSF) concentrations of Aβ1‐40, Aβ1‐42, total tau (tTau), pTau181, VILIP‐1, SNAP‐25, neurogranin (Ng), neurofilament light chain (NfL), and YKL‐40 were measured by immunoassay in 165 LEADS participants. The associations of biomarker concentrations with diagnostic group and standard cognitive tests were evaluated. Results: Biomarkers were correlated with one another. Levels of CSF Aβ42/40, pTau181, tTau, SNAP‐25, and Ng in EOAD differed significantly from cognitively normal and early‐onset non‐AD dementia; NfL, YKL‐40, and VILIP‐1 did not. Across groups, all biomarkers except SNAP‐25 were correlated with cognition. Within the EOAD group, Aβ42/40, NfL, Ng, and SNAP‐25 were correlated with at least one cognitive measure. Discussion: This study provides a comprehensive analysis of CSF biomarkers in sporadic EOAD that can inform EOAD clinical trial design. [ABSTRACT FROM AUTHOR]

41. After the Crash: Research-Based Theater for Knowledge Transfer

Abstract

Introduction: The aim of this project was to develop and evaluate a research-based dramatic production for the purpose of transferring knowledge about traumatic brain injury (TBI) to health care professionals, managers, and decision makers. Methods: Using results drawn from six focus group discussions with key stakeholders (consumers, informal caregivers, and health care practitioners experienced in the field of TBI) and relevant scientific literature, a 50-minute play was produced for the purpose of conveying the experiences of TBI survivors, informal care providers, and health practitioners and best practice for TBI care. A self-administered postperformance survey was distributed to audience members at the end of four performances in Ontario, Canada, to evaluate the play's efficacy. Two hundred ninety-one questionnaires were completed. The questionnaire had five questions scored on a 5-item Likert scale with space for open-ended comments. Results: Consistently high mean scores from the questionnaires indicate that theater is a highly efficacious and engaging method of knowledge transfer, particularly for complex material that deals with human emotion and interpersonal relationships. Discussion: Responses supported the effectiveness of drama as a knowledge translation strategy and identified its potential to impact practice positively. (Contains 1 table.)

Abstract

"Brain-predicted age" quantifies apparent brain age compared to normative neuroimaging trajectories. Advanced brain-predicted age has been well established in symptomatic Alzheimer disease (AD), but is underexplored in preclinical AD. Prior brain-predicted age studies have typically used structural MRI, but resting-state functional connectivity (FC) remains underexplored. Our model predicted age from FC in 391 cognitively normal, amyloid-negative controls (ages 18-89). We applied the trained model to 145 amyloid-negative, 151 preclinical AD, and 156 symptomatic AD participants to test group differences. The model accurately predicted age in the training set. FC-predicted brain age gaps (FC-BAG) were significantly older in symptomatic AD and significantly younger in preclinical AD compared to controls. There was minimal correspondence between networks predictive of age and AD. Elevated FC-BAG may reflect network disruption during symptomatic AD. Reduced FC-BAG in preclinical AD was opposite to the expected direction, and may reflect a biphasic response to preclinical AD pathology or may be driven by inconsistency between age-related vs. AD-related networks. Overall, FC-predicted brain age may be a sensitive AD biomarker.
