This research explores feasibility of adsorption method used for regulating polypenols content in wort with a view of improving beer quality. It examines adsorption of polyphenols (quercetin, gallic acid, rutin) from pure substance solutions, their mixtures and beer wort treated by sorbents that differ by origin, making, structure and surface chemical composition. The work determines patterns and specific features of polyphenols adsorption with activated carbons. To describe adsorption mechanism more precisely, we specified structure, surface chemical condition, and calculated adsorption parameters using equations of Langmuir, Freundlich and Dubinin-Radushkevich, and the multilayer adsorption theory (BET model). It is demonstrated that polyphenols adsorption mechanism depends on carbon characteristics and is of physical nature that reveals itself in dispersion interaction in micropores and in specific one with oxygen-containing functional group (OFG) on carbon surface. Polyphenol competitive adsorption in mixture and wort recognized. At polyphenols adsorption from model solutions and wort, carbon sorbents are identified to share sufficiently close sorption characteristics. We performed comparative evaluation of quality characteristics of beer produced from activated carbons treated and untreated worts. It is shown that beer samples produced from unhopped wort filtered through semi-coke adsorption, meet regulatory standards requirements of safety by organoleptic, physical and chemical indicators. Moreover, beer obtained from semi-coke treated wort exceeds control sample in terms of organoleptic and stability ensuring indicators.

One of the ways to improve colloidal stability and organoleptic properties of beer is to control polyphenol constituents in unhopped beer applying adsorption method with carbon sorbents, sorption progressing dynamically. The adsorption of polyphenol constituents from unhopped wort under dynamic conditions using carbonic sorbents has been studied in order to prove the possibility of using cheap carbon ABG and «Purolat Standard». According to the obtained data, carbon sorbent «Purolat Standard» can be recommended for practical realization and designing the process of adsorption under dynamic conditions. The possibility of designing the process of content control of unhopped wort using carbon sorbent «Purolat Standard» has been proved experimentally. The calculation method for filter parameters and the modes of adsorption of polyphenol constituents from unhopped wort has been suggested. The method is based on the outward diffusion equation of adsorption dynamics for isothermal line using the Dubinin-Radushkevich adsorption constants and the kinetic analysis data. The efficient filter parameters and the modes of adsorption of polyphenol constituents from unhopped wort, which lead to the improvement of colloidal stability and organoleptic properties of beer, have been calculated. The method of waste sorbent «Purolat Standard» recovery after adsorption of polyphenol constituents from unhopped wort has been suggested. The dependence of filtration rate on initial content of polyphenol compounds in wort has been found out. The required equipment and the operational modes of the apparatus providing the improvement in beer quality by the control of polyphenol constituents in wort have been recommended.

Pilonidal sinuses are a well-described surgical condition first recognised in the 19th century. They have most commonly been reported in the intergluteal cleft; involvement of other sites is limited to isolated case reports. Association of this condition with tuberculosis is rare and how the two conditions affect each other's causation is poorly understood. We report a rare case showing tubercular pathology in a chronic pilonidal sinus on the forehead of a young lady. A careful review of the lesion was required before labelling it as non-tubercular. The histopathological similarity between a foreign body reaction and tuberculosis in such lesions may confuse the clinician, and therefore differentiation is important to prevent morbidity associated with anti-tubercular therapy. The adsorption of gallic acid on active carbons of different ranks with different pore structure and chemical state of the surface is investigated. The regularities and features of the adsorption process are established. It is revealed that the adsorption isotherm of gallic acid from the aqueous solution with activated carbon of AG-OB-1 rank refers to the L-type isotherms by Giles's classification, and AC adsorption isotherm of ABG and Purolat-Standard ranks - to the S-type isotherms. L-type isotherm gives evidence concerning the flow of physical adsorption. With S-type isotherm is the adsorption described, at which the strength of the interaction between the solute and the adsorbent is less than the force of interaction between the adsorbed molecules, which can be explained by the formation of hydrogen bonds. It was found that the maximum adsorption of gallic acid with carbon sorbents changes in the following sequence - "Purolat-standard" > ABG > AG-OV-1. The mechanism of adsorption of gallic acid on active carbons is offered. Proceeding from the structure, the chemical state and the main adsorption parameters of active carbon, we can assume that the adsorption of gallic acid has physical nature. It can take place in pores, both due to dispersion interaction (of van der Waals forces) and due to the interaction between the active carbon surface functional groups, containing oxygen with carboxyl and hydroxyl groups of gallic acid. The values of adsorptive capacity testify to the dependence of the adsorption efficiency on the structure and physical-chemical properties of the sorbent. It has been determined that by the absorption of gallic acid specific interaction makes greater contribution. It is shown that the results obtained can be applied to create adsorption technologies on removing gallic acid from the sewage containing individual components and from the mixture with other polyphenols, including beer wort.

At present, one of the most important problems is the growing of environment pollution of Urban Development. Wastewater treatment generated in the manufacture of medicines and biologically active additives is of great importance for water bodies' sanitary protection. The sorption method provides a high degree of weakly concentrated wastewater purification, the absence of secondary pollution and the possibility of returning valuable components and water to the production. The purpose of the study was to develop an adsorption technology for wastewater treatment in the pharmaceutical industry containing flavonoids using semi-coke. Results of the study: The results of complex studies of the flavonoids - quercetin adsorption and rutin on active charcoals differing in raw materials, method of production, structure and chemistry of the surface are presented. The main equilibrium, kinetic and dynamic process parameters are calculated for engineering calculations. Importance of the research: The technology of wastewater adsorption treatment from pharmaceutical industries with subsequent regeneration of active coals is proposed, which will provide protection against water pollution and resource saving. The performed work showed the prospects of using the semi-coke "Puroolat-Standard", which is characterized by higher adsorption capacity and lower cost

The direct relationship between facilitative glucose transporters (GLUTs) and metabolic diseases opens new avenues for sensing metabolic deregulations and drives the development of molecular probes for GLUT-targeted detection of metabolic diseases. Radiotracer-based molecular imaging probes have been effectively utilized in reporting alterations in sugar uptake as an indication of metabolic deregulations, cancer development, or inflammation. Progress in developing fluorophore-based tools facilitated GLUT-specific analyses using more accessible fluorescence-based instrumentation. However, restrictions on the emission range of fluorophores and the requirement for substantial post-treatments to reduce background fluorescence have brought to light the critical directions for improvement of the technology for broader use in screening applications. Here we present turn-on GLUT activity reporters activated upon cells' internalization. We demonstrate a specific delivery of a sizable rhodamine B fluorophore through GLUT5 and showcase a stringent requirement in conjugate structure for maintaining a GLUT-specific uptake. With the turn-on GLUT probes, we demonstrate the feasibility of high-throughput fluorescence microscopy and flow cytometry-based GLUT activity screening in live cells and the probes' applicability for assessing sugar uptake alterations in vivo ., Competing Interests: The authors declare no competing financial interest.

Facilitative carbohydrate transporters (GLUTs, SLC2 gene family) are transmembrane proteins transporting hexoses and other sugars based on cellular metabolic demands. While a direct link between GLUTs and metabolic disorders has framed them as important biological and medicinal targets, targeting disease-relevant GLUTs remains challenging. In this study, we aimed to identify substrate-GLUT interactions that would discriminate between major fructose transporters. We examined the uptake distribution for conformational and configurational isomers of fructose using the corresponding conformationally locked fluorescently labeled mimetics as probes for assessing GLUT preferences in real time. Through comparative analysis of the uptake of the probes in the yeast-based single GLUT expression systems and the multi-GLUT mammalian cell environment, we established the ability of fructose transporters to discriminate between fructose conformers and epimers. We demonstrated that recreating the conformational and configurational mixture of fructose with molecular probes allows for the specific probe distribution, with fructofuranose mimetic being taken up preferentially through GLUT5 and β-d-fructopyranose mimetic passing through GLUT2. The uptake of α-d-fructopyranose mimetic was found to be independent of GLUT5 or GLUT2. The results of this study provide a new approach to analyzing GLUT5 and GLUT2 activity in live cells, and the findings can be used as a proof-of-concept for multi-GLUT activity screening in live cells. The research also provides new knowledge on substrate-GLUT interactions and new tools for monitoring alterations in GLUT activities.

Background and Objective: Traumatic brain injury (TBI) is a major health issue in developing nations such as India along with underreporting of TBI data because of a lack of major studies targeting the population with TBI. Various lacunae from field to tertiary centers are responsible for this significant burden of TBIs. We studied the epidemiologic profile of 1150 patients with TBI including the predictors of the outcome of TBIs in a tertiary health care institution in North India to identify the modifiable factors that could be used to improve the outcome and reduce the TBI burden., Methods: Patients presenting with TBI to a level I trauma center were identified and enrolled in the study. Data regarding patient information from accident to discharge or death were collected as per designed format and analyzed to determine outcome predictors., Results: Mean age was 36 ± 15.8 years and 84.6% of patients were male; road traffic accident was the mode of injury in 64.26% of cases. Lack of adequate prehospital care was seen in our study. Glasgow Coma Scale score on admission, Injury Severity Score, and Rotterdam CT score were found to be the 3 statistically significant predictors of outcome in patients with TBI., Conclusions: Knowledge about the causes, pattern, and distribution of patients with TBI from this study is helpful in policy making, research, health management, and rehabilitation at the national level in ours and in other nations., (Copyright © 2019 Elsevier Inc. Aim: Development of enzyme immunoassay detection of B and D factors of complement alternative pathway functional activity for solving diagnostic and prognostic problems of patient therapy. Study activity of these factors in blood sera of children with atopic dermatitis before and after therapy for elucidation of the role of complement alternative pathway in pathogenesis of this disease., Materials and Methods: Children aged 6 months to 18 years with atopic dermatitis were examined for functional activity of B and D factors in blood sera before and after therapy by the developed methods., Results: The developed enzyme immunoassay methods for determination of functional activity of B and D complement alternative pathway showed high sensitivity and reliability. In children with atopic dermatitis factor B and D activity was significantly lower than normal before treatment. After treatment these activity increased significantly (p < 0.004) and in the case of D factor--up to normal., Conclusion: The data obtained in the study indicates the presence of complement alternative pathway activation in atopic dermatitis in children and the possibility of use of factor B and D functional activity analysis for diagnostic and prognostic purposes.

Background/aim: In developing countries, enteric perforation due to typhoid continues to have very high morbidity and mortality rates irrespective of the type of operative procedure performed. The aim of the present study was to evaluate the effectiveness of a free omental sheet graft in perforated typhoid enteritis, in comparison to other methods of enteric perforation repair in terms of decreased morbidity, mortality and cost-effectiveness., Method: A non-randomized study of 114 patients with enteric perforations was carried out over a period of 4.5 years. The study was divided in two groups; group I includes 38 cases in which a free omental sheet graft was used in typhoid enteritis with perforation and group II includes 76 cases in which other surgical procedures for enteric perforation repair were used. The outcomes were measured in relation to postoperative complications and mortality., Results: The incidence of complications including faecal fistula was 2.6% in group I (free omental sheet graft group) versus 32.89% in group II. The mortality rate of 2.6% was also lower in group I versus 19.7% in group II., Conclusion: This study concludes that the use of free omental sheet graft on typhoid enteric perforation site is effective in lowering the repair leak rate and thereby decreases the morbidity and mortality associated with these procedures. By lowering the complication rates, it also entails reduction in financial burden.

Aim: Frequency of occurrence detection of C4A and C4B complement system deficiency in patients with chronic gastrointestinal tract (GIT) diseases including gastric ulcer (GU) and duodenal ulcer (DU)., Materials and Methods: 74 patients with chronic GIT diseases were examined. Endoscopy with stomach mucosa condition evaluation based on histobacterioscopic examination of gastroduodenal biopsy samples was used. Intestine microbiocenosis evaluation was performed by using microflora degree of manifestation according to Federal Standard of Russian Ministry of Health No 231 -91500.11.0004-2003. C4A and C4B isotypes in blood sera of patients were measured by using enzyme immunoassay., Results: Chronic gastroduodenitis was diagnosed in 35.1%, pangastritis B--in 41.9%, GU and DU--in 23% of patients. Histological evaluation of biopsy samples revealed marked inflammatory changes in stomach and duodenum mucosa in 77% of patients. Stomach mucosa infection rate by Helicobacter pylori reached 85%. Microbiological disorders manifestation in microflora of patients matched endoscopic and histobacteriscopic changes in it and was the highest for GU and DU. In 76.0% of cases C4A and C4B isotype deficiency in blood sera matches the development of erosive-ulcerous process in stomach and duodenum mucosa with marked background dysbiotic GIT microbiota disorders., Conclusion: Patients with functional deficiency of C4A and C4B isotypes have a genetic burden to susceptibility to chronic GIT diseases whereas H.pylori infection deteriorate the disease.

Aim: Development of new method of C4B isotype functional activity evaluation in enzyme immunoassay by using pharmaceutical preparation derinat as a classical pathway complement activator and its use for blood sera isotyping in confirmed urogenital tract chlamydia infection., Materials and Methods: Enzyme immunoassay was used to detect C4A and C4B isotype functional deficiency in blood sera of patients. Chlamydia etiology urogenital infection diagnosis was based on results of standard clinical-instrumental examination methods: vaginal clinical smear analysis, scrape sample light microscopy with consequent treatment by fluorescent monoclonal antibodies against Chlamydia trachomatis and PCR., Results: In acute form of the disease C4A deficiency frequency of occurrence was 0.36, and C4B deficiency - 0.55. In chronic form of the disease deficiency frequency of occurrence was 0.38 for both isotypes. In the group of healthy people isotype deficiency was 0.08 and 0.25, respectively., Conclusion: Innate masked C4 deficiency interfere with the normal immune defense of organism against chlamydia infection, and antigen carbohydrate pathogenicity may possibly be more significant for the development of immune response to which C4B isotype activity is necessary.

Representatives of 200 urban and 200 rural households were interviewed to determine the extent and appropriateness of the use of chloroquine obtained over-the-counter for malaria prophylaxis. Malaria prophylaxis was taken by 38 urban and 65 rural respondents. Chloroquine was the only drug used and was found in 7 urban and 40 rural households. The presence of chloroquine was detected in 4/130 urban and 11/136 rural urine samples tested. Knowledge about chloroquine was unsatisfactory with 6 urban and 10 rural respondents aware that chloroquine could have harmful adverse effects. The doses of chloroquine taken for prophylaxis varied widely, and of the 103 respondents who took prophylaxis only 16% (urban) and 31% (rural) were taking the correct dose. Self-medication accounts for a substantial, little acknowledged, portion of chloroquine use. To limit appropriate use of a powerful, potentially dangerous antimalarial drug and perhaps extend the useful therapeutic life of chloroquine clear recommendations and population education are needed.

1. A questionnaire examining the use of, and knowledge about over-the-counter (OTC) drugs was administered verbally to representatives from 200 urban and 200 rural households. Self-medication was common in both groups but there were more drugs present in rural (336 items) than urban homes (231 items) with the differences largely due to the more frequent presence of traditional medicines in rural homes. 2. Analgesics were the commonest items in the homes of both groups. A positive urine screening test for aspirin, chloroquine, or antibiotics was found in 25% of urban and 27% of rural samples tested. Respondents who elected to use an OTC drug for a particular symptom usually chose an appropriate drug. 3. Rural respondents were more likely to use traditional medicines particularly for the treatment of cough, abdominal pain, and diarrhoea. 4. Knowledge about dosages, the possibility of adverse reactions, and the seriousness of potential poisoning with OTC drugs was inadequate and needs to be corrected for more efficient self-medication.
