Lycium barbarum L. (goji), as an economic crop, has a high added value. However, the tender and fragile fruits are easily damaged during harvesting and transportation, leading to fruit bruising, which can cause rotting or black–brown spots after drying, seriously affecting the quality and price. In this study, two varieties of goji were used to determine and evaluate fruit bruising using a pendulum impact test, and the impact process was recorded using a high-speed camera and impact force sensor. This study discussed the energy changes during the impact process of fruits and conducted a correlation analysis of the impact energy, absorbed energy, restitution coefficient, impact force, and other indicators, analyzing the changes in each indicator with the falling height. The results showed that 0.2 m could be considered a critical height for damaging the fruit of goji. Furthermore, this study calculated the bruise susceptibility of the different varieties at different heights, which can be used for predicting bruising during the harvesting and collection of goji berries and ultimately for estimating the damage caused by mechanical harvesting. The leaf is a critical organ for assimilation in plants, and the anatomical structure of the leaf can reflect the adaptability of plants to their environment to a certain extent. The research objects in this investigation were 36 different cultivars (lines) of goji germplasm resources. The structure and stoma condition of healthy leaves were the subject of a comparative investigation. Significant variations in leaf structural indices and stoma were identified among various goji cultivars (lines). The leaves of goji were the thickest and possessed the lowest stoma density. The resilience of goji germplasm resources was assessed using the main component analysis approach and the membership function. The results showed that LR (L. ruthenicum Murr.) and LCy (L. cylindricum Kuang) exhibited the strongest resistance. Goji resistance is significantly influenced by thickness of the upper cuticle (TUC) and thickness of the lower cuticle (TLC), according to the analytical results for several leaf anatomical indicators and stress resistance. By analyzing the wax content of 13 different goji cultivars (lines) and field infection results, it was found that Tianjing 3 had the highest wax content, the smallest average number of galls, and the smallest infection area. This suggests that Tianjing 3 has the strongest resistance to Aceria pallida, and that its insect resistance is correlated with its wax content. This study offers a thorough assessment of 36 goji germplasm resources' resilience, providing a point of reference for the selection and propagation of resistant cultivars within these resources. Background: It is known that 60-80% of the population affected by various diseases, such as heart attack, stroke, and oncological diseases is associated with selenium (Se) deficiency. Therefore, enriching agricultural crops with Se is considered a current issue. Objective: This study investigates Se enrichment of Lycium barbarum (Lb) and its effectiveness in hydroponic and soil conditions in the Ararat Valley. The research focuses on the impact of adding exogenous Se on plant productivity, Se accumulation, and the content of biologically active substances (BAS) in Lb berries and leaves. Methods: The experiments were conducted from 2021 to 2023 using hydroponic and soil cultivation methods with varying Se concentrations in nutrient solutions and foliar nutrition. Results: The results demonstrate that Se enrichment significantly increased Se and BAS levels in both leaves and berries of Lb, with hydroponic cultivation exhibiting higher accumulation compared to soil conditions. Foliar feeding with a 0.005% Se solution and exogenous Se addition to nutrient solutions at 1.0 mg/L resulted in notable improvements in Se content and plant productivity. Furthermore, the analyses revealed significant differences between hydroponically and soil-grown Lb, emphasizing the potential of hydroponic cultivation for producing Seenriched plant material with enhanced BAS content. Conclusion: Hydroponic cultivation supplemented with Se and foliar nutrition is recommended for obtaining Se-rich Lb plant material, which is suitable for medicinal and functional food purposes. Abstract With the increasing trend of global aging, sarcopenia has become a significant public health issue. Goji berry, also known as “Gou qi zi” in China, is a traditional Chinese herb that can enhance the structure and function of muscles and bones. Otherwise, previous excellent publications illustrated that plant-derived exosome-like nanoparticles can exert good bioactive functions in different aging or disease models. Thus, we issued the hypothesis that Gouqi-derived nanovesicles (GqDNVs) may also have the ability to improve skeletal muscle health, though the effect and its mechanism need to be explored. Hence, we have extracted GqDNVs from fresh berries of Lycium barbarum L. (goji) and found that the contents of GqDNVs are rich in saccharides and lipids. Based on the pathway annotations and predictions in non-targeted metabolome analysis, GqDNVs are tightly associated with the pathways in metabolism. In muscle atrophy model mice, intramuscular injection of GqDNVs improves the cross-sectional area of the quadriceps muscle, grip strength and the AMPK/SIRT1/PGC1α pathway expression. After separately inhibiting AMPK or PGC1α in C2C12 cells with dexamethasone administration, we have found that the activated AMPK plays the chief role in improving cell proliferation induced by GqDNVs. Furthermore, the energy-targeted metabolome analysis in the quadriceps muscle demonstrates that the GqDNVs up-regulate the metabolism of amino sugar and nucleotide sugar, autophagy and oxidative phosphorylation process, which indicates the activation of muscle regeneration. Besides, the Spearman rank analysis shows close associations between the quality and function of skeletal muscle, metabolites and expression levels of AMPK and SIRT1. In this study, we provide a new founding that GqDNVs can improve the quality and function of skeletal muscle accompanying the activated AMPK/SIRT1/PGC1α signaling pathway. Therefore, GqDNVs have the effect of anti-aging skeletal muscle as a potential adjuvant or complementary method or idea in future therapy and research. Graphical Abstract

Abstract Background Goji (Lycium barbarum) is an important economic crop that is widely cultivated in Northwest China and is known as superfood. Goji plants are often affected by powdery mildew (Arthrocladiella mougeotii) in the field, resulting in considerable reduction in production and serious economic losses. The long-term reliance on agricultural chemicals to control powdery mildew not only leads to environmental pollution, but also causes excessive residues to accumulate in fruits, endangering human health. The plant epicuticular wax is the first physical barrier between land plants and the atmospheric environment, and plays an important role in the interactions of plants with pathogenic microorganisms. However, the relationship between epicuticular wax and powdery mildew resistance in goji plants is not well understood. Results The leaf epicuticular wax crystals of ‘Ningqi I’ goji were dense and small, and the wax load was 121.74 μg/cm2, which was 3.36 times greater than that of ‘Huangguo’ goji. The ratio of alcohol components in ‘Ningqi I’ goji was 121.18% greater than that in ‘Huangguo’ goji. The incidence rate and disease index of ‘Ningqi I’ goji inoculated with powdery mildew were significantly lower than those inoculated ‘Huangguo’ goji, showing high resistance and moderate susceptibility, respectively. After mechanical wax removal, the disease indices of the two goji varieties increased significantly, indicating extreme susceptibility. Seventeen differential expression genes showing significantly greater expression in ‘Ningqi’ goji leaves than in ‘Huangguo’ goji leaves were enriched in genes involved in fatty acid elongation and the wax biosynthesis pathway. The FAR activity and relative expression levels of LbaFAR and LbaWSD1 in ‘Ningqi I’ goji were significantly greater than those in ‘Huangguo’ goji. The relative expression levels of these genes significantly increased in the overexpression tobacco lines FAR-OE and WSD1-OE, and the wax load were augmented by 46.10% and 22.23%, respectively. Conclusions The increased expression of the LbaFAR and LbaWSD1 genes in ‘Ningqi I’ leaves led to increased FAR and WSD1 activity, increasing the wax load and alcohol content of the epicuticular of leaves, and improving leaf glossiness, resulting in strengthened resistance to powdery mildew. Therefore, the production of leaf epicuticular wax mediated by the LbaFAR and LbaWSD1 genes could be the main reason for the difference in powdery mildew resistance between the two goji varieties, which can provide a theoretical basis for the selection of resistant varieties to control goji powdery mildew. Abstract Metabolic syndrome (MetS) is a common disorder involving a cluster of metabolic abnormalities, such as abdominal obesity, hypertension, dyslipidemia, insulin resistance, and atherogenic profile. MetS is characterized by an increase in oxidative stress and a chronic proinflammatory state, which are directly related to the development and progression of this pathology. It has been seen how a healthy lifestyle and good dietary practices are key to improving the different metabolic parameters and, therefore, play a fundamental role in reducing the risk of developing diabetes. The present review focuses on the research evidence related to the therapeutic properties of Lycium barbarum L. in MetS gathered in the last years. Several preclinical studies suggest that L. barbarum extracts are a good dietary supplement for the prevention of cardiovascular diseases in people with MetS. This compound has been used for years in traditional Chinese medicine for the treatment of atrophic gastritis, problems related to the lungs, kidneys, and liver, and as a supplement for eye health. In addition, different in vitro and in vivo studies have been carried out that support the properties attributed to metabolites derived from L. barbarum, such as polysaccharides that have been shown diverse biological activities. In conclusion, L. barbarum extracts have multiple benefits to increase general well‐being and immune function. However, there are a limited number of studies related to effect of L. barbarum in MetS, but they demonstrated effectiveness in the treatment of obesity, diabetes mellitus type 2, and prevention of diabetes mellitus type 2 complication.

The waxy layer on the surface of the goji fruit increases the dehydration time. In this study, the novel use of ultrasound pretreatment (10, 20, and 30 min) assisted with osmotic dehydration was utilized to improve drying efficiency and product quality of goji. The results showed that goji treated with ultrasound‐assisted osmotic dehydration lost more water and gained more solids during osmotic dehydration than goji treated with osmotic dehydration only. Furthermore, the ultrasound‐assisted osmotic dehydration pretreatment method shortened the drying time for freezing drying goji and lowered the water activity of the dried goji. This may be due to the destruction of the wax layer on the surface of the goji by ultrasound. The rehydration rate of the goji treated with ultrasound for 20 min was significantly higher than that of the samples dried by osmosis dehydration only. The application of ultrasound‐assisted osmotic dehydration resulted in the best color and the highest glutathione and total free amino acid content of goji, especially the taurine. Besides the above, the antioxidant capacity of ultrasound‐assisted osmotic dehydration‐treated goji was enhanced, with the highest 1,1‐diphenyl‐2‐picrylhydrazyl (DPPH) radical scavenging capacity and ferric reducing antioxidant power (FRAP) values for the samples in the 20‐min ultrasound‐assisted group. The results demonstrate that ultrasound‐assisted osmotic dehydration not only reduces the drying time of goji but also enhances its product quality. Practical applications: This study explored the effects of different ultrasound‐assisted osmotic dehydration methods on the drying efficiency and quality of freeze‐dried goji. This study aimed to investigate the hepatoprotective effects of lyophilized powder of goji ferment (LPGF) against acetaminophen (APAP)-induced hepatic damage in Hep3B cells and in mice. Eleven strains of lactic acid bacteria (LAB) were selected and their hepatoprotection against APAP-induced cellular damage in Hep3B cell line was evaluated. Four strains of LAB, including BCRC11652 (Leuconostoc mesenteroides subsp. mesenteroides), BCRC14619 (Lactobacillus gasseri), KODA-1 (Pediococcus acidilactici), and KODA-2 (Limosilactobacillus fermentum), have hepatoprotective potential against APAP in vitro. Goji significantly stimulated the growth of individual and combined strains of LAB and the optimal fermented condition was the treatment of goji at 10% (w/w) for 24 h. The prepared lyophilized powder of goji ferment (LPGF) containing fifteen combinations of LAB strains was used to explore their hepatoprotection in vitro. LPGF containing all combinations of LAB strains, except for KODA-2, significantly restored APAP-reduced cell viability and improved APAP-increased cellular levels of alanine aminotransferase (ALT) and aspartate aminotransferase (AST). In mice model, LPGF containing BCRC11652, BCRC14619, and KODA-2 was chosen to evaluate its hepatoprotection against APAP-induced liver injury. LPGF at diverse doses have a tendency but no significant improvement on APAP-reduced body weight gain and liver weight. LPGF significantly decreased APAP-increased serum ALT and AST levels in a dose-dependent manner. At the end of experiment, LPGF significantly and dose-dependently reversed APAP-reduced activities of GSH and antioxidant enzymes, including glutathione peroxidase, superoxide dismutase, and catalase in hepatic tissue. In this paper, 46 fresh goji pulp products randomly selected from the market were compared and analyzed through the main components, color, flavor and other indicators, and the evaluation standard was preliminarily established. The results showed that the samples of fresh goji pulp showed good nutritional and flavor characteristics, but the quality of different products varied greatly. The pH ranged from 4.04~4.62, the viscosity ranged from 6.63~116.60 cP, the soluble solids ranged from 12.77%~30.63%, the total sugar content ranged from 93.19~220.26 mg/g, the flavonoid content ranged from 23.43~87.27 μg/g, and the total carotenoid content ranged from 3.23~1579.47 μg/g. The color of the samples was bright orange-red, and the samples responded to the eight flavor characteristics of umami, sour, astringency and saltness. Alkanes, alcohols, and volatile sulfides were the most varied compounds in the odor analysis. Based on principal component analysis, the correlation degree and contribution degree of each index and the main component were obtained, the quality evaluation index of fresh goji pulp was established, the grading standard of evaluation index was established by probability classification, and the evaluation standard of quality was constructed by analytic hierarchy process, and the comprehensive score of each sample of fresh goji pulp was obtained, which provided data reference for quality evaluation and standard formulation of fresh goji pulp.

This study compares the microbial diversity and content of bioactive compounds in dried goji berries available on the Polish market to those of the most highly valued goji berries from the Ningxia region in China. The content of phenols, flavonoids, and carotenoids were determined, as well as the antioxidant capacities of the fruits. The quantitative and qualitative composition of the microbiota inhabiting the fruits was assessed using metagenomics by high-throughput sequencing on the Illumina platform. The highest quality was demonstrated by naturally dried fruits from the Ningxia region. These berries were characterized by a high content of polyphenols and high antioxidant activity, as well as high microbial quality. The lowest antioxidant capacity was shown by goji berries cultivated in Poland. However, they contained a high amount of carotenoids. The highest microbial contamination was found in the goji berries available in Poland (>106 CFU/g), which is important in terms of consumer safety. Despite the widely accepted benefits of consuming goji berries, both the country of cultivation and the preservation method may influence their composition, bioactivity, and microbial quality. Carotenoids are key metabolites in goji (Lycium), a traditional Chinese medicine plant; however, the carotenoid content varies in fruits of different goji species, and the mechanism of this variation is not clear. Carotenoids participate in signal transduction and photosynthesis, and function as colorants and photoprotectors. Members of the carotenoid cleavage oxygenase (CCO) gene family are involved in the regulation of phytohormones, pigments, and aromatic substances, such as abscisic acid (ABA), β-carotenoid, and α-ionone, by degrading carotenoids. Some CCO genes are also related to an abiotic stress response. Here, a total of 12 LbCCO genes were identified and analyzed from the L. barbarum genome. CCO genes were divided into six subfamilies based on the constructed phylogenetic tree, including LbNCEDs, LbCCD1, LbCCD3, LbCCD4, LbCCD7, and LbCCD8. Among them, CCD3 was reported for the first time. The gene structure and motif analysis revealed the conservation of CCO subfamilies. Pseudogene generation and the importance of each subfamily in CCOs were revealed by collinearity analysis. The spatiotemporal transcriptomes of L. barbarum and L. ruthenicum were compared, suggesting that CCD4-1 may dominate carotenoid degradation in goji fruits. Cis-acting elements prediction and environment responsive gene expression analyses indicated that salt-alkali stress and photothermal conditions might influence the expression of CCOs in goji. This work aimed to boost the valorisation of Lycium intricatum Boiss. L. as a source of high added value bioproducts. For that purpose, leaves and root ethanol extracts and fractions (chloroform, ethyl acetate, n-butanol, and water) were prepared and evaluated for radical scavenging activity (RSA) on 2,2-diphenyl-1-picrylhydrazyl (DPPH) and 2,2′-azino-bis (3-ethylbenzothiazoline-6-sulfonic acid) (ABTS) radicals, ferric reducing antioxidant power (FRAP), and metal chelating potential against copper and iron ions. Extracts were also appraised for in vitro inhibition of enzymes implicated on the onset of neurological diseases (acetylcholinesterase: AChE and butyrylcholinesterase: BuChE), type-2 diabetes mellitus (T2DM, α-glucosidase), obesity/acne (lipase), and skin hyperpigmentation/food oxidation (tyrosinase). The total content of phenolics (TPC), flavonoids (TFC), and hydrolysable tannins (THTC) was evaluated by colorimetric methods, while the phenolic profile was determined by high-performance liquid chromatography, coupled to a diode-array ultraviolet detector (HPLC-UV-DAD). Extracts had significant RSA and FRAP, and moderate copper chelation, but no iron chelating capacity. Samples had a higher activity towards α-glucosidase and tyrosinase, especially those from roots, a low capacity to inhibit AChE, and no activity towards BuChE and lipase. The ethyl acetate fraction of roots had the highest TPC and THTC, whereas the ethyl acetate fraction of leaves had the highest flavonoid levels. Gallic, gentisic, ferulic, and trans-cinnamic acids were identified in both organs. In the current agro-economic context, for the rural development of Gorj County, it is important to identify new, attractive, but also economically efficient businesses. Starting from the fact that the interest in Goji fruits has increased significantly, along with their use in organic food and in traditional medicine, and the adaptation of this shrub to the pedoclimatic conditions in Gorj County, if the culture technology is respected, this study present the possibility of establishment and capitalization of a culture that is not specific to the area, but which, through its characteristics, can adapt to the pedoclimatic conditions in Gorj County. Following the economic analysis, it is demonstrated that this business has a positive impact on the sustainable development of the rural area of Gorj County, on the economy, on the social life and on the environment, by capitalizing of agricultural land, creating jobs, promoting the area and increasing the income of entrepreneurs. The present study emphasize the effect of R. crispus and L. barbarum 6% aqueous extract on blood sugar level in Alloxan induced diabetes in rats. The rats were divided in five groups: one non-diabetic control and four experimental groups with induced diabetes mellitus after 40 mg/kg b.w. intravenous administration of 2% Alloxan. One group was kept as diabetic control and in the other three groups was administered 6% aqueous extracts of R. crispus, L. barbarum or a combination of the extracts during seven weeks. The better results were obtained in case of L. barbatum (goji) extract administration followed by the R. crispus (yellow dock) extract. The combination of the two extracts has proven to have a weaker effect than the extracts given separately.

Goji berries, known for their health-promoting properties, are increasingly consumed around the world, often in the form of naturally- or freeze-dried fruits, further unprocessed. The aim of the study was to obtain dried goji berries extracts, characterized by high bioactivity and safety in terms of microbial contamination for the consumer. In the study, various solvents (water, ethanol, acetone, ethyl lactate, sunflower oil) and green extraction methods (heating and ultrasound-assisted extraction (UAE)) were used. In goji extracts, antioxidant activity and carotenoids content as bioactivity indicators, as well as total number of microorganisms were determined. Boiling of powdered dried goji fruits in water resulted in extracts with the best features, i.e., with high antioxidant properties (2.75–4.06 μmol of Trolox equivalent (TE)/mL), moderate to high content of carotenoids (0.67–1.86 mg/mL), and a reduced number of microorganisms compared with fruits. Extracts in 50% ethanol and 50% acetone were characterized primarily by very high antioxidant activity (3.09–4.90 μmol TE/mL). The high content of carotenoids (0.98–3.79 mg/mL) and high microbial quality (<10 CFU/g) were obtained by extraction in ethyl lactate by the UAE method. Taurine (Tau), a β-amino acid, exists in red goji fruit (Lycium barbarum L.). It exerts many cellular physiological functions such as anti-inflammation and oxidation resistance. The chemotherapy drug 5-fluorouracil (5FU) can cause intestinal mucositis. However, current therapeutic approaches for mucositis have limited efficacy and are associated with various side effects. It is still unknown whether Tau can alleviate intestinal mucositis. This study aimed to investigate the protective effect of the Tau in a mucositis mouse model and elucidate the underlying molecular mechanisms. The intestinal mucositis symptoms were alleviated by the Tau administration as evidenced by decreased body weight loss, histopathological score, oxidative stress, and improved glutathione (GSH). The Tau supplementation strengthened intestinal epithelial tight junction and reduced serum lipopolysaccharide (LPS) levels in intestinal mucositis mice. Moreover, the 5FU-induced inflammatory responses were alleviated by Tau treatment via the nuclear factor erythroid 2-related factor 2/heme oxygenase-1 (Nrf2/HO-1) and nuclear factor kappa-B/inducible nitric oxide synthase (NF-κB/iNOS) signaling pathway. Tau administration modulated short chain fatty acids (SCFAs) in the colon of mice. The results indicated that the Tau might be a new dietary strategy for intestinal mucositis caused by 5FU. Goji berry is a general term for various plant species in the genus Lycium. Goji has long been historically used in traditional Chinese medicines. Goji is a representative tonic medicine that has the effects of nourishing the liver and kidney and benefiting the essence and eyesight. It has been widely used in the treatment of various diseases, including tinnitus, impotence, spermatorrhea and blood deficiency, since ancient times. This study aims to comprehensively summarize the quality evaluation methods of the main compounds in goji, as well as the current research status of the phenolamides in goji and their pharmacological effects, to explore the feasibility of using phenolamides as quality control markers and thus improve the quality and efficacy in goji. Relevant literature from PubMed, Web of Science, Science Direct, CNKI and other databases was comprehensively collected, screened and summarized. According to the collected literature, the quality evaluation markers of goji in the Pharmacopoeia of the People's Republic of China are Lycium barbarum polysaccharide (LBP) and betaine. As a result of its structure complexity, only the total level of LBP can be determined, while betaine is not prominent in the pharmacological action of goji and lacks species distinctiveness. Neither of them can well explain the quality of goji. KuA and KuB are commonly used as quality evaluation markers of the Lycii cortex because of their high levels and suitable pharmacological activity. Goji is rich in polyphenols, carotenoids and alkaloids. Many studies have used the above compounds to establish quality evaluation methods but the results have not been satisfactory. Phenolamides have often been neglected in previous studies because of their low single compound levels and high separation difficulty. However, in recent years, the favorable pharmacological activities of phenolamides have been gradually recognized, and studies on goji phenolamides are greatly increasing. In addition, phenolamides have higher species distinctiveness than other compounds and can be combined with other compounds to better evaluate the quality of goji to improve its average quality. The phenolamides in the goji are rich and play a key role in antioxidation, anti-inflammation, neuroprotection and immunomodulation. As a result of their characteristics, it is suitable to evaluate the quality by quantitative analysis of multi-components by single-marker and fingerprint. This method can be combined with other techniques to improve the quality evaluation system of goji, which lays a foundation for their effectiveness and provides a reference for new quality evaluation methods of similar herbal medicines. Abstract Background Goji (Lycium spp., 2n = 24) is a fruit bearing woody plant popular as a superfood for extensive medicinal and nutritional advantages. Fruit size associated attributes are important for evaluating small-fruited goji berry and plant architecture. The domestication traits are regulated quantitatively in crop plants but few studies have attempted on genomic regions corresponding to fruit traits. Results In this study, we established high-resolution map using specific locus amplified fragment (SLAF) sequencing for de novo SNPs detection based on 305 F1 individuals derived from L. chinense and L. barbarum and performed quantitative trait loci (QTL) analysis of fruit size related traits in goji berry. The genetic map contained 3495 SLAF markers on 12 LGs, spanning 1649.03 cM with 0.47 cM average interval. Female and male parents and F1 individuals` sequencing depth was 111.85-fold and 168.72-fold and 35.80-fold, respectively. The phenotype data were collected for 2 successive years (2018–2019); however, two-year mean data were combined in an extra year (1819). Total 117 QTLs were detected corresponding to multiple traits, of which 78 QTLs in 2 individual years and 36 QTLs in extra year. Six Promising QTLs (qFW10–6.1, qFL10–2.1, qLL10–2.1, qLD10–2.1, qLD12–4.1, qLA10–2.1) were discovered influencing fruit weight, fruit length and leaf related attributes covering an interval ranged from 27.32–71.59 cM on LG10 with peak LOD of 10.48 and 14.6% PVE. Three QTLs targeting fruit sweetness (qFS3–1, qFS5–2) and fruit firmness (qFF10–1) were also identified. Strikingly, various traits QTLs were overlapped on LG10, in particular, qFL10–2.1 was co-located with qLL10–2.1, qLD10–2.1 and qLA10–2.1 among stable QTLs, harbored tightly linked markers, while qLL10–1 was one major QTL with 14.21 highest LOD and 19.3% variance. As LG10 harbored important traits QTLs, we might speculate that it could be hotspot region regulating fruit size and plant architectures. Conclusions This report highlights the extremely saturated linkage map using SLAF-seq and novel loci contributing fruit size-related attributes in goji berry. Our results will shed light on domestication traits and further strengthen molecular and genetic underpinnings of goji berry; moreover, these findings would better facilitate to assemble the reference genome, determining potential candidate genes and marker-assisted breeding.

Abstract Background Goji (Lycium) is a popular traditional health food, and its fruit and root extracts have been found to possess antioxidant, anti-inflammatory, and hypocholesterolemia-inducing abilities. Goji leaves also contain high amounts of phenolic compounds, similar to its fruit, and their extracts also exhibit several pharmaceutical effects. The induction of galls on Goji leaves reduces their photosynthetic ability and fruit yield, which raise their farming costs, thereby leading to economic loss. However, the defense mechanisms induced by infection may elevate the secondary metabolite content of the leaves, which might provide more nutritive compounds. Method Content of chlorophyll, carotenoids, polyphenols, and flavonoids in the extracts of normal and infected Goji leaves (L. chinense) were analyzed. The relative content of chlorogenic acid and rutin, two major phenolic compounds in Goji leaves, were determined by LC-MS/MS. Antioxidant activity was presented by demonstrating the DPPH scavenging percentage. The extract of Goji fruit (L. barbarum) was also analyzed to show a comparative result. Results In this study, we found that in infected Goji leaves, the polyphenol content was significantly increased. The level of chlorogenic acid was increased by 36% in galled leaves. The content of rutin in galled leaves was also elevated. Testing the antioxidant activities also showed that the extracts of galled leaves have higher DPPH scavenging abilities. Conclusions Our results demonstrated that galled Goji leaves have higher functional value, and may have potential as being consumed as health food.

O objetivo dessa revisão sistemática é demonstrar os resultados de estudos publicados sobre a ação do Lycium barbarum no metabolismo lipídico em humanos. Foram investigados artigos científicos publicados entre os anos de 2013 a 2018 nas bases de dados científicos PubMed, Science Direct, Cochrane Library e Web of Science. O método PICO (patient, intervention, comparison and outcomes) foi utilizado para nortear a pesquisa. A ferramenta de colaboração da Cochrane foi usada para avaliar o risco de viés dos estudos incluídos nessa revisão. Dos achados, foram selecionados 05 artigos relevantes para essa revisão. Os resultados dos estudos mostraram efeitos benéficos da suplementação do fruto do Lycium barbarum na diminuição de VLDL-C, LDL-C, colesterol total (CT) e aumento nos níveis de HDL-C. Contudo, novos estudos devem ser realizados para padronizar forma de preparo, tipo, dose e tempo de ingestão recomendado para padronizar ou assegurar o seu uso.

Introduction. Extract of goji berries (Lycium Barbarum L.) and buckwheat flour (Fagopýrum esculéntum L.) possess antioxidant and antimicrobial properties. As a result, they can be used to improve traditional Kazakh horse-meat formulations to obtain functional cooked and smoked meat products. These natural biologically active substances can improve the oxidative stability of pigments, lipids, and proteins of finished products. The research objective was to assess the potential of goji extract and buckwheat flour as additives that can improve the oxidative stability and general quality of Kanagat, a national Kazakh cooked and smoked horse-meat product. Goji extract and buckwheat flour were used in two concentrations – 0.5% and 1.0%. Study objects and methods. The research featured sensory evaluation of taste, smell, color, determination of color parameters (L*, a*, b*), pH, free amine nitrogen, total carbonyl proteins, acid value, peroxide value and thiobarbituric acid reactive substances (TBARS), as well as a histological analysis. Results and discussion. When 1.0% of goji extract and 1.0% of buckwheat flour were added to the traditional formulation, it improved the oxidative stability and quality of the modified horse-meat product while preserving its sensory properties and colour parameters. A set of microstructural studies showed that the processing of meat products with 1.0% of goji extract and 1.0% of buckwheat flour had a destructive effect on most fibers. The affected fibers showed multiple decays of myofibrillar substance, which turned into a finegrained protein mass. The abovementioned concentration caused effective inhibition of hydrolytic changes, as well as oxidation of proteins and lipids. Conclusion. The new technology made it possible to produce a new national horse-meat product fortified with 1.0% of goji extract and 1.0% of buckwheat flour. The specified amount of biologically active additives improved the oxidative stability and quality of the product, while maintaining its sensory and color characteristics.

Goji (Lycium barbarum L.) is one of the most popular medicinal plants, particularly as a typical nutritional fruit supplement rich with bioactive compounds. However, fruits browning, taste loss, and storage intolerance often occur during postharvest, because of tender and juicy tissue. Among them, the temperature is one of the most significant factors to maintain the quality and extend the shelf life of fruits. It is necessary to place fruits in a constant temperature environment after harvest since temperature fluctuations and variation tend to induce product quality deterioration. This study aims to investigate the effect of precise temperature control on storage quality and aroma components of fresh goji fruits. Three treatments of storage temperature at 0℃ were also set, including foam box (CK), foam box + cool storage agent (ice temperature), as well as the precise temperature control box and cool storage agent (phase temperature). The temperature was real-time recorded in different cabinets to evaluate the storage quality. Principal component analysis (PCA) was used to evaluate the aroma components of goji. Results showed that the foam box containing the self-made refrigerant performed better at ambient temperature, compared with that with the ice. Temperatures in the box of CK, ice, and phase temperature group were (0.12±0.17), (-0.04±0.07) and (-0.05±0.04)℃, respectively, during the whole storage period. A cold accumulator provided a cold source in the box to maintain a low-temperature environment. The temperature distribution was more uniform in the precision temperature control box, while the maintenance time of low temperature was longer than before, due to the excellent performance of heat preservation. The time of mildew and rot in the CK group, ice, and phase temperature group were 10, 20, and 40 d, respectively. At the storage of 40 d, the rot rate, mildew rate, and soft fruit rate in the phase temperature group were 4.11%, 4.50%, and 5.08%, respectively, much lower than 14.85%, 15.44%, and 15.01% in the CK group. Chromatic aberration ΔE was lower than 3, while Brightness L value reached 34.12. The content of soluble solid, titratable acid, Vitamin C conten, carotenoid were 0.65 percentage, 0.03 percentage, 4.34 mg/100g, 2.90 mg/g higher than CK group, respectively. Correspondingly, the treatment in the phase temperature group inhibited the increase of decay rate and weight loss rate, indicating the delay of ΔE and L, as well as a higher soluble solid, titratable acid, VC, and carotenoid content. The principal component analysis demonstrated that the goji quality in three groups was in the order of phase temperature group, ice temperature group and CK group. Moreover, the contents of favorable aroma components, such as lycium aldehydes and terpenoids, were relatively higher in the storage under precise temperature control. Consequently, the ice and phase temperature storage was the most effective in the long-term preservation of goji, where the color change of goji was delayed to facilitate the release of volatile substances, while effectively prolonging the storage period of goji to 30 days. Awareness of health benefits of goji berries coming from their bioactive compounds, mostly antioxidants like ascorbic acid, has grown. Recently, an ascorbic acid analogue from goji berries, the 2-O-?-D-glucopyranosyl-L-ascorbic acid has been reported. In rats, the analogue is absorbed intact and in the form of free vitamin C and consequently has been proposed as a provitamin C. Synthesis of the analogue is demanding and laborious and therefore reliable natural sources are searched. Knowledge concerning the analogue's occurrence in other parts of goji plant is lacking. The aim of this study was to evaluate the contents of 2-O-?-D-glucopyranosyl-L-ascorbic acid in rhizomes, stems and leaves from Lycium barbarum. Rhizomes, stems and leaveswere extracted and the content of 2-O-?-D-glucopyranosyl-L-ascorbic acid and non glucosylated, free ascorbic acid was determined by HPLC-DAD. 2-O-?-D-glucopyranosyl-L-ascorbic acid was found in all goji plant tissues investigated. Based on dry weight, 3.34 mg/100 g were found in the leaves, 4.05 mg/100 g in the stems and up to 12.6 mg/100 g in the rhizomes. Nevertheless, the analogue content in goji berries is much higher (40 to 280 mg/100 g dry weight). The present study confirmed the presence of 2-O-?-D-glucopyranosyl-L-ascorbic acid in rhizomes, stems and leaves of Lycium barbarum. However, their content compared to goji berries is considerably lower.

Chronic gut inflammation is an important risk factor for colorectal cancer (CRC). Our recent study suggests that dietary Goji reduces gut inflammation and increases Bifidobacteria and butyrate-producing bacteria in interleukin-10-deficient mice. This study profiled faecal metabolomic changes of the same cohort of mice. Thirty-four compounds were altered due to Goji supplementation, including polyunsaturated long-chain fatty acids (PUFAs), sugar and sugar alcohols, benzene derivatives, pyridine derivatives, dicarboxylic acids and short-chain hydroxy acids. Pathway analysis showed that α-linolenic acid and linoleic acid metabolism were strongly affected by Goji supplementation. Four ω-6 PUFAs, including linoleic acid and arachidonic acid were reduced due to Goji supplementation. Faecal levels of most amino acids, positively correlated with gut inflammation and CRC risk, tended to decrease in the Goji supplemented group along with decreased urease activity. Data provide valuable metabolic clues for unravelling the beneficial effects of Goji supplementation against gut inflammation and related disease.

The aim of the study was to analyze potential health-promoting components of 21 new cultivars of goji (Lycium barbarum L.). Polyphenols and carotenoids were identified by LC-MS-Q/TOF and quantified by UPLC-PDA. Moreover, vitamin C, sugar and organic acid profile, antioxidant (ABTS+·, FRAP) and in vitro antidiabetic potential (inhibition of α-amylase and α-glucosidase) and anti-aging (acetylcholinesterase (AChE) and butyrylcholinesterase BuChE) activity were evaluated. Some new cultivars contained high amounts of polyphenols (g8, g13 and g43) and carotenoids (g11, g26 and g12), and showed antioxidant (g8, g17, g22, g23), antidiabetic (g26), and anti-aging activity (g4, g6, g37). They were also content of vitamin C (2.39 to 6.24 mg/100 g), organic acids (1.95–4.30 g/100 g), and sugars (1.59–5.11 g/100 g). In conclusion, selected new cultivars might be used for the production of functional foods, as well as for medical or/and cosmetic purposes.

• RAMs is devised to enrich neonicotinoid pesticides while exclude proteins. • RAMs possess dual-function adsorption sites and zwitterionic polymer exclusion sites. • Offline and online RAMs couple with LC show the potential in the detection of neonicotinoid pesticides from Goji samples. • Online RAMs-LC is able to complete extraction-detection of neonicotinoid pesticides into one step by simple valve-switching. A method based on novel restricted access materials (RAMs) for the determination of neonicotinoid pesticides in Goji samples using offline and online solid phase extraction (SPE) coupled with high-performance liquid chromatography (LC). RAMs were synthesized using poly(chloromethylstyrene- co -divinylbenzene) (P VBC/DVB) microspheres as substrate, styrene (St) and n-vinylpyrrolidone (NVP) were first copolymerized on the interior to construct adsorption sites, and sulfobetaine methacrylate (SBMA) was then polymerized on the exterior to form exclusion sites via two-step surface initiated-atom transfer polymerization. The prepared P VBC/DVB @poly(St- co -NVP)@poly(SBMA) RAMs could efficiently extract neonicotinoid pesticides and automatically exclude proteins. Under the optimized conditions, the developed methods of offline (magnetic SPE and SPE column) and online extraction coupled with LC both using P VBC/DVB @poly(St- co -NVP)@poly(SBMA) RAMs as the extractant, exhibit a wide linearity, low limits of detection and limit of quantification and good inter-day and intra-day precision with satisfactory recoveries. Among these methods, online extraction coupled with LC based on novel RAMs exhibits clear advantages for the determination of neonicotinoid pesticides in Goji samples has clear advantages, such as simple operation by direct injection, short extraction times, and high accuracy with less human error. Goji berries have long been used for their nutritional value and medicinal purposes in Asian countries. In the last two decades, goji berries have become popular around the world and are consumed as a functional food due to wide-range bioactive compounds with health-promoting properties. In addition, they are gaining increased research attention as a source of functional ingredients with potential industrial applications. This review focuses on the antioxidant properties of goji berries, scientific evidence on their health effects based on human interventional studies, safety concerns, goji berry processing technologies, and applications of goji berry-based ingredients in developing functional food products.

With a great variety of foods on the market, the consumer’s responsibility is to choose food that can allow him to maintain his state of health and prevent chronic diseases. For this reason, several functional foods classified as fortified or enriched are currently being developed. In fact, the functional food market size is estimated to reach USD 267,924.4 million by 2027 [1]. Functional foods are able to provide essential nutrients such as vitamins, minerals and bioactive substances. The opportunities for incorporating these bioactive constituents into bread have grown rapidly, as bread is the staple food in many countries. Lycium barbarum (goji) berries are a source of phytochemicals with important biological functions and are designated as super-fruits [2]. The aim of the study was to add goji puree at different percentages (50% and 70%) to the bread dough. To evaluate the characteristics of bread enriched in goji puree, chemical–physical, sensory, rheological analysis and radical scavenging activity tests were carried out [3,4]. The best results were obtained with the sample enriched with 50% goji puree (B50G), showing a total phenol content of 42.07 mg gallic acid equivalent/100 g bread and an ABTS radical scavenging activity of 833.48 µmol Trolox/100 g bread. Bread sensory profile was identified by a trained panel using quantitative descriptive analysis, showing significant differences compared with the untreated sample in terms of crust, crumb colour and structural crunchiness, while olfactory and gustatory descriptors did not differ significantly. Furthermore, the enriched product is characterized by a higher content of bioactive substances, with particular reference to phenolic compounds, and by an interesting antioxidant activity. The absence of substantial changes in the sensory profile represents another fundamental aspect appreciated by the consumers. Collectively, our results demonstrate the potential health properties of this enriched bread.

Objective: Goji (fruits of Lycium spp.) is commonly consumed as food and medicine. The increasing market demand for goji has led to its wide cultivation and broad breeding, which might cause loss of genetic diversity. This study aims to uncover the genetic diversity of the cultivated and wild Lycium ., Methods: The chloroplast genome (CPG) of 34 accessions of Chinese food-medicinal Lycium spp., including the popular cultivars and their wild relatives, was re-sequenced and assembled, based on which the genetic diversity was evaluated., Results: Sequence structural comparison shows that CPG is comparatively conserved within species. Phylogenetic analysis indicates that CPG is sufficient for the discrimination of Lycium species; combined with nuclear ribosomal internal transcribed spacer (Nr ITS) sequences, materials with mixed genetic backgrounds can be identified. Nucleotide diversity analysis reveals that the modern cultivars are probably with a common maternal parent, while the wild accessions are with higher level of genetic diversity., Conclusion: For the first time this study reveals the intraspecies genetic diversity of Lycium spp. using CPG, highlighting the urgent conservation demand of wild genetic resources of Lycium . Our study also demonstrates that CPG provides crucial evidence for identification of Lycium species with mixed genetic backgrounds and highlights the importance of the wild relatives in genetic diversity conservation. The aim of this research was to evaluate the biochemical composition of the Nile tilapia meat in conditions of five phytobiotics administrated in feed. This experiment was performed in duplicate. The experimental variants were: V1 - control variant, V2 - 1% chilli pepper (Capsicum annuum) / kg feed, V3 - 1% black pepper (Pipper nigrum) / kg feed, V4 - 1% onion (Allium cepa) / kg feed, V5 - 1% goji fruits (Lycium barbarum) / kg feed and V6 - 1% basil (Ocimum basilicum) / kg feed. For biochemical analysis from muscle tissue, the sampling was performed at the beginning and at the end of the experiment from fresh meat. The results showed a significant differences (p<0.05), between variants in which were administered phytobiotics compared with the control variant, in case of the protein content (%), moisture content (%) and dry matter (%). Also, in this paper are presented the dynamics of the nutrient retention efficiency (PER-g/g, PUE-%, RP-g/fish and RLg/ fish) in the muscle tissue of the Nile tilapia. The best results were obtained in the case of the V3 variant regarding to retained protein (RP), PER and PUE and in V5 variant regarding to retained lipid (RL). In conclusion, the use of these phytobiotics in a concentration of 1% / kg feed in the Nile tilapia diet, led to significant changes (p <0.05) in the positive sense of the percentage moisture, protein and dry matter from muscle tissue and has contributed to the increasing the nutritional value of fish. The objective of this systematic review is to demonstrate the results of published studies on the action of Lycium barbarum on lipid metabolism. Scientific articles published between the years 2013 to 2018 were investigated in the scientific databases PubMed, Science Direct, Cochrane Library and Web of Science. The PICO (patient, intervention, comparison and outcomes) method was used to guide the research. The Cochrane Collaboration Tool was used to assess the risk of bias in the studies included in this review. Of the findings, we selected 05 articles relevant to this review. The results of the studies showed beneficial effects of supplementation of the Lycium barbarum fruit on the decrease of VLDL-C, LDL-C, TC (total cholesterol) and increase in HDL-C levels. However, further studies should be performed to standardize the preparation, type, dose and recommended intake time to standardize or ensure. [ABSTRACT FROM AUTHOR]

Novel food, super food, wolfberry, the Himalayan longevity fruit. Several different and, even more, colourful names define the fruits of Lycium barbarum L., one of the most popular food supplements and anti-aging remedies of the recent years. Goji derivatives find application at the interface between food and medicine thanks to the claimed antioxidant and immunomodulatory properties. Aiming at identifying the most effective extraction protocol for processing different goji berries samples (fresh and dried, commercial and from an Italian cultivar), combinations of extractive and analytical techniques were attempted. The concentration of bioactive compounds and of total phenols were measured and the antioxidant activity of the obtained extracts was studied. Fifteen phenylpropanoid glycosides, including six undescribed compounds were isolated from the fruit of Lycium barbarum L. (Solanaceae) (goji or wolfberry). Their structures were identified by detailed spectroscopic analyses. Seven known compounds were firstly isolated from the genus Lycium , in which the 1D and 2D NMR data of one compound were reported for the first time. Notably, two undescribed compounds were a pair of rare tautomeric glycoside anomers characterized by the presence of free anomeric hydroxy. Antioxidant and hypoglycemic activities of all these compounds were assessed using DPPH radical scavenging, oxygen radical absorbance capacity (ORAC), and α -glucosidase inhibitory assays, respectively. These compounds showed different levels of oxygen radical absorbance capacity, and some isolates exhibited potent antioxidant activity with greater ORAC values than the positive control (EGCG). In the current study, a carboxyl-rich polysaccharide purified from Lycium barbarum L. leaves (hereafter, LP) and its degradation with ascorbic acid and hydrogen peroxide were characterized. Degradation decreased the molecular weight of LP from 4.63 × 104 to 3.45 × 104 Da, and increased its zeta potential from −8.01 to −5.35 mV. In vitro experiments showed that degradation significantly increased the anticoagulant activity and, in particular, antiplatelet activity of LP (p < 0.05). The polysaccharide with the highest degree of degradation had higher inhibitory activity than aspirin against arachidonic acid- and thrombin-induced platelet aggregation at 0.5 g/mL. A reduction in uronic acids between LP and its degradation products significantly decreased their antiplatelet activity (p < 0.05). Further analysis confirmed that polysaccharides changed from a compact spherical structure to a random coil in aqueous solution following degradation, which facilitated the interaction of polysaccharides and platelets. • Polysaccharide from Lycium barbarum L. leaves (LP) was well characterized. • The degradation with Vc and H 2 O 2 potently increased LP's antiplatelet activity. • Less negative charges of degraded LP facilitate interaction of LP and platelets. • The degradation changes LP from a globular structure to a random coil conformation.
