Abstract

Abstract Background Metabolic dysfunction-associated steatotic liver disease (MASLD), previously known as non-alcoholic fatty liver disease (NAFLD), is a prevalent hepatic condition linked to metabolic alterations. It gradually causes liver damage and potentially progresses to cirrhosis. Despite its significance, research, especially in the pediatric population, is limited, leading to contradictory findings in diagnosis and treatment. This meta-analysis aims to synthesize existing literature on therapeutic interventions for MASLD in children and adolescents. Methods A comprehensive search of randomized controlled clinical trials yielded 634 entries from PubMed, Scopus, and Web of Science up to 2023. Interventions included medications, behavioral modifications, dietary changes, probiotics, supplements, surgical procedures, or combinations. The analysis focused on studies with treatment duration of at least 3 months, employing a random-effects REML meta-analysis model. Treatment effects on anthropometric measurements and biochemical components were examined and adjusted for heterogeneity factors analysis. A bibliometric analysis for insights into research contributors was performed. Results The systematic review incorporated 31 clinical trials, with 24 meeting criteria for meta-analysis. These comprised 3 medication studies, 20 with supplements, 4 focusing on lifestyle, and 4 centered on diets. Significant overall treatment effects were observed for ALT, AST, BMI, and HOMA-IR mainly by supplements and lifestyle. Meta-regression identified age, BMI changes, and treatment duration as factors modifying ALT concentrations. Bibliometric analysis involving 31 linked studies highlighted contributions from 13 countries, with the USA, Spain, and Chile being the most influential. Conclusions We conclude that supplementation and lifestyle changes can effectively impact ALT and AST levels, which can help address liver issues in obese children. However, the evaluation of risk bias, the high heterogeneity, and the bibliometric analysis emphasize the need for more high-quality studies and broader inclusion of diverse child populations to provide better therapeutic recommendations. Abstract

Abstract Background Apolipoprotein E (APOE) genotypes have been suggested to influence cognitive impairment and clinical onset in presenilin-1 (PSEN1) E280A carriers for autosomal dominant Alzheimer’s disease (ADAD). Less is known about their impact on the trajectory of biomarker changes. Neurofilament light chain (NfL), a marker of neurodegeneration, begins to accumulate in plasma about 20 years prior to the clinical onset of ADAD. In this study we investigated the impact of APOE ε4 and ε2 variants on age-related plasma NfL increases and cognition in PSEN1 E280A mutation carriers. Methods We analyzed cross-sectional data from PSEN1 E280A mutation carriers and non-carriers recruited from the Alzheimer’s Prevention Initiative Registry of ADAD. All participants over 18 years with available APOE genotype, plasma NfL, and neuropsychological evaluation were included in this study. APOE genotypes and plasma NfL concentrations were characterized for each participant. Cubic spline models using a Hamiltonian Markov chain Monte Carlo method were used to characterize the respective impact of at least one APOE ε4 or ε2 allele on age-related log-transformed plasma NfL increases. Linear regression models were estimated to explore the impact of APOE ε4 and ε2 variants and plasma NfL on a composite cognitive test score in the ADAD mutation carrier and non-carrier groups. Results Analyses included 788 PSEN1 E280A mutation carriers (169 APOE ε4 + , 114 ε2 +) and 650 mutation non-carriers (165 APOE ε4 + , 80 ε2 +), aged 18–75 years. APOE ε4 allele carriers were distinguished from ε4 non-carriers by greater age-related NfL elevations in the ADAD mutation carrier group, beginning about three years after the mutation carriers’ estimated median age at mild cognitive impairment onset. APOE ε2 allele carriers had lower plasma NfL concentrations than ε2 non-carriers in both the ADAD mutation carrier and non-carrier groups, unrelated to age, and an attenuated relationship between higher NfL levels on cognitive decline in the ADAD mutation carrier group. Conclusions APOE ε4 accelerates age-related plasma NfL increases and APOE ε2 attenuates the relationship between higher plasma NfL levels and cognitive decline in ADAD. NfL may be a useful biomarker to assess clinical efficacy of APOE-modifying drugs with the potential to help in the treatment and prevention of ADAD.

Abstract

Abstract Autosomal dominant Alzheimer’s disease (ADAD) is genetically determined, but variability in age of symptom onset suggests additional factors may influence cognitive trajectories. Although apolipoprotein E (APOE) genotype and educational attainment both influence dementia onset in sporadic AD, evidence for these effects in ADAD is limited. To investigate the effects of APOE and educational attainment on age-related cognitive trajectories in ADAD, we analyzed data from 675 Presenilin-1 E280A mutation carriers and 594 non-carriers. Here we show that age-related cognitive decline is accelerated in ADAD mutation carriers who also have an APOE e4 allele compared to those who do not and delayed in mutation carriers who also have an APOE e2 allele compared to those who do not. Educational attainment is protective and moderates the effect of APOE on cognition. Despite ADAD mutation carriers being genetically determined to develop dementia, age-related cognitive decline may be influenced by other genetic and environmental factors.

Abstract

Neogene sedimentary rocks cover extensive areas of the Cabo de Gata-Níjar UNESCO Global Geopark (Almería, SE Spain) although most of the outcropping rocks are Miocene volcanics. The post-volcanic sedimentary rocks include three successive Messinian coral reef units. The lower reefs consist of coral patches of varying dimensions comprising Porites and Tarbellastraea, which grew on carbonate ramps. The second unit is Porites fringing reefs that prograded from shorelines. These two lower units, early Messinian in age, were subaerially exposed and eroded during a relative sea-level drawdown, probably coeval with the Mediterranean Messinian salinity crisis. The last, late Messinian reef unit onlaps the erosion surface and consist of small Porites patches that grew dispersed in oolitic shoals together with microbial carbonates. These low-diversity reefs are the last zooxanthellate coral reefs in the Mediterranean basin and constitute the endpoint of decreasing coral diversity in the region during the Miocene due to disconnection from the Indian Ocean and global cooling. The good exposures and accessibility of these reefs in the arid landscape of the Geopark make them valuable sites for practical training in geology and paleontology at different levels and reference sites for geotourism.

Abstract

Abstract Background Pregnancy and the postpartum period are times of great change for women and their partners, often bringing substantial challenges and stress. Approximately 10%–20% of women suffer from mood disorders such as depression in the perinatal period. There are risks involved in using psychopharmacological interventions to treat perinatal depression. Mindfulness and compassion-based educational programs could be efficacious and cost-effective options for the prevention and treatment of perinatal mood disorders. The aim of this study is to assess the efficacy of an adapted Mindfulness-Based Childbirth and Parenting (MBCP) program that includes compassion training for pregnant women in primary care (PC) settings in the Spanish National Health System to decrease perinatal depression. Methods A multicenter randomized controlled trial (RCT) will be conducted. Participants will be pregnant women (n = 122) and their partners who wish to participate. They will be enrolled and assessed in PC settings and randomly assigned to either: (1) an adapted MBCP educational program tailored to the Spanish National Health System + treatment as usual (TAU); or (2) TAU only. The main outcome to be assessed will be depression, evaluated with the Edinburgh Postnatal Depression Scale (EPDS). Secondary outcomes will include self-reported measures of perceived stress, affects, mindfulness, self-compassion, maternal self-efficacy, and use of health and social services. Patients will be assessed at four timepoints: baseline; post-treatment; and at three and six months after childbirth. Intention-to-treat and per-protocol analyses will be carried out using linear regression mixed models. Effect sizes will be estimated using Cohen’s d. Discussion Perinatal depression is a significant health problem. An effective and low-cost childbirth education program that incorporates mindfulness and compassion practices may be a beneficial preventive complementary healthcare modality for expectant women and their partners. This study will be the first multicenter RCT in Spanish PC settings using adapted MBCP and compassion practices to reduce symptoms of depression during pregnancy and the postpartum period. En este artículo, con referentes de Thomas Popkewitz, se analiza la problemática de la equidad, específicamente las estrategias de inclusión mediante la administración social y normalización de los Derechos Básicos de Aprendizaje (DBA) de las matemáticas que operan el principio “Matemáticas para todos”. Se realiza el análisis documental en torno a tres problemáticas ligadas a la inclusión de todos: (1) las relaciones entre administración social del Estado, educación matemática y equidad; (2) la investigación en educación matemática que se ha ocupado de la problemática de equidad e inclusión; y (3) los aparatos de administración y mecanismos de regulación de prácticas de aprendizaje de las matemáticas inclusivas. El análisis se realizó sobre dos tipos de documentos, los del campo de investigación en educación matemática y de políticas nacionales y estudios internacionales, y los de materiales curriculares y técnicas de evaluación. El análisis posibilitó concluir que la gestión de estrategias de inclusión de los DBA sigue considerando que el aprendizaje es monolítico y la gestión de homogenización del aprendizaje allí promovido vuelve a reforzar la exclusión. A partir de este análisis nos interrogamos acerca de con qué políticas del conocimiento queremos incluir a todos los estudiantes para que aprendan matemáticas.

Abstract

The transcription factor TCF7L1 is an embryonic stem cell signature gene that is upregulated in multiple aggressive cancer types, but its role in skin tumorigenesis has not yet been defined. Here we document TCF7L1 upregulation in skin squamous cell carcinoma (SCC) and demonstrate that TCF7L1 overexpression increases tumor incidence, tumor multiplicity, and malignant progression in the chemically induced mouse model of skin SCC. Additionally, we show that downregulation of TCF7L1 and its paralogue TCF7L2 reduces tumor growth in a xenograft model of human skin SCC. Using separation-of-function mutants, we show that TCF7L1 promotes tumor growth, enhances cell migration, and overrides oncogenic RAS-induced senescence independently of its interaction with β-catenin. Through transcriptome profiling and combined gain- and loss-of-function studies, we identified LCN2 as a major downstream effector of TCF7L1 that drives tumor growth. Our findings establish a tumor-promoting role for TCF7L1 in skin and elucidate the mechanisms underlying its tumorigenic capacity.

El propósito de este artículo es compartir las posibilidades de reinvención de un camino de constitución de subjetividades sociales incluyentes en el aula de matemáticas, donde la exclusión del aprendizaje y la exclusión social coexisten. Lo haré desde investigaciones realizadas en aulas de matemáticas en las que la (in)exclusión en la participación de estudiantes es evidente. Desde la perspectiva sociopolítica de la educación matemática y con argumentos procedentes de la teoría crítica del currículo y de los estudios foucaultianos en educación, analizaré, primero, el relato de la inclusión de las matemáticas escolares en dos reformas curriculares de las matemáticas. Cuestiono los posibles significados de los sistemas de razón como prácticas históricas que normalizan y construyen las tesis culturales sobre la (in)exclusión. Propongo la dimensión cultural en el aula de matemáticas como una noción nodal en las prácticas de aprendizaje y enseñanza de las matemáticas para constituir subjetividades sociales incluyentes. A partir de una aproximación cualitativa, este artículo analiza las tensiones y negociaciones cotidianas que se producen en torno a la presencia y rol de la religión en las prisiones y hospitales contemporáneos en España. Más concretamente, el artículo examina la emergencia de nuevas formas de encaje entre la Iglesia católica, las minorías religiosas y las instituciones seculares; y sus implicaciones en cuanto a la naturaleza secular del Estado y las desigualdades entre grupos. La complejidad de las dinámicas religioso/secular, catolicismo/diversidad religiosa y religión/espiritualidad patentes en el marco de estas instituciones evidencia la imposibilidad de dibujar un modelo único para dar cuenta de cómo la religión y la diversidad religiosa son gestionadas en estos contextos.

Abstract

Understanding the molecular mechanisms of ultraviolet (UV) induced melanoma formation is becoming crucial with more reported cases each year. Expression of type II nuclear receptor Retinoid-X-Receptor α (RXRα) is lost during melanoma progression in humans. Here, we observed that in mice with melanocyte-specific ablation of RXRα and RXRβ, melanocytes attract fewer IFN-γ secreting immune cells than in wild-type mice following acute UVR exposure, via altered expression of several chemoattractive and chemorepulsive chemokines/cytokines. Reduced IFN-γ in the microenvironment alters UVR-induced apoptosis, and due to this, the survival of surrounding dermal fibroblasts is significantly decreased in mice lacking RXRα/β. Interestingly, post-UVR survival of the melanocytes themselves is enhanced in the absence of RXRα/β. Loss of RXRs α/β specifically in the melanocytes results in an endogenous shift in homeostasis of pro- and anti-apoptotic genes in these cells and enhances their survival compared to the wild type melanocytes. Therefore, RXRs modulate post-UVR survival of dermal fibroblasts in a "non-cell autonomous" manner, underscoring their role in immune surveillance, while independently mediating post-UVR melanocyte survival in a "cell autonomous" manner. Our results emphasize a novel immunomodulatory role of melanocytes in controlling survival of neighboring cell types besides controlling their own, and identifies RXRs as potential targets for therapy against UV induced melanoma.

Abstract

There are two important subjects concerning internationalization strategies that are of interest to academia, the business community and society in general. The first one is internationalization of small and medium-sized enterprises (SMEs); the other one is internationalization of local cultures. This article provides insights into the specific internationalization strategies used by Japanese SMEs to export traditional arts to foreign markets, focusing on Japanese pottery for the tea ceremony. SMEs encounter difficulties concerning limited resources in their internationalization process, but what is the situation of SMEs whose business activity is linked to indigenous cultural factors? The findings have been obtained from in-depth interviews with pottery artists as well as qualitative analysis of websites and other sources and they suggest a new previous phase in the export of products with a cultural meaning. I called this phase the 'reverse prior phase' and I argue that it might be a suitable way to start the internationalization process for Japanese traditional arts.

Abstract

Even though cognitive impairment in brain vascular disease has been widely described, there is not much known about specific neuropsychological profiles in affected families suffering from hereditary vascular dementia CADASIL. In Colombia we have reported for the first time two large kindreds suffering from CADASIL in the Antioquia region (Colombia). Those families carry mutations R1031C and C455R in notch 3 gene, respectively. In this study, we have develop a comparative analysis between 16 affected carriers of R1031C (family A) and 7 affected carriers of mutation C455R (family B). This includes a description of cognitive performance in these two groups applying specific protocols evaluating several cognitive functions, depression and everyday functionality. Analysis showed differences in age of onset, evolution time, dementia diagnosis and degree of leukoencephalopaty between R1031C and C455R carriers. Abstract

In the framework of the debate on high-frequency gravitational waves (GWs), after a review of GWs in standard General Relativity, which is due for completness, the possibility of merging such a traditional analysis with the Hyperspace formalism that has been recently introduced in some papers in the literature, with the goal of a better understanding of manifolds dimensionality also in a cosmological framework, is discussed. Using the concept of refractive index in the Hyperspace, spherical solutions are given and the propagation of GWs in a region of the Hyperspace with an unitary refractive index is also discussed. Propagation phenomena associated to the higher dimensionality are proposed, possibly including non-linear effects. Resumen Este artículo presenta una reflexión sobre las posibilidades de la diversidad, en contextos de desigualdad e injusticia en clases de matemáticas, al cuestionar el centro del currículo organizado alrededor de estructuras y conceptos o competencias matemáticas como principios de poder y control para la producción de identidades homogéneas y universales. Con el desplazamiento del centro del currículo a situaciones sociales políticas relevantes, enfatizamos la construcción de subjetividades sociales para conocer y generar visiones comunes de nuevas condiciones de vida futura. Abstract This article presents a reflection on the possibilities of Diversity, in contexts of inequality and injustice in mathematics classes by questioning the center of the curriculum organized around structures and concepts or mathematical competencies as principles of power and control for the production of homogeneous identities. and universal. Abstract

Plasma-measured tau phosphorylated at threonine 217 (p-tau217) is a potential non-invasive biomarker of Alzheimer's disease (AD). We investigated whether plasma p-tau217 predicts subsequent cognition and positron emission tomography (PET) markers of pathology in autosomal dominant AD.We analyzed baseline levels of plasma p-tau217 and its associations with amyloid PET, tau PET, and word list delayed recall measured 7.61 years later in non-demented age- and education-matched presenilin-1 E280A carriers (n = 24) and non-carrier (n = 20) family members.Carriers had higher plasma p-tau217 levels than non-carriers. Baseline plasma p-tau217 was associated with subsequent amyloid and tau PET pathology levels and cognitive function.Our findings suggest that plasma p-tau217 predicts subsequent brain pathological burden and memory performance in presenilin-1 E280A carriers. These results provide support for plasma p-tau217 as a minimally invasive diagnostic and prognostic biomarker for AD, with potential utility in clinical practice and trials.Non-demented presenilin-1 E280A carriers have higher plasma tau phosphorylated at threonine 217 (p-tau217) than do age-matched non-carriers. Higher baseline p-tau217 is associated with greater future amyloid positron emission tomography (PET) pathology burden. Higher baseline p-tau217 is associated with greater future tau PET pathology burden. Higher baseline p-tau217 is associated with worse future memory performance.

Abstract

Background Neurofilament light (NfL) is a promising biomarker of early neurodegeneration in Alzheimer's disease (AD). We examined whether plasma NfL was associated with in vivo amyloid beta and tau, and cognitive performance in non-demented presenilin-1 (PSEN1) E280A mutation carriers. Methods Twenty-five mutation carriers and 19 non-carriers (age range: 28 to 49 years) were included in this study. Participants underwent 11C Pittsburgh compound B (PiB)-PET (positron emission tomography), flortaucipir-PET, blood sampling, and cognitive testing. Results Mutation carriers exhibited higher plasma NfL levels than non-carriers. In carriers, higher NfL levels were related to greater regional tau burden and worse cognition, but not amyloid beta load. When we adjusted for age, a proxy of disease progression, elevated plasma NfL levels were only correlated with worse memory recall. Conclusions Findings support an association between plasma NfL, cognition, and tau pathology in non-demented individuals at genetic risk for developing AD dementia. Plasma NfL may be useful for selecting individuals at increased risk and tracking disease progression in AD.

Abstract

En la novela de autoficción Lo que hay (2022) la escritora Sara Torres realiza una indagación narrativa en la experiencia de un duelo doble: el amoroso por la pérdida de una amante, y el maternal, tras el fallecimiento de su madre a causa de un cáncer de mama. Con la ausencia como hilo conductor, la obra aborda otras temáticas como los discursos y la cultura hegemónica en torno al cáncer de mama, la vivencia de la sexualidad y el deseo (lésbicos) en un estado de duelo, la ética de los cuidados, y el potencial y las limitaciones de la literaturapara subjetivizar, elaborar y comunicar estas experiencias. Este artículo analiza las tres temáticas principales que vertebran la novela: el cáncer de mama, el duelo y el deseo, argumentando que la autora abre nuevas vías representacionales para imaginar la enfermedad y la pérdida desde una ética queer y feminista.

Abstract

El estudio de las emisiones de la BBC en diferentes idiomas durante la Segunda Guerra Mundial revela la extraordinaria confianza que el gobierno británico depositó en la radio, persuadido de su potencial influencia sobre la sociedad civil, como ya lo había hecho desde 1934 con la puesta en marcha del British Council, en un interesante avance de lo que a partir de los años 90 Joseph Nye denominaría Soft Power. Este trabajo aborda el estudio del departamento ocupado dentro de la BBC de las emisiones dirigidas a España y conocido como Servicio Español desde su creación en 1939 hasta el final de la Segunda Guerra Mundial. En este contexto, al Servicio Español se le encomienda la difícil labor de afianzar el vínculo de sus oyentes españoles con la causa aliada sin por ello inmiscuirse en la política española, en un difícil equilibrio que desencadenará no pocas tensiones políticas y que llevará al límite la política de contención hacia España marcada por Churchill. A fin de dar curso al enfoque narrativo de este trabajo, se ha hecho uso de Memorias, de fuentes de archivo del War Cabinet y del Foreign Office, de publicaciones periódicas, así como de fuentes bibliográficas.

Abstract

Blue Planet Law is the global and future-oriented environmental law that is necessary to face the global environmental crisis in the Anthropocene, assuming especially the link between climate action (SDG 13) and ocean sustainability (SDG 14). This open access book focuses on means of overcoming global environmental problems such as climate change, ocean degradation and biodiversity loss and the consequent risks for human life, health, food and wellbeing. It explores how environmental law, at the international, European and national levels, might set economic and technological development on a more sustainable path. Law must engage in dialogue with other areas such as philosophy, economics, ecology, and biology. This book highlights protection of the climate and the oceans and sustainable use of natural resources, through new policies, economies and technologies, including biotechnology, with a view to the preservation of life, health, food and a healthy environment for the present and future generations. The book may be seen as a contribution to the UN Sustainable Development Goals 13 and 14 and a tribute to the Declaration of the United Nations Conference on the Human Environment, also known as the Stockholm Conference (1972), on its 50th Anniversary.

Having a national geoheritage inventory is essential to plan effective geoconservation strategies. Since 2017, the Geological Survey of Brazil (CPRM) has been carrying out a project aimed at the Inventory of the Brazilian Geological Heritage and defined state coordinations to propose indicative lists of potential geosites based mainly on the scientific value (SV) according to the GEOSSIT platform. For the state of São Paulo, which was the first in Brazil to have a systematic geoheritage inventory, with 137 geosites already defined, this study intends to analyze them to propose some criteria to select the ones to compound the national list. Fifty-seven geosites were chosen according to both SV (≥ 300, following the requirements of GEOSSIT) and representativeness within each geological framework (when SV < 300). We also evaluated the selected geosites in other national initiatives, such as SIGEP (nine geosites) and the Geoparks Project (five geosites). The GEOSSIT public lists show only three of the 57 geosites already registered, a low number considering that these registrations are relevant indicators for the national inventory. The geosites were also analyzed according to the main thematic classification (eight main thematic categories, with a large number in the petrology theme - 35.10%) and general geological context (73.70% in the Mantiqueira, Paraná, and Tocantins provinces and 26.30% in Emerged Phanerozoic Basins - Paraná, Bauru, and São Paulo), according to the parameters available on GEOSSIT. The sites were also evaluated according to typology, being 33 points, 22 areas, and two sections. Regarding the statutory setting, 30% are in fully protected areas, 36% in public or private areas with non-effective statutory protection (APAs, marine land, paleontological sites, etc.), and 34% comprise public or private areas with no protection.

Abstract

Background Inactivated poliovirus vaccine (IPV) alone does not induce mucosal immunity. However, it was hypothesized that administration of IPV together with bivalent (types 1+3) oral poliovirus vaccine (bOPV) may stimulate mucosal cross-immunity to poliovirus type 2 (PV2). Methods Cuban infants were randomized to receive either one dose of IPV (Arm A); one dose of IPV with bOPV (Arm B) at about 6 months of age or no vaccine (Arm C). Subjects were challenged with one dose of trivalent OPV (tOPV); they were about 7 months old in arms A and B, and about 3 months old in arm C at a time of the tOPV challenge. Sera were collected before vaccination and 30 days after tOPV challenge and tested for presence of poliovirus neutralizing antibodies; stool samples were collected at days 0, 7, 14, 21 and 49 post-challenge and tested for presence of poliovirus. Results We enrolled 333 children. Excretion of PV2 following tOPV challenge was highest on day 7 (75 [CI 95% = 65-82%], 68 [CI 95% = 58-75%] and 73 [CI 95% = 63-80%] for study arms A, B, and C respectively); excretion decreased with every subsequent stool sampling; no significant differences either in proportion of PV2 excretion or in its duration were observed between study arms. Conclusions There was no reduction in excretion of PV2 after tOPV challenge in children who had received IPV with bOPV when compared to those who had received IPV alone or no vaccine. Polio eradication program cannot assume any PV2 mucosal response with the current polio immunization schedule.
