Anaerobic digestion (AD) of organic fraction of municipal solid waste (OFMSW) is prone to system breakdown under high organic loading rates (OLRs) condition, which subsequently reduces the efficiency of digestion process and results in substantial economic losses. In this perspective paper, the substances metabolisms, electrons flow, as well as microbial interaction mechanisms within AD process are comprehensively discussed, and the underlying bottleneck that causes inefficient methane production is identified, which is “electrons surplus”. Systems encountering severe electron surplus are at risk of process failure, making it crucial to proactively prevent this phenomenon through appropriate approaches. On this basis, the present perspective proposes three potential electronic regulation strategies to prevent electrons surplus, namely, electron shunt, accelerating electron transfer and regulating methanogenic metabolism pathway, and presents specific methodologies for each strategy. Furthermore, the potential solutions to challenges that may occur during the electronic regulation process are also presented in this paper. This perspective aims to provide innovative approaches to achieve the efficient and stable operation of OFMSW anaerobic digestion, especially under high OLRs condition.[Figure not available: see fulltext.].

Bioaugmentation has been recognized as a key strategy to improve the anaerobic digestion efficiency in organic waste treatment. Methanosarcina barkeri possesses direct interspecies electron transfer capability, a characteristic that allows it to outcompete other unwanted species such as sulfate-reducing bacteria. This study investigated the effects of bioaugmentation with Methanosarcina barkeri DSM800 on two continuous-stirred tank reactors fed with a sulfate-rich feedstock. One of the two reactors was supplemented with magnetite to facilitate direct interspecies electron transfer. Time series quantitative polymerase chain reactions were performed to evaluate the absolute abundance of crucial species, including the augmented Methanosarcina. Results showed increased and stabilized methane production of 22% and 21% in the reactor amended with magnetite and in the control reactor, respectively. Moreover, volatile fatty acids were almost completely consumed in the magnetite-supplemented reactor. The quantitative polymerase chain reaction was used to analyze the abundance of targeted species in response to bioaugmentation. Specifically, Methanosarcina barkeri was not retained in either reactor after one hydraulic retention time. Direct interspecies electron transfer-associated microorganisms showed opposite trends in the two reactors, highlighting the different interactions with Methanosarcina barkeri in the presence and absence of magnetite. Sulfate-reducing bacteria following the dissimilatory sulfate reduction pathway exhibited an opposite behavior in the reactor amended with magnetite, in contrast to those employing the assimilatory sulfate reduction pathway. Overall, the study demonstrated that bioaugmentation with exogenous archaea can considerably alter the microbial community, but the introduced species is not able to establish itself in a stable microbiome. It is known that the presence of sulfate decreases the methane yield in the anaerobic digestion systems. Sulfate-reducing bacteria can convert sulfate to hydrogen sulfide competing with methanogens for substrates such as H2 and acetate. The present work aims to elucidate the microbial interactions in biogas production and assess the effectiveness of electron-conductive materials in restoring methane production after exposure to high sulfate concentrations. The addition of magnetite led to a higher methane content in the biogas and a sharp decrease in the level of hydrogen sulfide, indicating its beneficial effects. Furthermore, the rate of volatile fatty acid consumption increased, especially for butyrate, propionate, and acetate. Genome-centric metagenomics was performed to explore the main microbial interactions. The interaction between methanogens and sulfate-reducing bacteria was found to be both competitive and cooperative, depending on the methanogenic class. Microbial species assigned to the Methanosarcina genus increased in relative abundance after magnetite addition together with the butyrate oxidizing syntrophic partners, in particular belonging to the Syntrophomonas genus. Additionally, Ruminococcus sp. DTU98 and other species assigned to the Chloroflexi phylum were positively correlated to the presence of sulfate-reducing bacteria, suggesting DIET-based interactions. In conclusion, this study provides new insights into the application of magnetite to enhance the anaerobic digestion performance by removing hydrogen sulfide, fostering DIET-based syntrophic microbial interactions, and unraveling the intricate interplay of competitive and cooperative interactions between methanogens and sulfate-reducing bacteria, influenced by the specific methanogenic group.

Four different types of ceramic gas distributors (Al2O3 of 1.2 μm and SiC of 0.5, 7 and 14 μm) were evaluated to increase biomethane formation during ex-situ biogas upgrading process. Each type of gas diffuser was tested independently at three different gas retention times of 10, 5 and 2.5 h, at thermophilic conditions. CH4 production rate increased by increasing input gas flow rate for all type of distributors, whereas CH4 concentration declined. Reactors equipped with SiC gas distributors effectively improved biomethane content fulfilling natural gas standards. Microbial analysis showed high abundance of hydrogenotrophic methanogens and proliferated syntrophic bacteria, i.e. syntrophic acetate oxidizers and homoacetogens, confirming the effect of H2 to alternate anaerobic digestion microbiome and enhance hydrogenotrophic methanogenesis. A detailed anaerobic bioconversion model was adapted to simulate the operation of the R1-R4 reactors. The model was shown to be effective for the simulation of biogas upgrading process in up-flow reactors.

Sufficient quantity of trace metals is essential for a well performing anaerobic digestion (AD) process. Among the essential trace elements in active sites of multiple important enzymes for AD are iron and nickel ions. In the present study, iron and nickel in the form of Fe2O3 and NiO were coated on TiO2 nanoparticles to be used in batch and continuous operation mode. The effect of TiO2, Fe2O3–TiO2, and NiO–TiO2 nanoparticles on each step of AD process was assessed utilizing simple substrates (i.e. cellulose, glucose, acetic acid, and mixture of H2–CO2) as well as complex ones (i.e. municipal biopulp). The hydrolysis rate of cellulose substrate increased with higher dosages of the coated TiO2 with both metals. For instance, the hydrolysis rate was increased up to 54% at Fe2O3–TiO2 and at a concentration of 23.5 mg/L for NiO–TiO2 it was increased up to 58%, while higher dosage suppressed the hydrolytic activity. Experimental results revealed that low dosages of NiO–TiO2 increased the accumulated methane production up to 24% probably by increasing the enzymatic activity of acetoclastic methanogenesis. Anaerobic digestion (AD) is a green technology that has been applied for many years. One of the main problems in this process is controlling these bioreactors to maximize methane production. A supervisory control strategy has been proposed to improve the methane production rate in an anaerobic digestion process while minimizing the risk of process failure in the presence of several drastic feedstock changes. The inner loop consisted of a feedback control that manipulated the feed flow rate for achieving the desirable methane production rate. A rule based control strategy was used as supervisory control loop. This controller received the total volatile fatty acids concentration in the reactor and the trends of methane production rate to calculate the set-point of the inner control loop. pH was used as a safety control parameter to prevent the system from acidification. The proposed control scheme was tested in the presence of several disturbances in the feedstock, including glucose overload, ammonia inhibition, substrate dilution and shifting feeding from manure to bio-pulp. The controller prevented VFA accumulation above 2.73, 2.94 and 4.09 g L−1, under glucose overload, ammonia inhibition and change of feedstock to bio-pulp, respectively. Thus, the supervisory control could successfully improve the methane production rate and keep the reactor stable against external severe disturbances.
