The career transition of lecturers and education staff has brought several organizational role changes, resulting in difficulty adapting and overcoming the impacts that arise from career transitions. The purpose of the study is to explore the adaptation experience and impact of the career transition of lecturers and education staff in Indonesia. This research uses qualitative methods with a grounded theory approach. Informants are recruited using the snowball sampling method. The number of informants interviewed was 18 people from 7 state universities in Indonesia. The results of the interview were coded with NVIVO software. The analysis results indicate that the consequences of a career transition consist of two phases, namely: adaptation and change, consisting of 23 dimensions and 50 indicators. The adaptation phase consists of 4 main categories, namely situation, self, support, and strategies. Schlossberg's Transition Theory can explain the adaptation process. The change phase includes four categories: behavior, role, learning, and perception. The findings show that during the change phase, informants use two career anchors, in contrast to Schein's Anchor Career Theory which states that individuals only use one main career anchor. This research enriches the theory of career transition in the context of higher education institutions in Indonesia.

This research focused on synthesizing CuO@NiO nanocomposites using a co-precipitation method to explore their antimicrobial, antioxidant, and melamine-sensing capabilities. Nickel oxide (NiO), known for its stability and optical properties, was doped with copper oxide (CuO) to enhance its performance. The synthesized nanocomposites showed strong antioxidant activity, achieving 72% inhibition for 2% CuO@NiO and 80% inhibition for 5% CuO@NiO, as determined by the DPPH assay. Antimicrobial tests demonstrated significant inhibition of both Gram-positive and Gram-negative bacteria, particularly with the 5% CuO@NiO composite. Electrochemical analysis, including cyclic voltammetry and differential pulse voltammetry, revealed high sensitivity for melamine detection, with a detection limit of 0.01 µM. These results suggest that CuO@NiO nanocomposites hold promise for applications in antioxidant therapies, antimicrobial treatments, and chemical sensing.

Abstract Background: Lysyl oxidase (LOX) family proteins have recently become a topic in cancer progression. Our recent study found a high expression of LOX-like 4 (LOXL4) in MDA-MB-231 cells. Objective: To reveal the impact of depleted LOXL4 in both in vitro and in vivo breast cancer models from a histological perspective. Material and Method: Endogenous LOXL4 was depleted using the CRISPR/Cas9 on MDA-MB-231 parental cells. Based on the LOXL4 protein expression, the clone was determined for the next experiment, thus generating MDA-MB-231 LOXL4 KO. Cell assay was conducted using colony formation assay (n=3) followed by crystal violet staining. The indicated cells were inoculated orthotopically to female BALB/c nude mice (n=5). At the end of the experiment, tumors were isolated, fixed, and prepared for Masson Trichrome staining. Result: CRISPR/Cas9 completely depleted LOXL4 expression on clone number #2-22. Background: Oral submucous fibrosis (OSMF) is an insidious chronic inflammatory progressive disease and one of the potentially malignant disorders, predominantly affecting the oral cavity. Objective: The current study aimed to compare the efficacy of curcumin and aloe vera along with ultrasound in the management of OSMF. Materials and Methods: In this intervention study, 40 participants with OSMF were allocated into two groups. Group A included 20 patients who received curcumin (curenext) oral gel for 3 months along with ultrasound therapy. Group B included 20 patients who received aloe vera gel (forever living gel) along with ultrasound therapy. Results: Following therapies, patients in the group A showed clinically significant improvements when compared to group B. Conclusion: The combined therapy of curcumin and ultrasound shows potential in managing OSMF by effectively alleviating clinical symptoms without any reported side effects. It is advised that future studies employ larger samples and longer treatment follow-up.

This prospective study investigated the association between lipoprotein (a) [Lp(a)] levels and adverse cardiac events in patients undergoing percutaneous coronary intervention (PCI) for coronary artery disease. Among 600 patients, 79.16 % were male. Kaplan Meier analysis revealed significantly higher incidence rates of cardiac death, major adverse cardiac events, myocardial infarction, revascularization and stroke in patients with elevated Lp(a) (≥30 mg/dL). Programmed ferroptotic death eliminates cells in all major organs and tissues with imbalanced redox metabolism due to overwhelming iron-catalyzed lipid peroxidation under insufficient control by thiols (Glutathione (GSH)). Ferroptosis has been associated with the pathogenesis of major chronic degenerative diseases and acute injuries of the brain, cardiovascular system, liver, kidneys, and other organs, and its manipulation offers a promising new strategy for anticancer therapy. This explains the high interest in designing new small-molecule-specific inhibitors against ferroptosis. Given the role of 15-lipoxygenase (15LOX) association with phosphatidylethanolamine (PE)-binding protein 1 (PEBP1) in initiating ferroptosis-specific peroxidation of polyunsaturated PE, we propose a strategy of discovering antiferroptotic agents as inhibitors of the 15LOX/PEBP1 catalytic complex rather than 15LOX alone. Here we designed, synthesized, and tested a customized library of 26 compounds using biochemical, molecular, and cell biology models along with redox lipidomic and computational analyses. We selected two lead compounds, FerroLOXIN-1 and 2, which effectively suppressed ferroptosis in vitro and in vivo without affecting the biosynthesis of pro-/anti-inflammatory lipid mediators in vivo. The effectiveness of these lead compounds is not due to radical scavenging or iron-chelation but results from their specific mechanisms of interaction with the 15LOX-2/PEBP1 complex, which either alters the binding pose of the substrate [eicosatetraenoyl-PE (ETE-PE)] in a nonproductive way or blocks the predominant oxygen channel thus preventing the catalysis of ETE-PE peroxidation. Our successful strategy may be adapted to the design of additional chemical libraries to reveal new ferroptosis-targeting therapeutic modalities.

As the development of green tribology in marine industries has gained traction, there has been a gradual shift in the utilization of water-lubricated bearings over oil lubrication in stern tube bearings. Water lubricants are preferred in marine applications because of their availability, non-compressibility, and better frictional and cooling properties. In this study, a computational fluid dynamics (CFD) approach was employed to simulate the water film pressure distribution and additional water film characteristics to analyze the performance of water-lubricated bearings (WLB) under various operational conditions. The design parameters of the WLB were obtained from a commercially available axial groove bearing. To enhance the current bearing design and operational capability, it is imperative to understand the behavior of water-lubricated bearings through numerical techniques. An 8-axial V shaped groove structure with L/D ratios of 1, 1.5, and 2 was considered for simulating WLB behavior at four different journal speeds. The numerical results indicate that a notable increase in the maximum water film pressure and bearing load capacity was observed for WLBs simulated at higher operating speeds. At 4000 RPM, an increase of approximately 2.17% in peak positive pressures was noted for long WLBs than for lower speeds. In the groove domain, the water flow dynamics are considerably complex. A certain quantity of lubricant inflow descends into the groove regions and collides with the groove boundaries, resulting in a recirculation flow and vortices. The simulation results provide guidance for improving groove designs to further enhance the lubrication characteristics of axially grooved WLB.

Photon-driven technologies encompass the utility of carefully engineered materials for myriad applications. Over the past decade, thin films of II–VI chalcogenide materials have emerged as versatile candidates for the efficient construction of photovoltaic devices. The buffer layer which serves as a lattice fixer in solar cells plays a role in determining the photon-driven processes between the absorber and the window layers, significantly affecting the performance of the working device. However, mostly used buffer layer, CdS is highly toxic. Recently, there is quest to replace the highly toxic cadmium sulfide (CdS) buffer layers with Zn-based materials to obtain high-efficiency solar cells. Among the II–VI compounds, zinc sulfide (ZnS) and zinc selenide (ZnSe) present promising futuristic scope for research in the photovoltaics field and are recognized as alternatives to the widely used toxic CdS buffer layer. Although significant research has been devoted in this direction, a comprehensive review capturing the latest advancements in photon-driven technologies pertaining to Zn based buffer layers in photovoltaics, has not yet been conducted. In light of these observations, we present a review focusing on the recent progress in the incorporation of Zn based buffer layers into different solar cells constituting CdTe, CIGS, and CZTS. The light matter interactions that depend on the thickness, and the band alignment of the buffer layer with the absorber is discussed in detail.

Machining of Incoloy 800 is challenging, which can be attributed to the low thermal conductivity, hot strength, accord for insert material and propensity to work harder. This study evaluates the influence of carbide tool-insert wear (rake, flank) and surface roughness (Ra) under process variables (cutting speed, feed rate and depth of cut) in high-speed dry turning of Incoloy 800. The essence of chip formation in Incoloy 800 in the context of coolant-less turning operation was also investigated. The turning experiments were performed in relation to Taguchi’s experimental design, with three factors with three levels: cutting speed (175, 200 and 225 m/min), feed rate (0.15, 0.2 and 0.25 mm/rev) and depth of cut (0.6, 0.8 and 1 mm). The experimental observations confirm that abrasion, adhesion and diffusion are the prime tool wear mechanisms responsible for tool failure. The formation of built-up edge (BUE) during the turning process was predominant, and excess BUE was observed at higher cutting parameters. The crater formation was found to be more severe at higher cutting conditions. The cutting speed has a firm hold on tool wear and surface roughness. The variation of surface roughness showed almost a cyclic trend and was found to decrease with an increase in the nose radius.

This study aims to set up a data-driven framework for optimization and performance prediction of a nanolubricant. Isoviscous approximation was applied to simplify analytical results. FDM methods were used to generate 1204 data points. A multilayer perceptron (MLP) was trained using this large dataset to achieve high testing accuracy (R = 1). The least mean squared error was found at 987 epochs for 23 hidden layer neurons. Multiple statistical tools were used to analyze the results. The error values in the load prediction (up to 7.34%) were observed to be higher than the friction coefficient (up to 0.041%) and side leakage (up to 0.05%). The JAYA algorithm, a parameter-independent machine learning algorithm was used for zone-specific optimization with the zones being low load region (eccentricity ratio: 0.1 to 0.3), medium load region (eccentricity ratio: 0.3–0.6), high load region (eccentricity ratio: 0.6–0.95) and whole range. The optimized combinations for low, medium, high and whole range of operation gave load capacity of 91 N, 357 N, 1328 N and 124 N, respectively. The optimized results highlight the tendency to minimize the eccentricity ratio and maximize volume fraction and aggregate particle size ratio.

This research is centered around the development and construction of a test apparatus for a counterflow evaporative cooler. The packing material utilized in the testing is coconut coir with wettability values of 300, 400 and 500 m2/m3. The experiments involve varying the air velocity. The study examined performance characteristics, including evaporation rates, cooler efficiency, coefficient of performance (COP) and energy consumption information, in relation to the standard Celdek 7090 packaging. The findings demonstrated that employing a coconut coir with optimal wettability resulted in a cooling efficiency and COP of 53.57% and 1.25, respectively. These values were slightly lower than those achieved with the usual Celdek 7090 packing. Coconut coir, as a biomass-derived material, possesses certain benefits that make it a suitable alternative for filling materials in evaporative cooling processes.

Precision machine tool structures require enhanced stability with higher static stiffness and increased damping. Granite epoxy (G-E) composite offers improved damping properties and is a promising alternative material. This study evaluates the mechanical strength and damping of G-E hybrid polymer composites with varying percentages of cast iron (CI) filler. Experimental and finite element (FE) analyses were conducted on hybrid composite samples incorporating 5, 10 and 15 wt% CI filler particles. Results show that a 70:30 ratio of G-E with 10% CI filler enhances compressive strength. Tensile and flexural behaviors are influenced by epoxy content, with minimal effects from CI filler. Both experimental and numerical simulations show significant improvement in stiffness and damping properties with higher quantities of granite particulates (80 wt%) and 5 wt% CI filler. Modal analysis confirms higher natural frequencies in all modes. This hybrid composite configuration presents a viable alternative for machine tool structures, offering increased structural damping.

Machining of Inconel 800 superalloy is associated with inherent issues like lower tool life and lower quality of machined surface owing to the work-hardening nature of the superalloy and increased mechanical and thermal stresses. The employment of cutting fluid in machining negatively affects the locale. Hence, dry machining is a feasible alternate solution. This work aims to optimize the cutting parameters (CP): cutting speed, feed rate and depth of cut in the high-speed dry turning of Inconel 800 employing an uncoated carbide insert to minimize the responses: tool wear (TW) and surface roughness (SR). The superalloy machining is conducted as per the experimental runs designed applying the Taguchi analysis. Then, the effects and contributions of the CP on the outputs were examined employing the signal-to-noise (S/N) ratio and the analysis of variance (ANOVA). Additionally, the multi-objective optimization (MOO) method grey relational analysis was employed to optimize CP. The results of the research work showed that cutting speed, feed rate, and depth of cut have noteworthy sway on TW and SR with a % contribution of 33.3, 13.8 and 23.7, respectively. Additionally, evaluation of SEM images of the cutting insert revealed that the abrasion, adhesion and diffusion are the primary wear mechanisms leading to abrasion groves, crater, chipping, built-up-edge and notch formation.

Background: Cardiovascular diseases (CVDs) are a leading cause of global mortality, motivating research into novel approaches for their management. Lipoprotein(a) (Lp(a)), a unique lipoprotein particle, has been implicated in atherosclerosis and thrombosis, suggesting its potential as a therapeutic target for CVDs. Aim: This study aimed to investigate the association of Lp(a) levels with various cardiovascular parameters and events among patients with confirmed cardiovascular disease. Methodology: A prospective study was conducted, enrolling 600 participants, predominantly comprising males (79%), with a mean age of 52.78 ± 0.412 years diagnosed with cardiovascular disease. The follow-up was done for 18 months. Patient demographics, blood investigations, and occurrence of major adverse cardiac events (MACE) were collected. SPSS version 21 was used to statistically analyze the relationships between elevated Lp(a) levels and factors such as age, glycated hemoglobin, mortality, MACE, cardiac death, target vessel revascularization, and stroke. Results: The study revealed significant (P < 0.05) associations between elevated Lp(a) levels and advanced age, increased glycated hemoglobin levels, as well as occurrences of all-cause mortality, MACE, cardiac death, target vessel revascularization, and stroke. Notably, a significant (P < 0.05), association between high Lp(a) levels and acute coronary syndrome (ACS) emerged, suggesting Lp(a)'s role in advanced cardiac events. Conclusion: The findings highlight the potential significance of Lp(a) as a notable risk factor in cardiovascular health. The observed associations between elevated Lp(a) and adverse cardiovascular events, including ACS, underscore its pathogenic role. Consequently, this study supports the rationale for further research into Lp(a)-specific therapeutic interventions, offering substantial promise in refining the management strategies for cardiovascular diseases.

Autism Spectrum Disorder (ASD) ialah sebuah bagian dari gangguan neurodevelopmental yang dicirikan melalui defisit pada komunikasi, interaksi sosial, pola perilaku atau keterbatsan aktivitas dan repetitif. ASD sedikit banyaknya memberikan disfungsi bagi kehidupan penyandangnya. Model Biopsikososial dapat membantu dalam membahas secara komprehensif dinamika psikologis dan faktor penyebab individu mengalami ASD. Penelitian ini memanfaatkan metode kualitatif melalui pendekatan studi kasus. Subjek pada penelitian ini ialah seorang anak laki-laki berusia 8 tahun yang mengidap ASD. Pengambilan data dilakukan melalui beberapa asesmen, yakni observasi, wawancara (orang tua dan guru), serta pemeriksaan psikologis. Adapun pemeriksaan psikologis yang diberikan adalah Vineland Social Maturity Scale (VSMS), Child Behavior Checklist (CBCL), Childhood Autism Rating Scale (CARS) dan Observasi terstruktur menggunakan Developmental Milestone Chart. Secara keseluruhan, penelitian ini dapat menggambarkan dinamika psikologis individu yang mengalami ASD. Tidak hanya penggambaran secara biologis, psikologis dan sosial, namun juga dijelaskan menggunakan area-area (predisposing, precipitating dan perpetuating) yang dapat menguraikan perjalanan gangguan pada ASD secara lebih komprehensif. Hasil penelitian juga menemukan bahwa peran keterlibatan orang tua sangat signifikan terhadap perkembangan positif anak dengan ASD, baik stimulasi maupun imitasi penerapan terapi di rumah.

Hyperemesis Gravidarum (HG) is a pregnancy complication that is often overlooked as it is typically considered normal. If HG is not properly treated, nutrition will not be fulfilled which can negatively affect maternal and fetal health and even maternal and fetal death. The exact cause of HG is not identified, so there are no effective preventive methods. However early detection can help for prompt and appropriate treatment. Therefore, a monitoring system for pregnancy conditions was designed for HG early detection. This system employs the MPX5050 DP pressure sensor for measuring blood pressure, the MAX30100 for assessing maternal heart rate and oxygen saturation, the MAX4466 sensor for monitoring fetal heart rate, and an expert system using the certainty factor method to diagnose the probability of hyperemesis gravidarum. The expert system achieves an accuracy of 93.33%. In comparison to the aneroid sphygmomanometer, the designed sphygmomanometer reveals a mean difference of 3.5 mmHg for diastolic pressure, with a standard deviation below 8 mmHg for both systolic and diastolic pressures. The measurement of heart rate and oxygen saturation has a deviation of 1.8 % and 1.02 % respectively. These deviations align with the standards specified by the Ministry of Health for medical devices. For the fetal heart rate, the mean deviation is 3.4 bpm, and the measurement error is 2.38%. Thus, this system can be utilized to monitor pregnancy conditions, enabling the early detection of hyperemesis gravidarum

While the prevailing treatment for corneal abrasions outside the athletic sphere is the application of a bandage contact lens (BCL), which lessens pain and allows for the maintenance of binocular vision, this is not the case during athletic play. This brief report highlights the advantage of BCLs in treating in-game corneal abrasions, ultimately allowing for an immediate return to play. Additionally, this report summarizes the mechanisms of bandage contact lenses, differentiates them from standard hydrogel contact lenses, and highlights the significant steps necessary to apply the bandage contact lens during an in-game corneal abrasion event. Overall, we link modern ophthalmology clinical practice and sports medicine, allowing for the attenuation of acutely-induced ocular pain to a manageable state.

FKIP Kindergarten is a Kindergarten with Accreditation B, and its address is at Jl. Inong Balee, PPG Dormitory, Kopelma Darussalam Village, Syiah Kuala District, Banda Aceh City, Aceh Province. The number of students in Kindergarten FKIP is 148 students and 16 teachers. FKIP Kindergarten has games in the schoolyard, but the focus of the games is more dominant on improving motor skills so that the increase in affective abilities becomes less honed. This dedication focuses on building children's character and increasing numeracy literacy through fun things. The method used in this service program is the direct instruction method, namely the service team consisting of 3 lecturers and 2 students assisted by partners, namely kindergarten teachers. With the giant snakes and ladders game, kindergarten students get an emphasis on character, literacy, and numeracy through games, so that character emphasis becomes easier to apply. The snakes and ladders provided have been specifically designed with additional characters and literacy and numeracy materials.

Persoalan premanisme pemalakan disertai kekerasan memenuhi kasus tindak pidana. Banyak kasus menganai hal tersebut di Semarang. Hal tersebut jelas melanggar KUHP. Diperlukan penanganan yang efektif terkait tindak pidana tersebut. Penelitian yuridis normatif ini menggunakan metode deskriptif analisis. Analisis data akan menggabungkan pendekatan kualitatif dengan analisis sekunder dari kasus-kasus dan pasal terkait tindak pidana premanisme. Hail penelitian menunjukan bahwa Tindak pidana premanisme pemalakan disertai dengan kekerasan merupakan permasalahan serius yang perlu ditangani dengan efektif oleh Polrestabes Semarang dan banyak kendala yang dihadapi dalam mewujudkannya diantaranya kendala internal, eksternal, serta faktor masyarakat.

In this investigation, Blue Cherry fruit was used to isolate potential probiotic isolate and characterized for their beneficial attributes. The main objective of the study was to evaluate the antagonistic activity of the isolated bacteria against plant pathogenic bacteria such as Ralstonia solanacearum, Pectobacterium carotovorum sub sp. carotovorum and Xanthomonas oryzae pv oryzae and also to evaluate the isolate for potential probiotic, plant growth promoting and biocontrol activity. A total of nine isolates were isolated from Blue Cherry fruit and initially screened for microscopic and biochemical characters. The agar-well diffusion method, paper disc, spot inoculation and microplate method were used to test the antibacterial activity from the cultured supernatants against plant pathogenic bacteria and zone of inhibition ranged from 0.6 to 2.1 cm and per cent inhibition of plant pathogenic bacteria ranged from 64.81 to 71.70 %. Effective isolate MYSBC14 was genotyped based on 16S rDNA and identified as Enterobacter faecium OL454757. Effective isolate E. faecium MYSBC14 was further characterized for probiotic properties, PGPR and bio-control attributes. MYSBC14 survived under pH 2 and pH 4 after 2 and 4 h after incubation (38.08 and 23.16 %, 87.90 to 82.02 % respectively) and was susceptible to majority of antibiotics evaluated. A survival rate of 52.67 and 43.01 % was recorded at 0.3 % oxgall salt concentration after 2 and 4 h after incubation. The cell surface hydrophobicity of 90.24 % and auto aggregation of 8.10 to 22.02 % represented the adhesion properties of the isolate. MYSBC14 showed indole acetic acid production (11.56 µg/mL), gibberellic acid production (10.18 µg/mL) and gave positive results for solubilization of phosphate, potash, protease and siderophore production and was found negative for zinc and silicon solubilization, cellulose degradation activity, HCN production activity and amylase activity. The findings of the present investigation showed that isolated probiotic organism possesses promising antibacterial, probiotic attributes, plant growth promoting characteristics and bio-control traits.

Tea plantation biomass wastes, such as tea plantation pruning, shade trees, and woody weeds have not been utilized. The waste can be used as renewable energy in the form of wood pellets. The problem is the feasibility of biomass waste to be used as material for making wood pellets as energy. This paper aims to analyze the potential of tea plantation biomass waste as wood pellet material to meet energy needs. The research was conducted in a tea plantation owned by the Tea and Kina Research Center (PPTK). Quantification of biomass waste potential per unit area was conducted in the plantation using direct measurement method. Proximate analysis of each wood pellet variant of biomass waste was conducted to match the quality of Indonesian Wood Pellet Standard. The biomass waste potential in PPTK is 14,281 tons per year which can produce 8,186 tons of wood pellets per year. This potential can meet the needs of wood pellet consumption from the tea production process at PPTK which is around 1.8 tons / day for the tea processing process of 13 tons/day. Based on proximate analysis, the wood pellets produced have a calorific value of 4425 cal/gram, density of 1.35 grams/cm3, fixed carbon content of 85.2%, and volatile matter of 3.72%. These results confirm that the wood pellets comply with the National Wood Pellet Standard (SNI 8021:2014). This can be a model for the application of the Green Circular Economy concept in the plantation sector.

Abstract UBA1 is the primary E1 ubiquitin-activating enzyme responsible for generation of activated ubiquitin required for ubiquitination, a process that regulates stability and function of numerous proteins. Decreased or insufficient ubiquitination can cause or drive aging and many diseases. Therefore, a small-molecule enhancing UBA1 activity could have broad therapeutic potential. Here we report that auranofin, a drug approved for the treatment of rheumatoid arthritis, is a potent UBA1 activity enhancer. Auranofin binds to the UBA1’s ubiquitin fold domain and conjugates to Cys1039 residue. The binding enhances UBA1 interactions with at least 20 different E2 ubiquitin-conjugating enzymes, facilitating ubiquitin charging to E2 and increasing the activities of seven representative E3s in vitro. Auranofin promotes ubiquitination and degradation of misfolded ER proteins during ER-associated degradation in cells at low nanomolar concentrations. It also facilitates outer mitochondrial membrane-associated degradation. These findings suggest that auranofin can serve as a much-needed tool for UBA1 research and therapeutic exploration.
