RATIONALE: The ability to recover the isotopic signature of water added to soil samples that have previously been ovendried decreases with the increasing presence of silt and clay. The effects on the isotopic signature of water associated with physicochemical soil properties are not yet fully understood, for either hydration or dehydration of soil samples. METHODS: The soil sample chemistry and the crystallinity of minerals were measured by X-ray fluorescence and X-ray diffraction. The organic carbon and the cation-exchange capacity were also determined. Water of known isotopic signature was used to spike an oven-dried substrate and subsequently extracted by cryogenic vacuum extraction at a temperature of 105°C. In addition, the soils were oven-dried at 205°C and water extractions were also performed at 205°C. The isotopic signatures of the water samples were determined by cavity-ring-down spectrometry. RESULTS: The isotope effects caused by the cryogenic vacuum extraction method applied to soils with elevated clay content were reduced. First, by increasing the extraction temperature to 205°C, we improved the precision of the cryogenic vacuum extraction method and the recovery of the known isotopic signature of the spike water. Secondly, the postcorrection of data based on the physicochemical soil properties and a common extraction temperature of 105°C improved the measurement trueness. CONCLUSIONS: The isotopic signature of soil water is influenced by mineral-water interaction. During the hydration of clay, different minerals deplete free water in heavy isotopes. The extracted soil water (dehydration water) gathered from clay-rich soils is generally more depleted in the heavy isotopes than the spike water, making results obtained for different soil types difficult to compare. Stable isotopes (deuterium, ²H, and oxygen-18, 18O) of soil pore water were measured directly in the field using tunable off-axis integrated cavity output spectroscopy (OA-ICOS) and commercially available soil gas probes in a semi-arid region of the Cuvelai-Etosha-Basin, Namibia. High spatial and temporal resolution was achieved in the study area with reasonable accuracy and measurements were in agreement with laboratory-based cryogenic vacuum extraction and subsequent cavity ring down laser spectroscopic isotope analysis (CRDS). After drift correction of the isotope data, mean precision for over 140 measurements of two consecutive field campaigns in June and November 2014 were 1.8 and 0.46 ‰ for δ²H and 18O, respectively. Mean Accuracy using quality check standards was 5 and 0.3 ‰ for δ²H and δ18O, respectively. Results support the applicability of an in-situ measurement system for the determination of stable isotopes in soil pore water. Spatio-temporal variability could be deduced with the observed data in an extremely dry evaporation dominated environment which was sporadically affected by intermittent rainfall. Since 1973, Kunene River water has been carried from the Calueque reservoir in Angola along a 160 km open concrete canal to the town of Oshakati in the central part of the Cuvelai-Etosha Basin and has been supplying drinking water to the most densely populated rural area of Namibia. Despite its importance for the region, intra-seasonal water quality and the technical condition of the canal are not routinely checked. Water samples were collected during four field campaigns right before the onset of the rainy season (November 2013 and 2014), and after the rainy season (June 2014 and May 2015), at 16 sites along the canal for stable water isotopes (deuterium, oxygen-17 and oxygen-18) and hydrochemical analyses. The isotope patterns and chemical composition of the canal water is discussed in comparison to local rain, Kunene source water, surface water and groundwater. Clear isotope enrichment indicates evaporative loss of water. A Craig-Gordon model was used to estimate water loss. The loss increases with distance from the source with a maximum of up to 10 %, depending on the season. The results are discussed in context of water availability, vulnerability and water resources management in this water-scarce area.

Rationale: The stable isotope compositions of hydrogen and oxygen in water (δ 2 H and δ 18 O values) have been widely used to investigate plant water sources, but traditional isotopic measurements of plant waters are expensive and labor intensive. Recent work with direct vapor equilibration (DVE) on laser spectroscopy has shown potential to side step limitations imposed by traditional methods. Here, we evaluate DVE analysis of plants with a focus on spectral contamination introduced by organic compounds. We present 17 O-excess as a way of quantifying organic compound interference in DVE., Methods: We performed isotopic analysis using the δ 2 H, δ 18 O and δ 17 O values of water on an Off-Axis Integrated Cavity Output Spectroscopy (IWA-45EP OA-ICOS) instrument in vapor mode. We used a set of methanol (MeOH) and ethanol (EtOH) solutions to assess errors in isotope measurements. We evaluated how organic compounds affect the 17 O-excess. DVE was used to measure the isotopic signatures in natural plant material from Pinus banksiana, Picea mariana, and Larix laricina, and soil from boreal forest for comparison with solutions., Results: The 17 O-excess was sensitive to the presence of organic compounds in water. 17 O-excess changed proportionally to the concentration of MeOH per volume of water, resulting in positive values, while EtOH solutions resulted in smaller changes in the 17 O-excess. Soil samples did not show any spectral contamination. Plant samples were spectrally contaminated on the narrow-band and were enriched in 1 H and 16 O compared with source water. L. laricina was the only species that did not show any evidence of spectral contamination. Xylem samples that were spectrally contaminated had positive 17 O-excess values., Conclusions: 17 O-excess can be a useful tool to identify spectral contamination and improve DVE plant and soil analysis in the laboratory and in situ. The 17 O-excess flagged the presence of MeOH and EtOH. Adding measurement of δ 17 O values to traditional measurement of δ 2 H and δ 18 O values may shed new light on plant water analysis for source mixing dynamics using DVE., (© 2019 John Wiley & Sons, Ltd.)

The study area is the Namibian part of the Cuvelai-Etosha Basin (CEB), located in central northern Namibia. The CEB is home to 40 % of Namibia's population, and most of the people live in rural areas. These people depend on both surface and groundwater resources which are limited in this dryland (mean annual rainfall ranging from 250 to 550 mm/a). The isotopic signatures of δ 18 O and δ 2 H from water samples (n = 61) collected over a course of 9 years from various research projects and existing (but mainly unpublished) data of meteoric water of the CEB (10 sites) were evaluated and local meteoric water lines (LMWLs) developed. Further, the data is discussed in the context of seasonal characteristics and trends and compared to available data from the Global Network of Isotopes in Precipitation (GNIP) for the southern African region. Our results extend the portfolio of previously published LMWLs for southern Africa and provide a more precise baseline for any isotope-based study in that region. The slope of the LMWL from the GNIP stations correlates with latitude. This correlation cannot be found within the CEB. The dominant control on the isotopic signature of the CEB of precipitation is seasonal.

TILLING (Targeting Induced Local Lesions IN Genomes) is a strategy used for functional analysis of genes that combines the classical mutagenesis and a rapid, high-throughput identification of mutations within a gene of interest. TILLING has been initially developed as a discovery platform for functional genomics, but soon it has become a valuable tool in development of desired alleles for crop breeding, alternative to transgenic approach. Here we present the Hor TILLUS ( Hor deum - TILL ING- U niversity of S ilesia) population created for spring barley cultivar "Sebastian" after double-treatment of seeds with two chemical mutagens: sodium azide (NaN 3 ) and N-methyl-N-nitrosourea (MNU). The population comprises more than 9,600 M 2 plants from which DNA was isolated, seeds harvested, vacuum-packed, and deposited in seed bank. M 3 progeny of 3,481 M 2 individuals was grown in the field and phenotyped. The screening for mutations was performed for 32 genes related to different aspects of plant growth and development. For each gene fragment, 3,072-6,912 M 2 plants were used for mutation identification using LI-COR sequencer. In total, 382 mutations were found in 182.2 Mb screened. The average mutation density in the Hor TILLUS, estimated as 1 mutation per 477 kb, is among the highest mutation densities reported for barley. The majority of mutations were G/C to A/T transitions, however about 8% transversions were also detected. Sixty-one percent of mutations found in coding regions were missense, 37.5% silent and 1.1% nonsense. In each gene, the missense mutations with a potential effect on protein function were identified. The Hor TILLUS platform is the largest of the TILLING populations reported for barley and best characterized. The population proved to be a useful tool, both in functional genomic studies and in forward selection of barley mutants with required phenotypic changes. We are constantly renewing the Hor TILLUS population, which makes it a permanent source of new mutations. We offer the usage of this valuable resource to the interested barley researchers on cooperative basis.

A hydrogeochemical and stable isotope study ( 2 H and 18 O) was carried out in the Cuvelai-Etosha Basin in order to characterize available groundwater and to identify possible recharge mechanisms for the perched aquifers. Data were collected during seven field campaigns between 2013 and 2015 from a total of 24 shallow and deep groundwater hand-dug wells. In the investigated groundwaters, hydrogencarbonate is the dominating anion in both well types, whereas cations vary between calcium and magnesium in deep wells, and sodium and potassium in shallow wells. Groundwater chemistry is controlled by dissolution of carbonate minerals, silicate weathering and ion exchange. Stable isotopic composition suggests that deep groundwater is recharged by high-intensity/large rainfall events, whereas the shallow wells can even be recharged by less-intense/small rainfall events. Water in deep wells reflect a mixture of water influenced by evaporation during or before infiltration and water that infiltrated through fast preferential pathways, whereas shallow wells are strongly influenced by evaporation. The findings of this research contribute to improve the understanding of hydrogeochemistry, recharge paths and temporal variations of perched aquifers.

The stable water isotope deuterium ((2)H) was applied as an artificial tracer ((2)H2O) in order to estimate groundwater recharge through the unsaturated zone and describe soil water movement in a semi-arid region of northern central Namibia. A particular focus of this study was to assess the spatiotemporal persistence of the tracer when applied in the field on a small scale under extreme climatic conditions and to propose a method to obtain estimates of recharge in data-scarce regions. At two natural sites that differ in vegetation cover, soil and geology, 500 ml of a 70% (2)H2O solution was irrigated onto water saturated plots. The displacement of the (2)H peak was analyzed 1 and 10 days after an artificial rain event of 20 mm as well as after the rainy season. Results show that it is possible to apply the peak displacement method for the estimation of groundwater recharge rates in semi-arid environments via deuterium labelling. Potential recharge for the rainy season 2013/2014 was calculated as 45 mm a(-1) at 5.6 m depth and 40 mm a(-1) at 0.9 m depth at the two studied sites, respectively. Under saturated conditions, the artificial rain events moved 2.1 and 0.5 m downwards, respectively. The tracer at the deep sand site (site 1) was found after the rainy season at 5.6 m depth, corresponding to a displacement of 3.2 m. This equals in an average travel velocity of 2.8 cm d(-1) during the rainy season at the first site. At the second location, the tracer peak was discovered at 0.9 m depth; displacement was found to be only 0.4 m equalling an average movement of 0.2 cm d(-1) through the unsaturated zone due to an underlying calcrete formation. Tracer recovery after one rainy season was found to be as low as 3.6% at site 1 and 1.9% at site 2. With an in situ measuring technique, a three-dimensional distribution of (2)H after the rainy season could be measured and visualized. This study comprises the first application of the peak displacement method using a deuterium labelling technique for the estimation of groundwater recharge in semi-arid regions. Deuterium proved to be a suitable tracer for studies within the soil-vegetation-atmosphere interface. The results of this study are relevant for the design of labelling experiments in the unsaturated zone of dry areas using (2)H2O as a tracer and obtaining estimations of groundwater recharge on a local scale. The presented methodology is particularly beneficial in data-scarce environments, where recharge pathways and mechanisms are poorly understood.

It has been pointed out that the meridional object and image foci as well as the meridional center of curvature will lie on the same straight line if the condition Delta (n cos(2)i/t) = Delta (n/t) is satisfied. The surfaces fulfilling this condition do not introduce astigmatism; hence they have been called minimal astigmatism surfaces. The generalized aplanatic points are defined and discussed, and the conditions for the incidence angle, which make it possible to replace the aspheric surfaces by spherical ones, are considered. A general method for the calculation of the minimal astigmatism surfaces is described.

In a paper probably to be published in Optika i Spektroskopiya the wave aberration for sagittal focus for the arbitrary surface of rotational symmetry has been carried out on the base of the astigmatic beam invariant D(s) = nu(s)d(s). The resulting expression for the wave aberration has been reformulated into three terms which, in the Seidel region, go over into astigmatism (the first) and into the Petzval curvature (the second) while the third disappears.
