This paper reports the behavior of 13X zeolite supported Ce, Cu, Co, Ag and Mn metal oxides toward ozone catalytic oxidation (OZCO) of toluene and the influence of moisture on the decomposition process. The simple impregnation method was adapted to disperse the metal oxides and found highly active for toluene oxidation in the presence of ozone. The steady-state activities and ozone decomposition data reveal that the activity is in the order of Mn/13X > Ce/13X > Cu/13X > Ag/13X > Co/13X and Mn/13X > Cu/13X > Ce/13X > Ag/13X > Co/13X, respectively. The extent of ozone decomposition is responsible for the degree of oxidative decomposition of toluene over the Mn/13X catalyst. The addition of moisture (Relative Humidity of 25%) to the reaction mixture considerably enhanced the conversion of toluene and selectivity to carbon oxides from 49% to 61% and 38% to 53% respectively, on the Mn/13X catalyst. The two sets of experiment results reveal that the surface adsorbed by-products such as benzene, benzaldehyde, p-methyl phenol and oxalic acid are considerably oxidized to CO2 in the presence of moisture whereas in the absence of moisture these by-products are slowly oxidized. The activity data in the presence of ozone and moisture also reveal that the moisture has considerably enhanced the activation of surface adsorbed by-products than that of initial toluene oxidation. Based on the temperature programmed desorption and temperature programmed oxidation studies, the addition of moisture decreased the by-products accumulation thereby, reduced the catalyst deactivation and enhanced the extended oxidation of toluene on the Mn/13X zeolite. The configuration about the central C=N bond is E, and there is an intramolecular O—H...N hydrogen bond which generates an S(6) ring. The molecular packing is stabilized by weak C—H...π interactions. The structure was refined as a two-component inversion twin.

An intramolecular O—H...N hydrogen bond generates an S(6) loop. In the crystal, molecules assemble into columns parallel to the a axis. The methyl group is disordered over two positions rotated from each other by 60°.

ABSTRACTColistin resistance in Acinetobacter baumannii is mediated by multiple mechanisms. Recently, mutations within pmrABC two-component system and overexpression of eptA gene due to upstream insertion of ISAba1 have been shown to play a major role. Thus, the aim of our study is to characterize colistin resistance mechanisms among the clinical isolates of A. baumannii in India. A total of 207 clinical isolates of A. baumannii collected from 2016 to 2019 were included in this study. Mutations within lipid A biosynthesis and pmrABC genes were characterized by whole-genome shotgun sequencing. Twenty-eight complete genomes were further characterized by hybrid assembly approach to study insertional inactivation of lpx genes and the association of ISAba1-eptA. Several single point mutations (SNPs), like M12I in pmrA, A138T and A444V in pmrB, and E117K in lpxD, were identified. We are the first to report two novel SNPs (T7I and V383I) in the pmrC gene. Among the five colistin-resistant A. baumannii isolates where complete genome was available, the analysis showed that three of the five isolates had ISAba1 insertion upstream of eptA. No mcr genes were identified among the isolates. We mapped the SNPs on the respective protein structures to understand the effect on the protein activity. We found that majority of the SNPs had little effect on the putative protein function; however, some SNPs might destabilize the local structure. Our study highlights the diversity of colistin resistance mechanisms occurring in A. baumannii, and ISAba1-driven eptA overexpression is responsible for colistin resistance among the Indian isolates.IMPORTANCEAcinetobacter baumannii is a Gram-negative, emerging and opportunistic bacterial pathogen that is often associated with a wide range of nosocomial infections. The treatment of these infections is hindered by increase in the occurrence of A. baumannii strains that are resistant to most of the existing antibiotics. The current drug of choice to treat the infection caused by A. baumannii is colistin, but unfortunately, the bacteria started to show resistance to the last-resort antibiotic. The loss of lipopolysaccharides and mutations in lipid A biosynthesis genes are the main reasons for the colistin resistance. The present study characterized 207 A. baumannii clinical isolates and constructed complete genomes of 28 isolates to recognize the mechanisms of colistin resistance. We showed the mutations in the colistin-resistant variants within genes essential for lipid A biosynthesis and that cause these isolates to lose the ability to produce lipopolysaccharides.

Outbreaks of monkeypox virus infections have imposed major health concerns worldwide, with high morbidity threats to children and immunocompromised adults. Although repurposed drugs and vaccines are being used to curb the disease, the evolving traits of the virus, exhibiting considerable genetic dynamicity, challenge the limits of a targeted treatment. A pan-genome-based reverse vaccinology approach can provide fast and efficient solutions to resolve persistent inconveniences in experimental vaccine design during an outbreak-exigency. The approach encompassed screening of available monkeypox whole genomes (n = 910) to identify viral targets. From 102 screened viral targets, viral proteins L5L, A28, and L5 were finalized based on their location, solubility, and antigenicity. The potential T-cell and B-cell epitopes were extracted from the proteins using immunoinformatics tools and algorithms. Multiple vaccine constructs were designed by combining the epitopes. Based on immunological properties, chemical stability, and structural quality, a novel multi-epitopic vaccine construct, V4, was finalized. The study found that the prevalence of hypertension among diabetics was 53.89%. Modifiable risk factors such as smoking and alcohol consumption were significantly associated with hypertension in diabetics, while non-modifiable risk factors such as age and duration of diabetes were not statistically significant. The study also found that certain biochemical parameters, such as HDL and cholesterol levels, were related to hypertension.
