In this work, we improve the performance of an optically pumped spintronic terahertz emitter (STE) by a factor of up to 6 in field amplitude through an optimized photonic and thermal environment. Using high-energy pump pulses (energy 5 mJ, fluence >1 mJ/cm2, wavelength 800 nm, duration 80 fs), we routinely generate terahertz pulses with focal peak electric fields above 1.5 MV/cm, fluences of the order of 1 mJ/cm2, and a spectrum covering the range 0.1-11 THz. Remarkably, the field and fluence values are comparable to those obtained from a state-of-the-art terahertz table-top high-field source based on tilted-pulse-front optical rectification in LiNbO3. The optimized STE inherits all attractive features of the standard STE design, for example, ease of use and the straightforward rotation of the terahertz polarization plane, without the typical 75% power loss found in LiNbO3 setups. It, thus, opens up a promising pathway to nonlinear terahertz spectroscopy. Pivotal degradation pathways of the most active non-heme iron epoxidation catalyst to date are investigated in detail. In-depth HR-ESI-MS and NMR spectroscopy decomposition studies and comparative catalytic experiments exclude the generally accepted literature paths of μ2-oxo bridged FeIII–O–FeIII dimer formation and methylene bridge oxidation. Instead, the Fe–NHC bond has been identified as the “weak spot”. It is shown that direct oxidation of one of the carbenes (ImCO) results in its de-coordination and induces protonation of the other NHC moieties, thus completely deactivating the catalyst. Evidence for protonation prior to carbene oxidation has not been found in this study.

The first diiron(III,IV)-μ2-oxo tetracarbene complex is isolated and characterized by SC-XRD, UV/Vis, EPR, Evans’ NMR and elemental analysis. CV indicates the presence of a transient high-valent diiron(IV)-μ2-oxo species. Its formation and decay is investigated via UV/Vis kinetics and NMR.

A straightforward modification route to obtain mono‐ and di‐substituted anthroyl ester bridge functionalized dinuclear Au(I) bis‐N‐heterocyclic carbene complexes is presented. The functionalization can be achieved starting from a hydroxyl‐functionalized ligand precursor followed by transmetallation of the corresponding Ag complex or via esterification of the hydroxyl‐functionalized gold complex. The compounds are characterized by NMR‐spectroscopy, ESI‐MS, elemental analysis and SC‐XRD. The mono‐ester Au complex shows quantum yields around 18%. In contrast, the corresponding syn‐di‐ester Au complex, exhibits significantly lower quantum yields of around 8%. Due to insufficient water solubility of the di‐ester, only the mono‐ester complex has been tested regarding its antiproliferative activity against HeLa‐ (cervix) and MCF‐7‐ (breast) cancer cell lines and a healthy fibroblast cell line (V79). IC50 values of 7.26 μM in the HeLa cell line and 7.92 μM in the MCF‐7 cell line along with selectivity indices of 8.8 (HeLa) and 8.0 (MCF‐7) are obtained. These selectivity indices are significantly higher than those obtained for the reference drugs cisplatin or auranofin., A simple modification route of hydroxyl functionalized bis‐NHC dinuclear Au(I) complexes with anthracene is presented, which leads to a significant improvement of the antiproliferative activity and quantum yields up to 18%. By controlling the reaction parameters, one hydroxyl group remains, allowing for further potential bioconjugation to obtain a selective theranostic metal complex.

A series of histidine derived Au(I) bis‐NHC complexes bearing different ester, amide and carboxylic acid functionalities as well as wingtip substituents is synthesized and characterized. The stability in aqueous media, in vitro cytotoxicity in a set of cancer cell lines (MCF7, PC3 and A2780/A2780cisR) along with the cellular uptake are evaluated. Stability tests suggest hydrolysis of the ester within 8 h, which might lead to deactivation. Furthermore, the bis‐NHC system shows a sufficient stability against cysteine and the thiol containing peptide GSH. The benzyl ester and amide show the highest activity comparable to the benchmark compound cisplatin, with the ester only displaying a slightly lower cytotoxicity than the amide. A cellular uptake study revealed that the benzyl ester and the amide could have different intracellular distribution profiles but both complexes induce perturbations of the cellular physiological processes. The simple modifiability and high stability of the complexes provides a promising system for upcoming post modifications to enable targeted cancer therapy., Novel histidine‐derived Au(I) bis‐NHCs with different functional groups (ester, amide and carboxylic acid) show antiproliferative activity in MCF7, PC3 and A2780/A2780cisR cancer cell lines. Two bio-inspired non-heme iron complexes bearing mixed tetradentate N-heterocyclic carbene/1,2,3-triazole ligands with cis labile coordination sites are reported. The compounds are studied in olefin epoxidation catalysis using H2O2 as oxidant. Sc(OTf)3, HClO4 and HOAc are applied as additives resulting in significant improvement of catalytic performance. Under optimized conditions the most active catalyst exhibits activities of 76,000 turnovers per hour, which is the highest reported value for an iron(II) catalyst. The complexes reveal comparably high stability and enable the challenging epoxidation of functionalized olefins. These results prove 1,2,3-triazoles to be promising and tunable ligands for iron catalyzed oxidation reactions.

Different models have been used to evaluate the interfacial Dzyaloshinskii-Moriya interaction (DMI) from the asymmetric bubble expansion method using magneto-optics. Here we investigate the most promising candidates over a range of different magnetic multilayers with perpendicular anisotropy. Models based on the standard creep hypothesis are not able to reproduce the domain wall (DW) velocity profile when the DW roughness is high. Our results demonstrate that the DW roughness and the interface roughness of the sample layers are correlated. Furthermore, we give guidance on how to obtain reliable results for the DMI value with this popular method. A comparison of the results with Brillouin light scattering (BLS) measurements on the same samples shows that the BLS approach often results in higher measured values of DMI.

Keplerian-Stacker is an algorithm able to combine multiple observations acquired at different epochs taking into account the orbital motion of a potential planet present in the images to boost the ultimate detection limit. In 2019, a total of 100 hours of observation were allocated to VLT VISIR-NEAR, a collaboration between ESO and Breakthrough Initiatives, to search for low mass planets in the habitable zone of the Alpha Cen AB binary system. A weak signal (S/N = 3) was reported around Alpha Cen A, at a separation of 1.1 a.u. which corresponds to the habitable zone. We have re-analysed the NEAR data using K-Stacker. This algorithm is a brute-force method able to find planets in time series of observations and to constrain their orbital parameters, even if they remain undetected in a single epoch. We scanned a total of about 3.5e+5 independent orbits, among which about 15 % correspond to fast moving orbits on which planets cannot be detected without taking into account the orbital motion. We find only a single planet candidate, which matches the C1 detection reported in Wagner et al. 2021. Despite the significant amount of time spent on this target, the orbit of this candidate remains poorly constrained due to these observations being closely distributed in 34 days. We argue that future single-target deep surveys would benefit from a K-Stacker based strategy, where the observations would be split over a significant part of the expected orbital period to better constrain the orbital parameters. This application of K-Stacker on high contrast imaging data in the mid-infrared demonstrates the capability of this algorithm to aid in the search for Earth-like planets in the habitable zone of the nearest stars with future instruments of the E-ELT such as METIS., 9 pages, 11 figures, K-Stacker github link

In recent years, femtosecond extreme-ultraviolet (XUV) and x-ray pulses from free-electron lasers have developed into important probes to monitor processes and dynamics in matter on femtosecond-time and angstrom-length scales. With the rapid progress of versatile ultrafast x-ray spectroscopy techniques and more sophisticated data analysis tools, accurate single-pulse information on the arrival time, duration, and shape of the probing x-ray and XUV pulses becomes essential. Here, we demonstrate that XUV pulses can be converted into terahertz electromagnetic pulses using a spintronic terahertz emitter. We observe that the duration, arrival time, and energy of each individual XUV pulse is encoded in the waveform of the associated terahertz pulses, and thus can be readily deduced from single-shot terahertz time-domain detection.

In antiferromagnets, the efficient transport of spin-waves has until now only been observed in the insulating antiferromagnet hematite, where circularly (or a superposition of pairs of linearly) polarized spin-waves diffuse over long distances. Here, we report long-distance spin-transport in the antiferromagnetic orthoferrite YFeO

The synthesis and antiproliferative activity of Mes- and iPr-substituted gold(I) bis(1,2,3-triazol-5-ylidene) complexes in various cancer cell lines are reported, showing nanomolar IC50 values of 50 nM (lymphoma cells) and 500 nM (leukemia cells), respectively (Mes < iPr). The compounds exclusively induce apoptosis (50 nM to 5 μM) instead of necrosis in common malignant blood cells (leukemia cells) and do not affect non-malignant leucocytes. Remarkably, the complexes not only overcome resistances against the well-established cytostatic etoposide, cytarabine, daunorubicin, and cisplatin but also promote a synergistic effect of up to 182% when used with daunorubicin. The present results demonstrate that gold(I) bis(1,2,3-triazol-5-ylidene) complexes are highly promising and easily modifiable anticancer metallodrugs.

Fluorescent Pd(II) and Pt(II) complexes bearing 4-methylene-7-methoxycoumarin (MMC) and 2,6-diispropylphenyl (Dipp) substituted NHC/1,2,3-triazole hybrid ligands are described. Depending on the reaction conditions two different ligand coordination modes are observed, i.e., bidentate solely coordinating via NHCs or tetradentate coordinating via NHCs and 1,2,3-triazoles. All Dipp substituted complexes show antiproliferative activity against cervix (HeLa) and breast (MCF-7) human carcinoma cells. The activity significantly depends on the coordination mode, with the tetradentate motif being notably more effective (HeLa: IC50 = 3.9 μM to 4.7 μM; MCF-7: IC50 = 2.07 μM to 2.35 μM). Amongst the MMC series, only the Pd(II) complex featuring the bidentate coordination mode is active against HeLa (IC50 = 6.1 μM). In contrast to its structurally related Dipp derivative (SI = 0.6), it shows a high selectivity for HeLa (SI > 16) compared to healthy skin cells (HaCaT). According to fluorescence microscopy, this compound is presumably located in late endosomes or lysosomes.

p300 and CREB-binding protein (CBP) are essential for a multitude of cellular processes. Dysregulation of p300/CBP histone acetyltransferase activity is linked to a broad spectrum of human diseases including cancers. A novel drug-like spirohydantoin (21) has been discovered as a selective orally bioavailable inhibitor of p300/CBP histone acetyltransferase. Lead compound 21 is more potent than the first-in-class lead A-485 in both enzymatic and cellular assays and lacks the off-target inhibition of dopamine and serotonin transporters, that was observed with A-485.

Within the field of cochlear implantation (CIs), the role of utilizing patient-specific cochlear anatomy for choosing the optimal implant electrode is becoming increasingly important. Unfortunately, performing detailed anatomical measurements of a cochlea using clinical imaging data is rather time consuming and hence difficult to implement into the clinical routine. In order to accelerate clinical cochlear anatomy evaluations, previously developed mathematical models can be adjusted to the patient-specific anatomy by measuring just a few overall cochlear dimensions. However, the accuracy of model-based cochlear anatomy estimations is unclear, and incorrect evaluations may lead to false conclusions regarding the suitability of specific implant electrodes.Based on 10 cochleae, an error evaluation of various commonly used curve fitting approaches for cochlear shape and duct length approximation was conducted. Spline tracings of the cochlear contours were used as reference values for the various approximations.Parameterized average cochlear helix models and two of five analytical approaches were found to be suitable for reconstructing the cochlear helical shape and estimating its length.Spline curve reconstructions are the most accurate and reliable method for assessing patient-specific cochlear geometry, especially in the case of anatomical irregularities. The most accurate results within the group of model-based evaluations still resulted in mean overall cochlear length deviations of approximately 5%.Spline curve reconstructions appear to be the best option for anatomical diagnostics in clinical practice. Retrospective studies can be performed to further evaluate model-based evaluations.

The dynamic and reversible acetylation of proteins, catalysed by histone acetyltransferases (HATs) and histone deacetylases (HDACs), is a major epigenetic regulatory mechanism of gene transcription and is associated with multiple diseases. Histone deacetylase inhibitors are currently approved to treat certain cancers, but progress on the development of drug-like histone actyltransferase inhibitors has lagged behind. The histone acetyltransferase paralogues p300 and CREB-binding protein (CBP) are key transcriptional co-activators that are essential for a multitude of cellular processes, and have also been implicated in human pathological conditions (including cancer). Current inhibitors of the p300 and CBP histone acetyltransferase domains, including natural products, bi-substrate analogues and the widely used small molecule C646, lack potency or selectivity. Here, we describe A-485, a potent, selective and drug-like catalytic inhibitor of p300 and CBP. We present a high resolution (1.95 Å) co-crystal structure of a small molecule bound to the catalytic active site of p300 and demonstrate that A-485 competes with acetyl coenzyme A (acetyl-CoA). A-485 selectively inhibited proliferation in lineage-specific tumour types, including several haematological malignancies and androgen receptor-positive prostate cancer. A-485 inhibited the androgen receptor transcriptional program in both androgen-sensitive and castration-resistant prostate cancer and inhibited tumour growth in a castration-resistant xenograft model. These results demonstrate the feasibility of using small molecule inhibitors to selectively target the catalytic activity of histone acetyltransferases, which may provide effective treatments for transcriptional activator-driven malignancies and diseases.

Herein we disclose SAR studies of a series of dimethylamino pyrrolidines which we recently reported as novel inhibitors of the PRC2 complex through disruption of EED/H3K27me3 binding. Modification of the indole and benzyl moieties of screening hit 1 provided analogs with substantially improved binding and cellular activities. This work culminated in the identification of compound 2, our nanomolar proof-of-concept (PoC) inhibitor which provided on-target tumor growth inhibition in a mouse xenograft model. X-ray crystal structures of several inhibitors bound in the EED active-site are also discussed.

Novel thermoelectric materials developed for operation at room temperature must have similar or better performance along with being as ecofriendly as those commercially used

Apolipoprotein E is a 299-residue lipid carrier protein produced in both the liver and the brain. The protein has three major isoforms denoted apoE2, apoE3, and apoE4 which differ at positions 112 and 158 and which occur at different frequencies in the human population. Genome-wide association studies indicate that the possession of two apoE4 alleles is a strong genetic risk factor for late-onset Alzheimer's disease (LOAD). In an attempt to identify a small molecule stabilizer of apoE4 function that may have utility as a therapy for Alzheimer's disease, we carried out an NMR-based fragment screen on the N-terminal domain of apoE4 and identified a benzyl amidine based fragment binder. In addition to NMR, binding was characterized using various other biophysical techniques, and a crystal structure of the bound core was obtained. Core elaboration ultimately yielded a compound that showed activity in an IL-6 and IL-8 cytokine release assay.

Protein lysine methyltransferases (PKMTs) regulate diverse physiological processes including transcription and the maintenance of genomic integrity. Genetic studies suggest that the PKMTs SUV420H1 and SUV420H2 facilitate proficient nonhomologous end-joining (NHEJ)-directed DNA repair by catalyzing the di- and trimethylation (me2 and me3, respectively) of lysine 20 on histone 4 (H4K20). Here we report the identification of A-196, a potent and selective inhibitor of SUV420H1 and SUV420H2. Biochemical and co-crystallization analyses demonstrate that A-196 is a substrate-competitive inhibitor of both SUV4-20 enzymes. In cells, A-196 induced a global decrease in H4K20me2 and H4K20me3 and a concomitant increase in H4K20me1. A-196 inhibited 53BP1 foci formation upon ionizing radiation and reduced NHEJ-mediated DNA-break repair but did not affect homology-directed repair. These results demonstrate the role of SUV4-20 enzymatic activity in H4K20 methylation and DNA repair. A-196 represents a first-in-class chemical probe of SUV4-20 to investigate the role of histone methyltransferases in genomic integrity.
