Giriş ve Amaç: Adipokinler ve disfonksiyonel yağ dokusu arasındaki ilişkide subkutan yağ dokusundan ziyade viseral yağ dokusundaki değişikliklerin rolüne dikkat çekilmiştir. Özellikle cilt altı yağ dokusunun etkilendiği lipödemde adipokinler hakkında bilgi yetersizdir. Bu çalışmada lipödemli hastalarda adiponektin, ghrelin, resistin ve visfatin düzeylerinin ve bunların yağ doku kalınlığı ile ilişkisinin araştırılması amaçlandı. Bu amaçla cilt altı yağ dokusu kalınlığı ultrasonografi ile objektif olarak değerlendirildi. Gereç ve Yöntemler: Lipödem tanısı almış toplam 19 kadın hasta ve yaş farkı olmayan 15 sağlıklı kadın çalışmaya dahil edildi. Deri ve deri altı yağ dokusu kalınlıkları ultrasonografik olarak ölçüldü. Tüm deneklerin serum adiponektin, ghrein, resistin ve visfatin seviyeleri sandviç ELISA protokolü kullanılarak ölçüldü. Sonuç: Bulgularımıza göre, serum adipokin düzeyleri ile deri altı yağ dokusu kalınlığı arasında anlamlı bir ilişki bulunmamakla birlikte, tamamen ilgisiz oldukları tartışmalıdır. Daha geniş serilerde yapılacak çalışmalar adipokinlerin cilt altı doku kalınlığı ile ilişkisine ve ultrasonografinin önemine ışık tutacaktır.

Objectives Learning approach in a certain learning process is based on the student’s intentions, behaviors, and habits according to his/her perceptions of the task of learning and determines the amount and quality of learning. The objective of this study is to evaluate the learning approaches of 1st and 4th grade midwifery students to biochemistry course and the change, if there is any, through their education. Methods This is an observational, cross-sectional study. The research population consisted of the voluntary students (86.47%, n:147) of the 2017–2018 season of the 1st and 4th class of Manisa Celal Bayar University, Health Science Faculty, Midwifery Department (n:170). Data were collected by using the “Introductory Information Form” and the “Learning Approach Scale”, and evaluated in the SPSS package program by performing number, percentile, mean, standard deviation, independent t test. Results The mean age of the students was 20.82 ± 1.81. Over 95% of the students stated that biochemistry lesson was necessary, while 59.9% reported that their biochemistry knowledge was insufficient. Over 87% of the student expressed their belief that the content of the biochemistry classes will help them in their professional career. Mean score of deep approach for Learning Approach Scale was 34.13 ± 6.07 (Min:19.00–Max:50.00), and mean score of superficial approach for Learning Approach Scale was 26.94 ± 6.37 (Min:15.00–Max:50.00). There was a significant relation between deep approach scale score and the perception of high importance of biochemistry in the professional life (p Conclusions Midwifery students, who believe that biochemistry is necessary for their professional career have a higher motivation for learning biochemistry, thus perform a deeper approach to learning. In general, creating effective and dynamic educational environments that support deep learning is necessary for enhancing the learning of biochemistry.

Giriş ve Amaç: Ten tene temas vajinal ve sezaryen doğumlarda tercih edilen ve pek çok olumlu etkisi bilinen bir uygulamadır. Bu çalışmada vajinal doğum sonrasında ten tene temasın plasenta ayrılma süresi ve doğum hormonları üzerine olan etkisini araştırnayı planladık. Yöntem ve gereçler: Çalışmaya XXXX kadın hastalıkları ve doğum kliniğine başvuran 40 gebe dahil edildiç çalışma grubunda doğum sonrası kesintisiz ten tene temas uygulandı, control grubunda anne ve bebek ayrıldı.Plasentanın ayrılma süresi not edildi ve doğum öncesi doğum sonrası kan örneklerinde oksitoain, β-endorfin ve katekolamin düzeyleri ölçüldü. Bulgular: Kadınların ortalam yaşı çalışma grubunda 28.55±5.97, control grubunda 26.75±6.58 idi. Plasentanın ayrılma süresi çalışma grubunda istatistiksel olarak anlamlı biçimde daha kısa bulundu. Oksitosin ve katekolamin seviyeleri hem çalışma hem control grubunda pre ve postpartum dönemde benzerdi. β-endorfin seviyeleri postpartum dönemde iki grup arasında benzerdi ancak prepartum dönemde çalışma grubunda istatistiksel olarak anlamlı biçimde daha yüksektir. Tartışma ve Sonuç: Doğum sonrası uygulanan ten tene temas doğum hormonları arasındaki dengeyi bozmadan plasenta ayrılma süresini kısaltmaktadır. Sağlık çalışanları ten tene temasın önemi hakkında bilgilendirilmeli ve uygulama erken postpartum dönemde yaygınlaştırılmalıdır. Doğum salonunda çalışanlar yani ebe, hemşire ve doktorlar ten tene temasın pek çok avantajının yanısıra plasenta yarılma süresini kısaltması ve doğum hormonlarına etki etmemesi konusunda da bilgilendirilmelidir.

To evaluate the effect of delivery in a vertical birth chair (VBC) and traditional delivery table (DT) supported by women’s movement during labor on the labor process, fetal outcome, maternal hormone levels, birth comfort, and satisfaction. This randomized controlled trial was conducted with 1:1:1 allocation. Group 1: in the VBC in upright position, Group 2: on the DT in supine position, these groups supported by freedom of movement, control group: on the DT in supine position, labor in bed. The duration of second stage of labor was not different between the groups (p = 0.246). The occurrence of instrumental birth, episiotomy, and perineal laceration was also not different among the groups (p = 0.772, p = 0.953, and p = 0.124). The use of uterotonic was observed in control group (p = 0.001). 1 and 5 APGAR scores of newborns were not different in all groups (p = 0.121, p = 0.268). The lowest pain score was observed in Group 1 (p = 0.001). Birth comfort and satisfaction were higher in Group 1 (p = 0.001 and p = 0.001). Decreased postpartum prolactin levels and increased postpartum oxytocin levels were observed in the control and Group 1 (p = 0.004, p = 0.006). Freedom of movement during labor and delivery using VBC in upright position can play birth-promoting and supporting role. There were no negative effects on the fetal outcome.

Objectives Postgraduate midwifery students learn the science and art of midwifery in clinical settings through the integration of evidence-based knowledge, practical skills, quality of care, and personal attitudes. These personal attitudes are confidence, intuition, problem solving, critical thinking, and self-efficacy. This study’s goal was to evaluate the clinical self-efficacy of midwifery students who took courses related to biochemistry in the postgraduate education program. Methods A cross-sectional design was used in the study. In the study, data collection was performed using surveys from students in the postgraduate midwifery education program. The study data were collected by employing the “Introductory Information Form” and the “Self-efficacy for Clinical Evaluation Scale.” Data evaluation was performed by number, percentage, mean, and correlation test. Results It was revealed that 67.7% of midwifery students who took the courses were postgraduate students in the thesis stage, 41% of them worked in secondary health services, and 51.6% of them had a clinical experience of 5 years and more. The total score of the self-efficacy perception subscale was 4.22 ± 0.49 (1–5), whereas the total score of the perceived confidence subscale was 4.57 ± 0.45 (1–5). There was a statistically significant correlation between the self-efficacy subscale and the importance subscale. Conclusions The results indicated that postgraduate midwifery students had high clinical self-efficacy scores. We think that the elective courses related to biochemistry in the postgraduate midwifery education program may have positively affected their clinical skills. We anticipate that it will be appropriate to conduct research with a larger sample size to examine the effect of postgraduate midwifery education on students’ clinical self-efficacy.

Objective:The most common cancer in smokers is bladder cancer (BC). Intravesical chemo immunotherapies are used to lower the risk of progression in patients who are at risk. Bacillus Calmette-Guérin immunotherapy is the most effective adjuvant therapy discovered. We aimed to evaluate the levels of arginine, citrulline, ornithine, symmetrically dimethylated arginine (SDMA), N-monomethyl-Larginine. (L-NMMA) or asymmetrically dimethylated arginine (ADMA) in patients with BC, as well as their relationship with methylarginine.Materials and Methods:Blood samples were collected from all patients (n=30) and controls (group 1) prior to transurethral resection of bladder tumour (TURBT) (group 2), 20 days after TURBT (group 3) and at the end of intravesical immunotherapy 74 (group 4). The levels of serum methylated arginine were measured using ABSCIEX API 3200 tandem mass spectrometry system in positive ESI mode.Results:In comparison to group 2, group 1’s ADMA and arginine/total methylated arginine levels were 98 significantly lower (p=0.035 and p=0.049, respectively), while SDMA/ADMA, L-NMMA and arginine/ADMA levels (p=0.001, p=0.008 and p=0.017) increased, and no statistical difference was found for other parameters (p>0.05). When compared to group 3, ADMA, arginine, citrulline, methylated arginines and L-NMMA levels in group 2 (p=0.035, p=0.001, p=0.015, p=0.032, p=0.032) increased, while SDMA/ADMA levels (p=0.041) decreased.Conclusion:The decrease in arginine and ADMA levels in non-muscle invasive BC patients is thought to be promising, and these markers may be useful in monitoring the diagnosis and treatment of patients.

Angiogenesis is the formation of new blood vessels. Angiogenesis affects cancer growth and is a useful target for cancer therapeutics. The effects of geldanamycin on angiogenesis in cases of gastric cancer are poorly understood. We investigated the effects of different doses of 17-allylamino-17-demethoxygeldanamycin (17-AGG), a semi-synthetic derivative of geldanamycin, on the interactions between cellular matrix proteins and angiogenesis factors in a gastric cancer cell line. We examined cancer cells on laminin and collagen I coated surfaces to determine their response to the angiogenic effect of these matrix molecules. We also evaluated the expression levels of VEGF, MMP-9, ES and TSP-1 using ELISA. We found that application of 17-AAG to the gastric cancer cell line on culture dish plastic decreased VEGF, TSP-1, ES and MMP-9 expression, whereas of all of these proteins were increased by laminin and collagen coating. 17-AAG currently is in clinical trial phase 2 and may be a promising drug for treatment of gastric cancer.

Buçalışmada mide kanseri hücre kültürüne uygulanan CAPE’ninVEGF, MMP, TSP-1 ve Endostatin gibi anjiyojenik ve antianjiyojenikbelirteçler arasındaki ilişkiyi araştırmayı amaçladık.CAPE uygulamasını takiben sitotoksisite, MTT testi ile ölçüldü.Hücrelerin anjiyojenik davranışı immünohistokimyasal olarakincelendi ve VEGF, MMP ve Endostatin ile TSP-1 seviyesi LCPCR testi ile ölçüldü. Even thoughthis process is useful in tissue repair and normal growth, it alsofacilitates the growth of cancer tissues and their metastasis viathe hematogenous route, and in this way, it speeds up metastasisand invasion. In the present study, we aimed to investigatethe relationship between angiogenesis and markers such asVEGF, MMP, TSP-1 and Endostatin in a CAPE-administeredgastric cancer cell in culture. Following CAPE administration,cytotoxicity was measured by MTT test. Angiogenic behavior ofcells was examined immunohistochemically by VEGF, MMP andEndostatin, and TSP – 1 level was measured by RT-PCR test. MTTtest showed that CAPE administered at a concentration of 0.5 μg/mL exerted a cytotoxic effect on a gastric cancer cell line. Geneexpression depicted by RT-PCR showed that VEGF, MMP and TSPlevels were decreased , while the endostatin level was increased.Changes in MMP, VEGF, TSP and endostatin suggest that CAPEhas a significant therapeutic effect on gastric cancer. In the lightof these findings, CAPE, which is a natural and inexpensivesubstance, could be effective in the treatment of cancer patientsand advanced studies need to be carried out.

The objective of this study is to evaluate the oxidative status of colitis-associated cancer induced by azoxymethane (AOM)/dextran sulfate sodium (DSS), and the effects of COX-2 inhibitor in mice. Totally 40 mice were randomized and divided to four groups. All animals except control and Cox-2 inhibitor alone group received AOM/DSS to establish colitis-associated cancer model as reported elsewhere. COX-2 preferential inhibitor meloxicam was used to minimize side effects such as gastrointestinal hemorrhage. Meloxicam were used (5mg/kg, intraperitoneal) three times a week with meloxicam alone and AOM/DSS + meloxicam group. Objective: Scientific studies are continuing on many molecules in order to be used in the diagnosis of diseases. The fact that even one of these molecules can be used for diagnosis is very important for clinicians in diagnosing the disease. For this purpose, intensive studies continue on homocysteine and arginine. For these reasons, in this study, we aim to determine whether there is any change in homocysteine and arginine levels in the serum of blood samples of breast patients and to explain its relationship with breast cancer.Material Materials and Methods: We carried out our work in Selcuk University Faculty of Medicine Biochemistry Metabolism Laboratory. Our study included 20 controls, 20 breast Ca, 20 metastasis patients. Blood collected from breast cancer patients stored at -80 ° C until biochemical analysis is performed. Homocysteine and arginine levels measured in the sera obtained from the blood samples taken. The levels of proteins were measured at 470 nm in the NMRspectrophotometer. To measure homocysteine levels were injected into the mass spectrometer for chromatography.Results: In the evaluation, a statistical increase was observed in Arginine, Arginine / ADMA levels in patients with breast Ca, and Homocysteine levels in patients with metastases compared to the control group. Bu nedenlerle biz de, bu çalışmamızda meme hastalarının kan örneklerinin serumlarında homosistein ve arjinin düzeylerinde herhangi bir değişiklik olup olmadığını belirleyebilmeyi ve bunun meme kanseri ile ilişkisini açıklayabilmeyi amaçlamaktayız. Gereç ve Yöntemler: Selçuk Üniversitesi Tıp Fakültesi Biyokimya Metabolizma Laboratuvarında çalışmalarımız gerçekleştirilmiştir. Çalışmamıza 20 kontrol, 20 meme Ca, 20 metastaz hastası dahil edildi. Sonuç: Elde ettiğimiz sonuçlara dayanarak, meme kanseri hastalarının tanı ve tedavileri sırasında bu markerlerın kullanılabileceği düşünülmektedir.

Objectives The aim of the present study was to investigate the effect of treatment on IL-1, IL-6, IL-8 and neopterin levels in patients with non-muscle invasive bladder cancer (NMIBC). Methods Thirty patients with NMIBC and 30 age-matched controls were included in the study. Preoperative, postoperative first control (at two weeks after second transurethral resection of bladder tumor (TURBT)) and second control (at the end of intravesical immunotherapy) blood samples were analyzed by ELISA to determine IL-1, IL-6, IL-8 and neopterin levels. The mean cytokine levels of the patients were statistically compared as well as comparing the patients’ and controls’ levels. Results There were no statistically significant differences between the mean IL-1, IL-6, IL-8 and neopterin levels of the patient and control group before initial TURBT. In the patient group, there were no statistically significant differences in the IL-6 and IL-8 levels after both TURBT and intravesical BCG therapy. In addition to the initiation and continuity of lactation, hormones transferred from mother plasma to milk during breastfeeding are also important. The aim of this study was to determine the effect of back massage on maternal serum ghrelin, leptin, adiponectin and visfatin levels in women giving birth. A total of 60 mothers, participated in this experimental study. Massage was applied in the first 3–4 h after birth. Blood sample was taken in the postnatal routine. It was determined that the intervention group did not make a statistically significant difference in ghrelin (1.37 ± 0.7, 1.00 ± 0.2), leptin (0.76 ± 0.5, 0.70 ± 0.1), adiponectin (3.55 ± 0.8, 3.05 ± 0.5) levels compared to the control group (p > 0.05). However, visfatin (10.53 ± 2.4, 9.97 ± 2.0) levels were statistically higher than the control group (p Back massage was found to be beneficial in terms of increased growth factors in breast milk production and content.

Purpose: This study examines the dyadic adjustment of married men and women with multiple sclerosis (MS). Method: This cross-sectional study consists of 40 patients with MS who were married and being followed in the neurology clinic of a university hospital. Data collection was performed by face-to-face interviews with only the participant and the researcher present. The participants filled in the Expanded Disability Status Scale, a sociodemographic form, and a dyadic adjustment scale. Results: The study group was 75% male, with age ranging from 27 to 65 years (mean, 44.40 ± 9.31 years). Expanded Disability Status Scale scores ranged from 0 to 7 (mean, 3.25 ± 1.94); 95% of the subjects were having relapses and remissions. Scores for patients with MS who were not considering divorce were statistically significantly higher than patients with MS who were considering divorce for dyadic adjustment (z = 3.845, P =.000), common view (z = 2.336, P =.020), endearment (z = 2.701, P =.007), and total scores (z = 3.399, P =.001). Conclusion: Most of the patients with MS who participated in this study were diagnosed with relapsing-remitting MS with a mean Expanded Disability Status Scale score of 3, indicating that patients had the physical ability to perform routine activities. The occurrence of prostate cancer in men is one of the most common types of cancer. Recent studies have found important links between cancer and adipocytokines. Adipocytokines are thought to be factors in the occurrence of a variety of diseases. In addition, adipocytokines studies in cancer patients have shown that these hormones may have an effect in the formation of cancer. The objective of this study was to evaluate the relationship between adiponectin, resistin, and leptin levels in BHP and prostate cancer patients. The study was conducted between September 2012 and April 2013 at the Department of Medical Biochemistry and the Department of Urology. Blood samples were collected from 20 people in the same age range who had been diagnosed by examination and biopsy as having BHP (benign prostatic patients) and prostate cancer patients who had not been operated on. Leptin, adiponectin, resistin, and human serum levels were measured using ELISA kit. In the prostate cancer group, serum adiponectin and resistin levels were significantly lower than in the BHP group. However, serum leptin levels in the prostate cancer group were not significantly different from those in the BHP group. This information and our own findings show that adiponectin and resistin, which are from the adipocytokine family, may play an important role in the progression of prostate cancer, and thus it may be possible to use them as diagnostic markers. Anti-inflamatuar ajanlar, selektif siklooksijenaz 2 inhibitörleri ve doğal moleküllerin karsinogenezin önlenmesinde kullanılması önemli araştırma ve ilgi alanını oluşturur. Bu çalışmanın amacı, farelerde azoksimetan (AOM)/dekstran sülfat sodyum (DSS) ile oluşturulan kolit ile ilişkili kanser modelinde oksidatif durum ve COX-2 inhibitörünün etkilerini araştırmaktır. Toplam 40 fare rastgele olarak dört gruba ayrıldı. Kontrol ve COX-2 inhibitörü alan gruplar hariç bütün gruplar daha önce bildirilen yönteme göre AOM/DSS uygulanarak kolit ile ilişkili kanser modeli oluşturuldu. Gastrointestinal hemoraji gibi yan etkileri minimize etmek için COX-2 seçici inhibitörü meloksikam kullanıldı. Meloksikam 5 mg/kg dozda haftada üç kez i.p. olarak uygulandı. Oksidatif stres belirteçleri süperoksit dismutaz (SOD), glutatyon peroksidaz (GPx), malondialdehit (MDA) ve ileri oksidasyon protein ürünleri (AOPP) spektrofotometrik olarak ölçüldü. Meloksikam ve AOM/DSS birlikte uygulaması farelerde SOD aktivitesine önemli düzeyde yükseltmiştir (P< 0.05). GPx aktivitesi meloksikam ve AOM/DSS tek ve kombine kullanımında önemli düzeyde artırmıştır (P< 0.05). MDA düzeyinde konrol grubu ile diğer gruplar arasında fark bulunmamıştır. AOPP seviyesi ise, meloksikam ve AOM/DSS kombine kullanıldığı grupta diğer gruplara göre daha yüksek bulundu. Meloksikam ve/veya AOM/DSS uygulaması lipid peroksidasyonuna neden olmazken, antioksidan enzim ve AOPP değerlerini yükselttiği görüldü.

The chaperone-binding drug, 17-allylamino-17-demethoxygeldanamycin, has recently come into clinical use. It is a derivative of geldanamycin, an ansamycin benzoquinone antibiotic with anti-carcinogenic effect. Understanding the effect of this drug on the cancer cells and their niche is important for treatment. We applied 17-allylamino-17-demethoxygeldanamycin to colon cancer cell line (Colo 205) on matrix molecules to investigate the relationship of apoptosis with terminal deoxynucleotidyl transferase dUTP nick end labeling immunocytochemistry and related gene expression. We used laminin and collagen I for matrix molecules and vascular endothelial growth factor for angiogenic structure. We also examined apoptosis-related signaling pathway including mitochondrial proteins, cytochrome c, Bcl-2, caspase-9, Apaf-1 expression using real-time polymerase chain reaction. There was clear effect of 17-allylamino-17-demethoxygeldanamycin that killed more cells on tissue culture plastic compared to matrix molecules. The IC50 value was 0.58 µg/mL for tissue culture plastic compared with 0.64 µg/mL for laminin and 0.75 µg/mL for collagen I. The analyses showed that more cells on matrix molecules underwent apoptosis compared to that on tissue culture plastic. Apoptosis-related gene expression was similar in which Bcl-2 expression decreased and proapoptotic gene expression of the cells on matrix molecules increased compared to that on tissue culture plastic. However, the application of 17-allylamino-17-demethoxygeldanamycin was more effective for the cells on collagen I compared to the cells on laminin. There was also a decrease in angiogenesis as shown by the vascular endothelial growth factor staining. This was more pronounced by coating of the tissue culture plastic with matrix molecules. Our results supported the anti-cancer effect of 17-allylamino-17-demethoxygeldanamycin, and this effect depended on matrix molecules. This effect occurs through apoptosis, and related genes were also altered. All these genes may serve for novel target under the effect of matrix substrate. AIM The apoptotic effect of geldanamycin derivative may be important for the colorectal cancer therapy. The mechanisms of apoptosis require understanding of the behavior of colon cancer cell line Colo-205 which mimics colon adenocarcinoma. Therefore, the effect of IC50 dose of 17-allylamino-17-demethoxygeldanamycin (17-AAG) on the colon cancer cells in vitro was studied for its anti-apoptotic activity. METHOD Apoptotic ratio of the Colo-205 cells was determined after 17-AAG application with terminal deoxynucleotidyl transferase dUTP nick end labeling (TUNEL) staining and apoptosis related genes. Apoptosis signal path related key mitochondrial proteins, cytochrome c, bcl-2, caspase 9 and Apaf-1 expression were examined with RT-PCR method. RESULTS 17-AAG caused induction of cell death. Apoptotic related genes such as cytochrome-c, Apaf-1 and caspase-9 protein expressions were increased significantly (p < 0.05) and anti-apoptotic bcl-2 expression was decreased significantly (p < 0.05). Our results indicated that the application of 17-AAG on Colo-205 cells showed anticancer effect by the apoptosis due to alteration of apoptotic genes. CONCLUSION The apoptotic effect of 17-AAG as an natural product for alternative medicine would be very important for the success and quality of life during the treatment of colon carcinoma with the combination of anticancer drugs (Tab. 1, Fig. 2, Ref. 32).

Amaç: Kas invaziv olmayan mesane kanserinde (KİOMK) tedavinin anjiyogenik [vasküler endotelyal büyüme faktörü-2 (VEGF-2) ve matriks metaloproteinaz-2 (MMP-2)] ve antianjiyogenik [endostatin (ES) and trombospondin-1 (TSP-1)] faktörler üzerine etkilerini araştırmayı amaçladık.Gereç ve Yöntem: KİOMK'li 30 hasta ve aynı yaş grubundaki 30 kontrol çalışmaya dahil edildi. Hastaların kan örnekleri üç defa alınarak yukarıda bahsedilen markerlar enzime bağlı immünosorbent testi yöntemiyle ölçüldü. İlk kan örneği transüretral rezeksiyon-mesane tümörü (TUR-MT) öncesi, ikinci örnek TUR-MT sonrası 20. günde ve son örnek intravezikal immünoterapi sonrası alındı. Hasta ve kontrol grubunun marker ölçüm ortalamaları karşılaştırıldı. Ayrıca hasta grubundan alınan üç ayrı ölçüm de kendi arasında karşılaştırıldı.Bulgular: Hasta grubunun TRU-MT öncesi ortalama VEGF-2 ve MMP-2 değerleri kontrol grubununkinden anlamlı olarak yüksel bulunurken (sırasıyla p=0,04 ve p=0,01), ortalama ES ve TSP-1 değerleri iki grup arasında istatistiksel olarak benzerdi (sırasıyla p=0,95 ve p=0,99). Bu anjiyogenik markerlar KİOMK tedavi sonrası takipte kullanılabilir Objective: We aimed to investigate possible effects of treatment on angiogenic [vascular endothelial growth factor-2 (VEGF-2) and matrix metalloproteinase-2 (MMP-2)] and antiangiogenic [endostatin (ES) and thrombospondin-1 (TSP-1)] factors in non-muscle invasive bladder carcinoma (NMIBC). Materials and Methods: Thirty NMIBC patients and 30 age-matched controls were included in the study. For the above-mentioned markers, peripheral blood samples were drawn at three time points to be studied by enzyme-linked immunosorbent assay: before transurethral resection of bladder tumor (TURBT), at first control (20 days after the operation) and second control (at the end of intravesical immunotherapy). The mean blood levels obtained in the three measurements and those in patients and controls were compared statistically.Results: The mean levels of VEGF-2 and MMP-2 in patients before TURBT were found to be statistically significantly higher than in controls (p=0.04 and p=0.01, respectively), while no significant differences were obtained between the mean ES and TSP-1 levels (p=0.95 and p=0.99, respectively). It was also found that the VEGF-2 and MMP-2 levels were significantly decreased after TURBT (p=0.03 and p=0.01, respectively), but the tendency of these decrease was not found to be statistically significant between the first and second controls. Conclusion: Elevated VEGF-2 and MMP-2 levels in patients with NMIBC were significantly decreased after and probably due to the TURBT, which leads to a conclusion that these angiogenic markers may be used for follow-up of NMIBC

Introduction Papillary thyroid cancer is a disease that has been associated with chronic inflammation. The purpose of this study is to measure the production of the proinflammatory cytokines IL-1β, IL-6 and IL-8 and neopterin, which is a novel biomarker for cellular immune response in papillary thyroid cancer. Materials and methods The serum IL-1β, IL-6, IL-8 and neopterin values of 31 papillary thyroid cancer patients undergoing bilateral total thyroidectomy were measured before and 20 days after surgery. The values were compared with those of 39 healthy controls. Results Serum IL-1β levels were similar across groups. IL-6 (pConclusions The proinflammatory cytokines IL-6 and IL-8, but not IL-1β, were produced in greater amounts in papillary thyroid cancer. Serum neopterin seems to be a valid biological marker supporting the presence of papillary thyroid cancer.

Adipose tissue-derived hormones (adipocytokines), such as adiponectin, leptin, resistin and visfatin, and the pancreatic hormone insulin, have been suggested to play a role in carcinogenesis. we therefore hypothesized that the oncological treatment of breast cancer may alter the serum levels of these adipocytokines and insulin. In this study, we aimed to compare the serum levels of adipocytokines and insulin between the pre- and post-treatment period in patients with breast cancer. In this prospective study, 20 consecutive patients with stage II and III breast cancer underwent breast-conserving surgery or total mastectomy and/or axillary dissection. The patients received adjuvant chemotherapy and radiotherapy, if necessary. Blood samples were obtained during the preoperative period and postoperatively after completion of the adjuvant therapy. There was no statistically significant difference between the pre- and post-treatment levels of visfatin, adiponectin and leptin. BackgroundLeptin has physiological roles in multiple systems, and has possible effects on several carcinogenesis steps. The aim of this study was to investigate the leptin levels in thyroid papillary carcinoma (TPC) patients.MethodsForty-three female TPC patients and 30 healthy female control subjects were recruited for the study. TPC was diagnosed by fine needle aspiration biopsy. TPC patients had a bilateral total thyroidectomy operation and their leptin levels were measured before and 20 days after the operation.ResultsSerum leptin levels of TPC patients were higher than in control group subjects (21.15 ± 14.12 ng/mL vs. 9.89 ± 0.21 ng/mL, p < 0.05). The leptin levels decreased after total thyroidectomy (13.92 ± 10.55 ng/mL) compared to prethyroidectomy levels (22.94 ± 14.67 ng/mL) in 34 patients who came to the follow-up visit (p < 0.05). However, the decreased post-thyroidectomy levels of leptin were still statistically significantly higher than the control group levels. Multivariate regression analysis showed that the leptin levels in TPC patients were not related to age, menopausal status or pathologic occult status but were directly related to the cancer group.ConclusionLeptin levels were elevated in thyroid cancer, decreased after total thyroidectomy, and might be associated with thyroid papillary carcinogenesis.

In this study, we aimed to evaluate the possible relations of serum leptin and thyroid hormones on insulin treatment of surgically thyroidectomized and streptozotosin induced diabetic group of rats and whether the thyroid hormones control the leptin levels or leptin levels affect the thyroid hormones in DM. The Sprague-Dawley rats were assigned to eight groups: group 1, control; group 2, diabetes (injected intraperitoneally (i.p.) with streptozotocin (stz) 55 mg/kg); group 3, diabetes + insulin (rats were treated with insulin, 7-10 U/kg/day, subcutaneously); group 4, surgically thyroidectomized control; group 5, thyroidectornized + diabetes (3 weeks after the surgical operation, injected i.p. with stz); group 6, thyroidectornized + diabetes + insulin; group 7, thyroidectomized + diabetes + insulin 4- thyroid hormone (after diabetes induction, rats were treated with insulin and thyroid hormone, levothyroxin sodium (T-4; 2.5 mu g/kg); group 8, thyroidectornized + diabetes + insulin + thyroid hormone (T-4; 5 mu g/kg). The free and total T-3 and T-4 levels were measured in serum samples by otoanalyzer, and leptin levels were determined by ELISA method. The main finding of our recent study is that the decreased levels of serum leptin during the diabetes, hypothyroidism, and hypothyroidism with diabetes can be regulated in different percentages with the treatment of insulin and various doses of thyroid hormone. The observations in our study suggest the idea that during diabetic hypothyroidism, without thyroid hormone treatment, insulin is not sufficient to balance the metabolic pathways so mediated effects of insulin in leptin regulation via thyroid hormones are an increased possibility.

The oxidation of protein plays an essential role in the pathogenesis of an important number of degenerative and cancer diseases, which is now widely recognized. The aim is to examine advanced oxidation protein products (AOPPs), lipid peroxidation products malondialdehyde (MDA), and ferrous oxidation in xylenol orange (FOX) in blood samples of papillary thyroid cancer patients compared with healthy controls to determine the oxidation status and the change after thyroidectomy.Thirty-five female thyroid cancer patients who underwent total thyroidectomy and 39 female control subjects were included into this study. Prethyroidectomy and postthyroidectomy, AOPP, FOX, and MDA levels were studied.Prethyroidectomy AOPP, FOX, and MDA levels were significantly higher compared to control (P.05). In postthyroidectomy AOPP, FOX, and MDA levels were significantly decreased compared with prethyroidectomy levels (P.05). However, postthyroidectomy levels on the 20th day were still significantly higher, compared to control subjects (P.05).In conclusion, all of AOPP, FOX, and MDA levels that are markers of protein oxidation and lipid hyperoxidation may induce thyroid cancer development and begin to decrease after thyroidectomy.

Migration, invasion, metastasis and angiogenesis associated with cancer depend on the surrounding microenvironment. Angiogenesis, the growth of new capillaries, is a regulator of cancer growth and a useful target for cancer therapy. We examined matrix protein interactions in a gastric cancer cell culture that was treated with different doses of caffeic acid (3,4-dihydroxycinnamic acid) phenethyl ester (CAPE). We also investigated the relations among the levels of vascular endothelial growth factor (VEGF), matrix metalloproteinase-9 (MMP-9), endostatin (ES) and trombospondin-1 (TSP-1). Cytotoxity of CAPE was measured using the 3-(4,5-dmethylthiazol-2-yl)-2, 5-diphenyltetrazolium bromide (MTT) assay. We examined the behavior of cells on laminin and collagen I coated surfaces in response to the angiogenic effect of these matrix molecules. We examined the protein alterations of these matrix molecules immunohistochemically and measured the levels of VEGF, MMP-9, ES and TSP-1 using the ELISA test. We showed that application of CAPE to the gastric cancer cell line on tissue culture plastic, laminin and collagen I significantly decreased the VEGF and MMP-9 protein levels. We found that TSP-1 levels were increased significantly in the gastric cancer cells after application of CAPE. The protein levels of gastric cancer cells also were increased significantly when tissue was cultured on laminin and collagen I. Application of CAPE to cells on laminin or collagen I coated surfaces significantly increased all of the proteins except ES. ES levels were increased on the collagen I covered surfaces, but the laminin surface decreased the levels of ES significantly. We demonstrated the beneficial effect of CAPE on a gastric cancer cell line including inhibition of proliferation and induction of some proteins that might be related to decreased angiogenesis.

Free radical-induced lipid peroxidation has been associated with numerous disease processes including diabetes mellitus. Glutathione-S-transferase (GST) catalyses the conjugation of glutathione with a variety of organic peroxides to form more water-soluble compounds. Glucose-6-phosphate dehydrogenase (G6PDH) is essential to control intracellular reductive potential by increasing glutathione intracellular levels, which in turn decrease the amount of reactive oxygen species. Glyburide decreases glucose production and enhances insulin action in liver. The aim of this study was to examine the effects of glyburide on the antioxidant enzyme activities in the liver tissue of diabetic rat. We investigated the activities of GST and G6PDH in the liver of both control and streptozotocin-induced diabetic rats. Forty male albino rats were included in this study. Liver GST and G6PDH activities decreased significantly in five-week diabetic rats (p < 0.001 and p < 0.001 respectively) compared to controls and glyburide therapy restored these activities (p < 0.001 for GST and p < 0.001 for G6PDH). Elevations of hepatic antioxidant enzymes with glyburide administration suggest that glyburide may directly alter hepatic enzyme activities.

The role of capsaicin in carcinogenesis is quite controversial. Although some investigators suspect that capsaicin is a carcinogen, co-carcinogen, or tumor promoter, others have reported that it has chemopreventive and chemotherapeutic effects. The present study aimed to evaluate the cytotoxicity and chemosensitizing activities of capsaicin alone and on 5-flourouracil (5-FU)-treated gastric cancer cells. In this study, the gastric cancer cell line HGC-27 was used and capsaicin used as a chemosensitizer and 5-flourouracil (5-FU) was used as chemotherapeutic. Cytotoxicity and chemosensitizing activities were analyzed with MTT assay; supernatant levels of LDH and glucose were detected as biochemical markers of cell viability; cytochrome c and AIF were evaluated with western blot; and additionally, wound-healing assays were employed. Results suggested that capsaicin had significant anticancer abilities; such capsaicin were capable of causing multifold decreases in the half maximal inhibitory concentration IC50 value of 5-FU. The continuing controversy surrounding consumption or topical application of capsaicin clearly suggests that more well-controlled epidemiologic studies are needed to evaluate the safety and efficacy of capsaicin use. In summary, the present study demonstrated that capsaicin has the potential to be used for treating gastric carcinoma with 5-FU in vitro.

Background: Rutin is a strong antioxidant molecule and it has advantageous over other flavonoids due to it is a nontoxic and nonoxidizable molecule. The concept of dual therapy of anti-cancer drugs with natural compounds has become a very promising approach in new strategy to treatment. The aim of this study was to evaluate the antiproliferative and apoptotic effects of rutin flavonoids and its efficacy in enhancing the anticancer effects of 5-FU (5-fluorouracil) and oxaliplatin against colon cancer cells. Material and Methods: Caco-2 human colon cancer cells were treated with rutin and/or anticancer drugs (5-FU and oxaliplatin), cell viability and apoptotic parameters were examined. Cell viability was determined by MTT assay. Apoptotic markers (cleaved caspase 3, cleaved PARP and phosphor-Bad) were determined using specific enzyme-linked immunosorbent assay kits. Cell viability assay shows that rutin inhibiting growth of Caco-2 cells at high concentrations (over 1000 μM). Combination with rutin markedly enhanced 5-FU and oxaliplatin growth inhibiting effects on Caco-2 cells. Results suggested that rutin had significant anticancer abilities; such rutin were capable of causing multifold decreases in the half maximal inhibitory concentration IC50 value of 5-FU and oxaliplatin. This flavonoid increased the levels of proteins associated with apoptotic cell death (phospho-Bad, cleaved caspase 3 and cleaved PARP) alone and combination. The activities of caspase 8 and caspase 9 were stimulated by the combination treatment of rutin and oxaliplatin then alone. DNA fragmentation was observed only on combination treatment. Conclusions: Combined treatment with rutin and anticancer drugs (5-FU and/or oxaliplatin) is more effective than the individual treatments of drugs at inhibiting growth of Caco-2 cells. The use of lower 5-FU and oxaliplatin doses, with similar effects, could be also useful to reduce possible adverse effects of these drugs. However, further studies at molecular level are required to elucidate chemopreventive and chemotherapeutic effects of rutin on colon cancer.
