The baroclinic annular mode (BAM) is the leading mode of variability in extratropical eddy activity characterized by its hemispheric-scale pulsing. Based on atmospheric reanalysis data for the Southern Hemisphere, this study reveals BAM-associated systematic modulations not only in fluxes associated with subweekly transient disturbances, as found by earlier studies, but also in their spatial structure involved in the dynamics of the BAM. Specifically, in the positive phase of the BAM characterized by enhanced activity of transient disturbances, their lower-tropospheric baroclinic structure becomes more distinct, and they tend to be more elongated meridionally in both the upper and lower troposphere. These BAM-associated structural modulations of the disturbances favor the more efficient baroclinic development via enhanced poleward heat transport and their downstream development, which can contribute to hemispheric-scale enhancement of kinetic energy associated with the disturbances. In addition, a tendency of the disturbances to exhibit horizontally tilting structure becomes more evident in the positive phase of the BAM, which is favorable for enhanced transport of westerly momentum from the subtropics to the midlatitude polar-front jet, or equivalently enhanced wave-activity propagation from the midlatitude storm track into the subtropics. This modulation lags the peak of anomalous kinetic energy of the disturbances, thus acting to contribute to the decay of the BAM signature. A set of numerical simulations suggests that the BAM-associated pulsing in storm-track activity and structural modulations are manifestations of atmospheric internal dynamics, which can be significantly amplified in the presence of a midlatitude oceanic frontal zone through the formation of more organized and coherent baroclinic wave packets.

Starting to resolve the oceanic mesoscale in climate models is a step change in model fidelity. This study examines how certain obstinate biases in the midlatitude North Atlantic respond to increasing resolution (from 1° to 0.25° in the ocean) and how such biases in sea surface temperature (SST) affect the atmosphere. Using a multimodel ensemble of historical climate simulations run at different horizontal resolutions, it is shown that a severe cold SST bias in the central North Atlantic, common to many ocean models, is significantly reduced with increasing resolution. The associated bias in the time-mean meridional SST gradient is shown to relate to a positive bias in low-level baroclinicity, while the cold SST bias causes biases also in static stability and diabatic heating in the interior of the atmosphere. The changes in baroclinicity and diabatic heating brought by increasing resolution lead to improvements in European blocking and eddy-driven jet variability. Across the multimodel ensemble a clear relationship is found between the climatological meridional SST gradients in the broader Gulf Stream Extension area and two aspects of the atmospheric circulation: the frequency of high-latitude blocking and the southern-jet regime. This relationship is thought to reflect the two-way interaction (with a positive feedback) between the respective oceanic and atmospheric anomalies. These North Atlantic SST anomalies are shown to be important in forcing significant responses in the midlatitude atmospheric circulation, including jet variability and the storm track. Further increases in oceanic and atmospheric resolution are expected to lead to additional improvements in the representation of Euro-Atlantic climate.

Atmospheric blocking events are persistent tropospheric weather patterns that are associated with extreme events, such as heatwaves. In the summer, they primarily occur at high latitudes, e.g., over northern Eurasia. However, blocking frequency over these regions is underestimated in climate models, and often overestimated over the midlatitudes, while causes for such discrepancies remain elusive. To improve model representation of blocking frequency, it is important to understand different processes that affect it. Here, we explore blocking frequency in a reanalysis and experiments with an atmospheric general circulation model forced with different sea surface temperature (SSTs) profiles. The configurations range from an idealized no SST-front experiment and prescribing idealized SST-front in different regions to prescribing realistic climatological SSTs. Surprisingly, this reveals that more idealized (realistic) experiments lead to more (less) realistic blocking frequency. We find that weaker (less realistic) blocking frequency over northern Eurasia is primarily caused by the circulation changes related to SST gradients over the North Pacific. This suggests an important role of inter-basin interactions between the Atlantic and the Pacific. Additionally, tropical teleconnections can also play a role. This may suggest that models struggle with circulation response to SSTs in boreal summer, especially over the North Pacific. Thus, this work has implications for simulating (future) summer heat (and other) extremes over the high latitudes.

We investigate the uncertainty (i.e., inter-model spread) in future projections of the boreal winter climate, based on the forced response of ten models from the CMIP5 following the RCP8.5 scenario. The uncertainty in the forced response of sea level pressure (SLP) is large in the North Pacific, the North Atlantic, and the Arctic. A major part of these uncertainties (31%) is marked by a pattern with a center in the northeastern Pacific and a dipole over the northeastern Atlantic that we label as the Pacific–Atlantic SLP uncertainty pattern (PA∆SLP). The PA∆SLP is associated with distinct global sea surface temperature (SST) and Arctic sea ice cover (SIC) perturbation patterns. To better understand the nature of the PA∆SLP, these SST and SIC perturbation patterns are prescribed in experiments with two atmospheric models (AGCMs): CAM4 and IFS. The AGCM responses suggest that the SST uncertainty contributes to the North Pacific SLP uncertainty in CMIP5 models, through tropical–midlatitude interactions and a forced Rossby wavetrain. The North Atlantic SLP uncertainty in CMIP5 models is better explained by the combined effect of SST and SIC uncertainties, partly related to a Rossby wavetrain from the Pacific and air-sea interaction over the North Atlantic. Major discrepancies between the CMIP5 and AGCM forced responses over northern high-latitudes and continental regions are indicative of uncertainties arising from the AGCMs. We analyze the possible dynamic mechanisms of these responses, and discuss the limitations of this work.

As a major mode of annular variability in the Southern Hemisphere, the baroclinic annular mode (BAM) represents the pulsing of extratropical eddy activity. Focusing mainly on subweekly disturbances, this study assesses the impacts of a midlatitude oceanic frontal zone on the BAM and its dynamics through a set of “aquaplanet” atmospheric general circulation model experiments with zonally uniform sea surface temperature (SST) profiles prescribed. Though idealized, one experiment with realistic frontal SST gradient reasonably well reproduces observed BAM-associated anomalies as a manifestation of a typical life cycle of migratory baroclinic disturbances. Qualitatively, these BAM features are also simulated in the other experiment where the frontal SST gradient is removed. However, the BAM-associated variability weakens markedly and shifts equatorward, in association with the corresponding modifications in the climatological-mean storm track activity. The midlatitude oceanic frontal zone amplifies and anchors the BAM variability by restoring near-surface baroclinicity through anomalous sensible heat supply from the ocean and moisture supply to cyclones, although the BAM is essentially a manifestation of atmospheric internal dynamics. Those experiments and observations further indicate that the BAM modulates momentum flux associated with transient disturbances to induce a modest but robust meridional shift of the polar-front jet, suggesting that the BAM can help maintain the southern annular mode. They also indicate that the quasi-periodic behavior of the BAM is likely to reflect internal dynamics in which atmospheric disturbances on both subweekly and longer time scales are involved.

The North Atlantic SLP response is better explained by the joint effect of SST and SIC perturbations, which is partly related to the Rossby wavetrain from the Pacific and the air–sea interactions over the North Atlantic. However, these responses shift westward relative to PAΔSLP, where in CAM4 it is related to the low-frequency transient eddy forcing. The magnitude of these responses in the two AGCMs is largely different. Thus, constraining only the SST and SIC projections might not alone help constrain future climate projections. We should investigate the role of other factors in these uncertainties, such as the atmosphere–SST–SIC coupled dynamics.

While the climatological-mean sensible and latent heat fluxes are remarkably well described using climatological-mean fields in the bulk flux formulas, this study shows that a significant fraction of the climatological-mean wind speed in the midlatitudes is associated with wind variations on synoptic time scales. Hence, the prevailing wind direction associated with the most intense air–sea heat exchange can differ from the mean wind direction. To pinpoint these striking differences between the climatological and synoptic viewpoint, this study presents a global climatology of the prevailing surface wind direction during air–sea heat exchanges calculated for instantaneous and time-averaged reanalysis data. The interpretation of the fluxes in the lower latitudes is basically unaffected by the different time averages, highlighting the time-mean nature of the circulation in the lower latitudes. In the midlatitudes, however, the prevailing wind direction features a significant equatorward component for subweekly time averages and reverts to pure westerlies for longer time averages. These findings pinpoint the necessity to consider subweekly time scales, in particular along the midlatitude SST fronts, to describe the air–sea heat exchange in a physically consistent way.

Early studies have suggested a linkage between the surface warming over the Barents-Kara Seas and the strength of the Siberian high in boreal winter. Here, we show that the linkage is not stable, and with an apparent interdecadal change in the late-1990s. Coinciding with Arctic surface warm anomalies in recent decades (1997–2017), the Siberian high has been significantly intensified, the East Asian jet stream has expanded westward, and an apparent Rossby wave has propagated from the Arctic to East Asia, suggesting an atmospheric teleconnection between midlatitudes and Arctic. In contrast, midlatitude atmospheric circulation anomalies coinciding with Arctic surface warm anomalies were barely statistically significant during 1979–1996. The associated strong anomalous ascending/descending motions and divergent/convergent upper troposphere air masses over the Arctic-Eurasian sector seem to have favored the midlatitude-Arctic linkage during 1997–2017. We further hypothesize that Arctic mid-tropospheric warming plays a crucial role for the linkage between midlatitudes and Arctic in boreal winter. Multi-model simulations support this, and also point to internal atmospheric variability as the cause for the interdecadal shift in the strength of the midlatitude-Arctic linkage.

The response of the Northern Hemisphere (NH) stratosphere to climate change has been usually studied within the classical Transformed Eulerian Mean framework, which focuses mainly on the impact of the resolved atmospheric waves. The role of the non-conservative (or wave-free) processes (like diabatic heating and diffusive potential vorticity mixing) in setting the stratospheric response to climate change remains poorly understood. Here we use different stand-alone atmospheric model experiments and the newly developed Finite Amplitude Local Wave Activity (FALWA) theory, in order to understand the role and the origins of the non-conservative processes in the NH stratospheric response to climate change.Our model response can reproduce the well-known weakening of the NH polar stratospheric vortex and strengthening of mid-latitude and subtropical stratospheric westerlies. It is shown that the overall structure of the wintertime response of the NH stratosphere to climate change is maintained mainly by the ocean-induced non-conservative processes with limited contribution of the wave-induced conservative dynamics. In particular, the tropical ocean warming due to climate change maintains the wave free component of the westerly wind, which setup the background wind for poleward wave propagation and the associated wave-induced weakening of the polar stratospheric vortex. The FALWA budget reveals that the weak response of the conservative (or wave induced) component of the stratospheric westerly is maintained mainly by the eddy meridional potential vorticity (PV) transport (or EP-flux divergence) against the non-conservative diffusive PV-mixing. Our work requires the consideration of the non-conservative processes for an accurate dynamical understanding of the stratospheric response to climate change.

Midlatitude oceanic fronts play an important role in the air-sea coupled weather and climate system. Created by the confluence of warm and cool oceanic western boundary currents, the strong sea-surface temperature (SST) gradient is maintained throughout the year. The climatological mean turbulent air-sea heat exchange maximizes along these SST fronts and collocates with the major atmospheric storm tracks. A recent study identified that the air-sea heat exchange along the SST front mainly occurs on sub-weekly time scales, associated with synoptic atmospheric disturbances. This implies a crucial role of air-sea moisture exchange along the SST fronts on the atmospheric water cycle through the intensification of atmospheric cyclones and the associated precipitation. In this study, we investigate this influence of the SST front on the atmospheric water cycle by analyzing the atmospheric response to different prescribed SST in the Atmospheric general circulation model For the Earth Simulator (AFES). Changing the latitude of the prescribed zonally symmetric SST in aqua-planet configuration, we find a distinctive response in convective and large-scale precipitation, surface latent and sensible heat fluxes, as well as diabatic heating and moistening with respect to the latitude of SST front. Upward surface latent heat flux and convective precipitation always maximize along the equatorward flank of SST front. On the other hand, large-scale precipitation is always located on the poleward flank of the SST front, in correspondence with the maximum atmospheric moisture flux convergence. The moisture flux convergence is mainly associated with midlatitude eddies and not with the time mean transport. This highlights the influence of mid-latitude SST fronts on the atmospheric water cycle through the organization of atmospheric storm track.

IPCC climate models (CMIP3/5) predict a poleward shift of the Southern Hemisphere (SH) jet stream under global warming, with a large spread across the models. Efforts to find emergent constraints for the future jet shift (response) have relied on the simulated present-day jet position (observable). However, this has been investigated primarily in a zonal-mean framework, which averages out important zonal asymmetries. In this study, we revisit the problem allowing for variations in the longitude, height and season of the response to gain a better physical understanding of the nature of the future jet shift in individual models. Results from a manual data analysis will help guide an exploration of the problem using a big data approach, in particular, the application of a genetic algorithm that finds optimal solutions based on iterative random selection within large sample data spaces.

Semi-idealized Atmospheric General Circulation-Model (AGCM) experiments are used, in order to study the different aspects of the hemisphere-scale wintertime troposphere/stratosphere-coupled circulation that are maintained by the North Atlantic and Pacific Ocean Western Boundary Currents (OWBCs). Here we show that the North Atlantic and Pacific OWBCs jointly maintain and shape the wintertime hemispheric circulation and its leading mode of variability Northern Annular Mode (NAM). The OWBCs energize baroclinic waves that reinforce quasi-annular hemispheric structure in the tropospheric eddy-driven jetstreams and NAM variability. Without the OWBCs, the wintertime NAM variability is much weaker and its impact on the continental and maritime surface climate is largely insignificant. Atmospheric energy redistribution caused by the OWBCs acts to damp the near-surface atmospheric baroclinicity and compensates the associated oceanic meridional energy transport in agreement with the Bjerknes compensation. Furthermore, the OWBCs substantially weaken the wintertime stratospheric polar vortex by enhancing the upward planetary wave propagation, and thereby affecting both stratospheric and tropospheric NAM-annularity. It is shown that the impact of OWBCs on northern hemisphere circulation has significant implication for stratosphere/troposphere dynamical coupling, time-scales on the NAM, frequency of Sudden stratospheric warming and potential formation of polar stratospheric clouds. Concurrent with the slowdown of global warming during 2002–2013, the wintertime land surface air temperatures over Eurasia, North America, Africa, Australia, South America, and Greenland experienced notable cooling trends. The oceanic effects on the continental cooling trends are here investigated using two sets of uncoupled experiments with six different climate models. Daily and annually varying sea ice is prescribed for both sets of experiments, while daily and annually varying SST is used in the first set (EXP1) and daily and annually repeating climatological mean SST in the second set (EXP2). All six models capture the slowdown of global-mean land surface air temperature during 2002–2013 winters in EXP1 only. The slowdown concurs with a negative phase of the Pacific Decadal Oscillation (PDO), indicating that PDO plays an important role in modulating the global warming signal. Not all ensemble members capture the cooling trends over the continents, suggesting additional contribution from internal atmospheric variability. The individual impact of North Atlantic and Pacific Ocean Western Boundary Currents (OWBCs) on the tropospheric circulation has recently been studied in depth. There is a growing demand for skillful prediction systems in the Arctic. Using the Norwegian Climate Prediction Model that combines the fully coupled Norwegian Earth System Model and the Ensemble Kalman filter, we present a system that performs both, weakly coupled data assimilation (wCDA) when assimilating ocean hydrography (by updating the ocean alone) and strongly coupled data assimilation when assimilating sea ice concentration (SIC) (by jointly updating the sea ice and ocean). We assess the seasonal prediction skill of this version of the Norwegian Climate Prediction Model, the first climate prediction system using strongly coupled data assimilation, by performing retrospective predictions (hindcasts) for the period 1985 to 2010. To better understand the origins of the prediction skill of Arctic sea ice, we compare this version with a version that solely performs wCDA of ocean hydrography. The reanalysis that assimilates just ocean data exhibits skillful hydrography in the upper Arctic Ocean and features an improved sea ice state, such as improved summer SIC in the Barents Sea, or reduced biases in sea ice thickness. Skillful prediction of SIE up to 10–12 lead months are only found during winter in regions of a relatively deep ocean mixed layer outside the Arctic basin. Additional DA of SIC data notably further corrects the initial seaice state, confirming the applicability of the results of Kimmritz et al. (2018) in a historical setting. The resulting prediction skill of SIE is widely enhanced compared to predictions initialized through wCDA of only ocean data. Particularly high skill is found for July‐initialized autumn SIE predictions.

International audience; Coordinated numerical ensemble experiments with six different state-of-the-art atmosphere models have been used in order to evaluate the respective impact of the observed Arctic sea ice and sea surface temperature (SST) variations on air temperature variations in mid and high latitude land areas. Two sets of experiments have been designed; in the first set (EXP1), observed daily sea ice concentration and SST variations are used as lower boundary forcing over 1982-2014 while in the second set (EXP2) the SST variations are replaced by the daily SST climatology. The observed winter 2 m air temperature (T2m) variations are relatively well reproduced in a number of mid and high latitude land areas in EXP1, with best agreement in southwestern North America and northern Europe. Sea ice variations are important for the interannual T2m variations in northern Europe but have limited impact on all other mid and high latitude land regions. In particular, sea ice variations do not contribute to the observed opposite variations in the Arctic and mid latitude in our model experiments. The spread across ensemble members is large and many ensemble members are required to reproduce the observed T2m variations over northern Europe in our models. The amplitude of T2m anomalies in the coldest observed winters over northern Europe is not reproduced by our multi-model ensemble means. However, the sea ice conditions in these respective winters and mainly the thermodynamic response to the ice anomalies lead to an enhanced likelihood for occurrence of colder than normal winters and extremely cold winters. Still, the main reason for the observed extreme cold winters is internal atmospheric dynamics. The coldest simulated northern European winters in EXP1 and EXP2 between 1982 and 2014 show the same large scale T2m and atmospheric circulation anomaly patterns as the observed coldest winters, indicating that the models are well able to reproduce the processes, which cause these cold anomalies. The results are robust across all six models used in this study.

A midlatitude oceanic frontal zone is a confluent region of warm and cool ocean currents, characterized by a strong meridional gradient in both sea surface temperature (SST) and surface air temperature (SAT). While recent observational and modeling studies indicate potential impacts of midlatitude oceanic fronts on the extratropical climatological circulation, including storm tracks and eddy-driven westerlies, their impacts on the atmospheric-dominant low-frequency variability (i.e., the annular mode) still remain to be understood. This study explores possible impacts of midlatitude oceanic frontal zones on annular mode signatures in the wintertime Southern Hemisphere (SH). To mimic the SH, sets of idealized aquaplanet experiments are conducted with zonally symmetric distributions of SST prescribed globally at the lower boundary of an atmospheric general circulation model. By systematically changing the latitude of frontal gradient in the SST profile, the experiments reveal that the characteristics of the wintertime annular mode exhibit strong sensitivity to the position of the SST front if situated at midlatitude or subpolar latitude. The annular mode may be interpreted as a manifestation of wobble of the extratropical tropospheric circulation between two “dynamical regimes”—one under the strong influence of SST gradient and the other under the strong control of atmospheric internal dynamics unrelated to the lower-boundary condition. In fact, this interpretation offers insight into the observed interbasin differences in the wintertime signature of the southern annular mode (SAM) that are embedded in the zonally symmetric anomalies. The findings suggest a possible reinterpretation of the climatological-mean state observed in the wintertime SH as the superposition of those two dynamical regimes.

Mitochondria are essential for neuronal function, providing the energy required to power neurotransmission, and fulfilling many important additional roles. In neurons, mitochondria must be efficiently transported to sites, including synapses, where their functions are required. Neurons, with their highly elongated morphology, are consequently extremely sensitive to defective mitochondrial trafficking which can lead to neuronal ill-health/death. We recently demonstrated that DISC1 associates with mitochondrial trafficking complexes where it associates with the core kinesin and dynein adaptor molecule TRAK1. We now show that the DISC1 interactors NDE1 and GSK3β also associate robustly with TRAK1 and demonstrate that NDE1 promotes retrograde axonal mitochondrial movement. GSK3β is known to modulate axonal mitochondrial motility, although reports of its actual effect are conflicting. We show that, in our system, GSK3β promotes anterograde mitochondrial transport. Finally, we investigated the influence of cAMP elevation upon mitochondrial motility, and found a striking increase in mitochondrial motility and retrograde movement. DISC1, NDE1, and GSK3β are implicated as risk factors for major mental illness. Our demonstration that they function together within mitochondrial trafficking complexes suggests that defective mitochondrial transport may be a contributory disease mechanism in some cases of psychiatric disorder.

Despite the strong association of azithromycin (AZM), a macrolide antibiotic, and levofloxacin (LVF), a quinolone antibiotic, to sediment, sorption data are scarce. We conducted sorption experiments with eight river sediments, their major clay minerals (illite and chlorite), a highly negatively charged clay mineral (montmorillonite), and an organic-matter-rich soil (Andosol). The sorption of AZM and LVF to the sediments was influenced by the concentration and type of coexisting inorganic cations as much as by reported organic cations. In addition, their linear sorption coefficients (Kd) to sediments were correlated with cation exchange capacity (CEC) but not organic carbon content, so cation exchange is the dominant sorption mechanism. Multiple linear regression analysis showed improved prediction of sediment Kd from CEC contributed by minerals and organic matter for AZM, but not for LVF. Kcec (= Kd/CEC) values of AZM were 2–3 orders of magnitude higher on minerals than on Andosol, but those of LVF ranged within a factor of 4. Therefore, mineral and organic components need to be separated in estimating AZM sorption to sediments. Sediment Kd values of AZM and LVF were satisfactorily predicted by a cation-exchange-based model using individual Kcec values on illite, chlorite, and Andosol (mean absolute error of 0.57 and 0.22 log units, respectively). Kcec values on montmorillonite and chlorite ranged within a factor of about 3 from those of illite for both antibiotics, and Kcec differences by mineral type would generally be negligible in model estimation. Because AZM was sorbed mostly to minerals in sediments, the model and sorption data can be applicable to various soils or sediments. Overall, the trend of LVF sorption corresponds to reported sorption of other organic cations, whereas remarkably higher AZM Kcec to minerals than to Andosol is attributable to its large lactone ring, higher molecular weight, or two charged amines.

The neuromodulatory gene DISC1 is disrupted by a t(1;11) translocation that is highly penetrant for schizophrenia and affective disorders, but how this translocation affects DISC1 function is incompletely understood. N-Methyl-D-Aspartate receptors (NMDAR) play a central role in synaptic plasticity and cognition, and are implicated in the pathophysiology of schizophrenia through genetic and functional studies. We show that the NMDAR subunit GluN2B complexes with DISC1-associated trafficking factor TRAK1, while DISC1 interacts with the GluN1 subunit and regulates dendritic NMDAR motility in cultured mouse neurons. Moreover, in the first mutant mouse that models DISC1 disruption by the translocation, the pool of NMDAR transport vesicles and surface/synaptic NMDAR expression are increased. Since NMDAR cell surface/synaptic expression is tightly regulated to ensure correct function, these changes in the mutant mouse are likely to affect NMDAR signalling and synaptic plasticity. Consistent with these observations, RNASeq analysis of translocation carrier-derived human neurons indicates abnormalities of excitatory synapses and vesicle dynamics. RNASeq analysis of the human neurons also identifies many differentially expressed genes previously highlighted as putative schizophrenia and/or depression risk factors through large-scale genome-wide association and copy number variant studies, indicating that the translocation triggers common disease pathways that are shared with unrelated psychiatric patients. Altogether our findings suggest that translocation-induced disease mechanisms are likely to be relevant to mental illness in general, and that such disease mechanisms include altered NMDAR dynamics and excitatory synapse function. This could contribute to the cognitive disorders displayed by translocation carriers.

Understanding the mechanism governing the ongoing global warming is a major challenge facing our society and its sustainable growth. Together with the CO2-forced warming, the concurrent polar sea ice loss might also have contributed to the observed Arctic warming amplification and also to the cooling trends over Eurasia through a dynamical teleconnection. However, previous individual modeling studies suggest widely different findings on the role of sea ice loss in Northern Hemisphere climate change. To help resolve this controversy, we used satellite-derived sea ice and sea-surface temperature to run coordinated hindcast experiments with five different atmospheric general circulation models. The multimodel ensemble-mean results presented in the paper reduce biases of each model and eliminate atmospheric internal unforced variability, and thus provide the best estimate to date of the signal of the polar sea ice loss. The results suggest that the impact of sea ice seems critical for the Arctic surface temperature changes, but the temperature trends elsewhere seem rather due to either sea-surface temperature changes or atmospheric internal variability. They give clear guidance on how to provide society with more accurate climate change attributions. Our work is of interest to stakeholders of countries in the Northern Hemisphere middle and high latitudes.

Disrupted-In-Schizophrenia 1 (DISC1) is a candidate risk factor for schizophrenia, bipolar disorder and severe recurrent depression. Here, we demonstrate that DISC1 associates robustly with trafficking-protein-Kinesin-binding-1 which is, in turn, known to interact with the outer mitochondrial membrane proteins Miro1/2, linking mitochondria to the kinesin motor for microtubule-based subcellular trafficking. DISC1 also associates with Miro1 and is thus a component of functional mitochondrial transport complexes. Consistent with these observations, in neuronal axons DISC1 promotes specifically anterograde mitochondrial transport. DISC1 thus participates directly in mitochondrial trafficking, which is essential for neural development and neurotransmission. Any factor affecting mitochondrial DISC1 function is hence likely to have deleterious consequences for the brain, potentially contributing to increased risk of psychiatric illness. Intriguingly, therefore, a rare putatively causal human DISC1 sequence variant, 37W, impairs the ability of DISC1 to promote anterograde mitochondrial transport. This is likely related to a number of mitochondrial abnormalities induced by expression of DISC1-37W, which redistributes mitochondrial DISC1 and enhances kinesin mitochondrial association, while also altering protein interactions within the mitochondrial transport complex.

Disrupted-In-Schizophrenia 1 (DISC1) was identified as a risk factor for psychiatric illness through its disruption by a balanced chromosomal translocation, t(1;11)(q42.1;q14.3), that co-segregates with schizophrenia, bipolar disorder and depression. We previously reported that the translocation reduces DISC1 expression, consistent with a haploinsufficiency disease model. Here we report that, in lymphoblastoid cell lines, the translocation additionally results in the production of abnormal transcripts due to the fusion of DISC1 with a disrupted gene on chromosome 11 (DISC1FP1/Boymaw). These chimeric transcripts encode abnormal proteins, designated CP1, CP60 and CP69, consisting of DISC1 amino acids 1–597 plus 1, 60 or 69 amino acids, respectively. The novel 69 amino acids in CP69 induce increased α-helical content and formation of large stable protein assemblies. The same is predicted for CP60. Both CP60 and CP69 exhibit profoundly altered functional properties within cell lines and neurons. Both are predominantly targeted to mitochondria, where they induce clustering and loss of membrane potential, indicative of severe mitochondrial dysfunction. There is currently no access to neural material from translocation carriers to confirm these findings, but there is no reason to suppose that these chimeric transcripts will not also be expressed in the brain. There is thus potential for the production of abnormal chimeric proteins in the brains of translocation carriers, although at substantially lower levels than for native DISC1. The mechanism by which inheritance of the translocation increases risk of psychiatric illness may therefore involve both DISC1 haploinsufficiency and mitochondrial deficiency due to the effects of abnormal chimeric protein expression. The late twentieth century was marked by a significant summertime trend in the Southern Annular Mode (SAM), the dominant mode of tropospheric variability in the extratropical Southern Hemisphere (SH). This trend with poleward shifting tropospheric westerlies was attributed to downward propagation of stratospheric changes induced by ozone depletion. However, the role of the ocean in setting the SAM response to ozone depletion and its dynamical forcing remains unclear. Here we show, using idealized experiments with a state-of-the-art atmospheric model and analysis of Intergovernmental Panel on Climate Change climate simulations, that frontal sea surface temperature gradients in the midlatitude SH are critical for translating the ozone-induced stratospheric changes down to the surface. This happens through excitation of wave forcing, which controls the vertical connection of the tropospheric SAM with the stratosphere and shows the importance of internal tropospheric dynamics for stratosphere/troposphere coupling. Thus, improved simulation of oceanic fronts may reduce uncertainties in simulating SH ozone-induced climate changes. publishedVersion

Disrupted-In-Schizophrenia 1 (DISC1) was identified as a candidate gene for schizophrenia. DISC1 is disrupted by a balanced t(1;11)(q42.1;q14.3) translocation segregating with schizophrenia and related psychiatric illness in a large Scottish family. Here, we show that DISC1 interacts via its globular domain with the p40 subunit of the eukaryotic translation initiation factor 3. Furthermore, we found that overexpression of DISC1 in SH-SY5Y cells induces the assembly of eIF3- and TIA-1-positive stress granules (SGs), discrete cytoplasmic granules formed in response to environmental stresses. Our findings suggest that DISC1 may function as a translational regulator and may be involved in stress response.

Background: hDLG, the human homologue of the Drosophila tumour suppressor dlg, functions as a scaffolding protein that facilitates the transmission of diverse downstream signals. hDLG possesses multiple protein-binding domains, including three PDZ domains, an SH3 domain, a HOOK domain and a guanylate kinase-like (GK) domain. Results: We studied the significance of the PDZ, SH3, HOOK and GK domains in the cytoplasmic localization of hDLG. We found that mutation of the SH3 or GK domain, but not the PDZ domain, resulted in a re-localization of hDLG to the nucleus. Furthermore, hDLG was found to possess a potential nuclear localization signal in the HOOK domain. Conclusion: These results suggest that the SH3, HOOK and GK domains of hDLG are important for its cytoplasmic localization.

Disrupted-In-Schizophrenia 1 (DISC1), a strong genetic candidate for psychiatric illness, encodes a multicompartmentalized molecular scaffold that regulates interacting proteins with key roles in neurodevelopment and plasticity. Missense DISC1 variants are associated with the risk of mental illness and with brain abnormalities in healthy carriers, but the underlying mechanisms are unclear. We examined the effect of rare and common DISC1 amino acid substitutions on subcellular targeting. We report that both the rare putatively causal variant 37W and the common variant 607F independently disrupt DISC1 nuclear targeting in a dominant-negative fashion, predicting that DISC1 nuclear expression is impaired in 37W and 607F carriers. In the nucleus, DISC1 interacts with the transcription factor Activating Transcription Factor 4 (ATF4), which is involved in the regulation of cellular stress responses, emotional behaviour and memory consolidation. At basal cAMP levels, wild-type DISC1 inhibits the transcriptional activity of ATF4, an effect that is weakened by both 37W and 607F independently, most likely as a consequence of their defective nuclear targeting. The common variant 607F additionally reduces DISC1/ATF4 interaction, which likely contributes to its weakened inhibitory effect. We also demonstrate that DISC1 modulates transcriptional responses to endoplasmic reticulum stress, and that this modulatory effect is ablated by 37W and 607F. By showing that DISC1 amino acid substitutions associated with psychiatric illness affect its regulatory function in ATF4-mediated transcription, our study highlights a potential mechanism by which these variants may impact on transcriptional events mediating cognition, emotional reactivity and stress responses, all processes of direct relevance to psychiatric illness.

[1] Major “storm tracks”, where migratory cyclones and anticyclones recurrently develop, are observed around midlatitude oceanic frontal zones with strong meridional gradient of sea-surface temperature (SST). A set of atmospheric general circulation model experiments is performed with zonally uniform SST prescribed at the model lower boundary. The latitudinal SST profile for each hemisphere is characterized by a single front. The frontal latitude is varied systematically from one experiment to another, while the intensity of the frontal gradient is kept unchanged. Though idealized, the experiments reveal a climatological tendency for a low-level storm track to be organized along or slightly poleward of the SST front if located in the subtropics or midlatitudes. As a surface manifestation of an eddy-driven polar-front jet (PFJ), surface westerly axis tends to form on the poleward flank of the front. This anchoring effect of the SST front is also hinted at upper levels, but the climatological positions of the storm track and PFJ are less sensitive to the frontal latitude. For the SST front at subpolar latitude, the joint primary axes of the upper-level storm track and PFJ form in midlatitudes away from the SST front. Their positions correspond to their counterpart simulated with a particular SST profile from which frontal gradient has been removed, suggesting that the anchoring effect of a subpolar SST front on the storm track and PFJ is overshadowed by atmospheric internal dynamics, namely, the self-maintenance mechanism of a midlatitude storm track and PFJ through their interactions.

Disrupted in schizophrenia 1 (DISC1) is a leading candidate susceptibility gene for schizophrenia, bipolar disorder and recurrent major depression, which has been implicated in other psychiatric illnesses of neurodevelopmental origin, including autism. DISC1 was initially identified at the breakpoint of a balanced chromosomal translocation, t(1;11) (q42.1;14.3), in a family with a high incidence of psychiatric illness. Carriers of the translocation show a 50% reduction in DISC1 protein levels, suggesting altered DISC1 expression as a pathogenic mechanism in psychiatric illness. Altered DISC1 expression in the post-mortem brains of individuals with psychiatric illness and the frequent implication of non-coding regions of the gene by association analysis further support this assertion. Here, we provide the first characterization of the DISC1 promoter region. Using dual luciferase assays, we demonstrate that a region -300 to -177 bp relative to the transcription start site (TSS) contributes positively to DISC1 promoter activity, while a region -982 to -301 bp relative to the TSS confers a repressive effect. We further demonstrate inhibition of DISC1 promoter activity and protein expression by forkhead-box P2 (FOXP2), a transcription factor implicated in speech and language function. This inhibition is diminished by two distinct FOXP2 point mutations, R553H and R328X, which were previously found in families affected by developmental verbal dyspraxia. Our work identifies an intriguing mechanistic link between neurodevelopmental disorders that have traditionally been viewed as diagnostically distinct but which do share varying degrees of phenotypic overlap.

Nuclear distribution factor E-homolog 1 (NDE1), Lissencephaly 1 (LIS1), and NDE-like 1 (NDEL1) together participate in essential neurodevelopmental processes, including neuronal precursor proliferation and differentiation, neuronal migration, and neurite out-growth. NDE1/LIS1/NDEL1 interacts with Disrupted in Schizophrenia 1 (DISC1) and the cAMP-hydrolyzing enzyme phosphodiesterase 4 (PDE4). DISC1, PDE4, NDE1, and NDEL1 have each been implicated as genetic risk factors for major mental illness. Here, we demonstrate that DISC1 and PDE4 modulate NDE1 phosphorylation by cAMP-dependent protein kinase A (PKA) and identify a novel PKA substrate site on NDE1 at threonine-131 (T131). Homology modeling predicts that phosphorylation at T131 modulates NDE1–LIS1 and NDE1–NDEL1 interactions, which we confirm experimentally. DISC1–PDE4 interaction thus modulates organization of the NDE1/NDEL1/LIS1 complex. T131-phosphorylated NDE1 is present at the postsynaptic density, in proximal axons, within the nucleus, and at the centrosome where it becomes substantially enriched during mitosis. Mutation of the NDE1 T131 site to mimic PKA phosphorylation inhibits neurite outgrowth. Thus PKA-dependent phosphorylation of the NDE1/LIS1/NDEL1 complex is DISC1–PDE4 modulated and likely to regulate its neural functions.

International audience; Disrupted-In-Schizophrenia 1 (DISC1) is a risk factor for schizophrenia and other major mental illnesses. Its protein binding partners include the Nuclear Distribution Factor E Homologs (NDE1 and NDEL1), LIS1, and phosphodiesterases 4B and 4D (PDE4B and PDE4D). We demonstrate that NDE1, NDEL1 and LIS1, together with their binding partner dynein, associate with DISC1, PDE4B and PDE4D within the cell, and provide evidence that this complex is present at the centrosome. LIS1 and NDEL1 have been previously suggested to be synaptic, and we now demonstrate localisation of DISC1, NDE1, and PDE4B at syn-apses in cultured neurons. NDE1 is phosphorylated by cAMP-dependant Protein Kinase A (PKA), whose activity is, in turn, regulated by the cAMP hydrolysis activity of phosphodiesterases, including PDE4. We propose that DISC1 acts as an assembly scaffold for all of these proteins and that the NDE1/ NDEL1/LIS1/dynein complex is modulated by cAMP levels via PKA and PDE4.

SummaryTo support the role of DISC1 in human psychiatric disorders, we identified and analyzed two independently derived ENU-induced mutations in Exon 2 of mouse Disc1. Mice with mutation Q31L showed depressive-like behavior with deficits in the forced swim test and other measures that were reversed by the antidepressant bupropion, but not by rolipram, a phosphodiesterase-4 (PDE4) inhibitor. In contrast, L100P mutant mice exhibited schizophrenic-like behavior, with profound deficits in prepulse inhibition and latent inhibition that were reversed by antipsychotic treatment. Both mutant DISC1 proteins exhibited reduced binding to the known DISC1 binding partner PDE4B. Q31L mutants had lower PDE4B activity, consistent with their resistance to rolipram, suggesting decreased PDE4 activity as a contributory factor in depression. This study demonstrates that Disc1 missense mutations in mice give rise to phenotypes related to depression and schizophrenia, thus supporting the role of DISC1 in major mental illness.

Extreme rainfall events in the South Pacific are widespread and affected by various factors on different time scales. We use daily rainfall data from 20 stations over the South Pacific to investigate the characteristics of extreme rainfall events from 1979 to 2018. For regional analysis, we group the stations into three clusters characterizing the western, the central, and the far eastern regions of the South Pacific Convergence Zone (SPCZ). Extreme rainfall events contribute to roughly 20% of the seasonal mean rainfall in all three clusters. Among all four factors considered, tropical cyclones (TCs) cause the highest increase in the probability (ΔpwesternSPCZ~286%, ΔpcentralSPCZ~84%, ΔpfareasternSPCZ~189%) of extreme rainfall events. The Madden-Julian Oscillation (MJO) is the second most important factor affecting the probability of extreme rainfall events, increasing it by 30–60% when the MJO is active over the western SPCZ in phases 5–6, over the central SPCZ in phases 6–7, and over the far eastern SPCZ in phases 8–1. The probability is reduced by the same order of magnitudes during the opposite dry phases of the MJO, i.e., phases 1–3 for the western and central SPCZ, and 3–6 for far the eastern SPCZ region. The probability of extreme rainfall events increases during La-Niña (El-Niño) conditions to the southwest (southeast) of the mean SPCZ by 27% (31%); however, the impact of the El-Niño and Southern Oscillation (ENSO) along the SPCZ is not apparent. Dynamical analysis shows that the favorable conditions for generating extreme rainfall events are associated with northwesterly moisture transport and its convergence. The impact of TCs, MJO, and ENSO on rainfall extreme events can be partly understood considering this dynamical analysis. Extratropical Rossby waves can trigger tropical disturbances, but their impact on extreme rainfall events is generally less important than of the TCs, MJO, and ENSO. publishedVersion
