Undaria pinnatifida (U. pinnatifida) polysaccharides (UPPS) are considered to be the major bioactive components of U. pinnatifida. The aim of the present study was to investigate the separation, sulfated modification, characterization and monosaccharide composition of UPPS. The optimal processing conditions were as follows: Distilled water-to-solid ratio, 50 ml/g; extraction time, 300 min; and extraction temperature, 90˚C. The major polysaccharide fraction of U. pinnatifida (UPPS-B1) was purified via DEAE-52 and Sephadex G-200 column chromatography. The chlorosulfonic acid-pyridine method was applied for sulfation modification. UPPS-B1 and sulfated (S)-UPPS-B1 were characterized via chemical analysis, ultraviolet-visible and Fourier-transformed infrared spectroscopy, gas chromatography and high-performance liquid chromatography. The total sugar content of UPPS-B1 and S-UPPS-B1 was 79.78 and 77.28%, respectively. The sulfate radical content of UPPS-B1 and S-UPPS-B1 was 8.53 and 29.12%, whilst the content of uronic acid was 9.29 and 7.98%, respectively. The average molecular weight of UPPS-B1 and S-UPPS-B1 was determined to be 37 and 110 kD, respectively. UPPS-B1 was considered to be a heteropolysaccharide composed of xylose, mannose, glucose and galactose at a ratio of 7.9:8.7:12.0:9.8. In addition, S-UPPS-B1 was a heteropolysaccharide composed of xylose, mannose, glucose and galactose at a ratio of 1.0:9.7:6.4:1.6. The results of the tumor growth inhibition experiment demonstrated that UPPS-B1 exhibited anti-tumor activity in vivo, which was improved following sulfation to yield S-UPPS-B1.

The aggregation-induced emission (AIE) mechanism of restriction of double-bond rotation (RDBR) was utilized to design an excellent solid emitter and sensor for the first time. Thus, cis-tetraphenylethylene (TPE) macrocycle diammoniums were synthesized and bound to a DNA chain by its two ammonium arms. The formed TPE dicycle at the cis position restricted the rotation of the double bond in both the ground and excited states, resulting in AIE enhancement, chiroptical performance enhancement, and sensing enhancement.

Purpose Laryngeal squamous-cell carcinoma (LSCC) is the second most common malignant tumor of head and neck squamous cell carcinoma. The study was aimed to identify key long non-coding RNAs (lncRNAs) biomarkers for LSCC. Methods Differentially expressed lncRNAs (DElncRNAs) and mRNAs (DEmRNAs) between LSCC and adjacent tissues were obtained based on The Cancer Genome Atlas. DElncRNA-DEmRNAs co-expression and DElncRNA-nearby-target DEmRNA interaction networks were constructed. Receiver operating characteristic and survival analysis were performed. A published dataset were as used to validate the result of bioinformatics analysis. Results We obtained 1103 DEmRNAs and 306 DElncRNAs between LSCC and adjacent tissues. A total of 338 DElncRNA-DEmRNA co-expression pairs and 229 DElncRNA-nearby-target DEmRNA pairs were obtained. Ten DElncRNAs and six DEmRNAs has great diagnostic value for LSCC. HOXB9 has potential prognostic value for LSCC. The results of GSE84957 validation were generally consistent with our results. Conclusion Our study provided clues for understanding the mechanism and developing potential biomarkers for LSCC.

The trypsin-assisted extraction of polysaccharides from Allium cepa L. was optimized using the response surface methodology (RSM). The optimum extraction conditions were extraction temperature, extraction time, extraction pH, and enzyme amount of 37.16°C, 180 min, 8.57, and 5.16%, respectively. Under the optimized conditions, the yield of A. cepa L. polysaccharides (ACP) reached 9.69%, which was comparable with the predicted yield (9.73%). Mid- and high-dose ACP significantly inhibited the tumor growth (43.93%) and the tumor inhibition percentage (38.05%), which were more than 30%. The ACP could extend the survival time of H22 ascites tumor-bearing mice. Furthermore, the ACP could reduce the thymus and the spleen atrophy and significantly promoted the Con A-induced proliferation of splenocytes and elevated the serum IFN-γ and IL-2 levels. Therefore, the ACP could inhibit the tumor growth in tumor-bearing mice and regulated the immune function of mice. Practical ApplicationsThe trypsin-assisted extraction has high efficiency, is carried out through the polysaccharide extraction and the deproteinization at the same time, and is more convenient and fast than traditional methods. No detailed study on the optimization of the trypsin extraction of onion polysaccharides is available. Thus, this experiment aims to use the BBD (4 factors and 3 levels) to optimize the roles of extraction temperature, extraction time, extraction pH, and amount of enzyme on the yield of polysaccharides obtained from the fruit of A. cepa L. In addition, when looking for high-quality biological functional principles for the pharmaceutical industry, the antitumor activity of ACP was evaluated. A. cepa L. is one of the most widely cultivated and consumed crops worldwide. Polysaccharides are the main active ingredient, and studies have shown that a high intake of Allium vegetables is associated with reduced risk of cancers.

Aberrant expression of beta-1,3-N-acetylglucosaminyltransferase-3 (B3GNT3) has been frequently clarified in various cancers, however, its role in endometrial cancer (EC) has not been assessed in detail. This study aimed to investigate the biological role of B3GNT3 in EC and simply explored the detailed mechanism. The EC RNA-Seq dataset from TCGA database was applied to evaluate the expression of B3GNT3 and assess its role on prognostic value. HEC-1-A and KLE cell lines of EC were used to perform loss- and gain-of-function B3GNT3 assays respectively. Quantitative real-time PCR (qRT-PCR) and western blot were used to measure the mRNA and protein levels of indicated molecules respectively. Cell counting kit-8, clone formation tests, and Transwell assay served to determine the changes of proliferative, invasive and migratory abilities of EC cells after altering the expression of B3GNT3. B3GNT3 was found to be highly expressed in EC tissues compared to normal tissues according to the online public databases, which confirmed by the following qRT-PCR in 3 EC cell lines. Besides, high B3GNT3 expression presented a worse overall survival in EC patients as compared with low B3GNT3 expression group. Furthermore, functional experiments in vitro indicated that B3GNT3 could facilitate the cell growth, invasion and migration. Moreover, we found that downregulation of B3GNT3 significantly reduced the expression level of GTP-RhoA and GTP-RAC1, whereas upregulation of B3GNT3 presented the opposite results. The results of current study demonstrate that B3GNT3 acts as an oncogene that promotes EC cells growth, invasion and migration possibly through regulating the RhoA/RAC1 signaling pathway-related markers, suggesting that B3GNT3 may be a candidate biomarker for EC therapeutic intervention.

Nasopharyngeal carcinoma (NPC) is one of the most malignant head and neck carcinomas with unique epidemiological features. In this study, we aimed to identify the novel NPC-related genes and biological pathways, shedding light on the potential molecular mechanisms of NPC. Based on Gene Expression Omnibus (GEO) database, an integrated analysis of microarrays studies was performed to identify differentially expressed genes (DEGs) and differentially methylated genes (DMGs) in NPC compared to normal control. The genes which were both differentially expressed and differentially methylated were identified. Functional annotation and protein–protein interaction (PPI) network construction were used to uncover biological functions of DEGs. Two DNA methylation and five gene expression datasets were incorporated. A total of 1074 genes were up-regulated and 939 genes were down-regulated in NPC were identified. A total of 719 differential methylation CpG sites (DMCs) including 1 hypermethylated sites and 718 hypomethylated sites were identified. Among which, 11 genes were both DEGs and DMGs in NPC. Pathways in cancer, p53 signaling pathway and Epstein–Barr virus infection were three pathways significantly enriched pathways in DEmRNAs of NPC. The PPI network of top 50 DEGs were consisted of 191 nodes and 191 edges. Our study was helpful to elucidate the underlying mechanism of NPC and provide clues for therapeutic methods.

Ionic liquid (IL)-assisted pretreatment of lignocellulosic biomass has been extensively studied. Cellulose and hemicelluloses are rich resources of sugars for biofuels. Lignin is a valuable feedstock for aromatic-based platform chemicals. In this study, a series of ionic liquids (ILs) were prepared with one-step synthesis and were investigated for their activity to pretreat lignocelluloses. High yields of lignin (61.0% and 60.4%) were achieved through [Hpy]Cl and [Hmim]Cl pretreatment of poplar. Lignin yields of 51.7% and 50.3% were obtained with a [Hpy]Cl and [Hnmp]Cl pretreatment of bamboo. Improving enzymatic hydrolysis was observed from the regenerated poplar with [Hpy]Cl pretreatment and the regenerated bamboo with [Hmim]Cl pretreatment. The isolated fractions were characterized by FTIR (Fourier transform infrared) spectroscopy. SEM (Scanning electron microscopy) and XRD (X-ray diffractometry) were employed to examine the cellulose-rich materials. A 2D 1H–13C heteronuclear single quantum coherence (HSQC) nuclear magnetic resonance (NMR) spectroscopy was employed to analyze lignin fraction. These ionic liquids were particularly effective to extract lignin from lignocelluloses to obtain cellulose-rich materials for biofuels.

Ionic liquid (IL)-assisted pretreatment is an important step in biochemical conversion of lignocellulosic biomass into biofuels. Design for low-cost ILs that efficiently work at a relatively low pretreatment temperature with a short processing time is of great interest. In this work, a functional acidic ionic liquid, 1-H-N-methyl-2-pyrrolidonium chloride ([Hnmp]Cl), was prepared by a simple synthetic procedure in a cost-effective manner and was then investigated for use in the pretreatment of lignocellulosic biomass. Factors including temperature, time, ratio of biomass to ionic liquid, and water content were studied to determine their impact on the pretreatment of lignocellulose; 91.39% of lignin content was recovered when the corn stalk was pretreated by pure [Hnmp]Cl at 100 °C for 45 min with a biomass loading of 5%. The highest glucose yield attained was 93.20% and the highest cellobiose yield was 18.76%, when the ratio of biomass to water was kept at 1:4. The pretreatment efficacy was dependent on the pretreatment temperature and processing time, which was more efficient for pure ionic liquid with respect to lignin recovery from lignocelluloses. However, more efficient enzymatic saccharification of cellulose-rich materials was achieved with aqueous ionic liquid.

The aim of the present study was to investigate the key genes, miRNAs and pathways in hypopharyngeal squamous cell carcinoma (HPSCC) and to elucidate the mechanisms underlying HPSCC development. The gene and microRNA (miRNA) expression profiles of HPSCC tissues and adjacent normal tissues from three subjects were obtained. Differentially expressed genes (DEGs) and differentially expressed miRNAs were identified in HPSCC. Functional annotation and protein-protein interaction (PPI) network were conducted to elucidate the biological functions of DEGs. A total of 160 DEGs (16 upregulated and 144 downregulated genes) and 79 differentially expressed miRNAs (48 upregulated and 31 downregulated miRNAs) were identified in HPSCC. The deregulated genes were significantly involved in spliceosome, cell cycle and RNA degradation. In the PPI network, S-phase kinase associated protein 1 (SKP1), non-POU domain containing octamer binding (NONO) and zinc activated ion channel (ZACN) were identified as hub proteins. On the whole, the present study may help to gain a comprehensive understanding of tumorigenesis in HPSCC and provide valuable information for early diagnosis and drug design of HPSCC in future research.

The aim of the present study was to elaborate the underlying pathogenesis of laryngeal squamous cell carcinoma (LSCC). Micro (mi) RNA and messenger (m) RNA expression profiling of patients with LSCC were downloaded from The Cancer Genome Atlas (TCGA) database. Differentially expressed miRNAs (DEMIs) and differentially expressed mRNAs (DEMs) were identified in LSCC compared to normal control tissues. The DEMs targeted by DEMIs were identified and the negative correlation between DEMs and DEMIs was subjected to visualization. The potential functions of DEMs targeted by DEMIs were annotated in Gene Ontology (GO) and Kyoto Encyclopedia of Genes and Genomes (KEGG) database. A total of 663 dysregulated DEMs (449 upregulated and 214 downregulated) and 33 DEMIs (24 upregulated and 8 downregulated) were identified in LSCC compared with normal controls. 502 negative correlations between DEMIs and DEMs were identified and subjected to construct interaction network. In the network, hsa‑miR‑486, ‑34c, ‑206 and ‑182 had the highest connectivity with DEMs, and respectively regulated 39, 33, 28 and 27 DEMs. DEMs targeted by DEMIs were significantly enriched in signal transduction, actin binding and extracellular region of GO terms and focal adhesion and extracellular matrix‑receptor interaction of KEGG pathways. The present study may provide valuable information for understanding the potential oncogenesis mechanism in LSCC and provide the foundation work for diagnosis biomarkers and therapeutic targets for LSCC.

The fuzzy clustering algorithm is sensitive to the m value and the degree of membership. Because of the deficiencies of traditional FCM clustering algorithm, we made specific improvement. Through the calculation of the value of m, the amendments of degree of membership to the discussion of issues, effectively compensate for the deficiencies of the traditional algorithm and achieve a relatively good clustering effect. Finally, through the analysis of temperature observation data of the three northeastern province of china in 2000, the reasonableness of the method is verified.

To explore the effect of prophylaxis on youth's chronic mountain sickness(CMS) who moved to an altitude of above 5 000 meters by long-term oxygen therapy (LTOT).Ninety-six male youth stationed at 5 070 m, 5 200 m and 5 380 m took oxygen continuously by nasal cannula (LTOT group) every body per day. One year later, epidemiological survey were carried out according to the international CMS diagnostic criteria consist of examining right ventricle end-diastolic dimension (RVED), right ventricular anterior wall (RVAW), right ventricular outflow tract (RVOT), main pulmonary artery (MPA), left ventricular end systolic diameter (LVSD) by ultrasonic diagnostic apparatus, and blood test of alanine aminotransferase (ALT), aspartate aminotransferase (AST), gamma-glutamyl transpeptidase (gamma-GT), creatine kinase (CK), lactate dehydrogenase (LDH), superoxide dismutase (SOD), malondialdehyde (MDA), nitric oxide (NO), nitric-oxide synthase (NOS) blood oxygen saturation (SaQ2). Then, they were compared with 91 males in the same group stationed at the same altitude (without any interventions, control group).(1) The epidemiological survey showed that, SaO2 were increased significantly (P0.05) and the prevalence rate of CMS were decreased compared with that of control group (P0.05). (2) Echocardiography showed that SOD, NO, NOS were increased (P0.05 or 0.01) and LVSD, MPA had no significant difference compared with that of control group (P0.05). (3) Biochemical index showed that, SOD, NO, NOS were increased (P0.05 or 0.01), MDA, ALT, AST, LDH were decreased (P0.05 or 0.01) and gamma-GT, CK had no significant difference compared with that of the control group.At high altitude, LTOT can reduce lipid peroxidation, improve the important organ injuries caused by hypoxia and protect the mitochondria respiratory function and play an important role on the prevention of chronic mountain sickness.

To investigate the role and mechanism of microRNA-21(miR-21) in the proliferation and apoptosis of ovarian epithelial carcinoma cells.A short-hairpin RNA specifically targeting miR-21 plasmid was constructed, and the recombinant was identified by restriction endonuclease analysis and DNA sequencing. Three experimental groups were included, transfection group (transfected with pSIREN-miR-21), negative control group (transfected with pSIREN-miR-21-neg) and blank control group (without transfection plasmid). The expression of miR-21 was detected by stem-loop real-time reverse transcription (RT)-PCR in OVCAR3 cells, and western blot was used to detect the expression of programmed cell death 4 (PDCD4) protein. Tethyl thiazolyl tetrazolium(MTT) and flow cytometry method were used respectively.Recombinant plasmid (pSIREN-miR-21) was constructed successfully and identified by restriction endonuclease analysis and DNA sequencing. The relative expression level of miR-21 in cells transfection, negative control and blank control group was 0.26 ± 0.08, 1.26 ± 0.21 and 1.00 respectively. The level of miR-21 in the cells in transfection group was significantly lower than those in the negative control and blank control group (P0.01). The gray scale of PDCD4 protein was 1443 ± 33, 858 ± 19 and 846 ± 16 in the transfection group, negative control and blank control group respectively. The value of PDCD4 in transfection group was higher than other control groups, and there were significantly difference among them(P0.01). Moreover, the optical density of the cells in transfection group was 0.661 ± 0.015, significantly lower than those in two control groups (0.848 ± 0.150 for negative control, 0.935 ± 0.133 for blank control, P0.01). Forty-eight hours after transfection, the rate of viable apoptotic cell was significantly higher than negative control and blank control group [(25.821 ± 0.763)% vs. (0.010 ± 0.003)% vs. (0.238 ± 0.023)%; P0.01]; 72 hours after transfection, the rates of viable apoptotic cell and necrotic cell were all higher than the two control groups [the rate of viable apoptotic cell was (30.480 ± 0.821)%, (7.792 ± 0.312)% and (7.033 ± 0.257)% respectively (P0.01); the rate of necrotic cell was (3.558 ± 0.211)%, (1.557 ± 0.067)% and (1.049 ± 0.028)%, respectively (P0.01)].miR-21 might play an important role in the proliferation and apoptosis of ovarian epithelial carcinoma cells through negatively control the expression of PDCD4.

To investigate the influence of miR-21 down-regulation on cell proliferation, apoptosis, invasion and migration of ovarian papillary adenocarcinoma cell lines (OVCAR3).Short-hairpin RNA (shRNA), specifically targeting miR-21, was constructed and transfected into OVCAR3 cells using the pSIREN-RetroQ linear vector (pSIREN-miR-21). The expression of miR-21 was detected with stem-loop real-time RT-PCR in OVCAR3 cells. Cell proliferation and apoptosis were monitored using the MTT assay and flow cytometry, respectively. Cell migration and invasion were assessed using the transwell migration and scratch-wound assay, respectively. Western-bloting was used for PDCD4 protein expression.pSIREN-miR-21 suppressed miR-21 expression in OVCAR3 cells. miR-21 expression levels in pSIREN-miR-21 cells was 0.3 ± 0.1, which was significantly lower when compared with pSIREN-miR-21-Neg and control groups (P0.01). Cell inhibition rate in the pSIREN-miR-21 group was higher than the control group (29.4% vs 9.0%, P0.01), as was the percentage of apoptotic and necrotic cells. By transwell migration assay, the number of cells migrating in the pSIREN-miR-21 group was significantly lower than in the control group. In addition, fewer cells were observed in the wounded area of the pSIREN-miR-21 group following the scratch-wound assay. PDCD4 expression was increased in OVCAR-3 cells transfected by pSIREN-miR-21 compared with vector-control transfected cells. Moreover, the optical density of the transfected cells was significantly lower than the two control groups.Down-regulation of miR-21 dramatically increased apoptotic cell death and decreased cell proliferation, invasion and migration in OVCAR3 cells. MiR-21 may play an important role in the biological behaviors of epithelial ovarian carcinoma cells through negative control of the expression of PDCD4.

Ovarian cancer, especially epithelial ovarian cancer (EOC), which accounts for 90% of ovarian cancer, continues to be the leading cause of death among gynecological malignancies. However, the factors associated with its malignant biological behavior are still poorly understood. Accumulating evidence suggests that microRNAs (miRNAs), regulating diverse biological processes, may play an important role in tumorigenesis and development. miR-21 has been frequently observed to be aberrantly overexpressed in various tumors. Using real-time PCR, we confirmed that miR-21 was significantly overexpressed in human EOC tissues and cell lines. The overexpression of miR-21 correlated with histological differentiation, clinicopathological stage, and lymph node metastasis, and we showed that knockdown of miR-21 by an inhibitor caused a significant reduction in cell proliferation and decrease in cell migration and invasion abilities. Furthermore, we demonstrated that knockdown of miR-21 significantly increased the expression of PTEN, a known tumor suppressor in ovarian cancer. Collectively, our findings suggest miR-21 may be important in the initiation and progression of EOC as an oncomiR, likely through regulating PTEN.

To investigate the expression of microRNA-21(miR-21) in ovarian epithelial carcinoma and its association with the clinicopathological features.The expression of miR-21 was detected by Stem-loop real-time RT-PCR in 48 cases of ovarian epithelial carcinomas, 24 cases of benign ovarian epithelial tumors and 15 cases of normal ovarian tissues.The relative expression level of miR-21(2-(DeltaDelta)CT) was 4.849-/+1.813 in the ovarian epithelial carcinomas, significantly higher than that in the benign ovarian tumors and normal ovarian tissues (P0.01), but comparable between the latter two groups. The expression of miR-21 was not correlated to the histological type, but increased significantly with the progression of the clinical stages and histological grading (P0.01), showing a close correlation to lymphatic metastasis.MiR-21 might play a role as an oncogene in the tumorigenesis and development of ovarian epithelial carcinoma, and is possibly correlated to the progression and prognosis of ovarian epithelial carcinoma.
