Periodontitis is a chronic inflammatory condition that affects the soft and hard tissue supporting the tooth, favoring the adhesion of bacteria that can generate dental laxity and bone resorption. This study aimed to test an experimental protocol for inducing periodontitis which allows a close clinical assessment and to evaluate the effectiveness of a photodynamic therapy (PDT) protocol to reverse the associated clinical signs. Fifteen Wistar male rats were used for this study, divided into three equal groups. Tooth scaling, rooting and planning of the first superior left molar were performed in all rats, causing the displacement of the gingiva in order to create an accumulation of plaque. A 5-0 cotton ligature was placed, provoking an inflammatory response removed after ten days. One group received one session of PDT; another group received 3 PDT; the last one represented the control group, with no treatment. Clinical evaluation was represented by assessing the body weight, mobility index and bleeding. A rat grimace scale was used to determine the pain. The results showed clinical aspects of periodontitis and healing tissue proved microscopically. The tested procedure can provide all the key biological factors present in periodontal disease and an option for reversing the clinical aspects.

Lipid-rich invasive human breast cancer is a rare enigmatic entity among special types of infiltrating duct carcinoma. Our paper reports a lipid-rich mammary carcinoma in a female cat with the gross, microscopic and immunohistochemical description of the tumor. A 13-year-old intact adult female, mixed-breed cat was presented by the owner to the Laboratory of Pathologic Anatomy from the Faculty of Veterinary Medicine Cluj-Napoca, Romania. A complete necropsy examination was performed in our laboratory. The tissue samples were collected and processed by paraffin technique for further histological, histochemical, immunofluorescence and immunohistochemical examination. During the necropsy examination, a subcutaneous mass was discovered on the chest. Several variably sized, well-demarcated neoplasms were noted in the right axillary lymph node, right thoracic wall, pleura, lungs, liver, spleen and kidney. Histologically, the cells frequently formed tubuloacinar structures. The morphology of the described tumor showed features of a poorly differentiated mammary carcinoma. Numerous tumoral cells were large and polygonal, with abundant cytoplasm that showed foam-like cytoplasm. The tumoral cells contained either multiple small or large and solitary vacuoles that pushed the nucleus to the periphery of the cell. Intracytoplasmic vacuoles of the neoplastic cells were positive for Oil-Red-O and negatively with Periodic Acid–Schiff. As for immunofluorescence/immunohistochemistry, nonvacuolated and vacuolated neoplastic cells were positive for cytokeratin and negative for vimentin. Histochemical and immunohistochemical analysis support a diagnosis of lipid-rich mammary carcinoma. This is the second reliable record of a lipid-rich mammary carcinoma in female cat and the first one with internal metastases.

Objectives/Hypothesis: It is acknowledged that the treatment of chronic rhinosinusitis (CRS) represents an important challenge for rhinology and for social and economic life. At present, one of the most common treatments for CRS is represented by local corticosteroids followed by endoscopic sinus surgery (ESS). Starting from the example of the mesenchymal stem cell’s (MSC) capacity to migrate and to modulate a real response in the nasal mucosa of an allergic rhinitis mouse model, we try to obtain a response in a CRS mouse model, using MSC derived by adipose tissue. The aim of this study is to demonstrate that the MSC can be used in CRS treatment and could change its priorities. Methods: Seventy female mice (6 MSC donor mice) were randomized in two stages of study, 32 Aspergillus fumigatus (Af) exposure mice (20 for histological comparison to 1st control mice and 12 for MSC administration, to CRS/MCS model) and 32 control mice (20 for histological comparison to CRS model and 12 for MSC administration and histological control to MSC model); in the first stage, the Aspergillus fumigatus (Af) CRS mouse model was targeted, in this section were included 64 (n = 32) mice (treated and control group). In order to assess the inflammation level (histological analysis), the animals were euthanized; in the second stage MSCs (1 × 106/animal) were administered intravenously to a total of 24 (n = 24) mice (12 mice from the exposed group and 12 mice from the second control group). Results: After 12 weeks of Af intranasal instillation, the inflammation parameters evaluated indicated a severe diffuse chronic inflammation, associated with diffuse severe hyperplasia and mature diffuse squamous metaplasia. The MSCs’ injection via the ophthalmic vein induced important histopathological changes in the CRS experimental group, starting with the presence of MSCs in all samples and continuing with the important degenerative character of inflammation. Conclusions: MSC administration demonstrated a real improvement of CRS evolution on the CRS mouse model.

Background: Establishing the diagnosis of COVID-19 and Pneumocystisjirovecii pulmonary coinfection is difficult due to clinical and radiological similarities that exist between the two disorders. For the moment, fungal coinfections are underestimated in COVID-19 patients. Case presentation: We report the case of a 52-year-old male patient, who presented to the emergency department for severe dyspnea and died 17 h later. The RT-PCR test performed at his admission was negative for SARS-CoV-2. Retesting of lung fragments collected during autopsy revealed a positive result for SARS-CoV-2. Histopathological examination showed preexisting lesions, due to comorbidities, as well as recent lesions: massive lung thromboses, alveolar exudate rich in foam cells, suprapleural and intra-alveolar Pneumocystisjirovecii cystic forms, and bilateral adrenal hemorrhage. Conclusion: COVID-19 and P.jirovecii coinfection should be considered, particularly in critically ill patients, and we recommend the systematic search for P. jirovecii in respiratory samples.

The silver content of the skin regeneration ointments can influence its regeneration process but in the meantime, it can take the benefit of the antibacterial properties of silver by avoiding the bacterial infection of an open wound. In the current study, the skin healing and regeneration capacity of bioactive glass with spherical gold nanocages (BGAuIND) in the Vaseline ointments were evaluated in vivo comparing the bioactive glass (BG)-Vaseline and bioactive glass with spherical gold (BGAuSP)-Vaseline ointments. Spherical gold nanocages are stabilized with silver and as a consequence the BGAuIND exhibits great antibacterial activity. Histological examination of the cutaneous tissue performed on day 8 indicates a more advanced regeneration process in rats treated with BGAuSP-Vaseline. The histopathological examination also confirms the results obtained after 11 days post-intervention, when the skin is completely regenerated at rats treated with BGAuSP-Vaseline compared with the others groups where the healing was incomplete. This result is also confirmed by the macroscopic images of the evolution of wounds healing. As expected, the silver content influences the wound healing process but after two weeks, for all of the post-interventional trials from the groups of rats, the skin healing was completely.

In this study, the antibacterial activity of cerium oxide nanoparticles on two Gram-negative and three Gram-positive foodborne pathogens was investigated. CeO2 nanoparticles (CeO2 nps) were synthesized by a Wet Chemical Synthesis route, using the precipitation method and the Simultaneous Addition of reactants (WCS–SimAdd). The as-obtained precursor powders were investigated by thermal analysis (TG–DTA), to study their decomposition process and to understand the CeO2 nps formation. The composition, structure, and morphology of the thermally treated sample were investigated by FTIR, Raman spectroscopy, X-ray diffraction, TEM, and DLS. The cubic structure and average particle size ranging between 5 and 15 nm were evidenced. Optical absorption measurements (UV–Vis) reveal that the band gap of CeO2 is 2.61 eV, which is smaller than the band gap of bulk ceria. The antioxidant effect of CeO2 nps was determined, and the antibacterial test was carried out both in liquid and on solid growth media against five pathogenic microorganisms, namely Escherichia coli, Salmonella typhimurium, Listeria monocytogenes, Staphylococcus aureus, and Bacillus cereus. Cerium oxide nanoparticles showed growth inhibition toward all five pathogens tested with notable results. This paper highlights the perspectives for the synthesis of CeO2 nps with controlled structural and morphological characteristics and enhanced antibacterial properties, using a versatile and low-cost chemical solution method.

The aim of the study was to present a review of the literature and research on the carotid body (CB) over the past years and update the latest findings. The purpose of this article is to present a general overview and comparative analysis of CB between species, from the microanatomy to the pathology of CB. This study gives information about the embryological development and physiological aspects of anatomical findings and their differences. The second part of the article gives a comparative analysis of the pathology of CB. Neoplasia of the CB in humans, namely, paraganglioma, in most cases, is part of a genetic MEN syndrome (multiple endocrine neoplasia). In dogs, paraganglioma is also involved with multiple neoplasia formations throughout the body, including endocrine and neuroendocrine glands. From this perspective, dogs are the most suitable specimens for studying carotid body tumors and their involvement in a MEN-like syndrome.

This study aimed to describe the gross and histological features of multiple endocrine and non-endocrine neoplasia, including multiple PGLs found in two Boxer dogs. Additionally, the identified PGLs were immunohistochemically evaluated, and the subunits 2, 3, and 4 of the SDHD gene were screened for possible mutations. The tumors identified include aortic and carotid body PGLs, thyroid follicular-compact carcinoma, and subcutaneous lipomas. One case also had a Leydig cell tumor and adrenal cortex hyperplasia, while the other had H-type pancreatic carcinoma. Three out of 4 PGLs appeared benign, but one aortic body tumor showed malignant features with neoplastic emboli at its edge. Immunohistochemical analysis confirmed the neuroendocrine origin of all PGLs, with positive staining for Chromogranin A, NSE, and variable positivity for S100. No somatic mutations were found in exons 2, 3, and 4 of the SDHD gene in any of the evaluated PGLs. The absence of mutations in the evaluated SDHD gene subunits suggests the involvement of other genetic factors or pathways in the development of these tumors, warranting further investigation in this field.

Abstract Pancreatic cancer is one of the most aggressive forms of cancer, and treatment options are limited. One therapeutic approach is to use nanoparticles to deliver the active agent directly to pancreatic cancer cells. Nanoparticles can be designed to specifically target cancer cells, minimizing damage to healthy tissues. Silver nanoparticles have the unique ability to absorb light, especially in the near-infrared (NIR) region. In this study, silver nanoparticles functionalized with IgG molecules were synthesized and administered to pancreatic cancer cell lines. Subsequently, the cells were photo-excited using a 2 W 808 nm laser and further examined in PANC-1 pancreatic cancer cell lines. Flow cytometry and confocal microscopy combined with immunochemical staining were used to examine the interaction between photo-excited silver nanoparticles and pancreatic cancer cells. The photothermal therapy based on IgG-functionalized silver nanoparticles in pancreatic cancer induces dysfunction in the Golgi apparatus, leading to the activation of the caspase-3 apoptotic pathway and ultimately resulting in cellular apoptosis. These findings suggest that our proposed IgG nanoparticle laser treatment could emerge as a novel approach for the therapy of pancreatic cancer.

Abstract Background Carbofuran is a widely used broad-spectrum pesticide that, despite strict regulation and being banned for more than a decade, is still encountered in cases of intentional poisoning in dogs and wildlife. The objective of the study was to provide a complete and detailed description of the pathological, histological and toxicological findings of 7 cases of intentional carbofuran poisoning in dogs. Results In this retrospective study, 7 cases of carbofuran intoxication recorded from July 2015 to June 2017 were analyzed. Following complete history recording, all cases were examined by complete necropsy and histopathology. Carbofuran intoxication was confirmed in all cases by gas chromatography. The postmortem examination revealed extensive hemorrhaging and congestion located mainly within the respiratory, nervous and cardiovascular systems, accompanied by degeneration and necrosis within the lungs, heart, and kidneys. Conclusions Although carbamates have been banned in the European Union, carbamate poisoning is still frequently encountered, especially in wild animals. This paper will contribute to a better understanding of the occurrence and pathogenesis of acute carbofuran exposure in dogs and contribute some peculiar pathological features of this type of poisoning to the current literature.

Abstract Background Left ventricular noncompaction (LVNC) is a rare form of cardiomyopathy currently described in humans and cats. It consists of a spongy myocardium characterized by prominent trabeculation and deep recesses involving more than 50% of the ventricular thickness. We describe the clinical and pathological features of LVNC combined with tricuspid valve dysplasia, double-orifice tricuspid valve and severe pulmonary stenosis in a puppy. In addition, we briefly review the LVNC causes, pathogenesis, forms and current diagnostic criteria. Case presentation A seven-week-old intact German Shorthaired Pointer-cross male was presented with a poor body condition, exercise intolerance and dyspnea. Clinical exam identified a bilateral systolic murmur (grade IV/VI over the right heart base and grade III/VI over the left heart base). Echocardiography identified tricuspid valve dysplasia, mild mitral regurgitation, and severe pulmonic stenosis with a trans-valvar systolic pressure gradient of 106 mmHg. Left ventricular noncompaction was diagnosed by necropsy and further confirmed histopathologically by the presence of two distinct myocardial layers: an inner noncompacted zone covering more than 50% of ventricular thickness containing prominent trabeculation and deep recesses, and an outer zone of compact myocardium. Conclusions This is the first case describing LVNC in a canine patient, supporting the introduction of this form of heart disease as a differential diagnosis for cardiomyopathies in juvenile and adult dogs.

Lafora Disease (LD) is a rare, fatal, late-onset, progressive form of myoclonic epilepsy, occurring in humans and dogs. Clinical manifestations of LD usually include seizures, spontaneous and reflex myoclonus with contractions of the neck and limb muscles. We studied the electroencephalogram (EEG) patterns of two beagles in whom LD was subsequently confirmed by genetic testing. In both cases, the EEG recordings, accompanied by electromyography (EMG), have shown similar uncommon patterns. The hypovoltaged background rhythm was interrupted by waxing “crescendo” polyspikes-slow wave complexes appearing 80–250 ms after the start of intermittent photic stimulation, followed by myoclonic jerks after 80–150 ms. This study highlights the value of EEG in establishing a presumptive diagnosis of LD in dogs.

Abstract Background Double chambered right ventricle (DCRV) is a congenital heart anomaly where the right ventricle is divided into two chambers. We describe, for the first time, an unusual combination of DCRV combined with some other congenital heart defects. Case presentation A 1.2-year-old Golden Retriever was presented with lethargy, exercise intolerance and ascites. Physical examination revealed an irregularly irregular pulse and a grade V/VI, systolic, right cranial murmur. Electrocardiography revealed widened and splintered QRS complexes with a right bundle-branch block pattern. Radiography demonstrated right-sided cardiomegaly. Two-dimensional echocardiography identified a DCRV with tricuspid valve dysplasia. The patient died despite abdominocentesis and 4 days of oral pharmacotherapy, and necropsy revealed an anomalous fibromuscular structure that divided the right ventricle into two compartments. Another finding was tricuspid valve dysplasia with hypoplasia of the posterior and septal leaflets. The anterior leaflet was prominent, being part of the anomalous structure that divided the right ventricle. Necropsy also identified a perimembranous ventricular septal defect and mild subaortic stenosis. Histopathological examination of the fibromuscular band that separated the right ventricle identified longitudinally oriented layers of dense fibrous connective tissue and myocardial cells arranged in a plexiform pattern. The muscular component was well represented at the ventral area of the fibromuscular band, and was absent in the central zone. Superficially, the endocardium presented areas of nodular hyperplasia covering mainly the fibrous part of the abnormal structure. The nodules were sharply demarcated and were composed by loosely arranged connective tissue with myxoid appearance, covered by discrete hyperplastic endocardium. Conclusions Concomitant cardiac malformations involving DCRV, tricuspid valve dysplasia, perimembranous ventricular septal defect and mild subaortic stenosis have not been previously described in veterinary medicine, and are reported here for the first time. Moreover, this is the first report of a canine patient with tricuspid valve dysplasia (TVD) and DCRV where the anterior leaflet is part of an anomalous structure dividing the right ventricle (RV) into two separate compartments.

Data regarding bitch mastitis is fairly scarce compared to the literature regarding ruminant mastitis. Neglecting clinical and subclinical cases of mastitis can be life threatening for both dam and puppies. The aim of this report is to present a case of Mastitis acuta complicated with a case of neonatal septicaemia in a Cane Corso (Canis lupus familiaris L.) pure breed female. Laboratory assays showed a milk pH value of 7.5, milk cytology revealed the presence of segmented neutrophils, while foamy cells, phagocytosis and highly pathogenic bacteria (i.e. Escherichia coli and Pseudomonas aeruginosa) were isolated from milk and puppy. To the author's knowledge, this is the first case of bitch mastitis linked to neonatal septicaemia with mixed E. coli and P. aeruginosa infection.

Round cell neoplasms (RCNs) are frequent cutaneous lesions in dogs, with high percentages among skin tumors. In this category are included histiocytoma, mast cell tumor, plasmacytoma, lymphoma and transmissible venereal tumor. The aim of the study was to perform an epidemiological study with reference to the cutaneous round cell tumors in a period of 10 years in the Department of Pathology (Faculty of Veterinary Medicine, Cluj-Napoca, Romania). Additionally, in the recorded cases with round cell tumors (mast cell tumor, histiocytoma and lymphoma) we described the main histological and cytological features. The epidemiological data were collected from the records of Pathology Department between 2005-2014. The investigation included dogs diagnosed with cutaneous round cell neoplasms, following necropsy analysis or assessment of biopsies or cytological samples. All collected specimens were analyzed by histopathological and/or cytological techniques. The staining used for histological investigation were Hematoxylin-eosin, Masson’s trichrome and Toluidine blue, whereas Diff Quik and Wright methods were utilized in cytological specimens. The distribution of the cutaneous round cell tumors in relation to age, breed and sex was also assessed. The most frequent round cell tumor type was the mast cell tumor (19.54%) followed by histiocytoma (11.33%) and lymphoma (1.98%). The round cell tumors recorded were equally distributed in both males and females. Concerning the distribution of cutaneous RCNs by age (average age), histiocytoma occurred in 5 years old subjects, mast cell tumor in 11.9 years old subjects, and lymphoma in 6 years old subjects. Mast cell tumor was more frequent in stray dogs and Boxer breed, while histiocytoma occurred more commonly in stray dogs. Histological and cytological analysis was mandatory to perform the differential diagnosis between RCNs. Microscopic details concerning cytoplasm and nucleus of tumoral cells, together with the displaying mode in histological specimens may reveal the type of RCN. Cutaneous round cell tumors had an increased incidence, aspect observed in other reports too. The differential diagnosis has to rely on some specific microscopical features that can suggest the type of RCN.

Mastitis in cows, one of the most common and economically important infectious diseases of dairy cattle, all over the world, with significant impact due to economic losses, occurs when the udder becomes inflamed because the leukocytes are released into the mammary gland usually in response to bacteria invasion of the teat canal. The main objective of this study was to evaluate the in vitro antimicrobial susceptibility of bacteria isolated from milk in order to design specific control programs for bovine mastitis in this area. A total of 204 milk samples aseptically collected both from farms and private owners were processed during May 2014 and March 2016 within the Microbiology Laboratory of the Faculty of Veterinary Cluj-Napoca, Romania. The microbiological examination was carried out by inoculation on blood agar and MacConkey medium. After the overnight incubation in aerobic conditions, the identification of the isolates was performed using microscopic, cultural and biochemical methods. Biochemical identification was based on API 20 Biomerieux system. Susceptibility to antibiotics was evaluated using Kirby Bauer disk diffusion method on Mueller-Hinton agar; the antibiotics were represented by Amoxicillin and Clavulanic Acid, Ceftiofur, Florfenicol, Mastidiscs, Enrofloxacin, Penicillin and Tetracycline. Staphylococcus spp. was the most common isolated pathogen, in 54.9% of the specimens, followed by Streptococcus spp. in 20.1%, Escherichia coli in 10.78%, Klebsiella spp. in 8.34%, Bacillus spp. in 5.88%. The most frequent associations were represented by staphylococci-streptococci in 62.7% of the samples, followed by streptococci-bacillus in 19.8% of the samples. The most important etiological agents identified were Staphylococcus aureus, S uberis, Streptococcus agalactiae, and Escherichia coli. Antimicrobial susceptibility test for the total isolates revealed good sensitivity to Enrofloxacin, Mastidiscs and Amoxicillin and Clavulanic Acid. Resistance was observed for Penicillin and Tetracycline. The major mastitis pathogens identified was Staphylococcus aureus, while recurrent mastitis treatment was based on systemic and local administration of Enrofloxacin and Mastidiscs respectively.

Recently, the veterinarians are dealing with a number of cases that require forensic expertise. Such a circumstance could be the accidental electrocution in animals, one of the causes of unnatural death. There is a scarcity with reference to the pathological findings in veterinary forensic medicine. In this paper, we present the main lesions that occurred in a horse with accidental electrocution that was presented for complete necropsy survey. A horse corpse was sent to the Pathology Department (Faculty of Veterinary Medicine, Cluj-Napoca, Romania) for a full medical survey. Preliminary results and external examination: the body was in rigor mortis; from the nasal cavities drained out reddish foam and in the mouth was observed the presence of ingested feed (straw) that was not chewed, suggesting a quick death. The findings detected after internal examination of the carcass were poor blood coagulability, haemorrhagic diathesis throughout the body, with haemorrhages of various sizes in different body regions (e.g., muscles of the withers, in the gluteal muscle, the mucosa of epiglottis, larynx, trachea, in the interstitium of the lung, and ecchymosis in the left kidney). Some other lesions detected were infarcts and haemorrhages in the fundic region of the stomach’s mucosa. In electrocution, haemorrhages are most often located in the respiratory tract, aspect observed in our case too. However, the diagnosis of electrocution has to corroborate the necropsy findings (which are not specific), with some other data such as the fulminant death and inspection of power source.

Plasmonic nanoparticles can be used as building blocks for the design of multifunctional systems based on polymeric capsules. The use of functionalised particles in therapeutics and imaging and understanding their effect on the cell functions are among the current challenges in nanobiotechnology and nanomedicine. The aim of the study was to manufacture and characterize polymeric microstructures by encapsulating plasmonic gold nanoparticles in biocompatible matrix of sodium alginate. The gold nanoparticles were obtained by reduction of tetracluoroauric acid with sodium citrate. To characterize the microcapsules, UV-Vis and FTIR spectroscopy, optical and confocal microscopy experiments were performed. In vitro cytotoxicity tests on HFL-1 cells were also performed. The capsules have spherical shape and 120 μm diameter. The presence of encapsulated gold nanoparticles is also shown by confocal microscopy. In vitro tests show that the microcapsules are not cytotoxic upon 24 h of cells exposure to microcapsules concentrations ranging from 2.5 to 25 capsules per cell. The obtained microcapsules of sodium alginate loaded with plasmonic gold nanoparticles could potentially be considered as release systems for biologically relevant molecules.

BACKGROUND:Melanoma therapy is challenging, especially in advanced cases, due to multiple developed tumor defense mechanisms. Photodynamic therapy (PDT) might represent an adjuvant treatment, because of its bimodal action: tumor destruction and immune system awakening. In this study, a combination of PDT mediated by a metal substituted phthalocyanine-Gallium phthalocyanine chloride (GaPc) and Metformin was used against melanoma. The study aimed to: (1) find the anti-melanoma efficacy of GaPc-PDT, (2) assess possible beneficial effects of Metformin addition to PDT, (3) uncover some of the mechanisms underlining cell killing and anti-angiogenic effects. METHODS:Two human lightly pigmented melanoma cell lines: WM35 and M1/15 subjected to previous Metformin exposure were treated by GaPc-PDT. Cell viability, death mechanism, cytoskeleton alterations, oxidative damage, were assessed by means of colorimetry, flowcytometry, confocal microscopy, spectrophotometry, ELISA, Western Blotting. RESULTS:GaPc proved an efficient photosensitizer. Metformin addition enhanced cell killing by mechanisms dependent on the cell line, namely apoptosis in the metastatic M1/15 and necrosis in the radial growth phase, WM35. Cell death mechanism relied on the inhibition of nuclear transcription factor (NF)-κB activation and tumor necrosis factor (TNF)-related apoptosis-inducing ligand (TRAIL) sensitization, leading to TRAIL and TNF-α induced apoptosis. Metformin diminished the anti-angiogenic effect of PDT. CONCLUSIONS:Metformin addition to GaPc-PDT increased tumor cell killing through enhanced oxidative damage and induction of proapoptotic mechanisms, but altered PDT anti-angiogenic effects. GENERAL SIGNIFICANCE:Combination of Metformin and PDT might represent a solution to enhance the efficacy, leading to a potential adjuvant role of PDT in melanoma therapy.

Thyroglossal duct cyst represents a congenital anomaly of the cervical region, rarely documented in animals. Although previously reported in dogs, cats, horses, goats, pigs, and calves, never in birds. This report describes a rare case of thyroglossal duct cyst in a hen. A necropsy of a Transylvanian Naked Neck hen carried following diphtheroid mucocutaneous lesions. The necropsy revealed a large, cyst-like structure measuring 0.5 cm at the level of the caudal edge of the left thyroid gland. Histologically, the cystic mass, bordered by 1–2 lines of well-differentiated ciliated cuboidal cells, presented nuclear immunoreactivity for Thyroid transcription factor 1. To the best of the authors’ knowledge, there are no previous records of thyroglossal duct cysts in avians. Moreover, this is the first case describing a thyroglossal duct cyst in a hen.

Hepatocellular carcinoma is a major global health concern and a leading cause of cancer-related mortality. The study explores chemically induced murine models, specifically the use of Diethylnitrosamine, to simulate human hepatocellular carcinoma. The focus is on understanding early hepatocellular alterations and distinguishing precursor changes from incidental phenomena. Traditionally, the assessment of hepatocellular alteration foci has relied on hematoxylin and eosin staining. However, this work introduces a novel approach by utilizing the fluorescence properties of eosin, a component of the hematoxylin and eosin stain, to investigate basophilic alteration foci in a Diethylnitrosamine-induced hepatocellular carcinoma murine model. Confocal laser scanning microscopy is employed to correlate changes in fluorescence intensity with tinctorial alterations, providing insights into the paraneoplastic changes in hepatocytes. Histologically, basophilic foci exhibit distinct nodular lesions with disrupted tissue architecture and intense basophilic cytoplasm. Nuclear alterations, including hyperchromasia and basophilia, contribute to the comprehensive understanding of cellular and nuclear changes in basophilic foci. The confocal laser scanning microscopy manifestation reveals a discernible reduction in eosin fluorescence intensity within basophilic foci compared to normal hepatic tissue. Statistical analysis demonstrates a markedly elevated median intensity of eosin fluorescence in normal hepatic tissue compared to tumoral tissue and illustrates a higher standard deviation in eosin intensity within normal hepatic tissue, indicating greater variability. Normal hepatic tissue exhibits a superior maximum intensity of eosin, suggesting a broader span of intensity values compared to tumoral tissue. These findings underscore the potential diagnostic utility of confocal laser scanning microscopy in toxicology pathology, establishing a link between tinctorial changes of basophilic foci and their fluorescence spectral shifts. This approach may contribute to a more nuanced understanding of early hepatocellular alterations and facilitate the identification of precursors to carcinomas in chemically induced hepatocellular carcinoma models.

This article describes a case of obstructive cholangioma (biliary adenoma) followed by severe hydrops (mucocelle of the gallbladder) in an adult bovine. The animal, a six year old female, crossbred cattle, was presented to the necropsy following an acute episode of postpartum septicemia. At necropsy the cholecyst was overdistended, with thick walls and field with 2 liters of clear-mucous content. Proximal to the emergence of the common bile duct, a well-circumscribed, sessile, cauliflower-shaped tumoral mass messuring 4/6 cm, obstruction to bile flow was identified. Cholangioma (cholangiocellular adenoma, biliary adenoma) was histologically diagnosticated bassed on the well-differentiated gland-like structures of the tumor, containing papillary projections covered by cuboidal and prismatic epithelium immunohistochemically positive at cytokeratin 19. In the rest of the gallbladder wall mucinous hyperplasia and discreet mononuclear inflammation was noticed. In this study we described, from our knowledge for the first time in cattle, a case of gallbladder mucocel due to obstructive cholangioma. [ABSTRACT FROM AUTHOR]

Ascites syndrome (AS) in chickens represents a major cause of mortality and the most frequent metabolic-related cause of abattoir carcass condemnation in broilers. Susceptibility of broiler chickens to ascites is determined by a complex interaction between genetically-controlled factors (as high-rate metabolism and peculiarity of the cardiovascular system) and exogenous factors such as nutrition, and temperature. Endocrine imbalances, especially thyroid-gland dysfunctions, are currently regarded as key-elements in the development of AS. The current case represents the first spontaneous case of hypothyroidism (goiter) associated with AS in broilers, bringing additional arguments to the theory which claims the role of thyroid dysfunction in this syndrome. Further studies of spontaneous cases which must include assessment of the thyroid hormones need be performed in order to better understand the link between thyroid gland dysfunction and the cardiopulmonary changes in the context of AS in birds.

The aim was to reveal the prevalence of post-mortem lesions in two large-scale bovine slaughterhouses found in Romania so as to point out the potential hazards for human health. The proper examination of animal carcasses and organs that are destined for human consumption is essential in order to ensure food safety but not all the time properly made due to lack of time during the process of slaughter. The results showed that the most frequent lesions found within the examined organs were caused by parasitic infestation (Echinococcus granulosus; Dyctiocaulus viviparus) and the highest prevalence was found in the lungs. The pathology of lesions found in bovine organs is very variable in the slaughterhouse, being able to report strange aspects such an ectopic liver in the lung parenchyma. Measures should be taken in the farms due to this high prevalence of parasites which causes both health problems as well as economical losses.

ABSTRACT In the present study, we aimed to assess the toxicity of artemisinin on the haematological system and its effect on the productive performance of broiler chickens. Eighteen-day-old chickens were randomly divided into four groups of 30 chickens (three replicates of 10 broilers/group): control group and three experimental groups: ART5 - diet with 5 ppm of artemisinin; ART50 - diet with 50 ppm of artemisinin; and ART500 - diet with 500 ppm of artemisinin. Artemisinin enhanced the productive performances of broiler chickens at the lowest concentration (5 ppm), but at the highest concentration (500 ppm), it negatively affected weight gain and the feed conversion ratio. The performance characteristics of the chickens whose diets were supplemented with 50 ppm artemisinin were similar to those of the control group. Additionally, 5 ppm artemisinin in feed did not significantly affect the haematological parameters of the chickens, but 50 and 500 ppm artemisinin induced a gradual decline of the total leukocytes, lymphopenia, monocytosis, and eosinopenia, and the highest concentration caused anaemia. Artemisinin at a low concentration could be used as a feed additive in the poultry industry to improve organic broiler production performance with no serious side effects.
