Abstract

Induction of the cell cycle is emerging as an intervention to treat heart failure. Here, we tested the hypothesis that enhanced cardiomyocyte renewal in transgenic mice expressing cyclin D2 would be beneficial during hemodynamic overload. We induced pressure overload by transthoracic aortic constriction (TAC) or volume overload by aortocaval shunt in cyclin D2expressing and WT mice. Although cyclin D2 expression dramatically improved survival following TAC, it did not confer a survival advantage to mice following aortocaval shunt. Cardiac function decreased following TAC in WT mice, but was preserved in cyclin D2-expressing mice. On the other hand, cardiac structure and function were compromised in response to aortocaval shunt in both WT and cyclin D2-expressing mice. The preserved function and improved survival in cyclin D2expressing mice after TAC was associated with an approximately 50% increase in cardiomyocyte number and exaggerated cardiac hypertrophy, as indicated by increased septum thickness. Aortocaval shunt did not further impact cardiomyocyte number in mice expressing cyclin D2. Following TAC, cyclin D2 expression attenuated cardiomyocyte hypertrophy, reduced cardiomyocyte apoptosis, fibrosis, calcium/calmodulin-dependent protein kinase IIS phosphorylation, brain natriuretic peptide expression, and sustained capillarization. Abstract

Accurate nuclear identification is crucial for distinguishing the role of cardiac myocytes in intrinsic and experimentally induced regenerative growth of the myocardium. Conventional histologic analysis of myocyte nuclei relies on the optical sectioning capabilities of confocal microscopy in conjunction with immunofluorescent labeling of cytoplasmic proteins such as troponin T, and dyes that bind to double-strand DNA to identify nuclei. Using heart sections from transgenic mice in which the cardiomyocyte-restricted [alpha]-cardiac myosin heavy chain promoter targeted the expression of nuclear localized [beta]-galactosidase reporter in >99% of myocytes, we systematically compared the fidelity of conventional myocyte nuclear identification using confocal microscopy, with and without the aid of a membrane marker. The values obtained with these assays were then compared with those obtained with anti-[beta]-galactosidase immune reactivity in the same samples. In addition, we also studied the accuracy of anti-GATA4 immunoreactivity for myocyte nuclear identification. Our results demonstrate that, although these strategies are capable of identifying myocyte nuclei, the level of interobserver agreement and margin of error can compromise accurate identification of rare events, such as cardiomyocyte apoptosis and proliferation. Thus these data indicate that morphometric approaches based on segmentation are justified only if the margin of error for measuring the event in question has been predetermined and deemed to be small and uniform. We also illustrate the value of a transgene-based approach to overcome these intrinsic limitations of identifying myocyte nuclei. Abstract

The ability to image calcium signals at subcellular levels within the intact depolarizing heart could provide valuable information toward a more integrated understanding of cardiac function. Accordingly, a system combining two-photon excitation with laser-scanning microscopy was developed to monitor electrically evoked [[Ca.sup.2+]].sub.i] transients in individual cardiomyocytes within noncontracting Langendorff-perfused mouse hearts. [[Ca.sup.2+]].sub.i] transients were recorded at depths [less than or equal to] 100 [micro]m from the epicardial surface with the fluorescent indicators rhod-2 or fura-2 in the presence of the excitation-contraction uncoupler cytochalasin D. Evoked [[Ca.sup.2+]].sub.i] transients were highly synchronized among neighboring cardiomyocytes. At 1 Hz, the times from 90 to 50% ([t.sub.90-50%]) and from 50 to 10% ([t.sub.50-10%]) of the peak [[Ca.sup.2+]].sub.i] were (means [+ or -] SE) 73 [+ or -] 4 and 126 [+ or -] 10 ms, respectively, and at 2 Hz, 62 [+ or -] 3 and 94 [+ or -] 6 ms (n = 19, P < 0.05 vs. 1 Hz) in rhod-2-loaded cardiomyocytes. [[Ca.sup.2+]].sub.i] decay was markedly slower in fura-2-loaded hearts ([t.sub.90-50%] at 1 Hz, 128 [+ or -] 9 ms and at 2 Hz, 88 [+ or -] 5 ms; [t.sub.50-10%] at 1 Hz, 214 [+ or -] 18 ms and at 2 Hz, 163 [+ or -] 7 ms; n = 19, P < 0.05 vs. rhod-2). Fura-2-induced deceleration of [[Ca.sup.2+]].sub.i] decline resulted from increased cytosolic [Ca.sup.2+] buffering, because the kinetics of rhod-2 decay resembled those obtained with fura-2 after incorporation of the [Ca.sup.2+] chelator BAPTA. Propagating calcium waves and [[Ca.sup.2+]].sub.i] amplitude alternans were readily detected in paced hearts. Abstract

An experiment was performed to examine the effect of tuberous sclerosis complex 2 (TSC2) gene product on cardiomyocyte growth and differentiation. Eker rats were used as samples because it was believed that they contain germ-line TSC2 mutation. Results of the experiment showed that TSC1 and TSC2 were significant regulators of cardiomyocyte cell cycle and terminal differentiation. Cardiomyocytes exhibited in the samples synthesized DNA after repeated passage in vitro suggesting that tuberin regulated cardiomyocyte cell-cycle withdrawal.

Abstract

Normal and injured transgenic mucine hearts were analyzed to determine the effects of cardiac dysfunction on cardiomyocyte deoxyribonucleic acid (DNA) synthesis. Analysis of normal and injured mucine hearts based on a cardiomyocyte nuclear labeling index indicated a maximum labeling index of 0.0005%. Furthermore, a single synthetic nucleus was detected after the examination of 180,000 ventricular heart cells indicating the rarity of cardiomyocyte synthesis.

Abstract

An investigation of cardiomyocytes isolated from inbred mice identified two phases of cardiomyocyte DNA synthesis during the murine cell cycle. The first phase occurred in early life phase and was characterized by cardiomyocyte proliferation or reduplication. The second phase occurred during early neonatal life and was characterized by binucleation of the cells. Northern and Western blot data of murine cardiomyocytes are presented.
