A tanulmány célja az automatizált szoftvertesztelés elméleti és gyakorlati hatékonyságának vizsgálata. Ehhez felmérjük, milyen szempontok szerint érdemes automatizált tesztelői szoftvert választani, valamint rávilágítunk, milyen esetekben hatékonyabb az automatizált tesztelés a manuális teszteléssel szemben. Előbb elméleti síkon vetjük össze a két tesztelési módszertant, majd egy valós üzleti esettanulmányon keresztül mutatjuk be a tesztelői szoftver kiválasztását és végzünk el egy gyakorlati összehasonlító elemzést. Öt automata tesztrendszer összehasonlítása alapján kiderül, hogy az alapvető technológiai és gazdasági követelményeken felül a rugalmasság és a megbízhatóság is kiemelkedő szempont a kiválasztási folyamatban. A tanulmányból kiderül az is, hogy az automatizálás csak ismétlődő tesztelések esetén lehet hatékony. Egy legalább tíz ügyféllel rendelkező szoftvergyártónak azonban megéri az automatizált tesztelés irányába mozdulni.

This article explores female entrepreneurs’ picture of self in the gaze of others. It relies on the narratives of female business owners gained via semi-structured interviews and focus groups, compiled in the framework of an international research project (iFEMPOWER)1 in Austria. The study reveals that the imagined and perceived gaze of others has a significant power on how businesswomen define both their professional and personal self and how they evaluate their self-worth. The gaze of others becomes a signifier of shame (for not being enough or being too much). The results of this study contribute to a more complex understanding of female entrepreneurship, and with the interdisciplinary character it aims at shaping the contemporary discourse on the gendered entrepreneurial sector.

Banks are usually active in creating and operating corporate social responsibility (CSR) programmes for different reasons. This paper analyses the Erste Bank SEEDS programme, which is dedicated to entrepreneurs promoting social welfare in Hungary. A total number of 68 social entrepreneurs were selected from 203 applicants to participate in the programme. To detect the success factors, the paper examines all proposals with content analysis. Then, using a bivariate logistic regression, the research estimates the probability of getting selected and identifies three critical factors: the potential social impact, financial sustainability and the life cycle of the product or service. These results hint to the bank caring about social impacts but only if financial sustainability is assured. Some social activities (community building, health or sustainable lifestyle) are less preferred than others (employment). The main contribution of the study is to present how corporate social responsibility programmes can be analysed and what hidden requirements may be incorporated within them.

A felsőfokú végzettséggel rendelkezőket kisebb mértékben érinti a szegénység. A végzettség megszerzése tehát létfontosságú lehet a szegénységből való kitöréshez. Magyarországon a felsőoktatás államilag finanszírozott, ingyenes elérése teljesítménytől függ. Mivel a leghátrányosabb helyzetű jelentkezők gyengébben teljesítenek, sokkal alacsonyabb eséllyel nyerhetnek felvételt valamely felsőoktatási intézmény képzésére. Az egyenlő esélyek javítása érdekében a hallgatók rendelkezésére áll egy jól megtervezett diákhitelrendszer hazánkban. A tanulmány bemutatja, hogy a felsőoktatási finanszírozási politika pillanatnyilag tovább növeli a szakadékot a szegények és tehetősebbek között. A Diákhitel 1 felvételével és annak Magyar Állampapír Plusz (MÁP Plusz) értékpapírba történő befektetésével arbitrázshozamot érhetnek el azok a hallgatók, akiknek nincs szükségük a tandíjuk és a megélhetésük hitelből történő finanszírozására. A magyar rendszer tehát ahelyett, hogy támogatná az alacsony jövedelmi hátterű diákok felzárkózási folyamatát, további támogatást nyújt a jobb körülményekkel rendelkező hallgatók számára. A nemzetközi diákhitelrendszerek példájára érdemes lenne univerzális helyett célzott megközelítést alkalmazni például kamattámogatás formájában, vagy célszerű lenne jövedelmi helyzethez kötni a hiteligénylést.

People without tertiary educational attainment are more affected by poverty. Thus, obtaining a higher degree of education can act as a key to pulling sections of society out of the poverty trap. In higher education, Hungary has a dual-track tuition policy, which offers restricted merit-based entry to state-funded universities, leading to entirely free higher education for a limited number of students. As the most disadvantaged students perform worse at secondary school, their chances of being enrolled in free tertiary education are much lower. To improve equity, students have access to a well-designed student loan system in Hungary. This study demonstrates that Hungary’s higher education funding policy is currently further widening the gap between the rich and the poor, as holding a Student Loan 1 and a Hungarian Government Security Plus (MÁP Plusz) position results in arbitrage for those who do not need to borrow to finance their tuition fee and living expenses during the academic years. Instead of supporting the catchingup process for students coming from low-income backgrounds, the Hungarian system provides additional financial aid to advantaged students. Based on an international comparison of student loan schemes, adopting a targeted approach instead of a universal one might be worthy of consideration, e.g. providing an interest rate subsidy to disadvantaged students or setting a lower income level of the student’s household as an eligibility criterion for borrowing.

Absztrakt: A kinureninek manapság intenzív érdeklődés tárgyát képezik, mivel számos fiziológiás és patológiás folyamatban részt vesznek. Az esszenciális aminosav triptofán elsősorban a kinurenin-útvonalon keresztül metabolizálódik. A lebomlás során kinurenin-aminotranszferázok segítségével keletkezik az egyik fontos termék, a kinurénsav. A kinurénsav excitatorikus receptorok ligandja, neuroprotektív tulajdonságú. A kinurénsav szintjének abnormális csökkenése vagy növekedése a neurotranszmitter-rendszerek egyensúlyának felborulásához vezethet, és ez számos neurodegeneratív és neuropszichiátriai betegségben megfigyelhető. A kinurénsav a poláros szerkezete miatt nehezen jut át a vér–agy-gáton, emiatt közvetlenül nem alkalmas terápiás célokra. Ezért kutatásunk célja olyan kinurénsav-analógok előállítása és farmakológiai tesztelése volt, melyek a vér–agy-gáton könnyebben átjutnak. Az újonnan szintetizált kinurénsav-analógok hatékonynak bizonyultak több idegrendszeri betegség (migrén, Huntington-kór) modelljében. A kinurénsav-származékokkal kapott eredmények szerint e vegyületek új terápiás célpontot jelenthetnek a neurodegeneratív betegségek kezelésében. Atopic dermatitis (AD) is one of the most common skin diseases, the prevalence of which is especially high among children. Although our understanding about its pathogenesis has substantially grown in recent years, and hence, several novel therapeutic targets have been successfully exploited in the management of the disease, we still lack curative treatments for it. Thus, there is an unmet societal demand to identify further details of its pathogenesis to thereby pave the way for novel therapeutic approaches with favorable side effect profiles. It is commonly accepted that dysfunction of the complex cutaneous barrier plays a central role in the development of AD; therefore, the signaling pathways involved in the regulation of this quite complex process are likely to be involved in the pathogenesis of the disease and can provide novel, promising, yet unexplored therapeutic targets. Thus, in the current review, we aim to summarize the available potentially AD-relevant data regarding one such signaling pathway, namely cutaneous opioidergic signaling.

Over the past decade, the rights of people whose sexual orientation does not conform to prevailing norms have become a divisive issue in many countries. Despite a long tradition of research on media and sexual minorities, the role of the media in these recent backlashes remains poorly understood. We argue that this is partly because work in this area is often underpinned by a simple, linear narrative that unambiguously links visibility to empowerment. We highlight the ambivalent impact of mediated visibility and argue that in the context of elite-driven polarization, illiberalism and low levels of media freedom, visibility can become a vehicle of control. To explore this proposition, we examine the link between media and public attitudes to same-sex relationships in four east European countries, combining a population survey with semi-structured interviews. The results confirm the need to consider the conditions of mediated visibility in particular socio-political contexts, showing that where control over the conditions of visibility remains in the hands of homophobic elites, both Public Service Media and digital media can contribute to negative attitudes.

Publicly available neural recordings obtained with high spatial resolution are scarce. Here, we present an electrophysiological dataset recorded from the neocortex of twenty rats anesthetized with ketamine/xylazine. The wideband, spontaneous recordings were acquired with a single-shank silicon-based probe having 128 densely-packed recording sites arranged in a 32 × 4 array. The dataset contains the activity of a total of 7126 sorted single units extracted from all layers of the cortex. Here, we share raw neural recordings, as well as spike times, extracellular spike waveforms and several properties of units packaged in a standardized electrophysiological data format. For technical validation of our dataset, we provide the distributions of derived single unit properties along with various spike sorting quality metrics. This large collection of in vivo data enables the investigation of the high-resolution electrical footprint of cortical neurons which in turn may aid their electrophysiology-based classification. Here, we devised a new strategy for eradicating cancer stem cells (CSCs), via a "synthetic-metabolic" approach, involving two FDA-approved antibiotics and a dietary vitamin supplement. This approach was designed to induce a "rho-zero-like" phenotype in cancer cells. This strategy effectively results in the synergistic eradication of CSCs, using vanishingly small quantities of two antibiotics. The 2 metabolic targets are i) the large mitochondrial ribosome and ii) the small mitochondrial ribosome. Azithromycin inhibits the large mitochondrial ribosome as an off-target side-effect. In addition, Doxycycline inhibits the small mitochondrial ribosome as an off-target side-effect. Vitamin C acts as a mild pro-oxidant, which can produce free radicals and, as a consequence, induces mitochondrial biogenesis. Remarkably, treatment with a combination of Doxycycline (1 μM), Azithromycin (1 μM) plus Vitamin C (250 μM) very potently inhibited CSC propagation by >90%, using the MCF7 ER(+) breast cancer cell line as a model system. The strong inhibitory effects of this DAV triple combination therapy on mitochondrial oxygen consumption and ATP production were directly validated using metabolic flux analysis. Therefore, the induction of mitochondrial biogenesis due to mild oxidative stress, coupled with inhibition of mitochondrial protein translation, may be a new promising therapeutic anti-cancer strategy. Consistent with these assertions, Vitamin C is known to be highly concentrated within mitochondria, by a specific transporter, namely SVCT2, in a sodium-coupled manner. Also, the concentrations of antibiotics used here represent sub-antimicrobial levels of Doxycycline and Azithromycin, thereby avoiding the potential problems associated with antibiotic resistance. Finally, we also discuss possible implications for improving health-span and life-span, as Azithromycin is an anti-aging drug that behaves as a senolytic, which selectively kills and removes senescent fibroblasts.

The tryptophan (TRP)-kynurenine (KYN) metabolic pathway is a main player of TRP metabolism through which more than 95% of TRP is catabolized. The pathway is activated by acute and chronic immune responses leading to a wide range of illnesses including cancer, immune diseases, neurodegenerative diseases and psychiatric disorders. The presence of positive feedback loops facilitates amplifying the immune responses vice versa. The TRP-KYN pathway synthesizes multifarious metabolites including oxidants, antioxidants, neurotoxins, neuroprotectants and immunomodulators. The immunomodulators are known to facilitate the immune system towards a tolerogenic state, resulting in chronic low-grade inflammation (LGI) that is commonly present in obesity, poor nutrition, exposer to chemicals or allergens, prodromal stage of various illnesses and chronic diseases. KYN, kynurenic acid, xanthurenic acid and cinnabarinic acid are aryl hydrocarbon receptor ligands that serve as immunomodulators. Furthermore, TRP-KYN pathway enzymes are known to be activated by the stress hormone cortisol and inflammatory cytokines, and genotypic variants were observed to contribute to inflammation and thus various diseases. The tryptophan 2,3-dioxygenase, the indoleamine 2,3-dioxygenases and the kynurenine-3-monooxygenase are main enzymes in the pathway. This review article discusses the TRP-KYN pathway with special emphasis on its interaction with the immune system and the tolerogenic shift towards chronic LGI and overviews the major symptoms, pro- and anti-inflammatory cytokines and toxic and protective KYNs to explore the linkage between chronic LGI, KYNs, and major psychiatric disorders, including depressive disorder, bipolar disorder, substance use disorder, post-traumatic stress disorder, schizophrenia and autism spectrum disorder.

Chronic pain is an unpleasant sensory and emotional experience that persists or recurs more than three months and may extend beyond the expected time of healing. Recently nociplastic pain has been introduced as a descriptor of mechanism of pain, which is due to disturbance of neural processing without actual or potential tissue damage, appearing to replace a concept of psychogenic pain. An interdisciplinary task force of the International Association for the Study of Pain (IASP) compiled a systematic classification of clinical conditions associated with chronic pain, which was published in 2018 and will officially come into effect in 2022 in the 11th revision of the International Statistical Classification of Diseases and Related Health Problems (ICD-11) by the World Health Organization. ICD-11 offers the option for recording the presence of psychological or social factors in chronic pain; however, cognitive, emotional, and social dimensions in the pathogenesis of chronic pain are missing. Earlier pain disorder was defined as a condition with chronic pain associated with psychological factors, but it was replaced with somatic symptom disorder with predominant pain in Diagnostic and Statistical Manual of Mental Disorders, 5th Edition (DSM-5) in 2013. Recently clinical nosology is trending toward highlighting neurological pathology of chronic pain, discounting psychological or social factors in the pathogenesis of pain. This review article discusses components of the pain pathway, the component-based mechanisms of pain, central and peripheral sensitization, roles of chronic inflammation, and the involvement of tryptophan-kynurenine pathway metabolites, exploring participations of psychosocial and behavioral factors in central sensitization of diseases progressing into development of chronic pain, comorbid diseases that commonly present a symptom of chronic pain, and psychiatric disorders that manifest chronic pain without obvious actual or potential tissue damage.

Deferiprone (DFP), also known as Ferriprox, is an FDA-approved, orally active, iron chelator that is currently used clinically for the treatment of iron-overload, especially in thalassaemia major. As iron is a critical factor in Fe-S cluster assembly that is absolutely required for the metabolic function of mitochondria, we hypothesized that DFP treatment could be used to selectively target mitochondria in cancer stem cells (CSCs). For this purpose, we used two ER(+) human breast cancer cell lines, namely MCF7 and T47D cells, as model systems. More specifically, a 3D tumorsphere assay was employed as a functional readout of CSC activity which measures anchorage-independent growth under low attachment conditions. Here, we show that DFP dose dependently inhibited the propagation of CSCs, with an IC-50 of ~100 nM for MCF7 and an IC-50 of ~0.5 to 1 &mu, M for T47D cells, making DFP one the most potent FDA-approved drugs that we and others have thus far identified for targeting CSCs. Mechanistically, we show that high concentrations of DFP metabolically targeted both mitochondrial oxygen consumption (OCR) and glycolysis (extracellular acidification rates (ECAR)) in MCF7 and T47D cell monolayers. Most importantly, we demonstrate that DFP also induced a generalized increase in reactive oxygen species (ROS) and mitochondrial superoxide production, and its effects reverted in the presence of N-acetyl-cysteine (NAC). Therefore, we propose that DFP is a new candidate therapeutic for drug repurposing and for Phase II clinical trials aimed at eradicating CSCs.

Currently kynurenines are considered a hot topic, because of their involvement in numerous physiological and pathological processes. The essential amino acid, tryptophan's main metabolism is through the kynurenine pathway. During the degradation of tryptophan, kynurenic acid is formed with the help of kynurenine aminotransferases. Kynurenic acid is an excitatory receptor ligand and it possesses neuroprotective properties. Abnormal decrease or increase in the kynurenic acid level can cause an imbalance in the neurotransmitter systems and it is associated with several neurodegenerative and neuropsychiatric disorders. Kynurenic acid has a poor penetration through the blood-brain barrier, so it is unfit for therapeutic purposes. For this reason, the aim of our research was the synthesis and pharmacological testing of kynurenic acid analogues with a better blood-brain barrier penetration. The newly synthetized kynurenic acid analogues proved to be effective in models of some nervous system disorders (migraine, Huntington's disease). According to our results with the novel kynurenic acid analogues, these molecules may represent a new therapeutic target in the treatment of neurodegenerative diseases. Short-chain halogenated aliphatic hydrocarbons (e.g. perchloroethene, trichloroethene) are among the most toxic environmental pollutants. Perchloroethene and trichloroethene can be dechlorinated to non-toxic ethene through reductive dechlorination by Dehalococcoides sp. Bioaugmentation, applying cultures containing organohalide-respiring microorganisms, is a possible technique to remediate sites contaminated with chlorinated ethenes. Application of site specific inocula is an efficient alternative solution. Our aim was to develop site specific dechlorinating microbial inocula by enriching microbial consortia from groundwater contaminated with trichloroethene using microcosm experiments containing clay mineral as solid phase. Our main goal was to develop fast and reliable method to produce large amount (100 L) of bioactive agent with anaerobic fermentation technology. Polyphasic approach has been applied to monitor the effectiveness of dechlorination during the transfer process from bench-scale (500 mL) to industrial-scale (100 L). Gas chromatography measurement and T-RFLP (Terminal Restriction Fragment Length Polymorphism) revealed that the serial subculture of the enrichments shortened the time-course of the complete dechlorination of trichloroethene to ethene and altered the composition of bacterial communities. Complete dechlorination was observed in enrichments with significant abundance of Dehalococcoides sp. cultivated at 8 °C. Consortia incubated in fermenters at 18 °C accelerated the conversion of TCE to ethene by 7–14 days. Members of the enrichments belong to the phyla Bacteroidetes, Chloroflexi, Proteobacteria and Firmicutes. According to the operational taxonomic units, main differences between the composition of the enrichment incubated at 8 °C and 18 °C occurred with relative abundance of acetogenic and fermentative species. In recent years, links between selective news exposure and political polarisation have attracted considerable attention among communication scholars. However, while the existence of selective exposure has been documented in both offline and online environments, the evidence of its extent and its impact on political polarisation is far from unanimous. To address these questions, and also to bridge methodological and geographical gaps in existing research, this paper adopts a media repertoires approach to investigate selective news exposure and polarisation in four Eastern European countries – the Czech Republic, Hungary, Poland and Serbia. Using a combination of population surveys, expert surveys and qualitative interviews, the data for the study were collected between November 2019 and May 2020. We identify five types of news repertoires based on their relative openness to counter-attitudinal sources, and show that selective news repertoires are present in 29% of the entire sample. Our findings also reveal significant cross-country differences, with the more selective news repertoires more prominent in countries characterised by higher levels of polarisation. Furthermore, while the selection of news sources is in line with people's electoral (and to a lesser extent ideological) preferences, our findings show that exposure to counter-attitudinal sources can also be strongly correlated with political and ideological leanings. Our qualitative data suggest that this is because exposure to counter-attitudinal sources can reinforce attitudes, and potentially contribute to polarisation. Qualitative data also highlight the influence of environmental factors (e.g., family), and suggest that selective news consumption is associated with normatively different conceptions of media trust.

The incidence of neurodegenerative diseases has increased greatly worldwide due to the rise in life expectancy. In spite of notable development in the understanding of these disorders, there has been limited success in the development of neuroprotective agents that can slow the progression of the disease and prevent neuronal death. Some natural products and molecules are very promising neuroprotective agents because of their structural diversity and wide variety of biological activities. In addition to their neuroprotective effect, they are known for their antioxidant, anti-inflammatory and antiapoptotic effects and often serve as a starting point for drug discovery. In this review, the following natural molecules are discussed: firstly, kynurenic acid, the main neuroprotective agent formed via the kynurenine pathway of tryptophan metabolism, as it is known mainly for its role in glutamate excitotoxicity, secondly, the dietary supplement pantethine, that is many sided, well tolerated and safe, and the third molecule, α-lipoic acid is a universal antioxidant. As a conclusion, because of their beneficial properties, these molecules are potential candidates for neuroprotective therapies suitable in managing neurodegenerative diseases.

A screening program was conducted between April 2010 and may 2011. the screening consisted ofmeasuring blood pressure, heart rate, cholesterol, blood glucose, weight, height, calculating Bmi, and determination of bodycomposition.Results. 2226 children aged 14-18 fullfilling the inclusion criteria participated in the program. 13% of the participantswere overweight (i.e. their Bmi was in between the 85th and 95th percentile of the Bmi for their age) and 4% of them wereobese (i.e. their Bmi was above the 95th percentile for their age). high body fat content was detected in 12.98% of girlsand 5.9% of boys that participated in the study. 66% of girls and 81% of boys with elevated Bmi had high body fat content.mean cholesterol level was significantly higher in the group of overweight and obese participants than in the group of participants with normal Bmi. mean systolic and diastolic blood pressure differed significantly between groups with differentBmi for both girls and boys, with overweight participants most likely to have pathologically high blood pressure.Conclusions. there was a significant increase in both systolic and diastolic blood pressure with an increase in Bmi. the resultsof our study may help design preventive programmes for obesity and hypertension in children.

Neurotensin (NT) and its analog neuromedin N (NN) are formed by the processing of a common precursor in mammalian brain tissue and intestines. The biological effects mediated by NT and NN (e.g. analgesia, hypothermia) result from the interaction with G protein-coupled receptors. The goal of this study consisted of the synthesis and radiolabeling of NN, as well as the determination of the binding characteristics of [(3)H]NN and G protein activation by the cold ligand. In homologous displacement studies a weak affinity was determined for NN, with IC50 values of 454nM in rat brain and 425nM in rat spinal cord membranes. In saturation binding experiments the Kd value proved to be 264.8±30.18nM, while the Bmax value corresponded to 3.8±0.2pmol/mg protein in rat brain membranes. The specific binding of [(3)H]NN was saturable, interacting with a single set of homogenous binding sites. In sodium sensitivity experiments, a very weak inhibitory effect of Na(+) ions was observed on the binding of [(3)H]NN, resulting in an IC50 of 150.6mM. In [(35)S]GTPγS binding experiments the Emax value was 112.3±1.4% in rat brain and 112.9±2.4% in rat spinal cord membranes and EC50 values of 0.7nM and 0.79nM were determined, respectively. NN showed moderate agonist activities in stimulating G proteins. The stimulatory effect of NN could be maximally inhibited via use of the NTS2 receptor antagonist levocabastine, but not by the opioid receptor specific antagonist naloxone, nor by the NTS1 antagonist SR48692. These observations allow us to conclude that [(3)H]NN labels NTS2 receptors in rat brain membranes.

Background Glioma is the most common highly aggressive, primary adult brain tumour. Clinical data show that therapeutic approaches cannot reach the expectations in patients, thus gliomas are mainly incurable diseases. Tumour cells can adapt rapidly to alterations during therapeutic treatments related to their metabolic rewiring and profound heterogeneity in tissue environment. Renewed interests aim to develop effective treatments targeting angiogenesis, kinase activity and/or cellular metabolism. mTOR (mammalian target of rapamycin), whose hyper-activation is characteristic for many tumours, promotes metabolic alterations, macromolecule biosynthesis, cellular growth and survival. Unfortunately, mTOR inhibitors with their lower toxicity have not resulted in appreciable survival benefit. Analysing mTOR inhibitor sensitivity, other metabolism targeting treatments and their combinations could help to find potential agents and biomarkers for therapeutic development in glioma patients. Methods In vitro proliferation assays, protein expression and metabolite concentration analyses were used to study the effects of mTOR inhibitors, other metabolic treatments and their combinations in glioma cell lines. Furthermore, mTOR activity and cellular metabolism related protein expression patterns were also investigated by immunohistochemistry in human biopsies. Temozolomide and/or rapamycin treatments altered the expressions of enzymes related to lipid synthesis, glycolysis and mitochondrial functions as consequences of metabolic adaptation; therefore, other anti-metabolic drugs (chloroquine, etomoxir, doxycycline) were combined in vitro. Results Our results suggest that co-targeting metabolic pathways had tumour cell dependent additive/synergistic effects related to mTOR and metabolic protein expression patterns cell line dependently. Drug combinations, especially rapamycin + doxycycline may have promising anti-tumour effect in gliomas. Additionally, our immunohistochemistry results suggest that metabolic and mTOR activity alterations are not related to the recent glioma classification, and these protein expression profiles show individual differences in patients’ materials. Conclusions Based on these, combinations of different new/old drugs targeting cellular metabolism could be promising to inhibit high adaptation capacity of tumour cells depending on their metabolic shifts. Relating to this, such a development of current therapy needs to find special biomarkers to characterise metabolic heterogeneity of gliomas.

We have shown previously that endocannabinoids promote sebaceous lipogenesis, and sebocytes are involved in the metabolism of the endocannabinoid-like substance oleoylethanolamide (OEA). OEA is an endogenous activator of GPR119, a recently deorphanized receptor, which currently is being investigated as a promising antidiabetic drug target. In this study, we investigated the effects of OEA as well as the expression and role of GPR119 in human sebocytes. We found that OEA promoted differentiation of human SZ95 sebocytes (elevated lipogenesis, enhanced granulation, and the induction of early apoptotic events), and it switched the cells to a proinflammatory phenotype (increased expression and release of several proinflammatory cytokines). Moreover, we could also demonstrate that GPR119 was expressed in human sebocytes, and its small interfering RNA–mediated gene silencing suppressed OEA-induced sebaceous lipogenesis, which was mediated via c-Jun N-terminal kinase, extracellular signal–regulated kinase 1/2, protein kinase B, and CRE-binding protein activation. Finally, our pilot data demonstrated that GPR119 was downregulated in the sebaceous glands of patients with acne, arguing that GPR119 signaling may indeed be disturbed in acne. Collectively, our findings introduce the OEA/GPR119 signaling as a positive regulator of sebocyte differentiation and highlight the possibility that dysregulation of this pathway may contribute to the development of seborrhea and acne.

Nicotinic acid (NA) activates hydroxycarboxylic acid receptor 2 (HCA2), and it is widely used in treating dyslipidaemias. Since its side effects include skin dryness, whereas its deficiency can be accompanied by dyssebacia, characterized by sebaceous gland enlargement, we asked if HCA2 is expressed on human sebocytes, and if NA influences sebocyte functions. By using human immortalized SZ95 sebocytes, we found that non‐cytotoxic (≤100 μmol/L; MTT‐assay) concentrations of NA had no effect on the homeostatic sebaceous lipogenesis (SLG; Nile Red), but normalized excessive, acne‐mimicking SLG induced by several lipogenic agents (arachidonic acid, anandamide, linoleic acid + testosterone; Nile Red; 48‐hr treatments). Moreover, it exerted significant anti‐proliferative actions (CyQUANT‐assay), and increased [Ca2+]IC (Fluo‐4 AM‐based Ca2+‐measurement). Although NA did not prevent the lipopolysaccharide‐induced pro‐inflammatory response (up‐regulation [Q‐PCR] and release [ELISA] of several pro‐inflammatory cytokines) of the sebocytes, collectively, these data support the concept that NA may be effective in suppressing sebum production in vivo. While exploring the mechanism of the sebostatic actions, we found that sebocytes express HCA2 (Q‐PCR, immunofluorescent labelling), siRNA‐mediated silencing of which prevented the NA‐induced Ca2+‐signal and the lipostatic action. Collectively, our data introduce NA, and HCA2 activators in general, as novel, potent and most likely safe sebostatic agents, with possible anti‐acne potential.

The purpose of the present study was to elucidate the role of protein kinase A and C in the mechanism of capsaicin inhibition on mu-opiate receptors. H89, a protein kinase A inhibitor and BIM (bisindolylmaleimide), a protein kinase C inhibitor were used for this purpose. BIM suspended the inhibition of capsaicin in endomorphin-1 competition binding. The addition of BIM alone had no effect itself on this reaction. H89 however, exerted a strong inhibitory effect on the endomorphin-1 binding. We can conclude that protein kinase C certainly plays a role in the inhibition of capsaicin. The role of protein kinase A in this reaction could not be established, owing to the blocking effect of H89 on the mu-opioid receptors.

Endogenously occurring opioid peptides are rapidly metabolized by different ectopeptidases. Human opiorphin is a recently discovered natural inhibitor of the enkephalin-inactivating neutral endopeptidase (NEP) and aminopeptidase-N (AP-N) (Wisner et al., 2006). To date, in vitro receptor binding experiments must be performed either in the presence of a mixture of peptidase inhibitors and/or at low temperatures, to block peptidase activity. Here we demonstrate that, compared to classic inhibitor cocktails, opiorphin dramatically increases the binding of [(3)H]MERF and [(3)H]MEGY ligands to rat brain membrane preparations. We found that at 0 °C the increase in specific binding is as high as 40-60% and at 24 °C this rise was even higher. In contrast, the binding of the control [(3)H]endomorphin-1, which is relatively slowly degraded in rat brain membrane preparations, was not enhanced by opiorphin compared to other inhibitors. In addition, in homologous binding displacement experiments, the IC(50) affinity values measured at 24 °C were also significantly improved using opiorphin compared to the inhibitor cocktail. In heterologous binding experiments the differences were less obvious, but still pronounced using [(3)H]MERF and MEGY compared to dynorphin(1-11), or naloxone and DAGO competitor ligands.

Tritium labelled endomorphins (Tyr-Pro-Trp-Phe-NH 2, EM1, Tyr-Pro-Phe-Phe-NH 2, EM2) and their analogs, Angiotensin-IV (Val- Tyr-Ile-His-Pro -Phe) and its analogs, Neuromedin N (LysIle-Pro-Tyr-Ile-Leu) and a new nociceptin analog (P he-Phe-Gly-pFPhe-Thr-Gly-Aib-Arg-LysSer-Ala-Arg-Lys-Arg-Lys-Asn-Gln-NH 2) and Substance P analogs (Arg-Pro-Lys-Pro-Gln-GlnPhe-Phe-Gly-Leu-Nle-NH 2, Arg-Pro-Lys-Pro-Gln-Gln-Phe) were synthesized by catalytic dehalogenation or saturation of the precursor pepti des with tritium gas. The specific radioactivity of the neuropeptides is high, ranging from 0.6 TBq/ mmol to 2.8 TBq/mmol. The labelled peptides were characterized by radioreceptor bindin g assays and used in mapping their metabolic pathway using rat brain membrane preparation.

Tyr-D-Ala-Gly-Phe-D-Nle-Arg-Phe (DADN) a synthetic analogue of the endogenous Met-enkephalin-Arg-Phe (Tyr-Gly-Gly-Phe-Met-Arg-Phe; MERF), was investigated in radioligand binding assays, [(35)S]GTPgammaS stimulation experiments as well as in in vivo algesiometric tests. Binding properties of [(3)H]DADN were measured in crude membrane fractions of rat spinal cord tissues and in homogenates of Chinese hamster ovary (CHO) cells selectively expressing delta-, kappa-or micro-opioid receptors. The highest affinity for [(3)H]DADN binding was observed in membranes from CHO cells transfected with micro-opioid receptors confirming the micro-selectivity of the peptide. Unlabeled DADN was also investigated in functional biochemical experiments by measuring opioid receptor-mediated G-protein activation in rat brain membrane fractions. The peptide stimulated the activity of the regulatory G-proteins in a concentration dependent manner, and the stimulation was efficiently inhibited in the presence of micro-receptor specific antagonist ligands further supporting the selectivity profile of DADN. Intrathecally administered DADN produced a dose-related, naloxone-reversible antinociception in rat hot water tail-flick tests. Among the selective opioid antagonists tested, the delta-selective naltrindole (NTI) and the kappa-specific norbinaltorphimine (norBNI) showed only slight blocking effects compared with naloxone. The results obtained in the in vitro agonist-stimulated [(35)S]GTPgammaS binding assays are in good agreement with the opioid agonist effect seen in the in vivo pain test.

The highly potent micro -opioid receptor agonist 14-methoxymetopon (4,5alpha-epoxy-3-hydroxy-14beta-methoxy-5beta,17-dimethylmorphinan-6-one) was prepared in tritium labelled form by a catalytic dehalogenation method resulting in a specific radioactivity of 15.9 Ci/mmol. Opioid binding characteristics of [3H]14-methoxymetopon were determined using radioligand binding assay in rat brain membranes. [3H]14-Methoxymetopon specifically labelled a single class of opioid sites with affinity in low subnanomolar range (Ki = 0.43 nm) and maximal number of binding sites of 314 fmol/mg protein. Binding of [3H]14-methoxymetopon was inhibited by ligands selective for the micro -opioid receptor with high potency, while selective kappa-opioids and delta-opioids were weaker inhibitors. 14-Methoxymetopon increased guanosine-5'-O-(3-[35S]thio)-triphosphate ([35S]GTPgammaS) binding with an EC50 of 70.9 nm, thus, providing evidence for the agonist character of this ligand. The increase of [35S]GTPgammaS binding was inhibited by naloxone and selective micro -opioid antagonists, indicating a micro -opioid receptor-mediated action.
