Background: Coffee hulls obtained from dry processing, have the potential to be used in ruminant diets. Objective: To evaluate the chemical composition and in situ degradability of dry matter (DM) and neutral detergent fiber (NDF) of coffee hulls treated with calcium oxide (CaO) in different environmental conditions (EC). Methods: Coffee hulls were subjected to treatments distributed in a 2 × 2 factorial arrangement, consisting of two levels of CaO (0 and 5%, on a DM basis) and two environmental conditions (aerobiosis and anaerobiosis) in a completely randomized design with four replicates, totaling sixteen experimental units. Five-gram samples were incubated in the rumen of two male cattle for 0, 3, 6, 9, 12, 24, 48, 72, 96, and 120 h. This work applies the Density Functional Theory (DFT) to study binuclear complexes of Mn2+ with the ligand 2-hydroxibenzilglycine and its substituted derivatives. The results showed the isomer 2 with multiplicity 11-ete as the most stable between all the structures and multiplicities obtained. Then, the most stable complex with the -OCH3, -Br, -Cl and -NO2 substituents were analyzed. Finally, the absolute hardness and the percentage of LUMO orbital participation for the substituted complexes were evaluated. Among them, the complexes with -NO2, -H, -Br groups were more likely to play the catalytic activity, respectively.

Pulmonary arterial hypertension (PAH) is a chronic cardiovascular disease that displays inflammatory components, which contributes to the difficulty of adequate treatment with the available therapeutic arsenal. In this context, the N-acylhydrazone derivative LASSBio-1359 was previously described as a multitarget drug candidate able to revert the events associated with the progression of PAH in animal models. However, in spite of having a dual profile as PDE4 inhibitor and adenosine A2A receptor agonist, LASSBio-1359 does not present balanced potencies in the modulation of these two targets, which difficult its therapeutic use. In this paper, we describe the design concept of LASSBio-1835, a novel structural analogue of LASSBio-1359, planned by exploiting ring bioisosterism. Using X-ray powder diffraction, calorimetric techniques, and molecular modeling, we clearly indicate the presence of a preferred synperiplanar conformation at the amide function, which is fixed by an intramolecular 1,5-N∙∙∙S σ-hole intramolecular interaction. Moreover, the evaluation of LASSBio-1835 (4) as a PDE4 inhibitor and as an A2A agonist confirms it presents a more balanced dual profile, being considered a promising prototype for the treatment of PAH.

Mercury (Hg) contamination of oceans is rapidly increasing, however Hg bioaccumulation in pinnipeds has been understudied. Here, we report for the first time Hg concentration in liver tissues of South American fur seals (Arctocephalus australis) in South and Southeast Brazil. Hg concentration was determined in twenty-five fur seals’ specimens found stranded along the coast of the Southwestern Atlantic Ocean. Samples were digested using the microwave technique and quantified by cold vapor generation atomic fluorescence spectrometry technique. The average Hg concentration was 6.37 mg kg-1 (wet weight), with a minimum concentration of 0.09 mg kg-1 and the highest concentration of 15.58 mg kg-1. No correlation between biological variables (sex, total length and weight) and Hg concentration in A. australis liver were found. The results presented here are of great importance to establish baselines for future evaluations of Hg contamination in marine mammals and the effects of this environmental problem in animal health.

Two greenhouse experiments were conducted in a randomized block design with a split plot scheme and eight repetitions. In the Ca experiment, the plots were the doses of 0, 100 and 200 mg kg(-1) of Ca, and in the P experiment, by the doses of 0, 150 and 300 mg kg(-1) of P. In both experiments, the subplots corresponded to the sampling time of seedlings (15, 30, 45, 60 and 75 days after planting). The Ca present in the substrate was sufficient to promote the proper rooting and growth of sweet potato seedlings in the trays. The Ca supply in excess (200 mg kg(-1)) adversely affected the growth of seedlings that remained in the tray for more than 60 days. Although the seedlings supplied with P showed higher root growth rate after 45 days, the initial P available in the substrate was sufficient to promote the adequate growth of the seedlings in the trays until 60 days. Acid digestions were performed for the samples and aliquots of them were analysed by ICP-MS and CV-AFS. To minimize the Hg memory effect in its determination by ICP-MS, gold (200 mu g L-1 in 5% v v(-1) HCl) was added to the sample digests. The detection limits were 0.02 mg kg(-1) for ICP-MS and 0.01 mg kg(-1) for CV-AFS. Accuracy was assessed by the calculation of the recovery obtained from TORT-3 (lobster hepatopancreas) certified reference material analysis. Accurate results were obtained for both CV-AFS and ICP-MS. The comparison of the results obtained for hepatic tissue with and without lyophilization showed a significant difference among them (at 95% of confidence level), evidencing losses of Hg in the lyophilization process. Objetivo: avaliar a composição química, a degradabilidade in situ da matéria seca (DM) e da fibra em detergente neutro (NDF) da casca de café tratada com óxido de cálcio (CaO) em diferentes condições de ambiente (EC). Métodos: a casca de café foi submetida aos tratamentos, distribuídos em esquema fatorial 2 × 2, sendo duas doses de CaO (0 e 5% base da matéria seca) e duas condições de ambiente (aeróbico e anaeróbico), em delineamento inteiramente casualizado, com quatro repetições, totalizando dezesseis unidades experimentais. Amostras de cinco gramas foram incubadas no rúmen de dois bovinos machos, nos tempos 0, 3, 6, 9, 12, 24, 48, 72, 96 e 120 horas. Resultados: para os parâmetros cinéticos de degradação da DM, à exceção da fração b, a degradabilidade potencial e efetiva foram afetados (p<0,05) pela interação CaO × EC. Quanto aos parâmetros estimados da degradação da fibra, a fração potencialmente degradável (b) apresentou efeito (p<0,05) para CaO e EC. O tratamento com CaO promoveu aumento da fração b em 4,08 pontos percentuais. Conclusões: a utilização do CaO no tratamento da casca de café promove aumento da degradabilidade efetiva e potencial da DM e redução da fração indegradável da fibra. Métodos: la cáscara de café fue sometida a tratamientos distribuidos en un diseño factorial 2 x 2, dos dosis de CaO (0 a 5% en base seca), y dos condiciones ambientales (aeróbica y anaeróbica) en un diseño completamente al azar con cuatro repeticiones, totalizando dieciséis unidades experimentales. Las muestras de cinco gramos se incubaron en el rumen de dos bovinos machos durante 0, 3, 6, 9, 12, 24, 48, 72, 96 y 120 horas. Resultados: para los parámetros cinéticos de degradación de la DM, a excepción de la fracción b, la degradabilidad potencial y efectiva fueron afectadas (p<0,05) por la interacción CaO × EC. En cuanto a los parámetros estimados de la degradación de la fibra, la fracción potencialmente degradable b presentó efecto (p<0,05) para CaO y EC. El tratamiento con CaO promovió un aumento de la fracción b en 4,08 puntos porcentuales. Conclusión: la utilización del CaO para el tratamiento de la cáscara de café aumenta la degradabilidad efectiva y potencial de la DM y reducción de la fracción no degradable de la fibra. Background: Individuals with type 2 diabetes mellitus are at increased risk for heart failure (HF), particularly those with coexisting atherosclerotic cardiovascular disease and/or kidney disease. Some but not all dipeptidyl peptidase-4 inhibitors have been associated with increased HF risk. We performed secondary analyses of HF and related outcomes with the dipeptidyl peptidase-4 inhibitor linagliptin versus placebo in CARMELINA (The Cardiovascular and Renal Microvascular Outcome Study With Linagliptin), a cardiovascular outcomes trial that enrolled participants with type 2 diabetes mellitus and atherosclerotic cardiovascular disease and/or kidney disease. Methods: Participants in 27 countries with type 2 diabetes mellitus and concomitant atherosclerotic cardiovascular disease and/or kidney disease were randomized 1:1 to receive once daily oral linagliptin 5 mg or placebo, on top of standard of care. All hospitalization for HF (hHF), cardiovascular outcomes, and deaths were prospectively captured and centrally adjudicated. In prespecified and post hoc analyses of HF and related events, Cox proportional hazards models adjusting for region and baseline history of HF were used. Recurrent hHF events were analyzed using a negative binomial model. In a subset of participants with left ventricular ejection fraction captured within the year before randomization, HF-related outcomes were assessed in subgroups stratified by left ventricular ejection fraction > or 50%. Results: CARMELINA enrolled 6979 participants (mean age, 65.9 years; estimated glomerular filtration rate, mL/min per 1.73m(2); hemoglobin A1c, 8.0%; 62.9% men; diabetes mellitus duration, 14.8 years), including 1873 (26.8%) with a history of HF at baseline. Median follow-up was 2.2 years. Linagliptin versus placebo did not affect the incidence of hHF (209/3494 [6.0%] versus 226/3485 [6.5%], respectively; hazard ratio [HR], 0.90; 95% CI, 0.74-1.08), the composite of cardiovascular death/hHF (HR, 0.94; 95% CI, 0.82-1.08), or risk for recurrent hHF events (326 versus 359 events, respectively; rate ratio, 0.94; 95% CI, 0.75-1.20). There was no heterogeneity of linagliptin effects on hHF by history of HF at baseline, baseline estimated glomerular filtration rate or urine albumin-creatinine ratio, or prerandomization left ventricular ejection fraction. Conclusions: In a large, international cardiovascular outcome trial in participants with type 2 diabetes mellitus and concomitant atherosclerotic cardiovascular disease and/or kidney disease, linagliptin did not affect the risk of hHF or other selected HF-related outcomes, including among participants with and without a history of HF, across the spectrum of kidney disease, and independent of previous left ventricular ejection fraction. - Abstract
IMPORTANCE Type 2 diabetes is associated with increased cardiovascular (CV) risk. Prior trials have demonstrated CV safety of 3 dipeptidyl peptidase 4 (DPP-4) inhibitors but have included limited numbers of patients with high CV risk and chronic kidney disease. OBJECTIVE To evaluate the effect of linagliptin, a selective DPP-4 inhibitor, on CV outcomes and kidney outcomes in patients with type 2 diabetes at high risk of CV and kidney events. DESIGN, SETTING, AND PARTICIPANTS Randomized, placebo-controlled, multicenter noninferiority trial conducted from August 2013 to August 2016 at 605 clinic sites in 27 countries among adults with type 2 diabetes, hemoglobin A(1c) of 6.5% to 10.0%, high CV risk (history of vascular disease and urine-albumin creatinine ratio [UACR] > 200mg/g), and high renal risk (reduced eGFR and micro-or macroalbuminuria). Participants with end-stage renal disease (ESRD) were excluded. Final follow-up occurred on January 18, 2018. INTERVENTIONS Patients were randomized to receive linagliptin, 5 mg once daily (n = 3494), or placebo once daily (n = 3485) added to usual care. Other glucose-lowering medications or insulin could be added based on clinical need and local clinical guidelines. MAIN OUTCOMES AND MEASURES Primary outcomewas time to first occurrence of the composite of CV death, nonfatalmyocardial infarction, or nonfatal stroke. Criteria for noninferiority of linagliptin vs placebo was defined by the upper limit of the 2-sided 95% CI for the hazard ratio (HR) of linagliptin relative to placebo being less than 1.3. Secondary outcome was time to first occurrence of adjudicated death due to renal failure, ESRD, or sustained 40% or higher decrease in eGFR from baseline. RESULTS Of 6991 enrollees, 6979 (mean age, 65.9 years; eGFR, 54.6 mL/min/1.73m2; 80.1% with UACR > 30mg/g) received at least 1 dose of study medication and 98.7% completed the study. Odometer information is an important feature of NDT systems for inspection procedures that involve mechanical scanning. Knowledge of transducer position during the inspection allows for the localization of flaws and the fusion (stitching) of several images into more sophisticated representations of the inspected object. Commercial encoders provide NDT systems with accurate realtime displacement information that can be integrated to obtain odometry. However, this information is typically limited to a single axis. Although composite schemes with more than one encoder can be built to provide 2-D or 3-D spatial information, they are mechanically intricate and lack flexibility and ease of use. We propose a 2-D position-tracking solution that is based on image processing. A miniature camera continuously captures images of the external surface of the inspected object, which are fed to an algorithm that detects and stores 2-D displacement between each pair of consecutive images. Additionally, the orientation quaternions provided by an Inertial Measurement Unit are stored, allowing for posterior 3-D path reconstruction over objects of known geometries, such as oil pipes. Besides logging position and orientation histories, the device also provides real-time displacement information to the NDT system, where it is perceived as a set of single-axis encoders, thus termed “the virtual encoder”. We demonstrate the applicability of the device to both contact and immersion ultrasonic inspections. The results show that the concept is promising, despite being based on simple principles and relatively easy to implement. Stereoscopic cameras, also known as depth cameras, are increasingly present in robotics navigation and 3D scene reconstruction. Normally, off-the-shelf depth cameras are not suited for underwater environments and exhibit considerable distortions in the provided 3D images when operating underwater. This is mainly due to the large difference in the refractive coefficients of air and water. This issue prevents their use in subsea applications such as the inspection of oil pipelines, valves and manifolds, where these devices could be used for odometry during ultrasonic inspections as well as for 3D scanning. Aiming to allow for such applications, we present a data-driven method for the correction of distortions caused by unaccounted diffractions in depth cameras. First, the distortion is modelled via a series of detections of control points from a standard target at known underwater positions, and an approximation of the inverse distortion is obtained. Then, at runtime, a procedure takes as input the (distorted) point cloud given by the camera at each frame, applies the inverse distortion, and yields a corrected point cloud. We apply the method to data provided by a RealSense D405 camera encased in a sealed acrylic container underwater. The correction makes it possible to use the camera for odometry underwater, clearing the path for the use of off-the-shelf depth cameras in a wide range of subsea applications. The method can be applied for any other transparent fluid.

A pandemia da Covid-19 exigiu medidas de isolamento social, acarretando implicações na saúde mental e no bem-estar dos indivíduos. A finalidade deste artigo é descrever a experiência do projeto de extensão da Unipampa – Campus São Borja (RS), “Encontrão Cultural Digital”, ocorrido entre outubro e dezembro de 2020. O objetivo principal foi proporcionar opções de arte e cultura para alunos, servidores e comunidade em geral, promovendo o bem-estar e a saúde mental. Foram realizados eventos online de apresentações, bate-papos e workshops com artistas locais da música, fotografia, artes plásticas, bordado, teatro e produção cultural. Os conteúdos de divulgação nas mídias institucionais e externas tiveram mais de 20 mil acessos, e as lives dos eventos, mais de 4 mil até o momento. A partir desses números e do feedback do público nos eventos, concluímos que a acessibilidade a arte e a cultura em tempos de crise é importante e pode ser reinventado, valorizando o trabalho de artistas locais. Pode-se afirmar que esta modalidade de projeto de extensão tem o potencial de contribuir para a geração de significativos impactos na saúde mental e no bem-estar das pessoas, podendo ser também papel da universidade a promoção dessas iniciativas.

O objetivo deste estudo foi avaliar as perdas gestacionais entre 30 e 120 dias de gestação e reconcepção em vacas de corte submetidas à inseminação artificial em tempo fixo (IATF). Foram analisadas 18.462 informações do arquivo zootécnico de animais submetidos a IATF no período da estação de monta de novembro de 2019 a abril de 2020 na região do semiárido de Minas Gerais. Os parâmetros avaliados foram perda gestacional, categoria animal, presença do bezerro ao pé da vaca, escore de condição corporal (ECC) e situação final da estação de monta das fêmeas que perderam gestação após a IATF, os dados foram analisados pelo teste qui-quadrado de Person (χ²) e teste de Kruskal-Wallis, com nível de significância de 5% com uso do programa SPSS. A taxa de prenhez final foi de 58,52%. A taxa de perdas entre o primeiro e segundo diagnóstico gestacional foi de 3,6%. Observou-se dependência entre as variáveis categoria e perda gestacional (χ²= 12,374, p< 0,05). In the course of ongoing research on the pseudoscorpion fauna in the northeastern region of Brazil, we compiled nine pseudoscorpion species with three of them, Geogarypus amazonicus, Apolpium ecuadorense, and Pachyolpium furculiferum, recorded for the first time from the Caatinga biome. Ecological comments are included, and the presence of G. amazonicus is discussed. Additionally, an updated checklist of all pseudoscorpion species and a map of the distribution of species richness for all biomes of Brazil are presented.

Hepatic tissue of Larus dominicanus sampled on the coastline of the state of Santa Catarina in Brazil between October 2016 and May 2018 was used to evaluate intraspecific trends and spatial distribution of essential trace elements (Mn, Co, Cu, Zn, Mo and Cr) and non-essential trace elements (As, Pb, Cd, Hg, Ba and V). Principal Component Analysis (PCA) indicated differences in the bioaccumulation of trace elements between female adults and male adults, differences to sex and age were indicated by Kruskal-Wallis test. Heat maps suggest hot spots in locals with high concentration of trace elements in liver of Larus dominicanus. In general, the concentration of trace elements were comparable with values reported in other studies carried out for this species in South America and other regions of the world. The heat maps showed to be a promising tool to identify influences of the locality on bioaccumulation of trace elements in Larus dominicanus.

Os óleos vegetais são bastante utilizados na culinária em frituras, porém sua reutilização não é recomendada para os mesmos fins, pois pode acabar trazendo malefícios à saúde do consumidor. Dessa forma, acaba-se por gerar uma grande quantidade de resíduo, que se descartado de maneira inadequada é capaz de poluir o solo e a água. Neste trabalho, usou-se a temática de obtenção de sabão a partir de óleo usado para conscientizar a comunidade interna e externa do Instituto Federal de Educação, Ciência e Tecnologia do Rio de Janeiro campus Duque de Caxias, sobre os problemas ambientais oriundos do descarte inadequado deste óleo. A ação foi desenvolvida por meio de palestras e oficinas temáticas para alunos, pais e comunidade do entorno com informações básicas acerca do descarte e reciclagem do óleo usado. A produção de sabão foi realizada em grupo pelos participantes durante as palestras tornando o aprendizado mais prazeroso e significativo. Portanto, a ação desenvolvida contribuiu significativamente para a formação de cidadãos conscientes quanto aos aspectos social e ambiental.

The objective of this study was to evaluate the sample size and area of the quadrats necessary to accurately estimate the forage mass (FM) of a fenced pasture of stargrass (Cynodon nlemfuensis cv. Florico) during the winter. Five metal quadrats were used: a 0.09 m² square (0.30 m side), a 0.25 m2 square (0.50 m side), a 0.25 m2 circle (0.28 m diameter), a 0.5 m2 rectangle (0.5 x 1.0 m), and a 1 m2 square (1.0 m side), each with eight replicates. The size and shape of the quadrats were determined based on cumulative variances to identify combinations that minimized the coefficient of variation (CV). The minimum sample size required to estimate the FM, morphological components and height was established by the CV maximum curvature method. The 0.25 m2 square quadrat (0.5 m side) presented the lowest cumulative CV in estimating the FM and the dry mass of dead material. However, for the estimation of the leaf and stem dry mass, the 1.00 m2 square quadrat (1.00 m side) presented the lowest CV. Using the 0.25 m2 square quadrat, a minimum number of six samples were required for the FM estimation, and eight samples were required for estimating the mean height of the stargrass pasture. Therefore, at least eight samples are recommended to obtain accurate results for the estimation of both variables.

This paper presents the optimization of aggregate mixture in the production of paver blocks through the formulation of a linear programming model. The proposed model aims at reducing manufacturing costs, respecting the compliance with the minimum quality requirements and mechanical/physical characteristics required by the technical standards. The results obtained with the model were used to produce a batch of products, which underwent a process of visual inspection of the quality and laboratory tests. The results showed the viability of using a linear programming to define the required quantities of raw materials to the manufacturing process of Concrete Paver Blocks, which generates economic benefits for the industry and allows the product quality control as well.

Primary initial teacher education courses in England have recently undergone radical reform particularly in relation to the time that students spend in schools. Primary pupils in schools working with student teachers from Homerton College, Cambridge were interviewed to find out what they thought about their student teachers, what impact the student teachers had had on their learning, what qualities they felt teachers should have and what understanding they had of the process by which people train to become teachers. The findings are discussed, and consideration is given to the implications for the future development of the partnership between primary schools and Higher Education Institutions. This paper presents the evaluation of two metaheuristics to solve the Unrelated Parallel Machine Scheduling Problem with Sequence Machine Dependent Setup Time. Considering such a problem, there is no relation between the time to process each task and the machine; and this is why the machines are referred to as unrelated. Furthermore, the setup time between the executions of two tasks depends on both, the task sequence and its associated machine. A metaheuristic genetic algorithm and a variable neighborhood search were used in order to solve the problem due to the difference among their characteristics. The maximal time for the schedule to be completed, also called makespan, was the performance measure used to evaluate the solutions. The results obtained by both metaheuristics were directly compared according to their performance to try to reduce this makespan. The results showed that the variable neighborhood algorithm search outperformed the genetic algorithm regarding the solutions quality and execution time.

Phoresy is a common dispersal behavior among pseudoscorpions. Neotropical pseudoscorpions, mainly from the North and Northeast regions of Brazil, are known for their dispersal relationships with beetles and flies. Here, we report phoretic association among nymphs of Sphenochernes camponoti (Chernetidae) and Fannia flies (F. pusio, F. yenhedi, and F. canicularis) (Diptera, Fanniidae). Twelve flies, each carrying a young pseudoscorpion, were collected in Caatinga vegetation in Pernambuco State, Brazil. Sphenochernes camponoti is a myrmecophilous pseudoscorpion that lives in Camponotus and Acromyrmex colonies. Despite its association with ants, this pseudoscorpion uses other winged arthropods to disperse. This is the first report of phoresy by Sphenochernes camponoti.

O objetivo desse estudo foi avaliar o tamanho e a área da moldura necessários para estimar a massa de forragem (MF) de um pasto vedado de capim-estrela (Cynodon nlenfluensis cv. Florico) durante o inverno. Foram utilizadas cinco molduras metálicas: quadrado de 0,09 m² (0,30 m de lado), quadrado de 0,25 m² (0,50 m de lado), circular de 0,25 m² (0,28 m de raio), retangular de 0,5 m² (0,5 x 1,0 m) e quadrado de 1 m² (1,0 m de lado), cada uma com oito repetições. A dimensão e o formato da moldura foram determinados com base nas variâncias acumuladas para escolha da combinação com menor coeficiente de variação (CV). O número mínimo de amostras necessárias para estimativa da MF, dos componentes morfológicos e da altura foi estabelecido pelo método da máxima curvatura do CV. A moldura de formato quadrado e 0,25 m2 de área (0,5 m de lado) apresentou menor CV acumulado para estimar a MF e a massa seca de material morto. No entanto, para estimativa da massa seca de folha e de colmo, a moldura de formato quadrado e área de 1,00 m2 (1,00 m de lado) apresentou menor CV. Com o uso da moldura de área 0,25 m2 de formato quadrado, foi estabelecido o número mínimo de seis amostras necessárias para estimativa da MF e oito amostras para estimativa da altura média do pasto de capim-estrela. Portanto, indicam-se no mínimo oito amostras para obter resultados acurados da estimativa de ambas variáveis.

Gonioterma sp. é uma espécie monófaga de lepidóptero que constrói seus abrigos foliares (AFs) em Roupala montana, uma espécie arbórea comum na vegetação dos cerrados do Brasil central. No Parque Estadual de Caldas Novas (PESCAN), estado de Goiás, foi realizado um estudo que procurou investigar: 1) preferência do lepidóptero em construir seu AF de acordo com o tamanho da planta; 2) a composição, riqueza e as guildas dos artrópodes associados aos AFs; e, 3) possível associação entre o tamanho das plantas com a abundância dos artrópodes presentes nesses AFs. Os principais resultados revelaram que as mariposas fêmeas de Gonioterma sp. podem depositar seus ovos em R. montana com diferentes classes de tamanhos, porém com preferência por plantas de maior porte. Os AFs apresentaram uma elevada riqueza e abundância de artrópodes de diferentes guildas, onde os mais representativos foram os predadores. Foi verificado associação positiva entre o tamanho de R. montana e a ocorrência de determinadas guildas de artrópodes. O papel exercido por Gonioterma sp. como engenheiro de ecossistema ficou destacado pela importância dos abrigos foliares produzidos por este animal como refúgios para a fauna de artrópodes locais.

Este artigo objetiva apresentar e descrever as ações do projeto de extensão Comunica Erechim, desenvolvido na Universidade Federal da Fronteira Sul (UFFS), Campus Erechim, desde março de 2011. O projeto parte das concepções de letramento como processo de valoração e do uso social da linguagem, constitutivo das práticas sociais e das subjetividades. Os resultados obtidos até agora demonstram a importância desse projeto que busca oferecer aos estudantes de graduação o desenvolvimento de habilidades e competências relativas à produção textual. Ademais, é um espaço para registrar a vida institucional da UFFS e expô-la à comunidade externa, estreitando as barreiras entre universidade e comunidade.

49. Synthesis, Pharmacological Profile and Docking Studies of New Sulfonamides Designed as Phosphodiesterase-4 Inhibitors.

Prior investigations showed that increased levels of cyclic AMP down-regulate lung inflammatory changes, stimulating the interest in phosphodiesterase (PDE)4 as therapeutic target. Here, we described the synthesis, pharmacological profile and docking properties of a novel sulfonamide series (5 and 6a-k) designed as PDE4 inhibitors. Compounds were screened for their selectivity against the four isoforms of human PDE4 using an IMAP fluorescence polarized protocol. The effect on allergen- or LPS-induced lung inflammation and airway hyper-reactivity (AHR) was studied in A/J mice, while the xylazine/ketamine-induced anesthesia test was employed as a behavioral correlate of emesis in rodents. As compared to rolipram, the most promising screened compound, 6a (LASSBio-448) presented a better inhibitory index concerning PDE4D/PDE4A or PDE4D/PDE4B. Accordingly, docking analyses of the putative interactions of LASSBio-448 revealed similar poses in the active site of PDE4A and PDE4C, but slight unlike orientations in PDE4B and PDE4D. LASSBio-448 (100 mg/kg, oral), 1 h before provocation, inhibited allergen-induced eosinophil accumulation in BAL fluid and lung tissue samples. Under an interventional approach, LASSBio-448 reversed ongoing lung eosinophilic infiltration, mucus exacerbation, peribronchiolar fibrosis and AHR by allergen provocation, in a mechanism clearly associated with blockade of pro-inflammatory mediators such as IL-4, IL-5, IL-13 and eotaxin-2. LASSBio-448 (2.5 and 10 mg/kg) also prevented inflammation and AHR induced by LPS. Finally, the sulfonamide derivative was shown to be less pro-emetic than rolipram and cilomilast in the assay employed. These findings suggest that LASSBio-448 is a new PDE4 inhibitor with marked potential to prevent and reverse pivotal pathological features of diseases characterized by lung inflammation, such as asthma.
