Ulidang Village, part of the Tameroddo Sub-district, has excellent potential in the fisheries and marine sector, although it still faces significant economic challenges. The potential of seaweed as a marine resource that has yet to be fully utilized is the focus of this activity. The main objective is to provide comprehensive counseling on the potential of seaweed, including cultivation methods, nutritional content, and business management aspects. This activity's implementation stage starts with planning, program implementation, and evaluation. The research results showed that socialization efforts, including explanations of seaweed potential, business management, and farming profit simulations, have provided the community with a deep understanding. The pretest and post)test evaluations illustrated the improvement in participants' understanding after the socialization activities. Thus, the development of seaweed farming in Ulidang Village can be a solution to improve community welfare, create jobs, and care for the marine environment. Strategic steps include strong partnerships, community empowerment, and adoption of innovations to ensure the sustainability of seaweed farming at the local and national levels. Eucheuma cottonii is a valuable marine algae species widely cultivated in Sumenep, Madura. Recent research on the biochemical characterization of various marine algae species has identified active anti-microbial polyphenols and their derivatives, such as flavonoids. These compounds possess anti-inflammatory and anti-microbial properties. Organisms with sufficient levels of polyphenols and flavonoids tend to have an enhanced immunity against bacterial and viral infections. As a result, Eucheuma cottonii has the potential to serve as a natural source for new medicines derived from the marine resources of Madura Island. This study aimed to optimize the use of marine biological resources, specifically Eucheuma cottonii, as the main ingredient for anti-microbial drugs. The research was conducted between May and November 2021 at the Integrated Laboratory of the University of Trunojoyo Madura. The stages of the research included sampling Eucheuma cottonii, extracting its active compounds, and testing the anti-microbial activity against Aeromonas hydrophila. The results showed that the average yield of Eucheuma cottonii extract ranged from 2.978% to 3.296%. The extract demonstrated anti-microbial properties, as evidenced by the formation of inhibition zones in Aeromonas hydrophila colonies. [ABSTRACT FROM AUTHOR]

4. Antioxidant activity of Eucheuma cottonii Seaweed Extracts from Central Mawasangka Waters

Eucheuma cottonii is one type of seaweed that has the potential to be an antioxidant. This seaweed grows and is distributed in various waters of Indonesia, especially in Southeast Sulawesi. However, information regarding the antioxidant activity and the compound content of E. cottonii in this area is still limited. Therefore, this research aims to determine the compound and antioxidant activity profile in E. cottonii. The study used laboratory experimental methods. The research stages were preparation (seaweed sampling), extraction, qualitative phytochemical screening, and antioxidant assay. The initial identification of compounds using qualitative phytochemical screening methods with three extraction solvents: ethanol, ethyl acetate, and n-hexane. Meanwhile, the antioxidant test employed the DPPH (2,2-diphenyl-1-picrylhydrazyl) method. The results of qualitative phytochemical screening revealed that the seaweed extract of E. cottonii contains saponins, tannins, and terpenoids. The antioxidant assay using the DPPH method indicated that among the three extraction solvents used, ethanol extract exhibited a very strong antioxidant activity (with an IC50 of 30,036 ppm), compared to ethyl acetate extract (IC50 of 182,179 ppm) and n-hexane extract (IC50 of 641,454 ppm). This study concluded that E. cottonii seaweed has the potential to be developed as a source of natural antioxidants or raw material for the discovery of antioxidant medicines in the future.

This study addresses concerns related to potential health risks associated with chlorine residues in traditional tea bags, which exhibit resistance to decomposition in soil. Consequently, the study utilized carrageenan, derived from Eucheuma cottonii seaweed, as a coating material for edible film. This research explores the impact of carrageenan concentration and drying temperature on the performance of tea bags. This study applied the randomized complete block design to analyze the impact of three carrageenan concentrations (1%, 3%, and 5%) at three different drying temperatures (80°C, 85°C, and 90°C) on carrageenan-based film. The investigation comprehensively evaluates the compliance of the carrageenan-based film with commercial-grade quality standards through critical factors such as thickness, solubility, mechanical strength and hedonic analysis. The findings indicate that the most effective carrageenan-based edible film can be produced at a concentration of 3% and a drying temperature of 90°C, highlighting its potential as an eco-friendly alternative in tea bag production. This research aims to assess the impact of adding Elkhorn Sea Moss Flour as a fiber source on the organoleptic characteristics and nutritional value of pasta. The treatments used are C0 = 0% addition of Elkhorn Sea Moss Flour; C1 = 30% ; C2 = 50%; C3 = 70%. The study utilized a completely randomized design (CRD) with one factor, comprising four concentration treatments replicated three times. The results showed that the addition of Elkhorn Sea Moss Flour to pasta resulted in significant increases in protein content, fiber, and antioxidant activity. Pasta with 70% Elkhorn Sea Moss Flour addition showed the highest fiber content and moderately high antioxidant activity, making it a potential functional food product. However, as Elkhorn Sea Moss Flour increased, there was a decrease in organoleptic parameters such as aroma, texture, and taste. Elkhorn Sea Moss Flour has great potential as an additive in functional food products, enriching fiber and antioxidant content, but its use should be carefully considered to ensure good consumer acceptance. This study aimed to determine the potential of Eucheuma cottonii seaweed extract as a natural antioxidant to improve blood profiles and kidney damage in male albino rats induced by NaNO2. This research used a completely randomized design. Thirty animals were divided into five groups, namely negative control, positive control, and three treatment doses of E. cottonii extract. NaNO2 was dissolved in distilled water, and E. cottonii extract was dissolved in 0.5% CMC-Na solution. The research was conducted for 45 days. The treatment group was given NaNO2 for 35 days; on days 16 to 45, E. cottonii extract was given at doses of 150, 300, and 450 mg/kg BW. The positive control was given NaNO2 for 35 days, then given the extract solvent. The negative controls were given the solvent only. At the end of the treatment, the rats were sacrificed to collect blood and kidney organs. The results showed that the extract at doses of 150 and 300 mg/kg BW increased the number of hemoglobin and erythrocytes. The dose of 150 mg/kg BW reduced various kidney damage, such as inflammatory cell infiltration, congestion, hemorrhage, fatty degeneration, necrosis, glomerular abnormalities, and protein deposits in the lumen of the renal tubules. Barru Regency is one of the largest seaweed producing areas in South Sulawesi. The sustainability of seaweed cultivation is influenced by various factors, including ecological, economic, social, institutional and technological. This study aims to analyze sustainability status of seaweed cultivation in Barru Regency. The method used is Multidimensional Scaling (MDS) approach which is analyzed by RAP SEAWEED. Several parameters from each dimension were analyzed to assess existing condition of seaweed cultivation in Barru Regency. The results showed that seaweed cultivation in Barru Regency was classified as quite sustainable on social and technological dimensions and was classified as less sustainable on institutional, economic and ecological dimensions. Parameter analysis using stress, R-Square, and monte carlo values shows accuracy of model and attributes are feasible for use in analysis. The results of sustainability status analysis which are still classified as lacking need to be addressed by taking into account several attributes that have high sensitivity in leverage analysis. Evaluation of attributes for increasing sustainability score can be a basic consideration in formulating a strategy for sustainable management of seaweed cultivation in Barru Regency. Desa Tirowali Kecamatan Ponrang Kabupaten Luwu terletak di sepanjang Teluk Bone sehingga menjadikan daerah ini memiliki sumber daya alam berupa hasil budidaya rumput laut terbesar kedua di Sulawesi Selatan. Hal ini tidak lepas dari dukungan kelompok-kelompok pembudidaya rumput laut, salah satunya yaitu UKM Mutiara Katoni. Kelompok ini membudidayakan rumput laut jenis Eucheuma cottonii menggunakan media tambak dengan metode monokultur maupun polikultur. Rumput laut jenis ini dipilih karena dapat dibudidayakan menggunakan peralatan sederhana, biaya yang murah, masa panen yang cepat, memiliki harga yang tinggi dan tingkat pemanfaatannya sangat luas. Dalam sekali masa panen, kelompok ini dapat menghasilkan sekitar 2 ton rumput laut basah. Namun, perkembangan budidaya rumput laut tidak seiring dengan peningkatan perekonomian masyarakat Desa Tirowali. Hal ini disebabkan oleh berbagai masalah yang dihadapi masyarakat pembudidaya, diantaranya keterbatasan pengetahuan budidaya masyarakat, belum adanya keterlibatan tenaga ahli dalam menyelesaikan masalah penanganan penyakit/hama rumput laut, kurangnya keahlian dan jiwa kewirausahaan masyarakat dalam mengelola usaha rumput laut, kurangnya modal dan bantuan sarana yang didapatkan pembudidaya, serta belum terciptanya kelembagaan kelompok pembudidaya rumput laut Eucheuma cottonii yang berdampak pada keterbatasan akses pasar dalam penjualan hasil panen. Solusi dari permasalahan ini yaitu dengan kegiatan pemberdayaan berbasis masyarakat berupa pendampingan budidaya, bantuan sarana dan permodalan, pelatihan manajemen usaha, dan penguatan kelembagaan. Kegiatan ini menghasilkan luaran berupa product knowledge berupa teknik budidaya dan manajemen usaha rumput laut serta penguatan kelembagaan untuk meningkatkan produktivitas rumput laut dengan kualitas tinggi dan membuka akses pasar dan jalur kordinasi pemasaran hasil panen rumput laut Eucheuma cottonii dalam rangka meningkatkan perekonomian pembudidaya di UKM Mutiara Katoni dan masyarakat umum di Desa Tirowali Kecamatan Ponrang Kabupaten Luwu.

11. Effects of Alkaline Solvent Type and pH on Solid Physical Properties of Carrageenan from Eucheuma cottonii.

The effects of alkali type and pH on the physical properties of carrageenan have been extensively studied. However, their effects on certain characteristics of solid-state properties of carrageenan have not been identified. This research aimed to investigate the effect of alkaline solvent type and pH on the solid physical properties of carrageenan isolated from Eucheuma cottonii. Carrageenan was extracted from the algae using NaOH, KOH, and Ca(OH)
2 at pHs of 9, 11, and 13. Based on the results of preliminary characterization, including yield, ash content, pH, sulphate content, viscosity, and gel strength, it was found that all samples followed Food and Agriculture Organization (FAO) specifications. The swelling capacity of carrageenan based on the type of alkali was KOH > NaOH > Ca(OH)2 . The FTIR spectra of all samples were in agreement with that of standard carrageenan. The molecular weight (MW) of carrageenan using KOH as the alkali followed the order pH 13 > pH 9 > pH 11, while using NaOH, the order was pH 9 > pH 13 > pH 11, and while using Ca(OH)2 , the order was pH 13 > pH 9 > pH 11. The results of the solid-state physical characterization of carrageenan with the highest MW in each type of alkali showed that the morphology of carrageenan using Ca(OH)2 has a cubic shape and is more crystal-like. The order of crystallinity of carrageenan using different types of alkali was Ca(OH)2 (14.44%) > NaOH (9.80%) > KOH (7.91%), while the order of density was Ca(OH)2 > KOH > NaOH. The order of solid fraction (SF) of the carrageenan was KOH > Ca(OH)2 > NaOH, while the tensile strength when using KOH was 1.17, when using NaOH it was 0.08, and while using Ca(OH)2 , it was 0.05. The bonding index (BI) of carrageenan using KOH = 0.04, NaOH = 0.02, and Ca(OH)2 = 0.02. The brittle fracture index (BFI) of the carrageenan was KOH = 0.67, NaOH = 0.26, and Ca(OH)2 = 0.04. The order of carrageenan solubility in water was NaOH > KOH > Ca(OH)2 . These data can be used as the basis for the development of carrageenan for excipients in solid dosage forms. Red seaweed species Eucheuma cottonii was thought to have potential as an anesthetic agent because it is shown to contain compounds with bioactivity as antimicrobial, antifungal, and also anticancer. There have been no studies regarding the active ingredients of anesthetized. The objective of the study was to examine the potential of E. cottonii extract as a candidate for fish anesthetic based on toxicity, phytochemistry, and active compounds contained in the extract. The stages of research include extraction, toxicity test extracts (BSLT), phytochemical test, quantitative test compound phytochemical results, and the identification of active compounds with LC-MS. Test results showed that E. cottonii extract contains a total of phenols of 4.60 ± 0.06 mg/GAE g which is one presentation of the active ingredient in the extract. The extract had bioactivity with LC50 of 221.71 ± 5.74 μg/mL. E. cottonii extract contained flavonoids compounds and saponins with a consecutive number of 24.48% and 0.22%. The results of identifying active compounds were found in several compounds that had activity asantidepressants or sedatives. E. cottonii extract can be used as a candidate for a fish anesthetic agent.

14. Improving congo red dye removal by modification layered double hydroxide with microalgae and macroalgae: Characterization and parametric optimation.

Currently, water contamination by dyes is increasing in water sources, so an appropriate method is needed to handle it. Adsorption is one of the appropriate methods for wastewater contamination. In this study, ZnAl/LDH composites with microalgae Spirulina platensis (SP) and macroalgae Eucheuma cottonii (EC) materials were synthesized using coprecipitation and hydrothermal methods. ZnAl/SP and ZnAl/EC were used for CR adsorption with parameters of pH, contact time, temperature, initial concentration of CR, and cycling adsorption-desorption (regeneration). The maximum adsorption of CR capabilities of ZnAl/SP and ZnAl/EC obtained were 185.185 mg/g (s = ± 1.24) and 144.928 mg/g (s = ± 1.03), respectively. The Langmuir and pseudo-second-order models demonstrate the best fit for both isotherm and kinetic analysis across all samples, suggesting a monolayer adsorption process and chemisorption mechanism. [Display omitted] • Preparation of ZnAl-EC was successful examination of the characterization data using XRD, FTIR, BET, TGA and XPS. • Excellent regeneration until 5 cycle with 48.83 % was adsorbed. • ZnAl-EC has a high thermal stability seen from the % loss weight of 3.64 % and left a residue of 95.96 % in TGA thermal analysis. • The maximum adsorption capacity of ZnAl-EC is 384.61 mg/g for Methyl Orange adsorption. This study synthesised ZnAl- Eucheuma cottonii (ZnAl-EC) composites made from ZnAl-Layered Double Hydroxide (ZnAl-LDH) and Eucheuma cottonii (EC) with the aim of making the composites have better stability. ZnAl-EC was tested for its ability to absorb anionic dyes. The thermal stability of ZnAl-EC was high % weight reduction decomposed by 3.64% and left a residue of 95.96%. The maximum adsorption capacity (Qmax) of ZnAl-EC was 384.61 mg/g. The ZnAl-EC can be reused up to 4–5 cycles with 4th and 5th cycle % adsorption of 75.10% and 48.83%. Kappa-carrageenan functional groups were identified by FTIR interacting on the surface and dyes adsorbed on the surface of ZnAl-EC seen from the d-space from 7.79 Å to 7.65 Å from XRD data analysis reinforced by XPS analysis in the Al2p spectrum with a binding energy of 72.91 eV and in S2p spectra showing a shift in binding energy and an increase in SO 3 - intensity. The success of this study shows the success of ZnAl-EC in regeneration and has a large adsorption capacity and shows the stability of the material as an adsorbent. [ABSTRACT FROM AUTHOR]

Macroalgae are attractive feeds for biobased platform chemicals like 5-hydroxymethylfurfural (HMF) and levulinic acid (LA). We here report an experimental study to obtain HMF from a representative red macroalgae using a biphasic water/MIBK system and a dual catalyst consisting of HCl and AlCl 3 in a batch set-up. The highest HMF yield was 64.0 mol% based on C6 sugars in the feed, corresponding with a carbon yield of 37.9 % (30 min, 153 °C, 0.04 M of AlCl 3 and 0.04 M of HCl, 5 wt% macroalgae (Eucheuma cottonii , EC), intake on water). A reaction network is proposed based on major and minor products identified in the liquid phase during reaction and involves both D-galactose and 3,6-anhydro-D-galactose. Red seaweed species Eucheuma cottonii was thought to have potential as an anesthetic agent because it is shown to contain compounds with bioactivity as antimicrobial, antifungal, and also anticancer. There have been no studies regarding the active ingredients of anesthetized. The objective of the study was to examine the potential of E. cottonii extract as a candidate for fish anesthetic based on toxicity, phytochemistry, and active compounds contained in the extract. The stages of research include extraction, toxicity test extracts (BSLT), phytochemical test, quantitative test compound phytochemical results, and the identification of active compounds with LC-MS. Test results showed that E. cottonii extract contains a total of phenols of 4.60 ± 0.06 mg/GAE g which is one presentation of the active ingredient in the extract. The extract had bioactivity with LC50 of 221.71 ± 5.74 µg/mL. E. cottonii extract contained flavonoids compounds and saponins with a consecutive number of 24.48% and 0.22%. The results of identifying active compounds were found in several compounds that had activity asantidepressants or sedatives. We could prepare disk-shaped fibers without particular mechanical treatments from Eucheuma cottonii , the commonly used red algae for obtaining carrageenan. After carrageenan extraction from cottonii , the residues were bleached using chlorine dioxide and hydrogen peroxide. The morphology of the bleached fiber was disk-shaped one with a very thin fiber wall thickness of less than 100 nm and a diameter of approximately 100 μm. The sugar analysis and X-ray diffraction of the bleached fibers showed that they consisted of mostly glucose and had the same pattern as cellulose I with more than 50% crystalline structure, respectively. Compared to one-dimensional cellulose micro- or nanofibrils, which exhibits slow drainage and possess intolerably high drying energy, these two-dimensional disk-shaped fibers, when formed a layer in water medium, exhibit fast drainage and low drying energy. The formed sheet resulted in excellent transparency and high oxygen barrier property. Therefore, by using these disk-shaped, thin fibers from cottonii , we expect that the biodegradable and transparent oxygen barrier layer can be produced at a paper machine, which is, if possible, extremely difficult in the case of cellulose micro- or nanofibrils due to their slow drainage and high drying energy. Green reinforcement in biopolymer for enhanced industrial application is on the increase. Bio-precipitated CaCO3 was synthesised via the process of microbially-induced calcium carbonate precipitation using Bacillus sp. strain. The bio-precipitated CaCO3 (B-PCC) and conventional CaCO3 precipitates (C-PCC) reinforcement effect on the properties of Eucheuma cottonii film was analysed and compared. The biocomposite films were fabricated using a solvent casting technique. The physicomechanical, thermal and biodegradability performances of B-PCC and C-PCC incorporated E. cottonii film with increasing fillers' contents [0.05, 0.10, 0.50, 1.00, 2.00 (wt.%)] were determined and compared through a range of testing include water vapour permeability (WVP), contact angle using sessile drop method, scanning electron microscopy (SEM), tensile, elongation, Young's modulus, thermogravimetric analysis (TGA), and soil burial test. A further comparison was made between the E. cottonii biocomposite film with the existing biodegradable mulch films to evaluate its potential application using wet cup method. Results revealed that biocomposite films embedded with 0.1% B-PCC appeared to exhibit higher water barriers and hydrophobicity behaviours than those embedded with C-PCC. However, the mechanical strength and thermal stability were slightly lower than those embedded with C-PCC. E. cottonii biocomposite films, especially those embedded with 0.1 wt. % B-PCC has shown the potential in agricultural mulch film as it promoted better biodegradation and tensile strength by 34.24% and 20% respectively compared to the existing biodegradable PLA/PBAT mulch film, making it a promising alternative to the conventional plastic to mitigate plastic pollution.

Seaweed has a high potential value after being processed into semi-pure carrageenan flour (Semi Refined Carrageenan, SRC) and pure carrageenan (Refined carageenan, RC). Carrageenan is a yellowish white flour from seaweed type Eucheuma cottonii, which can form a gel so that it plays a very important role in the food and medicine industry, including as a stabilizer, thickener and emulsifier. This research examines the comparision of the effectiveness of three solutions, namely soaking with rice washing water, well water, and CaCO3 2% solution in the initial immersion stage. The next stage examines the effect of different drying times using manual drying methods by exposing the carrageenan extract to the sun for 6 and 8 hours. The test results of sulfate level, moisture level, ash level, appearance, odor, texture, and color showed that the soaking treatment with rice washing water and drying for 6 hours (A2B1) with data were 403.4 mg / Kg (sulfate level); 6.42% (water levelt); 24.42% (ash level); 8,4 (appearance); 6.9 (odor); 8.5 (texture); and 8.1 (color). Further research can be focused on the closed SRC drying process to avoid environmental contamination and the effect of storage under vacuum and non-vacuum conditions on the chemical, physical, and organoleptic characteristics of SRC flour.

To screen, prepare, identify, and evaluate the activities of natural antioxidants for treating chronic diseases caused by oxidative stress. Two algal proteins, namely ZD10 and ZD60, precipitated with 10 and 60% (NH
4 )2 SO4 were extracted from red algae Eucheuma cottonii (E. cottonii) and hydrolyzed using five proteolytic enzymes. The results showed that ZD60 played the most significant role in the enhancement of 2,2-diphenyl-1-picrylhydrazyl radical (DPPH⋅) scavenging activity (25.91 ± 0.24%) among all protein hydrolysates. Subsequently, six antioxidant peptides (EP1-EP6) were isolated from the papain hydrolysate of ZD60 by ultrafiltration and chromatography methods. Their amino acid sequences were identified as Thr-Ala (EP1), Met-Asn (EP2), Tyr-Ser-Lys-Thr (EP3), Tyr-Ala-Val-Thr (EP4), Tyr-Leu-Leu (EP5), and Phe-Tyr-Lys-Ala (EP6) with molecular weights of 190.21, 263.33, 497.55, 452.51, 407.51, and 527.62 Da, respectively. Of which, EP3, EP4, EP5, and EP6 showed strong scavenging activities on DPPH⋅, hydroxyl radical (HO⋅), and superoxide anion radical (O- 2⋅). Moreover, EP4 and EP5 could significantly protect human umbilical vein endothelial cells (HUVECs) from H2 O2 -induced oxidative damage by increasing the levels of antioxidant enzyme systems including superoxide dismutase (SOD) and glutathione peroxidase (GSH-Px) to reduce the levels of reactive oxygen species (ROS) (60.51 and 51.74% of model group) and malondialdehyde (MDA) (75.36 and 64.45% of model group). In addition, EP4 and EP5 could effectively inhibit H2 O2 -induced apoptosis by preventing HUVECs from early apoptosis to late apoptosis. These results indicated that the antioxidant peptides derived from E. cottonii , especially EP4 and EP5, could serve as the natural antioxidants applied in pharmaceutical products to treat chronic cardiovascular diseases caused by oxidative damage, such as coronary heart disease, atherosclerosis, etc. The mechanisms of antioxidant peptides EP4 and EP5 on protecting HUVECs from oxidative damage. Bioactive compounds in Eucheuma cottonii include phenols or polyphenols with antioxidant, antibacterial, and antiviral properties. Seaweed quality, including phenolic content, is highly determined by several factors, including the growth location, cultivation technique, and harvest age. This study aimed to determine the effect of harvest age and solvent type on the phenolic content of E.cottonii. The effects of harvest age on water content and of simplicia on the extraction process were also determined. Phenolic active ingredients were extracted at three harvest ages (35, 40, and 45 days) and using four solvents (ethanol, ethyl acetate, hexane, and water). Analysis of variance was performed to determine the effects of both factors and their interactions. The older the seaweed, the lesser the water content. The powder produced at all ages is a good simplicia for materials extraction. Results showed that high phenolic content was obtained from the E. cottonii extract at the age of 35 days with ethyl acetate as the solvent.

Snakehead fish (Channa striata), a freshwater fish, is known to have functional properties in helping the process of healing burns and wounds after surgery. Besides that, the use of seaweed as a source of food fiber is also widely reported. This research was conducted to design fiber-rich fish nuggets. The purpose of this study was to determine the maximum levels of seaweed substitution in the processing of snakehead fish nuggets based on sensory properties. Single-factor experiment (seaweed, Eucheuma cottonii, substitution) arranged in a Completely Randomized Design with four replications was applied in this study. The parameters observed were hedonic and hedonic quality sensory properties for the attributes of color, aroma, texture, and taste of nuggets. The data obtained were analyzed by the Friedman Test, followed by the Wilcoxon Rank Test to detect the differences between treatments. Treatment of up to 75% seaweed substitution (0, 25, 50, and 75%) decreases significantly (p 0.05) the hedonic sensory acceptance of the snakehead fish nuggets for all attributes except the texture which shows an increase. Whereas, the treatment with substitution of seaweed in the narrow percentage of 0, 5, 10, 15, and 20% gave an expected result, which showed that there was no significant difference (p 0.05) of the hedonic sensory response for the overall acceptance. Nevertheless, 15% of seaweed substitution provides the best hedonic sensory response for overall acceptance. These results indicate that 15% of seaweed (Eucheuma cottonii) substitution is recommended for use in the processing of snakehead fish nuggets.

Semi-refined carrageenan (SRC) diproduksi secara komersial dari rumput laut jenis Eucheuma cottonii, melalui proses pemanasan menggunakan larutan alkali dan proses pengolahan proses pemisahan atau pemulihan, dan proses pengeringan SRC. Salah satu alternatif untuk proses pengeringan SRC adalah dengan menggunakan gelombang mikro, di mana proses pengeringan dapat berlangsung lebih cepat. Penelitian ini bertujuan untuk menentukan waktu optimal dan pemodelan matematis dari pengeringan menggunakan gelombang mikro dalam proses pengeringan SRC. Penelitian ini dilakukan dengan mengekstraksi 200 gr rumput laut E. cottoni dengan KOH 9% selama 1 jam, kemudian rumput laut dikeringkan dengan variasi waktu pengeringan 5, 10, 15, 20, 25, dan 30. Diperoleh model pengeringan halaman termodifikasi sebagai model yang layak dalam merepresentasikan model matematis pengeringan SRC, dimana nilai R adalah 0.9876 dan RSME adalah 0.00483. Waktu pengeringan optimum yang diperoleh dalam penelitian ini adalah 30 menit. Semi-refined carrageenan (SRC) was a processed seaweed product, with export value in 2018 reached 4.942 thousand tons. Semi-refined carrageenan (SRC) was produced commercially from Eucheuma cottonii type seaweed, through a heated process used alkali solution and the process of separation or recovery process, and the drying process of SRC. One alternative for the SRC drying process is by using microwaves, where the drying process can take place faster. The purpose of this studied was to determine the optimum time and mathematical modelling of drying used microwaves in the SRC drying process. This study was carried out by extracting 200 gr of E. Cottoni seaweed with 9% KOH for 1 hour, then dried seaweed with a variation of dried time 5, 10, 15, 20, 25, 30, 40, and 45 minutes. In this study, the modified page drying model was obtained as a feasible model in representing the mathematical model of SRC drying, where the values of R is 0.9876 and RSME is 0.00483. The optimum drying time obtained in this study was 30 minutes.

One of the professions in the marine sector that is mostly occupied by people living in coastal areas is seaweed cultivation. Seaweed is one of the marine product commodities with great potential to be developed in Indonesia because it has high economic value. One of the areas that are included as producers of Eucheuma Cottonii seaweed is Nunukan Island, which is located in Nunukan Regency, North Kalimantan Province. The main factor that determines success in seaweed cultivation activities is the selection of land locations. Errors in site selection can lead to crop failure and low quality of the seaweed produced. The purpose of this study is to create a decision support system to facilitate and assist the community in selecting the best location for seaweed cultivation quickly and precisely according to the criteria using the Analytical Hierarchy Process (AHP) method to calculate the criteria weights and the Simple Additive Weighting (SAW) method for performing alternative ratings. The criteria used were 7, namely depth, pH, current speed, brightness, temperature, salinity, and dissolved oxygen, while alternative data were 11 points of seaweed cultivation locations on Nunukan Island. Based on the results of the implementation of the two methods, recommendations for two locations for seaweed cultivation are Sei Banjar I and Sei Banjar II with the same preference value of 0.937 which is the highest value compared to other alternatives.

The effects of alkali type and pH on the physical properties of carrageenan have been extensively studied. However, their effects on certain characteristics of solid-state properties of carrageenan have not been identified. This research aimed to investigate the effect of alkaline solvent type and pH on the solid physical properties of carrageenan isolated from Eucheuma cottonii. Carrageenan was extracted from the algae using NaOH, KOH, and Ca(OH)2 at pHs of 9, 11, and 13. Based on the results of preliminary characterization, including yield, ash content, pH, sulphate content, viscosity, and gel strength, it was found that all samples followed Food and Agriculture Organization (FAO) specifications. The swelling capacity of carrageenan based on the type of alkali was KOH > NaOH > Ca(OH)2. The FTIR spectra of all samples were in agreement with that of standard carrageenan. The molecular weight (MW) of carrageenan using KOH as the alkali followed the order pH 13 > pH 9 > pH 11, while using NaOH, the order was pH 9 > pH 13 > pH 11, and while using Ca(OH)2, the order was pH 13 > pH 9 > pH 11. The results of the solid-state physical characterization of carrageenan with the highest MW in each type of alkali showed that the morphology of carrageenan using Ca(OH)2 has a cubic shape and is more crystal-like. The order of crystallinity of carrageenan using different types of alkali was Ca(OH)2 (14.44%) > NaOH (9.80%) > KOH (7.91%), while the order of density was Ca(OH)2 > KOH > NaOH. The order of solid fraction (SF) of the carrageenan was KOH > Ca(OH)2 > NaOH, while the tensile strength when using KOH was 1.17, when using NaOH it was 0.08, and while using Ca(OH)2, it was 0.05. The bonding index (BI) of carrageenan using KOH = 0.04, NaOH = 0.02, and Ca(OH)2 = 0.02. The brittle fracture index (BFI) of the carrageenan was KOH = 0.67, NaOH = 0.26, and Ca(OH)2 = 0.04. The order of carrageenan solubility in water was NaOH > KOH > Ca(OH)2. These data can be used as the basis for the development of carrageenan for excipients in solid dosage forms.

Eucheuma cottonii is red algae that contain high carbohydrates, especially carrageenan. Carrageenan is a sulfated polygalactan with 15-40% of ester-sulfate content. It is formed by alternate units of D-galactose and 3,6-anhydro-galactose joined by α-1,3 and β-1,4-glycosidic linkage. The galactose polymer therein can be hydrolyzed into Galactooligosaccharide (GOS)type prebiotics. Hydrolysis was done by low concentration of sulfuric acid and crude enzyme from inactivation of Vibrio algynoliticus cell broth. Combination of prebiotic and probiotic provide lactic acid bacteria (BAL) as well as its food sources in the colon. The aim of this research is to know the characteristics of BALgrowth and fermentation in hydrolysateof E. cottonii and to know the best method in producing synbiotic powdersfrom BAL grown in hydrolysate. Analysis conducted include the characteristics of synbiotic powders using two types of BAL, namely Lactobacillus casei and kefir bacteria (Lactobacillus spp.). Characteristics of the two types of BAL produced during fermentation were qualified as probiotics which were amounted to more than 108 cell/mL, pH value 0.2%. The results showed that the number of living bacteria was higherin the method of synbiotic productions after dryingof hydrolysates followed by inoculum addition, than the method of synbiotic productions which inoculum addition was donebefore drying.

Edible film karaginan merupakan salah satu alternatif pengganti kemasan plastik yang sulit terdegradasi di alam dan aman bagi bahan pangan. Karaginan merupakan hasil ekstraksi salah satu jenis rumput laut yaitu Eucheuma cottonii. Tujuan penelitian ini untuk mengetahui gugus fungsi dari edible film dengan variasi konsentrasi karaginan dan sifat permukaan dari edible film karaginan dengan SEM. Pemanfaatan karaginan hasil ekstraksi E.cottonii asal Aceh untuk pembuatan edible film diharapkan dapat meningkatkan potensi budidaya E.cottonii di Aceh. Tahapan penelitian dilakukan dengan dua tahap, tahap pertama proses ekstraksi karaginan dari E. cottonii dengan penggunaan KOH 14% dan tahap kedua pembuatan edible film dengan varisi konsentrasi karaginan 0,1%; 0,2%; 0,3% (b/v) dengan penambahan PEG sebagai plasticizer. Hasil FTIR menunjukkan gugus fungsi ester sulfat, ikatan glikosidik, 3,6 anhidro-d-galaktosa, gugus OH dan CH dengan bilangan gelombang berturut-turut pada rentang 1246,36-1246,9 cm-1 , 1072-1074 cm-1, 932-934 cm-1, 3370- 3382 cm-1, dan 2876-2880 cm-1. Perubahan bilangan gelombang edible film karaginan dibandingkan dengan FTIR karaginan hasil ekstraksi pada penelitian sebelumnya menunjukkan adanya interaksi antara karaginan dengan plasticizer PEG yang ditambahkan. Pengujian morfologi pada edible film pada konsentrasi karaginan 0,1% dan 0,2% dengan SEM menunjukkan permukaan yang homogen dan kontinyu, sedangkan pada konsentrasi 0,3% edible film tidak terbentuk.

Alkali treated cottonii (ATC) is a derivative product of Eucheuma cottoniithat is treated with alkali. This study used raw material of sap-free seaweed for ATC processing. Sap-free seaweed is a seaweed which thallus has been extracted with liquid known as sap. The use of sap-free seaweed as an ATC product is constrained by its low quality, so an effort to improve its quality is needed. Therefore the obejcetive of this Research was to get the optimal conditions of ATC processing. Experimental data were designed and analyzed using Response Surface MethodologyCentral Composite Design (RSM-CCD) using Design Expert 10.0.7® program. The optimization of ATC processing involved three components that were considered influential, namely KOH concentration, temperature, and processing time. Recommendation for optimal conditions issued by RSM-CCD on ATC processing from sap-free seawee d were 6 % KOH concentration at 75 oC for 120 minutes (93.1% desirability). The results of the response analysis showed a yield of 39.47% and a gel strength of 595.32 g/cm2. As a research control, the recommendation of RSMCCD used in ATC processing from nonsap-free seaweed was obtained yield of 36.81% and gel strength of 574.44 g/cm 2. ATC from sap-free seaweed has higher yield and gel strength than that from nonsap-free seaweed. Thismight be due to the sap-free seaweed was obtained using the proper sap extraction process and ATC processingunder optimal conditions, so that the quality of seaweed was better maintained.

Background: Eucheuma Cottonii is a type of red algae obtained from Sabah with main active component, sulfated polysaccharide or k‐carrageenan. Aims: The objective of this research was to evaluate the antioxidant, antibacterial and potential wound‐healing properties in aqueous extraction of E cottonii in order to meet the increasing demand for halal and natural cosmeceutical products. Methods and Results: Aqueous extract of E cottonii was investigated for active compounds by phytochemical screening and IR spectroscopy. Antioxidant activity was carried out using DPPH method, and the IC50 value was 1.99 mg/mL. Antibacterial activity was examined against Staphylococcus Aureus using Kirby‐Bauer disk diffusion method and showed 10.03 ± 0.06 mm zone of inhibition, achieved by 200 mg/mL of extracts. A wound was made by skin excision of area around 100 mm2 on each mouse. Test group was treated with aqueous extract gel (10% w/w); meanwhile, the mice that were treated with honey acted as the positive control group and the untreated mice as negative control group. Results showed that the wound contraction rate inclined to aqueous extracts as compared to untreated group (P <.05). Percentage of wound healing for aqueous extracts and untreated group were 87.7% ± 2.0% and 57.6% ± 5.3%, respectively. Water pollution can make many problems such as, the incidence of disease and poisoning. Pollution in water can produce free radicals and it is can trigger of disease for aquatic organisms. There is some efforts that can be done to provide this problem, such as chemical compound that can reduce the reaction of free radicals. Antioxidants are one of the chemical compounds that can reduce the activity of free radicals. Eucheuma cottonii is the one of a seaweed that has many in antioxidant compounds, such as phenol compounds, but it is also rich in iodine fiber and other important minerals. The method used in this research is descriptive explorative and experimental method. This research was conducted with several stages of seaweed extraction. Identification of Eucheuma cottonii extract is using FTIR test. The last stage is an antioxidant activity test that includes DPPH test (1,1-diphenyl-2-picrilhidrazil) and Inhibition Concentration 50 (IC 50). The results obtained in this study were based on FTIR test of antioxidant compound in Eucheuma cottonii seaweed extract. The one of compounds that have antioxidant activity include galaktosa-4-sulfat. Based on the results of antioxidant activity test using DPPH obtained that seaweed extract Eucheuma cottonii active as an antioxidant to ward off free radicals in the waters. The concentration of Eucheuma cottonii seaweed extract for preventive 50% concentration of DPPH is 39,926 ppm.

The objective of this work was to study the effect of seaweed (Eucheuma cottonii) flour addition on physicochemical and sensory characteristics of an Indonesian-style beef meatball. Seaweed flour (SF) was added to meatball batter at 0% (CON), 2.5% (SF2.5), 5.0% (SF5) and 7.5% (SF7.5) in weight/meat weight (w/w) basis. Proximate composition (moisture, crude protein, crude fat, crude fiber and ash content), pH, cooking loss, hardness, water-holding capacity, instrumental color and sensory properties (color, texture, aroma, taste and overall acceptability) of the boiled meatballs were evaluated. The effect of seaweed flour addition was very significant (P < 0.01) on moisture, crude fat, crude fiber, ash, cooking loss, hardness, water-holding capacity, lightness (L*), yellowness (b*) and all sensory properties, and was significant (P < 0.05) on crude protein, pH and the redness (a*) of a boiled Indonesian-style beef meatball. The addition of seaweed flour at more than 2.5% resulted in darker meatballs, with less protein and fat, and a stronger seaweed aroma. The results suggest that seaweed flour could be added at 2.5% (w/w) to produce an Indonesian-style beef meatball with enhanced fiber content. [ABSTRACT FROM AUTHOR]

Eucheuma cottonii (E. cottonii), Ganoderma lucidum (G. lucidum), and Gracilaria sp. are plants that contain high phytochemicals, such as flavonoids, polyphenols, saponins, and tannins. In this work, the phytochemicals were obtained using the subcritical water extraction (SWE) process. The SWE method uses water as a solvent in subcritical conditions. Therefore, the SWE process is an environmentally friendly process for extraction. In order to run the SWE process optimally, measurement of the extraction rate of SWE is needed. Calculation of the extraction rate of SWE process used first and second-order models according to Lagergren equation. SWE process was started by setting temperatures from 140 to 180oC at a pressure of 7 MPa and solvent flow rate of 1 ml/min. Before starting the extraction, the raw material was loaded into the extractor. The raw materials used were E. cottonii, G. lucidum, and Gracilaria sp. The extraction process was carried out for 3 hours, and the product was collected every 30 minutes. The collected product was put into a sample bottle and dried using a freeze dryer. After that, the products obtained were balanced by an analytical scale. Based on the result, the optimum temperature for the SWE process was 180oC for E. cottonii and G. lucidum and 160oC for Gracilaria sp. The yields of the SWE process under the optimum temperature were 85.37%, 58.42%, and 75.73% for E. cottonii, G. lucidum, and Gracilaria sp, respectively. The extract contained phytochemical compounds detected by highperformance liquid chromatography analysis. The kinetics model of extraction rate for all variables exhibited a second-order kinetics model that indicated that the extraction process was influenced by more than one factor. Bioactive compounds in Eucheuma cottonii include phenols or polyphenols with antioxidant, antibacterial, and antiviral properties. Seaweed quality, including phenolic content, is highly determined by several factors, including the growth location, cultivation technique, and harvest age. This study aimed to determine the effect of harvest age and solvent type on the phenolic content of E. cottonii. The effects of harvest age on water content and of simplicia on the extraction process were also determined. Phenolic active ingredients were extracted at three harvest ages (35, 40, and 45 days) and using four solvents (ethanol, ethyl acetate, hexane, and water). Analysis of variance was performed to determine the effects of both factors and their interactions. The older the seaweed, the lesser the water content. The powder produced at all ages is a good simplicia for materials extraction. Introduction: In this century, cancer has increased in incidence and become one of the deadliest disease in the world. However, to date, lung cancer treatments are still not fully effective, quite expensive and very exhaustive for the patient. Eucheuma cottonii is an abundant marine red macroalgae in Indonesia which have a potential anti-lung cancer properties. Aim of this research is to determine phytochemical profile of Eucheuma cottonii extracts, as well as to evaluate its antioxidant and cytotoxic effects on Lung A-549 cancer cells. Methods: Eucheuma cottonii obtained from Sorong beach, West Papua Province, Indonesia, were extracted with three different solvents, that is ethanol, ethylacetate, and n-hexane. These three Eucheuma cottonii extracts were identified for its phytochemical profiles, antioxidant activity by DPPH (2,2-diphenyl-1-picrylhydrazyl) assay, and cytotoxic activity on lung A-549 cells by MTT (3-[4,5-dimethylthiazol-2-yl]-2,5-diphenyltetrazolium bromide) assay. Results: Phytochemical analysis revealed that Eucheuma cottonii contains metabolites of triterpenoid and alkaloid. Antioxidant activity evaluation showed ethanol extract of Eucheuma cottonii has IC50 value of 559.76 µg/mL against DPPH free radical. Whereas cytotoxicity evaluation showed that ethanol extract and ethylacetate extract of Eucheuma cottonii have cytotoxic effects on Lung A-549 cancer cells, with IC50 value of 251.73 µg/mL and 261.41 µg/mL, respectively. Problems: The fishery potential that is often found in the waters of the city of Tarakan is Eucheuma cottonii seaweed. This type of seaweed is a potential fishery resource with high economic value. E cottonii seaweed is only sold in dry form and food processing, so the need for diversification of non-food products that can increase the selling value is higher. Purpose: The purpose of this study was to determine the formulation and characteristics of lip balm from Eucheuma cottonii seaweed. Methodology: The research was conducted in two stages, the first stage was the formulation and preparation of lip balm preparations and the second stage was the characterization of the lip balm formulations. Characterization includes smear test, organoleptic observation, homogeneity test, pH test, stability and melting point observation. Results/Findings: The results of this study showed that the pH measurement of seaweed lip balm preparations for each formula was 7.46 - 8.85. Based on the results of the homogeneity test the P1 formula showed the presence of coarse grains, the P2 formula showed medium and the P3 formula showed very fine grains. Based on the results of stability testing for each preparation, it showed that on days 0 and 7 it remained stable. However, the color faded during 14 and 21 days of observation at room temperature. Melt test of preparation P1 to 24 minutes, P2 to 27 minutes and P3 to 20 minutes., Masalah: Potensi perikanan yang banyak dijumpai di perairan kota Tarakan adalah rumput laut Eucheuma cottonii. Jenis rumput laut tersebut merupakan salah satu potensi sumber daya peikanan yang bernilai ekonomis tinggi. Rumput Laut E cottonii hanya dijual dalam bentuk kering dan pengolahan pangan, sehingga perlunya diversifikasi produk non pangan yang dapat meningkatkan nilai jual lebih tinggi. Tujuan: Tujuan dari penelitian ini adalah mengetahui formulasi dan karakteristik lip balm dari rumput laut Eucheuma cottonii. Metodologi: Penelitian dilakukan dengan dua tahapan, tahap pertama adalah formulasi dan penyediaan sediaan lip balm dan tahap kedua adalah karakterisasi formulasi lip balm. Karakterisasi meliputi uji oles, pengamatan organoleptik, pengujian homogenitas, pengujian pH, pengamatan stabilitas dan titik lebur. Temuan/Hasil Penelitian: Hasil penelitian ini menunjukkan pengukuran pH sediaan lip balm rumput laut tiap formula yaitu 7,46 - 8,85. Berdasarkan hasil pengujian homogenitas formula P1 memperlihatkan adanya butiran kasar, sediaan formula P2 memperlihatkannya sedang dan sediaan formula P3 memperlihatkan butiran sangat halus. Berdasarkan hasil pengujian stabilitas terhadap masing – masing sediaan menunjukkan bahwa pada hari ke 0 dan 7 tetap stabil. Namun, warna mengalami kepudaran dalam penyimpanan selama 14 dan 21 hari pengamatan pada suhu kamar. Uji lebur sediaan P1 menit ke 24, P2 menit ke 27 dan P3 menit ke 20. ABSTRAK Kondisi perairan laut yang fluktuatif dan cenderung ekstrim menyebabkan rumput laut mudah terserang penyakit ice-ice. Munculnya bercak putih pada thallus rumput laut Eucheuma cottonii yang terinfeksi penyakit ice-ice diduga disebabkan oleh bakteri patogen yaitu Pseudomonas spp. Tujuan yang ingin dicapai dalam pelaksanaan penelitian ini adalah untuk mengetahui pengaruh perbedaan salinitas terhadap viabilitas bakteri Pseudomonas spp dan untuk mengetahui pada salinitas berapakah bakteri Pseudomonas spp tidak dapat tumbuh. Bakteri diidentifikasi melalui uji biokimia, dan viabilitas diamati dengan menanam bakteri pada media TSA plate dengan salinitas 30, 32, dan 34 ppt dengan 3 kali pengulangan, dan untuk uji konfirmasi bakteri di tanam pada media TSA miring dengan kadar salinitas 0, 20, 40, 60, 80, dan 100 ppt. Hasil yang diperoleh menunjukan bakteri tumbuh dengan normal pada hampir semua media uji kecuali pada media 100 ppt, hal ini menunjukan bahwa bakteri Pseudomonas spp merupakan bakteri yang bersifat halofilik atau dapat tumbuh baik pada kadar salinitas yang tinggi. The Influence of Salinity Difference to Viability of Pseudomonas spp. Bacteria ABSTRACT The conditions of marine waters that fluctuate and tend to extremes can cause seaweed susceptible to ice-ice disease. The appearance of white patches on infected Eucheuma cottonii seaweed thallus is estimated as ice-ice disease which is caused by pathogenic bacteria of Pseudomonas spp The objectives of this research is to determine the effect of salinity difference to viability of bacteria Pseudomonas spp and to know on what salinity is the bacterium Pseudomonas spp that can’t grow. Bacteria were identified by biochemical tests. Viability was observed by planting bacteria on TSA plate medium with salinity 30 ppt, 32 ppt, and 34 ppt with 3 repetitions, and for bacterial confirmation test in planting on TSA tilting with salinity 0 ppt, 20 ppt, 40 ppt, 60 ppt, 80 ppt, And 100 ppt. The results showed that bacteria grew normally in almost all test media except in 100 ppt media, this showed that Pseudomonas spp bacteria were halophilic bacteria or could grow well at high salinity levels. Tujuan dari penelitian ini adalah untuk mengetahui kandungan senyawa aktif dan nilai SPF pada ekstrak rumput laut Turbinaria conoides dan Eucheuma cottonii yang dapat digunakan sebagai bahan aktif pada sediaan krim tabir surya. Hasil pengujian menunjukkan rumput laut Turbinaria conoides mengandung golongan senyawa fenol hidrokuinon, flavonoid, triterpenoid, steroid, dan saponin. Rumput laut Eucheuma cottonii hanya ditemukan senyawa alkaloid. Nilai SPF tertinggi pada rumput laut Turbinaria conoides terdapat pada ekstrak metanol (SPF 16.7), sedangkan untuk rumput laut Eucheuma cottonii terdapat pada ekstrak etil asetat (SPF 8.8). Hasil tersebut memperlihatkan bahwa ekstrak dari rumput laut Turbinaria conoides dan Eucheuma cottonii memiliki potensi yang baik jika digunakan sebagai bahan aktif pada sediaan krim tabir surya.

Bioethanol is one of the renewable resources derived from sugar fermentation process of carbohydrate substance with microorganismaid. Hydrolysis is the stage to get the simple sugar and is done with using kcarragenasethat is obtained from the isolation result of marine microbe in E.cottonii’s habitat. The objectives of this research were to gain time hydrolysis to produce the enzyme activity and reducing sugar highs by kkaragenase and to determine the best of the substrate concentration and k-carragenasein producing a reduction sugar and the highest ethanol. The research steps was started from isolate rejuvenation of IH22 microbe, enzyme production of crudeextraction k-carragenase and 80% acetone precipitation, determination of the hydrolysis timeand activity of k-carragenase. The next step was the substrate hydrolysis with amounts of 6%, 9%, and 12% (b/v) solids, by enzyme from acetone concentrations of 0%, 2.5%, and 5% (v/v) and then they were fermented for six days with the S.cereviseaeadapted yeast extract. Thek-carragenasse highest enzyme activity was obtained in incubation for 30-60 minutes while the highest reduction sugar was obtained inhydrolysis time for 120 minutes. During the hydrolysis,12% substrate concentration generated the highest reduction sugar and whole sugar of 3.21% and 9.89%, respectively, while 9% substrate with 5% enzyme treatment contained1.49% reduction sugar and 2.75% whole sugar, which was the best substrate in producing ethanol during fermentation process of 1.23% (v/v) and 0.82% (b/v) with 55.78% substrate efficiency and 70.44% fermentation efficieny.

Rumput laut jenis Ulva lactuca dan Eucheuma cottonii merupakan rumput laut yang dapat dijumpai di perairan Indonesia, akan tetapi pemanfaatannya sebagai produk nori belum dilakukan. Penelitian ini bertujuan untuk memproduksi nori dari rumput laut U. lactuca dan E. cottonii dan menguji karakteristik mutu fisik, kimia dan sensori. Dari hasil penelitian didapatkan bahwa campuran rumput laut U. lactuta dan E. cottonii dapat menghasilkan produk nori yang berkualitas. Hasil uji organoleptik menunjukkan produk nori yang dipanggang lebih dapat diterima dibandingkan nori tidak dipanggang. Karakteristik kimia dan fisik produk berbeda nyata pada taraf 5%. Hasil analisis karakteristik kimia menunjukkan bahwa produk nori rumput laut U. lactuca dan E. cottonii mempunyai kandungan protein yang lebih rendah (18.84%), kandungan karbohidrat yang lebih tinggi (62.31%), kandungan serat pangan yang lebih tinggi (36.76%) dan kapasitas antioksidan yang lebih rendah (43.01%) dibandingkan nori komersial (karbohidrat 41.8%, protein 40%, serat 21.3%, kapasitas antioksidan 51%).

Studi ini dimaksudkan untuk menginvestigasi karakteristik dan model matematika kurvapengeringan rumput laut Eucheuma cottonii. Hasil penelitian ini diharapkan dapat digunakanuntuk perancangan alat pengering rumput laut yang efisien. Pengeringan E.cottonii dilakukandengan menggunakan alat pengering laboratorium terkendali yang dilengkapi dengan sistemakuisisi data dengan kecepatan udara 0,5 m/detik. Empat variasi RH yang dilakukan pada suhu50 °C adalah 30%, 40%, 50% dan 60% serta empat variasi suhu yang dilakukan pada RH 40%adalah 40 °C, 50 °C, 60 °C dan 70 °C. Tiga model pengeringan yang diuji adalah model Newton, Henderson & Pabis dan Page. Simulasi model yang paling tepat ditentukan berdasarkan nilai R2 yang paling tinggi, serta nilai sum square error (SSE) dan root mean square error RMSE yang paling rendah. Laju pengeringan lapis tipis rumput laut E.cottonii umumnya berada pada periodelaju menurun. Hal ini sesuai dengan karakteristik pengeringan bahan-bahan biopolimer yang umumnya berlangsung dengan laju menurun. Hasil penelitian menunjukkan bahwa penurunan RH cukup efektif dalam menurunkan kadar air dibandingkan peningkatan suhu. Pada RH rendah yaitu 30% suhu 50 °C, penurunan kadar air 50% dicapai dalam waktu 80-85 menit jauh lebih cepat dari RH 60% suhu 50 °C yaitu 165-170 menit. Pada suhu tertinggi yaitu 70 °C RH 40%penurunan kadar air 50% dicapai dalam waktu 90 menit selisih 15 menit lebih cepat dibandingkan suhu terendah yaitu 40 °C RH 40%. Kajian ini mendapati model pengeringan E.Cotonii yang paling sesuai adalah model pengeringan Page dengan nilai R2, R2 terkoreksi, SSE and RMSE berturut - turut sebesar 0,98-0,99 ; 0,96-0,98 ; 0,0002-0,0126 dan 0,0002- 0,0206.

Background: Eucheuma cottonii (EC) is a rapidly growing red seaweed in Southeast Asia. Objectives: This study was performed to investigate the effects of EC extract on human keratinocyte migration. Materials and Methods: Cell migration was assessed using the scratch wound assay. Western blot analysis was performed to investigate the EC extract-induced signaling pathways. Results: EC extract promoted HaCaT keratinocyte migration in a concentration-dependent manner. To investigate the mechanism of EC extract-induced migration, we examined the migration-related signaling pathways. Western blot analysis showed that the EC extract showed no changes in extracellular signal-activated kinase (ERK) but showed slight Akt activation. In contrast, the EC extract strongly phosphorylated p38 mitogen-activated protein kinase (MAPK) and the subsequent downstream molecule, cAMP response element-binding protein (CREB). To examine the involvement of p38 MAPK pathway in EC extract-induced migration, SB203580, a specific inhibitor of p38 MAPK, was used. The results showed that EC extract-induced migration was abrogated by SB203580 pretreatment. In addition, SB203580 also blocked p38 MAPK and CREB activation. Conclusion: Taken together, these data suggest that EC extract promotes migration in human keratinocytes via p38 MAPK and CREB phosphorylation. [ABSTRACT FROM AUTHOR]

The effect of sonication pretreatment condition on Eucheuma cottonii and Gelidium amansii seaweed towards agar extraction wae studied. Four parameters were changed during sonication to investigate the effects on agar yield and quality. These parameters include the time interval, concentration ratio, frequency, and intensity. The highest amount of agar extracted from Eucheuma cottonii species could be obtained from the time interval of 30 min, seaweed weight to solvent volume ratio of 1:20, the frequency of 35 Hz, and the sonication power intensity of 30%. For Gelidium amansii species, the best agar yield also could be obtained from the time interval of 30 min, 1:20 of seaweed weight to water volume ratio, the frequency of 35 Hz, and power intensity of 30%. From the experiment, sonication pretreatment significantly influenced the yield and properties of extracted agar. The sonication with autoclaved seaweed produced agar containing less sulfate content, which is an excellent chemical property for gel electrophoresis applications. The gel strength of sonication with autoclaving for both seaweeds, Eucheuma and Gelidium species was the highest among those by sonication with direct heating, which proved that sonication pretreatment with autoclaving could enhance the physical properties of the agar. Abstract Response Surface Methodology (RSM) based on Central Composite Design (CCD) was employed to optimize the conditions of enzymatic hydrolysis and fermentation of the seaweed Eucheuma cottonii. The significant influence of cellulose loading, enzyme loading and incubation time in enzymatic hydrolysis that has been screened by Plackett-Burman Design (PBD) was optimized using CCD. The optimum glucose concentration of 24.24 g/L (81% glucose yield) was obtained at 3% (v/v) of cellulose loading, 4% (v/v) of enzyme loading and 54 h incubation time. Subsequently, PBD analysis showed the significant effects of inoculum concentration, pH, temperature and time on fermentation process. Further optimization study by CCD revealed that 12% (v/v) of inoculum concentration, pH 5.2, 32 °C and 72 h of fermentation time enhanced the bioethanol production up to 9.77 g/L with the yield of 0.40 g/g and 78% conversion efficiency. Thus, the RSM based optimization of bioethanol from Eucheuma cottonii showed satisfactory results in this research. In short, it can be concluded that this optimization approach will serve as a good foundation for the realization of a consistent bioethanol production in the future.
