The rapid range expansion of the toxigenic marine haptophytic alga Prymnesium parvum in inland aquatic systems across the southern USA and beyond has prompted great interest in the ecology and evolutionary biology of this invasive bloom-forming species. Researchers have speculated that increased toxicity and heterotrophy in suboptimal environments allow blooms to develop in these new inland habitats that seem to represent extremes relative to P. parvum's perceived optimal niche. We used a laboratory-based study to elucidate the roles of salinity and nutrient availabilities in P. parvum growth and toxicity under environmental conditions representative of hypereutrophic reservoirs of the southwestern USA in which P. parvum blooms are now common. We found evidence that nutrient conditions favoring toxigenesis in P. parvum are suboptimal for growth and bloom formation, whereas conditions conducive to high growth rates are less favorable for toxigenesis. In contrast, both growth and toxicity scaled positively with salinity. Taken in the context of the documented biogeography of P. parvum and that toxigenesis in P. parvum probably evolved in support of heterotrophic nutrient acquisition, our results suggest that P. parvum's apparent range expansion into and across the USA is driven by increasing availabilities of salty and nutrient-rich systems, rather than by its toxigenic abilities. Growing attention in aquatic ecology is focusing on biogeographic patterns in microorganisms and whether these potential patterns can be explained within the framework of general ecology. The long-standing microbiologist's credo 'Everything is everywhere, but, the environment selects' suggests that dispersal is not limiting for microbes, but that the environment is the primary determining factor in microbial community composition. Advances in molecular techniques have provided new evidence that biogeographic patterns exist in microbes and that dispersal limitation may actually have an important role, yet more recent study using extremely deep sequencing predicts that indeed everything is everywhere. Using a long-term field study of the 'invasive' marine haptophyte Prymnesium parvum, we characterize the environmental niche of P. parvum in a subtropical impoundment in the southern United States. Our analysis contributes to a growing body of evidence that indicates a primary role for environmental conditions, but not dispersal, in the lake-wide abundances and seasonal bloom patterns in this globally important microbe. Abstract: The toxigenic marine flagellate Prymnesium parvum was first recorded in Lake Texoma, OK-TX, USA, an impoundment of the Red and Washita Rivers, following a massive fish kill in January 2004. Results of a 4.5-year monitoring program, spanning five bloom periods, revealed that Prymnesium abundances in the lake were temporally and spatially variable—densities were higher in winter, near-shore, and in Red River-associated sampling sites; the largest blooms were in Lebanon Pool, a large backwater basin often disconnected from the main reservoir. Prymnesium blooms appeared to have been fueled by high nutrient concentrations, and winter-spring densities were positively correlated with chlorophyll a, conductivity, total phosphorus, total nitrogen, and microzooplankton biomass, and negatively correlated with molar total nitrogen:total phosphorus and cladoceran and total crustacean zooplankton biomass. Comparison of Prymnesium densities with hydrological data suggested that Prymnesium blooms in Lebanon Pool were highest when the pool was disconnected from the main reservoir; no bloom occurred in the winter of 2004–2005, the only year since the 2003–2004 invasion in which Lebanon Pool and Lake Texoma were connected during the winter months. Soil acidity or aluminum (Al) toxicity is a major limitation to crop production. In this paper, we examine the effects of surface-applied lime and gypsum on soil profile chemical properties that affect Al toxicity in short-term (1 year), medium-term (2 years and 8 months) and long-term (10 years) experiments. Sulfate applied to the soil surface as gypsum was leached rapidly to a depth of 40 cm in the short-term despite relatively low amounts (279 mm) of rainfall. In the medium and long-term experiments, 28–54% of the sulfate applied as gypsum was retained in the 0–50 cm soil layer due to adsorption and precipitation reactions. The combined application of lime and gypsum increased soil calcium, to a depth of 30 cm in the short-term and to a depth of 50 cm in the medium and long-terms. Increases in soil sulfate and calcium were associated with greater electrical conductivity to a depth of 50 cm for all sampling times. Application of lime alone had no impact on soil Al, pH, and calcium in the soil layers below 10 cm in the short and medium terms. In the long-term, increasing the rate of lime application from 2 to 8 t L ha−1 increased soil pH in the 10–20 cm soil layer while soil Al decreased to a depth of 30 cm. The combined use of lime and gypsum decreased soil Al in the 30–50 cm soil layer in the medium-term and the 20–30 cm soil layer in the long-term which was more than when only lime was applied. Hence, we recommend the use of lime plus gypsum for treating soils with subsoil Al toxicity. Surface (0–10 cm) and subsoil (soil layers below 10 cm) acidity and resulting aluminum (Al) toxicity reduce crop grain yields. In South Western Australia (SWA), these constraints affect 14.2 million hectares or 53% of the agricultural area. Both lime (L, CaCO3) and gypsum (G, CaSO4) application can decrease the toxic effect of Al, leading to an increase in crop grain yields. Within the region, it is unclear if G alone or the combined use of L and G has a role in alleviating soil acidity in SWA, due to low sulfate S (SO4–S) sorption properties of the soil. We present results from three experiments located in the eastern wheatbelt of SWA, which examined the short-term (ST, 2 growing seasons), medium-term (MT, 3 growing seasons), and long-term (LT, 7 growing seasons over 10 years) effects of L and G on grain yield and plant nutrient concentrations. Despite the rapid leaching of SO4–S and no self-liming impact, it was profitable to apply G, due to the significant ST grain yield responses. The grain yield response to G developed even following relatively dry years, but declined over time due to SO4–S leaching. At the LT experimental site had received no previous L application, whereas, at the ST and MT sites, L had been applied by the grower over the previous 5–10 years. For the LT site, the most profitable treatment for wheat (Triticum aestivum L.) grain yield, was the combined application of 4 t L ha−1 with 2 t G ha−1. At this site, the 0–10 cm soil pHCaCl2 was 4.6, and AlCaCl2 was greater than 2.5 mg kg−1 in the 10–30 cm soil layer. In contrast, at the ST and MT sites, the pHCaCl2 of 0–10 cm soil layer was ≥5.5; it was only profitable to apply G to the MT site where the soil compaction constraint had been removed by deep ripping. The use of L increases soil pHCaCl2, resulting in the improved availability of anions, phosphorus (P) in the LT and molybdenum (Mo) at all sampling times, but reduced availability of cations zinc (Zn) in the LT and manganese (Mn) at all sampling. Abstract: Within the last two decades, Prymnesium parvum (golden algae) has rapidly spread into inland waterways across the southern portion of North America and this organism has now appeared in more northerly distributed watersheds. In its wake, golden algae blooms have left an alarming trail of ecological devastation, namely massive fish kills, which are threatening the economic and recreational value of freshwater systems throughout the United States. To further understand the nature of this emerging crisis, our group investigated the chemical nature of the toxin(s) produced by P. parvum. We approached the problem using a two-pronged strategy that included analyzing both laboratory-grown golden algae and field-collected samples of P. parvum. Our results demonstrate that there is a striking difference in the toxin profiles for these two systems. An assemblage of potently ichthyotoxic fatty acids consisting primarily of stearidonic acid was identified in P. parvum cultures. While the concentration of the fatty acids alone was sufficient to account for the rapid-onset ichthyotoxic properties of cultured P. parvum, we also detected a second type of highly labile ichthyotoxic substance(s) in laboratory-grown golden algae that remains uncharacterized. In contrast, the amounts of stearidonic acid and its related congeners present in samples from recent bloom and fish kill sites fell well below the limits necessary to induce acute toxicity in fish. However, a highly labile ichthyotoxic substance, which is similar to the one found in laboratory-grown P. parvum cultures, was also detected. We propose that the uncharacterized labile metabolite produced by P. parvum is responsible for golden algae''s devastating fish killing effects. Moreover, we have determined that the biologically-relevant ichthyotoxins produced by P. parvum are not the prymnesins as is widely believed. Our results suggest that further intensive efforts will be required to chemically define P. parvum''s ichthyotoxins under natural bloom conditions. [Copyright &y& Elsevier]

Aluminum (Al) toxicity imposes a significant limitation to crop production in South Western Australia. This paper examines the impact of surface-applied lime and gypsum on soil solution chemistry in the short term (1 year) and the long-term (10 years) in water limited environments. In the experiments, we measured soil solution chemistry using a paste extract on soil profile samples collected to a depth of 50 cm. We then used the chemical equilibrium model MINTEQ to predict the presence and relative concentrations of Al species that are toxic to root growth (Al associated with Al3+ and AlOH2 or Toxic-Al) and less non-toxic forms of Al bound with sulfate, other hydroxide species and organic matter. A feature of the soils used in the experiment is that they have a low capacity to adsorb sulfate. In the short term, despite the low amount of rainfall (279 mm), sulfate derived from the surface gypsum application is rapidly leached into the soil profile. There was no self-liming effect, as evidenced by there being no change in soil solution pH. The application of gypsum, in the short term, increased soil solution ionic strength by 524–681% in the 0–10 cm soil layer declining to 75–109% in the 30–40 cm soil layer due to an increase in soil solution sulfate and calcium concentrations. Calcium from the gypsum application displaces Al from the exchange sites to increase soil solution Al activity in the gypsum treatments by 155–233% in the short term and by 70–196% in the long term to a depth of 40 cm. However, there was no effect on Toxic-Al due to Al sulfate precipitation. In the long term, sulfate leaching from the soil profile results in a decline in soil solution ionic strength. Application of lime results in leaching of alkalinity into the soil profile leading to a decreased Toxic-Al to a depth of 30 cm in the long term, but it did not affect Toxic-Al in the short term. Development of inhibitors for ubiquitin pathway has been suggested as a promising strategy to treat several types of cancers, which has been showcased by recent success of a series of novel anticancer drugs based on inhibition of ubiquitin pathways. Although the druggability of enzymes in ubiquitin pathways has been demonstrated, ubiquitin itself, the main agent of the pathway, has not been targeted. Whereas conventional enzyme inhibitors are used to silence the ubiquitination or reverse it, they cannot disrupt the binding activity of ubiquitin. Herein, we report that the scaffolds of sulfonated aryl diazo compounds, particularly Congo red, could disrupt the binding activity of ubiquitin, resulting in the activity equivalent to inhibition of ubiquitination. NMR mapping assay demonstrated that the chemical directly binds to the recognition site for ubiquitin processing enzymes on the surface of ubiquitin, and thereby blocks the binding of ubiquitin to its cognate receptors. As a proof of concept for the druggability of the ubiquitin molecule, we demonstrated that Congo red acted as an intracellular inhibitor of ubiquitin recognition and binding, which led to inhibition of ubiquitination, and thereby, could be used as a sensitizer for conventional anticancer drugs, doxorubicin.
