5. Relationship between indicators of faecal pollution and presence of pathogenic microorganisms in coastal seawater

Abstract

This study evaluated the relationship between the traditional indicators of faecal pollution, Total Coliforms (TC), Faecal Coliforms (FC) and Faecal Streptococci (FS), and the presence of Salmonellae in coastal seawater in addition, the sensitivity of coliforms and streptococci tests was calculated. Finally, the detection of Staphylococcus aureus was evaluated as a possible additional parameter of coastal seawater quality. The 308 samples tested were drawn from the coastal seawaters along the Lecce-Brindisi shoreline (Apulia-Italy). In accordance with the Italian regulations (DPR 470/82), routine detection of Salmonella spp in seawater is only performed when poor hygienic conditions are expected, whereas S. aureus is not a routine parameter. Of the samples tested, 20, 56 and 20 were found unsuitable for TC, FC and FS, respectively. Salmonella spp were isolated in 22 samples. These 14, 7 and 13 had low or absent TC, FC and FS densities. S. aureus was isolated in 41 samples; 16 of these 41 cases were observed in July. Sensitivity was 36.4%, 68.2% and 41% for total and faecal coliforms tests and streptococci test, respectively. Faecal coliforms seem therefore to better predict the presence of Salmonellae in seawater compared with TC and FS. Our data show that the presence of Salmonella spp is not necessarily correlated with great concentrations of faecal pollution indicators; therefore, it would be advisable to always perform the detection of Salmonella spp beside the traditional indicators. We also suggest, in accordance with others authors, including the detection of S. aureus as a supplementary indicator for the prediction of coastal seawater quality and associated health risks

Abstract

The Authors wanted to value the hygienic conditions of homegrown vegetables and vegetables grown for the market; they also wanted to establish if there was a relationship between fungi presence and faecal contamination level. The study was carried out on 100 samples of vegetables usually eaten raw; 70% of the latter was positive for the presence of fungi, 48% for the presence of FC and 8% for FS. Amongst the fungi, the most commonly isolated strains were Saccharomyces cerevisiae (21%) and Candida parapsilosis (11%). The conclusions drawn were that vegetables eaten raw were a real vehicle of pathogenic micro-organisms, or potentially so, mainly spread by the oral and faecal routes.

Abstract

I pesci, al momento della pesca, risultano più o meno inquinati in relazione alla qualità delle acque in cui vivono e al tipo di manipolazione che subiscono; tale contaminazione batterica oscilla tra 103ufc/cm2 di superficie corporea e tra 102-109 ufc/gr di intestino. Successivamente, si assiste ad una modificazione quali-quantitativa della microflora iniziale e molti batteri (spoilage flora) partecipano attivamente al processo di deterioramento.Essenzialmente, la contaminazione microbica del pesce, determina processi proteolitici, sebbene, soprattutto nella fase iniziale, siano i composti azotati non proteici ad essere metabolizzati dai microrganismi. Si formano così composti volatili come trimetilammina, dimetilammina, ammoniaca, che impartiscono odori anomali e associati al deterioramento. Scopo del nostro lavoro è stato quello di quantificare, in campioni di orate (Sparus aurata) provenienti da un allevamento di tipo intensivo (impianto a vasche) e in quelle provenienti da un allevamento di tipo semintensivo, alcune specie batteriche coinvolte nei processi di deterioramento in condizioni di refrigerazione. Su campioni di pesce appena pescati (conservati e trasportati in ghiaccio), è stata eseguita una cinetica di crescita microbica (tempo 0, 3 e 6 giorni) alla temperatura di 4°C su sezioni di pelle e muscolo, visceri e branchie. Sono stati valutati i seguenti parametri microbiologici: conta microbica totale, Pseudomonadaceae, Shewanella putrefaciens ed Enterobacteriaceae totali. Pur partendo da concentrazioni iniziali non significativamente diverse le orate provenienti da allevamento di tipo intensivo hanno evidenziato cariche microbiche più alte (superiori a 107ufc/g) a partire dal 3° giorno di conservazione per tutti i parametri considerati.Le orate provenienti da allevamento semintensivo sono risultate quindi microbiologicamente più stabili rispetto a quelle provenienti da allevamento intensivo. Pseudomonas fluorescens e Pseudomonas putida sono risultate le specie più abbondanti. Negli esemplari provenienti da allevamento intensivo sono state isolate anche Pseudomonas aeruginosa e Pseudomonas pseudomallei: l’isolamento di queste due specie fa ipotizzare una contaminazione da acque reflue nel sistema di alimentazione delle vasche dell’impianto di allevamento.

Abstract

A biomonitoring study using "Mytilus galloprovincialis" mussels was carried out, to estimate their possible employment as "natural indicators" of coastal bacterial pollution. The research was conducted from September 2001 to May 2002, twice in a month, near two coastal zones of Salento. We analysed both natural and cultivated mussels. The latter were placed in cages, after checking their starting level of microbiological pollution. The bacteriological analyses conducted simultaneously on mussels and their ambient water, were directed to the search of: total and faecal coliforms, faecal streptococci, Escherichia coli, Salmonella spp., Staphylococcus aureus and yeasts. In this preliminary study the mussels proved to be "sensitive indicators of faecal pollution", since they had an accumulation of the faecal indicators even if the water was not contaminated. Nevertheless a lower frequency in finding yeasts in the mussels (53.8%) in comparison with water (74.3%), the irregular presence of Salmonella spp. in water (3.75%) and its absence in bivalves, show the need of further studies, also through "in vitro" tests, in order to verify absorption/elimination kinetics of microorganisms.

Abstract

The survival of fecal coliforms, fecal streptococci, Salmonella enteritidis and Staphylococcus aureus in sea water (35@1000 salinity) in the dark and at room temperature was evaluated. Staphylococcus aureus has been regarded for a long time as the new parameter to add to the classic ones in order to define the quality and suitability to recreational use. At the sperimental condition considered the survival of fecal coliforms, fecal streptococci, Salmonella enteritidis, and Staphylococcus aureus resulted equal to 48 h, 88 h, 56 h, and 96 h respectively. Fecal coliforms resulted the most sensitive bacteria to osmotic shock while Staphylococcus aureus the most adaptable species. Our results seem to indicate the opportunity of reconsidering the microbiological parameters used for the control of beach water.
