Odorant binding proteins (OBPs) play key roles in the insect olfactory system by assisting the neuronal response to hydrophobic odor molecules, understanding their interaction with ligands will facilitate the virtual screening of behaviorally active compounds in insects. Here, we successfully cloned and confirmed CmedOBP13, an antennae-biased OBP from the rice leaffolder Cnaphalocrocis medinalis, as a secreted protein. Recombinant CmedOBP13 was obtained using the Escherichia coli system, and its binding affinities to 35 volatile compounds emitted by rice plants and three sex pheromone components from female moths were assessed by a competitive binding assay. The results revealed that CmedOBP13 exhibited binding affinity to 23 rice volatiles, while no binding affinity for sex pheromone components. Furthermore, the stability of its conformation was found to be dependent on the pH level. Finally, the interaction between CmedOBP13 and odorants was predicted and confirmed by molecular docking and mutation functional assays, respectively. The combination of multiple hydrophobic residues created an adequate hydrophobic setting for ligands, and three residues (Glu13, Arg34, and Tyr115) might form hydrogen bonds with 15 odorants. Single mutations of Glu13, Arg34, Leu72, and Tyr115 diminished the binding affinities of CmedOBP13 to corresponding odorants, respectively. Odorant-binding proteins (OBPs) and chemosensory proteins (CSPs), which are thought to play key roles in the olfactory recognition of insects, can be induced by the odorants they recognize, but little is known about the underlying regulatory mechanisms. Here, we found that NlOBP8 and NlCSP10 play coordinative roles in the chemoreception of brown planthoppers (BPHs) to the volatile component linalool. Also, the relative mRNA levels of NlObp8 and NlCp10 decreased upon exposure to linalool. Further, homeotic protein distal-less (Dll), which was also highly expressed in the antennae, was found to positively regulate the transcription of NlObp8 and NlCsp10 directly. Knocking down NlDll expression downregulated the expression of many additional olfactory functional genes and impaired the repellent behavior of BPHs to linalool. Our findings elucidate the direct regulatory role of Dll in BPHs' olfactory plasticity to linalool through modulating the olfactory functional gene expression and could provide guidance to sustainably control BPHs in the field.

Climate change has resulted in frequent heavy and prolonged rainfall events that exacerbate waterlogging stress, leading to the death of certain alpine Rhododendron trees. To shed light on the physiological and molecular mechanisms behind waterlogging stress in woody Rhododendron trees, we conducted a study of Rhododendron delavayi , a well-known alpine flower species. Specifically, we investigated the physiological and molecular changes that occurred in leaves of R. delavayi subjected to 30 days of waterlogging stress (WS30d), as well as subsequent post-waterlogging recovery period of 10 days (WS30d-R10d). Our findings reveal that waterlogging stress causes a significant reduction in CO 2 assimilation rate, stomatal conductance, transpiration rate, and maximum photochemical efficiency of PSII (Fv/Fm) in the WS30d leaves, by 91.2%, 95.3%, 93.3%, and 8.4%, respectively, when compared to the control leaves. Furthermore, the chlorophyll a and total chlorophyll content in the WS30d leaves decreased by 13.5% and 16.6%, respectively. Both WS30d and WS30d-R10d leaves exhibited excessive H 2 O 2 accumulation, with a corresponding decrease in lignin content in the WS30d-R10d leaves. At the molecular level, purine metabolism, glutathione metabolism, photosynthesis, and photosynthesis-antenna protein pathways were found to be primarily involved in WS30d leaves, whereas phenylpropanoid biosynthesis, fatty acid metabolism, fatty acid biosynthesis, fatty acid elongation, and cutin, suberin, and wax biosynthesis pathways were significantly enriched in WS30d-R10d leaves. Additionally, both WS30d and WS30d-R10d leaves displayed a build-up of sugars. Overall, our integrated transcriptomic, physiological, and metabolomic analysis demonstrated that R. delavayi is susceptible to waterlogging stress, which causes irreversible detrimental effects on both its physiological and molecular aspects, hence compromising the tree's ability to fully recover, even under normal growth conditions.

Odorant binding proteins (OBPs) play an important role in insect peripheral olfactory systems and exploring the physiological function of OBPs could facilitate the understanding of insects' chemical communication. Here, the functional analysis of an antenna-based NlugOBP8 from brown planthopper (BPH) Nilaparvata lugens (Stål) was performed both in vitro and in vivo. Recombinant NlugOBP8 exhibited strong binding affinity to 13 out of 26 rice plant volatiles and could form a stable complex with 9 of them according to the fluorescence binding and fluorescence quenching experiments. Circular dichroism spectra demonstrated that six volatiles could give rise to significant conformational change of recombinant NlugOBP8. H-tube olfactometer bioassay confirmed that BPHs were significantly attracted by nerolidol and significantly repelled by linalool, caryophyllene oxide, and terpinolene, respectively. Antennae of ds NlugOBP8 -injected BPHs exhibited significantly lower electrophysiological response to linalool and caryophyllene oxide. Moreover, the repellent responses of BPHs to these two volatiles were also impaired upon silencing NlugOBP8 . These data suggest that NlugOBP8 is involved in recognizing linalool and caryophyllene oxide and provide additional target for the sustainable control of BPHs.

Insects are highly reliant on their active olfactory system in which odorant binding proteins play a role to selectivity and sensitivity during odour perception and processing. This study sets out to determine whether and to which extent the antennal loaded SaveOBP10 in English grain aphid Sitobion avenae, contributes in olfactory processing during host selection. To understand this possible relationship, we purified the SaveOBP10 recombinant protein and performed fluorescence ligand binding tests, molecular docking, RNA interference (RNAi) and behavioural trials. The results showed that SaveOBP10 had strong binding affinities (Ki ≤5 μM) with most of wheat plant volatiles at pH 5.0 as compared to pH 7.4. In Y-tube olfactometer bioassays, the S. avenae was attracted behaviourally towards pentadecane, butylated hydroxytoluene, tetradecane and β-caryophyllene however repelled by naphthalene. After RNAi of SaveOBP10, the aphid showed nonattraction towards β-caryophyllene and nonsignificant behavioural response to pentadecane, butylated hydroxytoluene and tetradecane. Furthermore, the three-dimensional structure modelling and molecular docking of SaveOBP10 were performed to the volatiles with high binding abilities. Together these findings indicate that SaveOBP10 can bind more strongly to the volatiles that involved in S. avenae behaviour regulation and possibly will contribute effectively in S. avenae integrated pest management., (© 2022 Royal Entomological Society.)

Insect odorant-binding proteins (OBPs) are a class of small soluble proteins that can be found in various tissues wherein binding and transport of small molecules are required. Thus, OBPs are not only involved in typical olfactory function by specific activities with odorants but also participate in other physiological processes in non-chemosensory tissues. To better understand the complex biological functions of OBPs, it is necessary to study the transcriptional regulation of their expression patterns. In this paper, an apparent gradient expression pattern of Obp19, that was highly and specifically expressed in antennae and played an essential role in the detection of camphene, was defined in the antennae of the Japanese pine sawyer. Further, the transcription factor BarH1, that also presented gradient expression pattern in antennae, was found to regulate expression of Obp19 directly through binding to its upstream DNA sequence. The condition of BarH1 gene silence, the gene expression levels of Obp19 significantly decreased. At the same time, additional olfactory genes also were regulated and thus influence camphene reception. These findings provide us an opportunity to incorporate Obps in the gene regulatory networks of insects, which contribute to a better understanding of the multiplicity and diversity of OBPs and the olfactory mediated behaviors., (Copyright © 2021 Elsevier Ltd. Petal blight caused by fungi is among the most destructive diseases of Rhododendron, especially Rhododendron agastum. Nonetheless, the metabolite changes that occur during petal blight are unknown. We used untargeted gas chromatography time-of-flight mass spectrometry (GC-TOF-MS) and ultra-high performance liquid chromatography quadrupole time-of-flight tandem mass spectrometry (UHPLC-QTOF-MS/MS) to compare the metabolite profiles of healthy and petal blight R. agastum flowers. Using GC-TOF-MS, 571 peaks were extracted, of which 189 metabolites were tentatively identified. On the other hand, 364 and 277 metabolites were tentatively identified in the positive and negative ionization modes of the UHPLC-QTOF-MS/MS, respectively. Principal component analysis (PCA) and orthogonal projections to latent structures-discriminant analysis (OPLS-DA) were able to clearly discriminate between healthy and petal blight flowers. Differentially abundant metabolites were primarily enriched in the biosynthesis of specialized metabolites. 17 accumulated specialized metabolites in petal blight flowers have been reported to have antifungal activity, and literature indicates that 9 of them are unique to plants. 3 metabolites (chlorogenic acid, medicarpin, and apigenin) are reportedly involved in resistance to blight caused by pathogens. We therefore speculate that the accumulation of chlorogenic acid, medicarpin, and apigenin may be involved in the resistance to petal blight. The aim of this study was to fully understand the pollution characteristics and sources of PM 2.5 in Zhengzhou, and to investigate the differences in four seasons and between urban and suburban areas. At the Zhengzhou environmental monitoring center (urban areas) and Zhengzhou University (suburban areas), 1284 environmental PM 2.5 samples were collected in the four seasons of 2018. The concentrations of nine kinds of inorganic water-soluble ions, organic carbon, elemental carbon and 27 kinds of elements, were measured by ion chromatography, carbon analyzer, and X-ray fluorescence spectrometry, respectively. Enrichment factors (EF), index of geoaccumulation ( I geo ), potential ecological risk index (RI), chemical mass balance model (CMB), backward trajectory, and potential source contribution function were the methods used to study the chemical component characteristics and source differences of PM 2.5 in different seasons in the urban and suburban areas of Zhengzhou. The results showed that the annual average PM 2.5 concentration at the Zhengzhou environmental monitoring center and Zhengzhou University sites reached (59.7±24.0) μg·m -3 and (74.7±13.5) μg·m -3 , respectively. The PM 2.5 concentration at the suburban point was higher than at the urban point with the exception of winter, and the seasonal mean concentration decreased in the order of winter > autumn > spring > summer. Compared with the urban areas, the suburban areas were more affected by crustal substances in spring, and the concentrations of all components were higher in summer and autumn than the urban areas. Nevertheless, urban areas were more affected by coal burning sources and motor vehicle sources in winter. The component analysis results showed that the influences of soil dust and building dust were greater in the suburbs in spring than in the urban areas. In autumn, the suburbs were more affected by biomass sources than the urban areas, while the urban areas were more affected by building dust than were the suburbs. The concentrations of Cu, As, Zn, Pb, and Sb were strongly influenced by anthropogenic sources, and the enrichments of Zn, Cu, As, and Pb in urban areas were greater than in the suburbs. In addition, Zn, Cu, As, and Pb exhibited potential ecological risks. The outcomes of the CMB model showed that dust sources, secondary sulfate, secondary nitrate, and coal burning sources contributed significantly to PM 2.5 concentrations in spring, summer, autumn and winter, respectively. The contributions of secondary pollution sources (secondary organic aerosol, secondary sulfate, and secondary nitrate) and motor vehicle sources to urban areas were higher than to suburban areas, and the influences of biomass sources in autumn and winter were significantly higher than in spring and summer and urban areas. The backward trajectory results indicated that the local PM 2.5 concentration was affected by distant transmission from the northwest except in summer, was affected by neighboring provinces in the east in four seasons, and was affected by transmission from the south, with the exception of winter. Furthermore, the consequences of potential sources demonstrated that the local PM 2.5 concentration was mainly affected by the potential areas in Henan province and its boundary with neighboring provinces.

Odorant binding proteins play a key role in the olfactory system and are involved in the odor perception and discrimination of insects. To investigate the potential physiological functions of SaveOBP9 in Sitobion avenae , fluorescence ligand binding experiments, molecular docking, RNA interference, and behavioral tests were performed. Fluorescence binding assay results showed that SaveOBP9 had broad and high (Ki < 10 μM) binding abilities with most of the wheat volatiles, but was more obvious at pH 7.4 than pH 5.0. The binding sites of SaveOBP9 to the volatiles were predicted well by three-dimensional docking structure modeling and molecular docking. Moreover, S. avenae showed a strong behavioral response with the four compounds of wheat. The reduction in mRNA transcript levels after the RNA interference significantly reduced the expression level of SaveOBP9 and induced the non-significant response of S. avenae to the tetradecane, octanal, decanal, and hexadecane. This study provides evidence that SaveOBP9 might be involved in the chemoreception of wheat volatile organic compounds and can successfully contribute in the integrated management programs of S. avenae .

Chemosensory proteins (CSPs) play important roles in insects' chemoreception, although their specific functional roles have not been fully elucidated. In this study, we conducted the developmental expression patterns and competitive binding assay as well as knock-down assay by RNA interference both in vitro and in vivo to reveal the function of NlugCSP10 from the brown planthopper (BPH), Nilaparvata lugens (Stål), a major pest in rice plants. The results showed that NlugCSP10 messenger RNA was significantly higher in males than in females and correlated to gender, development and wing forms. The fluorescence binding assays revealed that NlugCSP10 exhibited the highest binding affinity with cis-3-hexenyl acetate, eicosane, and (+)-β-pinene. Behavioral assay revealed that eicosane displayed attractant activity, while cis-3-hexenyl acetate, similar to (+)-β-pinene significantly repelled N. lugens adults. Silencing of NlugCSP10, which is responsible for cis-3-hexenyl acetate binding, significantly disrupted cis-3-hexenyl acetate communication. Overall, findings of the present study showed that NlugCSP10 could selectively interrelate with numerous volatiles emitted from host plants and these ligands could be designated to develop slow-release mediators that attract/repel N. lugens and subsequently improve the exploration of plans to control this insect pest., (© 2018 Institute of Zoology, Chinese Academy of Sciences.)

We explore the characteristics and sources of water-soluble ions in aerosol fine particulate matter (PM 2.5 ) samples collected from Anyang, China, during typical seasonal months from 2018 to 2019. Nine water-soluble ions (Na + , NH 4 + , K + , Mg 2+ , Ca 2+ , F - , Cl - , NO 3 - , and SO 4 2- ) were analyzed. The analysis of PM 2.5 , water-soluble ion concentration levels, anion-cation balance, nitrogen oxidation rate (NOR), sulfur oxidation rate (SOR), and ion correlation showed that the annual average concentrations of PM 2.5 and water-soluble ions in Anyang were (85.81±45.43) μg·m -3 and (48.21±30.04) μg·m -3 , respectively. Concentrations of ions were ranked as:NO 3 - > SO 4 2- > NH 4 + > Cl - > K + > Ca 2+ > Na + > Mg 2+ > F - . The annual average concentration of the sum of NH 4 + , NO 3 - , and SO 4 2- was (42.72±27.87) μg·m -3 , which accounted for 87.14% of total water-soluble ions. Moreover, NH 4 + was highly related to SO 4 2- and NO 3 - . The mean values of the nitrogen oxidation rate (NOR) and sulfur oxidation rate (SOR) were 0.25 and 0.37, respectively. These results suggest that these ions were the result of secondary formation. The anion-cation charge equivalent value was 0.75-0.94, which indicates that the sampled aerosols were alkaline. NH 4 + mainly existed in the form of (NH 4 ) 2 SO 4 and NH 4 NO 3 in spring, summer, and autumn, whereas in winter it mainly existed in the form of NH 4 Cl. The results of principal component analysis indicated that secondary aerosols, coal combustion, biomass burning, and dust were the main sources of the water-soluble ions in Anyang during the sampled periods.

Online monitoring data of atmospheric pollutants and meteorological parameters in Zhengzhou from 2014 to 2017 were collected to analyze the concentration levels, seasonal variations, and the ratio characteristics of atmospheric pollutants, as well as the effects of meteorological conditions on these pollutants. Results show that the annual average concentrations of PM 2.5 and PM 10 in the four years were (88±49.8), (95.8±60.2), (78.6±70.3), and (72.0±53.5) μg·m -3 , and (158.5±65.3), (167.7±82.6), (144.5±91.5), and (132.7±70.3) μg·m -3 , respectively, and were approximately two times higher than the grade Ⅱ annual limits set by China. Fuel combustion and the formation of secondary pollutants were the main sources of PM 2.5 in Zhengzhou. Moreover, the contribution of coal combustion was found to be decreased, while that of traffic sources increased year by year. Low wind speed, high humidity, and reduced precipitation are important meteorological factors that contribute to serious air pollution. In addition, the Potential Source Contribution Function and the Concentration-Weighted Trajectory were used to analyze the potential sources, and the contribution of these sources to the distribution of PM 2.5 . The potential source areas of PM 2.5 were mainly distributed in neighboring cities and provinces, such as Shanxi, Shaanxi, Hubei, Shandong, and Hebei. The short-distance transmission was found to contribute significantly to the mass concentration of PM 2.5 .

Reverse chemical ecology based on insect functional odorant binding proteins has been extensively studied to screen behaviourally active compounds, whereas chemosensory proteins (CSPs), which are reportedly involved in olfactory chemical reception and could serve as molecular targets remain unclear. In the present study, two behaviourally active compounds for Cnaphalocrocis medinalis, a serious pest of rice in Asia, were successfully screened via an antenna-biased CSP, CmedCSP33. Fluorescence competitive binding assays showed that CmedCSP33 could bind to seven out of 32 rice volatiles. Fluorescence quenching experiments revealed that CmedCSP33 forms a stable complex with nerolidol and β-ionone, and circular dichroism (CD) spectra demonstrated that these two compounds cause conformational changes in CmedCSP33. Furthermore, H-tube olfactometer bioassays showed that C. medinalis displayed prominent attractant responses to nerolidol and prominent repellent responses to β-ionone. Additionally, binding assays and CD spectra at different pH values implied that extensive conformational changes may be a general feature of CSPs for triggering the subsequent chemical transduction. In this study, meteorological and air mass concentration data of Zhengzhou from December 2017 to February 2018 (winter) were used to quantify the influence of meteorological factors on the PM. The Hybrid Single-Particle Lagrangian Integrated Trajectory model was used to analyze the 48-hour backward trajectories and the cluster method was applied to classify the airflow backward trajectory. Moreover, the potential source contribution function and concentration-weighted trajectory analysis were applied to evaluate the transport pathways and sources of PM 2.5 in Zhengzhou. The results show that the heavy pollution in Zhengzhou during winter is mainly due to the low wind speed, high relative humidity, and low precipitation. The cluster analysis revealed that up to 60% of the back trajectories came from the northwest and 25.56% of the back trajectories came from the Beijing-Tianjin area. The airflow trajectories from the south and east account for 7.5% and 6.1% with higher PM 2.5 concentrations. The main potential sources of PM 2.5 in Zhengzhou during winter are located in Beijing-Tianjin-Hebei air pollution transmission channel cities including Jiaozuo, Kaifeng, Xinxiang, Hebi, Puyang, Anyang, Handan, and Xingtai. The adjacent provinces, including Shanxi, Hubei, and Anhui, also have great influence on the PM 2.5 in Zhengzhou.

Tumor-associated macrophages (TAMs) and their alternative activation contribute greatly to the development of hepatocellular carcinoma (HCC). Receptor-interacting protein 140 (RIP140) is widely expressed in macrophages and regulates macrophage-mediated energy metabolism, the inflammatory response and tumorigenesis. However, whether RIP140 is involved in the activation of TAMs has not been reported. In the present study, we determined the expression of RIP140 in macrophages after treatment with HCC-conditioned medium (HCM) for 24 h. We also analyzed the effects of RIP140 overexpression on macrophage polarization, invasion and apoptosis of HepG2 and Huh7 cells. Transwell and apoptosis assays were used to estimated cell invasion and apoptosis. In addition, we investigated the effects of RIP140 overexpression in macrophages on the growth of H22 cells by subcutaneous injection of H22 cells along with macrophages in BALB/c nude mice. Western blotting and qRT-PCR were used to detect protein and mRNA expression associated with the NF-κB/IL-6 axis in TAMs. Immunohistochemical and immunofluorescence staining were used to evaluate the protein expression of RIP140 or F4/80 in human HCC samples. The protein expression of RIP140 in peripheral blood mononuclear cells was detected by western blotting. Kaplan‑Meier survival curve estimation of overall survival for patients with HCC was based on RIP140 or F4/80 expression in HCC samples. We found that HCM inhibited RIP140 expression and fostered the alternative activation of macrophages. RIP140 overexpression in TAMs significantly inhibited the alternative activation of macrophages by inhibiting the NF-κB/IL-6 axis in TAMs, and suppressed HCC cell growth both in vitro and in vivo. In addition, the protein expression of RIP140 in peripheral blood monocytes was significantly lower in patients with HCC than in healthy people, and this result was consistent with the expression of RIP140 in HCC samples. Furthermore, low RIP140 expression and high F4/80 expression were found to be closely correlated with shorter survival time of the patients with HCC.

Objective: To investigate the effects of inhibiting TIM-4 function in Kupffer cells (KCs) on liver graft rejection in mice and explore the underlying mechanism., Methods: Mouse models of orthotopic liver transplantation were treated with a control mAb group and TIM-4 mAb. The activated KCs were assayed with immunohistochemistry after operation. The expression of TIM-4 in KCs were assayed with Western blotting and RT-PCR and the levels of AST, ALT, TBIL, TNF-α, IFN-γ and CCL2 were assayed detected. The expression of TIM-4 in KCs was observed with laser confocal microscopy. HE staining was used to observe the microstructure of the liver tissues, and the number of CD25 + Foxp3 + T cells was determined using with flow cytometry; the proteins levels of p-P65and p-P38 were assayed with Western blotting. The donor mice were treated with clodronate liposomes to destroy the KCs in the liver before transplantation, and the liver grafts were examined for graft rejection., Results: The number of activated KCs in the liver graft increased progressively over time. Compared with the sham-operated group, the liver graft showed significantly increased TIM-4 protein and mRNA levels at 1, 3, and 7 days after transplantation (P<0.05) and increased levels of AST, ALT, TBIL, TNF-α, IFN-γ and CCL2 at 7 days (P<0.05). The graft in TIM-4 mAb group showed mild pathological changes with a mean RAI score of 2.67∓0.75, which was significantly lower than that in control mAb group (P<0.05). The mean survival time of the recipient mice was 53.8∓6.4 days in TIM-4 mAb group, significantly longer than that in the control mAB group (14.5∓2.9 days, P<0.05). Donor treatment with clodronate liposomes resulted in comparable RAI scores in TIM-4 mAb and control mAb groups (8.01∓0.64 vs 7.93∓0.56, P>0.05). The protein levels of p-P65 and p-P38 in TIM-4 mAb group were significantly lower than those in control mAb group (P<0.05), and CD25 + Foxp3 + T cells in the liver graft increased significantly in TIM-4 mAb group., Conclusion: Inhibition of TIM-4 function in KCs reduces the production of inflammatory factors after liver transplantation possibly by inhibiting the NF-κB and MAPK signaling pathways and promoting the proliferation of Foxp3 + Treg cells to induce allograft tolerance.

SUMMARY To determine risk factors for sporadic Vibrio parahaemolyticus gastroenteritis, we conducted a population-based case-control study in sentinel hospital surveillance areas of Shanghai and Jiangsu province, China. Seventy-one patients with diarrhoea and confirmed V. parahaemolyticus infections were enrolled, and they were matched with 142 controls for gender, age and residential area. From the multivariable analysis, V. parahaemolyticus infections were associated with antibiotics taken during the 4 weeks prior to illness [odds ratio (OR) 7·6, 95% confidence interval (CI) 1·1–54·4)], frequent eating out (OR 3·3, 95% CI 1·0–10·4), and shellfish consumption (OR 3·4, 95% CI 1·0–11·1), with population-attributable fractions of 0·09, 0·24, and 0·14, respectively. Protective factors included keeping the aquatic products refrigerated (OR 0·4, 95% CI 0·2–1·0) and pork consumption (OR 0·2, 95% CI 0·1–0·9) [corrected] .

Stearoyl-acyl carrier protein desaturase (SAD), locating in the plastid stroma, is an important fatty acid biosynthetic enzyme in higher plants. SAD catalyzes desaturation of stearoyl-ACP to oleyl-ACP and plays a key role in determining the homeostasis between saturated fatty acids and unsaturated fatty acids, which is an important player in cold acclimation in plants. Here, four new full-length cDNA of SADs (ScoSAD, SaSAD, ScaSAD and StSAD) were cloned from four Solanum species, Solanum commersonii, S. acaule, S. cardiophyllum and S. tuberosum, respectively. The ORF of the four SADs were 1182 bp in length, encoding 393 amino acids. A sequence alignment indicated 13 amino acids varied among the SADs of three wild species. Further analysis showed that the freezing tolerance and cold acclimation capacity of S. commersonii are similar to S. acaule and their SAD amino acid sequences were identical but differed from that of S. cardiophyllum, which is sensitive to freezing. Furthermore, the sequence alignments between StSAD and ScoSAD indicated that only 7 different amino acids at residues were found in SAD of S. tuberosum (Zhongshu8) against the protein sequence of ScoSAD. A phylogenetic analysis showed the three wild potato species had the closest genetic relationship with the SAD of S. lycopersicum and Nicotiana tomentosiformis but not S. tuberosum. The SAD gene from S. commersonii (ScoSAD) was cloned into multiple sites of the pBI121 plant binary vector and transformed into the cultivated potato variety Zhongshu 8. A freeze tolerance analysis showed overexpression of the ScoSAD gene in transgenic plants significantly enhanced freeze tolerance in cv. Zhongshu 8 and increased their linoleic acid content, suggesting that linoleic acid likely plays a key role in improving freeze tolerance in potato plants. This study provided some new insights into how SAD regulates in the freezing tolerance and cold acclimation in potato.

Background: Acute gastrointestinal illness (AGI) is an important public-health problem worldwide. Previous national studies of the incidence of AGI in China were performed decades ago, and detailed information was not available. This study therefore sought to determine the magnitude, distribution, and burden of self-reported AGI in China., Methods: Twelve-month, retrospective face-to-face surveys were conducted in 20 sentinel sites from six provinces between July 2010 and July 2011., Results: In total, 39686 interviews were completed. The overall adjusted monthly prevalence of AGI was 4.2% (95% confidence interval, 4.0-4.4), corresponding to 0.56 episodes of AGI per person-year. Rates of AGI were highest in children aged < 5 years. Healthcare was sought by 56.1% of those reporting illness. Of the cases who visited a doctor, 32.7% submitted a stool sample. The use of antibiotics was reported by 49.7% of the cases who sought medical care and 54.0% took antidiarrhoeals. In the multivariable model, gender, age, education, household type, residence, season, province and travel were significant risk factors of being a case of AGI., Conclusions: This first population-based study in China indicated that AGI represents a substantial burden of health. Further research into the specific pathogens is needed to better estimate the burden of AGI and foodborne disease in China.

Background and Aims: Cancer cells are thought to possess immune evasion properties due to FasL overexpression in many types of human tumors. In the present study, we set out to investigate the role of MAPK-ERK pathway in 67-kDa laminin receptor induced FasL expression and FasL-mediated apoptosis in human cholangiocarcinoma cells., Methods: The expression of FasL and its promoter activity in cultured cholangiocarcinoma cells were examined after treatment with laminin or transfection with plasmids containing siRNA targeted to 67-kDa laminin receptor. The effects of MAPK-ERK cascade inhibitor and c-Myc inhibition by siRNA on 67-kDa laminin receptor-induced FasL expression were determined. Apoptosis assay was performed to analyze the apoptosis of lymphocytes cocultured with cholangiocarcinoma cells treated with or without MAPK-ERK cascade inhibitor., Results: Our results revealed that the specific MAPK-ERK cascade inhibitor, PD98059, significantly attenuated phosphorylation of c-Myc on Ser-62 and FasL upregulation in QBC-939 cells and these cells showed decreased cytotoxicity against Fas-sensitive Jurkat T cells. A luciferase reporter assay revealed that FasL promoter activity was significantly reduced in cells treated with PD98059 or transfected with c-Myc siRNA., Conclusions: Based on these results, we conclude that 67LR induces FasL expression and cytotoxicity against Fas-sensitive Jurkat T cells in human cholangiocarcinoma cells through the phosphorylation of c-Myc on Ser-62 and the subsequent activation of the FasL promoter through the ERK pathway.

To investigate the impacts of large hydropower station in Gufu River on benthic algae, monthly samplings were conducted from September 2004 to June 2007 at the site GF04 which was impacted by the hydropower station, with the site GL03 in Gaolan River as reference. During sampling period, no significant differences were observed in the main physicochemical variables between GF04 and GL03, but the hydrodynamics differed significantly. GL03 was basically at a status of slow flow; while GF04, owing to the discharging from the reservoir, was at a riffle status during more than 60% of the sampling period. Such a difference in hydrodynamics induced significant differences in the community similarity of benthic algae and the relative abundance of unattached diatoms, erect diatoms, and stalked diatoms between GF04 and GL03, which could better reflect the impacts of irregular draw-off by large hydropower station on river eco-system.
