The role that visual discriminative ability plays among giant pandas in social communication and individual discrimination has received less attention than olfactory and auditory modalities. Here, we used an eye-tracker technology to investigate pupil fixation patterns for eight captive male giant pandas Ailuropoda melanoleuca. We paired images (N = 26) of conspecifics against: 1) sympatric predators (gray wolves, tigers), and non-threatening sympatric species (golden pheasant, golden snub-nosed monkey, takin, red panda), 2) conspecifics with atypical fur coloration (albino, brown), and 3) zookeepers/non-zookeepers wearing either work uniform or plain clothing. For each session, we tracked the panda’s pupil movements and measured pupil first fixation point (FFP), fixation latency, total fixation count (TFC) and duration (TFD) of attention to each image. Overall, pandas exhibited similar attention (FFPs and TFCs) to images of predators and non-threatening sympatric species. Images of golden pheasant, snub-nosed monkey and tiger received less attention (TFD) than images of conspecifics, whereas images of takin and red panda received more attention, suggesting a greater alertness to habitat or food competitors than to potential predators. Pandas’ TFCs were greater for images of black-white conspecifics than for albino or brown phenotypes, implying that familiar color elicited more interest. Pandas reacted differently to images of men vs women. For images of women only, pandas gave more attention (TFC) to familiar combinations (uniformed zookeepers and plain-clothed non-zookeepers), consistent with the familiarity hypothesis. That pandas can use visual perception to discriminate intra-specifically and inter-specifically, including details of human appearance, has applications for panda conservation and captive husbandry.

Different responses of gut microbiota diversity affect host health in term of nutrient metabolism and disease immunity, the reason and the impact of low diversity level of giant panda needs to be studied. Therefore, to explore the impact of intestinal type and diet on gut microbiota diversity of giant pandas, we conducted a multi-species comparison (n=73), and the effects of different diets on gut microbiota function of giant panda were analyzed by PICRUSt method. The gut microbiota of giant panda was significantly lower than the same diets type herbivores and the same intestinal type carnivores. High-fiber bamboo culm diet significantly reduced the gut microbiota diversity of giant panda than bamboo shoot and leaf. The low diversity of gut microbiota led to the low nutritional metabolic function of giant panda, including energy metabolism and amino acid metabolism. These results showed that the short intestinal tract and high fiber diet caused the decrease of gut microbiota diversity of giant pandas.

Reproductive and life-history characteristics, such as age at first reproduction, interbirth interval (IBI), sex ratio of newborns, and infant mortality, are crucial for assessing the population dynamics and survival of threatened species. There are no longitudinal data on the life histories of the threatened northern white-cheeked gibbons (Nomascus leucogenys) and yellow-cheeked gibbons (Nomascus gabriellae). Here, we examine the reproductive parameters of these species in 19 Chinese zoos. Some of these captive individuals may be hybrids of N. leucogenys × N. gabriellae, but it is not possible to distinguish these hybrids from the parent species based on morphological characteristics. Thus, in this study we identified individuals to species level from the zoos’ archives. Based on breeding records, 46 infants were born to 14 female northern white-cheeked gibbons from 2000 to 2019 and 89 infants to 29 female yellow-cheeked gibbons from 1995 to 2018. The mean age at first reproduction was 10.1 ± SD 2.4 yr for northern white-cheeked gibbons and 10.0 ± SD 3.1 yr for yellow-cheeked gibbons. The mean IBI was 32.8 ± SD 11.7 mo for northern white-cheeked gibbons and 27.2 ± SD 14.8 mo for yellow-cheeked gibbons. There was no significant effect of parity on IBI in northern white-cheeked gibbons, but primiparous mothers had a significantly longer IBI than multiparous mothers in yellow-cheeked gibbons. There was no significant difference in infant mortality between primiparous and multiparous mothers for either species. The sex ratio of newborns did not deviate significantly from 1:1 for either species. Infant mortality was 13% for northern white-cheeked gibbons and 12% for yellow-cheeked gibbons. Infant death reduced the IBI by 19 mo in northern white-cheeked gibbons and 10 mo in yellow-cheeked gibbons. Our data suggest that the intrinsic reproduction rate is very low in these gibbons and that wild populations may continue to decline without suitable conservation intervention.

Mammal gastrointestinal tracts harbor diverse bacterial communities that play important roles in digestion, development, behavior, and immune function. Although, there is an increasing understanding of the factors that affect microbial community composition in laboratory populations, the impact of environment and host community composition on microbiomes in wild populations is less understood. Given that the composition of bacterial communities can be shaped by ecological factors, particularly exposure to the microbiome of other individuals, inter-specific interactions should impact on microbiome community composition. Here, we evaluated inter-population and inter-specific similarity in the fecal microbiota of Przewalski’s gazelle (Procapra przewalskii), an endangered endemic ruminant around Qinghai Lake in China. We compared the fecal bacterial communities of three Przewalski’s gazelle populations, with those of two sympatric ruminants, Tibetan gazelle (Procapra picticaudata) and Tibetan sheep (Ovis aries). The fecal bacterial community richness (Chao1, ACE) did not vary across the three Przewalski’s gazelle populations, nor did the composition vary between species. In contrast, the managed Przewalski’s gazelle population had higher bacterial diversity (Shannon and Simpson) and was more similar to its sympatric Tibetan sheep in beta diversity than the wild Przewalski’s gazelle populations. These results suggest that ecological factors like host community composition or diet affect Przewalski’s gazelle’s gastrointestinal bacterial community. The role of bacterial community composition in maintaining gastrointestinal health should be assessed to improve conservation management of endangered Przewalski’s gazelle. More broadly, captive breeding and reintroduction efforts may be impeded, where captive management results in dysbiosis and introduction of pathogenic bacteria. In free ranging populations, where wildlife and livestock co-occur, infection by domestic pathogens and diseases may be an underappreciated threat to wild animals.

Mars Surface Composition Detector (MarSCoDe) is a scientific payload onboard the Mars rover of China’s Tianwen-1 mission. With the capability of 1.6∼7 m remote detection and analysis, MarSCoDe instrument suite consists of a two dimensional pointing mirror and an optical head outside the rover cabin, a Laser Induced Breakdown Spectroscopy (LIBS) spectrometer and a Short Wave Infrared (SWIR) spectrometer collectively covering 240∼2400 nm and a master controller unit inside the rover body, calibration target sets, optical fibers and power cables connecting the internal and external units. Combining the techniques of active LIBS, passive SWIR and micro-imaging, MarSCoDe provides functions including elemental composition discrimination and quantitative determination, classification of rock and soil characteristics, sample texture imaging and characterization of plasma-excited area. This paper introduces MarSCoDe mainly in terms of scientific objectives, design requirements, assembly and implementation, spectral and radiation calibration, and performance verification. The LIBS laser irradiance on the target can soundly exceed 10 MW/mm2, and the performance of the LIBS module operated at different temperatures has been tested. The field of view of the SWIR spectrometer is 36.5 mrad. The micro-imager can extract the central pixel area of 320 × 320 and 1024 × 1024, and the former is binned into 64 × 64. The 2D pointing mirror has a wide forward detection range and a narrow backward calibration range, with the pointing pitch accuracy better than 0.133°.

Exposure to sexual stimuli can lead to increased aggression in male mammals, but it is unclear whether the aggression is related to the receptiveness of the females. Interactions with receptive females elicit testosterone (T) pulses that are important for sexual behaviors. We investigated the effects of male–female interactions on subsequent aggressive behaviors and T responses in the golden hamster (Mesocricetus auratus (Waterhouse, 1839)). Three groups (n = 18, 17, and 18) of males were exposed to receptive females, nonreceptive females, and blank (control), respectively. Then, we randomly chose eight animals from each group and measured their aggression toward an unfamiliar male conspecific; the remaining 29 males were used for a T assay (to avoid effects of aggression on T levels). The results show that interactions with females led to significantly higher male aggression and T levels than were found in control males. The increased aggression was not related to the receptiveness of the females, but receptive females elicited higher levels of T in males than nonreceptive females. Our findings suggest that the elevated aggression following the sexual encounter may serve to defend the female, by enhancing the ability of males to exclude other males from the vicinity of females, whereas the post-encounter T release may serve to assist mating behaviors and reproductive success.

Reintroduction plays a vital role in conservation for many endangered species, however, little long-term information is available on the population dynamics and conservation status. Here we provide a detailed account of the Chinese Milu (Elaphurus davidianus) conservation and reintroduction efforts over the past 35 years, and give updated information on current Milu distribution, population dynamics and conservation status based on long-term monitoring (1985–2020) and a detailed follow-up investigation (2013–2020) in 235 wildlife institutions throughout China. Milu conservation in China comprised three phases: i) establishing ex situ populations and increasing the number of Milu through captive breeding (1985–1992); ii) preparing captive Milu for life in the wild and establishing in situ conservation populations (1993–1997); and iii) reintroduction of Milu into the wild throughout their historic range (1998–ongoing). Currently, there are ca. 9062 Milu (including 2825 wild individuals) distributed across 83 sites with 7380 individuals living at Beijing Milu Park, Jiangsu Dafeng Milu Nature Reserve and Hubei Shishou Milu Nature Reserve. The average birth rates in three sites were all over 0.200, and the average adult mortality rates were below 0.085, resulting in a rapid population growth. We discuss a variety of factors that contributed to ex situ conservation success in the reintroduction of a species formerly extinct in the wild, and highlight past and present challenges of Milu conservation in China. Our results will provide helpful information on conservation and reintroduction for other endangered species around the world.

OBJECTIVE The objective of the present study was to examine whether wild rodents exhibit diverse obesity susceptibility and what factors predispose subjects to this divergence in response to a high-fat diet (HFD). METHODS Sixty male and female Brandt's voles (Lasiopodomys brandtii) were fed an HFD for 8 weeks, and the upper (obesity prone [OP]) and lower (obesity resistant [OR]) one-third for mass gain were selected. Energy budgets and pathologic changes were measured. Another 30 males were fed a low-fat control diet (LFD) for 10 weeks and then fed an HFD for 12 weeks. The energetic parameters of the rodents on an LFD were analyzed for the correlation with body mass of the rodents on an HFD. RESULTS OP voles had higher energy intakes, higher levels of noradrenaline-induced nonshivering thermogenesis, and a greater impairment of insulin tolerance than OR voles. Unlike laboratory rodents, there were no differences in physical activity or resting metabolic rate between these groups of voles. The thermogenic capacity during LFD feeding was the strongest predictor for mass gain during HFD feeding. CONCLUSIONS This study suggests that a wild rodent species of Brandt's voles exhibits diverse obesity susceptibility in reaction to an HFD, providing a natural model to give insight into the mechanisms for divergent obesity susceptibility. This study also indicates that maximum thermogenic capacity has a predictive power for the development of obesity when an HFD was available.

Elevated glucocorticoid (GC) concentration and increased vigilance are two common responses to predation risk in mammals. Chronic high-level GC concentration and vigilance occur at the expense of other life maintenance and reproduction activities, reflecting a trade-off between individual survival and future fecundity. Przewalski’s gazelle (Procapra przewalskii) is a group-living ungulate endemic to the high-altitude Qinghai Lake region of China. Group-size effect on gazelle vigilance has been examined, yet little is known about how their GC concentration is affected by group size or reproductive status. In this study, we examined the effect of group size and reproductive status on faecal glucocorticoid metabolite (FGM) concentrations and individual vigilance during different stages of the reproduction cycle (i.e. non-breeding, lambing and rutting) in free-ranging adult female Przewalski’s gazelles. Group size did not influence FGMs significantly, but mean vigilance duration increased with group size. The gazelles’ FGMs and vigilance peaked in lambing season. FGMs showed no difference between rutting season and non-reproductive season, but vigilance was lowest in the rutting season. FGMs correlated with vigilance frequency and vigilance duration. Antipredator responses of female Przewalski’s gazelles appear to change with reproductive status but not with group size in free-ranging females. Management measures should be taken in the lambing season to minimize stress on mother gazelles.

The giant panda (Ailuropoda melanoleuca) has evolved a large number of mucous glands in the intestinal lining to adapt to the digestion of high-fiber foods. However, in captive pandas, excessive mucus might form a mass and then be eliminated, which is often accompanied by discomfort and decreased activity. This event is called &lsquo, mucous excretion&rsquo, The causes of mucus excretions in captive pandas, however, remain unknown. The aims of this study were to document the occurrence of mucus excretion and to investigate its possible associations with pandas&rsquo, age, gender, and feces output. Eighteen giant pandas were studied at the Beijing Zoo from April 2003 to June 2017, and a total of 900 occurrences of mucous excretion and 32,856 daily defecation outputs in weight were recorded. The likelihood of mucous excretion occurrence decreased by 11.34% for each 1 kg of fecal output (Z = &minus, 4.12, p &lt, 0.0001), while it increased by 5.89% per year of age (Z = 4.02, p &lt, 0.0001). However, individual differences in gender had no significant effect on the mucous occurrence (Z = &minus, 0.75, p = 0.4508). A monthly change in mucus occurrence was also found. The mean frequency of mucus occurrence was significantly higher in October. In August, time (month) change showed the biggest negative influence on feces output but the biggest positive influence on mucus excretion (seasonal factors were &minus, 2.261 and 0.0126, respectively). Our results documented the occurrence of mucous excretions and confirmed their possible associations with the pandas&rsquo, age and fecal output based on a 15-year prospective study. This study not only adds to our knowledge of panda physiology but also suggests the need for further studies examining the causes of the excretion of mucous stools in captive pandas. Reducing the incidence of mucous excretion would promote ex situ conservation and enhance panda welfare.

Seasonally reproducing animals show many behavioral and physiological changes during the mating period, including increased signaling for intrasexual competition and mate attraction. We collected 102 anogenital gland secretions (AGS) from marking trees in Foping Nature Reserve, and used gas chromatography mass spectrometry analyze these chemical composition. Of these marks, all but one were from males, confirmed with DNA analysis. We found that several chemical constituents, especially volatile compounds, is present only during the mating season and that the relative abundance of many compounds changed as a function of breeding season, whereas nonvolatile compounds were lower in the mating season. This seasonal variation in chemical composition of AGS most likely plays an important role in governing giant panda reproduction, including mate location, attraction, and male-male competition. The chemical properties of many of these putative chemosignals-such as volatility and longevity-are suggestive of these roles, and undoubtedly contribute to successful reproduction for this species with a characteristically sophisticated chemical communication system. We also found a number of important differences between the chemical constituents of AGS from wild pandas and those found in previous studies with captive pandas, suggesting that inappropriate chemosignal composition may contribute to poor reproductive success in captive breeding programs.

Vocal individuality is a prerequisite for individual recognition, especially when visual and chemical cues are not available or effective. Vocalizations encoding information of individual identity have been reported in many social animals and should be particularly adaptive for species living in large and complexly organized societies. Here, we examined the individuality in coo calls of adult male golden snub-nosed monkeys (Rhinopithecus roxellana) living in a large and multilevel society. Coo calls are one of the most frequently occurring call types in R. roxellana and likely serve as the signals for contact maintenance or advertisement in various contexts including group movement, foraging, and resting. From April to October 2016, April to July 2017, and September to October 2017, we recorded a total of 721 coo calls from six adult males in a provisioned, free-ranging group and one adult male in captivity in Shennongjia National Park, China. We selected 162 high-quality recordings to extract 14 acoustic parameters based on the source-filter theory. Results showed that each of all parameters significantly differed among individuals, while pairwise comparisons failed to detect any parameter that was different between all pairs. Furthermore, a discriminant function analysis indicated that the correct assignment rate was 80.2% (cross-validation: 67.3%), greater than expected by chance (14.3%). In conclusion, we found evidence that coo calls of adult male R. roxellana allowed the reliable accuracy of individual discrimination complementarily enhanced by multiple acoustic parameters. The results of our study point to the selective pressures acting on individual discrimination via vocal signals in a highly gregarious forest-living primate. Vocal signaling represents a primary mode of communication for most nonhuman primates. A quantitative description of the vocal repertoire is a critical step in in‐depth studies of the vocal communication of particular species, and provides the foundation for comparative studies to investigate the selective pressures in the evolution of vocal communication systems. The present study was the first attempt to establish the vocal repertoire of free‐ranging adult golden snub‐nosed monkeys (Rhinopithecus roxellana) based on quantitative methods. During 8 months in Shennongjia National Park, China, we digitally recorded the vocalizations of adult individuals from a provisioned, free‐ranging group of R. roxellana across a variety of social‐ecological contexts. We identified 18 call types, which were easily distinguishable by ear, visual inspection of spectrograms, and quantitative analysis of acoustic parameters measured from recording samples. We found a great sexual asymmetry in the vocal repertoire size (females produced many more call types than males), likely due to the sex differences in body size and social role. We found a variety of call types that occurred during various forms of agonistic and affiliative interactions at close range. We made inference about the functions of particular call types based on the contexts in which they were produced. Studies on the vocal communication in R. roxellana are particularly valuable since they provide a case about how nonhuman primates, inhabiting forest habitats and forming complex social systems, use their vocalizations to interact with their social and ecological environments., We identified 18 call types in the vocal repertoire of free‐ranging adult Rhinopithecus roxellana, a primate living in forest habitats and forming a complex multilevel society. The social‐ecological contexts were recorded for each call type.

Wild animals are affected by growing human interference in their habitats and inevitably react internally to such stimuli. This study explores inherent physiological parameters to assess the effect of human interference on giant pandas in their wild habitat. Ninety-one fecal samples were collected within four nature reserves on Qinling Mountain. Fecal cortisol determinations were performed by radioimmunoassay. We categorized the intensity of local human disturbance and tested for correlations between the intensity of human disturbance and fecal cortisol metabolite levels. The results show a significant positive correlation between giant panda fecal cortisol metabolite levels and the degree of disturbance in their habitat. This study is the first to use a non-invasive monitoring method to analyze wild giant panda habitat disturbance, and demonstrates that cortisol metabolite levels in panda dung can objectively reflect the degree of panda habitat disturbance. The results provide a relatively objective means and method with which to evaluate the quality of wild giant panda habitat. This study highlights the need to monitor the effects of human disturbance on wildlife and to implement new policies in the management of nature reserves.

International audience; The giant panda Ailuropoda melanoleuca belongs to the family of Ursidae; however, it is not carnivorous, feeding almost exclusively on bamboo. Being equipped with a typical carnivorous digestive apparatus, the giant panda cannot get enough energy for an active life and spends most of its time digesting food or sleeping. Feeding and mating are both regulated by odors and pheromones; therefore, a better knowledge of olfaction at the molecular level can help in designing strategies for the conservation of this species. In this context, we have identified the odorant-binding protein (OBP) repertoire of the giant panda and mapped the protein expression in nasal mucus and saliva through proteomics. Four OBPs have been identified in nasal mucus, while the other two were not detected in the samples examined. In particular, AimelOBP3 is similar to a subset of OBPs reported as pheromone carriers in the urine of rodents, saliva of the boar, and seminal fluid of the rabbit. We expressed this protein, mapped its binding specificity, and determined its crystal structure. Structural data guided the design and preparation of three protein mutants bearing single-amino acid replacements in the ligand-binding pocket, for which the corresponding binding affinity spectra were measured. We also expressed AimelOBP5, which is markedly different from AimelOBP3 and complementary in its binding spectrum. By comparing our binding data with the structures of bamboo volatiles and those of typical mammalian pheromones, we formulate hypotheses on which may be the most relevant semiochemicals for the giant panda.

The postural origin hypothesis and the task complexity hypothesis propose that hand preference in non-human primates evolved in association with body posture and task complexity, respectively. The results of previous studies testing these two hypotheses, however, vary greatly with the different primate species and methods used. To investigate the effect of body posture and task complexity on hand preference, we recorded bouts of hand usage in nine captive northern white-cheeked gibbons (Nomascus leucogenys) housed at Beijing Zoo as they reached for food items in a ground-reaching task, a box task, and a tube task. The results showed that four to seven of the nine gibbons displayed a hand preference at the individual level in different tasks, and that hand preference in individuals was task-specific; there was no group-level hand preference in any task. The box task seemed to elicit a greater strength of hand preference than the ground-reaching task at the individual level. Although the small sample size rules out drawing any strong conclusions concerning hand preference at the group level, our results suggest that the suspensory reaching posture might increase the expression of hand preference at the individual level. This study provides preliminary information on hand preference in captive northern white-cheeked gibbons, and will be helpful for future studies addressing the origin and evolution of hand preference in small apes.

This study aimed to examine whether the vomeronasal organ (VNO) is a prerequisite in mice to acquire essential information from various social odors and whether long-term VNO dysfunction can elicit behavioral and physiological changes in mice. We used binary choice tests and habituation–dishabituation tests to measure the abilities of male mice to recognize social odors. We found that males with the VNO ablation failed to show olfactory preferences between the odors of mate versus non-mate females, offspring versus non-offspring pups, or opposite-sex conspecifics versus predators (cats or rats), but were capable of discriminating between the two treatments in each of the paired odors, suggesting that male mice with VNO ablation might smell out the chemical differences of the two types of odors, but could not extract the biological information contained in the odors. Furthermore, prolonged VNO deficiency resulted in a reduction in crossing behavior in a light/dark box, the frequency of urine marking, and the time spent in the center in an open field. These results indicate that chronic VNO dysfunction led to anxiety-like or submissive behavior. In addition, males with VNO ablation had atrophic adrenal glands and hypertrophic preputial glands, suggesting that VNO dysfunction could damage the physiological conditions to buffer the stress and that pheromone perception deficiency might enhance self-odor production in mice.

From 1997 to 2002, a female giant panda (Ailuropoda melanoleuca) was artificially stimulated and lactation was maintained, after her neonates were removed due to the female's inability to provide maternal care. Milk samples were collected and the amount of milk collected was quantified. The lactation curve of this animal was estimated based on the Gamma function: Y(t)=at(b)e(-ct). The amount of milk collected showed significant, positive relationships with the number of days after parturition both in 1999 and in the whole study period from 1998 to 2002. This female's lactation curves fit the type I pattern of a typical mammalian lactation curve. Daily milk collection (g) during the first 30 days after parturition, and from 31 to 60 days after parturition, showed a consistent pattern with one peak at around 8:00 hr. More milk was collected during the latter period than during the former period. The amount of milk (g) collected on mucus excretion days was significantly less than that on days after mucus excretion had ended, yet no significant difference was found between milk collected one day before mucus days and on mucus days, or between milk collected one day before and one day after mucus days. Mucus excretion from the gastrointestinal tract significantly impacted the amount of milk collected. The results from this study may aid the captive propagation and conservation of giant pandas and other endangered and rare captive mammal species.

It has been documented that social isolation imparts deleterious effects on gregarious rodents species, but caging in group imparts such effects on solitary rodents. This study was attempted at examining how kinship to affect body weight, behavioral interaction, mate choice and fitness when we caged male and female rat-like hamsters Tscheskia triton in pair, a solitary species. We found that females paired with nonsibling males became heavier than the females paired with sibling males, but both agonistic and amicable behavior between paired males and females did not differ between sibling and nonsibling groups. This indicated that kinship might reduce females’ obesity in response to forced cohabitation, and dissociation might exist between physiological and behavioral responses. Furthermore, binary choice tests revealed that social familiarity between either siblings or nonsiblings decreased their investigating time spent in opposite sex conspecific of cage mates and/ or their scents as compared with those of non-mates, suggesting effects of social association on mate and kin selection of the hamsters. On the other side, both females and males caged in pair with siblings show a preference between unfamiliar siblings or their scents and the counterparts of nonsiblings after two month separation, indicating that the kin recognition of the hamsters might also rely on phenotype matching. In addition, cohabitation (or permanent presence of fathers) elicited a lower survival of pups in nonsibling pairs than sibling pairs, but did not affect litter size, suggesting that kinship affects fitness when housing male and female ratlike hamsters together. Therefore, inbreeding might be adapted for rare and endangered animals.

Chemical communication plays an important role in kin selection and mate choice in mammals. The covariance of odor-genes of rodents has been documented and kinship odor has been proposed and termed, yet little is known of the relationship between genetic relatedness and chemical composition of kinship odors. Giant pandas (Ailuropoda melanoleuca) rely substantially on chemical communication to mediate their social interactions. To examine the relationship between genetic relatedness and compounds in the urine/anogenital gland secretions, we compared the similarities between genetic relatedness and the chemical profiles of anogenital gland secretions and urine via lineage construction and GC-MS (gas chromatography and mass spectrometry). We found that information about kinship odors was present only in the urine of male adults in the mating season but absent in the non-mating season. Adult females and all sub-adults did not have such kinship odors in either mating or non-mating season. Therefore, kinship odor in the panda was contingent on age, sex, and season. This is the first report about the condition-dependent expression of kinship odor, which may have a significant implication in the practice of panda conservation in relation to chemical communication and sexual selection.

This study investigated the impact of long-term paternal presence (cohabitation) on several physiological parameters such as body weight, adrenal weight, cortisol of parents, and the survival of pups compared with brief daily encounters (isolation) of male-female pairs in golden hamsters (Mesocricetus auratus). We showed that females were affected more by cohabitation as evidenced by increased body and adrenal weights, elevated cortisol concentrations, and heavier uteri and spleens as compared with cohabiting male and isolated females. Furthermore, we found that tetradecanoic and hexadecanoic acids of the flank glands were sexually dimorphic, for which they were putative female pheromones. These two compounds were suppressed in females and elevated in males by cohabitation, suggesting that cohabitation impaired sex chemosignals. Overall, we concluded that housing females and males together had deleterious effects on adults and the survival of their pups in the golden hamster.

With a combination of dichloromethane extraction and analysis by gas chromatography-mass spectrometry (GC-MS), we found 39 compounds (corresponding to 38 GC peaks) in the anogenital gland secretion (AGS) of captive adult giant pandas, Ailuropoda melanoleuca, during the non-mating season. In addition to indole, squalene, and some of the straight-chain fatty acids that had been characterized previously from the AGS of giant pandas, we identified several new compounds such as decenal, two isomers of decadienal, phenylacetic acid, 5-methylhydantoin, hydroquinone, phenylpropanoic acid, and erucic acid. Quantitative comparison of the relative abundances of the 20 main GC peaks revealed that 5-methylhydantoin, indole, and erucic acid are putative female pheromones, whereas squalene and hydroquinone are putative male pheromones. In addition to the presence of a few individual-specific compounds, the relative abundances of most of the 21 constituents varied more among individuals than within individuals. This suggests that individual identity might be coded in both digital and analog form. The chemical composition of different AGS samples from the same pandas consistently displayed a minimum cluster distance, much smaller than that between samples from different individuals in a hierarchical linkage cluster (average linkage) dendrogram. Our results indicate that the AGS might contain an "odor fingerprint." Although putative sex pheromones such as squalene and erucic acid should be assessed further by bioassay, our study suggests that synthetic chemosignals might be useful in modulating the behavior and physiology of giant pandas.

To test the hypothesis of whether anogenital gland secretions (AGS) of giant panda code for their sexual ability, we collected AGS samples of 11 male (5 adult and 6 subadult) and 14 female (7 adult and 7 subadult) captive giant pandas (Ailuropoda melanoleuca) from China Research and Conservation Center for the Giant Panda at Wolong, Sichuan, China from 1994 to 2002. The samples were analyzed by GC and GC-MS. The GC results showed that male and female pandas shared 20 constituents with equal capacity factors in the AGS. Further GC-MS analyses showed that there were a total of 95 compounds in the AGS. Nine constituents for the males and three for the females differed significantly in relative abundances between sexually active and inactive individuals, but no clear division between sexually active and inactive pandas by hierarchical cluster analyses on the relative abundances of a subset of 56 compounds. However, we were able to separate different age groups of male pandas by the constituents of AGS. 14 compounds were found significantly and negatively, and 8 compounds positively, correlated with pandas’ age by year. Our results suggest that the information contained in the AGS of panda might not code for pandas’ sexual ability but might for their age. These results will be valuable for captive breeding and conservation in the wild.

Stereotypic behavior is exhibited by a wide range of captive animals. Its association with hormones, especially elevated cortisol level and lack of naturalistic stimuli in the environment, has been little studied. This study hypothesizes that stereotypic behavior is caused by stress due to lack of appropriate, naturalistic stimuli in the environment. Using four adult pandas (Ailuropoda melanoleuca) in the Beijing Zoo from March–July in 2003, we tested the following predictions: 1) stereotypic behavior and fecal cortisol level will not change associated with the progress of reproductive state; 2) there is a positive correlation between the occurrence of stereotypic behavior and fecal cortisol level; and 3) environmental enrichment by adding a naturalistic stimulus will reduce both stereotypic behavior and fecal cortisol level. We did not find any significant differences in the occurrence of stereotypic behavior and fecal cortisol level but did find a significant difference in the total time engaged in displaying the stereotypic behavior among the three estrous periods. The level of stereotypic behavior was correlated with elevated fecal cortisol level. Enrichment simply by adding a naturalistic stimulus did not show significant effects on stereotypic behavior, or on fecal cortisol level. Our results supported the second prediction, but did not completely support the first and the third ones perhaps because of the small sample size. Anogenital gland secretions play a major role in chemical communication by giant pandas, Ailuropoda melanoleuca (David, 1869). We analyzed 45 samples of anogenital gland secretions collected from 24 captive pandas (5 male adults, 6 female adults, 6 male subadults, and 7 female subadults) by gas chromatography and mass spectrometry. The secretions contained over 95 compounds. Based on 56 common compounds (relative abundances >0.1%) shared by more than three individuals, we identified steroids, long-chain fatty acids, fatty-acid esters, aldehydes, alkanes, alkenes, amines, terpenes, and furans. The chemical composition of each secretion was individual-specific. Although none of these individual compounds was age- or sex-specific, the relative abundances of several compounds differed between males and females and between adults and subadults. This result shows that information about sex and age could be coded in analog form. Information about age but not gender could also be digitally coded by the presence or absence of some of the 56 compounds, in addition to the analog coding.

The behavior of giant pandas (Ailuropoda melanoleuca) was studied under captive conditions. Both male and female pandas spent similar amounts of time engaged in eating and locomotion. Males performed anogenital-marking more but rested less than females, which suggests a sexually dimorphic pattern of behavior. Furthermore, females housed in the seminatural environment spent significantly less time engaged in stereotyped behavior than did females housed in the traditional enclosure, indicating that an enclosure environment affects the behavior of giant pandas. Parker's sperm competition model predicts a negative relationship between pre-copulatory (social status) and post-copulatory (sperm quality and quantity) sexually selected traits, however, empirical studies have revealed considerable inconsistency in this relationship. We hypothesized that there was a trade-off between pre- and post-copulatory sexually selected traits, and hormones (corticosterone, CORT; testosterone, T) orchestrate this relationship. In this study, we measured energetic parameters in the dominant–subordinate Mongolian gerbils (Meriones unguiculatus), whose relationship was established under chronic social encounters in a neutral arena, and then tested the relationship between their social status and sperm quality and quantity. Our results showed that dominant males initiated attack sooner and displayed more aggression, self-grooming and locomotion behaviors in daily social encounters across seven consecutive days. Dominant gerbils also had more and better quality of sperm than that of subordinate males, yet showed no significant differences in energy intake and RMR in comparison with subordinate individuals. In addition, dominant males had higher concentrations of serum T than subordinate males, whereas the concentrations of CORT showed a reverse pattern. The frequency and duration of aggression (indicative of social status) increased with elevated T concentrations. Sperm quality in terms of number and activity were associated with higher concentrations of serum T in dominant gerbils, whereas small sperm counts and poor-quality sperm were associated with relatively higher concentrations of serum CORT in subordinate gerbils. Together, our data indicated that there was no trade-off between pre- and post-copulatory sexually selected traits but hormones orchestrated the relationship between these traits in male Mongolian gerbils.

Mirror self-recognition (MSR), i.e., the ability to recognize oneself in a mirror, is considered a potential index of self-recognition and the foundation of individual development. A wealth of literature on MSR is available for social animals, such as chimpanzees, Asian elephants and dolphins, yet little is known about MSR in solitary mammalian species. We aimed to evaluate whether the giant panda can recognize itself in the mirror, and whether this capacity varies with age. Thirty-four captive giant pandas (F:M = 18:16; juveniles, sub-adults and adults) were subjected to four mirror tests: covered mirror tests, open mirror tests, water mark control tests, and mark tests. The results showed that, though adult, sub-adult and juvenile pandas exposed to mirrors spent similar amounts of time in social mirror-directed behaviors (χ(2) = 0.719, P = 0.698), none of them used the mirror to touch the mark on their head, a self-directed behavior suggesting MSR. Individuals of all age groups initially displayed attacking, threatening, foot scraping and backwards walking behaviors when exposed to their self-images in the mirror. Our data indicate that, regardless of age, the giant pandas did not recognize their self-image in the mirror, but instead considered the image to be a conspecific. Our results add to the available information on mirror self-recognition in large mammals, provide new information on a solitary species, and will be useful for enclosure design and captive animal management.

Most studies support the viewpoint that the vomeronasal organ has a profound effect on conspecific odor recognition, scent marking and mating behavior in the golden hamster (Mesocricetus auratus). However, the role of the vomeronasal organ in social odor recognition, social interaction and fitness is not well understood. Therefore, we conducted a series of behavioral and physiological tests to examine the referred points in golden hamster. We found that male hamsters with vomeronasal organ lesion showed no preference between a predator odor (the anal gland secretion of the Siberian weasels (Mustela sibirica) and putative female pheromone components (myristic acid and palmitic acid), but were still able to discriminate between these 2 kinds of odors. In behavioral tests of anxiety, we found that vomeronasal organ removal causes female hamsters to spend much less time in center grids and to cross fewer center grids and males to make fewer crossings between light and dark boxes than sham-operated controls. This indicates that a chronic vomeronasal organ lesion induced anxious responses in females. In aggressive behavioral tests, we found that a chronic vomeronasal organ lesion decreased agonistic behavior in female hamsters but not in males. The pup growth and litter size show no differences between the 2 groups. All together, our data suggested that vomeronasal organ ablation disrupted the olfactory recognition of social chemosignals in males, and induced anxiety-like and aggressive behavior changes in females. However, a vomeronasal organ lesion did not affect the reproductive capacity and fitness of hamsters. Our studies may have important implications concerning the role of the vomeronasal organ in golden hamsters and also in rodents.

Environmental regionalization (ER) is the basis of environmental assessment, prediction, planning and decision-making. ER techniques were applied to the management of Township and Village Enterprises (TVEs) in China. TVEs are generally middle and small-scale enterprises widely distributed in rural areas with rapid development and high discharge rates of pollutants, which impact heavily on the environment. A set of 13 indicators was chosen to evaluate TVEs in China: (1) runoff depth (P = 75%); (2) rainfall; (3) atmospheric pollution coefficient; (4) percent of forest coverage; (5) national income per capita; (6) economic density of TVEs; (7) population density; (8) environmental management staff at county level; (9) cultivated area per capita; (10) technical advancement of TVEs; (11) environmental impact of TVE industrial structure; (12) ratio of waste water to runoff; and (13) coal consumption density. Fuzzy cluster analysis was employed to conduct quantitative classification. Four zones and seven subzones were obtained. The classification system was adapted to the requirements of environmental management for TVEs at national level.

Rufous-breasted Accentor (Prunella strophiata) is a small-sized bunting with an extremely geographical range in the world. Here, the complete mitochondrial genome of P. strophiata (16,830 bp in length) has been analyzed for building the database. Similar to the typical mtDNA of vertebrates, it contained 37 genes (13 protein-coding genes, two rRNA genes and 22 tRNA genes) and a non-coding region (D-loop). Overall base composition of the complete mitochondrial DNA is A (30.3%), G (14.7%), C (31.0%) and T (24.0%), the percentage of A and T (54.3%) is higher than G and C (45.7%). All the genes in P. strophiata were distributed on the H-strand, except for the ND6 subunit gene and 9 tRNA genes which were encoded on the L-strand. The phylogenetic relationships of 12 Passeriformes species were reconstructed based on the complete mtDNA sequences using the Bayesian inference method.

Males will alter their mating behavior to cope with the presence of their competitors. Even exposure to odors from potential competitors can greatly increase male ejaculate expenditure in a variety of animals including insects, fishes, birds and rodents. Major efforts have been made to examine males' plastic responses to sperm competition and its fitness benefits. However, the effects of competitor absence on male's sexual motivation and behaviors remain unclear, which has been proposed to be one of the causes for the poor sexual performance of some captive mammals. This study revealed that sexual motivation can be greatly enhanced in captive male giant pandas (Ailuropoda melanoleuca) by exposure to chemosensory cues from either one or three conspecifics males. It had been shown that potential rivals' odors increased males' chemosensory investigation behavior, as well as their observing, following and sniffing behaviors towards estrous females. Behaviors changed regardless of the number of rivals (one or three). Our results demonstrate the effects of potential competition on male giant pandas' sexual motivation and behavioral coping strategy. We anticipate that our research will provide a fresh insight into the mechanisms underlying poor sexual performance in male captive mammals, and valuable information for the practical management and ex situ conservation of endangered species.

The scent of a novel male can elicit pregnancy block in recently mated female mice (Mus musculus), a phenomenon known as the Bruce effect. Despite abundant literature on the Bruce effect in rodents, it remains unclear whether males related to a female's original mate can induce the Bruce effect in out-bred, communally living mice. We investigated this question using Kunming (KM) male mice of varying genetic relatedness. Recently mated females were subjected to three treatments: exposure to the urine of the mate, urine of the mate's male littermate, and urine of a male unrelated to the mate. It was found that the urine of male littermates of the females' mates did not elicit more pregnancy block than that of the females' mates. However, the urine of novel males caused a higher rate of female miscarriage than that of the females' mates. By using a habituation-dishabituation paradigm, we found that unmated females could discriminate the urine scents of two male littermates from those of a novel male unrelated to the littermates. To understand how females use urinary cues to discriminate between males with different genetic relationships, we used gas chromatography coupled with mass spectrometry (GC-MS) to examine the volatile composition of urine from males with varying relatedness. It was found that KM male littermates shared similar volatile compositions in their urine. Our results suggest that male kinship reduces the Bruce effect in female KM mice, and provide additional evidence for mate choice being partly mediated by the Bruce effect in KM mice.

The urinary volatile compounds of captive giant pandas, Ailuropoda melanoleuca, were analysed with GC-MS (gas chromatography and mass spectrometry). Samples were collected from adult males and females during the mating season, as well as from adult and sub-adult males during the non-mating season. We detected more than 96 compounds, 50 of which were common to all individuals. The major volatile constituents were ketones, phenols, alcohols, and fatty acids. No single compound was found to be specific for characterising sex, age, and season. MANOVA analysis on the relative abundance of compounds showed that adult males and females differ in four urinary compounds in the mating season and adult males differ in seven compounds between the mating and the non-mating seasons. We also found significant age effects on the relative abundance of urinary compounds in males in the non-mating season. This result suggests that those compounds might be the potential chemosignals signalling information about male pandas’ age, sex, and reproductive state.
