5. Aged brain and neuroimmune responses to COVID-19: post-acute sequelae and modulatory effects of behavioral and nutritional interventions

Abstract

Advanced age is one of the significant risk determinants for coronavirus disease 2019 (COVID-19)-related mortality and for long COVID complications. The contributing factors may include the age-related dynamical remodeling of the immune system, known as immunosenescence and chronic low-grade systemic inflammation. Both of these factors may induce an inflammatory milieu in the aged brain and drive the changes in the microenvironment of neurons and microglia, which are characterized by a general condition of chronic inflammation, so-called neuroinflammation. Emerging evidence reveals that the immune privilege in the aging brain may be compromised. Resident brain cells, such as astrocytes, neurons, oligodendrocytes and microglia, but also infiltrating immune cells, such as monocytes, T cells and macrophages participate in the complex intercellular networks and multiple reciprocal interactions. Especially changes in microglia playing a regulatory role in inflammation, contribute to disturbing of the brain homeostasis and to impairments of the neuroimmune responses. Neuroinflammation may trigger structural damage, diminish regeneration, induce neuronal cell death, modulate synaptic remodeling and in this manner negatively interfere with the brain functions.In this review article, we give insights into neuroimmune interactions in the aged brain and highlight the impact of COVID-19 on the functional systems already modulated by immunosenescence and neuroinflammation. We discuss the potential ways of these interactions with severe acute respiratory syndrome coronavirus 2 (SARS-CoV-2) and review proposed neuroimmune mechanisms and biological factors that may contribute to the development of persisting long COVID conditions. We summarize the potential mechanisms responsible for long COVID, including inflammation, autoimmunity, direct virus-mediated cytotoxicity, hypercoagulation, mitochondrial failure, dysbiosis, and the reactivation of other persisting viruses, such as the Cytomegalovirus (CMV). Finally, we discuss the effects of various interventional options that can decrease the propagation of biological, physiological, and psychosocial stressors that are responsible for neuroimmune activation and which may inhibit the triggering of unbalanced inflammatory responses. We highlight the modulatory effects of bioactive nutritional compounds along with the multimodal benefits of behavioral interventions and moderate exercise, which can be applied as postinfectious interventions in order to improve brain health.

Abstract

This paper is devoted to a brief presentation of the main results obtained within the FREEDAM research project. The project has been completed and, recently, also the dissemination of the results has reached the finish line. The project was devoted to the development of low-damage beam-to-column connections based on the use of friction dampers conceived to substitute the traditional dissipative zones of moment-resisting frames, i.e. the beam ends. The conception of these innovative connections is presented and the experimental tests carried out at the University of Salerno during the FREEDAM project are summarized dealing with the friction dampers, the beam-to-column joints sub-assemblages and the seismic simulation of a real scale one-bay two-storey building with the pseudo-dynamic testing method. Finally, the design procedures for seismic-resistant steel frames equipped with such connections are presented in the framework of Eurocode 8 and according to TPMC., Freedam Plus RFCS Project

11. Stereo bearings-only tracking of a wheelchair from a robot

Abstract

This project deals with technical assistance for people of reduced mobility. The goal of this work is to have a mobile platform with an embedded prehensive arm (MANUS®) track a wheelchair. The use of this mobile unit in relation to the patient’s wheelchair is carried out on a master-slave basis. This study, therefore, has a plural-disciplinary nature: Science for the Engineer, Human, Social and Clinical Sciences. To ensure the tracking of the wheelchair by the mobile platform, we applied a stereo bearings-only tracking (BOT) paradigm to a single moving target. The observer, the mobile platform (robot), is equipped with two omnidirectional vision sensors, that each provides output with which we compute two bearing angles. Having two bearing angles makes it possible to track a manoeuvring target. In this paper we will show how we take advantage of the multi-sensor data fusion to improve the BOT process and hence obtain a more reliable wheelchair tracking.

Abstract

“Successful aging” - defined as long life in a good mental and physical health, requires appropriate immune function. However, dysregulated immunity in later life, known as “immunosenescence,” results in compromised immune functions and disturbed pro- and anti-inflammatory balance. The impact of these age-related immune alterations on physical and mental health, and hence successful aging, is currently not well understood. With few exceptions, the studies on aging have not attempted to integrate psycho-neurological, metabolic, and endocrine parameters with immune function in order to dissect out which influences which. Few have taken CMV-infection into account and even gender has not always been considered. The present dissertation contributes to this developing research field both conceptually and empirically. It is publication-oriented and consists of four publications. Publication I presents data obtained from a subgroup of the Berliner Aging Study participants, which show significant age-associated differences of T-cell subset distribution. The modulatory impact of both sex and CMV-infection on T-cell naïve and memory phenotypes, but unaffected frequencies of T-stem cell-like memory cells, were found. For the first time, the frequency of the TSCM phenotype and of PD-1+ T-cells in peripheral circulation has been investigated, and the effects of age, CMV-serostatus, and sex on their frequency have been examined. Publication II reviews the relevant literature on the dynamic neuroimmune interactions between the immune and the nervous systems. Bringing these two strands of research together, we propose that immunosenescence and peripheral low-grade inflammation at least partly contribute to neuroinflammation inducing neurodegeneration and age-related cognitive impairments. We review and discuss possible interventions that can prevent or at least postpone these age-related changes. Publication III characterizes the baseline inflammatory status of aged individuals, recruited to undergo the cognitive, physical, and combined training interventions. By analyzing multiple circulating peripheral biomarkers and measures of objective cognitive function, we show that both CMV-serostatus and sex may modulate inflammatory immune factors, cognitive performance, and the relationship between the two domains, and should therefore be considered in comparative and interventional studies with elderly people. In study IV, we propose a new strategy that allows the quantitative investigation of multiple interactions between different cytokines, receptor molecules, metabolic and neurotrophic factors, hormones, immune cells, and measures of cognitive performance. Using a graph-theoretical approach enables us not only to visualize biologically meaningful interconnections between different variables but also to compare the network topology dynamics between different groups of CMV-seronegative and -seropositive men and women in a statistically sound manner. In summary, results obtained from the studies III and IV suggest that highly integrated and segregated networks have optimal neuroimmune interactions. Taken together, this dissertation contributes to the study of age-related functional alterations of immune and related physiological functions in two major ways. First, it expands the analysis methods that have been used to investigate markers of immunosenescence. Second, it has generated new findings on the modulatory effects of CMV-latency and sex on multiple peripheral biomarkers and cognitive function in older men and women.

Abstract

Temporomandibular disorders (TMD) are an heterogeneous group of disorders affecting temporomandibular joints (TMJ), masticatory muscles, or both. The aim of this study is to evaluate the relationship between general health and psychological distress variables and the clinical diagnosis of TMD in children between 8 - 13 years old. Moreover, it will be underlined the different perception of psychological distress existing between patients and their parents.Patients were divided in two groups: the "case group", which includes patients with TMD, and the "control group", which included patients asymptomatic for TMD. The "case group" is composed by 29 patients, while the "control group" is composed by 22 patients. The patients were asked to fill in a questionnaire called Child Health Questionnaire (CHQ), to evaluate general health and psychological condition and, eventually, the presence of symptoms of stress, anxiety and/or depression. The CHQ is 87 - item questionnaire developed in USA specifically for children.As widely reported during the present work, the link between TMD and psychological distress is existing and quite strong, because of lack of number of patients many aspects worth of attentions have not been analyzed as they deserved.That's way the conclusion must lead to the proposal of going on with the present study following, possibly, some aspects of research as an increase of the number of patients involved in the study; Individuate possible elements, internal and external, that could strongly influence differences between patient's results and parent's results, etc.

Abstract

Background: Albuminuria has been classically considered a marker of kidney damage progression in diabetic patients and it is routinely assessed to monitor kidney function. However, the role of a mild GFR reduction on the development of stage ≥3 CKD has been less explored in type 1 diabetes mellitus (T1DM) patients. Aim of the present study was to evaluate the prognostic role of kidney disease measures, namely albuminuria and reduced GFR, on the development of stage ≥3 CKD in a large cohort of patients affected by T1DM. Methods: A total of 4284 patients affected by T1DM followed-up at 76 diabetes centers participating to the Italian Association of Clinical Diabetologists (Associazione Medici Diabetologi, AMD) initiative constitutes the study population. Urinary albumin excretion (ACR) and estimated GFR (eGFR) were retrieved and analyzed. The incidence of stage ≥3 CKD (eGFR < 60 mL/min/1.73 m2) or eGFR reduction > 30% from baseline was evaluated. Results: The mean estimated GFR was 98 ± 17 mL/min/1.73m2 and the proportion of patients with albuminuria was 15.3% (n = 654) at baseline. About 8% (n = 337) of patients developed one of the two renal endpoints during the 4-year follow-up period. Age, albuminuria (micro or macro) and baseline eGFR < 90 ml/min/m2 were independent risk factors for stage ≥3 CKD and renal function worsening. When compared to patients with eGFR > 90 ml/min/1.73m2 and normoalbuminuria, those with albuminuria at baseline had a 1.69 greater risk of reaching stage 3 CKD, while patients with mild eGFR reduction (i.e. eGFR between 90 and 60 mL/min/1.73 m2) show a 3.81 greater risk that rose to 8.24 for those patients with albuminuria and mild eGFR reduction at baseline. Conclusions: Albuminuria and eGFR reduction represent independent risk factors for incident stage ≥3 CKD in T1DM patients. The simultaneous occurrence of reduced eGFR and albuminuria have a synergistic effect on renal function worsening.

Abstract

Background Albuminuria has been classically considered a marker of kidney damage progression in diabetic patients and it is routinely assessed to monitor kidney function. However, the role of a mild GFR reduction on the development of stage ≥3 CKD has been less explored in type 1 diabetes mellitus (T1DM) patients. Aim of the present study was to evaluate the prognostic role of kidney disease measures, namely albuminuria and reduced GFR, on the development of stage ≥3 CKD in a large cohort of patients affected by T1DM. Methods A total of 4284 patients affected by T1DM followed-up at 76 diabetes centers participating to the Italian Association of Clinical Diabetologists (Associazione Medici Diabetologi, AMD) initiative constitutes the study population. Urinary albumin excretion (ACR) and estimated GFR (eGFR) were retrieved and analyzed. The incidence of stage ≥3 CKD (eGFR 30% from baseline was evaluated. Results The mean estimated GFR was 98 ± 17 mL/min/1.73m2 and the proportion of patients with albuminuria was 15.3% (n = 654) at baseline. About 8% (n = 337) of patients developed one of the two renal endpoints during the 4-year follow-up period. Age, albuminuria (micro or macro) and baseline eGFR 90 ml/min/1.73m2 and normoalbuminuria, those with albuminuria at baseline had a 1.69 greater risk of reaching stage 3 CKD, while patients with mild eGFR reduction (i.e. eGFR between 90 and 60 mL/min/1.73 m2) show a 3.81 greater risk that rose to 8.24 for those patients with albuminuria and mild eGFR reduction at baseline. Conclusions Albuminuria and eGFR reduction represent independent risk factors for incident stage ≥3 CKD in T1DM patients. The simultaneous occurrence of reduced eGFR and albuminuria have a synergistic effect on renal function worsening. Abstract

Thanks to a coordinated funding by the European Space Agency (ESA) and the European Commission (EC) within the 7th framework program, the High-Speed Experimental Fly Vehicles – International (HEXAFLY-INT) project is aimed at the flight validation of hypersonics technologies enabling future trans-atmospheric flights. The project, which is currently involving partners from Europe, Russian Federation and Australia operating under ESA/ESTEC coordination, will achieve the goal of designing, manufacturing, assembling and flight testing an unpowered high speed vehicle in a glider configuration by 2018. The main technical challenges of the project are specifically related to the design of the vehicle gliding configuration and to the complexity of integrating breakthrough technologies with standard aeronautical technologies, e.g. high temperature protection system and airframe cold structures. Also, the sonic boom impact, which is one of the environmental challenges of the high speed flight, will be assessed. Abstract

Introduction. Temporomandibular disorders (TMD) are an heterogeneous group of disorders affecting temporomandibular joints (TMJ), masticatory muscles, or both. The aim of this study is to evaluate the relationship between general health and psychological distress variables and the clinical diagnosis of TMD in children between 8 - 13 years old. Moreover, it will be underlined the different perception of psychological distress existing between patients and their parents. Materials & Methods. Patients were divided in two groups: the "case group", which includes patients with TMD, and the "control group", which included patients asymptomatic for TMD. The "case group" is composed by 29 patients, while the "control group" is composed by 22 patients. The patients were asked to fill in a questionnaire called Child Health Questionnaire (CHQ), to evaluate general health and psychological condition and, eventually, the presence of symptoms of stress, anxiety and/or depression. The CHQ is 87 - item questionnaire developed in USA specifically for children. Results. As widely reported during the present work, the link between TMD and psychological distress is existing and quite strong, because of lack of number of patients many aspects worth of attentions have not been analyzed as they deserved. Conclusions: That's way the conclusion must lead to the proposal of going on with the present study following, possibly, some aspects of research as an increase of the number of patients involved in the study; Individuate possible elements, internal and external, that could strongly influence differences between patient's results and parent's results, etc. [ABSTRACT FROM AUTHOR]

Abstract

An advanced LES code, named SPARK-LES, is currently under development at CIRA within the frame of the HYPROB program. SPARK-LES is a structured, multiblock finite-volume code that solves the compressible Navier-Stokes equations with the final goal of performing accurate simulations of high-pressure combustion with suitable numerical/physical models. In this paper, the key features of the computational core are described. Centered high-order explicit and compact schemes for space integration are implemented; different solutions to improve numerical stability and robustness are analyzed and discussed. The capabilities of the code to reproduce the essential features of turbulent dynamics are proved by simulating the well-known Taylor-Green vortex problem.

As a fundamental step towards the development of a civil high-speed transport concept, the European Community (EC) co-funded HEXAFLY-INT international project (with partners from Europe, Russian Federation, Brazil and Australia), has the aim to design, manufacture and test in flight an innovative gliding hypersonic vehicle, based on the configuration developed in previous EC co-funded projects LAPCAT I, II and HEXAFLY. The flight experiment is focused on a self-controlled glider configuration characterized by high aerodynamic efficiency and equipped with several breakthrough technologies on-board, in order to provide valuable aerodynamic and aero-structural flight data to validate methods and technologies used to design the hypersonic vehicle. In this framework, this paper provides a detailed overview on research efforts carried out in assessing the aerodynamic performance of the expendable Experimental Flight Test Vehicle under development in the programme. Copyright © 2017 ASME. Over the last years, innovative concepts of civil high-speed transportation vehicles were proposed. In this framework, the Hexafly-INT project intends to test in free-flight conditions an innovative gliding vehicle with several breakthrough technologies on-board. This approach will help to gradually increase the readiness level of a consistent number of technologies suitable for hypervelocity flying systems. The vehicle design, manufacturing, assembly and verification is the main driver and challenge in this project. The prime objectives of this free-flying high-speed cruise vehicle shall aim at a conceptual design demonstrating a high aerodynamic efficiency in combination with high internal volume; controlled level flight at a cruise Mach number of 7 to 8;an optimal use of advanced high-temperature materials and structures. Present research describes the aero-thermal design process of the Experimental Flight Test Vehicle, namely EFTV. The glider aeroshape design makes maximum use of databases, expertise, technologies and materials elaborated in previously European community co-funded projects LAPCAT I & II [1][2], ATLLAS I & II [3][4] and HEXAFLY [5]. Abstract

Fashion is an important tool of knowledge of the intangible values that define the cultural profile of the places and, therefore, it is a theme of search useful to delineate the creative and experimental identity of the ample "Baia di Naples", beginning from the analysis of the activity developed by its protagonists in the most prolific season of the “Made in Italy”. In the history of Italian fashion, Naples and its “mythical” area have played a leading role, thanks to the work of some outstanding protagonists. This has been possible, not so much through the creative activity of those who left their hometown and moved to the capital city seeking fame, but rather thanks to those (Clarette Gallotti, Livio De Simone, Mario Valentino, Fausto Sarli and many other tailors and creative engaged in resort fashion), who have invested all their resources in the territories that they come from, contributing greatly to the consolidation of the role of the whole city of Naples in the larger frame of Italian fashion. Their success begins from the early of the Fifties, when the fashion, together with the tourism, it becomes an important tool to reviving Italy from the difficulties left by the war.

Abstract

E' ancora possibile nel mondo contemporaneo 'ritrarre', rappresentare, i drammi della storia? Il saggio cerca di dimostrare quanto nel panorama attuale l'arte tenda a 'presentare' il problema piuttosto che rappresentarlo dal momento che il 'conflitto' nell'arte contemporanea sembra una componente intrinseca a molte manifestazioni artistiche attuali. Prendendo spunto dal palindromo (RAW WAR) di Bruce Nauman del 1970, il saggio attraversa la storia dell'arte contemporanea internazionale, a partire dal secondo dopoguerra fino all'epoca attuale, focalizzando l'attenzione su alcuni casi di studio. Molte produzioni artistiche recenti affrontano il tema scegliendo quella sorta di 'zona grigia' che separa, ma anche unisce, le vittime e i carnefici, che problematizza la relazione fra 'buoni' e 'cattivi' e denuncia la 'guerra' ponendo in una luce diversa le parti in conflitto, al di là della contrapposizione col 'nemico'.

Abstract

There are two main challenges for law and policy to foster a sustainable development of agri-food systems through regulation. The first challenge is that the regulation of a certain aspect often does not lead to the intended outcomes. Regulative measures can only perturb, disturb or irritate agroecosystems because they are self-organising and autopoietic systems. All regulation of autopoietic systems depends on self-regulation because it is the ability of the agroecosystem to observe the disturbance and its internal schema of logic that will define the reaction. The second challenge is that regulation of one aspect often leads to unforeseen and unwanted side effects regarding other aspects. These unintended effects call for more regulations to deal with them, leading to a paradoxical situation of an increasingly growing web of regulation and effects, a situation that is concretely reflected in the exponential growth rate of the amount of positive law on agriculture and environment. These challenges are amplified by the increasing complexity created by specialisations in science, law and farming practice, a complexity that cannot be dealt with by further specialisation. In this chapter, we argue, based on social systems theory, that there is a need for a second-order platform of agroecological regulation where different scientific and law perspectives can meet and communicate about sustainable development and regulation of agroecosystems. But it requires that each perspective acknowledges its own blind spots and acknowledges that the agroecosystem can be seen from many other perspectives.
