Abstract

Item does not contain fulltext, Aberrations in nuclear size and shape are commonly used to identify cancerous tissue. However, it remains unclear whether the disturbed nuclear structure directly contributes to the cancer pathology or is merely a consequence of other events occurring during tumorigenesis. Here, we show that highly invasive and proliferative breast cancer cells frequently exhibit Akt-driven lower expression of the nuclear envelope proteins lamin A/C, leading to increased nuclear deformability that permits enhanced cell migration through confined environments that mimic interstitial spaces encountered during metastasis. Importantly, increasing lamin A/C expression in highly invasive breast cancer cells reflected gene expression changes characteristic of human breast tumors with higher LMNA expression, and specifically affected pathways related to cell-ECM interactions, cell metabolism, and PI3K/Akt signaling. Further supporting an important role of lamins in breast cancer metastasis, analysis of lamin levels in human breast tumors revealed a significant association between lower lamin A levels, Akt signaling, and decreased disease-free survival. These findings suggest that downregulation of lamin A/C in breast cancer cells may influence both cellular physical properties and biochemical signaling to promote metastatic progression.

20. Growth and production of Enterotoxin A by Staphylococcus aureus on 'home-style' French fries

Abstract

French fries produced in foodservice establishments are usually cut into strips and blanched in oil for 4 min at around 177 deg C before being held at room temperature prior to finish frying. Whilst they are being held, there is a potential for the fries to be contaminated with foodborne pathogens, such as Staphylococcus aureus. A new study investigates the growth potential of S. aureus on fresh-cut, oil-blanched French fries held at 21 deg C and 26.7 deg C. It is shown that there is a potential for S. aureus enterotoxin to occur on oil-blanched home-style French fries and it is recommended that they be finish-fried and served if stored at room temperature.

Abstract

The procedure for making 'home-style' French fries usually involves blanching prior to finish frying to cut preparation time, but it is possible for pathogens to contaminate blanched potato strips during the process. A new study investigates the growth potential of the pathogen Bacillus cereus on blanched 'home-style' French fries and determines whether the final frying process inactivates the microorganism. It is shown that oil-blanched French fry strips can support growth of B. cereus at temperatures of 21 deg C or 26.7 deg C, but growth is slowed at below 21 deg C. French fries should be fried within 3-4 hours or else discarded.

Abstract

Aim: The aim of this study was to describe factors affecting nursing education and labour markets in countries in East, Central, and Southern Africa, and critical areas for investment. Background: An understanding about the relationship between the supply of nurses (determined by types of educational programmes, and the quantity and quality of nurse graduates), and workforce demand is critical to health policy development. Methods: Six focus groups and 14 key informant interviews with nursing leaders and experts were conducted. Participants included government chief nursing officers, registrars of regulatory bodies, association leaders and heads of nursing education. The data were transcribed, coded and analysed using inductive techniques. Findings: Participants discussed challenges and strengths of nursing education, school and regulatory infrastructure, financing mechanisms for the nursing workforce, the state of nursing jobs and scope of nursing practice. Conclusion: Strengthened regulations and leadership are needed to improve investment in nursing, the quality of nursing education, and working conditions and to promote the achievement of better health outcomes. Implications for Nursing Policy: Clarifying scope of practice for nurses in the health sector and creating competency‐based requirements is important. Governments should establish positions that align with updated competencies and provide fair and safe working conditions. Abstract

Excited states in the neutron-deficient nuclides W8874162 and W9074164 were investigated by using the ?-ray spectrometer Jurogam. A change in structure is apparent from the first rotational alignments in W162 and W164, whose rotationally aligned bands are interpreted as ?(h9/2)2 and ?(i13/2)2 configurations, respectively. The level schemes have been extended using recoil (-decay) correlations with the observation of excited collective structures. Configuration assignments have been made on the basis of comparisons of the deduced aligned angular momentum, as a function of rotational frequency, with the predictions of the cranked shell model. © 2016 American Physical Society.

Abstract

Excited states in Re186 with spins up to J=12 were investigated in two separate experiments using W186(d,2n) reactions at beam energies of 12.5 and 14.5 MeV. Two- and threefold γ-ray coincidence data were collected using the CAESAR and CAGRA spectrometers, respectively, each composed of Compton-suppressed high-purity germanium detectors. Analysis of the data revealed rotational bands built on several two-quasiparticle intrinsic states, including a long-lived Kπ=(8+) isomer. Configuration assignments were supported by an analysis of in-band properties, such as gK-gR values. The excitation energies of the observed intrinsic states were compared with results from multi-quasiparticle blocking calculations, based on the Lipkin-Nogami pairing approach, that included contributions from the residual proton-neutron interactions.

Abstract

A study was conducted to obtain a bacteriological profile of fresh, frozen and spoiled white amur flesh as well as the storage conditions affecting the microbiological quality of these species. Acinetobacter, Citrobacter, Enterobacter, Escherichia, Micrococcus, Moraxella, Pseudomonas, Staphylococcus and Streptococcus were identified in the samples. It was shown that 200 micrograms/ml sodium hypochlorite dip and four degrees centigrade storage temperature are effective in controlling the coliforms.

Abstract

Six child care centers are surveyed to assess the microbiological quality of three food contact surfaces and one non-food contact surface to determine the effectiveness of cleaning and sanitization procedures within the facilities. The results from the survey studies demonstrated that the extent of bacterial contamination is dependent on the center, time of day, and the area sampled and have suggested the need to develop modified standard sanitation operating procedures to reduce potential hazards.

Abstract

The effect of ultimate pH (pHu) of pork on shelf life based on microbial growth, drip loss (WHC) and oxidative rancidity in vacuum-packaged loins stored at 4 degree centigrade is evaluated. The results have shown that ideal pork quality is achieved with a pHu between 5.6 and 5.9 and a further increase of pH might improve WHC but might concurrently reduce microbiological shelf life.

Contains fulltext : 171070.pdf (publisher's version ) (Closed access), During cancer metastasis, tumor cells penetrate tissues through tight interstitial spaces, which requires extensive deformation of the cell and its nucleus. Here, we investigated mammalian tumor cell migration in confining microenvironments in vitro and in vivo. Nuclear deformation caused localized loss of nuclear envelope (NE) integrity, which led to the uncontrolled exchange of nucleo-cytoplasmic content, herniation of chromatin across the NE, and DNA damage. The incidence of NE rupture increased with cell confinement and with depletion of nuclear lamins, NE proteins that structurally support the nucleus. Cells restored NE integrity using components of the endosomal sorting complexes required for transport III (ESCRT III) machinery. Our findings indicate that cell migration incurs substantial physical stress on the NE and its content and requires efficient NE and DNA damage repair for cell survival.

42. Essential Oil Emulsions as Postharvest Sanitizers To Mitigate SalmonellaCross-Contamination on Peppers

Abstract

Alternative postharvest sanitizers to chlorine are of increasing interest for many organic growers and consumers. An emulsion of clove bud oil (CBO; 0.2 and 0.5%) or thyme oil (0.2 and 0.5%) was evaluated as a sanitizer for produce washing against a five-serovar cocktail of Salmonellaon snacking peppers and compared for antimicrobial efficacy with sodium hypochlorite (200 ppm). To further evaluate these compounds, the sanitation efficacy of an emulsion was examined after the addition of 1% organic load (OL). Emulsion treatments at 0.2 and 0.5% thyme oil and 0.5% CBO were the least effected by OL and effectively reduced cross-contamination of Salmonellaon clean peppers, in many cases to below the limit of detection (1 CFU/10 g; P< 0.05). Chlorine and 0.2% CBO were rendered ineffective by the addition of OL in preventing cross-contamination and performed similarly to the water control. For surface-inoculated peppers, none of the evaluated treatments performed better than a water-only wash. The antimicrobial efficacy of the essential oil emulsions in the presence of OL indicates these emulsions may be suitable replacements for chlorine in postharvest produce wash systems.

Abstract

The National Heart Foundation of Australia assembled an expert panel to provide guidance on policy and system changes to improve the quality of care for people with chronic heart failure (CHF). The recommendations have the potential to reduce emergency presentations, hospitalisations and premature death among patients with CHF. Best-practice management of CHF involves evidencebased, multidisciplinary, patient-centred care, which leads to better health outcomes. A CHF care model is required to achieve this. Although CHF management programs exist, ensuring access for everyone remains a challenge. This is particularly so for Aboriginal and Torres Strait Islander peoples, those from non-metropolitan areas and lower socioeconomic backgrounds, and culturally and linguistically diverse populations. Lack of data and inadequate identification of people with CHF prevents efficient patient monitoring, limiting information to improve or optimise care. This leads to ineffectiveness in measuring outcomes and evaluating the CHF care provided. Expanding current cardiac registries to include patients with CHF and developing mechanisms to promote data linkage across care transitions are essential. As the prevalence of CHF rises, the demand for multidisciplinary workforce support will increase. Workforce planning should provide access to services outside of large cities, one of the main challenges it is currently facing. To enhance community-based management of CHF, general practitioners should be empowered to lead care. Incentive arrangements should favour provision of care for Aboriginal and Torres Strait Islander peoples, those from lower socioeconomic backgrounds and rural areas, and culturally and linguistically diverse populations.?Ongoing research is vital to improving systems of care for people with CHF. Future research activity needs to ensure the translation of valuable knowledge and highquality evidence into practice.

Abstract

Matière Nucléaire NESTER; International audience; The excitation energy of the lowest-energy superdeformed band in $^{196}$Pb is established using the techniques of time-correlated $\gamma$-ray spectroscopy. Together with previous measurements on $^{192}$Pb and $^{194}$Pb, this result allows superdeformed excitation energies, binding energies, and two-proton and two-neutron separation energies to be studied systematically, providing stringent tests for current nuclear models. The results are examined for evidence of a "superdeformed shell gap."

Abstract

γ-ray spectroscopy of high-spin states of the neutron-deficient nucleus Pb189 has been conducted with the Gd158(Ar36,5n) and Er164(Si29,4n) reactions. With the first of these, detection of evaporation residues and mass gating were used to unambiguously assign a number of prompt γ-ray transitions to Pb189. With the second reaction and a pulsed beam, an isomer with a mean life of 32 μs was found. Although inconclusive, the available evidence favors identification of the isomer with the 332+ state of the ν(i13/2)-3 configuration. The levels observed below the isomer can be identified with states involving three different structures: the neutron (i13/2)-3 multiplet in the spherical well; a prolate-deformed band involving mixed i13/2 neutron orbitals; and a state with the oblate π(2p-2h)0+ν(i13/ 2)-1 configuration. The evidence for structures associated with different shapes is supported by the observation of E0 components in some of the Jπ→Jπ transitions linking them.
