Abstract Heavy traffic jams are difficult to predict due to the complexity of traffic dynamics. Understanding the network dynamics of traffic bottlenecks can help avoid critical large traffic jams and improve overall traffic conditions. Here, we develop a method to forecast heavy congestions based on their early propagation stage. Our framework follows the network propagation and dissipation of the traffic jams originated from a bottleneck emergence, growth, and its recovery and disappearance. Based on large-scale urban traffic-speed data, we find that dissipation duration of jams follows approximately power-law distributions, and typically, traffic jams dissolve nearly twice slower than their growth. Importantly, we find that the growth speed, even at the first 15 minutes of a jam, is highly correlated with the maximal size of the jam. Our methodology can be applied in urban traffic control systems to forecast heavy traffic bottlenecks and prevent them before they propagate to large network congestions.

Abstract Background Interstitial lung disease (ILD) is a frequently observed comorbidity in autoimmune diseases such as dermatomyositis/polymyositis (DM/PM), and it is significantly associated with specific autoantibody types. One unique antibody type is the anti-transcription intermediate factor-1γ antibody (anti-TIF-1γ Ab), which has a positive rate of only 7%. It is often found in combination with malignancy and rarely with ILD, particularly rapidly progressive ILD (RPILD). In some cases, the presence of ILD in individuals with DM may indicate a paraneoplastic syndrome. Pneumocystis jiroveci pneumonia (PJP) typically occurs due to intensive immunosuppressive therapy, human immunodeficiency virus (HIV) infection, or malignancy, and rarely as an isolated condition. Case presentation A 52-year-old man with a history of rapid weight loss but non-HIV infected and not immunosuppressed who presented with fever, cough, dyspnea, weakness of the extremities, characteristic rash and mechanic's hand. Pathogenic tests suggested PJP, laboratory tests suggested a single anti-TIF-1γ Ab positive DM, imaging suggested ILD, and pathology revealed no malignancy. RPILD and acute respiratory distress syndrome (ARDS) developed after anti-infection and steroid hormone therapy. After mechanical support therapy such as Extracorporeal Membrane Oxygenation (ECMO), the patient developed late-onset cytomegalovirus pneumonia (CMVP), complicated bacterial infection, and ultimately death. Additionally, we discuss the potential causes of rapid weight loss, the mechanisms by which anti-TIF-1γ Ab may lead to ILD, and the possible connection between anti-TIF-1γ Ab positivity, rapid weight loss, immune abnormalities, and opportunistic infections. Conclusions This case emphasizes the importance of early recognition of malignant tumors and pulmonary lesions, assessment of the body's immune status, prompt initiation of immunosuppressive treatment, and prevention of opportunistic infections in individuals with single anti-TIF-1γ Ab positive DM presenting with rapid weight loss.

The models of k -core percolation and interdependent networks (IN) have been extensively studied in their respective fields. A recent study has revealed that they share several common critical exponents. However, several newly discovered exponents in IN have not been explored in k -core percolation, and the origin of the similarity still remains unclear. Thus, in this paper, by considering k -core percolation on random networks, we first verify that the two newly discovered exponents (fractal fluctuation dimension, $d^{^{\prime}}_\textrm{f}$ , and correlation length exponent, $\nu^{^{\prime}}$ ) observed in d -dimensional IN spatial networks also exist with the same values in k -core percolation. That is, the fractality of the k -core giant component fluctuations is manifested by a fractal fluctuation dimension, $\widetilde d_\textrm{f} = 3/4$ , within a correlation size N ʹ that scales as $N^{^{\prime}} \propto (p-p_c)^{-\widetilde\nu}$ , with $\widetilde\nu = 2$ . Here we define, $\widetilde\nu \equiv d\cdot \nu^{^{\prime}}$ and $\widetilde{d}_\textrm{f} \equiv d^{^{\prime}}_\textrm{f}/d$ . This implies that both models, IN and k -core, feature the same scaling behaviors with the same critical exponents, further reinforcing the similarity between the two models. Furthermore, we suggest that these two models are similar since both have two types of interactions: short-range (SR) connectivity links and long-range (LR) influences. In IN the LR are the influences of dependency links while in k -core we find here that for k = 1 and k = 2 the influences are SR and in contrast for $k\unicode{x2A7E}3$ the influence is LR. In addition, analytical arguments for a universal hyper-scaling relation for the fractal fluctuation dimension of the k -core giant component and for IN as well as for any mixed-order transition are established. Our analysis enhances the comprehension of k -core percolation and supports the generalization of the concept of fractal fluctuations in mixed-order phase transitions.

In the past decades, the properties of olfactory ensheathing cells (OECs) have been widely investigated. Studies have shown that transplantation of OECs cultured from the olfactory bulb mediates axonal regeneration, remyelination and restores lost functions in experimental central nervous system (CNS) injury models. Autologously sourcing the cells from the nasal mucosa or the olfactory bulb to treat patients with spinal cord injuries would be ideal, but the cell yield achieved may be inadequate to cover the surface area of the lesions typically encountered in human spinal cord contusion injuries. Therefore, banking allogenic cryopreserved olfactory bulb cells from donors or generating cell lines could provide a marked increase in cell stock available for transplantation. This study is undertaken in two control and two intervention groups. The control groups have lesions alone and lesions with collagen gel but without cells. The intervention groups have either transplantation of primary cultured olfactory bulb OECs (bOECs) encapsulated in collagen gel or cryopreserved bulb OECs (CbOECs) encapsulated in collagen gel. Here, we report that transplantation of cryopreserved rat bOECs encapsulated in collagen restored the loss of function in a vertical climbing test in a unilateral C6-T1 dorsal root injury model. The loss of function returns in 80% of rats with injuries in about 3 weeks comparable to that we observed after transplantation of primary cultured bOECs. The regeneration axons induced by the transplant are identified by neurofilament antibodies and ensheathed by OECs. Our results indicate that cryopreserved OECs retain their properties of inducing axon regeneration and restoring loss of function in the experimental model. This is a step forward to translate the research into future clinical applications.

Abstract A mouse model with cisplatin-induced ototoxicity was used in addition to human samples from the ITMAT Biobank at the University of Pennsylvania. Mouse auditory brainstem responses (ABR), inner ear histology, perilymph cisplatin sampling, and measurement of serum prestin via ELISA were performed. Human serum prestin level was measured via ELISA in patients with otological issues after cisplatin treatment and compared to matched controls. Serum prestin was significantly elevated before ABR threshold shifts in mice exposed to cisplatin compared to control mice. Prestin concentration also correlated with the severity of hearing threshold shifts in mice. After an extended rest post-cisplatin treatment, prestin returned to baseline levels in mice and humans. Prestin was significantly elevated in the serum before the onset of objective hearing loss and correlated with the severity of hearing damage indicating that prestin may function as an effective biomarker of cisplatin-induced ototoxicity. Human serum prestin levels responded similarly to mice > 3 weeks from ototoxic exposure with decreased levels of prestin in the serum.

Chitosan coatings could effectively increase the biostability and biocompatibility of biomaterials while maintaining their structural integrity. In this study, electrospun fibrous polyhydroxybutyrate (PHB) membranes were pre-treated with potassium hydroxide (KOH) or hydrogen peroxide (H2O2) and then modified with dopamine (DA) and glutaraldehyde (GA) to improve their adhesion with chitosan (CS). Scanning electron microscopy (SEM), water contact angles (WCA), and Fourier transform infrared spectroscopy (FTIR) were used to demonstrate the successful generation of DA and GA-modified PHB fibers. KOH pre-treated PHB membranes exhibited superior binding efficiency with CS at low concentrations compared to their H2O2 pre-treated counterparts. The thermal analysis demonstrated a considerable decrease in the degradation temperature and crystallinity of KOH pre-treated membranes, with temperatures dropping from 309 °C to 265.5 °C and crystallinity reducing from 100% to 25.59% as CS concentration increased from 0 to 2 w/v%. In comparison, H2O2 pre-treated membranes experienced a mild reduction in degradation temperature, from 309 °C to 284.4 °C, and a large decrease in crystallinity from 100% to 43%. UV-vis analysis using Cibacron Brilliant Red 3B-A dye (CBR) indicated similar binding efficiencies at low CS concentrations for both pre-treatments, but decreased stability at higher concentrations for KOH pre-treated membranes. Mechanical testing revealed a considerable increase in Young’s modulus (2 to 14%), toughness (31 to 60%), and ultimate tensile stress (UTS) (14 to 63%) for KOH-treated membranes compared with H2O2 pre-treated membranes as CS concentration increased from 0 to 2 w/v%.

In order to investigate the turbulent lubrication performance of a textured hydrodynamic journal bearing (THJB), a model of turbulent lubrication was established in this paper. The variations in the Reynolds number, oil film thickness, oil film pressure, bearing capacity, attitude angle, and side leakage flow with structural and working parameters were studied, and the axis whirl orbit was further analyzed. The results show that turbulent lubrication is suitable for the actual operating conditions of THJBs. The Reynolds number decreases with the eccentricity ratio in the pressure-bearing zone but increases with rotational speeds, whereas the variation in the maximum oil film pressure increases and the minimum oil film thickness decreases with the eccentricity ratio under various Reynolds numbers. The bearing capacity decreases with the dimple diameter, depth, oil film thickness, and clearance ratio but increases with the length/diameter ratio and dimple spacing. As the eccentricity ratio increases, the attitude angle decreases, but the side leakage flow increases. In addition, the system tends to be unstable as the rotational speed and length/diameter ratio increase, and the friction and wear on the surface are three-body friction. This work not only helps in analyzing the characteristics of a THJB under actual operating conditions but also provides support for research on the simulation of THJB’s lubrication mechanism of THJB via computational fluid dynamics.

Abstract Once the Internet of Things was proposed, it has received great attention from all walks of life and has become one of the top ten technologies that change the world. Therefore, more and more people are engaged in the research of the Internet of Things, after the unremitting efforts of all seniors. Now the Internet of Things has been applied to every aspect of our lives. However, in the application process of the Internet of Things, the protection of personal privacy will undoubtedly be involved. If this problem is not effectively resolved, it will become a major obstacle to the development of the Internet of Things. At present, the research of fully homomorphic technology has attracted great attention from the cryptography community. You can directly calculate the encrypted text encryption to obtain the output and decrypt the output. The result is the same as the output of the unencrypted plain text. This article first comprehensively describes the solution to the privacy protection problem in the existing Internet of Things, and then proposes to apply the fully homomorphic technology to the Internet of Things to make the services provided by the network more secure. Through the analysis of the basic composition and architecture of the existing Internet of Things system, a privacy protection interaction model for the Internet of Things is established, which uses a completely homomorphic technology. On this basis, the algorithm for implementing simple homomorphic encryption is improved, and general homomorphic encryption theory is proposed for some security issues. After using this method to encrypt privacy, the success rate of cracking dropped by 24%.

There is growing attention and effort focused on treating the root cause of sensorineural hearing loss rather than managing associated secondary characteristic features. With recent substantial advances in understanding sensorineural hearing-loss mechanisms, gene delivery has emerged as a promising strategy for the biological treatment of hearing loss associated with genetic dysfunction. There are several successful and promising proof-of-principle examples of transgene deliveries in animal models; however, there remains substantial further progress to be made in these avenues before realizing their clinical application in humans. Herein, we review different aspects of development, ongoing preclinical studies, and challenges to the clinical transition of transgene delivery of the inner ear toward the restoration of lost auditory and vestibular function.

The efficient evacuation of people from dangerous areas is a key objective of emergency management. However, many emergencies give little to no advanced warning, leading to spontaneous evacuation with no time for planning or management. For large emergencies, destinations become less certain, with traffic demand imbalanced and concentrated on a few oversaturated routes familiar to evacuees. Ultimately, this leads to rapid congestion and delay on some routes, while others remain barely used, extending clearance times with an accumulating population at risk. In this study we address these issues through incorporating spatio-temporal traffic resilience dynamics into a destination choice model utilizing the available capacity of the overall network. We validate our model through a post-concert egress event. The results suggest that our method can reduce total egress times and average travel time by 20%–43% over the no-guidance condition. Our method can be used to estimate and quantify emergency conditions to optimally guide destinations and routing choice for evacuees and/or autonomously moving vehicles during evacuations.

Abstract Social media can be a double-edged sword for society, either as a convenient channel exchanging ideas or as an unexpected conduit circulating fake news through a large population. While existing studies of fake news focus on theoretical modeling of propagation or identification methods based on machine learning, it is important to understand the realistic propagation mechanisms between theoretical models and black-box methods. Here we track large databases of fake news and real news in both, Weibo in China and Twitter in Japan from different cultures, which include their traces of re-postings. We find in both online social networks that fake news spreads distinctively from real news even at early stages of propagation, e.g. five hours after the first re-postings. Our finding demonstrates collective structural signals that help to understand the different propagation evolution of fake news and real news. Different from earlier studies, identifying the topological properties of the information propagation at early stages may offer novel features for early detection of fake news in social media.

The brain network is one specific type of critical infrastructure networks, which supports the cognitive function of biological systems. With the importance of network reliability in system design, evaluation, operation, and maintenance, we use the percolation methods of network reliability on brain networks and study the network resistance to disturbances and relevant failure modes. In this paper, we compare the brain networks of different species, including cat, fly, human, mouse, and macaque. The differences in structural features reflect the requirements for varying levels of functional specialization and integration, which determine the reliability of brain networks. In the percolation process, we apply different forms of disturbances to the brain networks based on metrics that characterize the network structure. Our findings suggest that the brain networks are mostly reliable against random or k-core-based percolation with their structure design, yet becomes vulnerable under betweenness or degree-based percolation. Our results might be useful to identify and distinguish brain connectivity failures that have been shown to be related to brain disorders, as well as the reliability design of other technological networks.

Abstract Prediction of traffic congestion is one of the core issues in the realization of smart traffic. Accurate prediction depends on understanding of interactions and correlations between different city locations. While many methods merely consider the spatio-temporal correlation between two locations, here we propose a new approach of capturing the correlation network in a city based on realtime traffic data. We use the weighted degree and the impact distance as the two major measures to identify the most influential locations. A road segment with larger weighted degree or larger impact distance suggests that its traffic flow can strongly influence neighboring road sections driven by the congestion propagation. Using these indices, we find that the statistical properties of the identified correlation network is stable in different time periods during a day, including morning rush hours, evening rush hours, and the afternoon normal time respectively. Our work provides a new framework for assessing interactions between different local traffic flows. The captured correlation network between different locations might facilitate future studies on predicting and controlling the traffic flows.

Peripheral nerve injury (PNI) is a neurological disorder that causes more than 9 million patients to suffer from dysfunction of moving and sensing. Using biodegradable polymers to fabricate an artificial nerve conduit that replicates the environment of the extracellular matrix and guides neuron regeneration through the damaged sites has been researched for decades and has led to promising but primarily pre-clinical outcomes. However, few peripheral nerve conduits (PNCs) have been constructed from controllable biodegradable polymeric materials that can maintain their structural integrity or completely degrade during and after nerve regeneration respectively. In this work, a novel PNC candidate material was developed via the electrospinning of polyhydroxy butyrate/chitosan (PHB/CS) composite polymers. An SEM characterisation revealed the resultant PHB/CS nanofibres with 0, 1 and 2 wt/v% CS had less and smaller beads than the nanofibres at 3 wt/v% CS. The water contact angle (WCA) measurement demonstrated that the wettability of PHB/CS electrospun fibres was significantly improved by additional CS. Furthermore, both the thermogravimetric analysis (TGA) and differentiation scanning calorimetry (DSC) results showed that PHB/CS polymers can be blended in a single phase with a trifluoracetic solvent in all compositions. Besides, the reduction in the degradation temperature (from 286.9 to 229.9 °C) and crystallinity (from 81.0% to 52.1%) with increasing contents of CS were further proven. Moreover, we found that the degradability of the PHB/CS nanofibres subjected to different pH values rated in the order of acidic > alkaline > phosphate buffer solution (PBS). Based on these findings, it can be concluded that PHB/CS electrospun fibres with variable blending ratios may be used for designing PNCs with controlled biodegradability.

The importance of understanding human mobility patterns has led many studies to examine their spatial-temporal scaling laws. These studies mainly reveal that human travel can be highly non-homogeneous with power-law scaling distributions of distances and times. However, investigating and quantifying the extent of variability in time and space when traveling the same air distance has not been addressed so far. Using taxi data from five large cities, we focus on several novel measures of distance and time to explore the spatio-temporal variations of taxi travel routes relative to their typical routes during peak and nonpeak periods. To compare all trips using a single measure, we calculate the distributions of the ratios between actual travel distances and the average travel distance as well as between actual travel times and the average travel time for all origin destinations during peak and nonpeak periods. In this way, we measure the scaling of the distribution of all single trip paths with respect to their mean trip path. Our results surprisingly demonstrate very broad distributions for both the distance ratio and time ratio, characterized by a long-tail power-law distribution. Moreover, all analyzed cities have larger exponents in peak hours than in nonpeak hours. We suggest that the interesting results of shorter trip lengths and times, characterized by larger exponents during rush hours, are due to the higher availability of travelers during rush hours. Thus, drivers are more motivated to shorten their trips in order to take new passengers in rush hours compared to non-rush hours. We also find a high correlation between distances and times, and the correlation is lower during peak hours than during nonpeak hours. The reduced correlations can be understood as follows. Due to the high availability of passengers in peak periods more drivers choose long distances to save time compared to nonpeak periods. Furthermore, we employed an indeterminate traffic assignment model, which supports our finding of the power-law distribution of the distance ratio and time ratio for human mobility. Our results can help to assess traffic conditions within cities and provide guidance for urban traffic management.

In a previous study, we reported that no axons were crossing from the severed dorsal roots to the spinal cord using the rat dorsal rhizotomy paradigm. The injury caused ipsilateral deficits of forepaw function. An attempt to restore the function by transplanting cells containing 5% olfactory ensheathing cells (OECs) cultured from the olfactory mucosa did not succeed. However, obtaining OECs from the olfactory mucosa has an advantage for clinical application. In the present study, we used the same rhizotomy paradigm, but rats with an injury received cells from a modified mucosal culture containing around 20% OECs mixed in collagen. The forelimb proprioception assessment showed that 80% of the rats receiving the transplants had functional improvement over six weeks of the study. The adhesive removal test showed that the time taken for the rats to notice the adhesive label and remove it almost returned to the normal level after receiving the transplants. Transplanted cells were identified with the expression of green fluorescent protein (ZsGreen). Some regeneration fibres immunostained for neurofilament (NF) or traced by biotinylated dextran amine (BDA) in the injury area were associated with the transplanted cells. The evidence in this study improves the prospect of clinical application using OECs from the olfactory mucosa to treat CNS injuries.

Abstract Existing studies have developed different indices based on various approaches including network connectivity, delay time and flow capacity, estimating the traffic reliability states from different angles. However, these indices mainly estimate traffic reliability from single view and rarely consider the combined effect of city traffic dynamics and underlying network structure. Based on percolation theory, Li et al. has developed a traffic reliability index to address this issue (Proc. Natl. Acad. Sci. USA 112(3):669-672, 2015) [1]. Here we compare this percolation-based index with one of the well-known index - congestion delay index (CDI). Using real traffic data of Beijing and Shenzhen (two large cities in China), we compare the two indices in the macroscopic trends and microscopic extreme values. The two indices are found to indicate the state of real-time traffic reliability in different consideration. Our results can be used for better evaluation of traffic system reliability and mitigation measures of traffic jams.

The dorsal root entry zone is often used in research to examine the disconnection between the central and peripheral parts of the nervous system which occurs following injury. Our laboratory and others have used transplantation of olfactory ensheathing cells (OECs) to repair experimental spinal cord injuries. We have previously used a four dorsal root (C6–T1) transection model to show that transplantation of OECs can reinstate rat forelimb proprioception in a climbing task. Until now, however, we have not looked in detail at the anatomical interaction between OECs and the peripheral/central nervous system regions which form the transitional zone. In this study, we compared short- and long-term OEC survival and their interaction with the surrounding dorsal root tissue. We reveal how transplanted OECs orient toward the spinal cord and allow newly formed axons to travel across into the dorsal horn of the spinal cord. Reconstruction of the dorsal root entry zone was supported by OEC ensheathment of axons at the injured site and also at around 3 mm further away at the dorsal root ganglion. Quantitative analysis revealed no observable difference in dorsal column axonal loss between transplanted and control groups of rats.

Overload failures propagate through hidden functional dependencies across networked systems. Here, the authors study the spatio-temporal propagation behaviour of cascading overload failures, and find that they spread radially from their origin with an approximately constant velocity.

Many complex systems in the real world can be modeled as complex networks, which has captured in recent years enormous attention from researchers of diverse fields ranging from natural sciences to engineering. The extinction of species in ecosystems and the blackouts of power girds in engineering exhibit the vulnerability of complex networks, investigated by empirical data and analyzed by theoretical models. For studying the resilience of complex networks, three main factors should be focused on: the network structure, the network dynamics and the failure mechanism. In this review, we will introduce recent progress on the resilience of complex networks based on these three aspects. For the network structure, increasing evidence shows that biological and ecological networks are coupled with each other and that diverse critical infrastructures interact with each other, triggering a new research hotspot of “networks of networks” (NON), where a network is formed by interdependent or interconnected networks. The resilience of complex networks is deeply influenced by its interdependence with other networks, which can be analyzed and predicted by percolation theory. This review paper shows that the analytic framework for Energies 2015, 8 12188 NON yields novel percolation laws for n interdependent networks and also shows that the percolation theory of a single network studied extensively in physics and mathematics in the last 60 years is a specific limited case of the more general case of n interacting networks. Due to spatial constraints inherent in critical infrastructures, including the power gird, we also review the progress on the study of spatially-embedded interdependent networks, exhibiting extreme vulnerabilities compared to their non-embedded counterparts, especially in the case of localized attack. For the network dynamics, we illustrate the percolation framework and methods using an example of a real transportation system, where the analysis based on network dynamics is significantly different from the structural static analysis. For the failure mechanism, we here review recent progress on the spontaneous recovery after network collapse. These findings can help us to understand, realize and hopefully mitigate the increasing risk in the resilience of complex networks.

Trauma causes spinal cord injury, and the devastating consequences of the injury are due to the failure of the damaged central nervous system (CNS) axons to regenerate. Previous studies have shown that olfactory ensheathing cells (OECs) are a unique type of glial cell and they can promote regeneration of CNS axons to aid recovery after spinal cord injury. Transplantation of OECs, in particular from the olfactory bulb (OB), is considered one of the most promising therapeutic strategies for the repair of CNS injuries, including spinal cord injury. Transplantation of OECs can be autologous or allogenic. Here we focused on the less invasive and more error-proof allograft approach which needs a collection of donor OB tissue for OEC production. In this study, we investigated the effects on the yield and proportions of OECs and olfactory nerve fibroblasts (ONFs) from storing OB tissue in various media for periods of 24 and 48 hours. The OEC yield contributes to the viability of a successful cell transplant. We concluded that storing OB tissue for a period longer than 24 hours negatively impacted the total cell number and subsequently the OEC population. This study provides useful information for future clinical applications.

Cell therapy has been shown to be a key clinical therapeutic option for central nervous system diseases or damage. Standardization of clinical cell therapy procedures is an important task for professional associations devoted to cell therapy. The Chinese Branch of the International Association of Neurorestoratology (IANR) completed the first set of guidelines governing the clinical application of neurorestoration in 2011. The IANR and the Chinese Association of Neurorestoratology (CANR) collaborated to propose the current version “Clinical Cell Therapy Guidelines for Neurorestoration (IANR/CANR 2017)”. The IANR council board members and CANR committee members approved this proposal on September 1, 2016, and recommend it to clinical practitioners of cellular therapy. These guidelines include items of cell type nomenclature, cell quality control, minimal suggested cell doses, patient-informed consent, indications for undergoing cell therapy, contraindications for undergoing cell therapy, documentation of procedure and therapy, safety evaluation, efficacy evaluation, policy of repeated treatments, do not charge patients for unproven therapies, basic principles of cell therapy, and publishing responsibility.

Putative gamete-derived progenies from two Hylocereus species, the diploid H. monacanthus and the tetraploid H. megalanthus, were studied with the dual aims to confirm their gamete origin and to evaluate their potential use as genetic resources. An additional goal was to determine the origin (allotetraploid vs. autotetraploid) of H. megalanthus by exploring morphological variations in the di-haploid (2x) H. megalanthus progeny. Gamete origin was proved in all five H. monacanthus lines obtained and in 49 of the 70 H. megalanthus lines by using flow cytometry and simple sequence repeat (SSR) markers. The five double-haploid (2x) H. monacanthus lines showed low vigor and abnormal flower development, with malformed ovules and aborted pollen grains. Only one flower set fruit, giving several viable seeds. For H. megalanthus, both abnormal ovules and defective anthers were observed in the di-haploid (2x) and double di-haploid (4x) lines. Among the 46 di-haploid lines, only 14 set fruit. Another 13 di-haploid lines formed flower buds that abscised before anthesis or soon after pollination. The severe sterility of the double-haploid H. monacanthus and the reduced fertility of all the di-haploid and double di-haploid H. megalanthus lines can be linked to their reduced heterozygosity, which drastically affected the development of normal female and male organs. We thus concluded that chromosome doubling, as occurred spontaneously in the double-haploid H. monacanthus and the double di-haploid H. megalanthus, is not sufficient to restore fertility in Hylocereus. We also observed very low gametoclonal variation among the di-haploid (2x) H. megalanthus lines, a finding that supported an autotetraploid, rather than an allotetraploid, origin of this species. Nonetheless, despite the above-described challenging limitations, these gamete-derived lines are currently being bred as the seed parent, offering unique possibilities for genetic research and additional breeding.

The central branches of the C7 and C8 dorsal roots were avulsed close to their entry point into the spinal cord in adult rats. The forepaw responses to heat and cold stimuli were tested at 1, 2, and 3 weeks after injury. Over this period, the paws were sensitive to both stimuli at 1-2 weeks and returned toward normal at 3 weeks. Immunohistology showed no evidence of axonal regeneration into the spinal cord in a control group of rats with avulsion only, implying that adjacent dorsal roots and their corresponding dermatomes were involved in the recovery. In a further group of rats, a mixture of bulbar olfactory ensheathing cells and olfactory nerve fibroblasts were transplanted into the gap between the avulsed roots and the spinal cord at the time of avulsion. These rats showed no evidence of either loss of sensation or exaggerated responses to stimuli at any of the time points from 1 to 3 weeks. Immunohistology showed that the transplanted cells formed a complete bridge, and the central branches of the dorsal root fibers had regenerated into the dorsal horn of the spinal cord. These regenerating axons, including Tuj1 and CGRP immunoreactive fibers, were ensheathed by the olfactory ensheathing cells. This confirms our previous demonstration of central regeneration by these transplants and suggests that such transplants may provide a useful means to prevent the development of abnormal sensations such as allodynia after spinal root lesions.

Cell transplantation is one of the most promising strategies for repair of human spinal cord injuries. Animal studies from a number of laboratories have shown that transplantation of olfactory ensheathing cells cultured from biopsies of the olfactory bulb mediate axonal regeneration and remyelination and restore lost functions in spinal cord injuries. For translation from small laboratory experimental injuries to the large spinal cord injuries encountered in human patients the numbers of cells that can be obtained from a patient's own olfactory bulb becomes a serious limiting factor. Furthermore, removal of an olfactory bulb requires invasive surgery and risks unilateral anosmia. We here report that xenografted mouse bulbar olfactory ensheathing cells immunoprotected by daily cyclosporine restore directed forepaw reaching function in rats with chronic C1/2 unilateral corticospinal tract lesions. Once function had been established for 10 days, cyclosporine was withdrawn. Thirteen out of 13 rats continued to increase directed forepaw reaching. Immunohistochemistry shows that in all cases neurofilament-positive axons were present in the lesion, but that the grafted cells had been totally rejected. This implies that once grafted cells have acted as bridges for axon regeneration across the lesion site their continued presence is no longer necessary for maintaining the restored function. This raises the possibility that in the future a protocol of temporary immunoprotection might allow for the use of the larger available numbers of immunoincompatible allografted cells or cell lines, which would avoid the need for removing a patient's olfactory bulb.

Networks can be used to describe the interconnections among individuals, which play an important role in the spread of disease. Although the small-world effect has been found to have a significant impact on epidemics in single networks, the small-world effect on epidemics in interconnected networks has rarely been considered. Here, we study the susceptible-infected-susceptible (SIS) model of epidemic spreading in a system comprising two interconnected small-world networks. We find that the epidemic threshold in such networks decreases when the rewiring probability of the component small-world networks increases. When the infection rate is low, the rewiring probability affects the global steady-state infection density, whereas when the infection rate is high, the infection density is insensitive to the rewiring probability. Moreover, epidemics in interconnected small-world networks are found to spread at different velocities that depend on the rewiring probability.

Treatment of patients sustaining a complete spinal cord injury remains an unsolved clinical problem because of the lack of spontaneous regeneration of injured central axons. A 38-year-old man sustained traumatic transection of the thoracic spinal cord at upper vertebral level Th9. At 21 months after injury, the patient presented symptoms of a clinically complete spinal cord injury (American Spinal Injury Association class A-ASIA A). One of the patient's olfactory bulbs was removed and used to derive a culture containing olfactory ensheathing cells and olfactory nerve fibroblasts. Following resection of the glial scar, the cultured cells were transplanted into the spinal cord stumps above and below the injury and the 8-mm gap bridged by four strips of autologous sural nerve. The patient underwent an intense pre- and postoperative neurorehabilitation program. No adverse effects were seen at 19 months postoperatively, and unexpectedly, the removal of the olfactory bulb did not lead to persistent unilateral anosmia. The patient improved from ASIA A to ASIA C. There was improved trunk stability, partial recovery of the voluntary movements of the lower extremities, and an increase of the muscle mass in the left thigh, as well as partial recovery of superficial and deep sensation. There was also some indication of improved visceral sensation and improved vascular autoregulation in the left lower limb. The pattern of recovery suggests functional regeneration of both efferent and afferent long-distance fibers. Imaging confirmed that the grafts had bridged the left side of the spinal cord, where the majority of the nerve grafts were implanted, and neurophysiological examinations confirmed the restitution of the integrity of the corticospinal tracts and the voluntary character of recorded muscle contractions. To our knowledge, this is the first clinical indication of the beneficial effects of transplanted autologous bulbar cells.

In an ongoing clinical trial, a spinal injured patient who received a transplant of autologous cells cultured from the olfactory bulb is showing greater functional benefit than three previous patients with transplants of mucosal origin. Previous laboratory studies of transplantation into rat spinal cord injuries show that the superior reparative benefits of bulbar over mucosal cultures are associated with regeneration of severed corticospinal tract fibers over a bridge of olfactory ensheathing cells (OECs) formed across the injury site. In a rat rhizotomy paradigm, we reported that transplantation of bulbar cell cultures also enables severed axons of the C6–T1 dorsal roots to regenerate across a bridge of OECs into the spinal cord and restore electrophysiological transmission and forepaw grasping during a climbing test. We now report a repeat of the same rhizotomy procedure in 25 rats receiving cells cultured from olfactory mucosal biopsies. In no case did the transplanted cells form a bridging pathway. No axons crossed from the severed roots to the spinal cord, and there was no restoration of forepaw grasping. This suggests that the superior clinical benefit in the patient receiving bulbar cell transplants is due to regeneration of severed fibers across the injury site, and this correlates with imaging and the pattern of functional recovery. Using present culture protocols, the yield of OECs from bulbar biopsies is around 50%, but that from mucosal biopsies is less than 5%. Improving the yield of OECs from mucosal biopsies might avoid the necessity for the intracranial approach to obtain bulbar cells.

The participation of individuals in multi-layer networks allows for feedback between network layers, opening new possibilities to mitigate epidemic spreading. For instance, the spread of a biological disease such as Ebola in a physical contact network may trigger the propagation of the information related to this disease in a communication network, e.g. an online social network. The information propagated in the communication network may increase the awareness of some individuals, resulting in them avoiding contact with their infected neighbors in the physical contact network, which might protect the population from the infection. In this work, we aim to understand how the time scale γ of the information propagation (speed that information is spread and forgotten) in the communication network relative to that of the epidemic spread (speed that an epidemic is spread and cured) in the physical contact network influences such mitigation using awareness information. We begin by proposing a model of the interaction between information propagation and epidemic spread, taking into account the relative time scale γ . We analytically derive the average fraction of infected nodes in the meta-stable state for this model (i) by developing an individual-based mean-field approximation (IBMFA) method and (ii) by extending the microscopic Markov chain approach (MMCA). We show that when the time scale γ of the information spread relative to the epidemic spread is large, our IBMFA approximation is better compared to MMCA near the epidemic threshold, whereas MMCA performs better when the prevalence of the epidemic is high. Furthermore, we find that an optimal mitigation exists that leads to a minimal fraction of infected nodes. The optimal mitigation is achieved at a non-trivial relative time scale γ , which depends on the rate at which an infected individual becomes aware. Contrary to our intuition, information spread too fast in the communication network could reduce the mitigation effect. Finally, our finding has been validated in the real-world two-layer network obtained from the location-based social network Brightkite.

System restoration from cascading failures is an integral part of the overall defense against catastrophic breakdown in networked critical infrastructures. From the outbreak of cascading failures to the system complete breakdown, actions can be taken to prevent failure propagation through the entire network. While most analysis efforts have been carried out before or after cascading failures, restoration during cascading failures has been rarely studied. In this paper, we present a modeling framework to investigate the effects of in-process restoration, which depends strongly on the timing and strength of the restoration actions. Furthermore, in the model we also consider additional disturbances to the system due to restoration actions themselves. We demonstrate that the effect of restoration is also influenced by the combination of system loading level and restoration disturbance. Our modeling framework will help to provide insights on practical restoration from cascading failures and guide improvements of reliability and resilience of actual network systems.

Protein interaction networks have become a tool to study biological processes, either for predicting molecular functions or for designing proper new drugs to regulate the main biological interactions. Furthermore, such networks are known to be organized in sub-networks of proteins contributing to the same cellular function. However, the protein function prediction is not accurate and each protein has traditionally been assigned to only one function by the network formalism. By considering the network of the physical interactions between proteins of the yeast together with a manual and single functional classification scheme, we introduce a method able to reveal important information on protein function, at both micro- and macro-scale. In particular, the inspection of the properties of oscillatory dynamics on top of the protein interaction network leads to the identification of misclassification problems in protein function assignments, as well as to unveil correct identification of protein functions. We also demonstrate that our approach can give a network representation of the meta-organization of biological processes by unraveling the interactions between different functional classes.

Recent studies have shown evidence for the preservation of colour in fossilized soft tissues by imaging melanosomes, melanin pigment containing organelles. This study combines geochemical analyses with morphological observations to investigate the preservation of melanosomes and melanin within feathers of the Early Cretaceous bird, Gansus yumenensis. Scanning electron microscopy reveals structures concordant with those previously identified as eumelanosomes within visually dark areas of the feathers but not in lighter areas or sedimentary matrices. Fourier transform infrared analyses show different spectra for the feathers and their matrices; melanic functional groups appear in the feather including carboxylic acid and ketone groups that are not seen in the matrix. When mapped, the carboxylic acid group absorption faithfully replicates the visually dark areas of the feathers. Electron Paramagnetic Resonance spectroscopy of one specimen demonstrates the presence of organic signals but proved too insensitive to resolve melanin. Pyrolysis gas chromatography mass spectrometry shows a similar distribution of aliphatic material within both feathers that are different from those of their respective matrices. In combination, these techniques strongly suggest that not only do the feathers contain endogenous organic material, but that both geochemical and morphological evidence supports the preservation of original eumelanic pigment residue.

Two-dimensional silicon-carbide (SixCy) materials stand out for their compatibility with current siliconbased technologies, offering unique advantages in nanoelectronics and photocatalysis. In this study, we employ density functional theory and nonequilibrium Green's function methods to investigate the electronic properties, electron transport characteristics, and optoelectronic qualities of experimentally synthesized monolayer Si9C15. Utilizing the modified deformation potential theory formula, we unveil Si9C15's significant directional anisotropy in electron mobility (706.42 cm2 V-1 s-1) compared to holes (432.84 cm² V-1 s-1) in the a direction. The electrical transport calculations reveal that configurations with a 3 nm channel length demonstrate an ON state when biased, reaching a peak current of 150 nA. Moreover, this maximum current value escalates to 200 nA under tensile strain, marking an increase of approximately 100 times compared to the 5 nm channel, which remains in an OFF state. Si9C15 exhibits high light absorption coefficients (B105 cm-1) and suitable band edge positions for water splitting at pH 0-7. Applying 1-5% tensile strain can tune the conduction band minimum and valence band maximum closer to the standard redox potentials, enhancing photocatalytic water splitting efficiency. Remarkably, under illumination at pH 0 and 7, Si9C15 can spontaneously catalyze water splitting, demonstrating its potential as a highly efficient photocatalyst. Our findings emphasize the importance of strain control and device length optimization for performance enhancement in nanoelectronics and renewable energy applications, positioning Si9C15 as a promising material for high-performance field-effect transistors and photocatalytic water splitting.
