Salinity adaptive responses in halophytes include energy-requiring processes, but the mechanism supplying ATP to meet the increased ATP demand remains unclear. In the present study, we examined the effects of NaCl on ATP synthesis in mitochondria isolated from a halophyte, the common ice plant (Mesembryanthemum crystallinum L.). The ATP synthesis rate was maintained or enhanced with increasing osmotic pressures ranged from 0.2 to 2.0 MPa in the assay mixtures containing NaCl, whereas it was decreased in the mixtures with the equivalent osmotic pressures generated by sorbitol only. Also, the ATP synthesis rate was enhanced with increasing NaCl concentrations ranged from 50 to 350 mM in the assay mixtures with fixed (2.0 MPa) osmotic pressure. These results suggested a mechanism of enhanced ATP synthesis in the mitochondria by the ionic effects of NaCl, which might play an important role in the adaptation of the ice plant under salinity.

A halophyte, the common ice plant (Mesembryanthemum crystallinum L.), shows the maximal growth under salinity, in which almost all crops die. The NaCl-stimulated growth, which is referred to as halophilism, is an important trait for adaptation to salinity, but the mechanism is still unclear. To elucidate factors contributing to the halophilism, we tested the effects of NaCl on growth, ion accumulation, and expression of ion homeostasis-related genes in suspension-cultured cells. Among nine ions analyzed, Na+, Cl−, K+, and NO3− were accumulated significantly in the cells showing halophilism than that in the salt-untreated cells, and the accumulation of these ions was positively correlated with the growth. Also, the expression of ion homeostasis-related genes for plasma membrane transporters and channels for incorporation of nitrate (McNRT1), sodium (McHKT1), potassium (McKmt1), cations/Cl− (McCCC1), for tonoplast antiporters H+/Cl− (McCLC1) and Na+/H+ (McNHX1), and V-ATPase subunit c (McVmac1) for sequestration of Cl− and Na+ into the vacuole, and for enzymes catalyzing biosynthesis of proline (McP5CS) and ononitol (McImt1) was higher in the cells showing halophilism than that in the salt-untreated cells. In this study, we investigated the feasibility of local tumor IR through UTMD to enhance the anti-tumor efficacy of doxorubicin (DOX) chemotherapy.Methods: UTMD was used to induce local tumor IR. After the major blood supply of the tumor was restored, DOX was intravenously injected into the tumor-bearing mice. The superoxide dismutase (SOD) and catalase (CAT) activity and ROS levels were examined, and the anti-tumor efficacy was evaluated.Results: UTMD blocked the circulation to the tumor for 30 mins. Slow reperfusion began to occur after 30 mins, and major blood supply was restored after 1 hr. The blood perfusion of the tumor completely recovered at 2 hrs. The activity of SOD in the tumors was significantly decreased at 2 hrs and 1 day after IR treatment with or without DOX treatment. The CAT activity showed no obvious changes at 2 hrs after IR treatment, whereas a significant decrease was found after 1 day in both the IR and DOX/IR groups. Moreover, higher levels of ROS were produced in the IR group and IR/DOX group. In vivo anti-tumor study indicated that the local tumor IR strategy may significantly enhance the anti-tumor efficacy of DOX chemotherapy.Conclusion: UTMD provides a novel, simple and non-invasive technique for tumor IR. Despite intensive lubrication, and attempts at surgical closure including an amniotic membrane patch graft, it progressed with subsequent adjacent corneal perforation. Thorough investigations revealed no underlying disease, except markedly delayed episcleral vessel filling on anterior segment fluorescein angiography.Conclusions: Neovascularisation is a known factor in the inhibition of ulceration. Abstract Background Although national policies in China are comprehensive and instructive, a wide disparity exists between different cities. The current status of public mental health services by region in China remains unclear. This study aimed to investigate policies related to public mental health services, the contact coverage of public mental health services and outcomes of service users. Methods A cross-sectional study was carried out in Southern China, between April 2021 and March 2022. Considering the geographical location, socioeconomic development levels, and prevalence of severe mental illness, four cities including Wuhan, Changsha, Guangzhou, and Shenzhen were selected. Relevant service providers were asked to report data on mental health policies and facility-related information, including mental health resources, registration rates of patients, management rates of patients, and medication rates of patients. Eligible patients were invited to report service user-related data, including contact coverage of public mental health services and their outcomes. SPSS 26.0 was used for data analysis. Results The four cities in Southern China have made different efforts to develop public mental health services, primarily focusing on socio-economically disadvantaged individuals. Community health centers in Guangzhou and Shenzhen reported having more professional human resources on mental health and higher mental health budgets for patients. The contact coverage rates of most public mental services were higher than 80%. Patients in Changsha (B = 0.3; 95%CI: 0.1–0.5), Guangzhou (B = 0.2; 95%CI: 0.1–0.3), and Shenzhen (B = 0.3; 95%CI: 0.1–0.4) who received social medical assistance services reported higher levels of medication adherence. Patients in Wuhan (B = -6.5; 95%CI: -12.9--0.1), Guangzhou (B = -2.8; 95%CI: -5.5--0.1), and Shenzhen who received community-based rehabilitation services reported lower levels of disability (B = -2.6; 95%CI: -4.6--0.5). Conclusions There have been advances in public mental health services in the four Southern cities. The contact coverage rates of most public mental health services were higher than 80%. Patients’ utilization of public mental services was associated with better health outcomes. To improve the quality of public mental health services, the government should try to engage service users, their family members, and supporters in the design, delivery, operationalization, and evaluation of these public mental health services in the future.

Abstract Human trophoblastic cell surface antigen 2 (Trop2) is a glycoprotein, a cellular marker of trophoblastic and stem cells, and a calcium signaling transducer involved in several signaling pathways, leading to the proliferation, invasion, and metastasis of tumors. It is expressed at a low level in normal epithelial cells, but at a high level in many tumors, making it an ideal target for cancer therapy. According to previous literature, Trop2 is broadly expressed in all breast cancer subtypes, especially in triple negative breast cancer (TNBC). Several clinical trials have demonstrated the effectiveness of Trop2-targeted therapy in breast cancer. Sacituzumab govitecan (SG) is a Trop2-targeted antibody-drug conjugate (ADC) that has been approved for the treatment of metastatic TNBC and hormone receptor-positive (HR+) and human epidermal growth factor receptor 2-negative (HER2-) breast cancer. This article reviews the structure and function of Trop2, several major Trop2-targeted ADCs, other appealing novel Trop2-targeted agents and relevant clinical trials to provide a landscape of how Trop2-targeted treatments will develop in the future.

Objective To detect endogenous heparin substance in patients with epidemic haemorrhagic fever (EHF) using thromboelastography (TEG) combined with conventional coagulation indices(CCTs), explore the value of TEG combined with CCTs in detecting coagulation function abnormalities in such patients, aiming at providing guidance for clinical treatment. Methods APTT, TT, TEG plain cup R value and TEG heparinase cup R value were tested in 35 EHF patients of mild/moderate stage, severe stage and recovery stage. Intergroup differences were analysed using Friedman ANOVA. Results The mean R and heparin cup R values were 7.5 and 6.7 for mild cases of EHF, and TT, R and heparinase cup R values were 16.9, 6.2 and 6.1 for recovering cases, respectively.The normal cup R and heparinase cup R values for mild cases, as well as the TT, R and heparinase cup R values for recovering cases, were normally distributed, and the rest were non-normally distributed. Abstract Limited data availability remains a significant challenge for Whisper’s low-resource speech recognition performance, falling short of practical application requirements. While previous studies have successfully reduced the recognition error rates of target language speech through fine-tuning, a comprehensive exploration and analysis of Whisper’s fine-tuning capabilities and the advantages and disadvantages of various fine-tuning strategies are still lacking. This paper aims to fill this gap by conducting comprehensive experimental exploration for Whisper’s low-resource speech recognition performance using five fine-tuning strategies with limited supervised data from seven low-resource languages. The results and analysis demonstrate that all fine-tuning strategies explored in this paper significantly enhance Whisper’s performance. However, different strategies vary in their suitability and practical effectiveness, highlighting the need for careful selection based on specific use cases and resources available.

Abstract Pleiotropy is frequently detected in agronomic traits of wheat (Triticum aestivum). A locus on chromosome 4B, QTn/Ptn/Sl/Sns/Al/Tgw/Gl/Gw.caas-4B, proved to show pleiotropic effects on tiller, spike, and grain traits using a recombinant inbred line (RIL) population of Qingxinmai × 041133. The allele from Qingxinmai increased tiller numbers, and the allele from line 041133 produced better performances of spike traits and grain traits. Another 52 QTL for the eight traits investigated were detected on 18 chromosomes, except for chromosomes 5D, 6D, and 7B. Several genes in the genomic interval of the locus on chromosome 4B were differentially expressed in crown and inflorescence samples between Qingxinmai and line 041133. The development of the KASP marker specific for the locus on chromosome 4B is useful for molecular marker-assisted selection in wheat breeding.

Abstract Crop wild relatives offer natural variations of disease resistance for crop improvement. Here, we report the isolation of broad-spectrum powdery mildew resistance gene Pm36, originated from wild emmer wheat, that encodes a tandem kinase with a transmembrane domain (WTK7-TM) through the combination of map-based cloning, PacBio SMRT long-read genome sequencing, mutagenesis, and transformation. Mutagenesis assay reveals that the two kinase domains and the transmembrane domain of WTK7-TM are critical for the powdery mildew resistance function. Consistently, in vitro phosphorylation assay shows that two kinase domains are indispensable for the kinase activity of WTK7-TM. Haplotype analysis uncovers that Pm36 is an orphan gene only present in a few wild emmer wheat, indicating its single ancient origin and potential contribution to the current wheat gene pool. Overall, our findings not only provide a powdery mildew resistance gene with great potential in wheat breeding but also sheds light into the mechanism underlying broad-spectrum resistance.

Abstract Grangea maderaspatana is a valuable medicinal plant growing in salt-affected areas, but its tolerance capability, physiological and biochemical responses to salinity is still unclear. To understand these traits, this study examined effects of salinity at different levels (50-400 mM NaCl) on plant growth and its responses. The results shown that the plant’s dry biomass decreased with increasing salinity levels of 100-400 mM NaCl, but its growth was maintained at 400 mM NaCl level with a dry biomass equal to 0.45 times that of the control, indicating that G. maderaspatana had a tolerance ability to high salinity. The plant also had adaptive responses to the salinity. The content of leaf chlorophylls and carotenoids were retained, even enhanced by 50-200 mM NaCl, suggesting a high adaptation of photosynthesis. Proline, Na+, and Cl- was highly accumulated while the accumulation of K+ and NO3 - was maintained with 200-400 mM NaCl, indicating that the plant had adaptive mechanisms for osmotic adjustment and ion homeostasis. Antioxidative activities of catalase, superoxide dismutase, and peroxidase were enhanced by the salinity. These findings are useful information for understanding salt tolerance mechanisms and for utilization of this medicinal plant in saline agriculture.

The role of the anchorage slot is to fix the anchorage and tension anchor rope, which is the weakest link of the concrete lining structure. The stress state of the anchorage slot area in the construction and operation stages of the 2# sand discharge of the Xiaolangdi Project was analyzed by modeling in this paper. The results showed that the stress state of the anchorage slot area was the most complex. The tensile stress was generated in the radial direction of the concrete lining during the construction period, and the circumferential pre-compression stress was unevenly distributed along the thickness direction. During the operation period, when the backfill concrete at the anchor slot and the lining jointly bore the internal water pressure, a tensile stress of 0.89 MPa was generated in the inner anchorage slot area of the lining, which was less than the tensile strength of the concrete itself. Finite element calculation results were compared with the field-measured data, and the results were essentially the same. Finite element analysis software was used to optimize the double-row asymmetric arrangement of the anchorage slots into a single-row symmetrical structure. The results showed that the angle of the complex stress area of the lining is reduced by about 75%. The optimized single-row symmetrical anchorage slot layout was safer, and the pre-stressing distribution was more uniform.

Phyla nodiflora is a valuable medicinal plant growing in coastal areas, hypothesizing its adaptability to salinity; however, it has not been investigated. This study, for the first time, elucidated responses in the growth of the shoots and its physiology to different soil salinity of 50–400 mM NaCl. The data showed that the shoot’s dry biomass was not affected by the salinity levels up to 100 mM, and it only decreased 33.50–56.33% compared to the control under 200–400 mM NaCl, indicating that P. nodiflora is a salt-tolerant plant that could survive under high salinity. In addition, the plant also had physiological responses which indicated its salt-induced injuries and adaptation to the salt stress. The chlorophyll a content was increased while the chlorophyll b remained unchanged under the salt stress. The proline and salt accumulation increased under the salinity, but the K+ and NO3− accumulation decreased. Moreover, increases in malondialdehyde and electrolyte leakage were observed, indicating salt-induced membrane damages. These responses suggested that the plant might evolve adaptive mechanisms to salinity. Our findings are useful information for further research in order to elucidate the salt-tolerant mechanisms and develop this plant for saline agriculture.

Abstract This study aimed to investigate the associations between the clinical characteristics and effectiveness of anti-PD-1 inhibitors in patients with EGFR-sensitive mutations, aiming to identify the potential subgroup of patients who might benefit from anti-PD-1 inhibitor treatment. Patients with advanced non-small cell lung cancer (NSCLC) harboring epidermal growth factor receptor (EGFR)-sensitive mutations who received subsequent anti-PD-1 inhibitors in combination with chemotherapy/antiangiogenic agents or alone after progression to tyrosine kinase inhibitors (TKIs) were screened. Clinical characteristics, including hematological parameters, were investigated for potential correlations with clinical outcomes. Subgroup and multivariate analyses were used for further confirmation of the relationship. Kaplan–Meier curves and Cox survival regression models using the log-rank test were used for progression-free survival (PFS) and overall survival (OS) assessments between the groups. Multiple regression analysis was performed using the standard regression coefficient values. The Wilcoxon test was used for the analysis of the variation in NLR. P ≤ 0.05 was considered to indicate statistical significance. This study was a retrospective study. Twenty-two patients met the inclusion criteria and were included in the study. The median PFS was 3.05 months (95% CI, 2.9–10.2 months). The median OS was 7.30 months (95% CI, 5.2–18.1 months). PFS in low neutrophil to lymphocyte ratio (NLR ≤ 4) was significantly longer than high NLR (NLR > 4, 5.7 months versus 2.0 months, HR, 0.35, 95% CI, 0.08–0.63, P = 0.0083). The OS in the low NLR group was also significantly better than that in the high NLR group (OS, 21.3 months versus 5.0 months, HR, 0.33; 95% CI, 0.09–0.74; P = 0.0163). In the multivariate analysis, NLR was the only significant factor for OS benefits (β = 3.535, 95% CI, 1.175–10.636, P = 0.025). Further investigation revealed that front-line TKIs exposure may contribute to the elevation or decrease of NLR, and finally lead to different efficacy outcomes by anti-PD-1 inhibitors. The findings suggest that a portion of advanced NSCLC patients with low NLR characteristics (NLR ≤ 4), even those harboring EGFR-sensitive mutations, could benefit from anti-PD-1 inhibitors as further line treatment after progression to EGFR-TKIs.

Lức dây (Phyla nodiflora) là loài cây thuốc có nhiều giá trị y học và có tiềm năng chịu mặn. Tuy nhiên, khả năng chống chịu của cây đối với mặn vẫn chưa được làm rõ. Mục tiêu của bài báo này là làm rõ khả năng chịu mặn và những thay đổi trong sinh trưởng, tình trạng nước và hàm lượng sắc tố quang hợp của cây ở các điều kiện mặn đất khác nhau (50, 100, 200, 400 mM NaCl). Kết quả cho thấy Lức dây có thể duy trì sự sinh trưởng với nồng độ mặn lên tới 400 mM NaCl. Sinh khối tươi và khô của cây không thay đổi đáng kể ở nồng độ 50–100 mM NaCl so với đối chứng, nhưng giảm tương ứng 51,68–71,15% và 34,96–58,03% ở các nồng độ 200–400 mM NaCl. Hàm lượng nước tương đối cũng đã giảm khi nồng độ muối tăng lên, tuy nhiên, hàm lượng các sắc tố quang hợp đã có xu hướng tăng có ý nghĩa ở nồng độ muối 400 mM. An active chitosan (CS) film containing a nano-hydroxyapatite–tea polyphenol (HAP-TP) complex was designed and prepared. The effects of HAP-TP loading on the structural and physicochemical properties of the CS-based film were evaluated. The mechanical and thermal properties of the film were significantly improved by the resulting intermolecular interactions and formation of hydrogen bonds between HAP-TP and CS, which reduced the water vapor and oxygen permeabilities of the film by 29.78 and 35.59 %, respectively. The CS-HAP-TP film exhibited excellent slow-release behavior and antioxidant activity, with a cumulative release rate at 700 h 6.79 % lower than that of CS-TP films. The CS-HAP-TP film significantly inhibited the deterioration of semi-dried golden pompano, and thus helped to retain the taste of umami and sweet amino acids in fish samples, while reducing off-flavor generation. The film therefore shows considerable potential as an active packaging material for the preservation of semi-dried fish products.

Bispecific antibodies, including bispecific IgG, are emerging as an important new class of antibody therapeutics. As a result, we, as well as others, have developed engineering strategies designed to facilitate the efficient production of bispecific IgG for clinical development. For example, we have extensively used knobs-into-holes (KIH) mutations to facilitate the heterodimerization of antibody heavy chains and more recently Fab mutations to promote cognate heavy/light chain pairing for efficient in vivo assembly of bispecific IgG in single host cells. A panel of related monospecific and bispecific IgG1 antibodies was constructed and assessed for immunogenicity risk by comparison with benchmark antibodies with known low (Avastin and Herceptin) or high (bococizumab and ATR-107) clinical incidence of anti-drug antibodies. Assay methods used include dendritic cell internalization, T cell proliferation, and T cell epitope identification by in silico prediction and MHC-associated peptide proteomics. Data from each method were considered independently and then together for an overall integrated immunogenicity risk assessment. In toto, these data suggest that the KIH mutations and in vitro assembly of half antibodies do not represent a major risk for immunogenicity of bispecific IgG1, nor do the Fab mutations used for efficient in vivo assembly of bispecifics in single host cells. Comparable or slightly higher immunogenicity risk assessment data were obtained for research-grade preparations of trastuzumab and bevacizumab versus Herceptin and Avastin, respectively. These data provide experimental support for the common practice of using research-grade preparations of IgG1 as surrogates for immunogenicity risk assessment of their corresponding pharmaceutical counterparts.

IntroductionObservational investigations have examined the impact of glucosamine use on the risk of cancer and non-neoplastic diseases. However, the findings from these studies face limitations arising from confounding variables, reverse causation, and conflicting reports. Consequently, the establishment of a causal relationship between habitual glucosamine consumption and the risk of cancer and non-neoplastic diseases necessitates further investigation.MethodsFor Mendelian randomization (MR) investigation, we opted to employ single-nucleotide polymorphisms (SNPs) as instruments that exhibit robust associations with habitual glucosamine consumption. We obtained the corresponding effect estimates of these SNPs on the risk of cancer and non-neoplastic diseases by extracting summary data for genetic instruments linked to 49 varied cancer types amounting to 378,284 cases and 533,969 controls, as well as 20 non-neoplastic diseases encompassing 292,270 cases and 842,829 controls. Apart from the primary analysis utilizing inverse-variance weighted MR, we conducted two supplementary approaches to account for potential pleiotropy (MR-Egger and weighted median) and assessed their respective MR estimates. Furthermore, the results of the leave-one-out analysis revealed that there were no outlying instruments.ResultsOur results suggest divergence from accepted biological understanding, suggesting that genetically predicted glucosamine utilization may be linked to an increased vulnerability to specific illnesses, as evidenced by increased odds ratios and confidence intervals (95% CI) for diseases, such as malignant neoplasm of the eye and adnexa (2.47 [1.34–4.55]), benign neoplasm of the liver/bile ducts (2.12 [1.32–3.43]), benign neoplasm of the larynx (2.01 [1.36–2.96]), melanoma (1.74 [1.17–2.59]), follicular lymphoma (1.50 [1.06–2.11]), autoimmune thyroiditis (2.47 [1.49–4.08]), and autoimmune hyperthyroidism (1.93 [1.17–3.18]). In contrast to prior observational research, our genetic investigations demonstrate a positive correlation between habitual glucosamine consumption and an elevated risk of sigmoid colon cancer, lung adenocarcinoma, and benign neoplasm of the thyroid gland.ConclusionCasting doubt on the purported purely beneficial association between glucosamine ingestion and prevention of neoplastic and non-neoplastic diseases, habitual glucosamine ingestion exhibits dichotomous effects on disease outcomes. Endorsing the habitual consumption of glucosamine as a preventative measure against neoplastic and non-neoplastic diseases cannot be supported.

In the evolving landscape of higher education, particularly in the post-pandemic era, it is crucial for college students to face societal challenges and achieve success by understanding and predicting psychological resilience. To deepen our understanding of psychological resilience, this study used a decision tree model to explore influencing factors. We surveyed 776 college students and collected data on demographic information, self-esteem, sense of school belonging, pro-environmental behavior, subjective well-being, internet game addiction, life autonomy, and academic procrastination using several scales. The decision tree model identified eight key predictors of psychological resilience, which are as follows in order of importance: self-esteem, sense of school belonging, pro-environmental behavior, subjective well-being, academic procrastination, life autonomy, internet game addiction, and academic achievement. This model's accuracy reached 73.985 %, emphasizing its potential utility in educational settings. The findings not only provide a novel and data-driven perspective to understand psychological resilience in college students compared to existing research but also provide practical guidance for educational practitioners and policymakers on how to develop psychological resilience in college students.

The utilization of in-situ resources will be crucial for the survival of astronauts in space. Therefore, plants and crops will be important for humans in space as they serve as food, provide oxygen, and remove carbon dioxide, enhancing habitability. The aim of this research is to explore the growth of crops on celestial bodies prior to human arrival. The paper outlines the creation of a novel capsule by Green Moon Project (GMP) designed to meet essential criteria for monitoring and enhancing crop cultivation on the lunar terrain, tackling key obstacles such as self-propagation, fluctuating light patterns, water provision, and monitoring germination and growth stages. The Center of Space Exploration (hereafter COSE) managed to sprout the first seed on another celestial body during the Chang’e 4 mission on the Moon in January 2019. This achievement means an important step in space agriculture and widens the biological research of crops that will sustain future crewed missions and human bases in space. Space farming requires greater understanding if humans are to survive in space without constant contact from Earth and that is why GMP goals are aligned COSE’s. Therefore, GMP and COSE work in synergy to boost the research of space farming, future crops, habitability, and close controlled environmental systems. Due to the similarities between both projects, both teams decided to join efforts and cooperate in future space missions. To provide scientific support and technical solutions for future long-term crewed exploration missions, it is mandatory to conduct ground verification experiments of controllable extraterrestrial ecosystems.

Intraoperative remifentanil administration has been linked to increased postoperative pain sensitivity. Recent studies have identified the involvement of euchromatic histone-lysine N-methyltransferase 2 (Ehmt2/G9a) in neuropathic pain associated with the transcriptional silencing of many potassium ion channel genes. This study investigates whether G9a regulates the potassium sodium-activated channel subfamily T member 1 (Slo2.2) in remifentanil-induced post-incisional hyperalgesia (RIH) in rodents. We performed remifentanil infusion (1 μg·kg-1·min-1 for 60 min) followed by plantar incision to induce RIH in rodents. Our results showed that RIH was accompanied by increased G9a and H3K9me2 production and decreased Slo2.2 expression 48 h postoperatively. Deletion of G9a rescued Slo2.2 expression in DRG and reduced RIH intensity. Slo2.2 overexpression also reversed this hyperalgesia phenotype. G9a overexpression decreased Slo2.2-mediated leak current and increased excitability in the small-diameter DRG neurons and laminal II small-diameter neurons in the spinal dorsal horn, which was implicated in peripheral and central sensitization. These results suggest that G9a contributes to the development of RIH by epigenetically silencing Slo2.2 in DRG neurons, leading to decreased central sensitization in the spinal cord. The findings may have implications for the development of novel therapeutic targets for the treatment of postoperative pain.

Background: X-linked hypophosphatemia (XLH, OMIM 307800) is a rare phosphorus metabolism disorder caused by PHEX gene variants. Many variants simply classified as missense or nonsense variants were only analyzed at the DNA level. However, growing evidence indicates that some of these variants may alter pre-mRNA splicing, causing diseases. Therefore, this study aimed to use bioinformatics tools and a minigene assay to ascertain the effects of PHEX variations on pre-mRNA splicing.Methods: We analyzed 174 variants in the PHEX gene described as missense or nonsense variants. Finally, we selected eight candidate variants using bioinformatics tools to evaluate their effects on pre-mRNA splicing using a minigene assay system. The complementary DNA (cDNA) sequence for the PHEX gene (RefSeq NM_000444.6) serves as the basis for DNA variant numbering.Results: Of the eight candidate variants, three were found to cause abnormal splicing. Variants c.617T>G p.(Leu206Trp) and c.621T>A p.(Tyr207*) in exon 5 altered the splicing of pre-mRNA, owing to the activation of a cryptic splice site in exon 5, which produced an aberrant transcript lacking a part of exon 5, whereas variant c.1700G>C p.(Arg567Pro) in exon 16 led to the activation of a cryptic splice site in intron 16, resulting in a partial inclusion of intron 16.Conclusion: Our study employed a minigene system, which has a great degree of flexibility to assess abnormal splicing patterns under the circumstances of patient mRNA samples that are not available, to explore the impact of the exonic variants on pre-mRNA splicing. Based on the aforementioned experimental findings, we demonstrated the importance of analyzing exonic variants at the mRNA level.

BackgroundAccumulating evidence suggests that the autism spectrum disorder (ASD) population exhibits altered hormone levels, including androgens. However, studies on the regulation of androgens, such as testosterone and dehydroepiandrosterone (DHEA), in relation to sex differences in individuals with ASD are limited and inconsistent. We conducted the systematic review with meta-analysis to quantitatively summarise the blood, urine, or saliva androgen data between individuals with ASD and controls.MethodsA systematic search was conducted for eligible studies published before 16 January 2023 in six international and two Chinese databases. We computed summary statistics with a random-effects model. Publication bias was assessed using funnel plots and heterogeneity using I2 statistics. Subgroup analysis was performed by age, sex, sample source, and measurement method to explain the heterogeneity.Results17 case-control studies (individuals with ASD, 825; controls, 669) were assessed. Androgen levels were significantly higher in individuals with ASD than that in controls (SMD: 0.27, 95% CI: 0.06–0.48, P=0.01). Subgroup analysis showed significantly elevated levels of urinary total testosterone, urinary DHEA, and free testosterone in individuals with ASD. DHEA level was also significantly elevated in males with ASD.ConclusionAndrogen levels, especially free testosterone, may be elevated in individuals with ASD and DHEA levels may be specifically elevated in males.

A recombinant inbred line (RIL) population derived from wheat landrace Qingxinmai and breeding line 041133 exhibited segregation in resistance to powdery mildew and stripe rust in five and three field tests, respectively. A 16K genotyping by target sequencing (GBTS) single-nucleotide polymorphism (SNP) array-based genetic linkage map was used to dissect the quantitative trait loci (QTLs) for disease resistance. Four and seven QTLs were identified for adult-plant resistance (APR) against powdery mildew and stripe rust. QPm.caas-1B and QPm.caas-5A on chromosomes 1B and 5A were responsible for the APR against powdery mildew in line 041133. QYr.caas-1B, QYr.caas-3B, QYr.caas-4B, QYr.caas-6B.1, QYr.caas-6B.2, and QYr.caas-7B detected on the five B-genome chromosomes of line 041133 conferred its APR to stripe rust. QPm.caas-1B and QYr.caas.1B were co-localized with the pleiotropic locus Lr46/Yr29/Sr58/Pm39/Ltn2. A Kompetitive Allele Specific Polymorphic (KASP) marker KASP_1B_668028290 was developed to trace QPm/Yr.caas.1B. Four lines pyramiding six major disease resistance loci, PmQ, Yr041133, QPm/Yr.caas-1B, QPm.caas-2B.1, QYr.caas-3B, and QPm.caas-6B, were developed. They displayed effective resistance against both powdery mildew and stripe rust at the seedling and adult-plant stages.

This research explores the application of an Empathy Design Thinking (EDT) curriculum within primary education, guided by the principles of Experience, Empathy, Exploration, and Evaluation, to examine its effect on fostering student creativity in a Chinese context. The curriculum was redesigned into a streamlined, modular format for ease of integration into existing educational frameworks. Findings demonstrate a positive impact on students' creative thinking abilities, particularly in enhancing fluency and flexibility, with notable mentions of empathy's vital role in the educational process. This study aligns with global educational trends emphasizing the need for empathetic and comprehensive learning experiences, offering insights into the potential of EDT to enrich creative education for primary students, educators, curriculum developers, and policymakers.

Background: The treatment of patellar tendon injury has always been an unsolved problem, and mechanical characterization is very important for its repair and reconstruction. Elastin is a contributor to mechanics, but it is not clear how it affects the elasticity, viscoelastic properties, and structure of patellar tendon.Methods: The patellar tendons from six fresh adult experimental pigs were used in this study and they were made into 77 samples. The patellar tendon was specifically degraded by elastase, and the regional mechanical response and structural changes were investigated by: (1) Based on the previous study of elastase treatment conditions, the biochemical quantification of collagen, glycosaminoglycan and total protein was carried out; (2) The patellar tendon was divided into the proximal, central, and distal regions, and then the axial tensile test and stress relaxation test were performed before and after phosphate-buffered saline (PBS) or elastase treatment; (3) The dynamic constitutive model was established by the obtained mechanical data; (4) The structural relationship between elastin and collagen fibers was analyzed by two-photon microscopy and histology.Results: There was no statistical difference in mechanics between patellar tendon regions. Compared with those before elastase treatment, the low tensile modulus decreased by 75%–80%, the high tensile modulus decreased by 38%–47%, and the transition strain was prolonged after treatment. For viscoelastic behavior, the stress relaxation increased, the initial slope increased by 55%, the saturation slope increased by 44%, and the transition time increased by 25% after enzyme treatment. Elastin degradation made the collagen fibers of patellar tendon become disordered and looser, and the fiber wavelength increased significantly.Conclusion: The results of this study show that elastin plays an important role in the mechanical properties and fiber structure stability of patellar tendon, which supplements the structure-function relationship information of patellar tendon. The established constitutive model is of great significance to the prediction, repair and replacement of patellar tendon injury. In addition, human patellar tendon has a higher elastin content, so the results of this study can provide supporting information on the natural properties of tendon elastin degradation and guide the development of artificial patellar tendon biomaterials.

Abstract Evidence on the associations between patient’s disability and caregiver burden among Chinese family caregivers of individual living with schizophrenia is lacking. This study aimed at explore the underlying mechanisms between patient’s disability and caregiver burden among Chinese family caregivers of individual living with schizophrenia. A cross-sectional study was carried out in four Chinese cities (Wuhan, Changsha, Guangzhou, and Shenzhen), between April, 2021 and March, 2022. A total of 493 patients and their family caregivers were invited to report related data. The Zarit burden interview, WHODAS 2.0, the Potentially harmful behavior scale, the Affiliate Stigma Scale, and the Multidimensional Scale of perceived social support were used to collect data. Linear regression analysis and bootstrapping analysis were conducted. The adjusted regression results showed that patients’ disability (B = 0.616; 95% CI: 0.479–0.753), potentially harmful behavior on caregivers (B = 0.474; 95% CI: 0.232–0.716), and caregiver’s low social support (B = −0.079; 95% CI: −0.158– −0.002), high level of affiliate stigma (B = 13.045; 95% CI: 10.227–15.864) were associated with higher level of caregiver burden (p
