Abstract: Single-cell ribonucleic acid sequencing is becoming widely employed to study biological processes at a novel resolution depth. The ability to analyse transcriptomes of multiple heterogeneous cell types in parallel is especially valuable for cell-focused lung research where a variety of resident and recruited cells are essential for maintaining organ functionality. We compared the single-cell transcriptomes from publicly available and unpublished datasets of the lungs in six different species: human (Homo sapiens), African green monkey (Chlorocebus sabaeus), pig (Sus domesticus), hamster (Mesocricetus auratus), rat (Rattus norvegicus) and mouse (Mus musculus) by employing RNA velocity and intercellular communication based on ligand–receptor co-expression, among other techniques. Specifically, we demonstrated a workflow for interspecies data integration, applied a single unified gene nomenclature, performed cell-specific clustering and identified marker genes for each species. Overall, integrative approaches combining newly sequenced as well as publicly available datasets could help identify species-specific transcriptomic signatures in both healthy and diseased lung tissue and select appropriate models for future respiratory research.

Abstract: Natural Killer (NK) cells play a key role in cancer immunosurveillance. However, NK cells from cancer patients display an altered phenotype and impaired effector functions. In addition, evidence of a regulatory role for NK cells is emerging in diverse models of viral infection, transplantation, and autoimmunity. Here, we analyzed clear cell renal cell carcinoma (ccRCC) datasets from The Cancer Genome Atlas (TCGA) and observed that a higher expression of NK cell signature genes is associated with reduced survival. Analysis of fresh tumor samples from ccRCC patients unraveled the presence of a high frequency of tumor-infiltrating PD-L1+ NK cells, suggesting that these NK cells might exhibit immunoregulatory functions. In vitro, PD-L1 expression was induced on NK cells from healthy donors (HD) upon direct tumor cell recognition through NKG2D and was further up-regulated by monocyte-derived IL-18. Moreover, in vitro generated PD-L1hi NK cells displayed an activated phenotype and enhanced effector functions compared to PD-L1- NK cells, but simultaneously, they directly inhibited CD8+ T cell proliferation in a PD-L1-dependent manner. Our results suggest that tumors might drive the development of PD-L1-expressing NK cells that acquire immunoregulatory functions in humans. Hence, rational manipulation of these regulatory cells emerges as a possibility that may lead to improved anti-tumor immunity in cancer patients.

3. Gamifying quantitative face-to-face interviews in rural India: An empirical evaluation based on the basic psychological needs theory.

Abstract: ObjectiveStandardized face-to-face interviews are widely used in low and middle-income countries to collect data for social science and health research. Such interviews can be long and tedious. In an attempt to improve the respondents' experience of interviews, we developed a concept of gamified interview format by including a game element. Gamification is reported to increase engagement in tasks, but results from rigorously developed research are equivocal, and a theory of gamification is still needed.Materials & methodsWe evaluated the proposed gamification with a randomized controlled trial based on self-determination theory, specifically on the basic psychological needs theory. In total, 1266 respondents were interviewed. Single and multiple mediation analyses were used to understand the effects of the gamified interview format.ResultsOur evaluation showed that the gamification we had developed did not improve the outcome, the experience of the interview reported by respondent. The effect of the gamified interview format depended on the ability of respondents: gamification can be counterproductive if it overburdens the respondents. However, the basic psychological needs theory explained the mechanisms of action of gamification well: feeling competent and related to others improved the reported experience of the interview.ConclusionWe emphasize the need to develop context-specific gamification and invite researchers to conduct equivalently rigorous evaluations of gamification in future studies.

4. Chk1 and the Host Cell DNA Damage Response as a Potential Antiviral Target in BK Polyomavirus Infection

Abstract: The human BK polyomavirus (BKPyV) is latent in the kidneys of most adults, but can be reactivated in immunosuppressed states, such as following renal transplantation. If left unchecked, BK polyomavirus nephropathy (PyVAN) and possible graft loss may result from viral destruction of tubular epithelial cells and interstitial fibrosis. When coupled with regular post-transplant screening, immunosuppression reduction has been effective in limiting BKPyV viremia and the development of PyVAN. Antiviral drugs that are safe and effective in combating BKPyV have not been identified but would be a benefit in complementing or replacing immunosuppression reduction. The present study explores inhibition of the host DNA damage response (DDR) as an antiviral strategy. Immunohistochemical and immunofluorescent analyses of PyVAN biopsies provide evidence for stimulation of a DDR in vivo. DDR pathways were also stimulated in vitro following BKPyV infection of low-passage human renal proximal tubule epithelial cells. The role of Chk1, a protein kinase known to be involved in the replication stress-induced DDR, was examined by inhibition with the small molecule LY2603618 and by siRNA-mediated knockdown. Inhibition of Chk1 resulted in decreased replication of BKPyV DNA and viral spread. Activation of mitotic pathways was associated with the reduction in BKPyV replication. Chk1 inhibitors that are found to be safe and effective in clinical trials for cancer should also be evaluated for antiviral activity against BKPyV.

Abstract: Abstract Background This article presents the development of a school handwashing programme in two different sub-Saharan countries that applies the RANAS (risk, attitudes, norms, ability, and self-regulation) systematic approach to behaviour change. Methods Interviews were conducted with 669 children enrolled in 20 primary schools in Burundi and 524 children in 20 primary schools in Zimbabwe. Regression analyses were used to assess the influence of the RANAS behavioural determinants on reported handwashing frequencies. Results The results revealed that, in both countries, a programme targeting social norms and self-efficacy would be most effective. In Burundi, raising the children’s perceived severity of the consequences of contracting diarrhoea, and in Zimbabwe, increasing the children’s health knowledge should be part of the programme. Conclusions The school handwashing programme should create awareness of the benefits of handwashing through educational activities, raise the children’s ability and confidence in washing hands at school through infrastructural improvements, and highlight the normality of washing hands at school through events and poster creation.

Abstract: BackgroundThe success of HER2-positive (HER2+) breast cancer treatment with trastuzumab, an antibody that targets HER2, relies on immune response. We demonstrated that TNFα induces mucin 4 (MUC4) expression, which shields the trastuzumab epitope on the HER2 molecule decreasing its therapeutic effect. Here, we used mouse models and samples from HER2+ breast cancer patients to unravel MUC4 participation in hindering trastuzumab effect by fostering immune evasion.MethodsWe used a dominant negative TNFα inhibitor (DN) selective for soluble TNFα (sTNFα) together with trastuzumab. Preclinical experiments were performed using two models of conditionally MUC4-silenced tumors to characterize the immune cell infiltration. A cohort of 91 patients treated with trastuzumab was used to correlate tumor MUC4 with tumor-infiltrating lymphocytes.ResultsIn mice bearing de novo trastuzumab-resistant HER2+ breast tumors, neutralizing sTNFα with DN induced MUC4 downregulation. Using the conditionally MUC4-silenced tumor models, the antitumor effect of trastuzumab was reinstated and the addition of TNFα-blocking agents did not further decrease tumor burden. DN administration with trastuzumab modifies the immunosuppressive tumor milieu through M1-like phenotype macrophage polarization and NK cells degranulation. Depletion experiments revealed a cross-talk between macrophages and NK cells necessary for trastuzumab antitumor effect. In addition, tumor cells treated with DN are more susceptible to trastuzumab-dependent cellular phagocytosis. Finally, MUC4 expression in HER2+ breast cancer is associated with immune desert tumors.ConclusionsThese findings provide rationale to pursue sTNFα blockade combined with trastuzumab or trastuzumab drug conjugates for MUC4+ and HER2+ breast cancer patients to overcome trastuzumab resistance.

Abstract: Malignancies during pregnancy are usually treated similarly to the malignancies in non-pregnant patients. Results of controlled therapeutic trials are typically not available. However, registered studies associated with some of the most common malignancies exist. In case of ovarian cancer, due to its rarity – especially with the primary metastatic cases – only individual case reports are available. The article presents a case with par aortal abdominal, thoracic and cervical lymph node metastasis in a 33-years-old pregnant female. She was treated with primary systemic chemotherapy since early pregnancy. At 32 weeks of gestation, the patient gave birth to a girl (1460g) by caesarean section. Subsequently, the patient received 3 more cycles of chemotherapy with carboplatin/paclitaxel and bevacizumab for 30 months as well as letrozole maintenance therapy. Three years after the initial diagnosis, complete instrumental assessment showed unremarkable results without tumour recurrence.

Abstract: This study reports findings from a multisite cluster randomized controlled trial designed to validate and scale up an existing successful professional development program that uses a cognitive strategies approach to text-based analytical writing. The Pathway to Academic Success Project worked with partner districts affiliated with 4 National Writing Project (NWP) sites in southern California. Informed by a wide body of research on the efficacy of strategy instruction to enhance students’ academic literacy, the intervention aimed to help secondary school students, particularly Latinos and mainstreamed English learners, to develop the academic writing skills called for in the rigorous Common Core State Standards for English Language Arts. Two hundred thirty teachers from partner districts affiliated with the NWP sites were stratified by school and grade and then randomly assigned to the treatment or control group. Treatment teachers participated in 46 hrs of training and learned how to apply cognitive strategies by using an on-demand writing assessment to help students understand, interpret, and write analytical essays about nonfiction texts. Multilevel models revealed significant effects on a holistic measure of an on-demand writing assessment (d = .32) as well as on 4 analytic attributes: content (d = .31), structure (d = .29), fluency (d = .27), and conventions (d = .32). Abstract: Lipoteichoic acid (LTA) from Gram-positive bacteria exerts different immune effects depending on the bacterial source from which it is isolated. Lacticaseibacillus rhamnosus GG LTA (LGG-LTA) oral administration reduces UVB-induced immunosuppression and skin tumor development in mice. In the present work, we evaluate the immunomodulatory effect exerted by LGG-LTA in dendritic cells (DC) and T cells, both in vitro and in the gut-associated lymphoid tissue (GALT). During cell culture, LTA-stimulated BMDC increased CD86 and MHC-II expression and secreted low levels of pro and anti-inflammatory cytokines. Moreover, LTA-treated BMDC increased T cell priming capacity, promoting the secretion of IL-17A. On the other hand, in orally LTA-treated mice, a decrease in mature DC (lamina propria and Peyer’s patches) was observed. Concomitantly, an increase in IL-12p35 and IFN-γ transcription was presented (lamina propria and Peyer’s Patches). Finally, an increase in the number of CD103+ DC was observed in Peyer’s patches. Together, our data demonstrate that LGG-LTA activates DC and T cells. Moreover, we show that a Th1-biased immune response is triggered in vivo after oral LTA administration. These effects justify the oral LTA activity previously observed.

Abstract: Today's variety of photovoltaic (PV) technologies imposes new challenges to laboratories and industries to precisely measure the performance of devices and, consequently, to accurately estimate the energy yield once installed in a specific location. Spectroradiometry has become a key discipline for metrology applied to PV: Spectral irradiance is one of the three parameters according to which solar simulators are classified according to IEC 60904-9; precise spectrum measurements are a key factor in the spectral mismatch calculation. Finally, energy rating calculations according to IEC 61853 involve spectral irradiance conditions different than the AM1.5G standard spectrum. To tackle these issues, since 2011, the International Spectroradiometer Interlaboratory Comparison (ISRC) takes place annually in different locations of Europe with the participation of laboratories, research institutes, and industry partners to assess spectral measurement capabilities and share good measurement practices and protocols. In this paper, several results of the 9th ISRC 2019 are presented, looking in particular at the impact on characterization of new technologies like organic devices (OPV), dye-sensitized (DSSC), and perovskites.

Abstract: Escherichia coli pathotypes (i.e., enteropathogenic and enterotoxigenic) have been identified among the pathogens most responsible for moderate-to-severe diarrhea in low- and middle-income countries (LMICs). Pathogenic E. coli are transmitted from infected human or animal feces to new susceptible hosts via environmental reservoirs such as hands, water, and soil. Commensal E. coli, which includes nonpathogenic E. coli strains, are widely used as fecal bacteria indicator, with their presence associated with increased likelihood of enteric pathogens and/or diarrheal disease. In this study, we investigated E. coli contamination in environmental reservoirs within households (N = 142) in high-population density communities of Harare, Zimbabwe. We further assessed the interconnectedness of the environmental compartments by investigating associations between, and household-level risk factors for, E. coli contamination. From the data we collected, the source and risk factors for E. coli contamination are not readily apparent. One notable exception is the presence of running tap water on the household plot, which is associated with significantly less E. coli contamination of drinking water, handwashing water, and hands after handwashing. In addition, E. coli levels on hands after washing are significantly associated with handwashing water contamination, hand contamination before washing, and diarrhea incidence. Finally, we observed that animal ownership increases E. coli contamination in soil, and E. coli in soil are correlated with contamination on hands before washing. This study highlights the complexity of E. coli contamination in household environments within LMICs. More, larger, studies are needed to better identify sources and exposure pathways of E. coli—and enteric pathogens generally—to identify effective interventions.

Abstract: Rationale Consistent hand hygiene prevents diarrheal and respiratory diseases, but it is often not practiced. The disease burden is highest in low-income settings, which need effective interventions to promote domestic handwashing. To date, most handwashing campaigns have focused on promoting frequent handwashing at key times, whereas specifically promoting handwashing techniques proven to be effective in removing microbes has been confined to healthcare settings. Methods We used a cluster-randomized, factorial, controlled trial to test the effects of two handwashing interventions on the behavior of primary caregivers in Harare, Zimbabwe. One intervention targeted caregivers directly, and the other targeted them through their children. Outcome measures were surveyed at baseline and six weeks’ follow-up and included observed handwashing frequency and technique and fecal hand contamination before and after handwashing. Results Combining the direct and indirect interventions resulted in observed handwashing with soap at 28% of critical handwashing times, while the corresponding figure for the non-intervention control was 5%. Observed handwashing technique, measured as the number of correctly performed handwashing steps, increased to an average of 4.2, while the control averaged 3.4 steps. Demonstrated handwashing technique increased to a mean of 6.8 steps; the control averaged 5.2 steps. No statistically significant group differences in fecal hand contamination before or after handwashing were detected. Conclusions The results provide strong evidence that the campaign successfully improved handwashing frequency and technique. It shows that the population-tailored design, based on social-cognitive theory, provides effective means for developing powerful interventions for handwashing behavior change. We did not find evidence that children acted as strong agents of handwashing behavior change. The fact that the microbial effectiveness of handwashing did not improve despite strong improvements in handwashing technique calls for critical evaluation of existing handwashing recommendations. The aim of future handwashing campaigns should be to promote both frequent and effective handwashing.

Abstract: This paper presents results of an all-sky search for periodic gravitational waves in the frequency range [50, 1 190] Hzand and with frequency derivative range of approx. [-20,1.1] x 10(exp-10) H(sub 0)z s(exp-1) for the fifth LIGO science run (S5). The search uses a noncoherent Hough-transform method to combine the information from coherent searches on time scales of about one day. Because these searches are very computationally intensive, they have been carried out with the Einstein@Home volunteer distributed computing project. Postprocessing identifies eight candidate signals; deeper follow-up studies rule them out. Hence, since no gravitational wave signals have been found, we report upper limits on the intrinsic gravitational wave strain amplitude ho. For example, in the 0.5 Hz-wide band at 152.5 Hz, we can exclude the presence of signals with h(exp 0) greater than 7.6 x 10(exp-25) at a 90% confidence level. This search is about a factor 3 more sensitive than the previous Einstein@Home search of early S5 LIGO data.

Abstract: The human BK polyomavirus (BKPyV) is latent in the kidneys of most adults, but can be reactivated in immunosuppressed states, such as following renal transplantation. If left unchecked, BK polyomavirus nephropathy (PyVAN) and possible graft loss may result from viral destruction of tubular epithelial cells and interstitial fibrosis. When coupled with regular post-transplant screening, immunosuppression reduction has been effective in limiting BKPyV viremia and the development of PyVAN. Antiviral drugs that are safe and effective in combating BKPyV have not been identified but would be a benefit in complementing or replacing immunosuppression reduction. The present study explores inhibition of the host DNA damage response (DDR) as an antiviral strategy. Immunohistochemical and immunofluorescent analyses of PyVAN biopsies provide evidence for stimulation of a DDR in vivo. DDR pathways were also stimulated in vitro following BKPyV infection of low-passage human renal proximal tubule epithelial cells. The role of Chk1, a protein kinase known to be involved in the replication stress-induced DDR, was examined by inhibition with the small molecule LY2603618 and by siRNA-mediated knockdown. Inhibition of Chk1 resulted in decreased replication of BKPyV DNA and viral spread. Activation of mitotic pathways was associated with the reduction in BKPyV replication. Chk1 inhibitors that are found to be safe and effective in clinical trials for cancer should also be evaluated for antiviral activity against BKPyV.

Abstract: Background Consistent domestic hand hygiene can reduce diarrhea-related morbidity and mortality and the spread of other communicable diseases. However, it remains uncertain which technique of handwashing is most effective and practicable during everyday life. The goal of this study is to determine how the handwashing technique, as performed in the daily life by the participants of this case study in Harare, Zimbabwe, influences microbial handwashing effectiveness. Methods Handwashing technique of 173 primary caregivers was observed in their homes and hand rinse samples were collected before and after handwashing. Samples were analyzed for Escherichia coli and total coliform concentrations. Generalized linear models were used to predict fecal hand contamination after washing from observed handwashing technique. Results Cleaning under fingernails, scrubbing the fingertips, using soap, and drying hands through rubbing on clothes or a clean towel statistically significantly reduced E coli contamination of hands after washing. Tap use, scrubbing fingertips, and rubbing hands on clothes to dry them statistically significantly reduced total coliform contamination. Conclusions Recommendations for effective and practicable domestic handwashing in Harare, Zimbabwe, should include performing specific handscrubbing steps (ie, cleaning under the fingernails and rubbing the fingertips), and soap and tap use. This calls for further research to develop behavior change interventions that explicitly promote effective handwashing technique at critical times.

Abstract: Main text The EURAMET.PR-S5 intercomparison of the short circuit current of reference solar cells at standard test conditions was carried out as star type comparison. In total 6 participants with seven facilities took part, 1 from Europe (PTB/Germany with two facilities: the established DSR facility as pilot lab and the new Laser-DSR facility as participant) and 5 outside Europe (NIM/China, VNIIOFI/Russia, KRISS/South Korea, A-STAR/Singapore, ITRI/Chinese Taipei). Therefore, this comparison can be considered as a worldwide one as all NMIs world-wide that had built up facilities for the primary ISTC-calibration of reference solar cells when the measurements started, participated. For the calculation of the reference value of the supplementary comparison no participant had to be excluded and the used weighted mean with cut-off has been supported by all participants. The unilateral Degrees of Equivalence (DoE) calculated for each participant are fully 100 % consistent with their standard uncertainties. The largest En-value is 0.61 and therefore significant smaller than 1. The chi-square test is fully satisfied with 2.09 < χ2(5) = 11.07. Thus, the DSR method is highly suitable for the calibration of reference solar cells according to the short circuit current at standard test conditions. Abstract: This paper reports on the results of the fifth spectral irradiance measurement intercomparison and the impact these results have on the spread of spectral mismatch calculations in the outdoor characterization of reference solar cell and photovoltaic (PV) devices. Ten laboratories and commercial partners with their own instruments were involved in the comparison. Solar spectral irradiance in clear sky condition was measured with both fast fixed and slow rotating grating spectroradiometers. This paper describes the intercomparison campaign, describes different statistical analysis used on acquired data, reports on the results, and analyzes the impact these results would have on the primary calibration of a c-Si PV reference cell under natural sunlight.

Abstract: Recent years have seen unparalleled efforts by the Swatchh Bharat Mission (SBM) to make India open defecation free. While latrine coverage has been boosted very successfully, latrine use has remained low in many areas of the country. Consequently, the aim of this study was to use robust psychological theory to develop and rigorously evaluate low-cost and scalable behaviour change interventions to promote latrine use in rural India. This study reports findings from a cluster-randomized controlled trial (N = 1945) conducted in rural Karnataka, India, from January 2017 to February 2019. The evaluated behaviour change interventions were developed using the risks, attitudes, norms, abilities, and self-regulation (RANAS) approach. Results showed that latrine use changed by more than 15% in both treatment and control arms. The intervention triggered an additional, statistically significant increase in latrine use of approximately 5% to reach 97% use at endline. The results suggest that external factors had a strong influence on latrine use, with intensive efforts by SBM likely to be among these. The added value of the campaign was to increase latrine use to almost complete uptake and to successfully tackle the most change-resistant individuals. This intervention or selected components could complement future latrine use promotion in India.

Abstract: Traceable measurements of PV devices is essential ensuring worldwide comparability of PV products. However, a continuously growing market and increasing diversity of PV products and their application raises the demand for calibrations and testing services beyond the well-established standard test conditions (STC). Examples for this development is energy rating of PV modules and the operation of solar cells in space. The PV-calibration laboratory at PTB meets this demand by extending its calibration services using the differential spectral responsivity (DSR) method with expanded uncertainties of 0,4% to 0,7% (k=2).

Abstract: Bradykinin-mediated, drug-induced edema like ACE-inhibitor-induced angioedema (ACEi AE) is almost exclusively located in the head and neck region and is potentially life threatening. To date, there are no guidelines or officially-approved treatments available for this pathology.We sought to provide a structured therapeutic algorithm for the acute treatment of drug-induced bradykinin-mediated angioedema.We analyzed data (especially the course of disease and therapy) of all patients with acute angioedema, who presented to the Department of Otorhinolaryngology, Head and Neck Surgery at the University of Ulm (2010-2015). We also conducted a literature review on PubMed with the terms "acute angioedema", "angioedema emergency", "ACE angioedema", "bradykinin angioedema" and "angioedema therapy". Other fundamental references were the recent German guidelines "hereditary angioedema", "anaphylaxis" and "airway management".An emergency algorithm was generated as a flowchart for the acute therapy of bradykinin-mediated drug-induced angioedema was generated. We focused on the decision criteria for intubation/airway management and pharmacological therapy: antihistamines and glucocorticoids versus anti-bradykinin treatment. Furthermore, recommendations for inpatient monitoring have been derived.To date, therapy of drug-induced bradykinin-mediated angioedema is performed according to an "off-label" use and without officially-approved guidelines. The presented emergency algorithm provides a first approach for a structured therapeutic concept for a potentially life-threatening pathology.

Abstract: The influences of varying charge storage capabilities of EEPROM transistors in a MemFlash configuration are presented. The effect a thinning of the tunnelling oxide has on the retention and the corresponding hysteretic current–voltage curves of MemFlash cells is investigated through measurements as well as simulations. Furthermore, the variation of charging and discharging voltages along with different cycle frequencies is explored in respect to the change in hysteretic behavior. Finally, the influences of changing device parameters on the behavior as artificial synapses were investigated by emulating LTP for different MemFlash cells. Here, we found a strong dependency of the learning rate and the memory capabilities from the tunnel oxide thickness, which allows flexible application in neuromorphic computing schemes.

Abstract: An intercomparison of terrestrial photovoltaic (PV) calibrations was performed among a number of European calibration and testing laboratories that participated in the European Metrology Research Program (EMRP) project “PhotoClass”. The purpose of this intercomparison was to evaluate the comparability of calibration and testing services within the stated uncertainties of the individual laboratories. The calibration objects were two world photovoltaic scale (WPVS)-type reference solar cells, one made from crystalline silicon and one made from GaAs. The calibration value (CV) was the short-circuit current under standard test conditions (ISTC). In conclusion, it was found that the CVs are all consistent within the stated uncertainties. This result strengthens the reliance in the calibration chain and in the PV calibration infrastructure in Europe.

Abstract: Spectroradiometry is a key metrological topic for accurate testing of photovoltaic (PV) devices, particularly relevant both for indoor testing on solar simulators and for outdoor testing. The relevance of accurate measurements of solar spectral irradiance has led the most renowned European solar PV test centres to take part to a series of International Spetroradiometer Intercomparisons that has taken place every year so far since 2011 in various localities in the Mediterranean Basin. This paper revisits the performance of participant laboratories and highlights the importance of inter-laboratory comparisons, showing the possible improvements in measurement reproducibility. © 2018 IEEE.

Abstract: Periprosthetic joint infection is a rare and devastating complication. Management of this complication often requires a multidisciplinary approach similar to that used for the care of patients with cancer. Several studies have reported better outcomes following total joint arthroplasties performed at specialized hospitals than those performed at general hospitals. Specialized institutions use care pathways that aid the multidisciplinary team in decision making. During the recent Musculoskeletal Infection symposium, specific issues were discussed with regard to the treatment of periprosthetic joint infection, including medical optimization, systematic approaches to infection management, and the importance of establishing registries to aid in the creation of Centers of Excellence. A Center of Excellence in periprosthetic infection could provide better overall outcomes with lower financial, physical, and emotional costs to patients.

Abstract: A completely newly designed multi-functional facility for the primary calibration of reference solar cells and the spectral characterization of all solar cell types has been developed and built at PTB. The new facility is based on the successfully applied Differential Spectral Responsivity (DSR) method that allows the determination of the absolute spectral responsivity and nonlinearity of solar cells with the lowest uncertainties. By using a tunable laser system, the new setup avoids the main problem of monochromator-based systems: the low optical power level of the monochromatic beam. Thus it enables a significant reduction of the uncertainty for the short circuit current under standard test conditions ISTC of solar cells. It enables the calibration of World Photovoltaic Scale (WPVS) reference solar cells with an uncertainty of 0.4% (k = 2), the lowest value stated by any WPVS laboratory. 2013 The Authors. Due to the high market volume of solar installations of 50 billion US dollars per year, an uncertainty of 1% in efficiency measurement causes an uncertainty of 500 million US dollars per year in the product value. Hence PTB has repeatedly been approached with the request of a lower measurement uncertainty, even though PTB – as a qualified World Photovoltaic Scale (WPVS) laboratory – already belongs to the institutes providing the lowest uncertainty for the primary calibration of reference solar cells worldwide while being traceable to the SI. Abstract: Many differentiated plant cells can dedifferentiate into stem cells, reflecting the remarkable developmental plasticity of plants. In the moss Physcomitrella patens, cells at the wound margin of detached leaves become reprogrammed into stem cells. Here, we report that two paralogous P. patens WUSCHEL-related homeobox 13-like (PpWOX13L) genes, homologs of stem cell regulators in flowering plants, are transiently upregulated and required for the initiation of cell growth during stem cell formation. Concordantly, Δppwox13l deletion mutants fail to upregulate genes encoding homologs of cell wall loosening factors during this process. During the moss life cycle, most of the Δppwox13l mutant zygotes fail to expand and initiate an apical stem cell to form the embryo. Our data show that PpWOX13L genes are required for the initiation of cell growth specifically during stem cell formation, in analogy to WOX stem cell functions in seed plants, but using a different cellular mechanism.

42. Microwave conductance and electrochemical characterization of Si/a-SiN :H heterojunctions in contact to aqueous electrolyte

Abstract: In the present work, electrochemical impedance measurements in combination with conductance measurements in the microwave frequency range were applied to semiconductor/insulator heterojunctions in an electrochemical cell configuration. It is shown that the combination of both techniques yields a more reliable and detailed interpretation of the experimental results. The Si/a-SiNx:H interface was characterized by three different methods. Quasi-stationary photoconductance measurements in combination with photocurrent measurements were performed as function of the external cell polarization. Potential modulated conductance and impedance measurements were used to characterize the electrical properties of the Si/a-SiNx:H interface. The reliability of the Mott–Schottky procedure was successfully tested for two different electrochemical cells. Transient photoconductance measurements as a function of the applied potential were used to determine the surface recombination kinetics at the active interface. The experimental results led to the conclusion that the fixed charge in a-SiNx:H is responsible for the electrical passivation of Si wafer. Furthermore, it was observed that the electronic configuration of this interface presents certain instabilities when driven in aqueous media.

Abstract: Handwashing has been shown to considerably reduce diarrhea morbidity and mortality. To decontaminate hands effectively, the use of running water, soap, and various scrubbing steps are recommended. This study aims to identify the behavioral determinants of effective handwashing. Everyday handwashing technique of 434 primary caregivers in high-density suburbs of Harare, Zimbabwe, was observed and measured as an 8-point sum score of effective handwashing technique. Multiple linear and logistic regression analyses were performed to predict observed handwashing technique from potential contextual and psychosocial determinants. Knowledge of how to wash hands effectively, availability of a handwashing station with functioning water tap, self-reported frequency of handwashing, perceived vulnerability, and action planning were the main determinants of effective handwashing technique. The models were able to explain 39% and 36% of the variance in overall handwashing technique and thoroughness of handscrubbing. Memory aids and guided practice are proposed to consolidate action knowledge, and personalized risk messages should increase the perceived vulnerability of contracting diarrhea. Planning where, when, and how to maintain a designated place for handwashing with sufficient soap and water is proposed to increase action planning. Since frequent self-reported handwashing was associated with performing more effective handwashing technique, behavior change interventions should target both handwashing frequency and technique concurrently.

Abstract: Image analysis problems such as feature tracking, edge detection, image enhancement, or texture analysis require the detection of multi-oriented patterns which can appear at arbitrary orientations. Direct rotated matched filtering for feature detection is computationally expensive, but can be sped up with steerable filters. So far, steerable filter approaches were limited to only one direction. Many important low-level image features are, however, characterized by more than a single orientation. We therefore present here a framework for efficiently detecting specific multi-oriented patterns with arbitrary orientations in grayscale images. The core idea is to construct multisteerable filters by appropriate combinations of single-steerable filters. We exploit that steerable filters are closed under addition and multiplication. This allows to derive a design guide for multisteerable filters by means of multivariate polynomials. Furthermore, we describe an efficient implementation scheme and discuss the use of weighting functions to reduce angular oscillations. Applications in camera calibration, junction analysis of images from plant roots, and the discrimination of L, T, and X-junctions demonstrate the potential of this approach.

Abstract: The placenta is a major barrier that prevents potentially infectious agents from causing fetal diseases or related complications during pregnancy. Therefore, we postulated that the placenta might express a broad repertoire of antimicrobial proteins as well as inflammatory chemokines and cytokines to combat invading microorganisms. Here we demonstrate that placental cells indeed express a wide range of AMPs (antimicrobial peptides and proteins) including bactericidal/permeability-increasing protein (BPI), secretory leukocyte protease inhibitor (SLPI), human β-defensin 2 (hBD2), acyloxyacyl hydrolase (AOAH), and cathelicidin (CAP18). In addition, these cells also secrete pro-inflammatory cytokines and chemokines upon stimulation with bacterial ligands. Notably, we show that BPI expression by placental cells could be completely attributed to granulocytes while highly purified placental trophoblasts expressed only a subset of the AMPs like SLPI. Unexpectedly, trophoblast AMPs did not exhibit inducible secretion in response to various TLR ligands and further investigations showed that the unresponsiveness of trophoblasts to lipopolysaccharide (LPS) was due to a lack of TLR4 expression. In summary, we have shown that the expression of different AMPs can be allocated to various cells in the placenta and the repertoire of the AMPs expressed by placental cells is a result of a cooperation of leukocytes as well as cells from embryonic origin.

Abstract: Estimating the dynamics of furbearer populations is challenging because their elusive behavior and low densities make observations difficult. Statistical population reconstruction is a flexible approach to demographic assessment for harvested populations, but the technique has not been applied to furbearers. We extended this approach to furbearers and analyzed 8 yr of age-at-harvest data for American marten (Martes americana) in the Upper Peninsula of Michigan. Marten abundance estimates showed a general downward trend from an estimate of = 1,733.3 ( = 861.3) animals in 2000 to =1,163.9 ( = 520.1) in 2007. The harvest probability of martens increased nearly 5-fold from 0.0542 ( = 0.0250) in 2000 to 0.2637 ( = 0.1154) in 2007, which corresponded to a 5-fold increase in trap-nights. Continued monitoring of martens in the Upper Peninsula, Michigan, and a reassessment of current harvest regulations are necessary given the estimated decreases.
