3. Got Sick of Surveys or Lack of Social Capital? An Investigation on the Effects of the COVID-19 Lockdown on Institutional Surveying

- Abstract
The use of surveys to better understand students' experiences and teaching quality in higher education has a long history of implementation and critical review. Although research on student feedback surveys has highlighted a number of areas of concern, a well-designed student survey on teaching and learning will produce a strong foundation of evidence to inform action by academics and universities. Prior to the pandemic of 2020, analysis of the student surveys on teaching and learning highlighted lower rates of participation than in previous decades. This article examines the impacts of COVID-19 on the uptake of student surveys on teaching and learning within an Australian university. While students are affected by survey fatigue, some are more affected than others, especially those who are suffering from lack of academic and peer interaction. Using Bourdieu's concept of social capital, this article finds that first year students who started university during COVID-19 had a higher drop in response rate than other cohorts, highlighting the importance of interactions with peers when completing surveys. This article also complements the expanding literature on the impacts of COVID-19 on the experiences of higher education students.

Carrier-protein-dependent metabolic pathways biosynthesize fatty acids, polyketides, and non-ribosomal peptides, producing metabolites with important pharmaceutical, environmental, and industrial properties. Recent findings demonstrate that these pathways rely on selective communication mechanisms involving protein-protein interactions (PPIs) that guide enzyme reactivity and timing. While rational design of these PPIs could enable pathway design and modification, this goal remains a challenge due to the complex nature of protein interfaces. Computational methods offer an encouraging avenue, though many score functions fail to predict experimental observables, leading to low success rates. Here, we improve upon the Rosetta score function, leveraging experimental data through iterative rounds of computational prediction and mutagenesis, to design a hybrid fatty acid-non-ribosomal peptide initiation pathway. By increasing the weight of the electrostatic score term, the computational protocol proved to be more predictive, requiring fewer rounds of iteration to identify mutants with high in vitro activity. This allowed efficient design of new PPIs between a non-ribosomal peptide synthetase adenylation domain, PltF, and a fatty acid synthase acyl carrier protein, AcpP, as validated by activity and structural studies. This method provides a promising platform for customized pathway design, establishing a standard for carrier-protein-dependent pathway engineering through PPI optimization.

7. Evaluating the Research Productivity of a State University in Central Luzon, Philippines: Basis for Policy Recommendations

- Abstract
This descriptive study evaluated the research productivity of a state university in Central Luzon, Philippines, for the past five years (2016-2020) as basis for policy formulation. The study used document analysis to ascertain the research productivity in terms of: (1) Number of papers published in refereed international journals such as Scopus and Commission on Higher Education (CHED) accredited journals; (2) Number of faculty researchers with publication to these journals; and (3) Total number of citations. The data were obtained primarily from online publications found in the Google Scholar and Scopus databases. Results revealed that the state university's research productivity is relatively high in terms of published papers in refereed international journals for the past five years. However, the published papers in Scopus-indexed journals and CHED accredited journals are relatively low. An average number of faculty researchers publish their works in reputable journals, but very few faculties publish in the journals recommended by the CHED. Likewise, the university had a remarkable research citation record for the past five years. The findings of this research have important implications for policy to improve research productivity and enhance the research culture in higher education institutions (HEIs). Such policies include the need to have strong support to faculty researchers, forge research collaborations, source external research funding, and establish a sound incentive mechanism.

Abstract Background While pluripotent stem cell (PSC) therapies move toward clinical and commercial applications at a rapid rate, manufacturing reproducibility and robustness are notable bottlenecks in regulatory approval. Therapeutic applications of PSCs require large cell quantities to be generated under highly robust, well-defined, and economically viable conditions. Small-scale and short-term process optimization, however, is often performed in a linear fashion that does not account for time needed to verify the bioprocess protocols and analysis methods used. Design of a reproducible and robust bioprocess should be dynamic and include a continuous effort to understand how the process will respond over time and to different stresses before transitioning into large-scale production where stresses will be amplified. Methods This study utilizes a baseline protocol, developed for the short-term culture of PSC aggregates in Vertical-Wheel® bioreactors, to evaluate key process attributes through long-term (serial passage) suspension culture. This was done to access overall process robustness when performed with various commercially available media and cell lines. Process output variables including growth kinetics, aggregate morphology, harvest efficiency, genomic stability, and functional pluripotency were assessed through short and long-term culture. Results The robust nature of the expansion protocol was demonstrated over a six-day culture period where spherical aggregate formation and expansion were observed with high-fold expansions for all five commercial media tested. Profound differences in cell growth and quality were revealed only through long-term serial expansion and in-vessel dissociation operations. Some commercial media formulations tested demonstrated maintenance of cell growth rates, aggregate morphology, and high harvest recovery efficiencies through three bioreactor serial passages using multiple PSC lines. Exceptional bioprocess robustness was even demonstrated with sustained growth and quality maintenance over 10 serial bioreactor passages. However, some commercial media tested proved less equipped for serial passage cultures in bioreactors as cultures led to cell lysis during dissociation, reduction in growth rates, and a loss of aggregate morphology. Conclusions This study demonstrates the importance of systematic selection and testing of bioprocess input variables, with multiple bioprocess output variables through serial passages to create a truly reproducible and robust protocol for clinical and commercial PSC production using scalable bioreactor systems.

9. Association between pelvic lymph node dissection and survival among patients with prostate cancer treated with radical prostatectomy

Introduction: Although the clinical benefits of pelvic lymph node dissection (PLND) at the time of radical prostatectomy for prostate cancer remain uncertain, major guidelines recommend PLND based on risk profile. Thus, the objective of this study was to examine the association between PLND and survival among patients undergoing RP stratified by Gleason grade group (GG) with the aim of allowing patients and physicians to make more informed care decisions about the potential risks and benefits of PLND. Materials and methods: From the SEER-17 database, we examined overall (OS) and prostate cancer-specific (PCSS) survival of prostate cancer patients who underwent RP from 2010 to 2015 stratified by GG. We applied propensity score matching to balance pre-operative characteristics including race, age, PSA, household income, and housing status (urban/rural) between patients who did and did not undergo PLND for each GG. Statistical analyses included log-rank test and Kaplan-Meier curves. Results: We extracted a matched cohort from 80,287 patients with GG1-5 who underwent RP. The median PSA value was 6.0 ng/mL, and the median age was 62-years-old. 49,453 patients underwent PLND (61.60%), while 30,834 (38.40%) did not. There was no difference in OS and PCSS between patients who received PLND and those who did not for all Gleason GG (OS–GG1: P = 0.20, GG2: P = 0.34, GG3: P > 0.05, GG4: P = 0.55, GG5: P = 0.47; PCSS–GG1: P = 0.11, GG2: P = 0.96, GG3: P = 0.81, GG4: P = 0.22, GG5: P = 0.14). Conclusions: In this observational study, PLND at the time of RP was not associated with improved OS or PCSS among patients with cGS of 3 + 3, 3 + 4, 4 + 3, 4 + 4, 4 + 5, and 5 + 4. These findings suggest that until definitive clinical trials are completed, prostate cancer patients who have elected RP should be appropriately counseled on the potential risks and lack of proven survival benefit of PLND.

10. Integration of Environmental Education (EE) in Teacher Education Programs: Toward Sustainable Curriculum Greening

Environmental education (EE) is vital in addressing global environmental problems. Educational institutions are integrating EE into their curricula, but studies indicated the need for its intensification. This mixed method study assessed the integration of EE in Teacher Education Programs (TEPs) in Region III, Philippines. Respondents were 10 program deans, 10 chairs, 171 teachers, and 344 students. Data were gathered using validated questionnaires. Findings revealed that the New Teacher Education Curriculum (NTEC) integrated EE in program outcomes and performance indicators; in mandated courses -Physical Education (PE) and National Service Training Program (NSTP); and in Science Technology and Society (STS), a General Education (GE) course. Likewise, teachers integrated EE in their lessons and in community extension programs. Seminars/webinars included environmental care and promoted environmental awareness in social media. As to support for EE integration, administrative, physical facilities and financial support were provided. On student outcomes, positive findings were shown on attitude towards environment care, but application of knowledge and skills needs improvement. Students' knowledge was significantly correlated to attitude (Pearson's r value is 0.593 and p value <0.05) as well as skills acquisition and attitude (Pearson's r value is 0.647 and p value <0.05). Teachers' challenges in the EE integration included inadequate environmental knowledge and lack of time to lead students in environmental action. The study concludes that EE is integrated into the TEPs but the outcomes on the learners' knowledge and skills acquisition still need strengthening. Also, EE should be integrated in professional education courses, not only in GE and mandated courses.

Background: Public health threats can significantly impact mass gatherings and enhancing surveillance systems would thus be crucial. Epidemic Intelligence from Open Sources (EIOS) was introduced to Qatar to complement the existing surveillance measures in preparation to the FIFA World Cup Qatar 2022 (FWC22). This study estimated the empirical probability of EIOS detecting signals of public health relevance. It also looked at the factors responsible for discerning a moderate-high risk signal during a mass gathering event. Methods: This cross-sectional descriptive study used data collected between November 8th and December 25th, 2022, through an EIOS dashboard that filtered open-source articles using specific keywords. Triage criteria and scoring scheme were developed to capture signals and these were maintained in MS Excel. EIOS’ contribution to epidemic intelligence was assessed by the empirical probability estimation of relevant public health signals. Chi-squared tests of independence were performed to check for associations between various hazard categories and other independent variables. A multivariate logistic regression evaluated the predictors of moderate-high risk signals that required prompt action. Results: The probability of EIOS capturing a signal relevant to public health was estimated at 0.85 % (95 % confidence interval (CI) [0.82 %−0.88 %]) with three signals requiring a national response. 14. Moving toward a Digital Competency-Based Approach in Applied Education: Developing a System Supported by Blockchain to Enhance Competency-Based Credentials

- Abstract
A competency-based approach to education (CBE) has emerged in recent years, fostering curriculum by tracking and indicting students' acquired skills and competencies. Since applied education is moving away from a theoretical approach to its application, employers are eager to be empowered with graduates' full e-profiles, which demonstrate candidates' competency-based strengths and weaknesses. This study considered a new digital system for competency-based learning, enhanced by Blockchain and badge technologies, to improve and indicate practical classes' quality in applied programs. Our core objectives were to promote the digitalization of competency-based education and students' e-portfolios as a proposed system in applied education. We also assessed its implementation, beginning with a learning gap analysis and moving on to discuss the digital CBE to support employers' ability to validate graduates' competency-based credentials acquired through their signature learning experience. We found that the digitalization of skills and competency-based credentials should be enhanced to foster knowing-by-doing and practical capabilities, which should be incorporated in applied education to achieve optimum CBE results and support recruitment and professional development processes. Further research and study are recommended to develop and unify standards adopted by the Higher Education Institutions (HEIs), that are recognized by the industries.

The primary objective of this study is to review and characterize research-based music assessments that might be used to guide and assist teachers and students in transforming their attitudes and perceptions, ability to express their ideas and concepts during the assessment process openly. This study employed descriptive and autoethnographic methods to elicit information about current conditions and practices through the library approach and literature review. The identified music assessment techniques were used to determine how well students comprehend important points in a session. It includes the Music Concept Memory Exercise and Instrumental and Vocal Schematic Processing for music lesson instructions and the Five-Letter Name Pitch Memory Test and Three-Chord Familiarization Assessment for basic instrumental music instructions. They are used as a core component deliberately designed to expose classroom teachers and prospective teachers to the essentials for effective instructional practices. Effective evaluation ensures that learners' knowledge, understanding, and skills are transferred effectively in future contexts.

Non-ribosomal peptides play a critical role in the clinic as therapeutic agents. To access more chemically diverse therapeutics, non-ribosomal peptide synthetases (NRPSs) have been targeted for engineering through combinatorial biosynthesis; however, this has been met with limited success in part due to the lack of proper protein-protein interactions between non-cognate proteins. Herein, we report our use of chemical biology to enable X-ray crystallography, molecular dynamics (MD) simulations, and biochemical studies to elucidate binding specificities between peptidyl carrier proteins (PCPs) and adenylation (A) domains. Specifically, we determined X-ray crystal structures of a type II PCP crosslinked to its cognate A domain, PigG and PigI, and of PigG crosslinked to a non-cognate PigI homologue, PltF. The crosslinked PCP-A domain structures possess large protein-protein interfaces that predominantly feature hydrophobic interactions, with specific electrostatic interactions that orient the substrate for active site delivery. MD simulations of the PCP-A domain complexes and unbound PCP structures provide a dynamical evaluation of the transient interactions formed at PCP-A domain interfaces, which confirm the previously hypothesized role of a PCP loop as a crucial recognition element. Finally, we demonstrate that the interfacial interactions at the PCP loop 1 region can be modified to control PCP binding specificity through gain-of-function mutations. This work suggests that loop conformational preferences and dynamism account for improved shape complementary in the PCP-A domain interactions. Ultimately, these studies show how crystallographic, biochemical, and computational methods can be used to rationally re-engineer NRPSs for non-cognate interactions.

Since the beginning of the coronavirus disease 2019 (COVID-19) pandemic, numerous severe acute respiratory syndrome coronavirus 2 (SARS-CoV-2) variants have emerged, some leading to large increases in infections, hospitalizations and deaths globally. The virus's impact on public health depends on many factors, including the emergence of new viral variants and their global spread. Consequently, the early detection and surveillance of variants and characterization of their clinical effects are vital for assessing their health risk. The unprecedented capacity for viral genomic sequencing and data sharing built globally during the pandemic has enabled new variants to be rapidly detected and assessed. This article describes the main variants circulating globally between January 2020 and June 2023, the genetic features driving variant evolution, and the epidemiological impact of these variants across countries and regions. Second, we report how integrating genetic variant surveillance with epidemiological data and event-based surveillance, through a network of World Health Organization partners, supported risk assessment and helped provide guidance on pandemic responses. In addition, given the evolutionary characteristics of circulating variants and the immune status of populations, we propose future directions for the sustainable genomic surveillance of SARS-CoV-2 variants, both nationally and internationally: (i) optimizing variant surveillance by including environmental monitoring; (ii) coordinating laboratory assessment of variant evolution and phenotype; (iii) linking data on circulating variants with clinical data; and (iv) expanding genomic surveillance to additional pathogens. Experience during the COVID-19 pandemic has shown that genomic surveillance of pathogens can provide essential, timely and evidence-based information for public health decision-making. Depuis le debut de la pandemie de coronavirus survenue en 2019 (COVID-19), de nombreux variants du coronavirus 2 du syndrome respiratoire aigu severe (SARS-CoV-2) sont apparus, certains entrainant une forte augmentation du nombre d'infections, d'hospitalisations et de deces dans le monde. L'impact du virus sur la sante publique depend de nombreux facteurs, notamment l'emergence de nouveaux variants viraux et leur propagation a l'echelle mondiale. Par consequent, la detection precoce et la surveillance des variants ainsi que la caracterisation de leurs effets cliniques sont essentielles pour evaluer leur risque pour la sante. La capacite sans precedent de sequencage du genome viral et de partage des donnees, capacite mise en place a l'echelle mondiale pendant la pandemie, a permis de detecter et d'evaluer rapidement de nouveaux variants. Le present article decrit les principaux variants circulant dans le monde entre janvier 2020 et juin 2023, les caracteristiques genetiques a l'origine de leur evolution et leur impact epidemiologique dans les differents pays et regions. Ensuite, nous expliquerons comment l'integration de la surveillance des variants genetiques aux donnees epidemiologiques et a la surveillance fondee sur les evenements, par l'intermediaire d'un reseau de partenaires de l'Organisation mondiale de la sante, a permis de faciliter l'evaluation des risques et de fournir des orientations sur les mesures a prendre en periode de pandemie. En outre, compte tenu des caracteristiques evolutives des variants en circulation et de l'etat immunitaire des populations, nous proposons des orientations futures pour une surveillance genomique durable des variants du SARS-CoV-2, au niveau tant national qu'international: (i) optimiser la surveillance des variants en incluant le suivi environnemental; (ii) coordonner l'evaluation en laboratoire de l'evolution des variants et du phenotype; (iii) etablir un lien entre les donnees sur les variants en circulation et les donnees cliniques; et (iv) etendre la surveillance genomique a d'autres agents pathogenes. Lexperience de la pandemie de COVID-19 a mis en evidence que la surveillance genomique des agents pathogenes peut fournir en temps utile des informations essentielles fondees sur des preuves en vue de la prise de decisions en matiere de sante publique. Desde el inicio de la pandemia de la enfermedad por coronavirus de 2019 (COVID-19), han aparecido numerosas variantes del coronavirus de tipo 2 causante del sindrome respiratorio agudo severo (SRAS-CoV-2), algunas de las que han provocado un gran aumento de las infecciones, hospitalizaciones y muertes en todo el mundo. El impacto del virus en la salud publica depende de muchos factores, entre ellos la aparicion de nuevas variantes viricas y su propagacion mundial. En consecuencia, la deteccion y vigilancia tempranas de las variantes y la caracterizacion de sus efectos clinicos son vitales para evaluar su riesgo sanitario. La capacidad sin precedentes de secuenciacion genomica viral y de intercambio de datos creada a nivel mundial durante la pandemia ha permitido detectar y evaluar rapidamente variantes nuevas. En este articulo se describen las principales variantes que circulan a nivel mundial entre enero de 2020 y junio de 2023, la caracteristica genetica que impulsa la evolucion de las variantes y el impacto epidemiologico de estas variantes en los diferentes paises y regiones. En segundo lugar, se informa de como la integracion de la vigilancia de variantes geneticas con los datos epidemiologicos y la vigilancia basada en eventos, a traves de una red de asociados de la Organizacion Mundial de la Salud, apoyo la evaluacion de riesgos y ayudo a proporcionar orientacion sobre las respuestas a la pandemia. Ademas, dadas las caracteristicas evolutivas de las variantes circulantes y el estado inmunitario de las poblaciones, se proponen orientaciones futuras para la vigilancia genomica sostenible de las variantes del SRAS-CoV-2, tanto a nivel nacional como internacional: (i) optimizar la vigilancia de las variantes mediante la inclusion de la monitorizacion ambiental; (ii) coordinar la evaluacion de laboratorio de la evolucion y el fenotipo de las variantes; (iii) vincular los datos sobre las variantes circulantes con los datos clinicos; y (iv) ampliar la vigilancia genomica a patogenos adicionales. La experiencia durante la pandemia de la COVID-19 ha demostrado que la vigilancia genomica de patogenos puede proporcionar informacion esencial, oportuna y basada en evidencias para la toma de decisiones en materia de salud publica. Food and water are rewarding in part because they satisfy our internal needs1,2. Dopaminergic neurons in the ventral tegmental area (VTA) are activated by gustatory rewards3-5, but how animals learn to associate these oral cues with the delayed physiological effects of ingestion is unknown. Here we show that individual dopaminergic neurons in the VTA respond to detection of nutrients or water at specific stages of ingestion. A major subset of dopaminergic neurons tracks changes in systemic hydration that occur tens of minutes after thirsty mice drink water, whereas different dopaminergic neurons respond to nutrients in the gastrointestinal tract. We show that information about fluid balance is transmitted to the VTA by a hypothalamic pathway and then re-routed to downstream circuits that track the oral, gastrointestinal and post-absorptive stages of ingestion. To investigate the function of these signals, we used a paradigm in which a fluid's oral and post-absorptive effects can be independently manipulated and temporally separated. We show that mice rapidly learn to prefer one fluid over another based solely on its rehydrating ability and that this post-ingestive learning is prevented if dopaminergic neurons in the VTA are selectively silenced after consumption. These findings reveal that the midbrain dopamine system contains subsystems that track different modalities and stages of ingestion, on timescales from seconds to tens of minutes, and that this information is used to drive learning about the consequences of ingestion.

Background: COVID-19 is primarily known as a respiratory illness; however, many patients present to hospital without respiratory symptoms. The association between non-respiratory presentations of COVID-19 and outcomes remains unclear. We investigated risk factors and clinical outcomes in patients with no respiratory symptoms (NRS) and respiratory symptoms (RS) at hospital admission. Methods: This study describes clinical features, physiological parameters, and outcomes of hospitalised COVID-19 patients, stratified by the presence or absence of respiratory symptoms at hospital admission. RS patients had one or more of: cough, shortness of breath, sore throat, runny nose or wheezing; while NRS patients did not. Results: Of 178,640 patients in the study, 86.4 % presented with RS, while 13.6 % had NRS. NRS patients were older (median age: NRS: 74 vs RS: 65) and less likely to be admitted to the ICU (NRS: 36.7 % vs RS: 37.5 %). NRS patients had a higher crude in-hospital case-fatality ratio (NRS 41.1 % vs. RS 32.0 %), but a lower risk of death after adjusting for confounders (HR 0.88 [0.83–0.93]). Conclusion: Approximately one in seven COVID-19 patients presented at hospital admission without respiratory symptoms. These patients were older, had lower ICU admission rates, and had a lower risk of in-hospital mortality after adjusting for confounders.

Lesson Study provides an opportunity for teachers to engage in layers of individual and collaborative reflections in, on, and for practice to improve the teaching and learning process. This paper reports on reflections on teaching ratio and proportion online using Lesson Study, which could improve the teaching-learning process, specifically on pedagogy. The study consisted of three major stages: Lesson Planning, Lesson Study Proper, and Reflection through post-lesson discussion. The research lesson study proper was participated in by Grade 9 students in a school in the Philippines and six teachers—one taught the lesson, and the other five observed the class via Zoom. Also, the six teachers were participants in the post-lesson discussion. The data gathered from this descriptive qualitative study were analyzed using thematic analysis. It was concluded that in teaching ratio and proportion, the distinction between terminologies must be emphasized so there will be no confusion and for the students to use what is proper. Moreover, it is advised that a real-life problem involving ratio and proportion be included in the lesson so that students will appreciate its uses and lessen its abstraction. Teachers must also go beyond routine activities to increase students' critical thinking and employ the Higher Order Thinking Skills. Thus, it is recommended that teachers undergo training to learn different and new pedagogies to improve their teaching by going beyond routine activities and incorporating real-life problems.

Abstract The RGD (Arg-Gly-Asp)-binding integrins αvβ6 and αvβ8 are clinically validated cancer and fibrosis targets of considerable therapeutic importance. Compounds that can discriminate between homologous αvβ6 and αvβ8 and other RGD integrins, stabilize specific conformational states, and have high thermal stability could have considerable therapeutic utility. Existing small molecule and antibody inhibitors do not have all these properties, and hence new approaches are needed. Here we describe a generalized method for computationally designing RGD-containing miniproteins selective for a single RGD integrin heterodimer and conformational state. We design hyperstable, selective αvβ6 and αvβ8 inhibitors that bind with picomolar affinity. CryoEM structures of the designed inhibitor-integrin complexes are very close to the computational design models, and show that the inhibitors stabilize specific conformational states of the αvβ6 and the αvβ8 integrins. In a lung fibrosis mouse model, the αvβ6 inhibitor potently reduced fibrotic burden and improved overall lung mechanics, demonstrating the therapeutic potential of de novo designed integrin binding proteins with high selectivity.

The implementation of e-jeepneys stands as a change process that will eventually transition to the modernization of the public transport system in the Philippines. To address concerns about jeepneys’ effects on the environment, energy use, society, the economy, and policies, their acceptability in the Philippines must be considered. This research study aims to identify the sources of influence on Filipinos’ adoption of e-jeepney utilization as a mode of transportation using the extended Pro-Environmental Planned Behavior (PEBP) model. A total of 502 commuters voluntarily answered the survey questionnaire. Based on the findings, perceived environmental concern (PEC) is the most significant determinant influencing attitude (AT) and, thus, affecting the Filipinos’ behavioral intention (BI) towards the adoption of e-jeepneys. Conversely, AT was the primary determinant of BI, which strongly supported the notion of AT as a strong driving force shaping behavioral decisions. Moreover, perceived authority support (PAS) emerged as a strong predictor of subjective norms (SNs), demonstrating the influence of institutional support on societal perceptions. As a result, more environmentally conscious people are more likely to view e-jeepneys positively and intend to use them as a mode of transportation. The endorsement or support from authoritative figures or institutions notably influences subjective norms, which are individuals’ perceptions of social pressures regarding the use of e-jeepneys.

- Abstract
Stakeholder engagement is central to California's vision for how the Local Control Funding Formula (LCFF), with its Local Control Accountability Plan (LCAP), can improve outcomes for K-12 students. On February 1, 2019, PACE's annual conference, Putting Evidence Into Action to Advance Equity in California, brought together policymakers, educators, researchers, and interested citizens to build on the "Getting Down to Facts II" research released in 2018. With the rigorous and comprehensive research as a shared evidence base, participants discussed the future of PK-12 education policy in California. This brief details promising practices shared in a conference session on local democratic engagement in school governance. The panelists varied in their roles and experiences working to improve stakeholder engagement. They included the county office official leading the state's new Community Engagement Initiative, a school district administrator with a story to tell about his own district's engagement efforts, a leader of the state's largest parent advocacy organization, and a community organizer who works to make sure student voices are part of the LCFF conversation. Overall, panelists shared how their organizations are engaging a broad and diverse set of stakeholders and how they are building stakeholder capacity to engage productively in school governance. Reflections from session attendees are also taken into account in this brief.

Abstract Background Osteoarthritis (OA) is a chronic debilitating disease impacting a significant percentage of the global population. While there are numerous surgical and non-invasive interventions that can postpone joint replacement, there are no current treatments which can reverse the joint damage occurring during the pathogenesis of the disease. While many groups are investigating the use of stem cell therapies in the treatment of OA, we still don’t have a clear understanding of the role of these cells in the body, including heterogeneity of tissue resident adult mesenchymal progenitor cells (MPCs). Methods In the current study, we examined MPCs from the synovium and individuals with or without a traumatic knee joint injury and explored the chondrogenic differentiation capacity of these MPCs in vitro and in vivo. Results We found that there is heterogeneity of MPCs with the adult synovium and distinct sub-populations of MPCs and the abundancy of these sub-populations change with joint injury. Furthermore, only some of these sub-populations have the ability to effect cartilage repair in vivo. Using an unbiased proteomics approach, we were able to identify cell surface markers that identify this pro-chondrogenic MPC population in normal and injured joints, specifically CD82LowCD59+ synovial MPCs have robust cartilage regenerative properties in vivo. Conclusions The results of this study clearly show that cells within the adult human joint can impact cartilage repair and that these sub-populations exist within joints that have undergone a traumatic joint injury. Therefore, these populations can be exploited for the treatment of cartilage injuries and OA in future clinical trials.

WHO works, on a daily basis, with countries globally to detect, prepare for and respond to acute public health events. A vital component of a health response is the dissemination of accurate, reliable and authoritative information. The Disease Outbreak News (DON) reports are a key mechanism through which WHO communicates on acute public health events to the public. The decision to produce a DON report is taken on a case-by-case basis after evaluating key criteria, and the subsequent process of producing a DON report is highly standardised to ensure the robustness of information. DON reports have been published since 1996, and up to 2022 over 3000 reports have been published. Between 2018 and 2022, the most frequently published DON reports relate to Ebola virus disease, Middle East respiratory syndrome, yellow fever, polio and cholera. The DON web page is highly visited with a readership of over 2.6 million visits per year, on average. The DON report structure has evolved over time, from a single paragraph in 1996 to a detailed report with seven sections currently. WHO regularly reviews the DON report process and structure for improvements. In the last 25 years, DON reports have played a unique role in rapidly disseminating information on acute public health events to health actors and the public globally. They have become a key information source for the global public health response to the benefit of individuals and communities.

47. No evidence for relationship between paternal post-partum depressive symptoms and testosterone or cortisol in first-time fathers

Male life history strategies are regulated by the neuroendocrine system. Testosterone (T) and cortisol regulate male behaviors including parenting and facilitate managing tradeoffs at key transitions in development such as first-time fatherhood. Both hormones demonstrate marked fluctuations in the postnatal period, and this presents an opportunity to investigate the role of T and cortisol in postpartum depressive symptoms—comparably less studied in fathers than in mothers in the evolutionary literature. Prior work on depressive symptoms has yet to integrate insights from the “dual hormone hypothesis (DHH)” which has focused on how T and cortisol interact to jointly regulate traits associated with dominance and status-seeking (i.e., mating effort) but has yet to be included in models of parenting effort. In this research, we use secondary data to investigate the relationship between DHH and traits ostensibly opposed to status seeking (i.e., depressive symptoms). First-time fathers (n = 193) provided morning saliva samples 10 months following parturition and reported on the presence of depressive symptoms (BDI-II). Responses were decomposed into three factors: cognitive, affective, and somatic. Using hybrid latent variable structural equation modeling, we did not find evidence that T predicted variability in cognitive, affective, or somatic depressive symptom factors. We found a null effect for cortisol as well. Finally, we could not find evidence that the DHH variable (T × cortisol interaction) predicted any variability in cognitive, affective, or somatic depressive symptoms. While we did not find evidence to support our hypotheses using a secondary data set, this study contributes to research on the neuroendocrinology of depression in fathers. Discussion focuses on the limitations of sample demographics, timing of saliva and self-report collection, and the lack of extant theory specific to paternal postpartum depression.

Aim. To assess the acceptability of a symptom self-management booklet among older Chinese Americans receiving kidney replacement therapy. Background. In previous work, we identified commonly occurring, bothersome symptoms and strategies used in this population to ameliorate symptoms. We used these data to develop a symptom self-management booklet in English, traditional, and simplified Chinese. Introduction. In the United States, the prevalence of kidney disease is 1.5 times higher in Asians compared to whites. With the many symptoms associated with this disease, self-management of symptoms would be particularly helpful. Methods. Seven older Chinese Americans receiving kidney replacement therapy and their caregivers were interviewed to assess the acceptability of the booklets. We reviewed participant feedback on content, graphics, and design, reading experience, suggestions for improvement, and health information sources using the inductive thematic method. Results. Overall, patients confirmed acceptability of these self-management booklets across all domains. Discussion. This study validated the booklet as a source of health information for older Chinese American patients with kidney disease, which some studies suggest are preferred to electronic materials or methods in this population. Health care providers can use the resultant booklets when caring for these patients to provide culturally sensitive information on self-management of symptoms. Conclusion and Implications for Nursing. These booklets provide a free resource tailored to an underserved population and may help nurses and nurse practitioners provide care with cultural humility. Implications for Health Policy. Embracing community-based participatory research, as was done in this study, can help create culturally appropriate patient education materials that empower patient symptom self-management and promote informative and culturally sensitive conversations between patients, families, and providers.

49. Exploring the Antidiarrheal Properties of Papaya Leaf: Insights In Vivo Study in Mice-Model and In Silico Analysis at M3 Muscarinic Acetylcholine Receptor Interaction

Diarrhea caused by gut motility involves 5-HT3 serotonin receptor (5-HT3R) antagonist, M3 muscarinic acetylcholine receptor (M3-AchR), and H1 histamine receptor (H1R) via their respective agonist. Papaya (Carica papaya L.) leaf is an herbal medicine to treat diarrhea in Indonesia, though this has not yet been proven scientifically. This study aimed to determine the antidiarrheal properties of papaya leaf through in vivo and in silico studies. In the mice model, papaya leaves were infused with distilled water and subjected to in vivo antidiarrheal study using castor oil-induced diarrhea. In silico molecular docking study of nineteen secondary metabolites was carried out on the M3-AchR (PDB ID: 5ZHP) using AutoDock Tools 1.5.6, while ADMET was predicted by pre-ADMET. The results showed that papaya leaf infusion caused a decrease in the total number of feces, an increase in the onset time of diarrhea, a reduction in the frequency of diarrhea, and an increase in the percentage of inhibition of diarrhea. Quercetin 3-rutinoside, a flavonoid glycoside, is potentially an antidiarrheal remedy at M3-AchR. ADMET prediction showed good distribution on the target and was not toxic, but absorption needed to be considered. We concluded that the antidiarrheal activity of papaya leaf infusion was dose-dependent. Based on a molecular docking study, the flavonoid glycoside was very effective as an antidiarrheal remedy. ADMET prediction showed a specific distribution to the target and was not toxic.
