The endothelium was the first non-hematopoietic tissue to be identified as a physiological target for erythropoietin (EPO). EPO is involved in recruitment and mobilization of endothelial progenitors and stimulates the production of erythroid cell regulatory factors in endothelial cells. Production of these EPO-dependent factors is inhibited by IL-3 in vitro. Furthermore, EPO-dependent red cell formation in anemic mice is equally repressed by IL-3. The number of IL-3 receptors on endothelial cells increases in chronic inflammation and IL-3 may be one of the inflammatory cytokines, together with TNF-alpha, IFN-gamma or IL-6, which prevents optimal red cell formation in many patients with kidney failure receiving high doses of EPO. These patients could benefit from the administration of some of the EPO-stimulated endothelial factors, such as C21 (the C-terminal segment thrombospondin-4), thrombospondin-1 and chaperonin 10, because these proteins bypass EPO receptors and signaling pathways that are usually compromised in EPO resistance. C21 stimulates red cell formation in anemic mice, increases human hematopoietic cell proliferation in vitro and could eventually fight inflammation, because it is an osteopontin antagonist. Thrombospondin-1 prevents inflammation, stimulates erythroblast proliferation and counteracts IGFBP-3-mediated erythroid inhibition. Finally, chaperonin 10 stimulates hemoglobin synthesis and has anti-inflammatory properties through the inhibition of Toll-like receptor signaling pathways., (Copyright 2010 Elsevier Ltd. C21, the C-terminal residue of thrombospondin-4 (TSP-4), was identified as a peptide growth factor during an investigation concerning erythropoietin-dependent, erythroid stimulating factors of endothelial origin. It is active in cultures of several human hematopoietic stem cells, skin fibroblasts and kidney epithelial cells and stimulates red cell formation in anemic mice. A method of affinity chromatography in the presence of high concentrations of Triton X-100, previously developed for identifying proteins associated with the TSP-1 receptor CD47, was utilized for the detection of C21 binding molecules and their detergent-resistant, associated partners. These experiments helped to delineate two different mechanisms of C21 action, which are compatible with its cell proliferating activity. As a cell matrix peptide, C21 binds to the osteopontin receptor CD44 and could act as an osteopontin antagonist, preventing the inhibition of primitive hematopoietic stem cell proliferation. TSP-1, another matrix protein, binds to C21 and could indirectly act as an antagonist, by shunting C21-CD44 interactions. The second mechanism is a direct effect of C21 on cell proliferation. The extremely rapid internalization and nuclear localization of the peptide could be explained by CD44-mediated internalization, followed by a microtubule-mediated transport towards the nucleus, or, eventually, direct membrane insertion. These alternative hypotheses are supported by previously observed membrane insertion of similar synthetic and viral acidic amphipathic peptides, the presence of microtubule-associated protein 1B (MAP1B) and dynactin in the triton-soluble complexes associated with C21 and the presence in such complexes of dual compartment proteins for nuclei and plasma membranes, such as MAP1B, AHNAK and CD44., (Copyright (c) 2009 Elsevier Inc. Background: C21, the C-terminal peptide of thrombospondin-4, has growth promoting activity and was discovered as one of several erythropoietin-dependent endothelial proteins. C21 stimulates red cell formation in anemic mice and is a growth factor for CD34+ and CD36+ hematopoietic cells, skin fibroblasts and kidney epithelial cells. ROD1 has been identified as an intracellular mediator. Nothing is known about the existence of putative C21 receptors on plasma membranes of target cells., Findings: We analyzed the nature of C21-binding proteins in cell lysates of skin fibroblasts using C21 affinity columns. The membrane receptor CD44 was identified as C21-binding protein by mass spectrometry. We were unable to demonstrate any direct involvement of CD44 on cell growth or the effect of C21 on cell proliferation. A soluble form of CD44 was synthesized in insect cells and purified from culture supernatants with a combination of PVDF filtration in the presence of ammonium sulphate and HPLC. Both osteopontin and hyaluronic acid competitively displaced Biotin-C21 binding to CD44. In a colony-forming assay using primitive CD133+ hematopoietic stem cells from cord blood, osteopontin and C21 had opposite effects and C21 reduced the inhibitory action of osteopontin., Conclusion: CD44 is a C21-binding membrane protein. We could not demonstrate an involvement of CD44 in the proliferative action of C21. Nevertheless, based on the antagonism of C21 and osteopontin in hematopoietic precursors, we speculate that C21 could indirectly have a major impact on hematopoietic stem cell proliferation, by hindering osteopontin membrane binding at the level of the bone marrow niche.

The matrix protein thrombospondin-4 has an acidic amphipathic C-terminal peptide (C21) which stimulates erythroid cell proliferation. Here we show that C21 stimulates red cell formation in anemic mice in vivo. In vitro experiments indicated that the peptide-mediated increase of erythroid colony formation in cultures of human CD34+ hematopoietic progenitor cells was possible only under continuous presence of erythropoietin. In the absence of this cytokine, C21 stimulated exclusively myeloid colony formation. Therefore, the peptide is not a specific erythroid differentiation factor. In fact, it is mitogenic in non-erythroid cells, such as skin fibroblasts and kidney epithelial cells. In erythroleukemic TF-1 cells, it actually decreased the production of the erythroid differentiation marker glycophorin A. C21-affinity chromatography revealed regulator of differentiation 1 (ROD1) as a major C21-binding protein. ROD1 is the hematopoietic cell paralog of polypyrimidine tract binding proteins (PTBs), RNA splice regulators which regulate differentiation by repressing tissue-specific exons. ROD1 binding to C21 was strongly inhibited by synthetic RNAs in the order poly A > poly U > poly G = poly C and was weakly inhibited by a synthetic phosphorylated peptide mimicking the C-terminal domain of RNA polymerase II. Cellular overexpression or knockdown experiments of ROD1 suggest a role for this protein in the mitogenic activity of C21. Since the nuclear proteins ROD1 and PTBs regulate differentiation at a posttranscriptional level and there is a fast nuclear uptake of C21, we put forward the idea that the peptide is internalized, goes to the nucleus and maintains cells in a proliferative state by supporting ROD1-mediated inhibition of differentiation.

The anti-inflammatory peptide early pregnancy factor/chaperonin 10 (cpn10) was identified by 2D-electrophoresis/mass spectrometry as one of the proteins increased in human umbilical cord endothelial cells (HUVEC) after treatment with erythropoietin (EPO). EPO increased the amount of cpn10 released into the medium of HUVEC cultures, despite the absence of a secretory signal peptide. Although immunosupressive agents would represent an indirect advantage for red cell formation under conditions of infection and inflammation, it is possible that cpn10 may have direct effects on erythroid cells. We show that the chaperonin decreased cell proliferation in cultures of the erythroleukemia cell line K562 and increased the amounts of the erythroid differentiation markers glycophorin A and hemoglobin in TF-1 cells. Nevertheless, cpn10 is not a specific erythroid cell differentiation factor, because monolayers of skin fibroblasts overexpressing cpn10 had significantly higher levels of the differentiation marker collagen I than normal fibroblasts. Nothing is known about the mechanism of action of cpn10 in its capacity as a general differentiation factor. An analysis of early changes taking place in K562 cells after incubation with cpn10 using antibody microarrays identified several phosphorylation events, including a decrease of GSK-3alpha phosphorylation. Further studies in TF-1 cells indicated that cpn10 decreased the phosphorylation of cofilin-1 while stimulating that of GSK-3beta. Furthermore, glycophorin A production decreased in the presence of a GSK-3 inhibitor in the same cells. These experiments support the idea that GSK-3-regulated signal transduction pathways are not only important for stem cell maintenance but may be involved in events controlling cell differentiation.

Serpin A1 (alpha1-antitrypsin, alpha1-proteinase inhibitor), a potent neutrophil elastase inhibitor, has therapeutic potential as a wound-healing agent. We compared the in vitro wound-healing action of serpin A1-IGF, a recombinant fusion protein of serpin A1(M351E-M358L) and insulin-like growth factor I with that observed in the presence of natural serpin A1 or A1-C26, the synthetic C-terminal 26 residue peptide of serpin A1, previously shown to have mitogenic and antiviral activities. All agents reduced wound sizes in monolayers of the kidney epithelial cell line LLC-PK1 and in primary cultures of human skin fibroblasts. Wound reduction in primary human keratinocytes was only observed with the serpin A1-IGF chimera. None of the factors stimulated cell proliferation using a colorimetric assay, with the exception of the serpin A1-IGF chimera, which caused a significant increase of cell proliferation and thymidine incorporation in human skin fibroblasts. However, wound healing by the A1-IGF chimera was reduced in keratinocytes in the presence of mitomycin C, suggesting a role of cell proliferation in wound reduction. The hydrophobic A1-C26 peptide significantly increased the production of collagen I in skin fibroblasts, an appealing asset for skin care applications.

Serpin A1 (alpha1-antitrypsin, alpha1-proteinase inhibitor) has been shown to be a non-cytolytic antiviral factor present in blood and effective against HIV infection. The best known physiological role of serpin A1 is to inhibit neutrophil elastase, a proteinase which is secreted by neutrophils at sites of infection and inflammation. Decreased HIV-infectivity is associated with decreased density of membrane-associated elastase. The enzyme may facilitate binding of the HIV membrane protein gp120 to host cells, and it specifically cleaves SDF-1, the physiological ligand of the HIV-1 co-receptor CXCR4. It has been suggested that one of the actions of serpin A1 as antiviral agent is to reduce HIV infectivity, and this property could be due to elastase inhibition. However, the most dramatic effect of serpin A1 is inhibition of HIV production. In vitro experiments indicate that the C-terminal peptide of serpin A1, produced during the formation of the complex of serpin with serine proteinases, may be responsible for the inhibition of HIV-1 expression in infected cells. This peptide, an integral part of the serpin-enzyme complex, is internalized by several scavenger receptors. Peptides corresponding to the C-terminal section of serpin A1 inhibit HIV-1 long-terminal-repeat-driven transcription and interact with nuclear proteins, such as alpha1-fetoprotein transcription factor. LDL-receptor-related protein 1 (LRP1/CD91), the best known receptor for serpin-enzyme complexes, is up-regulated in monocytes of HIV-1-infected true non-progressors. CD91 could be one of the major players in host resistance against HIV-1. It has the capacity of internalizing antiviral peptides such as serpin C-terminal fragments and alpha-defensins, and is at the same time the receptor for heat-shock proteins in antigen-presenting cells, in which chaperoned viral peptides could lead to the induction of cytotoxic T-cell responses.

Serpin A1 (alpha1-proteinase inhibitor) inhibits human immunodeficiency virus 1 (HIV-1) production by mechanisms which remain to be elucidated. The complex formation of serpin A1 with proteinases eliminates the proteolytic activity and generates a fragment corresponding to the serpin C-terminal 36-residue peptide. Here, we show that the C-terminal 26-residue peptide of serpin A1 (A1-C26) inhibits HIV-long terminal repeat (LTR)-driven transcription in epithelial cells transfected with HIV-1 LTR promoter-driven genes. A1-C26 increased STAT1 phosphorylation and strongly reduced viral expression in a monocytic cell line infected with HIV-1 NL4-3. This reduction of expression was also observed in HIV-1 infected, PHA-activated peripheral blood mononuclear cells. In HIV-1 infected cells, the inhibitory activity of HIV-1 caused by B9-C23 and C1-C26, the A1-C26 homologues corresponding to the C-terminal sections of serpin B9 and serpin C1, was much lower than that obtained with A1-C26. These serpin peptides represent a novel class of antiviral agents.

There is a close association between the growth hormone (GH)-insulin-like growth factor I (IGF-I) axis, infection and immunity. Infection with the human immunodeficiency virus (HIV) is often associated with a decrease of the concentrations of IGF-I, IGF-II, IGF-binding protein 3 (IGFBP-3) and an increase of IGFBP-1 and -2. Many investigators have studied the relationship between the GH-IGF-I system and some of the most common characteristics of disease progression, such as decreased CD4 cell counts, weight loss and fat redistribution. Although conditions for restoration of thymic function and lymphopoiesis with GH or IGF-I are still not well defined, many studies led to the development of clinical trials on the therapeutic use of GH, IGF-I and GHRH for the treatment of weight loss or fat redistribution, two problems which persist despite the introduction of highly active antiretroviral therapy. Monitoring IGF-I concentrations during treatment with GH and GHRH is likely to become an essential component of their therapeutic use. IGF-I levels are the first indicator of treatment efficacy and can be used to monitor compliance. High levels of IGF-I are a warning sign for the increased risk of potential adverse effects, such as acromegalic-like symptoms or malignancy. This could lead to a reduction of the therapeutic dose or the temporary interruption of treatment until IGF levels reach a safe range. IGF-I levels are also likely to increase with other hormones used in HIV patients, such as erythropoietin for the treatment of anemia or anabolic androgens in HIV-infected women.

The nature of erythropoietin (EPO)-dependent, erythroid cell regulatory factors secreted by endothelial cells is largely unknown. The production of thrombospondin 1 (TSP-1) and insulin-like growth factor binding protein 3 (IGFBP-3) is increased in cultures of human umbilical vein endothelial cells (HUVEC) incubated with erythropoietin (EPO). Simultaneous incubation of HUVEC with EPO and interleukin 3 (IL-3) resulted in a decreased production, suggesting that both TSP-1 and IGFBP-3 belong to the EPO- and IL-3-dependent erythroid regulatory factors previously described in cultures of bone marrow endothelial cells. TSP-1 and TSP-1 derived synthetic peptides based on the CD36 and CD47 binding sites of TSPs increased thymidine incorporation into bovine erythroid cells of fetal liver. IGBBP-3 inhibited thymidine incorporation in the same cells. Preincubation of erythroid cells with TSP-1 eliminated the inhibitory activity of IGFBP-3. We suggest that EPO-dependent, endothelial-derived TSP-1 may play a positive role in red cell production by acting directly on erythroid cells, stimulating DNA synthesis and preventing the inhibitory action of IGFBP-3.

Erythropoietin (EPO) stimulates the production of small erythroid cell stimulating factors (molecular weight <5 kDa) in cultures of bone marrow endothelial cells. We identified a fragment of thrombospondin-4 (TSP-4) as an EPO-stimulated protein in endothelial cell lysates. Pre-incubation of the low molecular weight fractions from supernatants of EPO-treated umbilical cord endothelial cells (HUVEC) with antibodies against the C-terminal residues of TSP-1,2 and TSP-4 decreased the erythroid cell stimulating activity. The C-terminal TSP-1 section corresponding to a molecular weight lower than 6 kDa has the integrin-associated protein binding motif VVM. The corresponding TSP-4 fragment, lacking the three residue sequence VVM, has a distinctive acidic peptide comprising the last 21 amino acids (C21) with the characteristics of an amphipathic helix. C21 stimulated thymidine incorporation into bovine erythroid cells, increased cell numbers in cultures of cord blood CD36+ erythroid precursors and skin fibroblasts, and decreased HUVEC proliferation. SC21, a homologous peptide of identical amino acid composition but with interchanged residues, was non-amphipathic and had no erythroid cell stimulating activity.

C26, the C-terminal 26 residue peptide of serpin A1, significantly increased cell proliferation in cultures of hepatoma cells, but not in porcine kidney epithelial cells, human skin fibroblasts or keratinocytes. The mitogenic activity of C26 was preferentially inhibited with a protein kinase C (PKC) inhibitor, an antibody against CD47 and CD47 short interfering RNA. The mutant C26-K19R,N22M, imitating a thrombospondin-like cell adhesion motif, increased the mitogenic activity in both Hep G2 cells and MCF-7 breast cancer cells. Phosphorylation of C26 at T24 (a putative PKC phosphorylation site) resulted in a 1.9-2.5 increase in mitogenic activity over C26 in MCF-7 cells.

It has been previously reported that insulin-like growth factor I (IGF I) decreases in AIDS patients with wasting, a condition that is partially prevented by combined IGF I growth hormone therapy. By generating bifunctional proteins of IGF I and stromal cell-derived factor 1alpha (SDF-1alpha) or alpha1 proteinase inhibitor (API), two proteins known to prevent HIV infection, it may be possible to improve the therapeutic effectiveness of these compounds for the treatment of AIDS-mediated wasting. SDF-1alpha or the M351E-M358L mutant of API were attached at the C-terminal end of IGF I and synthesized by a stable insect cell expression technique. The IGF I-SDF-1alpha chimera reduced the enhancement of thymidine incorporation into bovine fetal erythroid cells observed in the presence of insect cell produced IGF I alone. It also decreased the SDF-1 and IGF I-stimulated hematopoietic cell migration, without losing the capacity to compete with the binding of HIV-1 (IIIB)-surface glycoprotein gp120. The IGF I-API chimera displayed the same mitogenic activity and a similar, but lower chemotactic activity than IGF I in the assays mentioned above. It had a comparable anti-elastase activity to that observed with a previously described IGF II-API fusion protein with the single mutation M351E. The binding of gp120 to a murine hematopoietic cell line was stimulated by human neutrophil elastase (25-100 nM) and inhibited by IGF I-API. In conclusion, the linkage of IGF I with SDF-1 or API can alter some biological functions of the single components of the chimera while keeping their ability to compete with HIV-1-gp120 binding.

We compared the antiapoptotic activity of a recombinant chimera of insulin-like growth factor II (IGF-II) and interleukin (IL)-3 with the corresponding equimolar mixture of the individual components based on changes in several factors associated with survival in the CD34+ human hematopoietic cell line TF-1. Propidium iodide-stained cells analyzed by fluorescein-activated cell sorter indicated that the chimera was more effective than the corresponding equimolar mixture in decreasing the amounts of apoptotic cells and increasing the proportion of cells in the S-phase of the cell cycle. The chimera was more effective in increasing the antiapoptotic protein Bclx(L) and produced a significant increase in signal transducer and activator of transcription-5 phosphorylation and in phosphatidylinositol-3 kinase (PI-3K) activity. The PI-3K inhibitor LY294002 specifically inhibited cell survival in the presence of the chimera, suggesting a key role of this enzyme in the potentiation of survival caused by the linkage of IGF and IL-3. This potentiation of survival and its preferential inhibition by LY294002 were also observed in a nontransformed, primary culture of human umbilical cord endothelial cells.

A chimera of an N-terminally modified insulin growth factor (IGF)-II, NQPQMVHTY-hIGF-II(9-67) (BOMIGF), fused to interleukin-3 (IL-3) significantly improved the migration of CD34(+) human hematopoietic cells with respect to the effects observed during co-stimulation with BOMIGF and IL-3. A phosphatidylinositol-3 (PI-3) kinase inhibitor specifically inhibited migration in the presence of the chimera, while no significant difference in the inhibition of migration was observed in the presence of a Rho kinase inhibitor. These results suggest a key role of the PI-3 kinase pathway in the potentiation of migration caused by the linkage of BOMIGF and IL-3.

Alpha(1)-proteinase inhibitor (API) was coupled at the C-terminus of a human insulin-like growth factor (IGF) analog to facilitate its production in insect cells. This fusion protein significantly increased thymidine incorporation into HL-60 cells as compared with the incorporation observed with an equivalent molar mixture of the IGF analog and API. The M351E variant of API has been previously shown to reduce aggregate formation in prokaryotic expression systems. When the oxidation-sensitive methionine 351 of the inhibitor was changed to glutamate, the M351E variant was secreted in larger amounts from insect cells than the corresponding fusion protein with wild-type API. The M351E fusion protein and the corresponding chimera containing the wild-type API were tested for their capacity to inhibit human neutrophil elastase. The M351E variant was a more potent elastase inhibitor than the fusion protein containing the wild-type analog, whereas the proliferative activity of both chimeras was identical. The described mitogenic effect of the chimera and the improved anti-elastase activity of the M351E variant are two ideal properties for therapeutic agents acting in pathological situations where cell proliferation and inhibition of neutrophil elastase have to take place simultaneously, such as during wound healing.

Azidothymidine (AZT)-induced anemia in mice can be reversed by the administration of IGF-IL-3 (fusion protein of insulin-like growth factor II (IGF II) and interleukin 3). Although interleukin 3 (IL-3) and erythropoietin (EPO) are known to act synergistically on hematopoietic cell proliferation in vitro, injection of IGF-IL-3 and EPO in AZT-treated mice resulted in a reduction of red cells and an increase of plasma EPO levels as compared to animals treated with IGF-IL-3 or EPO alone. We tested the hypothesis that the antagonistic effect of IL-3 and EPO on erythroid cells may be mediated by endothelial cells. Bovine liver erythroid cells were cultured on monolayers of human bone marrow endothelial cells previously treated with EPO and IGF-IL-3. There was a significant reduction of thymidine incorporation into both erythroid and endothelial cells in cultures pre-treated with IGF-IL-3 and EPO. Endothelial cell culture supernatants separated by ultrafiltration and ultracentrifugation from cells treated with EPO and IL-3 significantly reduced thymidine incorporation into erythroid cells as compared to identical fractions obtained from the media of cells cultured with EPO alone. These results suggest that endothelial cells treated simultaneously with EPO and IL-3 have a negative effect on erythroid cell production., (Copyright 2002 Elsevier Science Ltd. alpha(1)-proteinase inhibitor (API) is a potential therapeutic agent in all diseases in which elastase released by neutrophils has to be effectively neutralized. We ligated the cDNA of human API to the C-terminal section of an insulin-like growth factor II analogue (BOMIGF), known to be properly folded and secreted in insect cells using the baculovirus expression system. The BOMIGF-API chimera was recovered from the incubation medium of the infected cells. It shared the properties of both IGFs and API. It inhibited neutrophil elastase and formed SDS-stable complexes with the enzyme. The attachment of the large API protein to the C-terminal end of the 10 kDa IGF analogue did not destroy the IGF-mediated stimulation of thymidine incorporation into bovine fetal erythroid cells. We tested the capacity of the chimera to affect fibronectin-dependent TF-1 cell migration. BOMIGF-API significantly restored TF-1 cell migration in the presence of elastase, which is the enzyme of burn wound fluid most probably involved in fibronectin degradation. Some of the beneficial uses for this chimera may include all instances for which inhibition of elastase-mediated extracellular matrix destruction as well as stimulation of cell migration and proliferation are required for tissue repair.

The effect of 406, a novel fusion protein between the N-terminal sequence of the insect insulin-like peptide, bombyxin, human insulin-like growth factor II and mouse interleukin 3 was investigated in its capacity to abrogate the toxic effects of azidothymidine (AZT) in C57BL/6 mice. Mice receiving 2.5 mg/ml AZT in their drinking water were concurrently treated with daily s.c. injections of 14, 140 or 1400 ng 406 for 4 wk. AZT-treated mice had a lower total weight, hemoglobin content and white blood cells than non treated controls. 406 significantly increased the number of circulating white blood cells at all doses, and the optimal effects were observed at a dose of 140 ng/mouse. Using this optimal dose, 406 completely abrogated the AZT-mediated weight loss. The effects on erythroid cells depended on the severity of the AZT-induced anemia. The amounts of hemoglobin were equal or slightly lower than those of controls under conditions of mild anemia, but were significantly higher than controls under conditions of severe anemia. 406 significantly increased the number of all hematopoietic colony-forming cells in bone marrow and spleen, but the effects were particularly striking in granulocyte-macrophage precursors. Blood glucose levels did not change at optimal or suboptimal 406 doses but increased at a dose of 1.4 microg/mouse. These experiments demonstrate the usefulness of these IGF-cytokine fusion proteins, whose low cost production represents a significant advantage for future in vivo studies.

We have found that a slightly modified insulin-like growth factor II (IGF II) consisting of a chimaera of bombyxin and human IGF II (BOMIGF) is properly secreted in insect cells by using the baculovirus expression system. Human interleukin 3 (IL-3) was attached to the C-terminal amino acid residue of BOMIGF with peptide linkers containing five or twelve residues. Only the chimaera with the 12-residue linker had biological activities of both IGF II and IL-3. The chimaera had a significantly higher mitogenic activity than IL-3 in cell cultures of the human haemopoietic cell line TF-1 and its effect could be observed even at femtomolar concentrations. It was also able to stimulate thymidine incorporation in IGF II-dependent bovine fetal erythroid cells. The chimaera significantly increased the number of macroscopic haemopoietic colonies in cultures of human peripheral blood in comparison with IL-3 or mixtures of IL-3 and BOMIGF in vitro. Subcutaneous injection of a BOMIGF-mouse IL-3 chimaera in normal C57BL/6 mice resulted in a significant increase of the number of spleen stem cells producing macroscopic haemopoietic colonies. This new system for the biosynthesis of IGF-cytokine fusion proteins in insect cells might prove advantageous for the low-cost and high-yield production of molecules with complementary or synergistic biological activities.

A technique for the optimal synthesis of secreted fusion proteins between insulin-like growth factors (IGFs) and cytokines is described. The cDNA of BOMIGF, a fusion protein between the insect insulin-like peptide bombyxin and IGF II, has been linked to the gene of interleukin-3. The BOMIGF-interleukin 3 fusion gene was cloned downstream of the promoter regions of the p10 and polyhedrin proteins within baculovirus transfer vectors. A third, dual transfer vector was constructed with the gene inserted simultaneously behind p10 and polyhedrin promoters. Two different lepidopteran cell lines, Spodoptera frugiperda (Sf9) and Trichoplusia ni (BTI-TN-5B1-4) were infected with the recombinant baculoviruses obtained from the three transfer vectors. Trichoplusia ni cells produced the largest amount of recombinant protein. Although the efficiency of the three recombinant viruses was remarkably similar, the baculovirus with the gene present behind both promoters produced relatively more recombinant protein in host cells than those viruses driven with the polyhedrin or p10 promoters alone. The BOMIGF-interleukin-3 chimera was stable and continuously increased in the culture medium up to 5-6 days postinfection. Therefore the addition of a protease inhibitor was useful only at the stage of massive host cell death. Medium supplemented with copper sulfate was detrimental for the long-term production of the fusion protein.

BOMIGF, a chimera between bombyxin and insulin-like growth factor II (IGF II) has been modified and extended to a total of 85 amino acids including 7 additional basic amino acids. The new peptide bound heparin as efficiently as GM-CSF and much better than IGF II or BOMIGF. It could be retained on albumin-coated tissue culture wells without loosing its capacity to stimulate thymidine incorporation into erythroid cells of fetal bovine liver and on fibronectin-heparin coated wells keeping its mitogenic activity towards L6 muscle cells.

Stromal cells are one of the components of the hematopoietic microenvironment, which is crucial for the proliferation and differentiation of hematopoietic cells. We have obtained a bovine fetal-liver stromal cell line that supports erythroid colony formation in the presence of erythropoietin. The cells are cytokeratin-negative and vimentin-positive, indicating a mesenchymal origin. Furthermore, they have phagocytic activity and show endothelial-like morphology. Erythropoietin at a concentration of 2 mU/mL was sufficient to increase erythropoiesis in the presence of these stromal cells, and the effect could be further enhanced in the presence of physiological concentrations of insulin-like growth factor II. The supportive role of stromal cells on erythroid colony formation could still be observed, although to a lesser extent, when the erythroid precursors were physically separated from the stromal monolayer by an agar layer or with serum-free media conditioned by stromal cells. Ultrafiltration of the conditioned media indicated that the active factors have a nominal molecular weight smaller than 3 kD, which does not correspond to any of the known cytokines with erythroid-cell stimulating activities. Our data suggest that these fetal-liver stromal cells play an important role in fetal erythropoiesis, a function which was previously thought to be played exclusively by fetal hepatocytes.

A synthetic gene encoding the signal peptide and the N-terminal sequence of bombyxin, an insect insulin-like peptide, and the 58 amino acids of the C-terminal sequence of human insulin-like growth factor II (IGF II) has been expressed using the baculovirus system. This synthetic chimaera was obtained by amplification of four overlapping oligonucleotides using Taq polymerase and cloning into the transfer vector pBluebac. The construct was integrated by homologous recombination into the Autographa californica nuclear polyhedrosis genome. Spodoptera frugiperda Sf9 insect cells infected with the recombinant baculovirus secreted an accurately processed peptide consisting of the ten N-terminal amino acids of bombyxin and the 58 C-terminal amino acids of IGF II. The N-terminal glutamine of bombyxin was changed to asparagine to facilitate sequencing of the synthetic peptide. The chimaera was five times more potent than human recombinant IGF II in its capacity to stimulate thymidine incorporation into erythroid cells of fetal bovine liver in a serum-free medium. It stimulated erythroid colony formation in the presence of 2 microunits/ml erythropoietin in cells cultured over a monolayer of stromal cells of fetal liver. Artificial chimaeras as described here may prove useful for the production of insulin, IGF I and other peptides as secreted proteins in insect cells.

Based on the primary structure of the rat corticostatin R4 (antimicrobial defensin RatNP-1), a synthetic gene was designed, synthesized, and inserted into the IPTG-inducible prokaryotic expression vector pFLAG, with an additional 13 codons between the FLAG sequence and the synthetic R4 sequence. This construct, N-p1.2, was further developed by inclusion of multiple cloning sites right after the FLAG sequence, forming a new plasmid pSCV-1. Escherichia coli transformants containing pSCV-1 or N-p1.2 could only be propagated on agar plates in the absence of IPTG due to the detrimental expression of R4 fusion peptide to the growth of bacteria upon IPTG induction. A 214-bp bovine IGF-II cDNA and a 700-bp Ly-6C.2 cDNA fragment were subcloned into pSCV-1 and N-p1.2 respectively. Only the E. coli cells transformed with recombinant plasmids grew on IPTG agar plates. This "suicide" selection against nonrecombinants was further tested in cDNA library construction using pSCV-1. Analysis of plasmid DNA prepared from randomly picked colonies growing on ampicillin agar plates containing IPTG showed all plasmids contained cDNA inserts. The lambda Hind III fragments were used for comparing the cloning efficiency of pSCV-1 to pBluescript. Four of the 60 (6.6%) analyzed white colonies transformed with pBluescript were false positives. All of the analyzed pSCV-1-transformed colonies growing on IPTG plates contained recombinant forms of plasmid. The percentage recovery of each ligatable lambda Hind III fragment was similar in both pBluescript and pSCV-1.(ABSTRACT TRUNCATED AT 250 WORDS)

1. cDNA for insulin-like growth factor II (IGF II) was synthesized from poly A(+)-RNA from fetal bovine intestine and amplified. 2. The sequence corresponding to amino acids 6-62 was identical to a published ovine IGF II sequence cDNA with the exception of a single nucleotide change (G to A). 3. Northern blot analysis of intestine, liver, kidney and spleen from bovine fetuses showed multiple IGF II RNA species which are more similar to the human than to the rodent mRNAs. 4. Under the hybridization conditions used, synthesis of antisense strands, as described in embryonic chicken IGF II transcripts, was undetectable.

The recently isolated cysteine- and arginine-rich peptides of the neutrophil primary granule and the murine intestinal Paneth cell, named defensins or corticostatins, have a number of distinct biological properties. We report the effects of three of these peptides HP-1, HP-1-56 and HP-4 on the incorporation of [3H]thymidine into the DNA of the leukemic cell line HL60. HP-1 and HP-1-56, but not HP-4, inhibited DNA synthesis at 1-50 nM without causing cell death. At higher concentrations the effect was reversed, resulting in a small, but statistically significant increase in DNA synthesis at 1 microM. In contrast HP-4 had no effect on HL60 cells at nanomolar concentrations but was strongly cytotoxic at micromolar concentrations.

The erythropoietic factors present in an anephric patient with nearly normal hematocrit were isolated from plasma by reversed-phase and gel permeation HPLC. The most active fraction was purified and the analysis of its N-terminal sequence was identical to the published sequence of the human insulin-like growth factor I (IGF I). Recombinant human IGF I had identical elution positions as the isolated erythropoietic factor on reversed-phase HPLC and the same molecular weight on gel permeation HPLC. Furthermore, hrIGF I stimulated erythroid colony formation in human bone marrow cultures as was previously shown for the isolated human erythropoietic factor. These results suggest that IGF I may replace erythropoietin as a stimulator of erythropoiesis in some patients with anemia and renal failure.

During the isolation and purification of erythroid cell-stimulating factors from fetal tissues and blood, we found that they were almost invariably contaminated with substances that inhibited thymidine incorporation into erythroid cells of fetal bovine liver. We have isolated and partially sequenced three of these inhibitory factors. The first one was a 46-kDa heparin-binding protein from fetal bovine serum with 80% sequence identity with human apolipoprotein H (apo H). Although human apo H had no inhibitory activity on thymidine incorporation, the bovine apo H-like protein inhibited thymidine incorporation with an ID50 of 36 nM. It probably belongs to a group of heparin-binding apolipoproteins such as apo B and E, which have been reported to inhibit hematopoietic cells. The second inhibitor isolated from fetal bovine serum was clearly cytotoxic at a concentration of 1 nM. This 11-kDa peptide seems to be structurally related to the anaphylatoxins. The third inhibitor was isolated from human fetal intestine. The amino-terminal sequence of this protein was nearly identical to the amino-terminal sequence of human phospholipase A2 isolated from pancreas or lung. Bovine liver erythroid cell membranes are particularly sensitive to phospholipases. Since the synthesis and secretion of phospholipase A2 has been reported to be under the control of interleukin-1 or tumor necrosis factor in different cells, it is possible that this enzyme may be secreted locally and play an important role in tissue remodeling during injury or fetal development.

Extracts of human fetal intestine contain factors that can stimulate or inhibit thymidine incorporation into fetal bovine erythroid cells. An inhibitory factor was purified to homogeneity by gel-permeation and reversed-phase high performance liquid chromatography. The inhibitory action was due to cell lysis. The first 25 amino acids of the N-terminal segment were identical to the human lung and pancreatic phospholipase A2. The isolated protein released arachidonic acid from 2-arachidonyl phosphatidylcholine. Porcine phospholipase A2 had the same effects as the intestinal protein, including its tissue-specific lysis of fetal bovine liver erythroid cells. No decrease of thymidine incorporation was seen in fetal bovine intestinal cells, 3T3 cells, or K562 cells incubated with the porcine enzyme. No release of hemoglobin or cell lysis was observed with human erythrocytes or fetal bovine erythrocytes. Porcine and bee phospholipases, which have low sequence homology, are nearly equipotent in inhibiting thymidine incorporation, whereas melittin and beta-bungarotoxin were less active than the pancreatic enzyme. These results support the tissue-specific effects observed with other phospholipases A2. The high sensitivity of liver erythroid cells towards some phospholipases A2 suggest that these enzymes may be involved in the elimination of hepatic erythroid cells at the end of gestation.

A 46 kDa heparin-binding protein which inhibits thymidine incorporation in cultures of fetal calf liver erythroid cells was isolated from fetal bovine serum by affinity chromatography on heparin-Sepharose, ion-exchange chromatography, gel filtration and reversed-phase h.p.l.c. The N-terminal sequence of the first 22 amino acids showed 81% identity with the published sequence of human apolipoprotein H. The isolated protein inhibited thymidine incorporation with an ED50 (concn. producing 50% of maximal effect) of 36 nM. A 100% inhibition of thymidine incorporation and a 40% decrease in cell numbers in cultures of fetal calf erythroid cells were observed at a protein concentration of 840 nM. No effects could be seen in cultures of 3T3 cells used as controls. Human apolipoprotein H had no inhibitory activity in any of the cell cultures tested, suggesting a species-specificity or a different structure or function for the bovine heparin-binding protein.

An 8 kd heparin-binding peptide which stimulates thymidine incorporation in cultures of fetal calf liver erythroid cells was isolated from fetal bovine serum by affinity chromatography on Heparin-Sepharose, ion exchange chromatography, gel filtration and reversed-phase HPLC. The N-terminal sequence of the isolated peptide was identical to the N-terminal sequence of bovine erythrotropin or insulin-like growth factor II (IGF II). The potential heparin-binding site of IGF II is probably situated in the arginine-rich C-peptide region. The affinities of human recombinant IGF I and II were compared with those of apolipoprotein H (a plasma heparin-binding protein) and bovine insulin in a heparin-affinity column. The retention times were in the order: Apolipoprotein H greater than hrIGF II greater than hrIGF I greater than insulin (no retention). This unusual property of IGF II suggests that it may be captured in the extracellular matrix in a similar way to fibroblast growth factor, interleukin 3 or granulocyte/macrophage colony-stimulating factor.

A method is described for the rapid preparation of erythrotropin from commercially available fetal bovine serum. It consists of the reversed-phase extraction of fetal bovine serum using octadecylsilyl-silica cartridges followed by reversed-phase high-pressure liquid chromatography (HPLC). Further purification can be achieved by gel-permeation HPLC. Serum erythrotropin coelutes with erythrotropin I from fetal bovine intestine on reversed-phase HPLC. It has a molecular weight and an amino acid composition very similar to those reported for the intestinal erythrotropins. The easy preparation of serum erythrotropin with specific activities higher than commercially available erythropoietin preparations will facilitate the study of the physiological role of this factor in vivo and in vitro.

The addition of erythropoietin to cell cultures of erythroid cells of human fetal liver resulted in an increased incorporation of thymidine, adenine, and uridine into trichloroacetic acid-insoluble cell fractions and in an increased uptake of adenine and uridine into the cell. Although the effects of testosterone and erythropoietin on heme synthesis in these cells are known to be very similar, there was no effect of testosterone on the total incorporation of radioactive precursors into DNA or RNA. The RNA synthesized after short pulses of radioactive uridine, when analyzed on sucrose gradients containing 1% sodium dodecyl sulfate, consisted of a homogeneous peak sedimenting at 10 plus or minus 2 S, which is quite different from the heterogeneous, high-molecular-weight RNA synthesized under identical conditions in primary cultures of human fetal lung, kidney, or liver parenchymal cells. Addition of testosterone to liver erythroid cells in cultures for 5 hr followed by a 1-hr uridine pulse resulted in a 3-fold increase of RNA species with an average sedimentation coefficient of 14 plus or minus 3 S. The similarity with the sedimentation coefficient of the globin mRNA described in other systems and the high degree of specialization of the erythroid cells suggest that this RNA may be a stable intermediate involved in the synthesis of hemoglobin.

The separation of alpha, beta, G gamma and A gamma globin chains by reversed phase high pressure liquid chromatography has been studied using columns of octadecylsilyl-silica. All separations were performed in solvent mixtures of water and acetonitrile acidified with phosphoric acid or the hydrophobic ionpairing reagent trifluoroacetic acid. The addition of trifluoroacetic acid or the chaotropic agent NaClO4 to the mobile phase increased the resolution and the peak sharpness of the eluted globin chains. The use of decreasing gradients of TFA or NaClO4 and chromatography at 40 degree were useful steps for the separation of alpha and beta globin chains, a prerequisite for the successful application of this method for the prenatal diagnosis of beta-thalassemia. The beta/gamma synthetic ratio obtained from blood samples taken by fetoscopy from normal fetuses and fetuses with beta-thalassemia trait were measured simultaneously by CM-cellulose chromatography and high pressure liquid chromatography. The values obtained by HPLC were very similar but slightly lower than those obtained by CM-cellulose chromatography. The new method is fast and accurate and will prove to be useful for prenatal diagnosis involving globin chain separation.

We have studied the effect of different growth factors on thymidine incorporation in cell cultures of erythroid cells from fetal calf and rat livers, a system which has been used in the past as a bioassay for the purification of erythropoietin and erythropoietin-like factors. Insulin-like growth factor I significantly stimulated thymidine incorporation in calf and rat liver cells. Its action in both cell types was practically identical to the effect of bovine serum erythrotropin, a peptide structurally similar to insulin-like growth factor II. The synergistic effect between insulin-like growth factor I and erythropoietin could be observed with partially purified sheep plasma erythropoietin but not with recombinant human erythropoietin. The highest thymidine incorporation was observed when both erythropoietin and insulin-like growth factor I were added simultaneously. Platelet-derived growth factor had a lower thymidine incorporation-stimulating activity than insulin-like growth factor and did not have any synergistic effect with erythropoietin in rat liver cells. Fibroblast growth factor (0.4-6.0 ng/ml) was completely inactive in the thymidine incorporation assay of calf liver cells. Transforming growth factor-beta alone at a concentration of 1 ng/ml significantly inhibited thymidine incorporation into rat liver cells. It seems that from all growth factors tested so far, those belonging to the insulin family of peptides are the most likely to be detected in the thymidine incorporation assays using calf or rat liver cells.

A bioassay based on the measurement of thymidine incorporation into trichloroacetic acid-insoluble materials in erythroid cell suspensions from fetal calf liver was used as the assay for purification of two small peptides (erythrotropins I and II) from fetal calf intestine. The peptides were purified using reversed-phase and gel permeation high performance liquid chromatography (HPLC). The two peptides have very similar amino acid compositions and a molecular weight of about 3500 daltons. Erythrotropin II stimulated thymidine incorporation and potentiated the action of erythropoietin in cultures of erythroid cells from fetal rat liver.

An erythrotropin-like peptide (ELP) and bovine serum erythrotropin (ET) coeluted after more than four chromatographic steps. ELP was separated from ET on gel permeation high pressure liquid chromatography due to its large molecular weight. The effect of ELP on the stimulation of [3H] Thymidine incorporation in cell cultures of fetal bovine liver was much lower than the effect of ET. The N-terminal amino acid sequence of ELP indicated that it has a group of amino acids identical to portions of the LDL-receptor and the hypothetical proteins BHLF1 and BOLF1 deducted from the DNA sequence of the Epstein-Barr virus. These results are consistent with the idea that cell surface proteins have structural homologies with secreted proteins or some growth factor-related peptides.

Three techniques for analysing hemoglobin synthesis in blood samples obtained by fetoscopy were evaluated. Of the fetuses studied, 12 were not at risk of genetic disorders, 10 were at risk of beta-thalassemia, 2 were at risk of sickle cell anemia and 1 was at risk of both diseases. The conventional method of prenatal diagnosis of hemoglobinopathies, involving the separation of globin chains labelled with a radioactive isotope on carboxymethyl cellulose (CMC) columns, was compared with a method involving globin-chain separation by high-pressure liquid chromatography (HPLC) and with direct analysis of labelled hemoglobin tetramers obtained from cell lysates by chromatography on ion-exchange columns. The last method is technically the simplest and can be used for diagnosing beta-thalassemia and sickle cell anemia. However, it gives spuriously high levels of adult hemoglobin in samples containing nonlabelled adult hemoglobin. HPLC is the fastest method for prenatal diagnosis of beta-thalassemia and may prove as reliable as the CMC method. Of the 13 fetuses at risk for hemoglobinopathies, 1 was predicted to be affected, and the diagnosis was confirmed in the abortus. Of 12 predicted to be unaffected, 1 was aborted spontaneously and was unavailable for confirmatory studies, as were 3 of the infants; however, the diagnosis was confirmed in seven cases and is awaiting confirmation when the infant in 6 months old in one case. Couples at risk of bearing a child with a hemoglobinopathy should be referred for genetic counselling before pregnancy or, at the latest, by the 12th week of gestation so that prenatal diagnosis can be attempted by amniocentesis, safer procedure, with restriction endonuclease analysis of the amniotic fluid cells.

During a study on the role of cyclic nucleotides on the RNA polymerase activities of isolated fetal calf liver nuclei, it was observed that cGMP enhanced the RNA polymerase activity in the presence of Mg++ at low ionic strength, conditions which are appropriate for the measurement of the RNA polymerases I and III. However, the cGMP-dependent stimulation was sensitive to low concentrations of alpha-amanitin, indicating an effect on the activity of the RNA polymerase II. For this reason the effect of cGMP was tested again using purified RNA polymerase II preparations from calf thymus devoid of RNA polymerases I and III. cGMP stimulated activity of the purified RNA polymerase II at concentrations of 1 and 10 nM. cGMP also caused a small but significant stimulation of the protein kinase activity associated with the enzyme preparations of the RNA polymerase used.

The role of isoproterenol in the synthesis of heme associated with hemoglobin has been studied in primary cultures of human fetal liver. This drug (10-10M) stimulated the incorporation of 59Fe into heme associated with hemoglobin in cell cultures of livers obtained from human fetuses 8 to 10 weeks gestation but was inactive in cell cultures prepared from livers of older fetuses. Isoproterenol has its optimal activity at a much earlier time in gestation than that previously reported for testosterone or erythropoietin. The interpretation of these results is that there is present a cell population in human fetal liver early in gestation with the appropriate isoproterenol-receptors and capable of erythroid differentiation after exposure to the drug.

The effect of streptozocin (STZ)-induced maternal diabetes in rats on fetal erythropoiesis was studied in short-term cultures of fetal liver cells at the time of switch from embryonic to adult hemoglobins. Liver erythroid cell functions were monitored by measuring the incorporation of [3H]-uridine in trichloroacetic acid (TCA)-soluble and -insoluble cell fractions and of [3H]-leucine in hemoglobin chains. Fetal liver cells of diabetic rats showed a higher incorporation of [3H]-uridine compared with controls when the cells were obtained from 14-day-old fetuses. However, there were no significant differences in the uptake of uridine when cells were obtained from 16-day-old fetuses. In parallel cell cultures, incorporation of [3H]-leucine into adult and embryonic globin chains was studied by separation of the globin chains by high-performance liquid chromatography (HPLC). The overall globin chain synthesis was higher in the fetuses of diabetic mothers compared with controls on day 14 of gestation. Erythropoietin had similar effects on the stimulation of globin chains in the two groups of fetuses. However, in the fetuses of diabetic mothers, erythropoietin had a specific stimulatory effect on embryonic-type globins that was significantly higher in the fetuses of diabetic mothers compared with controls. Differences between fetuses of control and diabetic mothers completely disappeared at 16 days of gestation. It is concluded that maternal diabetes has an effect on the cells synthesizing embryonic hemoglobins on day 14 of gestation, but by the time the switch from embryonic to adult-type hemoglobins is complete, these differences are abolished.

The effect of sheep plasma erythropoietin preparations on the incorporation of [5(-3)H]uridine into erythroid cells has been studied using cells of fetal calf liver cultured in serum-free medium. The cells were incubated for 20 h with the hormone, followed by a 1-h incubation with [3H]uridine. Erythropoietin caused a 2.5-fold increase in the incorporation of uridine into cold trichloroacetic-acid-insoluble cell extracts and a 70% increase in the incorporation of uridine into the cold acid-soluble cell extracts. The phosphorylated metabolites of labeled uridine present in the cold acid-soluble fraction were analyzed by anion-exchange high performance liquid chromatography (HPLC). Erythropoietin increased the amounts of labeled UDP and UTP per cell. However, the specific activity of UTP and the labeled amounts of UDP-glucose in erythropoietin-treated cells were not significantly different from those in control cell cultures. After chromatography of the crude erythropoietin preparations on reversed-phase and gel-permeation HPLC, there was a perfect coincidence of the fractions stimulating uridine incorporation into acid-soluble and acid-insoluble cell extracts. The protein fractions from crude erythropoietin which stimulated uridine incorporation after purification by reversed-phased HPLC were also able to stimulate globin chain synthesis in fetal calf liver cells. These experiments suggest that the multiple effects on uridine metabolism described above are due to erythropoietin, rather than other proteins contaminating the crude hormone preparations.

alpha-Fetoprotein and the synthesis of heme associated with hemoglobin were measured simultaneously in short-term cultures of human fetal liver cells to correlate the relationship of alpha-fetoprotein to erythroid cell function. Both synthetic processes decreased exponentially during the first 5 days of culture. The use of media supplemented with different batches of fetal calf serum and porcine portal vein serum indicated that the optimal conditions for the production of alpha-fetoprotein were different from those required for the synthesis of heme associated with hemoglobin. Moreover, the alpha-fetoprotein-producing cells could be separated from erythroid cells after velocity sedimentation in Ficoll gradients. Although it is well known that erythropoiesis and alpha-fetoprotein production occur simultaneously during ontogenesis, alpha-fetoprotein itself (0.01-100 micron g/ml) did not stimulate heme synthesis in liver erythroid cells. Erythropoietin did not stimulate alpha-fetoprotein production. It is concluded that there is no cause-effect relationship between alpha-fetoprotein production and erythroid cell fuction in human fetal liver cells and that the two processes occur independently in different cell types.

A serum-free culture system of erythroid cells of fetal rabbit liver was used for the determination of the erythropoietin-like activity present in fetal calf plasma. The activity found was too high to be explained solely by the presence of erythropoietin. For this reason we decided to investigate the nature of factors present in fetal calf tissues which could have an erythropoietin-like effect in a serum-free medium. The use of fetal calf liver cells and the replacement of the 59Fe incorporation with thymidine incorporation into acid-insoluble materials improved the sensitivity of the assay. This method was used for the rapid screening for the presence of erythropoietin-like factors in reversed-phase extracts of livers, kidneys, lungs, intestines and brains from fetal calves. Intestine extracts stimulated thymidine incorporation and were therefore used as the source for the isolation by reversed-phase and gel permeation high performance liquid chromatography (HPLC) of two active factors (1 and 2). They had different retention times on reversed-phase HPLC but had very similar amino acid compositions and a molecular weight of about 3500 daltons. Both factors added to cultures of calf liver cells caused an increase of alpha and gamma globin chain synthesis equivalent to that observed in the presence of erythropoietin. However, there were important functional differences between both factors and erythropoietin in a rat liver bioassay of thymidine incorporation similar to the one described above. Factors 1 or 2 could not increase the number of cells after a 20h incubation whereas erythropoietin caused a modest increase in cell numbers. Both factors were less potent than erythropoietin in stimulating thymidine incorporation in rat liver cells. Finally, factors 1 and 2 were synergistic with erythropoietin. It is concluded that fetal calf intestine contains factors acting on erythroid cells which have different physical and biological properties from those of erythropoietin. The names erythrotropins I and II are proposed for these factors.

A reversed-phase high performance liquid chromatography (HPLC) method for the direct separation of globin chains from cell lysates of peripheral blood and fetal liver from rat fetuses is described. Partial amino acid analysis of the globins eluted from the HPLC columns as well as comparison with the known elution positions of the adult globin chains in carboxymethyl cellulose chromatography indicated that alpha-chains are eluted first, followed by adult beta chains. The last chains to be eluted are considered as embryonic globins because of their absence in adult rats and their rapid disappearance from the liver after the 14th day of gestation. Liver erythroid cells isolated from rat fetuses on day 14 of gestation mainly synthesized alpha-chains and embryonic globin chains, whereas cells prepared from 16-day old fetuses synthesized almost exclusively alpha-chains and adult beta chains. When the fetal rat liver cells were cultured for 20h with erythropoietin there was a significant stimulation in the synthesis of alpha-globins and adult beta chains but not on the synthesis of embryonic globin chains. It is concluded that HPLC can be useful for the study of rat globin chain synthesis during fetal development, because it separates the globin chains in the three groups of globins, namely alpha, beta and embryonic chains which are important in the switch occurring in the liver.
