Precise gamma-ray spectral analysis is crucial in high-stakes applications, such as nuclear security. Research efforts toward implementing machine learning (ML) approaches for accurate analysis are limited by the resemblance of the training data to the testing scenarios. The underlying spectral shape of synthetic data may not perfectly reflect measured configurations, and measurement campaigns may be limited by resource constraints. Consequently, ML algorithms for isotope identification must maintain accurate classification performance under domain shifts between the training and testing data. To this end, four different classifiers (Ridge, Random Forest, Extreme Gradient Boosting, and Multilayer Perceptron) were trained on the same dataset and evaluated on twelve other datasets with varying standoff distances, shielding, and background configurations. A tailored statistical approach was introduced to quantify the similarity between the training and testing configurations, which was then related to the predictive performance. Wilcoxon signed-rank tests revealed that the OVR-wrapped XGB significantly outperformed the other algorithms, with confidence levels of 99.0% or above for the 133Ba, 60Co, 137Cs, and 152Eu sources. The findings from this work are significant as they outline techniques to promote the development of robust ML-based approaches for isotope identification.

Abstract Purpose Trailing-edge populations at the low-latitude, receding edge of a shifting range face high extinction risk from climate change unless they are able to track optimal environmental conditions through dispersal. Methods We fit dispersal models to the locations of 3165 individually-marked black-throated blue warblers (Setophaga caerulescens) in the southern Appalachian Mountains in North Carolina, USA from 2002 to 2023. Black-throated blue warbler breeding abundance in this population has remained relatively stable at colder and wetter areas at higher elevations but has declined at warmer and drier areas at lower elevations. Results Median dispersal distance of young warblers was 917 m (range 23–3200 m), and dispersal tended to be directed away from warm and dry locations. In contrast, adults exhibited strong site fidelity between breeding seasons and rarely dispersed more than 100 m (range 10–1300 m). Consequently, adult dispersal kernels were much more compact and symmetric than natal dispersal kernels, suggesting adult dispersal is unlikely a driving force of declines in this population. Conclusion Our findings suggest that directional natal dispersal may mitigate fitness costs for trailing-edge populations by allowing individuals to track changing climate and avoid warming conditions at warm-edge range boundaries.

Abstract Obtaining rigorous baseline density estimates of species of conservation interest is key when assisting landowners to achieve management goals on private lands. Northern bobwhite (Colinus virginianus) populations are declining throughout their range and despite being the focus of numerous private land conservation initiatives, baseline density estimates in privately owned pine forests are lacking. We sought to address this knowledge gap across the Southeastern United States by sampling 105 privately owned pine stands throughout 2018 to 2020 using observer point count and autonomous recording unit (ARU) sampling data. Using Bayesian hierarchical models, we investigated the influence of stand management (brush management or applied fire) on bobwhite density, as well as four landscape‐scale environmental variables. These included percentage cover of forest, herbaceous, agricultural or burnt land area across six different spatial scales ranging from 500‐m to 10‐km around each pine stand. Baseline density on sites with no management was estimated to be 2.24 coveys per 100 ha (1.00–5.03, 95% BCI), with little impact of applying brush management, but a trend for a positive effect of fire management (0.19, −0.01 to 0.38 95% BCI). This impact of fire was seen at both the stand‐scale, correlated with an increase in acreage of applied prescribed burn management, and across the greater landscape area, correlated with cover of burnt area within a 2‐km buffer around each site. There were also strong positive influences of herbaceous vegetation and a strong negative influence of forest cover on bobwhite density. Practical implication: our sampling efforts fill an important information gap regarding densities throughout private lands in the Southeastern United States. Our study also highlights the necessity of landscape scale planning for Northern Bobwhite conservation initiatives because the efficacy of conservation practices (i.e. prescribed fire and brush management) could be altered by the landscape surrounding the treated forest stand.

Free-ranging nilgai antelope (Boselaphus tragocamelus) are an understudied species, both on their native ranges of India, Pakistan, and Nepal and on their introduced ranges in southern Texas. Basic data related to population sizes, survival, reproduction, and recruitment are needed throughout their range to inform management and conservation decisions. We collected nilgai fetuses from 3 ranches in southern Texas, including East Foundation’s El Sauz and Santa Rosa ranches, and the Norias Division of the King Ranch® from 2018–2021. We calculated the percentage of individuals that were pregnant in each of the sample years and overall. We determined monthly average, maximum, and minimum fetus length. Of 488 nilgai cows, we found 386 to be pregnant (79%) and 214 to be pregnant with twins (56%). We found nilgai cows as young as 1-year old to have fetuses and therefore to have reached sexual maturity. Sex ratios of fetuses during any sampling year did not differ. We found ample evidence supporting our hypothesis that nilgai are fecund on their introduced range of southern Texas. To prevent nilgai overpopulation and associated problems, harvest management strategies should be implemented, specifically on nilgai cows.

LC-MS/MS analyses have been reported as challenging for the reliable separation and quantification of cyanogenic glycosides (CNGs), especially (R)-prunasin and sambunigrin isomers found in American elderberry (Sambucus nigra L. subsp. canadensis (L.) Bolli). Hence, a novel multiple reaction monitoring (MRM)-based ultra-high-performance liquid chromatography–tandem mass spectrometry (UHPLC-MS/MS) method was developed and validated in the present study for simultaneous separation and quantification of five CNGs, including amygdalin, dhurrin, linamarin, (R)-prunasin, and (S)-prunasin (commonly referred to as sambunigrin). Initially, the role of ammonium formate was investigated as an aqueous mobile-phase additive in developing MRM-based UHPLC-MS/MS. Later, chromatographic conditions for the resolved separation of (R)-prunasin and sambunigrin were identified. Validation studies confirmed that the developed method has good linearity and acceptable precision and accuracy. A noticeable matrix effect (mainly signal enhancement) was observed in leaf samples only. This method was used to detect and quantify CNGs, including (R)-prunasin and sambunigrin, in leaf and fruit samples of American elderberry. Among the studied CNGs, only (R)-prunasin was detected in the leaf samples. Interestingly, (S)-prunasin (sambunigrin) was not detected in the samples analyzed, even though it has been previously reported in elderberry species.

Decreased genetic diversity and possible inbreeding depression have recently been documented in the last wild ocelot (Leopardus pardalis) population in the United States. One consequence of inbreeding depression in felids may be reduced semen quality which can adversely affect reproductive potential. Detailed assessments of reproductive parameters in wild individuals and populations can be conducted using assisted reproductive technologies, such as semen collection and analysis. For most felid species, semen has traditionally been collected via electroejaculation (EEJ22 Electroejaculation); however, an alternative method has been developed using alpha-2 agonist drugs to induce direct sperm release into the urethra, allowing collection by catheterization without requiring specialized equipment. The goal of this study was to characterize normative reproductive traits in free-ranging ocelots and co-occurring bobcats (Lynx rufus) in southern Texas and assess the effectiveness of urethral catheterization (UC33 Urethral Catheterization) for semen recovery in both species. For semen collection, free-ranging cats were live-captured and anesthetized using intramuscular ketamine and medetomidine/dexmedetomidine (alpha-2 agonist) with UC conducted 20–40 minutes post-induction. In ocelots only, EEJ was subsequently performed if UC failed to recover a viable sample. Semen collection was attempted in 31 felids (n=9 ocelots; n=22 bobcats), with sperm recovery by UC in seven of nine ocelots (78 %) and 14 of 22 bobcats (66 %), and by EEJ in four of five ocelots (80 %). Abstract Small water bodies (i.e., ponds; 0.01 km2). We leverage high‐resolution (3 m) optical satellite imagery from Planet Labs and deep learning methods to map seasonal changes in pond and lake areal extent across four regions in Alaska. Our water maps indicate that changes in open water extent over the snow‐free season are especially pronounced in ponds. To investigate potential impacts of seasonal changes in pond area on carbon emissions, we provide a case study of open water methane emission budgets using the new water maps. Our approach has widespread applications for water resources, habitat and land cover change assessments, wildlife management, risk assessments, and other biogeochemical modeling efforts.

Although the current campaign finance system in the US allows private donations to campaigns, an increasing body of evidence suggests that these contributions influence policy and could potentially lead to social injury. This leads to an important question: Do campaign contributions constitute crime? The present article takes up this question. After an overview of the famous debate between Tappan and Sutherland on the role of social injury in determining whether something is a crime—and a concise summary of different types of political crime (e.g. corruption, bribery, state crime, and state-corporate crime)—an analysis is conducted to assess the connection, if any, between campaign contributions and two types of social injury: economic harm/inequality and environmental harm. Findings from the analysis show that campaign contributions can, indeed, cause social injury. It is therefore concluded that campaign contributions sometimes constitute crime under Sutherland’s framework. Numerous campaign finance reform options are discussed—all with the intent of limiting the social injury created by campaign contributions.

Abstract The alkylating agent, temozolomide (TMZ), is the most commonly used chemotherapeutic for the treatment of glioblastoma (GBM). The anti-glioma effect of TMZ involves a complex response that includes G2-M cell cycle arrest and cyclin-dependent kinase 1 (CDK1) activation. While CDK1 phosphorylation is a well-described consequence of TMZ treatment, we find that TMZ also robustly induces CDK1 expression. Analysis of this pathway demonstrates that CDK1 is regulated by NF-κB via a putative κB-site in its proximal promoter. CDK1 was induced in a manner dependent on mature p50 and the atypical inhibitor κB protein, BCL-3. Treatment with TMZ induced binding of NF-κB to the κB-site as assessed by gel shift analysis and chromatin immunoprecipitation. Examination of a CDK1 promoter-reporter demonstrated the functional relevance of the κB-site and underlined the requirement of p50 and BCL-3 for activation. Targeted knockdown of CDK1 or chemical inhibition with the selective CDK1 inhibitor, RO-3306, potentiated the cytotoxic effect of TMZ. These results identify CDK1 as an NF-κB target gene regulated by p50 and BCL-3 and suggest that targeting CDK1 may be a strategy to improve the efficacy of TMZ against GBM.

ABSTRACT Free-ranging white-tailed deer (Odocoileus virginianus) across the United States are increasingly recognized for infection and transmission of SARS-CoV-2. Through a cross-sectional study of 80 deer at three captive cervid facilities in central and southern Texas, we provide evidence of 34 of 36 (94.4%) white-tailed deer at a single captive cervid facility seropositive for SARS-CoV-2 by neutralization assay (PRNT90), with endpoint titers as high as 1,280. In contrast, all tested white-tailed deer and axis deer (Axis axis) at two other captive cervid facilities were seronegative, and SARS-CoV-2 RNA was not detected in respiratory swabs from deer at any of the three facilities. These data support transmission among captive deer that cannot be explained by human contact for each infected animal, as only a subset of the seropositive does had direct human contact. The facility seroprevalence was more than double of that reported from wild deer, suggesting that the confined environment may facilitate transmission. Further exploration of captive cervids and other managed animals for their role in the epizootiology of SARS-CoV-2 is critical for understanding impacts on animal health and the potential for spillback transmission to humans or other animal taxa. IMPORTANCE As SARS-CoV-2 vaccine coverage of the human population increases and variants of concern continue to emerge, identification of the epidemiologic importance of animal virus reservoirs is critical. We found that nearly all (94.4%) of the captive white-tailed deer at a cervid facility in central Texas had neutralizing antibodies for SARS-CoV-2. This seroprevalence is over double than that which has been reported from free-ranging deer from other regions of the United States. Horizontal transmission among deer may be facilitated in confinement. Tracking new infections among wild and confined deer is critical for understanding the importance of animal reservoirs for both veterinary and human health.

p50 is a constitutively produced NF-κB subunit that modulates the response to DNA damage. Here, the authors show that activation of ATR during S phase induces p50 interaction with BARD1 resulting in p50 mono-ubiquitination, facilitating cell cycle progression and promoting chromosome integrity.

Abstract Background Nuclear factor-κB (NF-κB) plays a prominent role in promoting inflammation and resistance to DNA damaging therapy. We searched for proteins that modulate the NF-κB response as a prerequisite to identifying novel factors that affect sensitivity to DNA damaging chemotherapy. Results Using streptavidin-agarose pull-down, we identified the DExD/H-box RNA helicase, DDX39B, as a factor that differentially interacts with κB DNA probes. Subsequently, using both RNA interference and CRISPR/Cas9 technology, we demonstrated that DDX39B inhibits NF-κB activity by a general mechanism involving inhibition of p65 phosphorylation. Mechanistically, DDX39B mediates this effect by interacting with the pattern recognition receptor (PRR), LGP2, a pathway that required the cellular response to cytoplasmic double-stranded RNA (dsRNA). From a functional standpoint, loss of DDX39B promoted resistance to alkylating chemotherapy in glioblastoma cells. Further examination of DDX39B demonstrated that its protein abundance was regulated by site-specific sumoylation that promoted its poly-ubiquitination and degradation. These post-translational modifications required the presence of the SUMO E3 ligase, PIASx-β. Finally, genome-wide analysis demonstrated that despite the link to the PRR system, DDX39B did not generally inhibit interferon-stimulated gene expression, but rather acted to attenuate expression of factors associated with the extracellular matrix, cellular migration, and angiogenesis. Conclusions These results identify DDX39B, a factor with known functions in mRNA splicing and nuclear export, as an RNA-binding protein that blocks a subset of the inflammatory response. While these findings identify a pathway by which DDX39B promotes sensitization to DNA damaging therapy, the data also reveal a mechanism by which this helicase may act to mitigate autoimmune disease.

Northern high-latitude lakes are critical sites for carbon processing and serve as potential conduits for the emission of permafrost-derived carbon and greenhouse gases. However, the fate and emission pathways of permafrost carbon in these systems remain uncertain. Here, we used the natural abundance of radiocarbon to identify and trace the predominant sources of methane, carbon dioxide, dissolved inorganic and organic carbon in nine lakes within the Yukon Flats National Wildlife Refuge in interior Alaska, a discontinuous permafrost region with high landscape heterogeneity and susceptibility to climate, permafrost, and hydrological changes. We find that although Yukon Flats lakes primarily process young carbon (modern to 1290 ± 60 years before present), permafrost-derived carbon is present in some of the sampled lakes and contributes, at most, 30 ± 10% of the dissolved carbon in lake surface waters. Apportionment of young carbon and legacy carbon (carbon with radiocarbon age ⩾5000 years before present) is decoupled among the dissolved inorganic and organic carbon species, with methane showing a stronger legacy signature. Our observations suggest that permafrost-thaw-related transport of carbon through Yukon Flats lacustrine ecosystems and into the atmosphere is small, and likely regulated by surficial sediments, permafrost distribution, wildfire occurrence, or masked by contemporary carbon processes. The heterogeneity of lakes across our study area and northern landscapes more broadly cautions against using any one region (e.g. Yedoma permafrost lakes) to upscale their contribution across the pan-Arctic.

Rapid warming in Arctic tundra may lead to drier soils in summer and greater lightning ignition rates, likely culminating in enhanced wildfire risk. Increased wildfire frequency and intensity leads to greater conversion of permafrost carbon to greenhouse gas emissions. Here, we quantify the effect of recent tundra fires on the creation of methane (CH _4 ) emission hotspots, a fingerprint of the permafrost carbon feedback. We utilized high-resolution (∼25 m ^2 pixels) and broad coverage (1780 km ^2 ) airborne imaging spectroscopy and maps of historical wildfire-burned areas to determine whether CH _4 hotspots were more likely in areas burned within the last 50 years in the Yukon–Kuskokwim Delta, Alaska, USA. Our observations provide a unique observational constraint on CH _4 dynamics, allowing us to map CH _4 hotspots in relation to individual burn events, burn scar perimeters, and proximity to water. We find that CH _4 hotspots are roughly 29% more likely on average in tundra that burned within the last 50 years compared to unburned areas and that this effect is nearly tripled along burn scar perimeters that are delineated by surface water features. Our results indicate that the changes following tundra fire favor the complex environmental conditions needed to generate CH _4 emission hotspots. We conclude that enhanced CH _4 emissions following tundra fire represent a positive feedback that will accelerate climate warming, tundra fire occurrence, and future permafrost carbon loss to the atmosphere.

We derive the spatial and wavelength behavior of dust attenuation in the multiple-armed spiral galaxy VV 191b using backlighting by the superimposed elliptical system VV 191a in a pair with an exceptionally favorable geometry for this measurement. Imaging using the James Webb Space Telescope and Hubble Space Telescope spans the wavelength range 0.3–4.5 μ m with high angular resolution, tracing the dust in detail from 0.6–1.5 μ m. Distinct dust lanes continue well beyond the bright spiral arms, and trace a complex web, with a very sharp radial cutoff near 1.7 Petrosian radii. We present attenuation profiles and coverage statistics in each band at radii 14–21 kpc. We derive the attenuation law with wavelength; the data both within and between the dust lanes clearly favor a stronger reddening behavior ( R = A _V / E _B _− _V ≈ 2.0 between 0.6 and 0.9 μ m, approaching unity by 1.5 μ m) than found for starbursts and star-forming regions of galaxies. Power-law extinction behavior ∝ λ ^− ^β gives β = 2.1 from 0.6–0.9 μ m. R decreases at increasing wavelengths ( R ≈ 1.1 between 0.9 and 1.5 μ m), while β steepens to 2.5. Mixing regions of different column density flattens the wavelength behavior, so these results suggest a different grain population than in our vicinity. The NIRCam images reveal a lens arc and counterimage from a background galaxy at z ≈ 1, spanning 90° azimuthally at 2.″8 from the foreground elliptical-galaxy nucleus, and an additional weakly lensed galaxy. The lens model and imaging data give a mass/light ratio M / L _B = 7.6 in solar units within the Einstein radius 2.0 kpc.

Little information exists on songbird roosting habits and the types of habitats that songbirds select. To better understand a species' habitat requirements, all aspects of its biology should be studied. The Cerulean Warbler (Setophaga cerulea, Wilson) is a Nearctic-Neotropical migrant that is a species of conservation concern across its range. It is one of the fastest declining species of wood-warbler (Parulidae) in North America. Since 2007, a breeding population of Cerulean Warblers has been monitored in southern Indiana, as part of the Hardwood Ecosystem Experiment. The Hardwood Ecosystem Experiment is a 100 yr project that studies the effects of different forest management practices on plant and animal communities. During the 2017 breeding season, 10 male Cerulean Warblers were tracked to roost locations. Roost sites selected by male Cerulean Warbler were characterized with less basal area, higher canopy cover, greater grapevine (Vitis spp., L.) presence, less shrubs, steeper slopes, and less white oak (Quercus alba, L.) abundance. With this new knowledge we can incorporate additional features of habitat in the formulation of a management plan for this declining species.

Beavers have established themselves as a key component of low arctic ecosystems over the past several decades. Beavers are widely recognized as ecosystem engineers, but their effects on permafrost-dominated landscapes in the Arctic remain unclear. In this study, we document the occurrence, reconstruct the timing, and highlight the effects of beaver activity on a small creek valley confined by ice-rich permafrost on the Seward Peninsula, Alaska using multi-dimensional remote sensing analysis of satellite (Landsat-8, Sentinel-2, Planet CubeSat, and DigitalGlobe Inc./MAXAR) and unmanned aircraft systems (UAS) imagery. Beaver activity along the study reach of Swan Lake Creek appeared between 2006 and 2011 with the construction of three dams. Between 2011 and 2017, beaver dam numbers increased, with the peak occurring in 2017 (n = 9). Between 2017 and 2019, the number of dams decreased (n = 6), while the average length of the dams increased from 20 to 33 m. Between 4 and 20 August 2019, following a nine-day period of record rainfall (>125 mm), the well-established dam system failed, triggering the formation of a beaver-induced permafrost degradation feature. During the decade of beaver occupation between 2011 and 2021, the creek valley widened from 33 to 180 m (~450% increase) and the length of the stream channel network increased from ~0.6 km to more than 1.9 km (220% increase) as a result of beaver engineering and beaver-induced permafrost degradation. Comparing vegetation (NDVI) and snow (NDSI) derived indices from Sentinel-2 time-series data acquired between 2017 and 2021 for the beaver-induced permafrost degradation feature and a nearby unaffected control site, showed that peak growing season NDVI was lowered by 23% and that it extended the length of the snow-cover period by 19 days following the permafrost disturbance. Our analysis of multi-dimensional remote sensing data highlights several unique aspects of beaver engineering impacts on ice-rich permafrost landscapes. Our detailed reconstruction of the beaver-induced permafrost degradation event may also prove useful for identifying degradation of ice-rich permafrost in optical time-series datasets across regional scales. Future field- and remote sensing-based observations of this site, and others like it, will provide valuable information for the NSF-funded Arctic Beaver Observation Network (A-BON) and the third phase of the NASA Arctic-Boreal Vulnerability Experiment (ABoVE) Field Campaign.

Background Fledglings of many mature forest-dependent Neotropical songbirds move from mature forest habitats into areas of thick vegetation such as regenerating clearcuts. The Cerulean Warbler (Setophaga cerulea), a Neotropical migratory songbird, is a species of conservation concern across its range and it is listed as endangered in Indiana. This species has declined faster than any other species of wood-warbler in North America. Most prior research on Cerulean Warblers has examined the breeding biology, but there are no data on habitat use by fledgling Cerulean Warblers. Our research aimed to determine where fledgling Cerulean Warblers dispersed after they left their nest, but before they migrated to their wintering grounds. Methods Since 2007, Cerulean Warbler breeding populations have been monitored in Yellowwood and Morgan–Monroe state forests in southern Indiana as part of a 100-year study called the Hardwood Ecosystem Experiment. To identify habitats used by fledgling Cerulean Warblers, we captured by hand or mist-nets, adult and juvenile Cerulean Warblers once young had fledged from a nest. We attached radio-transmitters to individuals and tracked each bird daily using radio-telemetry. Radio-telemetry data were collected from May to July 2015–2017, and microhabitat data on fledgling locations and random locations were collected during the same years in the month of July. Results Fledgling presence, when compared to random non-use sites, was positively correlated to presence of grapevines, greater vertical vegetation density, and greater ground and canopy cover. Fledgling presence was negatively correlated with white oak abundance, aspect, basal area, and the abundance of mature trees that Cerulean Warbler adults use for nesting. Conclusions Our study is the first to demonstrate that Cerulean Warbler fledglings occupy habitats that are characterized by specific habitat components. Fledgling sites were located in areas with high vegetation density, such as clusters of grapevine, which provided cover from predators. Identifying Cerulean Warbler habitats throughout the breeding season can better inform natural resource personnel on how to manage forests to meet the habitat needs of this rapidly declining migratory songbird.

Modafinil has been used as an adjunctive medication in the treatment of bipolar 1 depression with reported success. Case reports have been published demonstrating modafinil induced mania in bipolar patients and modafinil induced psychosis in schizophrenic patients. To our knowledge, we report the only case of modafinil induced psychosis in a patient with bipolar depression treated with both mood stabilizers and antipsychotics. In addition, it is the quickest onset to psychosis (2 days) at the lowest dosage of modafinil (100 mg/day) reported in the literature. Although favorable outcomes using modafinil for treatment of bipolar depression have been reported in literature, clinicians should remain cautious of the potential to rapidly induce psychosis with modafinil at low dosages in patients with bipolar depression despite being treated with mood stabilizers and antipsychotics.

Background Nitric oxide donors are widely used to treat cardiovascular disease, but their major limitation is the development of tolerance, a multifactorial process to which the in vivo release of nitric oxide is thought to contribute. Here we describe the preclinical and clinical results of a translational drug development effort to create a next‐generation nitric oxide donor with improved pharmacokinetic properties and a unique mechanism of nitric oxide release through CYP3A4 metabolism that was designed to circumvent the development of tolerance. Methods and Results Single‐ and multiple‐dose studies in telemetered dogs showed that MK‐8150 induced robust blood‐pressure lowering that was sustained over 14 days. The molecule was safe and well tolerated in humans, and single doses reduced systolic blood pressure by 5 to 20 mm Hg in hypertensive patients. Multiple‐dose studies in hypertensive patients showed that the blood‐pressure–lowering effect diminished after 10 days, and 28‐day studies showed that the hemodynamic effects were completely lost by day 28, even when the dose of MK‐8150 was increased during the dosing period. Conclusions The novel nitric oxide donor MK‐8150 induced significant blood‐pressure lowering in dogs and humans for up to 14 days. However, despite a unique mechanism of nitric oxide release mediated by CYP3A4 metabolism, tolerance developed over 28 days, suggesting that tolerance to nitric oxide donors is multifactorial and cannot be overcome solely through altered in vivo release of nitric oxide. Mountain caribou are an endangered ecotype of woodland caribou (Rangifer tarandus caribou) that live in highprecipitation, mountainous ecosystems of southeastern British Columbia and northern Idaho. The distribution and abundance of these caribou have declined dramatically from historical figures. Results from many studies have indicated that mountain caribou rely on old conifer forests for several life-history requirements including an abundance of their primary winter food, arboreal lichen, and a scarcity of other ungulates and their predators. These old forests often have high timber value, and understanding mountain caribou ecology at a variety of spatial scales is thus required to develop effective conservation strategies. Here we summarize results of studies conducted at three different spatial scales ranging from broad limiting factors at the population level to studies describing the selection of feeding sites within seasonal home ranges of individuals. The goal of this multi-scale review is to provide a more complete picture of caribou ecology and to determine possible shifts in limiting factors across scales. Our review produced two important results. First, mountain caribou select old forests and old trees at all spatial scales, signifying their importance for foraging opportunities as well as conditions required to avoid alternate ungulates and their predators. Second, relationships differ across scales. For example, landscapes dominated by roads and edges negatively affect caribou survival, but appear to attract caribou during certain times of the year. This juxtaposition of fine-scale behaviour with broad-scale vulnerability to predation could only be identified through integrated multi-scale analyses of resource selection. Consequently we suggest that effective management strategies for endangered species require an integrative approach across multiple spatial scales to avoid a focus that may be too narrow to maintain viable populations. Mange studier har vist at denne økotypen er avhengig av vinterføden hengelav i gammel barskog hvor det også er få andre klovdyr og dermed få predatorer. Slik skog er også viktige hogstområder, og å forstå økologien til fjellvillreinen i forskjellige skaleringer er derfor nødvendig for å utvikle forvaltningsstrategier som kan berge og ta vare på denne reinen. Artikkelen gir en oversikt over slike arbeider: fra studier av begrensende faktorer på populasjonsnivå til studier av sesongmessige beiteplasser på individnivå. Hensikten er å få frem et mer helhetlig perspektiv på fjellvillreinen og finne hvordan de begrensende faktorene varierer etter skaleringen som er benyttet i studiet. Oversikten vår frembragte to viktige resultater; 1) Uansett skalering så velger dyrene gammel skog og gamle trær. 2) Dyrenes bruk av et område kan variere med benyttet skalering, for eksempel vil landskap utbygd med veier og hogstflater være ufordelaktig for overlevelsen, men synes likevel å kunne tiltrekke fjellvillreinen til visse tider av året. Forholdet mellom atferd ut fra fin-skalering og stor-skalering sårbarhet hva gjelder predasjon, ville kun blitt avdekket ved flere-skaleringsanalyse av hvordan ressursene benyttes. Ut fra dette foreslår vi at forvaltningsstrategier for truete bestander som eksempelvis fjellvillreinen, må baseres på tilnærminger ut fra ulike skaleringer for å hindre at et for snevert perspektiv kan begrense muligheten for vedvarende levedyktighet.

The Purcell Mountains of southeast British Columbia support a population of mountain caribou near the southernmost extension of their range. This ecotype is dependent upon late-successional forests, largely because such stands provide arboreal lichen for winter forage. Recent provincial forest practices legislation and land-use planning initiatives have provided the impetus for developing an interim caribou habitat assessment model for use as a planning tool. We applied an HSI (habitat suitability index) model developed for a nearby population as a testable hypothesis of caribou habitat selection in the southern Purcells. In a study area of about 6000 km2, 512 radiolocations were obtained for 22 animals from 1993 through 1995. Seasonal selectivity was assessed for the following model variables: elevation, slope, habitat type/current cover type, overstory size class, canopy closure, and age of dominant overstory. Caribou were most selective for stand age, which the model also defined as the greatest determinant of habitat suitability. However, we did not judge overall model output to be an adequate predictor of habitat selection by southern Purcell caribou. Seasonal ratings for each variable were therefore modified to better reflect selection patterns by animals in this study, and subjectively adjusted to ensure that potentially limiting habitat types were rated highly. An evaluation of the adjusted model established its efficacy as an interim decision-support tool. Selection analyses of spatial habitat distribution levels indicated a preference by caribou for landscapes with at least 40% suitable habitat per 250 ha and per 5000 ha. From this, it is apparent that suitable habitat is highly fragmented in this study area.

Alkylating chemotherapy is a central component of the management of glioblastoma (GBM). Among the factors that regulate the response to alkylation damage, NF-κB acts to both promote and block cytotoxicity. In this study, we used genome-wide expression analysis in U87 GBM to identify NF-κB-dependent factors altered in response to temozolomide and found the long noncoding RNA (lncRNA) MALAT1 as one of the most significantly upregulated. In addition, we demonstrated that MALAT1 expression was coregulated by p50 (p105) and p53 via novel κB- and p53-binding sites in the proximal MALAT1 coding region. Temozolomide treatment inhibited p50 recruitment to its cognate element as a function of Ser329 phosphorylation while concomitantly increasing p53 recruitment. Moreover, luciferase reporter studies demonstrated that both κB and p53 cis-elements were required for efficient transactivation in response to temozolomide. Depletion of MALAT1 sensitized patient-derived GBM cells to temozolomide cytotoxicity, and in vivo delivery of nanoparticle-encapsulated anti-MALAT1 siRNA increased the efficacy of temozolomide in mice bearing intracranial GBM xenografts. Despite these observations, in situ hybridization of GBM specimens and analysis of publicly available datasets revealed that MALAT1 expression within GBM tissue was not prognostic of overall survival. Together, these findings support MALAT1 as a target for chemosensitization of GBM and identify p50 and p52 as primary regulators of this ncRNA. SIGNIFICANCE: These findings identify NF-κB and p53 as regulators of the lncRNA MALAT1 and suggest MALAT1 as a potential target for the chemosensitization of GBM.

Despite the global significance of the subsurface biosphere, the degree to which it depends on surface organic carbon (OC) is still poorly understood. Here, we compare stable and radiogenic carbon isotope compositions of microbial phospholipid fatty acids (PLFAs) with those of in situ potential microbial C sources to assess the major C sources for subsurface microorganisms in biogeochemical distinct shallow aquifers (Critical Zone Exploratory, Thuringia Germany). Despite the presence of younger OC, the microbes assimilated 14C-free OC to varying degrees; ~31% in groundwater within the oxic zone, ~47% in an iron reduction zone, and ~70% in a sulfate reduction/anammox zone. The persistence of trace amounts of mature and partially biodegraded hydrocarbons suggested that autochthonous petroleum-derived hydrocarbons were a potential 14C-free C source for heterotrophs in the oxic zone. In this zone, Δ14C values of dissolved inorganic carbon (-366 ± 18‰) and 11MeC16:0 (-283 ± 32‰), an important component in autotrophic nitrite oxidizers, were similar enough to indicate that autotrophy is an important additional C fixation pathway. In anoxic zones, methane as an important C source was unlikely since the 13C-fractionations between the PLFAs and CH4 were inconsistent with kinetic isotope effects associated with methanotrophy. In the sulfate reduction/anammox zone, the strong 14C-depletion of 10MeC16:0 (-942 ± 22‰), a PLFA common in sulfate reducers, indicated that those bacteria were likely to play a critical part in 14C-free sedimentary OC cycling. Results indicated that the 14C-content of microbial biomass in shallow sedimentary aquifers results from complex interactions between abundance and bioavailability of naturally occurring OC, hydrogeology, and specific microbial metabolisms.

Pronghorn (Antilocapra americana) are considered a keystone species of North American grasslands and an important economic source for many landowners in Texas. Pronghorn restoration projects routinely capture and translocate individuals from surplus populations to restoration areas. The objective of this study was to generate normal hematological and biochemical reference intervals (RI) for free-ranging pronghorn populations in Texas as a health monitoring tool for pronghorn restoration efforts. Blood samples were collected by jugular venipuncture and divided among an EDTA tube, serum separator tube, and a single blood smear on site. Complete blood counts and biochemical profiles were completed at the Texas Veterinary Medical Diagnostic Laboratory. In total, 417 individuals (41 males, 376 females) were included in the analysis. RI were determined by robust methods (R Studio) and mixed models' analysis of variance (SPSS 28) to examine differences in blood parameters due to fever, sex, age (adult versus yearling [<1 yr of age]), cell abnormalities, and pathogen exposure reported by the testing laboratory. Sex, age, and pathogen exposure affected mean blood values, but did not warrant development of separate RI by class. Bluetongue virus was identified in 46.8% (195/417) of pronghorns and epizootic hemorrhagic disease in 89.4% (194/217) of pronghorns; 84.8% (184/217) of the pronghorns tested positive for both diseases. This information provides baseline hematology and biochemical parameters to assess the health of free-ranging pronghorn and guide wildlife managers in decision-making for future translocations and restoration objectives. Precise gamma-ray spectral analysis is crucial in high-stakes applications, such as nuclear security. Research efforts toward implementing machine learning (ML) approaches for accurate analysis are limited by the resemblance of the training data to the testing scenarios. The underlying spectral shape of synthetic data may not perfectly reflect measured configurations, and measurement campaigns may be limited by resource constraints. Consequently, ML algorithms for isotope identification must maintain accurate classification performance under domain shifts between the training and testing data. To this end, four different classifiers (Ridge, Random Forest, Extreme Gradient Boosting, and Multilayer Perceptron) were trained on the same dataset and evaluated on twelve other datasets with varying standoff distances, shielding, and background configurations. A tailored statistical approach was introduced to quantify the similarity between the training and testing configurations, which was then related to the predictive performance. Wilcoxon signed-rank tests revealed that the OVR-wrapped XGB significantly outperformed the other algorithms, with confidence levels of 99.0% or above for the 133Ba, 60Co, 137Cs, and 152Eu sources. Simple Summary: To better inform nilgai antelope management decisions, basic life history data are needed. We collected reproductive tracts of free-ranging female nilgai from 2018 to 2021 in Southern Texas. We found high pregnancy and twinning rates. Moreover, we found nilgai as young as 1 year old to be pregnant and nilgai as old as 12 years old to be pregnant. Our findings demonstrate the high reproductive potential of free-ranging nilgai in Southern Texas. To prevent nilgai overpopulation and associated damage to the land, harvest management strategies should be used, particularly in introduced areas where there are no natural predators. Free-ranging nilgai antelope (Boselaphus tragocamelus) are an understudied species, both on their native ranges of India, Pakistan, and Nepal and on their introduced ranges in southern Texas. Basic data related to population sizes, survival, reproduction, and recruitment are needed throughout their range to inform management and conservation decisions. We collected nilgai fetuses from 3 ranches in southern Texas, including East Foundation's El Sauz and Santa Rosa ranches, and the Norias Division of the King Ranch® from 2018–2021. We calculated the percentage of individuals that were pregnant in each of the sample years and overall. We determined monthly average, maximum, and minimum fetus length. Of 488 nilgai cows, we found 386 to be pregnant (79%) and 214 to be pregnant with twins (56%). We found nilgai cows as young as 1-year old to have fetuses and therefore to have reached sexual maturity. Sex ratios of fetuses during any sampling year did not differ. We found ample evidence supporting our hypothesis that nilgai are fecund on their introduced range of southern Texas.
