3. A step towards measuring connectivity in the deep-sea: elemental fingerprints of mollusk larval shells discriminate hydrothermal vent sites

Abstract

Deep-sea hydrothermal-vent systems are under investigation for base and precious metal exploitations. The impact of mining will depend critically on the ability of larval dispersal to connect and replenish endemic populations. However, assessing connectivity is extremely challenging, especially in the deep sea. Here, we investigate the potential of elemental fingerprinting of mollusc larval shells to discriminate larval origins between multiple hydrothermal sites in the Southwest Pacific Ocean. The gastropodShinkailepas tollmannirepresents a suitable candidate as it uses capsules to hold larvae before dispersal, which facilitates sampling. Multielemental microchemistry was performed using cutting-edge femtosecond laser ablation Inductively Coupled Plasma Mass Spectrometry analysis to obtain individual measurements on 600 encapsulated larval shells. We used classification methods to discriminate the origin of individuals from 14 hydrothermal sites spanning over 3,500 km, with an overall success rate of 70%. When considering less sites within more restricted areas, reflecting dispersal distances reported by genetic and modelling approaches, the success rate increased up to 86%. We conclude that individual larval shells register site-specific elemental signatures that can be used to assess their origin. These results open new perspectives to get direct estimates on population connectivity from the geochemistry of pre-dispersal shell of recently settled juveniles.

Abstract

A multimethod approach aimed at characterizing carbonate parietal deposits and at proposing a chronology for these carbonate crusts is described. Dating was performed by radiometric methods (C-14 for recent samples and U series) on samples that had been characterized beforehand using optical and cathodoluminescence microscopy and Fourier transform infrared microspectroscopy. For the U series, high precision on U–Th ages was achieved using liquid phase multicollector inductively coupled plasma mass spectrometry (ICP-MS) applied to large samples, while laser ablation single collector inductively coupled plasma sector field mass spectrometry (ICP-SFMS) provided information on the reliability of the sampling with a high spatial resolution. This methodology, based on the combination of these two techniques reinforced by the information obtained by the calcite characterization methods, was applied to carbonate deposits from the cave of Trou du Renard (Soyons, France). The ages obtained with the two U–Th dating techniques are comparable and illustrate that different laminae were deposited at different rates in the samples. In the future, this procedure, based on the mineralogical and geochemical characterization of the samples and their dating by radiometric methods, will be applied to the layers of parietal carbonates deposited on Palaeolithic decorated walls. When the crystallization is slow, the U/Th dating method by imaging technique is of interest, as well as that by multicollector ICP-MS in liquid phase. Abstract

The specific stock composition and dispersion of anadromous fish species aggregations in the marine environment are poorly known, while they can play a major role in the metapopulation dynamics. Otolith microchemistry has proven to be a powerful tool to address natal origins of anadromous fish. We used archived otolith microchemistry to investigate the population-specific composition of subadult European shads (Alosa alosa and Alosa fallax) in the ocean during the 1980s. The allocation of natal origin was addressed relying on contemporary water and juveniles' signatures within a Bayesian model. A great discrimination of natal origin was obtained at the Biscay Gulf scale. However, the discrimination of 1980s natal origin for the southern rivers with similar geology based on 2013 water and juveniles' baselines was doubtful. Our results showed that the most abundant southern populations were dominant, suggesting that population-specific composition was related to population relative abundance. The dispersion in the marine environment was plastic; alternatively, shads were found large distances away from their natal rivers, while others remained in the vicinity of their natal river plume. La composition specifique de stocks et la dispersion de regroupements d'especes de poissons anadromes dans le milieu marin demeurent meconnues, bien qu'elles puissent jouer un important role dans la dynamique de metapopulation. La microchimie des otolithes s'avere etre un outil puissant pour examiner les origines natales de poissons anadromes. Nous utilisons la microchimie d'otolithes archives pour examiner la composition propre a la population d'aloses europeennes (Alosa alosa and Alosa fallax) sous-adultes dans l'ocean dans les annees 1980. L'affection de l'origine natale repose sur des signatures d'eau et de juveniles contemporaines dans un modele bayesien. Une excellente discrimination de l'origine natale est obtenue a l'echelle du golfe de Gascogne. Cependant, la discrimination de l'origine natale dans les annees 1980 pour les rivieres meridionales de geologie semblable basee sur des donnees de reference de 2013 pour l'eau et les juveniles est incertaine. Abstract

A method has been developed for the direct and simultaneous multielement determination of Cu, Zn, Sn, and Pb in soil and sediment samples using femtosecond laser ablation inductively coupled plasma mass spectrometry (fs-LA-ICPMS) in combination with isotope dilution mass spectrometry (IDMS). The in-cell isotope dilution fs-LA-ICPMS method proposed in this work was based on the quasi-simultaneous ablation of the natural abundance sample and the isotopically enriched solid spike, which was performed using a high repetition rate laser and a fast scanning beam device in a combined manner. Both the sample preparation procedure and the total analysis time have been drastically reduced, in comparison with previous approaches, since a unique multielement isotopically enriched solid spike was employed to analyze different powdered samples. Numerous experimental parameters were carefully selected (e.g., carrier gas flow rate, inlet diameter of the ablation cell, sample translation speed, scanner speed, etc.) in order to ensure the complete mixing between the sample and the solid spike aerosols. The proposed in-cell fs-LA-ICP-IDMS method was tested for the analysis of two soil (CRM 142R, GBW-07405) and two sediment (PACS-2, IAEA-405) reference materials, and the analysis of Cu, Zn, Sn, and Pb yielded good agreement of usually not more than 10% deviation from the certified values and precisions of less than 15% relative standard deviation. Furthermore, the concentrations were in agreement not only with the certified values but also with those obtained by ICP-IDMS after the microwave-assisted digestion of the solid samples, demonstrating therefore that in-cell fs-LA-ICP-IDMS opens the possibility for accurate and precise determinations of trace elements in powdered samples reducing the total sample preparation time to less than 5 min. Additionally, scanning electron microscope measurements showed that the aerosol generated by in-cell fs-LA-ICP-IDMS predominantly consisted of linear agglomerates of small particles (in the order of few tens of nanometers) and a few large spherical particles with diameters below 225 nm.

Abstract

We report the results of a multiproxy study that combines structural analysis of a fracture–stylolite network and isotopic characterization of calcite vein cements and/or fault coating. Together with new paleopiezometric and radiometric constraints on burial evolution and deformation timing, these results provide a first-order picture of the regional fluid systems and pathways that were present during the main stages of contraction in the Tuscan Nappe and Umbria–Marche Apennine Ridge (northern Apennines). We reconstruct four steps of deformation at the scale of the belt: burial-related stylolitization, Apenninic-related layer-parallel shortening with a contraction trending NE–SW, local extension related to folding, and late-stage fold tightening under a contraction still striking NE–SW. We combine the paleopiezometric inversion of the roughness of sedimentary stylolites – that constrains the range of burial depth of strata prior to layer-parallel shortening – with burial models and U–Pb absolute dating of fault coatings in order to determine the timing of development of mesostructures. In the western part of the ridge, layer-parallel shortening started in Langhian time (∼15 Ma), and then folding started at Tortonian time (∼8 Ma); late-stage fold tightening started by the early Pliocene (∼5 Ma) and likely lasted until recent/modern extension occurred (∼3 Ma onward). The textural and geochemical (δ18O, δ13C, Δ47CO2 and 87Sr∕86Sr) study of calcite vein cements and fault coatings reveals that most of the fluids involved in the belt during deformation either are local or flowed laterally from the same reservoir. However, the western edge of the ridge recorded pulses of eastward migration of hydrothermal fluids (>140 ∘C), driven by the tectonic contraction and by the difference in structural style of the subsurface between the eastern Tuscan Nappe and the Umbria–Marche Apennine Ridge.

Abstract

A laser ablation-ICPMS method using an infrared (1030 nm), low-energy (39 [micro]J/pulse), high repetition rate (10 kHz), femtosecond laser was developed to improve the sensitivity of detection of heteroatom-containing proteins in 1D polyacrylamide gels. A 2-mm-wide lane was ablated by ultrafast (10 cm [s.sup.-1]) back-and-forth movement of a 20-[micro]m laser beam parallel to the protein bands while the gel advanced perpendicularly. This procedure resulted in a considerable increase in detection sensitivity (> 40-fold) compared to the nanosecond 266-nm laser ablationICPMS, mainly because of the much larger amount of ablated material introduced into the plasma on the time scale of the dwell time of the mass spectrometer. The method was applied to the specific detection in the gel of formate dehydrogenase expressed in Escherichia coli and of selenoproteins in Desulfococcus multivorans with detection limits at the low-femtomolar levels.

However, the western edge of the ridge recorded pulses of eastward migration of hydrothermal fluids (>140 ∘C), driven by the tectonic contraction and by the difference in structural style of the subsurface between the eastern Tuscan Nappe and the Umbria–Marche Apennine Ridge.

International audience; Les datations U-Th (méthode radiométrique), bien que théorisées depuis les années 1950 etlargement employées en géologie, ne sont encore que très peu appliquées en archéologie.Alors qu’une large gamme de matériaux archéologiques est en principe datable par cette méthode(coquilles, dents, dépôts calcitiques etc), son application fait aujourd’hui face à trois limitationsmajeures. La première est que le protocole actuel requiert une grande quantité de matière(plusieurs dizaines de mg) - quantité souvent indisponible pour ces échantillons petits et uniques. Ladifficulté est alors de rendre compte des potentielles incorporations et/ou départs en radioéléments,ces matériaux se présentant fréquemment comme des systèmes ouverts. Enfin, les limites dedétection des instruments aujourd’hui dédiés aux datations U-Th ne permettent pas l’étude dematériaux ne contenant que des traces voire ultra-traces d’uranium.Le protocole présenté ici a été développé pour lever ces verrous technologiques, tirant parti desavantages offerts par le couplage d’un laser UV femtoseconde à haute cadence de tir et d’unspectromètre de masse haute résolution. Les mesures isotopiques directes des ultra-traces (quelquesatogrammes) d’uranium et de thorium permettent de réaliser des cartographies isotopiques, icisur des échantillons de nature diverses issus de sites préhistoriques clés dans l’évolution humaine.Ainsi, des datations U-Th peuvent être réalisées, directement à partir de ces images, de manièrerapide (quelques heures), et ce grâce à des quantités infimes de matières (100 à 1000 fois moinsque pour les protocoles couramment utilisés), le tout en rendant compte de la distribution spatialede ces isotopes au sein du matériau, à l’échelle de quelques micromètres seulement. Dès lors, leprotocole de datation U-Th que nous avons développé ici permet d’appréhender les datations parles séries de l’uranium d’une nouvelle manière, sur une grande quantité d’échantillons négligésou rejetés jusqu’à présent.

80ko hamarkadaz geroztik, laser bidezko ablazioa eta akoplamendu induktibozko plasma-masa espektrometria (LA-ICPMS) konbinatzen dituen teknika analitikoa sakonki ikertu eta garatu da, eta, gaur egun, lagin solidoen analisi elemental zein isotopiko zuzena egiteko erreferentziazko teknika bilakatu da. Ezaugarri bereizgarrienak hauexek dira: erabilpen erraza, analisi-denbora laburra, laginari eragindako kalte urriak, sentsibilitate handia eta elementu nagusi zein minoritarioen eta ratio isotopikoen aldibereko neurketa ahalbidetzen duen tarte dinamiko zabala. Lan honetan, LA-ICPMS teknikaren oinarriak eta konfigurazio orokorra azaldu eta femtosegundo/ nanosegundo laser bidezko ablazioaren erabileraren arteko desberdintasun nagusiak nabarmentzen dira. Azkenik, teknika honen egungo egoeraz jabetzeko, jatorri askotariko laginen analisi elementala laburbildu eta berrikusten da biologian eta ingurumenean, osasunean, geokimikan zein auzitegi zientzien alorretan.; Since the early 80s, laser-ablation inductively-coupled-plasma mass spectrometry (LA-ICPMS) has been widely explored and nowadays is considered to be one of the most versatile analytical techniques for the direct trace elemental and isotopic analyses of solid samples. The most remarkable features are ease of use, fast sample throughput, limited sample damages, high sensitivity and wide dynamic range which allows the simultaneous acquisition of major and trace elements, as well as the measurement of isotope ratios. Throughout this manuscript, the principles and general setup of LA-ICPMS will be extensively explained and the performance of femtosecondLA in comparison to nanosecond-LA discussed. In order to show the state of the art in this field a variety of examples for elemental analysis of solid samples in biological/environmental science, geochemistry and forensic science applications will be presented.

Abstract

Laser ablation inductively coupled plasma mass spectrometry is more and more considered as a tool of choice for solid analysis. The use of femtosecond laser sources permit to improve analytical performances of the coupling thanks to the non-thermal ablation obtained in comparison with the more conventional nanosecond laser used. Through a collaboration between the LCABIE (Laboratoire de Chimie Analytique Bio-Inorganique et Environnement) of Pau and Novalase SA (a company specialized in optical and laser integration), the first integrated femtosecond laser ablation system was designed in 2003. Its unusual characteristics (high repetition rate and scanning beam device) allow to perform some new ablation strategies and open encouraging analytical perspectives in terms of sensitivity and quantification. The potential of this prototype was evaluated through fundamental analyses such as elemental fractionation and characterization of the aerosol produced by laser ablation and through the development of different applications such as analysis of soils and sediments by in-cell isotope dilution, detection of selenoproteins in gel electrophoresis, and determination of uranium isotopic composition of micrometric particles.; L'ablation laser couplée à une détection par spectrométrie de masse à plasma induit apparait de plus en plus comme un outil de choix pour l'analyse des solides. L'utilisation de sources laser femtoseconde permet d'améliorer les performances analytiques du couplage, l'interaction laser-matière privilégiant les phénomènes athermiques contrairement aux lasers nanoseconde plus classiquement utilisés. Une collaboration entre le Laboratoire de Chimie Analytique Bio-Inorganique et Environnement de Pau et Novalase SA (spécialisé dans l'intégration optique et laser) a fait naitre la première station d'ablation laser femtoseconde intégrée. Ses caractéristiques particulières, que sont la haute cadence de tir et un optique de balayage rapide du faisceau, permettent d'explorer de nouvelles stratégies d'ablation et ouvrent des perspectives analytiques encourageantes en terme de sensibilité et de mode de quantification. Le potentiel de ce prototype a été évalué au travers d'analyses fondamentales, telles que le fractionnement élémentaire et l'étude de la taille des particules de l'aérosol produit par ablation, et de différentes applications telles que l'analyse de sols et de sédiments par dilution isotopique réalisée par le laser dans la chambre d'ablation, la détection de sélénoprotéines dans des gels d'électrophorèse et la détermination de l'isotopie de l'uranium dans des particules micrométriques.
