The effectiveness of state-of-the-art systemic treatments for neurological disorders is hampered not only by the difficulty in crossing the blood brain barrier but also off-target drug interactions. In this study, a delivery method is simulated for a novel U-shaped microtube locally infusing drugs directly into the extracellular space of the brain and relying on diffusion as a transport mechanism. The influence of flow rate, drug properties and device geometry are investigated. It is anticipated that these findings will accelerate progress on both developmental and applied drug delivery and materials research. Graphical abstract

Bioelectronics hold the key for understanding and treating disease. However, achieving stable, long-term interfaces between electronics and the body remains a challenge. Implantation of a bioelectronic device typically initiates a foreign body response, which can limit long-term recording and stimulation efficacy. Techniques from regenerative medicine have shown a high propensity for promoting integration of implants with surrounding tissue, but these implants lack the capabilities for the sophisticated recording and actuation afforded by electronics. Combining these two fields can achieve the best of both worlds. Here, we show the construction of a hybrid implant system for creating long-term interfaces with tissue. We create implants by combining a microelectrode array with a bioresorbable and remodellable gel. These implants are shown to produce a minimal foreign body response when placed into musculature, allowing us to record long-term electromyographic signals with high spatial resolution. Electronics that change shape inside the body enable keyhole surgery of spinal implants that can treat severe pain., Spinal cord stimulation is one of the oldest and most established neuromodulation therapies. However, today, clinicians need to choose between bulky paddle-type devices, requiring invasive surgery under general anesthetic, and percutaneous lead–type devices, which can be implanted via simple needle puncture under local anesthetic but offer clinical drawbacks when compared with paddle devices. By applying photo- and soft lithography fabrication, we have developed a device that features thin, flexible electronics and integrated fluidic channels. This device can be rolled up into the shape of a standard percutaneous needle then implanted on the site of interest before being expanded in situ, unfurling into its paddle-type conformation. The device and implantation procedure have been validated in vitro and on human cadaver models. This device paves the way for shape-changing bioelectronic devices that offer a large footprint for sensing or stimulation but are implanted in patients percutaneously in a minimally invasive fashion.

Implantable electrophoretic drug delivery devices have shown promise for applications ranging from treating pathologies such as epilepsy and cancer to regulating plant physiology. Upon applying a voltage, the devices electrophoretically transport charged drug molecules across an ion‐conducting membrane out to the local implanted area. This solvent‐flow‐free “dry” delivery enables controlled drug release with minimal pressure increase at the outlet. However, a major challenge these devices face is limiting drug leakage in their idle state. Here, a method of reducing passive drug leakage through the choice of the drug co‐ion is presented. By switching acetylcholine's associated co‐ion from chloride to carboxylate co‐ions as well as sulfopropyl acrylate‐based polyanions, steady‐state drug leakage rate is reduced up to sevenfold with minimal effect on the active drug delivery rate. Numerical simulations further illustrate the potential of this method and offer guidance for new material systems to suppress passive drug leakage in electrophoretic drug delivery devices., Drug leakage is a safety concern for long‐term implantable electrophoretic drug delivery devices. In this work, co‐ion engineering is proposed as a clinically viable method to drastically reduce drug leakage rate, without any sacrifices on the device active performance. The co‐ion engineering method is drug‐independent and can be readily applied to existing electrophoretic drug delivery devices.

The advancement of bioelectronics depends greatly on new material development and engineering solutions. Redox polymers are promising candidates to contribute to this advancement of biointerfacing devices. For such devices to be clinically useful, they must fulfill an assortment of requirements, including biocompatibility, stability, mechanical compliancy and the ability to effectively monitor or influence biological systems. The use of redox polymers in bioelectronic research has demonstrated a great deal of potential in satisfying these constraints. In this chapter, we consider the advantageous aspects of polymer electronics for biomedical applications including electrophysiological recording, neuromodulation, biosensor technologies and drug delivery. Particular emphasis is given to PEDOT-based systems as these have demonstrated the highest degree of bioelectronic device success to date, however, other polymers are also discussed when pertinent.

Here we report recent advances in precise engineering to incorporate advanced electrophoretic drug delivery system into implantable devices for treating neurological disorders including both depth probes and cortical arrays with recording capabilities. The efficacy of the platform is demonstrated by stopping epileptic seizures in vivo. This is the first in vivo demonstration of an electrophoretic implant treating a neurological disorder and offers a glimpse of what can be achieved by tailored engineering of electrophoretic devices. We anticipate this work to be the starting point for new stimulation, recording and drug delivery paradigms in chronic neural implantation.

Electrophoretic drug delivery devices are able to deliver drugs with exceptional temporal and spatial precision. This technology has emerged as a promising platform for treating pathologies ranging from neuropathic pain to epilepsy. As the range of applications continues to expand, there is an urgent need to understand the underlying physics and estimate materials and device parameters for optimal performance. Here, computational modeling of the electrophoretic drug delivery device is carried out. Three critical performance indices, namely, the amount of drug transported, the pumping efficiency and the ON/OFF ratio are investigated as a function of initial drug concentration in the device and fixed charge concentration in the ion exchange membrane. The results provide guidelines for future materials and device design with an eye towards tailoring device performance to match disease-specific demands.

Epilepsy is a group of neurological disorders which affects millions of people worldwide. Although treatment with medication is helpful in 70% of the cases, serious side effects affect the quality of life of patients. Moreover, a high percentage of epileptic patients are drug resistant; in their case, neurosurgery or neurostimulation are necessary. Therefore, the major goal of epilepsy research is to discover new therapies which are either capable of curing epilepsy without side effects or preventing recurrent seizures in drug-resistant patients. Neuroengineering provides new approaches by using novel strategies and technologies to find better solutions to cure epileptic patients at risk. As a demonstration of a novel experimental protocol in an acute mouse model of epilepsy, a direct in situ electrophoretic drug delivery system is used. Namely, a neural probe incorporating a microfluidic ion pump (µFIP) for on-demand drug delivery and simultaneous recording of local neural activity is implanted and demonstrated to be capable of controlling 4-aminopyridine-induced (4AP-induced) seizure-like event (SLE) activity. The γ-aminobutyric acid (GABA) concentration is kept in the physiological range by the precise control of GABA delivery to reach an antiepileptic effect in the seizure focus but not to cause overinhibition-induced rebound bursts. The method allows both the detection of pathological activity and intervention to stop seizures by delivering inhibitory neurotransmitters directly to the epileptic focus with precise spatiotemporal control. As a result of the developments to the experimental method, SLEs can be induced in a highly localized manner that allows seizure control by the precisely tuned GABA delivery at the seizure onset.

Local drug delivery directly to the source of a given pathology using retrodialysis is a promising approach to treating otherwise untreatable diseases. As the primary material component in retrodialysis, the semipermeable membrane represents a critical point for innovation. This work presents a new ionic hydrogel based on polyethylene glycol and acrylate with dopamine counterions. The ionic hydrogel membrane is shown to be a promising material for controlled diffusive delivery of dopamine. The ionic nature of the membrane accelerates uptake of cationic species compared to a nonionic membrane of otherwise similar composition. It is demonstrated that the increased uptake of cations can be exploited to confer an accelerated transport of cationic species between reservoirs as is desired in retrodialysis applications. This effect is shown to enable nearly 10-fold increases in drug delivery rates from low concentration solutions. The processability of the membrane is found to allow for integration with microfabricated devices which will in turn accelerate adaptation into both existing and emerging device modalities. We discuss the ramifications of this finding and propose a simple model that describes capacitance in terms of sites in which ions injected from the electrolyte replace holes that are extracted from the film by a metal contact. This study presents a novel brain implant capable of electrophoretically delivering drugs and recording local neural activity on the surface of the brain. The drug delivery is made possible by the integration of a microfluidic ion pump (µFIP) into a conformable electrocorticography (ECoG) device with recording cites embedded next to the drug delivery outlets. The µFIP ECoG device can deliver a high capacity of several biologically important cationic species on demand. The therapeutic potential of the device is demonstrated by using it to deliver neurotransmitters in a rodent model while simultaneously recording local neural activity. These developments represent a significant step forward for cortical drug-delivery systems.

Minimally invasive electrodes of cellular scale that approach a bio-integrative level of neural recording could enable the development of scalable brain machine interfaces that stably interface with the same neural populations over long period of time.In this paper, we designed and created NeuroRoots, a bio-mimetic multi-channel implant sharing similar dimension (10µm wide, 1.5µm thick), mechanical flexibility and spatial distribution as axon bundles in the brain. A simple approach of delivery is reported based on the assembly and controllable immobilization of the electrode onto a 35µm microwire shuttle by using capillarity and surface-tension in aqueous solution. Once implanted into targeted regions of the brain, the microwire was retracted leaving NeuroRoots in the biological tissue with minimal surgical footprint and perturbation of existing neural architectures within the tissue. NeuroRoots was implanted using a platform compatible with commercially available electrophysiology rigs and with measurements of interests in behavioral experiments in adult rats freely moving into maze. We demonstrated that NeuroRoots electrodes reliably detected action potentials for at least 7 weeks and the signal amplitude and shape remained relatively constant during long-term implantation.This research represents a step forward in the direction of developing the next generation of seamless brain-machine interface to study and modulate the activities of specific sub-populations of neurons, and to develop therapies for a plethora of neurological diseases.

An implantable microfluidic ion pump prevents seizures in a mice by delivery of inhibitory neurotransmitters to the seizure source., The persistence of intractable neurological disorders necessitates novel therapeutic solutions. We demonstrate the utility of direct in situ electrophoretic drug delivery to treat neurological disorders. We present a neural probe incorporating a microfluidic ion pump (μFIP) for on-demand drug delivery and electrodes for recording local neural activity. The μFIP works by electrophoretically pumping ions across an ion exchange membrane and thereby delivers only the drug of interest and not the solvent. This “dry” delivery enables precise drug release into the brain region with negligible local pressure increase. The therapeutic potential of the μFIP probe is tested in a rodent model of epilepsy. The μFIP probe can detect pathological activity and then intervene to stop seizures by delivering inhibitory neurotransmitters directly to the seizure source. We anticipate that further tailored engineering of the μFIP platform will enable additional applications in neural interfacing and the treatment of neurological disorders.

Implantable devices offer an alternative to systemic delivery of drugs for the treatment of neurological disorders. A microfluidic ion pump (µFIP), capable of delivering a drug without the solvent through electrophoresis, is developed. The device is characterized in vitro by delivering γ-amino butyric acid to a target solution, and demonstrates low-voltage operation, high drug-delivery capacity, and high ON/OFF ratio. It is also demonstrated that the device is suitable for cortical delivery in vivo by manipulating the local ion concentration in an animal model and altering neural behavior. These results show that µFIPs represent a significant step forward toward the development of implantable drug-delivery systems.

Organic solar cells utilizing the small molecule donor 7,7′-(4,4-bis(2-ethylhexyl)-4H-silolo[3,2-b:4,5-b′]dithiophene-2,6-diyl)bis(6-fluoro-4-(5′-hexyl-[2,2′-bithiophen]-5-yl)benzo[c][1,2,5] thiadiazole) (p-DTS(FBTTh2)2 and the polymer acceptor poly{[N,N′-bis(2-octyldodecyl)-1,4,5,8-naphthalenedicarboximide-2,6-diyl]-alt-5,5′-(2,2′-bithiophene)}(P(NDI2OD-T2)) are investigated and a power conversion efficiency of 2.1% is achieved. By systematic study of bulk heterojunction (BHJ) organic photovoltaic (OPV) quantum efficiency, film morphology, charge transport and extraction and exciton diffusion, the loss processes in this blend is revealed compared to the blend of [6,6]-phenyl-C71-butyric acid methyl ester (PC71BM) and the same donor. An exciton diffussion study using Forster resonant energy transfer (FRET) shows the upper limit of the P(NDI2OD-T2) exciton diffusion length to be only 1.1 nm. The extremely low exciton diffusion length of P(NDI2OD-T2), in combination with the overlap in donor and acceptor absorption, is then found to significantly limit device performance. These results suggest that BHJ OPV devices utilizing P(NDI2OD-T2) as an acceptor material will likely be limited by its low exciton diffusion length compared to devices utilizing functionalized fullerene acceptors, especially when P(NDI2OD-T2) significantly competes with the donor molecule for photon absorption.

Analysis of measured charge-carrier mobilities and fill factors in solution-processable small-molecule bulk-heterojunction solar cells reveals that in order to achieve a high FF, the hole and electron mobilities must be >10(-4) cm 2 V(-1) s(-1) . Neat-film mobility measurements are also found to be a useful predictor of the maximum blend film mobility and FF obtained in blend film solar cells.

Solution deposited bulk heterojunction organic solar cells are viewed as one of the most promising alternative energy sources because of their ease of processing and their potential to be produced using large scale techniques such as roll-to-roll, newspaper style, coating. Since organic materials have a relatively low dielectric constant the dissociation of an excited electron–hole pair into free collectable charge carriers is inefficient in many cases. Often the excited electron–hole pairs recombine back to the ground state in a process known as geminate recombination before they ever fully dissociate into free charge carriers. Even after dissociation, free holes and electrons can encounter each other once more and subsequently recombine back to the ground state in a process known as nongeminate recombination. In both cases the incident photon energy is lost and fewer carriers are collected at the electrodes. Hence, charge carrier recombination is one of the key loss mechanisms in organic solar cells. In this review the latest on geminate and nongeminate recombination is discussed.

By combining the molecular donor p-DTS(FBTTh2 )2 with a readily produced perylene diimide acceptor we are able to achieve a power conversion efficiency of 3.0%, making this one of the most efficient non-fullerene organic solar cells to date. The reduced power conversion efficiency of the present system compared to the use of phenyl-C71 -butyric acid methyl ester as an electron acceptor is shown to primarily be related to a significant reduction in the internal quantum efficiency. These results indicate the potential of small-molecule:non-fullerene bulk-heterojunction organic photovoltaics.

Morphological control over the bulk heterojunction (BHJ) microstructure of a high-efficiency small molecule photovoltaic system is demonstrated using both thermal treatment and solvent additive processing. Single crystal X-ray diffraction is utilized to understand molecular interactions in the solid state and the BHJ morphology is examined using bright field, high-resolution, and cross-section transmission electron microscopy techniques. Controlling the domain size, while maintaining good molecular order within the semiconducting donor material, is found to be crucial in achieving high performance and over 90% internal quantum efficiency exhibited under the optimized conditions.

Advantages of semiconducting small molecules-as opposed to semiconducting polymers-include synthetic simplicity, monodispersity, low cost, and ease of purification. One purported disadvantage of small-molecule films is reduced mechanical robustness. This paper measures the tensile modulus and crack-onset strain for pure films of the high-performance solution-processable small-molecule donors 7,7'-[4,4-bis(2-ethylhexyl)-4H-silolo[3,2-b:4,5-b']dithiophene-2,6-diyl]bis[6-fluoro-4-(5'-hexyl-[2,2'-bithiophen]-5-yl)benzo[c][1,2,5]thiadiazole] (DTS(FBTTh2)2), 2,5-di(2-ethylhexyl)-3,6-bis(5″-n-hexyl-[2,2',5',2″]terthiophen-5-yl)-pyrrolo[3,4-c]pyrrole-1,4-dione (SMDPPEH), and 6,13-bis(triisopropylsilylethynyl)pentacene (TIPS-pentacene), the acceptor 5,5'-(2,1,3-benzothiadiazole-4,7-diyldi-2,1-ethenediyl)bis[2-hexyl-1H-isoindole-1,3(2H)-dione] (HPI-BT), blends of DTS(FBTTh2)2 and SMDPPEH with [6,6]-phenyl C71 butyric acid methyl ester (PC71BM) and with HPI-BT, and bulk heterojunction films processed with the additives 1,8-diiodooctane (DIO) and polystyrene (PS). The most deformable films of solution-processed organic semiconductors are found to exhibit tensile moduli and crack-onset strains comparable to those measured for conjugated polymers. For example, the tensile modulus of as-cast DTS(FBTTh2)2 is 0.68 GPa (i.e., comparable to poly(3-hexylthiophene) (P3HT), the common polymer), while it exhibits no cracks when stretched on an elastomeric substrate to strains of 14%. While this high degree of stretchability is lost upon the addition of PC71BM (4.2 GPa, 1.42%), it can be partially recovered using processing additives. Tensile modulus and crack-onset strain are highly correlated, which is typical of van der Waals solids. Increased surface roughness was correlated to increased modulus and brittleness within films of similar composition. Decreased stiffness for soluble molecular semiconductors can be rationalized by the presence of alkyl side chains, which decrease the van der Waals attraction between molecules in the crystalline grains. These measurements and observations could have important consequences for the stability of devices based on molecular semiconductors, especially those destined for stretchable or ultraflexible applications, or those demanding mechanical robustness during roll-to-roll fabrication or use in the outdoor environment.

KGaA, Weinheim Studies of the living tissue via optical means are able to monitor biological activities such as metabolism, gene expression, and variations in ionic concentration. Organic optoelectronic devices have numerous advantages over traditional inorganic technologies, yet limited examples of their capabilities exist in biomedical applications. An organic photodetector (OPD) with a simple structure acts as a highly sensitive optical sensor for detecting intrinsic optical signals of a living brain tissue. The signals are related to cell volume variations and are essential in detecting biological events, such as metabolism and hypoxia. This work demonstrates for the first time the capability of OPDs to assess biooptical events in neuroscience. Their simple fabrication and the capability for selective absorption of an optical event via tunable chemistry pave the way for their integration in biomedical prostheses with broad applications in bioelectronics.

Connections are delineated between solar-cell performance, charge-carrier mobilities, and morphology in a highperformance molecular solar cell. The observations show that maximizing the relative phase purity and structural order while simultaneously limiting the domain size may be essential for achieving optimal solar-cell performances in solution-processed small-molecule solar cells .

Electroluminescence (EL) from the charge-transfer state and singlet excitons is observed at low applied voltages from high-performing small-molecule bulk-heterojunction solar cells. Singlet emission from the blends emerges upon altering the processing conditions, such as thermal annealing and processing with a solvent additive, and correlates with improved photovoltaic performance. Low-temperature EL measurements are utilized to access the physics behind the singlet emission.

A mechanistic study of the DNA interfacial layer that enhances the photoresponse in n-type field-effect transistors (FET) and lateral photoconductors using a solution-processed fullerene derivative embedded with disperse-red dye, namely PCBDR, is reported. Incorporation of the thin DNA layer simultaneously leads to increasing the electron injection from non-Ohmic contacts into the PCBDR active layer in dark and to increasing the photocurrent under irradiation. Such features lead to the observation of the enhancement of the photoresponsivity in PCBDR FETs up to 10(3) . Kelvin probe microscopy displays that in the presence of the DNA layer, the surface potential of PCBDR has a greater change in response to irradiation, which is rationalized by a larger number of photoinduced surface carriers. Transient absorption spectroscopy confirms that the increase in photoinduced carriers in PCBDR under irradiation is primarily ascribed to the increase in exciton dissociation rates through the PCBDR/DNA interface and this process can be assisted by the interfacial dipole interaction.

Two new high-performance DPP-containing donor molecules employing a molecular architecture with three DPP chromorphores (tri-DPP) in conjugated backbones are synthesized and characterized. The two tri-DPP molecules with only a structural difference on alkyl substitutions, when blended with PC71 BM, lead to power conversion efficiencies up to 4.8 and 5.5%, respectively.

Charge transport and nongeminate recombination are investigated in two solution-processed small molecule bulk heterojunction solar cells consisting of diketopyrrolopyrrole (DPP)-based donor molecules, mono-DPP and bis-DPP, blended with [6,6]-phenyl-C71-butyric acid methyl ester (PCBM). While the bis-DPP system exhibits a high fill factor (62%) the mono-DPP system suffers from pronounced voltage dependent losses, which limit both the fill factor (46%) and short circuit current. A method to determine the average charge carrier density, recombination current, and effective carrier lifetime in operating solar cells as a function of applied bias is demonstrated. These results and light intensity measurements of the current-voltage characteristics indicate that the mono-DPP system is severely limited by nongeminate recombination losses. Further analysis reveals that the most significant factor leading to the difference in fill factor is the comparatively poor hole transport properties in the mono-DPP system (2 × 10-5 cm2 V-1 s-1 versus 34 × 10-5 cm2 V-1 s-1). These results suggest that future design of donor molecules for organic photovoltaics should aim to increase charge carrier mobility thereby enabling faster sweep out of charge carriers before they are lost to nongeminate recombination. Neuroinspired device architectures offer the potential of higher order functionalities in information processing beyond their traditional microelectronic counterparts. Here we demonstrate a neuromorphic function of orientation selectivity, which is inspired from the visual system, with a combination of organic photodetectors and a multi-gated organic electrochemical transistor based on poly(3,4ethylenedioxythiophene):poly(styrene sulfonate) (PEDOT:PSS). The device platform responds preferably to different orientations of light bars, a behaviour that resembles orientation selectivity of visual cortex cells. These results pave the way for organic-based neuromorphic devices with spatially correlated functionalities and potential applications in the area of organic bioelectronics.

The light intensity dependence of the main photoelectrical parameters of the nonfullerene small-molecule bulk heterojunction (BHJ) solar cells p-DTS(FBTTh2)2:perylene diimide (T1:PDI) shows that the nongeminate recombination losses play an important role in this system. A simple approach for the quantitative analysis of capacitance spectroscopy data of the organic BHJ solar cells, which allows to determine the density of free charge carriers as a function of applied bias under standard working conditions, is demonstrated. Using the proposed capacitance spectroscopic technique, the nongeminate recombination losses in the T1:PDI solar cells are quantitatively characterized in the scope of the bimolecular- and trap-assisted recombination mechanisms. Their contributions are separately analyzed within a wide range of the applied bias.

The field of organic photovoltaics has recently produced highly efficient single-junction cells with power conversion efficiency >10%, yet the open-circuit voltage (VOC) remains relatively low in many high performing systems. An accurate picture of the density of states (DOS) in working solar cells is crucial to understanding the sources of voltage loss, but remains difficult to obtain experimentally. Here, the tail of the DOS is characterized in a number of small molecule bulk heterojunction solar cells from the charge density dependence of VOC, and is directly compared to the disorder present within donor and acceptor components as measured by Kelvin probe. Using these DOS distributions, the total energy loss relative to the charge transfer state energy (ECT)—ranging from ≈0.5 to 0.7 eV—is divided into contributions from energetic disorder and from charge recombination, and the extent to which these factors limit the VOC is assessed.

Whereas the role of molecularly mixed domains in organic photovoltaic devices for charge generation is extensively discussed in the literature, the impact on charge recombination and thus fill factor is largely unexplored. Here, a combination of soft X-ray techniques enables the quantification of phases at multiple length scales to reveal their role regarding charge recombination in a highly efficient solution processed small molecule system 7,7′-(4,4-bis(2-ethylhexyl)-4H-silolo[3,2-b:4,5-b′]dithiophene-2,6-diyl)bis(6-fluoro-4-(5′-hexyl-[2,2′-bithiophen]-5-yl)benzo[c][1,2,5]thiadiazole) (p-DTS(FBTTh2)2). A quantitative (linear) relationship between the average composition variations and the device fill-factor is observed. The results establish the complex interrelationship between average phase purity, domain size, and structural order and highlight the requirement of achieving sufficient phase purities to diminish bimolecular and geminate recombination in solution processed small molecule solar cells.

The effects of copper metalation of tetrabenzoporphyrin on the properties and performance of organic solar cells are studied. Tetrabenzoporphyrin (BP) and copper tetrabenzoporphyrin (CuBP) are both solution processed from soluble precursor materials and thermally converted in the thin film. Despite high field-effect hole mobility above 1 cm2 V−1 s−1, the power conversion efficiency (PCE) of solar cell devices with CuBP is severely diminished compared to those with BP. Conducting atomic force microscopy (c-AFM) is used to show that CuBP films are highly conductive in the direction perpendicular to the substrate, relative to those comprising BP. By analyzing the donor absorption characteristics as well as the external quantum efficiency and short-circuit current density of bilayer OPV devices as a function of donor layer thickness, it is determined that the differences in performance are likely due to a prohibitively short effective exciton diffusion length (LD) in the metalated derivative. By modeling the external quantum efficiency of bilayer OPV devices, we are able to approximate this difference in effective LD to be 15 nm for BP and 2 nm for CuBP.
