Context: Diabetic retinopathy (DR) is one of the leading causes of vision impairment and blindness among diabetic patients globally. Despite advancements in conventional treatments, the quest for more holistic approaches and fewer side effects persists. traditional chinese medicine (tcM) has been used for centuries in managing various diseases, including diabetes and its complications. Objective: this review evaluated the efficacy and underlying mechanisms of tcM in the management of DR, providing information on its potential integration with conventional treatment methods. Methods: A comprehensive literature review was conducted using PubMed, web of Science, and the china National Knowledge infrastructure (cNKi) with the search terms 'traditional chinese medicine', 'diabetic retinopathy', 'clinical efficacies' and their combinations. Studies published before 2023 without language restriction were included, focusing on clinical trials and observational studies that assessed the effectiveness of tcM in DR treatment. Results: the review synthesized evidence of empirical traditional chinese formulas, traditional chinese patent medicines, and isolated phytochemicals on DR treatment. the key mechanisms identified included the reduction of oxidative stress, inflammation, and neovascularization, as well as the improvement in neurovascular functionality and integrity of the retinal blood barrier. Conclusions: tcM shows promising potential to manage DR. More large-scale, randomized controlled trials are recommended to validate these findings and facilitate the integration of tcM into mainstream DR treatment protocols. [ABSTRACT FROM AUTHOR]

Considerable efforts have been devoted to treating gastric ulcers. Attempts in this field tend to develop drug delivery systems with prolonged gastric retention time. Herein, we develop novel Chinese herb pollen-derived micromotors as active oral drug delivery system for treating gastric ulcer. Such Chinese herb pollen-derived micromotors are simply produced by asymmetrically sputtering Mg layer onto one side of pollen grains. When exposed to gastric juice, the Mg layer can react with the hydrogen ions, resulting in intensive generation of hydrogen bubbles to propel the micromotors. Benefiting from the autonomous motion and unique spiny structure, our micromotors can move actively in the stomach and adhere to the surrounding tissues. Besides, their special architecture endows the micromotors with salient capacity of drug loading and releasing. Based on these features, we have demonstrated that our Chinese herb pollen-derived micromotors could effective deliver berberine hydrochloride and show desirable curative effect on the gastric ulcer model of mice. Therefore, these Chinese herb pollen-derived micromotors are anticipated to serve as promising oral drug delivery carriers for clinical applications.

Traditional Chinese Medicine (TCM), originating over 3,000 years ago, incorporates herbal medicine, acupuncture, and other techniques. The term ''carminative'' is derived from the Latin word ''carminare,'' meaning to cleanse. Carminatives can reduce lower esophageal pressure, thereby potentially increasing the susceptibility to gastroesophageal reflux disease or heartburn. Methods: The online databases including Scopus, PubMed, Web of Science, and Google Scholar were searched using different via keywords: Carminatives, Chinese herb, and Herbal. The objective of this review was to provide a concise summary of the herbal ingredients that have been previously documented for their gastro-stimulating properties in carminative products. Results: Medicinal plants are widely recognized as a crucial and valuable resource for the development of pharmaceutical products. These herbal products possess carminative properties and offer additional health benefits in a wide range of therapeutic uses of carminative like as Anti-microbial, Diuretic, Irritable bowel syndrome, etc. Discussion: With the increasing global interest in natural medicines, there is a potential for higher demand for TCM as carminatives. Novel formulations with a blend of TCM can be created to improve their effectiveness and convenience. This review can provide chances for more research and increase understanding of TCM for usage as a carminative. Conclusion: In conclusion traditional Chinese Medicine (TCM) incorporates herbal remedies, like those with carminative properties, to address gastrointestinal discomfort and promote overall well-being. Abstract Honokiol is a neolignan biphenol found in aerial parts of the Magnolia plant species. The Magnolia plant species traditionally belong to China and have been used for centuries to treat many pathological conditions. Honokiol mitigates the severity of several pathological conditions and has the potential to work as an anti-inflammatory, anti-angiogenic, anticancer, antioxidant, and neurotherapeutic agent. It has a long history of being employed in the healthcare practices of Southeast Asia, but in recent years, a greater scope of research has been conducted on it. Plenty of experimental evidence suggests it could be beneficial as a neuroprotective bioactive molecule. Honokiol has several pharmacological effects, leading to its exploration as a potential therapy for neurological diseases (NDs), including Alzheimer's disease (AD), Parkinson’s disease (PD), cerebral ischemia, anxiety, depression, spinal cord injury, and so on. So, based on the previous experimentation reports, our goal is to discuss the neuroprotective properties of honokiol. Besides, honokiol derivatives have been highlighted recently as possible therapeutic options for NDs. So, this review focuses on honokiol's neurotherapeutic actions and toxicological profile to determine their safety and potential use in neurotherapeutics.

Chinese herb Radix sophorae tonkinensis extract oxymatrine shows anticancer effects. This study evaluated the role of oxymatrine in colorectal cancer (CRC) and the underlying molecular events in vitro and in vivo. CRC cells were treated with different doses of oxymatrine to assess cell viability, reactive oxygen species production, gene expression, and gene alterations. Meanwhile, mouse xenograft and liver metastasis models were used to assess the effects of oxymatrine using histology examination, transmission electron microscopy, and Western blot, respectively. Our results showed that oxymatrine treatment triggered CRC cell mitophagy to inhibit CRC cell growth, migration, invasion, and metastasis in vitro and in vivo. At the gene level, oxymatrine inhibited LRPPRC to promote Parkin translocation into the mitochondria and reduce the mitophagy‐activated NLRP3 inflammasome. Thus, oxymatrine had an anticancer activity through LRPPRC inhibition, mitophagy induction, and NLRP3 inflammasome suppression in the CRC cell xenograft and liver metastasis models. In conclusion, the study demonstrates the oxymatrine anti‐ CRC activity through its unique role in regulating CRC cell mitophagy and NLRP3 inflammasome levels in vitro and in vivo. The removal of heavy metals through adsorption represents a highly promising method. This study focuses on the utilization of an abundant cellulose-rich solid waste, licorice residue (LR), as a natural material for hydrogel synthesis. To this end, LR-EPI hydrogels, namely, LR-EPI-5, LR-EPI-6 and LR-EPI-8, were developed by crosslinking LR with epichlorohydrin (EPI), specifically targeting the removal of Pb, Cu, and Cr from aqueous solutions. Thorough characterizations employing Fourier-transform infrared spectroscopy (FTIR) and scanning electron microscopy confirmed the successful crosslinking of LR-EPIs by EPI, resulting in the formation of porous and loosely structured hydrogels. Batch studies demonstrated the high efficacy of LR-EPI hydrogels in removing the three heavy metal ions from aqueous solutions. Notably, LR-EPI-8 exhibited the highest adsorption capacity, with maximum capacities of 591.8 mg/g, 458.3 mg/g, and 121.4 mg/g for Pb2+, Cr3+, and Cu2+, respectively. The adsorption processes for Pb2+ and Cu2+ were well described by pseudo-second-order kinetics and the Langmuir model. The adsorption mechanism of LR-EPI-8 onto heavy metal ions was found to involve a combination of ion-exchange and electrostatic interactions, as inferred from the results obtained through X-ray photoelectron spectroscopy and FTIR. This research establishes LR-EPI-8 as a promising adsorbent for the effective removal of heavy metal ions from aqueous solutions, offering an eco-friendly approach for heavy metal removal and providing an environmentally sustainable method for the reutilization of Chinese herb residues. Introduction: Chinese medicinal herbs play important roles in anti-inflammatory, antioxidant, and antibacterial activities. However, the effects of Chinese herb ultrafine powder (CHUP) on laying hens still need to be elucidated. Therefore, this study aimed to evaluate the effects of dietary CHUP supplementation on jejunal morphology, physical barrier function, and microbiota in laying hens. Methods: A total of 576 Xinyang black-feather laying hens (300 days old) were randomly assigned into eight groups, with eight replicates per group and nine hens per replicate. The hens were fed a basal diet (control group) and a basal diet supplemented with 0.5% Leonuri herba (LH group), 0.25% Ligustri lucidi fructus (LF group), 0.25% Taraxaci herba (TH group), 0.5% LH + 0.25% LF (LH-LF group), 0.5% LH + 0.25% TH (LH-TH group), 0.25% LF + 0.25% TH (LF-TH group), and 0.5% LH + 0.25% LF + 0.25% TH (LH-LF-TH group), respectively, for 120 days. Results: The results showed that dietary LH-LF and LH-LF-TH supplementation increased (p < 0.05) the jejunal villus height to crypt depth ratio of laying hens. Dietary LF-TH supplementation up-regulated jejunal claudin-5 expression, while LH supplementation up-regulated jejunal claudin-1 expression and increased the jejunal abundances of potentially beneficial bacteria related to short-chain fatty acids and bacteriocins production, such as Blautia, Carnobacterium, Clostridiales, and Erysipelotrichales (p < 0.05). In addition, dietary LH supplementation enriched (p < 0.05) the tetracycline biosynthesis, butirosin/neomycin biosynthesis, and D-arginine/D-ornithine metabolism, whereas steroid biosynthesis and limonene/pinene degradation were enriched (p < 0.05) in the LH-LF and LH-LF-TH groups. Moreover, Spearman's correlation analysis revealed the potential correlation between the abundance of the jejunal microbiota and jejunal morphology and the physical barrier function of laying hens. Discussion: Collectively, these findings suggest that dietary CHUP supplementation could enhance the beneficial bacteria abundance, physical barrier function, and metabolic function associated with short-chain fatty acids and bacteriocins production. Upstream treatment of atrial fibrillation (AF, for short) is a new approach to the prevention and treatment of AF with non-antiarrhythmic drugs, which is essentially primary and secondary prevention of AF. The former refers to the prevention of AF by controlling risk factors such as diabetes, hypertension, and heart failure before AF occurs, and the latter mainly refers to targeting ion channels, inflammation, oxidative stress, and other pathways to reduce or reverse atrial electrical and structural remodeling, reduction of AF load, and reduction of the chance of AF occurrence or progression. More and more studies have shown that many traditional Chinese medicines, active ingredients of Chinese medicines, and Chinese herbal formulas have definite effects on the upstream treatment of AF, but their mechanisms of action are different. Therefore, we summarized the relevant literature on the application and mechanisms of Chinese medicine on the upstream treatment of AF in recent years, hoping to be helpful for subsequent studies.

Alpiniae Oxyphyliae Fructus (AOF) (yizhi) is a frequently medicated Chinese herb for Alzheimer disease (AD) treatment. The present study investigated the components and potential mechanisms of AOF through network pharmacology analysis and molecular docking. The results showed that AOF contains at least 20 active ingredients and involves 184 target genes. A total of 301 AD-related genes were obtained from the DisGeNET, GeneCards, GEO, OMIM, and Alzheimer Disease: Genes databases. A total of 41 key targets were identified from the topology analysis of the AOF-AD target network. These key targets are involved in 105 signal pathways, such as the PI3K-Akt, HIF-1, and MAPK pathways, and can regulate gene transcription, cell death, cell proliferation, drug response, and protein phosphorylation. AOF's active ingredients, Chrysin, Isocyperol, Izalpinin, Linolenic acid, CHEMBL489541, Oxyphyllenone A, Oxyphyllenone B, and Oxyphyllol C, show high affinity to targets, including PPARG, ESR1, and AKT1. Eucommia ulmoides Oliver (EuO) is a traditional Chinese herb with effects on repairing disorders of liver lipid metabolism and enhancing immunity in mice. In this study, the effects of EuO on immune enhancement and liver protection of M. salmoides were tested by adding 3% EuO to a high-starch diet and were evaluated by histopathology, enzymology, flow cytometry, and molecular biology. The results showed that the high starch diet promoted hepatocytes apoptosis, damaged the liver, and led to impaired immune function. However, the integrality of liver was recoverd, innate response was enhanced, hepatocyte apoptosis was inhibited, and antioxidant capacity was improved after treatment with EuO. These findings reveal that EuO could improve the immune function while also repairing the M. salmoides liver injury induced by a high starch diet. This study provides a theoretical basis for EuO to solve the liver disease of M. salmoides caused by a high starch diet.

Background: Current data indicates the incidence of neuropathic pain after surgical nerve injury is as high as 50%, thus representing a major problem for patients and for the medical system. Triptolide, a traditional Chinese herb, has anti-inflammatory effects on various neurodegenerative and neuroinflammatory diseases. This agent also reduces peripheral nerve injury-induced neuropathic pain, although the mechanism underlying this effect is still unknown. Materials and Methods: The effects of triptolide on spinal nerve ligation (SNL) injury-induced neuropathic pain was studied in an animal model using behavioral, morphological and molecular biological methods. Results: Repeated administration of intrathecal triptolide was found to alleviate SNL- or Poly(I:C) (toll-like receptor 3 agonist) injectioninduced mechanical allodynia without any motor impairment. The mechanism by which triptolide reduces SNL- and Poly(I:C) injectioninduced microglial activation appears to be via the inhibition of OX42 expression, which is a microglial-specific marker. Intrathecal triptolide also suppressed SNL- and Poly(I:C) injection-induced expression of spinal TRIF. TRIF transmits signals from activated TLR3 and is the downstream adaptor of TLR3 in microglia. In addition, intrathecal triptolide inhibited the expression of spinal pro-inflammatory IL-1β following SNL or Poly(I:C) injection. Conclusions: Intrathecal triptolide can suppress the TLR3/TRIF/IL-1β pathway in spinal microglia following SNL. In addition, the plasma estradiol level in the L-LF, LF-T, and L-LF-T groups and unsaturated fatty acids concentrations in egg yolk of the CHUP groups (except LF-T group) were increased, whereas total cholesterol (T, L-LF, L-T, and L-LF-T groups) in egg yolk and the atherogenicity (T, L-T, and L-LF-T groups) and thrombogenicity (T, L-LF, L-T, and L-LF-T groups) indexes were decreased ( p  < 0.05) on day 60 of the trial compared with the control group. Moreover, bitter amino acids in egg albumen were decreased ( p  < 0.05) in the L-LF group on day 60 and the L-LF-T group on day 120 of the trial. Collectively, these findings indicate that dietary CHUP supplementation could improve eggshell quality and increase plasma reproductive hormone, fatty acid and amino acid composition, and nutritional values of eggs, especially L-LF and L-LF-T.

Traditional Chinese medicine and Chinese herbs have a demonstrated value for disease therapy and sub-health improvement. Attempts in this area tend to develop new forms to make their applications more convenient and wider. Here, we propose a novel Chinese herb microneedle (CHMN) patch by integrating the herbal extracts, Premna microphylla and Centella asiatica, with microstructure of microneedle for wound healing. Such path is composed of sap extracted from the herbal leaves via traditional kneading method and solidified by plant ash derived from the brine induced process of tofu in a well-designed mold. Because the leaves of the Premna microphylla are rich in pectin and various amino acids, the CHMN could be imparted with medicinal efficacy of heat clearing, detoxicating, detumescence and hemostatic. Besides, with the excellent pharmaceutical activity of Asiatic acid extracted from Centella asiatica, the CHMN is potential in promoting relevant growth factor genes expression in fibroblasts and showing excellent performance in anti-oxidant, anti-inflammatory and anti-bacterial activity. Taking advantages of these pure herbal compositions, we have demonstrated that the derived CHMN was with dramatical achievement in anti-bacteria, inhibiting inflammatory, collagen deposition, angiogenesis and tissue reconstruction during the wound closure. These results indicate that the integration of traditional Chinese herbs with progressive technologies will facilitate the development and promotion of traditional Chinese medicine in modern society.

Patients and Methods: A total of 108 patients with primary osteopenic LBP were randomly divided into the JYMGG group and placebo group. Both groups took 600 mg of oral Caltrate D daily; in addition, the JYMGG group was given oral JYMGG, while the placebo group was given placebo granules. The treatment period was 6 months for both groups. The pre- to post-treatment changes in the bone mineral density (BMD), visual analogue scale (VAS) score, Oswestry disability index (ODI), and bone turnover markers were compared between the two groups. Results: The post-treatment VAS score and ODI were significantly lower than baseline in both groups (P< 0.05). In the JYMGG group, the lumbar BMD increased from 0.88± 0.07 g/cm2 to 0.90± 0.13 g/cm2 and the hip BMD increased from 0.77± 0.08 g/cm2 to 0.78± 0.10 g/cm2, giving increases of 2.70% and 1.96% respectively, but the differences were not statistically significant. The post-treatment levels of ALP, osteocalcin, P1NP, and β-CTX were increased compared with baseline in both groups, but the differences were not statistically significant. The thyrotropin level was significantly increased after treatment in the placebo group (P< 0.05). There were no abnormalities detected in routine blood and kidney function tests performed during the observation period. Some patients showed elevated liver enzymes and gastrointestinal reactions. Conclusion: JYMGG effectively relieved the bone pain, and improved the quality of life of patients with primary osteopenic LBP. [ABSTRACT FROM AUTHOR]

An effective analytical method to quantify glyphosate (Gly), aminomethylphosphonic acid (AMPA) and glufosinate (Glu) in four kinds of Chinese herbs by derivatization was developed. The method was studied in four steps: extraction, purification, derivatization and enrichment. The extraction and purification steps were optimized. The sample was added with isotope-labeled internal standards and extracted with 30% acetonitrile. The MCX solid-phase extraction cartridge was chosen to reduce interferences and the purification solution was derivatized with 9-fluoromethoxycarbonyl chloride. 4 M HCl was selected as reaction terminator. The supernatant was enriched by salting-out using NaCl and analyzed by LC–MS/MS in heated positive electrospray ionization mode by multiple reactions monitoring. To verify the method methodically, matrix effect, linearity, limits of quantitation and detection, accuracy and precision all met SANTE/12682/2019 requirements. The modified method was applied to 152 batches of 4 different Chinese herbs. The overall detection rate of Gly was 79.61%, AMPA was 72.37% and Glu was 16.45%. [ABSTRACT FROM AUTHOR]

TaohongSiwu decoction (THSWD), a traditional Chinese recipe, has been widely used to treat hypertension since ancient times. However, the mechanisms of its action are still unclear. Herein, we aimed to explore the gut microbial activity of THSWD in high-salt diet-induced hypertensive mice. Eight percent high-salt (NaCl) diet was used to induce hypertension for 4 weeks in a mouse model. Meanwhile, THSWD was used to intervene in the high-salt diet-induced mice, and the efficacy was evaluated by different parameters. Here, we found that THSWD significantly restored blood pressure compared with the model group. Moreover, THSWD effectively protected endothelial function by significantly upregulating the level of nitric oxide (NO) and downregulating the level of endothelin-1 (ET-1), angiotensin I (AngI), and vascular endothelial growth factor (VEGF) in serum compared with the model group. Notably, THSWD significantly upregulated the relative abundance of Dubosiella and downregulated that of Cyanobium_PCC-6307 and DNF00809 at the genus level compared with the model group. The results of PCA and microbial distance calculation further exhibited that THSWD treatment resulted in significant regulation of the microbial community. Furthermore, compared with the model group, THSWD increased the level of vitamin k2 (VK2) in serum. These findings indicate that THSWD could protect blood pressure and endothelial function by regulating gut microbiota and promoting microbial metabolite VK2. The antiplatelet effects of GBE50, aspirin and 1:1 combination of GBE50 and aspirin were detected by microplate method using rabbit platelets. Synergy finder 2.0 was used to analyze the synergistic antiplatelet effect. The compounds in GBE50 were identified by UPLC-Q/TOF-MS analysis and the candidate compounds were screened by TCMSP database. The targets of candidate compounds and aspirin were obtained in TCMSP, CCGs, Swiss target prediction database and drugbank. Targets involving platelet aggregation were obtained from GenCLiP database. Compound-target network was constructed and GO and KEGG enrichment analyses were performed to identify the critical biological processes and signaling pathways. The levels of thromboxane B2 (TXB2), cyclic adenosine monophosphate (cAMP) and PAF receptor (PAFR) were detected by ELISA to determine the effects of GBE50, aspirin and their combination on these pathways.Results: GBE50 combined with aspirin inhibited platelet aggregation more effectively. The combination displayed synergistic antiplatelet effects in AA-induced platelet aggregation, and additive antiplatelet effects occurred in PAF, ADP and collagen induced platelet aggregation. Seven compounds were identified as candidate compounds in GBE50. Enrichment analyses revealed that GBE50 could interfere with platelet aggregation via cAMP pathway, AA metabolism and calcium signaling pathway, and aspirin could regulate platelet aggregation through AA metabolism and platelet activation. ELISA experiments showed that GBE50 combined with aspirin could increase cAMP levels in resting platelets, and decreased the levels of TXB2 and PAFR.Conclusion: Our study indicated that GBE50 combined with aspirin could enhance the antiplatelet effects. They exerted both synergistic and additive effects in restraining platelet aggregation. Heart failure (HF) is the end stage of several cardiovascular diseases with high mortality worldwide; however, current chemical drugs have not beneficial effect on reducing its mortality rate. Due to its properties of multiple targets components with multiple targets, natural products derived from traditional Chinese medicine (TCM) have exerts unique effects on the amelioration of the clinical symptoms of HF, yet, TCM is not widely used in the clinic since the potential therapeutic targets have not been fully investigated. Therefore, in this review, we briefly summarized the pathophysiological mechanism of HF and reviewed the published clinical evaluations of TCM and natural products from Chinese herbs to treat HF. Then, the therapeutic potential and the underlying mechanisms by which the natural products from Chinese herb exert their protective effects were further summarized. We concluded from this review that natural products from Chinese herbs have been shown to be more effective in treating HF by targeting multiple signaling pathways, including anticardiac hypertrophy, antifibrotic, anti-inflammatory, antioxidative and antiapoptotic activities. However, the major limitations of these compounds is that there are a lack of large scale, multicenter, randomized and controlled clinical trials for their use in treatment of HF, and the toxic effects of natural products from Chinese herbs also needed further investigation. Despite these limitations, further clinical trials and experimental studies will provide a better understanding of the mechanism of natural products from Chinese herbs and promote their wide use to treat HF. Sarcopenia has become a key challenge for healthy aging in older adults. However, it remains unclear whether traditional Chinese medicine can effectively treat sarcopenia. This systematic review analyzes the current evidence for the effect of traditional Chinese medicine (TCM) on sarcopenia. We searched for articles regarding sarcopenia treated by TCM in Cochrane library, PubMed, SinoMed, Web of Science, Embase, and the China National Knowledge Infrastructure (from inception until 10 December 2021). Two researchers independently screened the literature in accordance with the inclusion and exclusion criteria designed by PICOS principles. The risk of bias was assessed by the Cochrane Risk of Bias (ROB) tool. The quality of evidence was assessed by the grading of recommendations, assessment, development, and evaluation (GRADE). Participants' characteristics, interventions, and the relevant results of the included studies were extracted and synthesized in a narrative way. The total number of participants in the 21 included studies was 1,330. Most of the studies evaluated physical function (n = 20) and muscle strength (n = 18), and a small number of studies (n = 6) assessed muscle mass. Overall, it was found that TCM had a positive impact on muscle strength (grip strength, chair stand test) and physical function (6-m walking speed, timed up and go test, sit and reach) in patients with sarcopenia, inconsistent evidence of effects on muscle mass. However, the small sample size of the included studies led to imprecision in the results, and the presence of blinding of the studies, allocation concealment, and unreasonable problems with the control group design made the results low grade. Among these results, the quality of evidence for grip strength (n = 10) was of medium grade, and the quality of evidence related to the remaining indicators was of low grade. This systematic review showed that traditional Chinese Qigong exercises and Chinese herbal medicine have a positive and important effect on physical performance and muscle strength in older adults with sarcopenia. Future high-quality multicenter randomized controlled trials (RCTs) with large samples are needed to determinate whether acupuncture and other therapies are effective in treating sarcopenia. Parkinson's disease (PD) is characterized by progressive degeneration of dopaminergic neurons, leading to misbalance and loss of coordination. Current therapies are claimed only for symptomatic relief, on long-term use, which causes alteration in basal ganglia, and give rise to various adverse effects like dyskinesia and extra pyramidal side effects, which is reversed and proved to be attenuated with the help of various herbal approaches. Therefore, in order to attenuate the dopaminergic complications, focus of current research has been shifted from dopaminergic to non-dopaminergic strategies. Herbs and herbal remedies seems to be a better option to overcome the complications associated with current dopaminergic therapies. In recent years, various herbs and herbal remedies based on Ayurveda, traditional Chinese and Korean remedies, have become the target of various researches. These herbs and their bioactive compound are being extensively used to treat PD in India, China, Japan, and Korea. Abstract Background Dyslipidemia is a major risk factor for atherosclerotic cardiovascular disease and a leading cause of death worldwide. The clinical utility of commonly used lipid-lowering drugs such as statins and fibrates is sometimes limited by the occurrence of various adverse reactions. Recently, berberine (BBR) has received increasing attention as a safer and more cost-effective option to manage dyslipidemia. Thus, a high-quality randomized controlled trial to evaluate the efficacy and safety of BBR in the treatment of dyslipidemia is deemed necessary. Methods/design This is a randomized, double-blind, and placebo-controlled clinical trial. A total of 118 patients with dyslipidemia will be enrolled in this study and randomized into two groups at a ratio of 1:1. BBR or placebo will be taken orally for 12 weeks. The primary outcome is the percentage of low-density lipoprotein cholesterol reduction at week 12. Other outcome measures include changes in other lipid profiles, high sensitivity C-reactive protein, blood pressure, body weight, Bristol Stool Chart, traditional Chinese medicine symptom form, adipokine profiles, and metagenomics of intestinal microbiota. Safety assessment includes general physical examination, blood and urine routine test, liver and kidney function test, and adverse events. Discussion This trial may provide high-quality evidence on the efficacy and safety of BBR for dyslipidemia. Importantly, the findings of this trial will help to identify patient and disease characteristics that may predict favorable outcomes of treatment with BBR and optimize its indication for clinical use. Glaucoma is an eye disease characterized by progressiveretinal nerve damage and impaired vision, which is the top one irreversible blinding eye disease. The pathologic intraocular pressure elevation is its key risk. At present, the clinical medicine with intraocular pressure reducing and retinal nerve protection effects focused on symptomatic therapy with unsatisfied effects. Chinese herb monomers have advantages of both Chinese herbs and chemical drugs. Chinese herbs and Chinese herb monomers have favorable effects on glaucoma therapy, especially on retinal nerve protection, which provides a vast room for new drug development. The paper summarized applications and mechanism of representative anti-glaucoma Chinese herbal formulas, Chinese herbs and especially Chinese herb monomers, which would provide references for clinical therapy and new drug development for glaucoma.

Abstract Background Shanzhuyu (the dried mature sarcocarp of Cornus officinalis Sieb. et Zucc., DMSCO) is a Chinese herb that can be used for the treatment of Alzheimer’s disease (AD), but its mechanism remains unknown. The present study aimed to investigate the active ingredients and effective mechanisms of DMSCO for the treatment of AD based on a network pharmacology approach. Methods The active components of DMSCO were collected from the TCMSP and ETCM databases and the target proteins of these compounds were predicted using TCMSP, SwissTargetPrediction and the STITCH database. The AD-related target proteins were identified from the OMIM, DisGeNet, GEO and GeneCards databases. The network interaction model of the compound-target-disease was established and was used to obtain the key targets of DMSCO on AD through network topology analysis. Subsequently, gene enrichment in Gene Ontology (GO) and KEGG pathways were conducted using the David 6.8 online tool. Results A total of 30 DMSCO effective compounds and 209 effective drug targets were obtained. A total of 172 AD-related genes and 37 shared targets of DMSCO and AD were identified. A total of 43 key targets for the treatment of AD were obtained from the topological analysis of the DMSCO-AD target network. These key targets were involved in a variety of biological processes, including amyloid deposition, apoptosis, autophagy, inflammatory response and oxidative stress and pathways, such as the PI3K-AKT, MAPK and TNF pathways. Three key compounds, namely ursolic acid, anethole and β-sitosterol were obtained from the analysis of the key targets. Conclusions Ursolic acid, anethole and β-sitosterol may be the main active components of DMSCO in the treatment of AD. DMSCO can treat AD by regulating amyloid deposition, apoptosis, autophagy, inflammatory response and oxidative stress via the PI3K-AKT, MAPK and other signaling pathways.

Abstract Background Oxidative stress mediates the nerve injury during the pathogenesis of Alzheimer’s disease (AD). Protecting against oxidative stress damage is an important strategy to prevent and treat AD. Di-Huang-Yi-Zhi (DHYZ) is a Chinese medicine used for the treatment of AD, but its mechanism remains unknown. This study is aimed to investigate the effect of DHYZ on H2O2 induced oxidative damage in PC12 cells. Methods PC12 cells were treated with H2O2 and DHYZ. Cell proliferation was detected by Cell counting kit-8 (CCK-8) assay. Cytotoxicity of H2O2 was measured by lactate dehydrogenase (LDH) release assay. Apoptosis were identified by Annexin V-FITC/PI staining. Caspase 3 activity was detected by commercial kit. Mitochondrial membrane potential (MMP) was detected by JC-1 staining. Reactive oxygen species (ROS) was 2′, 7′-Dichlorodihydrofluorescein diacetate (DCFH-DA) staining. Protein expression and phosphorylation was identified by western blot. Results The results showed that DHYZ antagonized H2O2-mediated cytotoxicity and proliferation inhibition. DHYZ reduced ROS production, stabilize mitochondrial membrane potential, inhibit Caspase-3 activity and apoptosis induced by H2O2. In addition, DHYZ inhibited the phosphorylation of ASK1, JNK1/2/3 and p38 MAPK which were up-regulated by H2O2. Conclusions The present study suggested that DHYZ protected PC12 cells from H2O2-induced oxidative stress damage and was related to inhibition of ROS production and ASK1-JNK/p38 MAPK signaling. The present study provides experimental evidence for the application of DHYZ for the management of oxidative stress damage and AD.

Various techniques such as data mining, network pharmacology, molecular docking and molecular dynamics simulation were used in this study to screen and validate effective herbal medicines for the treatment of idiopathic pulmonary fibrosis (IPF) and to reveal their mechanisms of action at the molecular level. The use of this approach will provide new tools and ideas for future drug screening, especially for the application of herbal medicines in the treatment of complex diseases. Among them, the five identified core targets, including IL6, TP53, AKT1, VEGFA, and TNF, as well as a series of major active compounds, will be important references for future anti-IPF drug development. This information will accelerate the discovery and development of relevant drugs. Meanwhile, this study further confirmed the potential value of four Chinese herbal medicines, including Gancao, Danshen, Huangqin, and Sanqi, in the treatment of IPF. This will promote more clinical trials and practices to confirm and optimise the application of these herbs. Finally, this study is an important theoretical guide to enhance the advantages of Chinese herbal medicines in the prevention and treatment of major and difficult diseases, as well as to understand and utilise the potential efficacy of Chinese herbal medicines. Chinese herb hydrogel has outstanding values in wound treatment. Tremendous effort is dedicated to developing functional and intelligent Chinese herb hydrogels for accelerating healing process. Herein, a novel multifunctional bilayer Chinese herb hydrogel patch for wound healing is presented. The patches are composed of a Ginsenoside compound K (GCK)-integrated chitosan inverse opal scaffold layer and a Bletilla striata polysaccharide (BP) hydrogel filler layer. As the photothermal conversion capacity of BP can trigger faster degradation of its hydrogel layer under near-infrared (NIR) light, the encapsulated BP can be controllably released to wound area and exhibit photothermal antibacterial and antioxidant effects. Besides, the inverse opal scaffold layer shows great mechanical property and moisturizing performance, and its released GCK can promote angiogenesis for long term therapeutic effects. Attractively, due to responsive structural color, the patch is endowed with self-reporting performance, which can real-time visually monitor the drug release degree. Taking advantages of these properties, the patches have been demonstrated to achieve satisfying therapeutic effects for in vivo wound treatment. ABSTRACT: This experiment examined the separate and combined effects of daidzein (Da) and Chinese herbs (CH) on laying performance and egg quality of post-peak laying hens. Additionally, we explored potential mechanisms of action for these 2 additives by examining plasma hormone levels. After 4 wk of acclimation to caging, 60-week-old Hyline Brown hens (360) were selected and randomly divided into 4 groups with 6 replicates and 15 chickens per replicate. The following 4 dietary groups were utilized: 1) control group (basal diet); 2) Da group (basal diet + 0.03 kg/t DA); 3) CH group (basal diet +0.6 kg/t CH); 4) Da + CH group (basal diet + 0.03 kg/t Da + 0.6 kg/t CH). Data were analyzed in a completely randomized design with a 2×2 factorial arrangement of treatments. Egg production and FCR treatment averages were analyzed in the following 3 phases: wk 1-4, 5-8, and 1-8 of treatment administration. Results revealed that Da increased egg production but decreased FCR (P < 0.05) for wk 1-8 and especially during wk 5-8 (P < 0.05). CH decreased FCR in wk 1-4 and 5-8 (P < 0.05), but increased egg production only during wk 5-8 (P < 0.05). Da increased Haugh units (P < 0.05) on wk 4 and 8; CH increased Haugh units (P < 0.05) but decreased yolk ratio (P < 0.05) on wk 4 and 8. Da increased the plasma levels of T3, PROG, FSH, LH and E2 (P < 0.05); CH increased the plasma level of T3 (P < 0.05). Additionally, Da x CH interactions existed for albumen height, Haugh units, albumen ratio and the level of T3 on wk 8 (P < 0.05), indicating that the combination of Da and CH was more effective than administration of either of these dietary components independently. In conclusion, Da and CH, both independently and in combination, increase laying performance, egg quality and plasma hormones levels in post-peak laying hens. Therefore, these treatments may be able to provide prolonged economic benefits to aged laying hens.

BACKGROUND: Previous studies have shown that Tougu Xiaotong Capsules can reduce the expression of chondrocyte inflammatory factors, reduce chondrocyte apoptosis, promote chondrocyte proliferation, and delay cartilage and subchondral bone degeneration, thereby alleviating the process of osteoarthritis and reducing inflammation responses. However, the mechanism of this prescription on synovial lesions is unclear. OBJECTIVE: To further explore the mechanism of anti-inflammatory and anti-swelling effect of Tougu Xiaotong Capsule on early osteoarthritis with synovial edema based on the regulation of aquaporins. METHODS: Twenty-four Sprague-Dawley rats were randomly divided into three groups: blank group, model group and traditional Chinese medicine (TCM) group. Osteoarthritis animal models were made by papain injection. After 1 week, animals were gavaged respectively with Tougu Xiaotong Capsule in the TCM group or saline in the blank and model groups for 4 weeks. The morphology and structure of the knee joints were observed by MRI. Interleukin-1β and tumor necrosis factor α in serum and joint fluid were detected by radioimmunoassay. Synovial tissue microstructure was observed through paraffin section, and the ultrastructure of synovial cells observed by transmission electron microscope. Aquaporins 1 and 3 were quantified and located by immunohistochemical staining/fluorescence staining. The study protocol was approved by the Laboratory Animal Ethics Committee of Fujian University of Traditional Chinese Medicine (approval No. [2019]024). RESULTS AND CONCLUSION: Compared with the model group, synovial lesion was alleviated and synovial secretion was decreased in the TCM group; the contents of interleukin-1β and tumor necrosis factor α in the serum and joint fluid were decreased significantly in the TCM group; synovial cell hyperplasia was alleviated, tissue fibrosis and vascular hyperplasia was significantly reduced in the TCM group; the number of rough endoplasmic reticula, Golgi apparatus, vesicles and lysosomes was decreased in synovial cells in the TCM group; and the expressions of aquaporin 1 and aquaporin 3 were significantly downregulated, and they were scattered in the cell membrane and cytoplasm, without aggregation. To conclude, by decreasing aquaporin 1 and aquaporin 3 expression in synovial tissue, Tougu Xiaotong Capsule can reduce synovial secretion and alleviate joint edema, which may be one of the anti-inflammation and anti-swelling mechanisms of the prescription. BACKGROUND: MicroRNA is involved in intervertebral disc degeneration, and Yaotu Granule can significantly delay the progress of intervertebral disc degeneration. OBJECTIVE: To investigate the effect of Yaotu Granule in the proliferation and apoptosis of nucleus pulposus cells by regulating the expression of microRNA-221 (miR-221). METHODS: MiR-221 mimics and inhibitors were transfected into rat nucleus pulposus cells to establish miR-221 overexpression and inhibition models. Tumor necrosis factor-α was used to induce nucleus pulposus cell degeneration model. The degenerated nucleus pulposus cells were treated with 100 and 200 mg/L Yaotu Granule freeze-dried powder (low and high dose groups). In the miR-221 mimics + Yaotu Granule group, miR-221 mimic and 200 mg/L Yaotu Granule freeze-dried powder were added. Cell counting kit-8 and TUNEL assay were used to detect the cell proliferation and apoptosis, RT-qPCR was used to detect mRNA expression of miR-221, and western blot was used to detect the expression of Bax and Bcl-2. RESULTS AND CONCLUSION: MiR-221 mimics reduced cell prolifecation at 24, 48, 72 hours (P < 0.05) and promoted cell apoptosis at 48 hours (P < 0.05); miR-221 inhibitors had the opposite results (P < 0.05). Compared with the normal group, tumor necrosis factor-α inhibited cell proliferation, increased cell apoptosis, and upregulated miR-221 expression (all P < 0.05). Compared with the model group, low- and high-dose Yaotu Granules increased cell proliferation, reduced cell apoptosis, and downregulated miR-221 expression (all P < 0.05). Compared with the mimics group, cell proliferation improved, apoptosis rate decreased, the protein expression of Bax reduced, and the Bcl-2 protein expression up-regulated in the miR-221 mimics + Yaotu Granule group (all P < 0.05). To conclude, miR-221 has an effect on the proliferation and apoptosis of nucleus pulposus cells, and Yaotu Granule can delay the progression of intervertebral disc degeneration by downregulating the expression of miR-221. Background: Myocardial ischaemia/reperfusion (I/R) results in myocardial injury via excessive autophagy. Huoxue Jiedu Formula (HJF) has been widely applied in China for the treatment of ischaemic heart disease. However, the mechanisms of HJF are still poorly understood. Thus, the present experiment was designed to observe the effects of HJF on myocardial I/R injury and explore the possible mechanism.Methods: Myocardial injury in rats subjected to myocardial I/R was reflected by nitrotetrazolium blue chloride staining, thioflavin S staining, serum creatine kinase-MB (CK-MB) and cardiac troponin T (cTnT). Autophagy was determined by electron microscopy, laser confocal microscopy, Q-PCR and western blot. The possible pathway was predicted by network pharmacology and validated in vivo and in vitro.Results: Pretreatment of HJF decreased the no-reflow area, infarcted area, serum CK-MB levels and serum cTnT levels in I/R rat model. In addition, pretreatment of HJF decreased autophagy in heart tissues (decrease in Beclin-1 and LC3-II, and increase in Bcl-2, p62 and ratio of LC3-I/LC3-II). In the vivo study, pretreatment of HJF significantly decreased hypoxia/reoxygenation (H/R)-induced autophagy in H9C2 cells. Network pharmacology was applied to predict the possible mechanism by which HJF affects cardiac autophagy, and the PI3K/AKT/mTOR signalling pathway was the most significantly enriched pathway. And experimental studies demonstrated that pretreatment of HJF increased the phosphorylation of AKT and mTOR, and the effects of HJF on autophagy would be offset by PI3K inhibitor LY294002.Conclusion: Pretreatment of HJF ameliorates myocardial I/R injury by decreasing autophagy through activating PI3K/AKT/mTOR pathway.

BACKGROUND: Studies have shown that serum homocysteine concentration is an independent predictor of the prevalence and severity of coronary artery disease for patients with normal hypersensitivity C-reactive protein levels. OBJECTIVE: To investigate the protective effect of Wenban Decoction on the apoptosis of rat cardiac microvascular endothelial cells (CMECs) induced by homocysteine by regulating PI3K/Akt signaling pathway. METHODS: Rat CMECs were primarily cultured in vitro, and the cells were randomly divided into control group, model group and Wenban Decoction group (50 mg/L Wenban Decoction). The cells in the latter two groups were injured by 10 mmol/L homocysteine prior to the treatment. Cell counting kit-8 was used to detect the cell viability of each group. ELISA was used to determine serum lactate dehydrogenase, malondialdehyde, whole blood catalase, superoxide dismutase, glutathione peroxidase, interleukin 6, intercellular adhesion molecule 1, interleukin 1β, and tumor necrosis factor α. Flow cytometry was used to detect cell apoptosis after addition of LY294002 based on the treatment with Wenban Decoction. Western blot was used to detect the expression of PI3K, Akt, p-Akt, Bax, Bcl-2 and caspase3 protein in the cells. An ethic approval was given by the Animal Experiment Ethics Committee of Liaoning University of Traditional Chinese Medicine (approval No. 21000092018010). RESULTS AND CONCLUSION: Compared with the control group, the survival ability of CMECs in the model group was significantly reduced, the leakage of lactate dehydrogenase was significantly increased, which caused oxidative stress and the release of inflammatory factors, and finally led to a large number of apoptosis. Compared with the model group, Wenban Decoction improved the survival ability of CMECs, reduced the leakage of lactate dehydrogenase, significantly decreased the intracellular levels of interleukin 1β, intercellular adhesion molecule 1, interleukin 6 and tumor necrosis factor α (P < 0.05), as well as reduced the number of apoptotic cells. PI3K inhibitor reversed the inhibitory effect of Wenban Decoction on homocysteine-induced apoptosis of CMECs. To conclude, Wenban Decoction can significantly improve the survival ability of CMECs, reduce the leakage of lactate dehydrogenase, inhibit the level of oxidative stress and the release of inflammatory factors, and ultimately reduce the number of apoptotic cells, which is related to the inhibition of PI3K/Akt signaling pathway. BACKGROUND: Osteoporotic fractures are the most common serious complications caused by osteoporosis, and their repair is more difficult, which seriously threatens the health and quality of life of patients. OBJECTIVE: To investigate the interventional effect of Panax Notoginsenosides in ovariectomized osteoporotic fracture rats and the relevant mechanism. METHODS: Fifty healthy female rats were selected. Ten of them were normal group, and the other 40 rats were used to make ovariectomized osteoporotic fracture models. Model rats were divided into model group, low, medium and high dose group. Normal group and model group rats were intragastrically administered normal saline, rats in the low, medium and high dose group were given intragastric administration of 10, 20 and 40 mg/kg Panax Notoginsenosides solution. The study protocol was approved by the Animal Ethic Committee of the First Affiliated Hospital of Guizhou University of Traditional Chinese Medicine, No. (2019)58 on September 23, 2019. RESULTS AND CONCLUSION: The bone mineral density and bone cell index of the high dose group were higher than those of low and middle dose groups (P < 0.05). The bone volume fraction, the number of callus trabeculae and the thickness of trabeculae in the high dose group were all higher than those in the model group, the low dose group and the middle dose group, and the resolution of trabeculae was lower than that in these three groups (P < 0.05). The levels of glutathione peroxidase, bone morphogenetic protein 2, and vascular endothelial growth factor in the high dose group were lower than those in the normal group, but higher than those in the model group, low and medium dose groups. The levels of lipid peroxide, malondialdehyde, osteocalcin, type I procollagen carboxy terminal propeptide, bone-specific alkaline phosphatase in the high dose group were higher than those in the normal group, but lower than those in the model group, low and medium dose group (P < 0.05). The expression of PI3K, Akt and mTOR in the low, middle and high dose groups was lower than that in the normal group and higher than that in the model group; and the expression of PI3K, Akt and mTOR in the high dose group was higher than that in the low and middle dose groups (P < 0.05). Therefore, treatment with Panax Notoginsenosides can increase the bone miner density and bone cell index, promote the growth of bone trabecula at the callus, alleviate oxidative stress injury, regulate the PI3K / Akt / mTOR signal pathway, and accelerate the formation of new blood vessels in the callus and fracture healing in ovariectomized osteoporotic fracture rats. Abstract Background Based on clinical medications and related studies, we established a Yang-Gan Jie-Du Sang-Jie (YGJDSJ) herbal formula for hepatocarcinoma treatment. In present study, we evaluated the anti-cancer potential of YGJDSJ on suspension-grown human hepatocellular carcinoma Bel-7402 cells. Methods Bel-7402 cells were cultured in poly(2-hydroxyethyl methacrylate) (poly-HEMA) coated plates and treated with YGJDSJ. Anchorage-independent cell growth was detected by cell Counting Kit-8 (CCK-8) assay and soft agar colony formation assay. Anoikis was detected by ethdium homodimer-1 (EthD-1) staining and flow cytometry analysis. Caspases activities were detected by the cleavage of chromogenic substrate. Reactive oxygen species (ROS) was detected by 2′,7′-dichlorofluorescin diacetate (DCFH-DA) staining. Protein expression and phosphorylation was identified by western blot. Protein expression was knocked-down by siRNA. Results YGJDSJ inhibited the proliferation of Bel-7402 cells in poly-HEMA coated plates and anchorage-independent growth of Bel-7402 cells in soft agar. YGJDSJ also induced anoikis in Bel-7402 cells as indicated by EthD-1 staining and flow cytometry analysis. YGJDSJ activated caspase-3, − 8, and − 9 in suspension-grown Bel-7402 cells. The pan-caspase inhibitor Z-VAD-FMK significantly abrogated the effects of YGJDSJ on anoikis in suspension-grown Bel-7402 cells. In addition, YGJDSJ increased ROS in suspension-grown Bel-7402 cells. The ROS scavenger N-acetyl-L-cysteine (NAC) partially attenuated YGJDSJ-induced activation of caspase-3, − 8 and − 9 and anoikis in suspension-grown Bel-7402 cells. Furthermore, YGJDSJ inhibited expression and phosphorylation of protein tyrosine kinase 2 (PTK2) in suspension-grown Bel-7402 cells. Over-expression of PTK2 significantly abrogated YGJDSJ induced anoikis. Conclusions YGJDSJ inhibits anchorage-independent growth and induce caspase-mediated anoikis in Bel-7402 cells, and may relate to ROS generation and PTK2 downregulation.

Background: Oxidative stress mediates the nerve injury during the pathogenesis of Alzheimer’s disease (AD). Protecting against oxidative stress damage is an important strategy to prevent and treat AD. Di-Huang-Yi-Zhi (DHYZ) is a Chinese medicine used for the treatment of AD, but its mechanism remains unknown. This study is aimed to investigate the effect of DHYZ on H2O2 induced oxidative damage in PC12 cells. Methods: PC12 cells were treated with H2O2 and DHYZ. Cell proliferation was detected by Cell counting kit-8 (CCK-8) assay. Cytotoxicity of H2O2 was measured by lactate dehydrogenase (LDH) release assay. Apoptosis were identified by Annexin V-FITC/PI staining. Caspase 3 activity was detected by commercial kit. Mitochondrial membrane potential (MMP) was detected by JC-1 staining. Reactive oxygen species (ROS) was 2′, 7′-Dichlorodihydrofluorescein diacetate (DCFH-DA) staining. Protein expression and phosphorylation was identified by western blot. Results: The results showed that DHYZ antagonized H2O2-mediated cytotoxicity and proliferation inhibition. DHYZ reduced ROS production, stabilize mitochondrial membrane potential, inhibit Caspase-3 activity and apoptosis induced by H2O2. In addition, DHYZ inhibited the phosphorylation of ASK1, JNK1/2/3 and p38 MAPK which were up-regulated by H2O2. Conclusions: The present study suggested that DHYZ protected PC12 cells from H2O2-induced oxidative stress damage and was related to inhibition of ROS production and ASK1-JNK/p38 MAPK signaling. Eucommia leaf extract (ELE) is a traditional Chinese herbal medicine. We investigated the effect of ELE on the development of atherosclerosis and changes in peritoneal macrophage function in apolipoprotein E knockout (ApoE–/–) mice. At 8 weeks of age, ApoE–/– mice were randomly divided into three groups that were fed a high‐fat diet blended with 0% (control), 5% or 10% ELE for a period of 7 weeks. The 10% ELE dose caused an approximately 36% reduction in atherosclerotic lesions, as estimated by oil red O staining. Real‐time PCR analysis showed that the 1‐week treatment with ELE reduced mRNA levels of Tnf‐alpha, Il‐1, and Mif in peritoneal macrophages isolated from the ApoE–/– mice. Furthermore, a 1‐week treatment with the 10% ELE diet significantly reduced migration and adhesion functions in peritoneal macrophages. These results suggest that a 10% ELE diet reduces atherosclerotic lesions and modulates macrophage function by reducing cytokine expression. The study aimed to compare the pharmacokinetic properties of quercitrin, astragalin, afzelin and taxifolin, four major bioactive components of Polygonum orientale inflorescence extracts, between sham-operated and myocardial ischemia–reperfusion injury (MIRI) rats. Rats were divided into two groups: MIRI model and sham-operated. The blood samples were collected according to the time schedule. The levels of quercitrin, astragalin, afzelin and taxifolin in the plasma at designated time points were determined using an HPLC–MS/MS method. Various pharmacokinetic parameters were estimated from the plasma concentration versus time data using non-compartmental methods. After the administration of the Chinese herb Polygonum orientale inflorescence extracts, the Cmax, AUC, as well as MRT, increased, while CL decreased, in MIRI model compared to the sham-operated animals. These results suggest that the pathological damage of ischemia–reperfusion had a significant impact on the pharmacological effects of Polygonum orientale inflorescence extracts on ischemic heart disease. The method had been successfully applied to evaluate the pharmacokinetics of quercitrin, astragalin, afzelin and taxifolin in rat plasma after the oral administration of Chinese herb Polygonum orientale inflorescence extracts in rats. Osthole is an active coumarin extracted from Cnidium monnieri (L.) Cusson, a traditional Chinese herb which possesses multiple pharmacological activities. Asthma is a long term disease characterized by chronic airway inflammation and hyperresponsiveness. Worldwide, around 339 million people are reported to be suffering from asthma. Coumarin osthole has been known to exhibitanti-inflammatory and antiallergic activities. Here, we investigated mechanisms underlying the protective effects of osthole in ovalbumin (OVA) induced asthmatic mice. A total 40 C57BL/6 male mice were used as experimental model animals. The results showed that osthole treatment significantly reduces OVA-induced elevated levels of serum IgE and inflammatory cytokines (IL-4, IL-5, IL-6, IL-13) except IL-10 in bronchoalveolar lavage fluid (BALF), and decreased the recruitment of inflammatory cells in BALF. The CCK-8 assay was performed to examine the viability of cells. The enzyme linked immunosorbent assay (ELISA) was used to determine the expression of inflammatory factors. Western blot analysis was conducted to examine TLR9 and JNK protein expression. Results showed that osthole significantly inhibits the recruitment of inflammatory cells and Th1/Th2 cytokines expression in BALF and airway hyper-responsiveness. Furthermore, osthole attenuated upregulation of systemic immunoglobulin production derived from OVA sensitization. Results also suggest that osthole can reverse the imbalance of Thlfrh2 in asthma, thus alleviating the symptoms of asthma to a certain extent. In conclusion, the study demonstrated that osthole, activecoumarin extract of Cnidium monnieri, attenuated the allergic airway inflammation via inhibition of TLR9/JNK pathway in mice model indicating that it could be a potential therapeutic target for asthma. Dengue fever is an important epidemic disease with a high prevalence in tropical and subtropical countries. We aimed to investigate the effects of a treatment integrating traditional Chinese (TCM) and Western medicines on dengue inpatients with warning signs (i.e., group B) according to the World Health Organization dengue classification in this retrospective cohort study of medical records. Inpatients who were treated with conventional Western therapies in the absence or presence of TCM were assigned to the control and treatment groups, respectively. Data were compared using an analysis of variance, general linear analysis, and chi-square test. The most common clinical symptoms and signs of dengue fever were fever and muscle ache. The treatment group patients were significantly more likely to present general weakness and poor appetite than the control group patients. Patients in the treatment group were more likely to experience stomachache than those in the control group. Moreover, comparisons of the changes in hemoglobin and alanine aminotransferase levels over time revealed significant differences between the patient groups. Zhu Ye Shi Gao Tang, Gui Pi Tang, Paeonia suffruticosa, and Clerodendrum cyrtophyllum were the most commonly administered TCM formula and single herbs in this study. Patients in the treatment group experienced a resolution of symptoms, signs, and laboratory data and were discharged smoothly, without deterioration to death or critical care. The clinical efficacy of the hospital preparation compound granules of Hedyotis diffusa (CGHD), which is composed of Hedyotis diffusa Willd, Smilax china L., Solanum lyratum Thunb., has accumulated a good reputation over the past decades. However, because it is a hospital preparation, few researchers have paid attention to it, resulting in a lack of systematic basic research studies. Thus, it is not clear whether there are safety concerns that restrict its clinical application, and toxicological evaluation of CGHD is needed. The aim of this study was to evaluate the safety of CGHD by conducting acute toxicity and long-term toxicity experiments, with the objective of providing evidence for its clinical safety and a theoretical foundation for its clinical application. KM mice were selected for the acute toxicity experiment and were administered water or CGHD-E 3 times within 24 h. The reactions of the animals to CGHD treatment were observed and recorded within 1 h after administration and then once a day for 14 consecutive days. SD rats were selected to conduct the long-term toxicity experiment. The drug-treated groups were administered different doses of CGHD-E, which were equivalent to 10 times, 20 times and 50 times the clinical dose in humans. The rats were administered the drug for 28 consecutive days. After 28 days, the animals were sacrificed, and routine blood tests, blood coagulation function analysis, liver and kidney function tests, and glycolipid metabolism related tests were conducted. The major organs of the rats were collected to calculate organ coefficients and perform hematoxylin-eosin (HE) staining. In the CGHD-E acute toxicity experiment, the drug-treated groups did not show adverse reactions or poisoning symptoms, and the maximum tolerated dose of CGHD-E in mice was greater than 45.072 g/kg. In the long-term toxicity experiment, drug-treated rats generally exhibited a good condition, but continuous administration decreased on body weight and food intake, especially in male rats. Coagulation function alterations and the impact on the liver during long-term drug administration were also assessed, which should be emphasized in clinical applications. No significant toxic effects were observed according to routine blood tests or test of liver and kidney function, glucose and lipid metabolism, or ion metabolism. The results of this study showed that CGHD was nontoxic or had low toxicity, providing not only a scientific basis for its clinical application, determining the appropriate clinical dose and monitoring clinical toxicity but also theoretical support for subsequent clinical drug trials.
