2. Single cell profiling of circulating autoreactive CD4 T cells from patients with autoimmune liver diseases suggests tissue imprinting.

- Abstract
Autoimmune liver diseases (AILD) involve dysregulated CD4 T cell responses against liver self-antigens, but how these autoreactive T cells relate to liver tissue pathology remains unclear. Here we perform single-cell transcriptomic and T cell receptor analyses of circulating, self-antigen-specific CD4 T cells from patients with AILD and identify a subset of liver-autoreactive CD4 T cells with a distinct B-helper transcriptional profile characterized by PD-1, TIGIT and HLA-DR expression. These cells share clonal relationships with expanded intrahepatic T cells and exhibit transcriptional signatures overlapping with tissue-resident T cells in chronically inflamed environments. Using a mouse model, we demonstrate that, following antigen recognition in the liver, CD4 T cells acquire an exhausted phenotype, play a crucial role in liver damage, and are controlled by immune checkpoint pathways. Our findings thus suggest that circulating autoreactive CD4 T cells in AILD are imprinted by chronic antigen exposure to promote liver inflammation, thereby serving as a potential target for developing biomarkers and therapies for AILD. Dysregulated CD4 T cells have been implicated in autoimmune liver disease, but their phenotypes and origin are still unclear. Here the authors profile circulating, autoreactive CD4 T cells to find transcription signatures similar to tissue-activated, exhausted T cells, thereby hinting a tissue origin for these circulation CD4 T cells. Background Patients with chronic hepatitis C virus (HCV) infection are at greater risk of developing metabolic disorders. Obesity is a major risk factor for these disorders, and therefore, managing body weight is crucial. Cannabis use, which is common in these patients, has been associated with lower corpulence in various populations. However, this relationship has not yet been studied in persons with chronic HCV infection. Methods Using baseline data from the French ANRS CO22 Hepather cohort, we used binary logistic and multinomial logistic regression models to test for an inverse relationship between cannabis use (former/current) and (i) central obesity (i.e., large waist circumference) and (ii) overweight and obesity (i.e., elevated body mass index (BMI)) in patients from the cohort who had chronic HCV infection. We also tested for relationships between cannabis use and both waist circumference and BMI as continuous variables, using linear regression models. Results Among the 6348 participants in the study population, 55% had central obesity, 13.7% had obesity according to their BMI, and 12.4% were current cannabis users. After multivariable adjustment, current cannabis use was associated with lower risk of central obesity (adjusted odds ratio, aOR [95% confidence interval, CI]: 0.45 [0.37–0.55]), BMI-based obesity (adjusted relative risk ratio (aRRR) [95% CI]: 0.27 [0.19–0.39]), and overweight (aRRR [95% CI]: 0.47 [0.38–0.59]). This was also true for former use, but to a lesser extent. Former and current cannabis use were inversely associated with waist circumference and BMI. Conclusions We found that former and, to a greater extent, current cannabis use were consistently associated with smaller waist circumference, lower BMI, and lower risks of overweight, obesity, and central obesity in patients with chronic HCV infection. Longitudinal studies are needed to confirm these relationships and to assess the effect of cannabis use on corpulence and liver outcomes after HCV cure. International audience; Background: Chronic hepatitis B virus (HBV) infection results in a high risk of cirrhosis and its complications, cirrhosis decompensation (DC), hepatocellular carcinoma (HCC), liver transplantation (LT), death or any of these outcomes (composite endpoint [CE]). Nucleos(t)ide analogues (NUCs) such as tenofovir or entecavir are associated with a reduction in these complications.Aim: To compare the impact of tenofovir and entecavir on these outcomes in patients treated for HBV infection and included in the prospective Hepather cohort.Methods: All patients with HBV infection who had received tenofovir or entecavir for more than 6 months at or after entry in the ANRS CO22 cohort were selected. Patients with HDV and HCV co-infection or prior liver event were excluded. Incidence rates of events were compared using inverse probability of treatment weighting (IPW).Results: The cohort included 1800 patients (986 tenofovir and 814 entecavir). Median follow-up was 4.2 years. The incidences of HCC, DC, LT, ACD, LRD and CE were not different between tenofovir- (1.8 (0.9; 3.2), 0.6 (0.2; 1.6), 0.2 (0.0; 0.8), 1.7 (0.8; 3.0), 0.8 (0.2, 1.8) and 4.1 (3.0; 5.4) per 1000 person-years) and entecavir-treated patients (1.6 (0.7; 3.0), 0.7 (0.2; 1.8), 0.2 (0.0; 1.0), 3.0 (1.7, 4.8), 0.5 (0.1; 1.5) and 5.0 (3.3; 7.2)) per 1000 person-years, respectively.Conclusion: The risk of liver-related events or death was not different between tenofovir- and entecavir-treated patients in this large prospective cohort of predominantly non-cirrhotic French patients.Trial registration number: NCT019553458.

International audience; Background: Chronic hepatitis B virus (HBV) infection may evolve into cirrhosis and hepatocellular carcinoma, and this progression may be accelerated by specific risk factors, including overweight and obesity. Although evidence for a protective effect of cannabis use on elevated body weight has been found for other populations, no data are available for HBV-infected patients.Aims: We aimed to identify risk factors (including cannabis use) for overweight and obesity in patients with HBV chronic infection.Methods: Using baseline data from the French ANRS CO22 Hepather cohort, we performed two separate analyses, one using “central obesity” (based on waist circumference) and the other “overweight” and “obesity” (based on body mass index) as outcomes. Logistic and multinomial regressions were used to model central obesity and overweight/obesity, respectively.Results: Among the 3706 patients in the study population, 50.8% had central obesity, 34.7% overweight, and 14.4% obesity. After multivariable adjustment, current cannabis use was associated with a 59% lower risk of central obesity compared with no lifetime use (adjusted odds ratio [95% CI]: 0.41 [0.24 to 0.70]). It was also associated with a 54% and 84% lower risk of overweight (adjusted relative risk ratio [95% CI]: 0.46 [0.27 to 0.76]) and obesity (0.16 [0.04 to 0.67]), respectively.Conclusions: Cannabis use was associated with lower risks of overweight and obesity in patients with HBV chronic infection. Future studies should test whether these potential benefits of cannabis and cannabinoid use translate into reduced liver disease progression in this high-risk population.

Le Statut de Rome a été conclu le 17 juillet 1998 et il est entré en vigueur le 1er juillet 2002. Depuis, les chambres de la Cour pénale internationale ont déjà fait œuvre d’un intense travail judiciaire en parallèle d’une politique pénale développée par les procureurs Luis Moreno Ocampo et Fatou Bensouda. Internationale.; International audience; Le développement de nouvelles solutions de protection balistique nécessite des simulations numériques, afin de prédire leurs performances et leurs effets secondaires. Les mécanismes induits dans les tissus balistiques lors d'un impact dépendent des paramètres de tissage mais aussi des paramètres internes des fils utilisés dans ces structures. Ainsi, connaître le comportement dynamique du fil est essentiel pour déterminer le comportement balistique des tissus lors d'un impact. Ce travail vise à étudier le comportement mécanique d'un fil para-aramide en charge dynamique et longitudinale. À cette fin, nous avons conçu un nouveau dispositif expérimental pour réaliser ces essais à l'aide d'un canon à gaz: le Dispositif de Traction par Impact sur Fil (DTIF). Des campagnes d'essais ont été réalisées pour identifier les paramètres dynamiques d'un fil vierge, ainsi que de caractériser l'influence de l'arrangement structurel du tissu sur le comportement dynamique d'un fil.

- Abstract
Political rights of refugees are subject to several limitations in the international legal system. The present research ponders the conformity of such limitations with respect to the condition of legality, legitimacy and proportionality accepted in human rights. The intention is to reconcile political rights with refugee status from Hannah Arendt’s political thinking. The approach distinguishes reasonable from unreasonable limitations. On the one hand, the limitations to right of political nature (freedom of expression, association and assembly) are described as unreasonable due to the lack of conformity with respect to legality, legitimacy and proportionality conditions. This consequently reconcile refugee status with rights of a political nature. On the other hand, limitations to political rights in the strict sense (right to vote and eligibility) are considered reasonable due to their compliance with the conditions of citizenship and residence. This gives rise to an hindrance as for refugees to exercise their political rights in the strict sense, both in the host country and in the home country. To bypass this obstacle, this research suggests the cessation of refugee status., Les droits politiques des réfugiés sont soumis à plusieurs limitations dans l’ordre juridique international. La présente recherche s’interroge sur la conformité de telles limitations aux conditions de légalité, de légitimité et de proportionnalité admises en droits de l’homme. Elle part Le but est de concilier les droits politiques avec le statut de réfugié à partir de la pensée politique d’Hannah Arendt. La démarche distingue les limitations raisonnables de celles qui sont déraisonnables. D’un côté, les limitations aux droits de nature politique (libertés d’expression, d’association et de réunion) sont qualifiées de déraisonnables en raison de l’absence de conformité avec les conditions de légalité, de légitimité et de proportionnalité. Cela a pour effet de concilier le statut de réfugié avec les droits de nature politique. De l’autre, les limitations aux droits politiques au sens strict (droit de vote et d’éligibilité) sont jugées raisonnables en raison de leur conformité aux conditions de citoyenneté et de résidence. Il s’ensuit une entrave à l’exercice des droits politiques au sens strict des réfugiés aussi bien dans le pays d’accueil que dans celui d’origine. This work aims at studying the mechanical behavior of a para-aramid yarn in dynamic and longitudinal load. To that end, we designed a new experimental device to achieve these tests with the use of a gas gun: the Tensile Impact Test for Yarn (TITY). This device met numerous modifications in the aim to reach optimal test conditions. It is associated with a measurement device using laser and allowing obtaining the velocity profile of the projectile and the Strength-Strain characteristic curves of the tested yarn. The validity of the measure is confirmed by comparing our results with those obtained with a laser Doppler vélocimètre, device allowing measuring directly and precisely the velocity profile of the projectile. Many test campaigns are performed. Both in the aim to identify the dynamic parameters of a single and virgin yarn, and in the aim to characterize the impact of the fabric structural arrangement on the dynamic behavior of a yarn. This study reveals the intervention of three main structural parameters, which are the total inter-yarns contact surface, the number of binding points and the transvers and lateral inter-yarns pressures, having an impact on four main yarn break criteria, which are the structural defects and hysteresis phenomena inside the filament, stress concentrations and frictional forces concerning the interactions between the tested yarn and the rest of the fabric.; Ce travail a pour objectif d’étudier le comportement mécanique d’un fil de para-aramide en sollicitation longitudinale et dynamique. Pour ce faire, nous avons conçu un nouveau dispositif expérimental permettant de réaliser ces essais à l’aide d’un canon à gaz : le Dispositif de Traction par Impact sur Fil (DTIF). Ce dispositif a subi de nombreuses modifications dans le but d’obtenir des conditions d’essais optimales. Celui-ci est associé à un dispositif de mesure par laser permettant d’obtenir le profil de vitesse du projectile ainsi que les courbes caractéristiques de Force-Déformation du fil testé. La validité de la mesure est confirmée par comparaison aux résultats obtenus avec un vélocimètre à effet Doppler, dispositif permettant de mesurer directement et de façon précise le profil de vitesse du projectile. Plusieurs campagnes d’essais sont réalisées, à la fois dans le but d’identifier les caractéristiques dynamiques d’un fil seul, et dans le but de caractériser l’influence de l’arrangement structural d’un tissu sur le comportement dynamique d’un fil. Cette étude révèle l’intervention de trois principaux paramètres structuraux, qui sont la surface de contact inter-fils totale, le nombre de points de liage et les pressions inter-fils transverses et latérales au sein du tissu, influant sur quatre principaux critères de rupture du fil, qui sont les défauts de structure et les phénomènes d’hystérésis au sein du filament, les concentrations de contraintes ainsi que les forces de frottement au niveau des interactions entre le fil testé et le reste de la structure tissée.

International audience; Fabrics have been widely used as part of ballistic protections since the 1970s and the development of new ballistic solutions made from fabrics need numerical simulations, in order to predict the performance of the ballistic protection. The performances and the induced mechanisms in ballistic fabrics during an impact depend on the weaving parameters and also on the inner parameters of the yarns used inside these structures. Thus, knowing the dynamic behaviour of yarn is essential to determine the ballistic behaviour of fabrics during an impact.Two major experimental devices exist and are used to test ballistic yarns in a dynamic uniaxial tension. The first one corresponds to the Split Hopkinson Tensile Bars device, which is commonly used to characterize the mechanical properties of materials in uniaxial tension and under high loading. The second one is the transversal impact device. The real conditions of ballistic impact can be realized with this device.Then, this paper deals with a new experimental setup developed in our laboratory and called the ‘tensile impact test for yarn’ (TITY) device. With this device, specific absorbed energy measurements of para-aramid yarns (336 Tex, Twaron™, 1000 filaments) have been carried out and revealed that static and dynamic properties of para-aramid are different.

International audience; This paper deals with the study of weaving parameters and their influence on the ballistic performances of 3D fabrics. The goal is to better understand how such fabrics behave when they undertake impact loadings and to find some ways to improve their capacity to absorb the kinetic energy of projectiles. The study is focused on the layer-to-layer interlocks. As varying parameters, we study the material (aramid and polyethylene), the yarn density, the number of inner layer of the structure and the total areal density of the samples. Ballistic tests with 5 mm steel balls at different velocities are performed. Residual velocities after perforation are evaluated by high speed camera or X-ray pictures. Analysis of the experimental results and perspectives on future works are then given.

We surveyed evaluators who conduct sexually violent predator evaluations ( N = 95) regarding the frequency with which they use the Psychopathy Checklist-Revised (PCL-R), their rationale for use, and scoring practices. Findings suggest that evaluators use the PCL-R in sexually violent predator cases because of its perceived versatility, providing information about both mental disorder and risk. Several findings suggested gaps between research and routine practice. For example, relatively few evaluators reported providing the factor and facet scores that may be the strongest predictors of future offending, and many assessed the combination of PCL-R scores and sexual deviance using deviance measures (e.g., paraphilia diagnoses) that have not been examined in available studies. There was evidence of adversarial allegiance in PCL-R score interpretation, as well as a "bias blind spot" in PCL-R and other risk measure (Static-99R) scoring; evaluators tended to acknowledge the possibility of bias in other evaluators but not in themselves. Findings suggest the need for evaluators to carefully consider the extent to which their practices are consistent with emerging research and to be attuned to the possibility that working in adversarial settings may influence their scoring and interpretation practices. REVERSE is a randomized, double-masked, sham-controlled, multicenter, phase III clinical trial that evaluated the efficacy of a single intravitreal injection of rAAV2/2 ND4 in subjects with visual loss from Leber hereditary optic neuropathy (LHON). A total of 37 subjects carrying the m.11778G>A (MT-ND4) mutation and with duration of vision loss between 6 to 12 months were treated. Each subject’s right eye was randomly assigned in a 1:1 ratio to treatment with rAAV2/2 ND4 (GS010) or sham injection. The left eye received the treatment not allocated to the right eye. Unexpectedly, sustained visual improvement was observed in both eyes over the 96-week follow-up period. At Week 96, rAAV2/2 ND4-treated eyes showed a mean improvement in best-corrected visual acuity (BCVA) of -0.308 LogMAR (+15 ETDRS letters). A mean improvement of 0.259 (0.068) LogMAR (+13 ETDRS letters) was observed in the sham treated eyes. Consequently, the primary endpoint, defined as the difference in the change in BCVA from baseline to Week 48 between the two treatment groups, was not met (p = 0.894, ANCOVA). At Week 96, 25 subjects (68%) had a clinically relevant recovery in BCVA from baseline in at least one eye and 29 subjects (78%) had an improvement in vision in both eyes. A non-human primate study was conducted to investigate this bilateral improvement. Evidence of transfer of viral vector DNA from the injected eye to the anterior segment, retina and optic nerve of the contralateral non-injected eye supports a plausible mechanistic explanation for the unexpected bilateral improvement in visual function after unilateral injection.

- Abstract
We surveyed evaluators who conduct sexually violent predator evaluations ( N = 95) regarding the frequency with which they use the Psychopathy Checklist-Revised (PCL-R), their rationale for use, and scoring practices. Findings suggest that evaluators use the PCL-R in sexually violent predator cases because of its perceived versatility, providing information about both mental disorder and risk. Several findings suggested gaps between research and routine practice. For example, relatively few evaluators reported providing the factor and facet scores that may be the strongest predictors of future offending, and many assessed the combination of PCL-R scores and sexual deviance using deviance measures (e.g., paraphilia diagnoses) that have not been examined in available studies. There was evidence of adversarial allegiance in PCL-R score interpretation, as well as a "bias blind spot" in PCL-R and other risk measure (Static-99R) scoring; evaluators tended to acknowledge the possibility of bias in other evaluators but not in themselves. Findings suggest the need for evaluators to carefully consider the extent to which their practices are consistent with emerging research and to be attuned to the possibility that working in adversarial settings may influence their scoring and interpretation practices.

45. Development of a geographic information-driven real-time surveillance system for disease surveillance.

- Abstract
To respond to emerging public health threats such as West Nile virus, an advanced geographic information systems (GIS) -driven Web-based real-time surveillance system was developed to serve the National West Nile virus dead bird surveillance programme in Canada. The development of this system uses real-time Web GIS technologies and services to enhance conventional real-time surveillance systems based on real-time GIS requirements. The system has three modules: QuickTrack, QuickMap and QuickManage. QuickTrack is the real-time surveillance module that supports data collection, edit and transfer. QuickMap is the real-time Web GIS module that provides comprehensive real-time GIS supports and services in public health surveillance and information sharing. The QuickManage module is a Web-based system management package used to manage the entire system. This system offers an effective approach to enhance real-time public health surveillance systems by integrating real-time Web GIS technologies and services. The system demonstrates that real-time Web GIS technologies can play an important role in enhancing public health surveillance systems.
