Mutations in C8orf37 cause Bardet-Biedl syndrome (BBS), retinitis pigmentosa (RP), and cone–rod dystrophy (CRD), all manifest in photoreceptor degeneration. Little is known about which proteins C8orf37 interacts with to contribute to photoreceptor survival. To determine the proteins that potentially interact with C8orf37, we carried out a yeast two-hybrid (Y2H) screen using C8orf37 as a bait. FAM161A, a microtubule-binding protein localized at the photoreceptor cilium required for photoreceptor survival, was identified as one of the preys. Double immunofluorescence staining and proximity ligation assay (PLA) of marmoset retinal sections showed that C8orf37 was enriched and was co-localized with FAM161A at the ciliary base of photoreceptors. Epitope-tagged C8orf37 and FAM161A, expressed in HEK293 cells, were also found to be co-localized by double immunofluorescence staining and PLA. Furthermore, interaction domain mapping assays identified that the N-terminal region of C8orf37 and amino acid residues 341-517 within the PFAM UPF0564 domain of FAM161A were critical for C8orf37–FAM161A interaction. These data suggest that the two photoreceptor survival proteins, C8orf37 and FAM161A, interact with each other which may contribute to photoreceptor health. [ABSTRACT FROM AUTHOR]

Background: The benefits of performing open versus laparoscopic gubernaculum-sparing second-stage Fowler-Stephens orchiopexy (FSO) remain unclear. We compared the two techniques to answer this question. Methods: We retrospectively studied a cohort of patients who underwent laparoscopic first-stage FSO and open versus laparoscopic gubernaculum-sparing second-stage FSO at our institution between September 2004 and June 2020 (all patients underwent surgery by a single surgeon). We evaluated both procedures based on the incidence of testicular atrophy, testicular ascent, and other complications. Results: The age at initial surgery was 45.7 ± 28.2 months (median, 39). One hundred nine cases were treated with open second-stage gubernaculum-sparing FSO (OFSO), and 96 cases were treated with laparoscopic second-stage gubernaculum-sparing FSO (LFSO). The mean follow-up period was 59.8 months (median, 54; standard deviation, +35). The overall testicular atrophy rate was 1.5%. Atrophy was observed in 2 and 1 patient in the OFSO and LFSO groups, respectively (1.8% versus 1.0%, P > .05). There was no significant difference in the incidence of testicular ascent between both groups (2.8% versus 3.1%). Five and four complications were noted in the OFSO and LFSO groups, respectively (P > .05). Conclusions: Second-stage gubernaculum-sparing FSO achieved high testicular survival rates and satisfactory testicular positions. Neither the open nor laparoscopic approach appeared superior, because the overall testicular survival rates and incidence of testicular ascent and other complications were equivalent between both groups. Background: Negative body image is a common psychological phenomenon among Chinese college students meriting investigation. Peers and the media are important factors that negatively influence body image. This study explored the mechanisms of media and peers promoting negative body image among Chinese college students. Method: Data from 407 college students (173 men and 234 women) were collected using the Revised Social and Cultural Attitude Questionnaire of Appearance, Appearance Comparison Scale, Peer Impact Scale, and Negative Body Image Scale. Results: Correlational analysis results demonstrated that media attention was not significantly correlated with negative body image. All other variables were significantly positively correlated. Path analysis results indicated that the direct effect of media attention on negative body image was not significant, while the direct effect of peer impact was significant. In addition, appearance comparison and internalization of the thin ideal acted as a chained indirect effect between negative body image, media attention, and peer impact. Conclusions: The research revealed that focusing on perfect bodies displayed in the media did not produce a negative body image. However, focusing on the media and peer conversations regarding the body caused the participants to compare appearances and internalize ideal body shape standards, leading to negative self-evaluations. Plain English Summary: Adolescents' peers and media have far-reaching influence. This study explored their impact on the body image of Chinese college students. A questionnaire survey was distributed to 461 Chinese college students. The results showed that peers directly affected body image, while media attention did not. Background: Although the morbidity and mortality rates associated with idiopathic pulmonary fibrosis (IPF) are high, there is still lack of powerful and precise therapeutic options for IPF. Object: Through in vitro model, this study sought to determine whether binding of acetylated CCAAT/enhancer binding protein β (C/EBPβ) to alpha-smooth muscle actin (α-SMA) promoter could affect the activity of the latter as well as assess if it is essential for epithelial-to-mesenchymal transition (EMT) and extracellular matrix deposition in IPF. Methods: The expression of EMT and C/EBPβ in A549 cells treated with transforming growth factor-beta (TGF-β) as pulmonary fibrotic model was detected by western blotting and qPCR. Collagen-I expression using ELISA was performed. The luciferase activity was used to examine the activity of C/EBPβ. Knockdown of C/EBPβ was performed by siRNA. We also investigated the effect of deacetylation of C/EBPβ on EMT using sirtuin 1 (SIRT1). The binding ability of C/EBPβ with α-SMA promoter was affirmed via chromatin immunoprecipitation (ChIP) and electrophoresis mobility shift assay (EMSA). The relationship between α-SMA and acetylated C/EBPβ was determined with co-immunoprecipitation (Co-IP). SiRNA-mediated knockdown of C/EBPβ in A549 cells attenuated TGF-β1-induced myofibroblast differentiation and ECM deposition. The extent of association between acetylated C/EBPβ and α-SMA promoter was dynamically monitored. Results: It was confirmed that deacetylation of C/EBPβ in A549 cells successfully ameliorated TGF-β1-induced EMT, as shown by reduction in α-SMA expression and excessive collagen-I accumulation. Conclusion: The EMT and fibrotic effect of TGF-β1 is dependent on acetylated C/EBPβ-mediated regulation of α-SMA gene activity. Thus, C/EBPβ acetylation may play a central role in pulmonary fibrosis. Extracellular protease Vpr (Vpr), gamma-glutamyltranspeptidase (GGT; EC 2.3.2.2) and glyoxal/methylglyoxal reductase (YvgN; EC 1.1.1.21) are extracellular enzymes involved in feather degradation, which were identified by secretome analyses from an efficient feather-degrading strain Bacillus subtilis CH-1. The encoding sequences corresponding to the three secretory enzymes were cloned into vector pET22b for recombinant expression in Escherichia coli strain BL21 (DE3). Afterward, the proteins containing the C-terminal His-tag were purified using a Ni-IDA column. The optimal temperatures and pH values for protease activity of recombinant Vpr, GGT, and YvgN were identified as 45 °C/pH 7.0, 40 °C/pH 8.0, and 50 °C/pH 6.0 respectively when casein is the substrate. Furthermore, the synergistic effects of the three enzymes were studied using feather powder as substrate. Vpr was the core enzyme to hydrolyze keratin, while GGT and YvgN were coenzymes providing reducing activities for keratin decomposition. The keratinolytic activity was enhanced to about 1.4-folds when YvgN and Vpr applied together in comparison to Vpr alone. And the keratinolytic activity almost reached to 1.5-folds when all the three enzymes were combined to use. The study provides a novel perspective of the mechanism of keratin degradation by microorganisms, and thereby may also be of relevance for the design of an industrial process for enzymatic keratin degradation; however, additional experiments must be done to substantiate this conclusion. In this study, TiO2 nanorod arrays (TNR), Ag quantum dots (QDs) sensitized with TNR TiO2/Ag, bismuth oxyhalide (BiOI) nanosheets, and Ag QDs co‐modified with TNR and TiO2/BiOI/Ag (TBA) were prepared by a stepwise process. The morphological, structural, compositional, optical, photocatalytic (PC), and photoelectrochemical (PEC) properties of the samples were investigated. The TBA‐2 sample exhibited the highest photocurrent density (281.8 μA/cm2) and photodegradation efficiency (93.3%), with values 9.7 times and 2.25 times higher than those for TNR, respectively. The improvement in sample performance can be attributed to the formation of a heterojunction between BiOI and TiO2, thereby enhancing the absorption of visible light and improving the charge separation efficiency; Ag QDs limit interfacial electron‐hole pair recombination. The experimental results show that TBA can effectively promote light‐induced carrier transport and visible light absorption, while inhibiting the recombination rate of the electron‐hole pairs, PEC, and PC. The mixed-phase heterostructured TiO 2 nanotree arrays were synthesized by a secondary hydrothermal route, and Ti3+ ions were introduced into the arrays for enhanced photoelectrochemical performance. The results show that the arrays consist of heterostructures with anatase and rutile phases. XPS results indicate that during the hydrogenation process, part of Ti4+ ions transform to Ti3+ ions. Although the band gap shifts by only 0.1 eV, introduced defects and Ti3+ ions can greatly increase the efficiency of electron-hole separation. Photocurrent of the heterostructured TiO 2 nanotree arrays with introduced Ti3+ ions increases by 41 times, and the photocurrent density enhances from 0.0055 to 0.2272 mA/cm2. • Ti3+ ions were introduced into the arrays for enhanced photoelectrochemical performance. • It is a clean and convenient way to construct the inner structure of TiO 2. • Heterostructured TiO 2 nanotree arrays with large specific surface area were fabricated successfully. Abstract In this paper, highly ordered titanium dioxide nanorod (TNR) arrays decorated with Ag-Ag 2 S quantum-dots (QDs) were synthesized on FTO substrate through hydrothermal treatment and in-situ vulcanization. The content of Ag 2 S QDs on Ag/TNR was controlled by changing the immersion time of Ag/TNR in the N-N-dimethyl formamide (DMF) solution containing dissolved sulfur element. The morphology, structure, composition and optical, photoelectric and photocatalytic properties of QDs/TNR arrays were studied in detail. In the visible range, the QDs/TNR array with a soaking time of 12 h exhibits the maximal photocurrent density (0.082 mA/cm2) and photodegradation rate (62.2%), which is 5.5 and 1.85 times higher than that of pure TNR. The improvement of these properties could be attributed to synergy effect between Ag and Ag 2 S materials. That is, Ag QDs can stably capture and transmit photo-generated electrons from the TNR surface, whereas the Ag 2 S promoter is regarded as the interface active sites to enhance photocatalytic reactions. Therefore, the synergistic effect of Ag-Ag 2 S can significantly enhance the photoelectrochemical and photocatalytic properties of the TNR arrays. The authors report the genetic composition and stoichiometry of endogenous subfamilies of PSD95‐containing supercomplexes in the mouse brain using an innovative strategy of combining gene‐tagging knock‐in, targeted mutations, and quantitative biochemical assays. Big sensing data is prevalent in both industry and scientific research applications where the data is generated with high volume and velocity. Cloud computing provides a promising platform for big sensing data processing and storage as it provides a flexible stack of massive computing, storage, and software services in a scalable manner. Current big sensing data processing on Cloud have adopted some data compression techniques. However, due to the high volume and velocity of big sensing data, traditional data compression techniques lack sufficient efficiency and scalability for data processing. Based on specific on-Cloud data compression requirements, we propose a novel scalable data compression approach based on calculating similarity among the partitioned data chunks. Instead of compressing basic data units, the compression will be conducted over partitioned data chunks. To restore original data sets, some restoration functions and predictions will be designed. MapReduce is used for algorithm implementation to achieve extra scalability on Cloud. With real world meteorological big sensing data experiments on U-Cloud platform, we demonstrate that the proposed scalable compression approach based on data chunk similarity can significantly improve data compression efficiency with affordable data accuracy loss. Discussion on the role of DEHP in the critical period of gonadal development in pregnant rats (F0), studied the evolution of F1-F4 generation of inter-generational inheritance of cryptorchidism and the alteration of DNA methylation levels in testis. Pregnant SD rats were randomly divided into two groups: normal control group and DEHP experimental group. From pregnancy 7d to 19d, experimental group was sustained to gavage DEHP 750mg/kg bw/day, observed the incidence of cryptorchidism in offspring and examined the pregnancy rate of female rats through mating experiments. Continuous recording the rat’s weight and AGD value, after maturation (PND80) recording testis and epididymis’ size and weight, detected the sperm number and quality. Subsequently, we examined the evolution morphological changes of testicular tissue for 4 generation rats by HE staining and Western Blot. Completed the MeDIP-sequencing analysis of 6 samples (F1 generation, F4 generation and Control). DEHP successfully induced cryptorchidism occurrence in offspring during pregnancy. The incidence of cryptorchidism in F1 was 30%, in F2 was 12.5%, and there was no cryptorchidism coming up in F3 and F4. Mating experiment shows conception rate 50% in F1, F2 generation was 75%, the F3 and F4 generation were 100%. HE staining showed that the seminiferous epithelium of F1 generation was atrophy and with a few spermatogenic cell, F2 generation had improved, F3 and F4 generation were tend to be normal. The DNA methyltransferase expression was up-regulated with the increase of generations by Real Time-PCR, immunohistochemistry and Western Blot. MeDIP-seq Data Analysis Results show many differentially methylated DNA sequences between F1 and F4. DEHP damage male reproductive function in rats, affect expression of DNA methyltransferase enzyme, which in turn leads to genomic imprinting methylation pattern changes and passed on to the next generation, so that the offspring of male reproductive system critical role in the development of imprinted genes imbalances, and eventually lead to producing offspring cryptorchidism. Cloud computing opens a new era in IT as it can provide various elastic and scalable IT services in a pay-as-you-go fashion, where its users can reduce the huge capital investments in their own IT infrastructure. In this philosophy, users of cloud storage services no longer physically maintain direct control over their data, which makes data security one of the major concerns of using cloud. Existing research work already allows data integrity to be verified without possession of the actual data file. When the verification is done by a trusted third party, this verification process is also called data auditing, and this third party is called an auditor. However, such schemes in existence suffer from several common drawbacks. First, a necessary authorization/authentication process is missing between the auditor and cloud service provider, i.e., anyone can challenge the cloud service provider for a proof of integrity of certain file, which potentially puts the quality of the so-called ‘auditing-as-a-service’ at risk; Second, although some of the recent work based on BLS signature can already support fully dynamic data updates over fixed-size data blocks, they only support updates with fixed-sized blocks as basic unit, which we call coarse-grained updates. As a result, every small update will cause re-computation and updating of the authenticator for an entire file block, which in turn causes higher storage and communication overheads. In this paper, we provide a formal analysis for possible types of fine-grained data updates and propose a scheme that can fully support authorized auditing and fine-grained update requests. Based on our scheme, we also propose an enhancement that can dramatically reduce communication overheads for verifying small updates. Theoretical analysis and experimental results demonstrate that our scheme can offer not only enhanced security and flexibility, but also significantly lower overhead for big data applications with a large number of frequent small updates, such as applications in social media and business transactions. Microplastic pollution has gradually become a major global concern, due to the widespread use of plastics. Microplastics enter the environment and are degradated, while also being ingested by organisms, affecting various physiological functions and adversely affecting the health of organisms. Microplastic pollution is currently a wide concern, but data on the impact on organisms is still not sufficient. Therefore, this review summarizes the research on microplastic pollution in marine, soil and fresh water, and its impact on organisms, focusing on the effects of microplastics on organisms' feeding behavior and oxidative stress responses, intestinal microbes and reproductive function, and the combined effects of microplastic pollutants on organisms. Abstract: Temporal constraints are often set when complex e-science processes are modelled as scientific workflow specifications. However, many existing processes such as climate modelling often have only a few coarse-grained temporal constraints globally. This is not sufficient to control overall temporal correctness as we can not find temporal violations locally in time for handling. Local handling affects fewer workflow activities, hence more cost effective than global handling with coarse-grained temporal constraints. Therefore, in this paper, we systematically investigate how to localise a group of fine-grained temporal constraints so that temporal violations can be indentified locally for better handling cost effectiveness. The corresponding algorithms are developed. The quantitative evaluation demonstrates that with local fine-grained temporal constraints, we can improve handling cost effectiveness significantly than only with coarse-grained ones. Abstract: The wolf spider Lycosa singoriensis is a hunting spider with a widespread distribution in northwest China. The venom gland of spiders, which is a very specialized secretory tissue, can secrete abundant and complex toxin components. To extensively examine the transcripts expressed in the venom glands of L. singoriensis, we generated 833 expressed sequence tags (ESTs) from a directional cDNA library. Toxin-like sequences account for 69.1% of these ESTs, 17.3% are similar to cellular transcripts and 13.6% have no significant similarity to any known sequences. Here, we identified 223 novel toxin-like sequences, which can be classified into six different superfamilies; that means a novel potential source of ligands for varied ion channels was discovered. With the aid of Gene Ontology terms and homology to eukaryotic orthologous groups, the annotation of cellular transcripts revealed some cellular processes important for the toxin secretion of venom glands including protein synthesis, protein folding, tuned post-translational processing and trafficking, etc. [Copyright &y& Elsevier]

Abstract: Jingzhaotoxin-34 (JZTX-34) is a 35-residue polypeptide from the venom of Chinese tarantula Chilobrachys jingzhao. Our previous work reported its full-length cDNA sequence encoding a precursor with 87 residues. In this study we report the protein expression and biological function characterization. The toxin was efficiently expressed by the secretary pathway in yeast. Under whole-cell patch-clamp mode, the expressed JZTX-34 was able to inhibit tetrodotoxin-sensitive (TTX-S) sodium currents (IC50 ∼85nM) while having no significant effects on tetrodotoxin-resistant (TTX-R) sodium currents on rat dorsal root ganglion neurons. The inhibition of TTX-S sodium channels was completely reversed by strong depolarization (+120mV). Toxin treatment altered neither channel activation and inactivation kinetics nor recovery rate from inactivation. However, it is interesting to note that in contrast to huwentoxin-IV, a recently identified receptor site-4 toxin from Ornithoctonus huwena venom, 100nM JZTX-34 caused a negative shift of steady-state inactivation curve of TTX-S sodium channels by approximately 10mV. The results indicated that JZTX-34 might inhibit mammalian sensory neuronal sodium channels through a mechanism similar to HWTX-IV by trapping the IIS4 voltage sensor in the resting conformation, but their binding sites should not overlay completely. Abstract: Hepatitis B surface antigen (HBsAg) loss under antiviral therapy is rare in chronic hepatitis B patients and the dynamics of serum HBsAg in these patients are not available. The changes in serum HBsAg following treatment with adefovir (n =31) or peg-interferon-α-2a (n =23) were studied in hepatitis B e-antigen (HBeAg) positive chronic hepatitis B patients. Abbott Architect HBsAg assay was used to quantify serum HBsAg. HBsAg levels were significantly decreased during the first 12 weeks of treatment with median change of −397.0IU/ml and −555.4IU/ml, respectively for adefovir and peg-interferon-α-2a (p =0.005 and 0.001, respectively). Beyond 12 weeks, no further significant HBsAg reductions were found even in patients with sustained viral replication inhibition in either group. Three distinct patterns of HBsAg changes were observed in most patients in both treatment groups: biphasic pattern (rapid HBsAg reduction from baseline to week 12); assurgent pattern (higher HBsAg level at week 12 than at baseline); and wavy pattern (HBsAg reduction from baseline to week 12, followed by relapse at week 24 or week 28). These results might offer insights into the possible mechanism(s) underlying the unusual occurrences of HBsAg loss under antiviral therapy. [Copyright &y& Elsevier]

Abstract: The tarantula Chilobrachys jingzhao is one of the most venomous spiders with a specialized organ, venom gland, which synthesizes and secretes the complex and abundant toxin proteins. The components of the venom have been extensively studied. As far as the molecular mechanism of toxin secretion and metabolism is concerned, we still knew a little. To obtain a comprehensive view of function of its venom gland we constructed a non-normalized cDNA library of the venom gland and generated 788 expressed sequence tags (ESTs). All ESTs were assembled into 356 non-redundancy sequences including 85 clusters and 271 singlets, of which 31.4% of total unique sequences belong to secretion protein coding sequences including cystine knot toxins (29.1%) and other secretion proteins (2.3%); 54.0% are similar to common cellular transcripts; and 14.6% have no significant similarity to any known sequences. Annotation of these ESTs revealed some novel possible venom components and cellular processes important for venom gland functions, including protein posttranslation processing, cell motility, protein synthesis, energy supply, etc. This study contains a transcriptome analysis of spider venom gland focusing on its cellular structural and functional aspects, and confirms the very specialized nature of the spider venom gland as toxin producer. Abstract: The bird spider Ornithoctonus huwena is one of the most venomous spiders in China. Its venom is a mixture of various compounds with diverse bioactivities. Ninety proteins and 47 peptides have been identified, and 67 cDNA sequences encoding different toxin precursors have been cloned. However, the genomic DNA of them is seldom reported. To characterize the genomic DNA structure of huwentoxins, the genomic DNA encoding toxins of three superfamilies were cloned by using sequence specific or partially degenerate primers based on their cDNA sequences. An unexpected finding was that the intron was lacking in the genomic sequences of three superfamilies. The genomic DNA information has predictive value for better understanding the relationship of spider toxin evolution. In addition, we have cloned and analyzed 19 novel genes encoding toxin-like precursors by using the genomic DNA of the spider O. huwena. [Copyright &y& Elsevier]

The plant pathogenesis-related (PR) proteins play a crucial role in the defense of plants against pathogens and orchestrate the innate immune system of plants. In this paper, a non-normalized cDNA library of the leaf was constructed to obtain a comprehensive view of PR proteins of Macleaya cordata. Specifically, 511 expressed sequence tags (ESTs) were generated using Sanger sequencing. All ESTs were assembled into 364 non-redundancy sequences, including 78 clusters and 286 singlets. The PR protein expression profile of the medicinal herb M. cordata has been investigated and is represented by defensin, lipid-transfer protein, (S)-norcoclaurine synthase, and major allergen protein, suggesting that the herb contains rich active proteins against pathogens. Furthermore, two defensins were selected for recombinant expression in yeast, and the antimicrobial activities were explored. Since they both present a broad antimicrobial spectrum, they are of particular importance for agricultural and medicinal applications. Our study describes defensins in Papaveraceae for the first time and provides novel insights into the effective components. • This work proposes a parallel and distributed deep learning framework for IoT data analytics. • The proposed method is implemented and deployed with a real-world problem of energy consumption forecasting. • Comprehensive comparative study has been conducted to show the outperformance of the proposed method. Energy consumption forecasting based on IoT data and deep learning algorithm inheriting distributed and collaborative learning is a widely studied topic both in engineering and computer science fields. For different households with drastically different energy consumption patterns, the traditional centralized machine learning (ML) and deep learning (DL) methods suffer problems including inaccuracy, inefficiency and laggings of the prediction performance. In this study, we propose a sophisticated multi-channel bidirectional nested LSTM framework (MC-BiNLSTM) combined with discrete stationary wavelet transform (SWT) for highly accurate and efficient energy consumption forecasting. The main contributions of this study include the decomposition using SWT for accuracy improvement and the collaborative BiNLSTM structure for efficiency improvement. Microplastics pollution poses a new threat to the environment of intertidal zone. The sea forest, mangrove, has been polluted by a large number of plastic debris worldwide. To fill the gaps in knowledge of mangrove rhizosphere microbes connected with the 'plasticsphere', a semi-controlled in situ exposure experiment for nine different types of microplastics were conducted in mangrove ecosystem. A sign of biodegrading was observed on polyethylene, polyamide 6 and polyvinyl chloride microplastics surface after 3 months exposure. We discovered that the metabolic activities of the dominant bacteria on certain microplastics were related to the specific groups on polymers molecule. The selective colonization may be driven by the chemotaxis of bacteria. Specially, microplastics biofilms of polyethylene, polyamide 6, polyvinyl chloride and expanded polystyrene possess distinctive dominant bacteria assemblages which have great significance in ecosystem processes involving carbon cycle or sulfur cycle. Community of mangrove soil microorganism and microplastic biofilm varies as the seasons changes. As a new niche, microplastics has higher inclusivity to bacteria than surrounding soil. Additionally, pathogens for human beings (Vibrio parahaemolyticus and Escherichia - Shigella) were detected both in microplastics and soil. We stress that the interaction between microplastics and rhizosphere microorganisms may affect the growth and health of mangrove plants. Besides, we point out that mangrove rhizosphere microorganism can be an ideal candidate for plastics-degradation. Bacterial quorum sensing (QS) and bacterial extracellular polymeric substances (EPS) are the major challenges in treating biofilm chronic infection. Herein, we proposed targeting intervention phase separation of EPS by nano-antibiotics (Ir(ppy) 2 dmb-PAZ-HA) as an effective strategy for combating bacterial biofilm infection, in which Iridium complex (Ir(ppy) 2 dmb-CHO) and antibiotic (Pazufloxacin, PAZ) for synergistic antibacterial, and hyaluronic acid (HA) was used to enhance Ir(ppy) 2 dmb-PAZ release on-demand in biofilm microenvironment. In vitro results showed that Ir(ppy) 2 dmb-PAZ-HA holds excellent antibacterial activity and antibiofilm capacity. Importantly, Ir(ppy) 2 dmb-PAZ-HA can be target to regulate phase separation of EPS, and then interfere with biofilm formation, which is a competitive mechanism between Ir(ppy) 2 dmb-PAZ-HA and Methicillin-resistant Staphylococcus aureus (MRSA) itself. In addition, in vivo results confirmed that Ir(ppy) 2 dmb-PAZ-HA was able to accelerate biofilm chronic wound healing, and effectively alleviate bacterial acute lung infection. Background & Aims: aMAP score, as a hepatocellular carcinoma risk score, is proven to be associated with the degree of chronic hepatitis B‐related liver fibrosis. We aimed to evaluate the ability of aMAP score for metabolic dysfunction‐associated steatotic liver disease (MASLD; formerly NAFLD)‐related fibrosis diagnosis and establish a machine‐learning (ML) model to improve the diagnostic performance. Methods: A total of 946 biopsy‐proved MASLD patients from China and the United States were included in the analysis. The aMAP score, demographic/clinical indices and liver stiffness measurement (LSM) were included in seven ML algorithms to build fibrosis diagnostic models in the training set (N = 703). The performance of ML models was evaluated in the external validation set (N = 125). Results: The AUROCs of aMAP versus fibrosis‐4 index (FIB‐4) and aspartate aminotransferase‐platelet ratio (APRI) in cirrhosis and advanced fibrosis were (0.850 vs. 0.857 [P = 0.734], 0.735 [P = 0.001]) and (0.759 vs. 0.795 [P = 0.027], 0.709 [P = 0.049]). When using dual cut‐off values, aMAP had a smaller uncertainty area and higher accuracy (26.9%, 86.6%) than FIB‐4 (37.3%, 85.0%) and APRI (59.0%, 77.3%) in cirrhosis diagnosis. The seven ML models performed satisfactorily in most cases. In the validation set, the ML model comprising LSM and 5 indices (including age, sex, platelets, albumin and total bilirubin used in aMAP calculator), built by logistic regression algorithm (called LSM‐plus model), exhibited excellent performance. In cirrhosis and advanced fibrosis detection, the LSM‐plus model had higher accuracy (96.8%, 91.2%) than LSM alone (86.4%, 67.2%) and Agile score (76.0%, 83.2%), respectively. Additionally, the LSM‐plus model also displayed high specificity (cirrhosis: 98.3%; advanced fibrosis: 92.6%) with satisfactory AUROC (0.932, 0.875, respectively) and sensitivity (88.9%, 82.4%, respectively). Simple Summary: Rhodotorula mucilaginosa is widely detected in the environment, and it produces broad metabolites such as carotenoid pigments, amino acids, polyunsaturated fatty acids, polysaccharides, and enzymatic substances. We previously isolated a strain of Rhodotorula mucilaginosa ZTHY2 from the coastal waters of the South China Sea, which showed probiotic properties. Rhodotorula mucilaginosa ZTHY2 promoted the growth of experimental animals, increased the abundance of beneficial bacteria in gut, and enhanced immune function. Given that immunosuppression is common in livestock, environmental stresses, such as overpopulation, food contamination, and chronic diseases, contribute to immunosuppression of livestock associated with declining animal production, failed immunoprophylaxis, and economic losses. Therefore, in this study, cyclophosphamide was used to generate immunosuppressed models to further evaluate the immunomodulatory effects of Rhodotorula mucilaginosa ZTHY2. Rhodotorula mucilaginosa (R. mucilaginosa) can enhance the immune and antioxidant function of the body. However, whether R. mucilaginosa has an immunoregulatory effect on cyclophosphamide (CTX)-induced immunosuppressed animals remains to be clarified. In this study, the R. mucilaginosa ZTHY2 that we isolated from the coastal waters of the South China Sea previously was prepared in order to investigate its immunoprotective effect on CTX-induced immunosuppression in mice, and the effects were compared to those of Lactobacillus acidophilus (LA) (a well-known probiotic). Seventy-two male SPF mice were divided into six groups: The C group (control); IM group (immunosuppressive model group) (+CTX); Rl, Rm, and Rh groups (+CTX+low, medium, and high concentration of R. mucilaginosa, respectively); and PC (positive control) group (+CTX+LA). After a 28-day feeding trial, blood samples were taken for biochemical and serum immunological analysis, and the thymus and spleen were collected to analyze the organ index, lymphocyte proliferation and differentiation, and antioxidant capacity. The findings showed that R. mucilaginosa ZTHY2 improved the spleen and thymus indices, effectively attenuated immune organ atrophy caused by CTX, and enhanced the proliferation of T and B lymphocytes induced by ConA and LPS. R. mucilaginosa ZTHY2 promoted the secretion of cytokines and immunoglobulins and significantly increased the contents of IL-2, IL-4, IL-6, TNF-α, IFN-γ, IgA, IgG, IgM, CD4, CD8, CD19, and CD20 in serum. The proportion of CD4+, CD8+, CD19+, and CD20+ lymphocytes in spleen, thymus, and mesenteric lymph nodes were increased. In addition, R. mucilaginosa ZTHY2 reduced the reactive oxygen species (ROS) and malondialdehyde (MDA) levels and increased glutathione (GSH), total superoxide dismutase (SOD), and catalase (CAT) levels. Our results indicated that R. mucilaginosa ZTHY2 can significantly enhance the immune function of immunosuppressed mice, and improving antioxidant capacity thus attenuates CTX-induced immunosuppression and immune organ atrophy. Blockchain has received a widespread attention because of its decentralized, tamper-proof, and transparent nature. Blockchain works over the principle of distributed, secured, and shared ledger, which is used to record, and track data within a decentralized network. This technology has successfully replaced certain systems of economic transactions in organizations and has the potential to overtake various industrial business models in future. Blockchain works over peer-to-peer (P2P) phenomenon for its operation and does not require any trusted-third party authorization for data tracking and storage. The information stored in blockchain is distributed throughout the decentralized network and is usually protected using cryptographic hash functions. Since the beginning of blockchain technology, its use in different applications is increasing exponentially, but this increased use has also raised some questions regarding privacy and security of data being stored in it. Protecting privacy of blockchain data using data perturbation strategy such as differential privacy could be a novel approach to overcome privacy issues in blockchain. In this article, we cover the topic of integration of differential privacy in each layer of blockchain and in certain blockchain based scenarios. (1) Background: Hepatic venous occlusion type of Budd–Chiari syndrome (BCS-HV) and pyrrolizidine alkaloid-induced hepatic sinusoidal obstructive syndrome (PA-HSOS), share similar clinical features, and imaging findings, leading to misdiagnoses; (2) Methods: We retrospectively analyzed 139 patients with BCS-HV and 257 with PA-HSOS admitted to six university-affiliated hospitals. We contrasted the two groups by clinical manifestations, laboratory tests, and imaging features for the most valuable distinguishing indicators.; (3) Results: The mean patient age in BCS-HV is younger than that in PA-HSOS (p < 0.05). In BCS-HV, the prevalence of hepatic vein collateral circulation of hepatic veins, enlarged caudate lobe of the liver, and early liver enhancement nodules were 73.90%, 47.70%, and 8.46%, respectively; none of the PA-HSOS patients exhibited these features (p < 0.05). DUS showed that 86.29% (107/124) of patients with BCS-HV showed occlusion of the hepatic vein, while CT or MRI showed that only 4.55%(5/110) patients had this manifestation (p < 0.001). Collateral circulation of hepatic veins was visible in 70.97% (88/124) of BCS-HV patients on DUS, while only 4.55% (5/110) were visible on CT or MRI (p < 0.001); (4) Conclusions: In addition to an established history of PA-containing plant exposure, local hepatic vein stenosis and the presence of collateral circulation of hepatic veins are the most important differential imaging features of these two diseases. Background the incidence of acute-on-chronic liver disease (AoCLD) is increasing. Objective to investigate the clinical features and risk factors of AoCLD and construct an effective prognostic nomogram model for older patients with AoCLD. Methods data from 3,970 patients included in the CATCH-LIFE study were used, including 2,600 and 1,370 patients in the training and validation sets, respectively. Multivariate Cox regression analyses were performed to identify predictive risk factors in older individuals, and an easy-to-use nomogram was established. Performance was assessed using area under the curve, calibration plots and decision curve analysis (DCA). Results of the 3,949 patients with AoCLD, 809 were older with a higher proportion of autoimmune-related abnormalities, hepatitis C viral infection and schistosomiasis. In the older patient group, the incidence of cirrhosis, hepatic encephalopathy (HE), infection, ascites and gastrointestinal bleeding; neutrophil-to-lymphocyte ratio (NLR), aspartate-to-alanine transaminase ratio (AST/ALT), creatinine and blood urea nitrogen levels were higher, whereas incidence of acute-on-chronic liver failure, white blood cell, platelet and haemoglobin levels; albumin, total bilirubin (TB), AST and ALT levels; international normalised ratio (INR), estimated glomerular filtration rate and blood potassium levels were lower than in the younger group. The final nomogram was developed based on the multivariate Cox analysis in training cohort using six risk factors: ascites, HE grades, NLR, TB, INR and AST/ALT. Liver transplantation-free mortality predictions were comparable between the training and validation sets. DCA showed higher net benefit for the nomograph than the treat-all or treat-none strategies, with wider threshold probabilities ranges. Background: Whether serum hepatitis B virus (HBV) RNA associates with hepatocellular carcinoma (HCC) development in chronic hepatitis B (CHB) patients has not been fully elucidated.Methods: We enrolled 2974 patients receiving nucleos(t)ide analogues (NAs) from a prospective, observational CHB cohort to investigate the effect of serum HBV RNA, measured at study entry (baseline), on HCC development, using Cox regression analyses.Results: During median follow-up of 4.4 years, 90 patients developed HCC. Patients with detectable baseline HBV RNA (n = 2072) exhibited significantly higher HCC risk than those with undetectable level (5-year HCC incidence estimated by Kaplan-Meier method: 4.1% versus 1.8%, P = .009; adjusted hazard ratio [aHR] = 2.21, P = .005). HBV RNA levels of 609-99 999 and ≥100 000 copies/mL were associated with incrementally increasing HCC risk (aHR = 2.15 and 3.05, respectively; P for trend = .003), compared to undetectable level (<609 copies/mL). Moreover, patients with single-detectable either HBV DNA or RNA and double-detectable DNA and RNA had 1.57- and 4.02-fold higher HCC risk, respectively, than those with double-undetectable DNA and RNA (P for trend = .001).Conclusions: High-level HBV RNA is associated with increased HCC risk in NAs-treated patients. Cloud computing has rendered its ever-increasing advantages in flexible service provisions, which attracts the attentions from large-scale enterprise applications to small-scale smart uses. For example, more and more multimedia services are moving towards cloud to better accommodate people's daily uses on various smart devices that support cloud, some of which are similar or equivalent in their functionality (e.g., more than 1,000 video services that share similar 'video-play' functionality are present in APP Store). In this situation, it is necessary to discriminate these functional-equivalent multimedia services, based on their Quality of Service (QoS) information. However, due to the abundant information of multimedia content, dozens of QoS criteria are often needed to evaluate a multimedia service, which places a heavy burden on users' multimedia service selection. Besides, the QoS criteria of multimedia services are usually not independent, but correlated, which cannot be accommodated very well by the traditional selection methods, e.g., traditional simple weighting methods. In view of these challenges, we put forward a multimedia service selection method based on weighted Principal Component Analysis (PCA), i.e., Weighted PCA-based Multimedia Service Selection Method (W_PCA_MSSM). The advantage of our proposal is two-fold. First, weighted PCA could reduce the number of QoS criteria for evaluation, by which the service selection process is simplified. Second, PCA could eliminate the correlations between different QoS criteria, which may bring a more accurate service selection result. The computation offloading problem in mobile edge computing (MEC) has received a lot of attention, but service caching is also a research topic that cannot be ignored in MEC. Due to the limited resources available on the Edge Server (ES), a wise computation offloading and service caching policy must be formulated in order to maximize system offload efficiency. In this paper, a many-objective joint optimization computation offloading and service caching model (MaJOCOSC) is designed. The model takes into account the limited computing and storage resources of ES, the delay and energy consumption constraints of different types of tasks, and multiple processing modes of user tasks, and sets delay, energy consumption, task hit service rate, service cache balancing, and load balancing as the five optimization objectives of MaJOCOSC. Meanwhile, a non-dominated sorting genetic algorithm (NSGAIII-ASF&WD) based on achievement scalar function (ASF) and the k-nearest neighbor weighted distance mating selection strategy is proposed for better solving the model. The ASF ensures that the given strategy performs well for each objective value, and the k-nearest neighbor weighted distance provides the user with a diversity of strategies. Simulation results show that NSGAIII-ASF&WD can obtain better objective values when solving the model compared with other many-objective evolutionary algorithms, and a suitable computation offloading and service caching strategy is obtained. Leaves and flowers of Amorphophallus konjac do not develop simultaneously thus unique features can be elucidated through study of flowering transformation in A. konjac. In this study, transcriptome libraries of A. konjac leaf buds (LB) and flower buds (FB) were constructed followed by high-throughput sequencing. A total of 68,906 unigenes with an average length of 920 bp were obtained after library assembly. Out of these genes, 24,622 unigenes had annotation information. A total of 6859 differentially expressed genes (DEGs) were identified through differential expression analysis using LB as control. Notably, 2415 DEGs were upregulated whereas 4444 DEGs were downregulated in the two transcriptomes. Go and KEGG analysis showed that the DEGs belonged to 44 functional categories and were implicated in 98 metabolic pathways and 38 DEGs involved in plant hormone signal transduction. Several genes were mined that may be involved in A. konjac flower bud differentiation and flower organ development. Eight DEGs were selected for verification of RNA-seq results using qRT-PCR analysis. Two FLOWERING LOCUS T (FT) genes named AkFT1 and AkFT2 were identified though homologous analysis may be the florigen gene implicated in modulation of A. konjac flowering. These genes were significantly upregulated in flower buds compared with the expression levels on leaf buds. Overexpression of AkFT genes though heterologous expression in Arabidopsis showed that the transgenics flowered at a very early stage relative to wild type plants. These findings indicate that AkFT1 and AkFT2 function as regulation genes in A. konjac flowering development and the two genes may present similar functions during flowering transition. Abstract: We propose an asymmetric AlGaAs/GaAs double quantum wells (QWs) structure for realizing the enhancement of self-Kerr nonlinearity. It is found, with resonant tunneling, that the self-Kerr nonlinearity can be clearly enhanced, while the absorption of probe field is very small and can be safely neglected.
