The hydraulic fracturing in-situ stress testing technology was used to test two boreholes (500-meter and 520-meter deep) at the Taiyuan pumped storage power station in Shanxi Province. The in-situ stress state of critical areas was obtained, and the ground stress level, underground building layout, and lining form in the project area were analyzed. The results show that the maximum horizontal principal stress ranges from 10.98 to 18.09 MPa, the minimum horizontal principal stress from 6.79 to 11.32 MPa, and the vertical principal stress from 9.61 to 13.57 MPa. Compared with the high and low in-situ stress values at the north and south ends of Shanxi Province, respectively, the measured values are between; Compared with the simulated in-situ stress field in the Qinshui Basin, the test results are basically consistent. The vertical stress values are between the maximum horizontal principal stress values and the minimum horizontal principal stress values (SH>Sv>Sh), which means the maximum horizontal stress at the measuring point is the maximum principal stress and is in the strike-slip stress state. Its lateral pressure coefficient Kav is between 0.92 and 1.09, reflecting that the tectonic action in the engineering area is not intense. In the range of 330–506 meters, the saturated uniaxial compressive strength of the two boreholes is betwwen 35 and 107 MPa, with an average of 63.79 MPa, and the ratio of the saturated strength to the maximum principal stress (\begin{document}$ {{R}_{\mathrm{b}}} $\end{document}/σm) is between 3.54 and 5.81, belonging to the medium–high stress level. The direction of the maximum horizontal principal stress in the project area is NE 43° to NE 70.5°, and the average is NE 59.5°, consistent with the regional focal mechanism solution and GPS displacement data. From the perspective of in-situ stress orientation, the average direction of the maximum principal stress in the engineering area is NE 59.5°, and the direction of the long axis of the underground powerhouse is between NE 29.5° and NE 89.5°, which is conducive to the stability of the surrounding rock of the powerhouse. Krüppel‐like factor 7 is a transcriptional activator and acts as an oncogene in human cancers, including hepatocellular carcinoma (HCC). Tryptophan metabolism is important for HCC cell proliferation, metastasis, and invasion. It is unclear whether KLF7 could regulate Trp metabolism in HCC. In this study, we found that Trp metabolism was suppressed in HCC cells with KLF7 knockdown. The mRNA and protein levels of SLC1A5, SLC7A5, and TPH1, as well as the content of Trp and serotonin, were reduced after KLF7 knockdown, and were potentiated following KLF7 overexpression. Increasing the content of serotonin could restore the malignancy of tumour cells in vitro and tumour growth in vivo. Conversely, decreasing the content of serotonin suppressed HCC cell proliferation. The binding activity of KLF7 was on the promoter of SLC1A5, and KLF7 positively regulated the expression of SLC1A5. KLF7 contributed to the proliferation and migration of HCC cells by up‐regulation of SLC1A5. Collectively, KLF7 promotes the progression of HCC through regulating Trp metabolism. Although the potent antibacterial ability of radezolid against Staphylococcus aureus has been widely reported worldwide, its antibacterial and anti-biofilm activity against the S. aureus clinical isolates from China remains elusive. In this study, the minimum inhibitory concentration (MIC) of radezolid was determined in S. aureus clinical isolates from China using the agar dilution method, and the relationship between radezolid susceptibility and ST distribution was also investigated. The anti-biofilm activity of radezolid against S. aureus was determined by a crystal violet assay and compared with that of linezolid and contezolid. The quantitative proteomics of S. aureus treated with radezolid was analyzed, and the genetic mutations in radezolid-induced resistant S. aureus were determined by whole-genome sequencing. The dynamic changes in transcriptional expression levels of several biofilm-related genes were analyzed by quantitative RT-PCR. Our data showed that radezolid MIC ranged from ≤0.125 to 0.5 mg/L, which was almost 1/4 × MIC of linezolid against S. aureus, indicating the greater antibacterial activity of radezolid than linezolid. The S. aureus clinical isolates with radezolid MICs of 0.5 mg/L were most widely distributed in ST239 of MRSA and ST7 of MSSA. Moreover, the more robust anti-biofilm activity of radezolid with subinhibitory concentrations (1/8 × MIC and 1/16 × MIC) was demonstrated against S. aureus when compared with that of contezolid and linezolid. Genetic mutations were found in glmS, 23S rRNA, and DUF1542 domain-containing protein in radezolid-induced resistant S. aureus selected by in vitro induction of drug exposure. Quantitative proteomic analysis of S. aureus indicated that the global expression of some biofilm-related and virulence-related proteins was downregulated. Quantitative RT-PCR further confirmed that the expressions of some downregulated biofilm-related proteins, including sdrD, carA, sraP, hlgC, sasG, spa, sspP, fnbA, and oatA, were decreased after 12 h and 24 h of exposure to radezolid. Conclusively, radezolid shows robust antibacterial and anti-biofilm activity against S. aureus clinical isolates from China when compared with contezolid and linezolid.

Background: Rosacea is a chronic inflammatory dermatological condition in humans, and its pathogenesis remains unclear. However, the development of rosacea is suspected to be related to Demodex, a microscopic commensal organism that resides in or near hair follicles and sebaceous glands. Although Demodex is known to be a host-specific, obligate commensal organism, it is currently difficult to be cultured in vitro to parasitize and infect other animal hosts. Therefore, direct evidence for a pathogenic role of Demodex in rosacea is currently lacking. Summary: As circumstantial evidence, non-invasive skin-detecting techniques have shown abnormally elevated numbers of Demodex in rosacea patients. Increased cytokine levels such as IL-10, IL-8, and IL-12p70 have been observed in human sebocytes following the Demodex challenge, and acaricides have been found to be effective in rosacea therapy, all point to a close relationship between Demodex and rosacea. Based on these findings, we conducted a comprehensive literature review to summarize the current state of knowledge, research insights, and clinical treatment recommendations for Demodex-associated rosacea, with the ultimate goal of improving patient outcomes.
