Abstract Background Neither a TYRP1-mediated highly conserved genetic network underlying skin color towards optimum defense nor the pathological tendency of its mutation is well understood. The Oujiang Color Common Carp (Cyprinus carpio var. color) as a model organism, offering valuable insights into genetics, coloration, aquaculture practices, and environmental health. Here, we performed a comparative skin transcriptome analysis on TYRP1 mutant and wild fishes by applying a conservative categorical approach considering different color phenotypes. Results Our results reveal that an unusual color phenotype may be sensitized with TYRP1 mutation as a result of upregulating several genes related to an anti-inflammatory autoimmune system in response to the COMT-mediated catecholamine neurotransmitters in the skin. Particularly, catecholamines-derived red/brown, red with blue colored membrane attack complex, and brown/grey colored reduced eumelanin are expected to be aggregated in the regenerated cells. Conclusions It is, thus, concluded that the regenerated cells with catecholamines, membrane attack complex, and eumelanin altogether may contribute to the formation of the unusual (coffee-like) color phenotype in TYRP1 mutant.

Depression, a global health problem with growing prevalence, brings serious impacts on the daily life of patients. However, the antidepressants currently used in clinical are not perfectly effective, which greatly reduces the compliance of patients. Berberine is a natural quaternary alkaloid which has been shown to have a variety of pharmacological effects, such as hypoglycemic, lipid-regulation, anticancer, antibacterial, anti-oxidation, anti-inflammatory, and antidepressant. This review summarizes the evidence of pharmacological applications of berberine in treating depression and elucidates the mechanisms of berberine regulating neurotransmitter levels, promoting the regeneration of hippocampal neurons, improving hypothalamic-pituitary-adrenal axis dysfunction, anti-oxidative stress, and suppressing inflammatory status in order to provide a reference for further research and clinical application of berberine. Nanoparticles with small diameters and large surface areas have potential advantages and are actively utilized in various fields related to biomedical and catalytic applications. Multifunctional applications can be achieved by endowing nanoparticles with piezoelectric, quantum dot, magnetothermal, and piezoluminescent properties. In particular, multiferroic magnetoelectric nanoparticles (MENPs) can generate electricity by coupling piezoelectric and magnetostrictive properties when an external magnetic field, which is harmless to the human body, is applied. In this regard, magnetoelectricity (ME) induced by a magnetic field makes MENPs useful for various biomedical and electrocatalytic applications. The ME voltage coefficients, which express the efficiency of energy conversion from magnetic field to electricity, show differences depending on the setup for ME measurements of MENPs. Therefore, numerous attempts have been made to optimize the ME characterization method to reduce measurement errors resulting from charge leakages caused by the specimen preparation, as well as to investigate the ME effect of a single nanoparticle. Our review is focused on the structures, syntheses (hydrothermal and sol–gel methods), activation mechanism, and measurement of magnetoelectricity, as well as applications, of core–shell MENPs. The biological process of healing of wounds deals with the regeneration of cells and is of utmost importance mainly because of the morphological and functional attributes of the tissue. One of the modern approaches is to use novel nanomaterial based wound dressings and other therapeutic treatments that have higher efficiency and specificity. Researchers all around the world have established the competence of various organic, inorganic or polymer-based and biogenically synthesized nanomaterials as a weapon for wound curing. Various matrices carrying nanoparticles like hydrogels, hydrocolloids are used in nano based drug delivery system. The biocompatibility, antimicrobial efficacy and sustained rate of drug delivery are the three main features that are considered for designing a nano drug for effective wound healing. The modification of the nanosized liposome using propylene glycol increases cell-permeation and retention in skin layer. Curcumin loaded liposomes show high anti-inflammatory effect, carbon nanodots (CNDs) and graphene oxide based nanosheets have also shown an immense importance in possessing high surface area. They also show potency in promoting fibroblast growth factors and collagen deposition for speedy recovery of wound. Moreover, the chronic wounds, that could not be cured by common antibiotics, can be addressed by nano based drugs not only for their easy penetration to deeper layer of tissues but also for their efficiency in killing some biofilm associated bacterial strains. Hence, nowadays nano based dressing materials are widely used for rapid recovery of wounds of various types.

Abstract Background The study was conducted to assess the characterization, differentiation, and in vitro cell regeneration potential of canine mesenteric white adipose tissue-derived mesenchymal stem cells (AD-MSCs). The tissue was harvested through surgical incision and digested with collagenase to obtain a stromal vascular fraction. Mesenchymal stem cells isolated from the stromal vascular fraction were characterized through flow cytometry and reverse transcription-polymerase chain reaction. Assessment of cell viability, in vitro cell regeneration, and cell senescence were carried out through MTT assay, wound healing assay, and β-galactosidase assay, respectively. To ascertain the trilineage differentiation potential, MSCs were stained with alizarin red for osteocytes, alcian blue for chondrocytes, and oil o red for adipocytes. In addition, differentiated cells were characterized through a reverse transcription-polymerase chain reaction. Results We observed the elongated, spindle-shaped, and fibroblast-like appearance of cells after 72 h of initial culture. Flow cytometry results showed positive expression for CD44, CD90, and negative expression for CD45 surface markers. Population doubling time was found 18–24 h for up to the fourth passage and 30±0.5 h for the fifth passage. A wound-healing assay was used to determine cell migration rate which was found 136.9 ± 4.7 μm/h. We observed long-term in vitro cell proliferation resulted in MSC senescence. Furthermore, we also found that the isolated cells were capable of differentiating into osteogenic, chondrogenic, and adipogenic lineages. Conclusions Mesenteric white adipose tissue was found to be a potential source for isolation, characterization, and differentiation of MSCs. This study might be helpful for resolving the problems regarding the paucity of information concerning the basic biology of stem cells. The large-scale use of AD-MSCs might be a remedial measure in regenerative medicine.

Damage to proximal tubules due to exposure to toxicants can lead to conditions such as acute kidney injury (AKI), chronic kidney disease (CKD) and ultimately end‐stage renal failure (ESRF). Studies have shown that kidney proximal epithelial cells can regenerate particularly after acute injury. In the previous study, we utilized an immortalized in vitro model of human renal proximal tubule epithelial cells, RPTEC/TERT1, to isolate HRTPT cell line that co‐expresses stem cell markers CD133 and CD24, and HREC24T cell line that expresses only CD24. HRTPT cells showed most of the key characteristics of stem/progenitor cells; however, HREC24T cells did not show any of these characteristics. The goal of this study was to further characterize and understand the global gene expression differences, upregulated pathways and gene interaction using scRNA‐seq in HRTPT cells. Affymetrix microarray analysis identified common gene sets and pathways specific to HRTPT and HREC24T cells analysed using DAVID, Reactome and Ingenuity software. Gene sets of HRTPT cells, in comparison with publicly available data set for CD133+ infant kidney, urine‐derived renal progenitor cells and human kidney‐derived epithelial proximal tubule cells showed substantial similarity in organization and interactions of the apical membrane. Single‐cell analysis of HRTPT cells identified unique gene clusters associated with CD133 and the 92 common gene sets from three data sets. In conclusion, the gene expression analysis identified a unique gene set for HRTPT cells and narrowed the co‐expressed gene set compared with other human renal–derived cell lines expressing CD133, which may provide deeper understanding in their role as progenitor/stem cells that participate in renal repair. Sensory hair cells (HCs) are highly susceptible to damage by noise, ototoxic drugs, and aging. Although HCs cannot be spontaneously regenerated in adult mammals, previous studies have shown that signaling pathways are involved in HC regeneration in the damaged mouse cochlea. Here, we used a Notch antagonist (DAPT), a Wnt agonist (QS11), and recombinant Sonic hedgehog (SHH) protein to investigate their concerted actions underlying HC regeneration in the mouse cochlea after neomycin-induced damage both in vivo and in vitro. With DAPT, the numbers of HCs increased, and supporting cell (SC) proliferation was seen in both the intact and damaged cochlear sensory epithelia, while these numbers were unchanged in the presence of QS11. When simultaneously treated with DAPT and QS11, the number of HCs increased dramatically, and much greater SC proliferation was seen in the cochlear epithelium. In transgenic mice with both Notch1 conditional knockout and β-catenin over-expression, cochlear SC proliferation and HC regeneration were more obvious than in either Notch1 knockout or β-catenin over-expressing mice separately. When cochleae were treated with DAPT, QS11, and SHH together, SC proliferation was even greater, and this proliferation was seen in both the HC region and the greater epithelial ridge. High-throughput RNA sequencing was used to identify the differentially expressed genes between all groups, and the results showed that the SHH and Wnt signaling pathways are involved in SC proliferation. Our study suggests that co-regulation of the Notch, Wnt, and SHH signaling pathways promotes extensive cell proliferation and regeneration in the mouse cochlea. [ABSTRACT FROM AUTHOR]

[Elaeagnaceae]) plant parts, especially berries, are well characterized and repeatedly tested for antioxidant activity and regenerative properties, in various cell types and tissues. However, fatty acids (FA) have been less investigated in term of biological effects, although, they are important bioactive components of the sea buckthorn fruit and oil. The aim of our work was to determine whether sea buckthorn seed oil is a suitable source of FA with regenerative properties on normal skin cells. Using high-performance liquid chromatography (HPLC) and liquid chromatography – mass spectrometry (LC-MS), we purified and characterized four fractions enriched in saturated (palmitic) and non-saturated (linoleic, alfa-linolenic, oleic) FA, which were tested for cytotoxicity, cytokine and growth factor production, and regenerative effect on normal keratinocytes and skin fibroblasts. Evidence is presented that the palmitic acid enriched fraction was a suitable sea buckthorn seed oil derived product with cell proliferation properties on both skin cell types.

With the globally increasing prevalence, cardiovascular diseases (CVDs) have become the leading cause of mortality. The transplantation of endothelial progenitor cells (EPCs) holds a great promise due to their potential for vasculogenesis, angiogenesis, and protective cytokine release, whose mechanisms are essential for CVD therapies. In reality, many investigations have attributed the therapeutic effects of EPC transplantation to the secretion of paracrine factors rather than the differentiation function. Of note, previous studies have suggested that EPCs could also release exosomes (diameter range of 30–150 nm), which carry various lipids and proteins and are abundant in microRNAs. The EPC-derived exosomes (EPC-EXs) were reported to act on the heart and blood vessels and were implicated in anti-inflammation, anti-oxidation, anti-apoptosis, the inhibition of endothelial-to-mesenchymal transition (EndMT), and cardiac fibrosis, as well as anti-vascular remodeling and angiogenesis, which were considered as protective effects against CVDs. In this review, we summarize the current knowledge on using EPC-EXs as therapeutic agents and provide a detailed description of their identified mechanisms of action to promote the prognosis of CVDs.

Background: Mesenchymal stem cell-based therapy is known to have the potential to induce angiogenesis. However, there are still some limitations regarding their clinical application. Photomodulation/photobiomodulation is non-invasive and non-toxic phototherapy able to stimulate cell viability, proliferation, differentiation, and migration, when the right irradiation parameters are applied. A review of the published articles on human conditioned-by-photobiomodulation mesenchymal cells in an in vitro set up was carried out. Our aim was to describe the studies' results and identify any possible tendency that might highlight the most suitable procedures. Methods: A search in English of the PubMed database was carried out with the search criteria: photobiomodulation or photoactivation or photomodulation, and mesenchymal cells. All irradiations applied in vitro, on human mesenchymal cells, with wavelengths ranged from 600 to 1000 nm. Results: The search yielded 42 original articles and five reviews. Finally, 37 articles were selected with a total of 43 procedures. Three procedures (7.0%) from 620 to 625 nm; 26 procedures (60.5%) from 625 to 740 nm; 13 procedures (30.2%) from 740 to 1000 nm; and one procedure (2.3%) with combinations of wavelengths. Of the 43 procedures, 14 assessed cell viability (n = 14/43, 32.6%); 34 cell proliferation (n = 34/43, 79.1%); 19 cell differentiation (n = 19/43, 44.2%); and three cell migration (n = 3/43, 7.0%). Conclusions: Photobiomodulation is a promising technology that can impact on cell viability, differentiation, proliferation, or migration, leading to enhance its regenerative capacity. Currently, the predominant method for the industrial production of 1,3-dihydroxyacetone (DHA) from glycerol involves fed-batch fermentation. However, previous research has revealed that in the biocatalytic synthesis of DHA from glycerol, when the DHA concentration exceeded 50 g·L−1, it significantly inhibited microbial growth and metabolism, posing a challenge in maintaining prolonged and efficient catalytic production of DHA. In this study, a new integrated continuous production and synchronous separation (ICSS) system was constructed using hollow fiber columns and perfusion culture technology. Additionally, a cell reactivation technique was implemented to extend the biocatalytic ability of cells. Compared with fed-batch fermentation, the ICSS system operated for 360 h, yielding a total DHA of 1237.8 ± 15.8 g. The glycerol conversion rate reached 97.7 %, with a productivity of 3.44 g·L−1·h−1, representing 485.0 % increase in DHA production. Objectives Recurrent urinary tract infection (rUTI) is a common infectious disease in women. This study investigated the urothelial cell proliferation, the cytoskeleton, barrier proteins, and inflammatory protein expression in women with rUTIs. Methods: Female patients with recurrent or persistent UTIs were recruited. Bladder mucosal specimens were investigated by Western blot and immunohistochemical staining for the urothelial cytoskeleton proteins cytokeratin 5 (CK5), CK14, and CK20; proteins involved in cellular proliferation, including CD34, sonic hedgehog (SHH), and tumor protein 63 (TP63); barrier proteins zonula occludens 1 (ZO‐1) and E‐cadherin; inflammatory proteins p38 and tryptase; and proapoptotic proteins Bcl2‐associated agonist of cell death protein (BAD), Bcl2‐associated X protein (BAX), and caspase‐3. Women with stress urinary incontinence without bladder symptoms served as controls. Bladder specimens from 18 recurrent UTI patients with rUTIs and 12 persistent UTIs, and 17 controls were analyzed, and protein expressions were compared between the three groups. Results: Cell proliferation protein expression for CD34, SHH, and TP63 was significantly lower in the urothelium of patients with rUTIs than in controls. Expression of CK5 increased, whereas CK20 decreased significantly in rUTIs compared with those of controls. Apoptotic proteins BAD, BAX, and caspase‐3 were significantly higher in patients with rUTIs. However, barrier proteins ZO‐1 and E‐cadherin, and tryptase were not significantly lower in patients with rUTIs. Conclusion: Deficits in expression of proteins involved in urothelial cell proliferation, cytoskeleton, and barrier function were noted in patients with rUTIs. Hypoxia is of great significance for stem cells to maintain the proliferation and differentiation capacity. As a specialized mesenchymal component of the hair follicle (HF), the dermal papilla cell (DPC) not only regulates HF cycle, but also plays a pivotal role in differentiating hair follicle stem cell(HFSC) into HF. However, whether hypoxia could affect DPCs on proliferation or metabolism remains unclear. In our study, DPCs were cultured in normoxia (20%O2) or hypoxia (5%O2). Cell viability assays were performed, and lactate dehydrogenase (LDH) activity and lactate level in DPCs were detected. After that, LDH was overexpressed or knocked down in DPCs; then, the expression of protein markers (ALP, Ki‐67) was assessed by Western blotting, and cell proliferation was also detected after overexpression or knockdown of LDH. Hypoxia did show positive effect on proliferation of DPCs. The LDH activity of DPCs cultured under hypoxic condition was significantly higher than that of cultured under normoxic condition. Overexpression of LDH significantly up‐regulates the expression of ALP and Ki‐67 compared with knockdown and negative control. Cell proliferation was also promoted in DPCs with elevated LDH. Our findings showed that the proliferation activity of DPCs could be stimulated under hypoxia. Meanwhile, LDH plays an important role in maintaining the activity of DPCs in hypoxic condition. Polyhydroxylated derivatives of 6-keto,7-dehydrocholesterol (ecdysteroids) are common constituents of various plants. In 1965, they were accidentally discovered in the search for the insect moulting hormone. These biologically important natural compounds are neither insect hormones nor inducers of insect ecdysis. Due to their strong anabolic, vitamin D-like effects in insects, domestic animals and humans, I propose the use of the arbitrary term vitamin D1. The present paper describes the effects of vitamin D1 on the growth and regeneration of excised epidermal cells of the tobacco hornworm, Manduca sexta (Sphingidae). The periods of programmed cell death and cell proliferation (histolysis and histogenesis, respectively) exactly coincide in insects with endogenous peaks of increased concentration of vitamin D1. Epidermal cells communicate with each other, creating a mutually integrated tissue, connected by mechanical, chemical, electrical, ionic or other so far incompletely known factors. After natural cell death, or after the artificial removal of some epidermal cells, the neighbouring cells that lose communication integrity, begin to divide mitotically to replace the disconnected part. Cell divisions are arrested as soon as the integrity of the living tissue is established. During insect ontogeny, the application of juvenile hormone causes regenerating epidermal cells to repeat the previous morphogenetic programme (i.e., development of patches of larval tissue on the body of a pupa, or metathetely). Conversely, the application of vitamin D1 (20-hydroxyecdysone) caused the regenerating cells to prematurely execute a future morphogenetic programme (i.e., development of patches of pupal tissue on the body of a larva, or prothetely). Among the key features of insect regeneration, is the arrest of cell divisions when tissues resume living cell-to-cell integrity. This prevents the formation of aberrant groups of cells, or tumours. It is well established that the main physiological systems of insects (e.g., circulatory, respiratory, neuro-endocrine) are structurally and functionally similar to corresponding systems in humans. Thus the basic principles of cell regeneration and the role of vitamin D1 in insects may also be valid for humans. The common vitamins D2 (ergocalciferol) or D3 (cholecalciferol), are exclusively lipid soluble secosterols, which require activation by UV irradiation and hydroxylation in the liver. By contrast, the neglected vitamin D1 is a natural derivative of polyhydroxylated 7-dehydrocholesterol of predominantly plant origin, which is both partly a water and partly a lipid soluble vitamin. It neither requires UV irradiation, nor hydroxylation due to 6 or 7 already built-in hydroxylic groups. Like other vitamins, it enters insect or human bodies in plant food or is produced by intestinal symbionts. Vitamin D1 causes strong anabolic, vitamin D-like effects in domestic animals and in humans. I am convinced that avitaminosis associated with a deficiency of vitamin D1 in human blood may be responsible for certain hitherto incurable human diseases, especially those related to impaired nerve functions and somatic growth, aberrant cell regeneration

Introduction: MicroRNA (miRNA) is known to be involved in nerve injury. Our study aimed to identify the role and mechanism of miR-1b in rat neuron proliferation, regeneration and apoptosis. Material and methods: Neurons were successfully separated and identified using a microscope and immunofluorescence staining of microtubule-associated protein 2 (MAP-2). The expressions of miR-1b and Krüppel-like factor 7 (KLF7) were detected by quantitative real-time polymerase chain reaction (qRT-PCR). Neuron viability and apoptosis were detected by MTT assay and flow cytometry, respectively. Neuron regeneration states were observed using a microscope and analysed by the ImageJ software. Expressions of C-caspase-3 and cell regeneration-related proteins (nerve growth factor [NGF], ciliary neurotrophic factor [CNTF] and brain-derived neurotrophic factor [BDNF]) were measured by Western blot. Target genes and potential binding sites of KLF7 and miR-1b were predicted by TargetScan 7.2 and confirmed by dual luciferase reporter assay. Results: Neurons were identified as MAP-2-positive. Up-regulation of miR-1b reduced neuron viability and regenerative ability, promoted neuron apoptosis and C-caspase-3 expression, and down-regulated the expressions of cell regeneration-related proteins. KLF7 was the target gene of miR-1b. Overexpressed KLF7 rescued the effects of up-regulation of miR-1b on neuron viability, regeneration and apoptosis. Expressions of NGF, CNTF and BDNF were suppressed yet C-caspase-3 expression was up-regulated by miR-1b mimic, which was partially rescued by overexpressed KLF7. Conclusions: Up-regulation of miR-1b promoted rat neuron proliferation and regeneration yet inhibited apoptosis via targeting KLF7. [ABSTRACT FROM AUTHOR]

Objectives: To investigate the role of dibenzazepine (DBZ) in promoting supporting cell (SC) proliferation and hair cell (HC) regeneration in the inner ear. Materials and Methods: Postnatal day 1 wild‐type or neomycin‐damaged mouse cochleae were cultured with DBZ. Immunohistochemistry and scanning electron microscopy were used to examine the morphology of cochlear cells, and high‐throughput RNA‐sequencing was used to measure gene expression levels. Results: We found that DBZ promoted SC proliferation and HC regeneration in a dose‐dependent manner in both normal and damaged cochleae. In addition, most of the newly regenerated HCs induced by DBZ had visible and relatively mature stereocilia bundle structures. Finally, RNA sequencing detected the differentially expressed genes between DBZ treatment and controls, and interaction networks were constructed for the most highly differentially expressed genes. Conclusions: Our study demonstrates that DBZ can significantly promote SC proliferation and increase the number of mitotically regenerated HCs with relatively mature stereocilia bundles in the neonatal mouse cochlea by inhibiting Notch signalling and activating Wnt signalling, suggesting the DBZ might be a new therapeutic target for stimulating HC regeneration. BACKGROUND: Exosomes are vesicles with phospholipid bilayers containing lots of proteins, lipid and RNA secreted by various types of cells, which might play important roles in cell-to-cell communication and signal transduction and participate in the regulation of important physiology or pathological processes. Recent studies have discovered that exosomes derived from mesenchymal stem cells, a promising treatment for acute lung injury, exert an anti-inflammation effect and improve the microvascular endothelial barrier permeability and enhance lung epithelial tissue repair. OBJECTIVE: To review the recent advances in mesenchymal stem cells derived exosomes as a novel approach to the treatment of acute lung injury. METHODS: Databases of PubMed and CNKI were searched for the articles related to the mechanism of mesenchymal stem cells-derived exosomes in the treatment of acute lung injury between 1981 and 2018. The keywords were “mesenchymal stem cells, exosomes, acute lung injury” in English and Chinese, respectively. Fifty-eight articles were enrolled for analysis. RESULTS AND CONCLUSION: (1) Exosomes plays an important role in mediating intercellular communication and regulating immune system by transferring bioactive proteins, lipids and nucleic acids to target cells. (2) Mesenchymal stem cells-derived exosomes carry certain proteins, lipids, nucleic acids from mesenchymal stem cells. They might contribute to attenuating the inflammatory response and apoptosis, and promoting cell regeneration. (3) The existing researches summarize the main mechanism of treatment of acute lung injury by mesenchymal stem cell-derived exosomes, and provide reference for the future application of mesenchymal stem cell-derived exosomes in the treatment of acute lung injury. Ideal heart valve solutions aim to provide thrombosis-free durability. A scaffold-based polycarbonate urethane urea tissue-engineered heart valve designed to mimic native valve microstructure and function was used. This study examined the acute in vivo function of a stented tissue-engineered heart valve in a porcine model. Trileaflet valves were fabricated by electrospinning polycarbonate urethane urea using double component fiber deposition. The tissue-engineered heart valve was mounted on an AZ31 magnesium alloy biodegradable stent frame. Five 80-kg Yorkshire pigs underwent open tissue-engineered heart valve implantation on cardiopulmonary bypass in the pulmonary position. Tissue-engineered heart valve function was echocardiographically evaluated immediately postimplant and at planned study end points at 1, 4, 8, and 12 hours. Explanted valves underwent biaxial mechanical testing and scanning electron microscopy for ultrastructural analysis and thrombosis detection. All 5 animals underwent successful valve implantation. All were weaned from cardiopulmonary bypass, closed, and recovered until harvest study end point except 1 animal that was found to have congenital tricuspid valve dysplasia and that was euthanized postimplant. All 5 cases revealed postcardiopulmonary bypass normal leaflet function, no regurgitation, and an average peak velocity of 2 m/s, unchanged at end point. All tissue-engineered heart valve leaflets retained microstructural architecture with no platelet activation or thrombosis by scanning electron microscopy. There was microscopic evidence of fibrin deposition on 2 of 5 stent frames, not on the tissue-engineered heart valve. Biaxial stress examination revealed retained postimplant mechanics of tissue-engineered heart valve fibers without functional or ultrastructural degradation. A biodegradable elastomeric heart valve scaffold for in situ tissue-engineered leaflet replacement is acutely functional and devoid of leaflet microthrombosis. The complete polycarbonate urethane urea (PCUU) valve on a magnesium (Mg) stent before implant is shown both after implantation, with related echocardiography revealing no regurgitation, and after explant, revealing no evidence of microthrombosis on scanning electron micrograph. Highlights • Clinical researchers have found that coinfection of the respiratory tract can cause distinct disease outcome, sometimes leading to long-lasting infection, compared to single viral infection. • The impact of coinfections in human respiratory tract have not yet been evaluated in either theoretical or experimental studies on a large scale. • A few experiments confirm that different respiratory viruses can infect the same cell (superinfection). • Superinfection alone cannot cause long-lasting viral coinfections. • The combined mechanism of superinfection and cell regeneration provides a plausible mechanism for chronic viral coinfections. Abstract Molecular diagnostic techniques have revealed that approximately 43% of the patients hospitalized with influenza-like illness are infected by more than one viral pathogen, sometimes leading to long-lasting infections. It is not clear how the heterologous viruses interact within the respiratory tract of the infected host to lengthen the duration of what are usually short, self-limiting infections. We develop a mathematical model which allows for single cells to be infected simultaneously with two different respiratory viruses (superinfection) to investigate the possibility of chronic coinfections. We find that a model with superinfection and cell regeneration has a stable chronic coinfection fixed point, while superinfection without cell regeneration produces only acute infections. This analysis suggests that both superinfection and cell regeneration are required to sustain chronic coinfection via this mechanism since coinfection is maintained by superinfected cells that allow slow-growing infections a chance to infect cells and continue replicating. Tissue engineering is known to encompass multiple aspects of science, medicine and engineering. The development of systems which are able to promote the growth of new cells and tissue components are vital in the treatment of severe tissue injury and damage. This can be done through a variety of different biofabrication strategies including the use of hydrogels, 3D bioprinted scaffolds and nanotechnology. The incorporation of stem cells into these systems and the advantage of this is also discussed. Biopolymers, those which have a natural original, have been particularly advantageous in tissue engineering systems as they are often found within the extracellular matrix of the human body. The utilization of biopolymers has become increasing popular as they are biocompatible, biodegradable and do not illicit an immune response when placed into the body. Tissue engineering systems for use with the eye are also discussed. This is of particular interest as the eye is known as an immune privileged site resulting in an extremely limited ability for natural cell regeneration.

Depression, a global health problem with growing prevalence, brings serious impacts on the daily life of patients. However, the antidepressants currently used in clinical are not perfectly effective, which greatly reduces the compliance of patients. Berberine is a natural quaternary alkaloid which has been shown to have a variety of pharmacological effects, such as hypoglycemic, lipid-regulation, anti-cancer, antibacterial, anti-oxidation, anti-inflammatory, and antidepressant. The COVID-19 pandemic generated interest in the medicinal applications of messenger RNA (mRNA). It is expected that mRNA will be applied, not only to vaccines, but also to regenerative medicine. The purity of mRNA is important for its medicinal applications. However, the current mRNA synthesis techniques exhibit problems, including the contamination of undesired 5'-uncapped mRNA and double-stranded RNA. Recently, our group developed a completely capped mRNA synthesis technology that contributes to the progress of mRNA research. The introduction of chemically modified nucleosides, such as N1-methylpseudouridine and 5-methylcytidine, has been reported by Karikó and Weissman, opening a path for the practical application of mRNA for vaccines and regenerative medicine. Yamanaka reported the production of induced pluripotent stem cells (iPSCs) by introducing four types of genes using a retrovirus vector. iPSCs are widely used for research on regenerative medicine and the preparation of disease models to screen new drug candidates. Among the Yamanaka factors, Klf4 and c-Myc are oncogenes, and there is a risk of tumor development if these are integrated into genomic DNA. Therefore, regenerative medicine using mRNA, which poses no risk of genome insertion, has attracted attention. In this review, the author summarizes techniques for synthesizing mRNA and its application in regenerative medicine.

Increasing evidences have demonstrated that the invertebrate gill is a predominant tissue participating in the immune response during pathogen challenge. In the present study, the hematopoiesis and immune activities in gill of Pacific oyster Crassostrea gigas were investigated. Stem-like cells with big nuclei and thin cytoplasm were found in the tubules of gill filaments, where DNA synthesis is active and hemocytes production are exuberant. The oysters primarily stimulated by formaldehyde-killed Vibrio splendidus exhibited stronger immune responses and enhanced cell regeneration in gill when they encountered the secondary challenge of live V. splendidus . After the secondary stimulation with V. splendidus , the expression levels of Cg Clec-4 and Cg IFN in the gill of oysters pre-stimulated with formaldehyde-killed V. splendidus were significantly higher ( p < 0.05) than that in the oysters pre-stimulated with filter-sterilized (0.22 μm pore size) sea water, while the expression level of Cg IL-17 was significantly decreased ( p < 0.05). Meanwhile, the protein expression level of hematopoietic transcription factor Cg GATA3 and immune-related protein Cg EcSOD in gill increased apparently after the secondary challenge with V. splendidus . ROS production was also enhanced ( p < 0.05) at 6 h and 24 h after the secondary challenge. The phagocytic rate in gill of oysters pre-stimulated with formaldehyde-killed V. splendidus was significantly increased ( p < 0.05) at 6 h after the secondary challenge with live V. splendidus , showing faster response than that pre-stimulated with filter-sterilized sea water. These results collectively showed that the immune parameters in gill were apparently enhanced after secondary challenge with live V. splendidus , indicating that hematopoiesis might participate in immune priming in Pacific oyster C. gigas . The anterior lobe of the pituitary gland is a master endocrine tissue composed of five types of endocrine cells. Although the turnover rate of pituitary endocrine cells is as low as about 1.6% per day, recent studies have demonstrated that Sex-determining region Y-box 2 (SOX2)+-cells exist as pituitary stem/progenitor cells in the adult anterior lobe and contribute to cell regeneration. Notably, SOX2+-pituitary stem/progenitor cells form two types of niches in this tissue: the marginal cell layer (MCL-niche) and the dense cell clusters scattering in the parenchyma (parenchymal-niche). However, little is known about the mechanisms and factors for regulating the pituitary stem/progenitor cell niches, as well as the functional differences between the two types of niches. Elucidation of the regulatory mechanisms in the niches might enable us to understand the cell regeneration system that acts in accordance with physiological demands in the adult pituitary. In this review, so as to reveal the regulatory mechanisms of the two types of niche, we summarize the regulatory factors and their roles in the adult rodent pituitary niches by focusing on three components: soluble factors, cell surface proteins and extracellular matrixes. This study proposes a novel electrolytic method for simultaneous removal of trace estrogens and regeneration of electrolytic cells for long-term wastewater treatment. Continuous treatments of estrogens estrone (E1), 17β-estradiol (E2) and 17α-ethinyl estradiol (EE2) were theoretically and experimentally studied using an electrolytic reactor equipped with a multi-packed granular glassy carbon electrode reactor. Experimental results demonstrated that E1, E2 and EE2 were effectively removed through electro-polymerization on the granular glassy carbon (and Pt/Ti) anode counter. Polymer formed during continuous treatment was quickly decomposed and electrodes were regenerated completely by OH radicals produced through the reduction of ozone. Calculated overall energy consumptions were less than 10 Wh/m 3 , demonstrating extremely low energy consumptions. In addition, a mathematical model developed based on the limiting mass transfer rate and post-regeneration could represent general trends in time series data observed in experiments. The development of therapeutic treatments to regenerate urothelium, manufacture tissue equivalents or neourethras for in-vivo application is a significant challenge in the field of tissue engineering. Many studies have focused on urethral defects that, in most cases, inadequately address current therapies. This article reviews the primary tissue engineering strategies aimed at the clinical requirements for urothelium regeneration while concentrating on promising investigations in the use of grafts, cellular preparations, as well as seeded or unseeded natural and synthetic materials. Despite significant progress being made in the development of scaffolds and matrices, buccal mucosa transplants have not been replaced. Recently, graft tissues appear to have an advantage over the use of matrices. These therapies depend on cell isolation and propagation in vitro that require, not only substantial laboratory resources, but also subsequent surgical implant procedures. The choice of the correct cell source is crucial when determining an in-vivo application because of the risks of tissue changes and abnormalities that may result in donor site morbidity. Addressing an appropriately-designed animal model and relevant regulatory issues is of fundamental importance for the principal investigators when a therapy using cellular components has been developed for clinical use. Purpose of review Cell based muscle tissue engineering carries the potential to revert the functional loss of muscle tissue caused by disease and trauma. Although muscle tissue can be bioengineered using various precursor cells, major limitations still remain. Recent findings In the last decades several cellular pathways playing a crucial role in muscle tissue regeneration have been described. These pathways can be influenced by external stimuli and they not only orchestrate the regenerative process after physiologic wear and muscle trauma, but also play an important part in aging and maintaining the stem cell niche, which is required to maintain long-term muscle function.
