- Abstract
Cubiu is a vegetable of Solanaceae family, native to the Amazon, which is widely distributed through Brazil, Peru, and Colombia. It is used in food, medicine, and cosmetics by native populations. Research has shown that cubiu extracts have antioxidant activities with great biological relevance. We performed a phytochemical screening to identify the main chemical groups that could confer antioxidant activity to this extract. Several tests and qualitative precipitation specific staining for major classes of secondary metabolites were used. Antioxidant capacity in vitro tests (DPPH and ABTS) were also used to assess the extract’s ability to sequester free radicals of 70% hydroethanolic and aqueous extracts of cubiu flour. Alkaloids, organic acids, phenols, flavonoid glycosides, and coumarins were found in the hydroethanolic extract while the aqueous extract presented anthocyanins, gums, tannins and mucilage, amino groups, and volatile and fixed acids. For in vitro tests, the IC
50 value obtained in the DPPH assay was 606.3 ± 3.5 μg/mL while that for the ABTS assay was 290.3 ± 10.7 µg/mL. Although cubiu extracts present chemical compounds directly related to antioxidant activity, our results show that it has a low antioxidant activity. Additional studies will be needed to isolate and characterize specific compounds to further assess antioxidant activity. [ABSTRACT FROM AUTHOR]- Published

- Abstract
An in vivo study was conducted to assess the effects of the consumption of Astrocaryum aculeatum Amazon Meyer (tucumã) in the treatment of diet-induced dyslipidemia in sedentary and exercised Wistar rats. With an average weight of 350 grams, 40 male rats were divided into 4 subgroups of 10. The sedentary control group (SCG) was fed with commercial feed, while the sedentary treatment group (STG) was fed with a ration of tucumã. In addition to the sedentary groups, two exercise groups were formed. The Exercised control group (ECG) was fed with commercial food and the exercised treatment group (ETG) was fed with a ration of tucumã. Body weight gain and food intake were monitored during the experiment. Plasma was analyzed for cholesterol, triglycerides, HDL-C, LDL-C, VLDL, total protein, glucose, insulin, and leptin concentrations. Our results show that the ECG group tended to consume more food, while the groups that were fed with tucumã pulp (STG and ETG) presented a greater tendency to gain body mass. ECG group showed a tendency towards a higher concentration of cholesterol in plasma, while STG and ETG presented higher absolute values for triglycerides and VLDL. No hypolipiemic effect was observed related to tucuma ingestion. [ABSTRACT FROM AUTHOR]

Endopleura uchi (Huber) Cuatrec is a native Amazonian tree species with proven herbal properties in several works by researchers from the region and around the world. From its shell is isolated a substance called Bergenin, which has among its properties, antioxidant, hepatoprotective and hypolipidemic activities. The aim of this study was to evaluate the protective effect of bergenin and dry extract of endopleura uchi on the treatment of induced NAFLD in mice and obesity. NAFLD and obesity were induced in male C57BL / 6J mice by Hypercaloryc and Hyperlipid Feed (HF). Sixty male C57BL / 6J mice were divided into 10 groups of six animals each, with the following characteristics: GM1, Negative Control; GM2, Sham Group: Positive Control; GM3, fed HF for eight weeks and then treated with dry Endopleura uchi extract, 200mg/kg/day for seven weeks; GM4, similar to the above, treated with 100mg/kg/day of E. uchi extract; GM5, HF-fed for seven weeks and treated concomitantly with 200mg/kg/day of Endopleura uchi dry extract; GM6, similar to GM5, treated with 100mg/kg/day to verify a potential prophylactic effect at both doses. Groups GM7 and GM8, fed HF for eight weeks and then treated with 50 and 25mg/kg/day for seven weeks with aqueous Bergenin solution respectively; GM9 and GM10, similar to GM5 and GM6 prophylactic groups and treated with aqueous Bergenin solution at doses of 50 and 25 mg/kg/day, respectively. Biochemical examinations of liver function tests and histopathology were performed to prove the anti-obesity effects and treatment of NAFLD of both solutions. Descriptive data analysis was performed and the results are presented in tables, graphs/figures and descriptive measures, such as mean, standard error, among others. To verify the influence of treatments, ANOVA with post hoc test by Student Newman Keuls was used. Having significant influence of the treatments, the Tukey test was used to verify the differences between the means of the groups. In addition, it is associated with modification of the immune response, leading in increased production of inflammatory mediators, triggering chronic inflammation. Natural products are sources of new antidiabetic and anti-inflammatory drugs, thus being fundamental for the development of prototypes that may serve as a basis for drugs development in the future. The aim of this work was synthesize a new derivative from reaction between naringenin and aminoguanidine and to elucidate its anti-diabetic and anti-inflammatory properties. The condensation reaction between aminoguanidine and naringenin produced a derivative characterized as a hydrazine carboximidamide (DCHA) in a post-reaction yield of 81%. DCHA was chemically characterized by mass spectrometry, NMR and IV. The DCHA didn???t exhibited toxicity in murine fibroblasts up to 50 ug / ml. The DCHA demonstrated antioxidant activity upper to naringenin and satisfactory glycation inhibition in vitro. In silico assays. DCHA showed affinity to iNOS, COX and PDE4, thus suggesting that molecule could exhibit anti-inflammatory and hypoglycemic activities. In the antidiabetic activity tests, DCHA decreased the glycemic levels of diabetic mice after 21 days of oral treatment (30mg / kg), thus wielding a hypoglycemic effect. In the anti-inflammatory activity assays, DCHA showed a significant anti-inflammatory response by suppressing the leukocytes migration into the peritoneal cavity in LPS-mice exposed and treated at a dose of 1mg / kg, inhibiting the activation of macrophages in vitro and decreasing edema in a model of zimozan-induced arthritis in mice when treated orally (5 mg / kg). The results obtained demonstrate that it was possible to obtain a new chemical entity from reaction between naringenin and aminoguanidine with antioxidative, hypoglycemic and anti - inflammatory activity which can be further explored in the development of a prototype for treatment of chronic diseases such as diabetes or other processes inflammatory drugs. O DCHA apresentou atividade antioxidante superior a naringenina e apresentou inibi????o satisfat??ria da glica????o, demonstrando elevado potencial antiglica????o in vitro.

Phytochemical studies of the species, which elucidate their chemical composition, fractionated in sesquiterpenes and diterpenes, have been performed. Although crude oil is well studied, it is scarce to know the effect of its isolated substances, and there are few studies on the structural modifications of these diterpene acids, seeking to improve their biological activities. In this sense, the present work had as objectives the isolation of the majority diterpenic acids present in the Copaifera multijuga oil: copalic and 3-hydroxy-copalic acids and Copaifera reticulata: hardwickiic acid, with structural modifications in the 2 (two) lowcost and optimized chemistries, as well as in vitro and in vivo assays of the substance with the best anti-inflammatory screening and subsequent in silico studies. After isolation, the substances were semi-synthesized and their structures elucidated by 1H and 13C NMR analysis, and therefore, tested in cell viability assay and TNF-?? inhibition activity check test in vitro. The in vivo model of pleurisy/pleuritis in balb-c mice was performed with the best anti-inflammatory substance (ACD13B-esterification of ibuprofen with 3-hydroxy-copalic acid), as well as expression assay of IKB-?? protein. In addition, the gastrointestinal test was also performed with the same substance. The semi-synthesized derivatives were: ACD4- epoxidation of copalic acid, ACD13B-esterification of ibuprofen with 3-hydroxy-copalic acid and ACD51-acylation of phthalic anhydride with 3-hydroxy-copalic acid. ACD13B showed no cytotoxic potential in the THP-1 line when tested at the concentration of 2 ??M. The other substances did not present significant cytotoxicity at concentrations tested. In addition, ACD13B significantly inhibited TNF-?? production in LPS-stimulated THP-1 cells. In animals with induced pleurisy, ACD13B reduced the total number of leukocytes by 40% and the number of mononuclear cells at a dose of 100 mg/kg intraperitoneally (IP) by up to 50%. There was also inhibition of pIKB-??. ACD13B presented a lower gastrointestinal effect when compared to ibuprofen in mice. The docking study revealed the affinity and mode of interaction of the substance ACD13B in COX-2 and COX-1, identifying hydrophobic and hydrophilic interactions. The results indicate that the ibuprofen derivative with 3-hydroxy copalic acid is a prototype of an anti-inflammatory drug with a lower gastrointestinal effect than ibuprofen, which may be explored in the future for other functional and therapeutic properties.

Foods presented in capsules or pills constituted by edible pieces of fruits and vegetables which, have been submitted to drying or dehydration processing, must be assessed as being novel. Zingiber zerumbet L. Smith (ZZ), an important member of family Zingiberaceae, used traditionally in folk medicine throughout the world, especially in the south of Asia including India. This species is marketed worldwide, mostly through the manufacture of the shampoo elaborated from its flower and, it presents countless medicinal properties, such as anti-inflammatory, antitumor, anti-allergic, antipyretic and anti-platelet aggregation activities. Zerumbona, the major active component extracted from the plant rhizome is a highly therapeutic-powered substance. Under this context, the present work proposes to encapsulate the aqueous and hydroalcoholic extracts from the rhizome of ZZ for being utilized as functional food, therefore, it was necessary to assess some their medicinal and toxic activities. ZZ rhizomes were processed for attaining the gross extract (ZZGE), and ground and submitted to maceration for achieving hydroalcoholic extracts. Determination of the minerals held by rhizomes of ZZ was determined by the AOAC (1995) method. The phytochemical screening of the extracts was carried out through Mattos (1997) methodology. Thin Layer Chromatography (TLC) in silica gel utilized iodine, serum sulfate, UV 254nm and 365nm as developers. The test with Artemia salina according to Meyer (1982) was employed for the cytotoxicity assay. Mice were utilized for the acute toxicity test, and followed up for 14 days. The ethanol-induced gastric model was utilized to determine the anti-ulcer activity. The antinociceptive activity was ascertained through acetic acid-induced abdominal writhing, hot plate and hyperalgesia. The anti-inflammatory activity was verified through Paw edema test in rats and ear edema test in mice. From the preliminary results we obtained a extractive rate of 60g from ZZGE and 10g from ZZHE, in a percentile of 1% and 0.16%, respectively. The extracts hold a characteristic odor. In the determination of ZZ rhizomes, there is a larger quantity of iron (231 mg/kg). The ZZGE and ZZHE extracts phytochemical screening revealed the presence of the following substances: xanthones, flavonoids, phenols, tannins and alkaloids. The TLC of ZZGE indicated a spot developed in iodine with a retention factor of approximately 0.7 cm similar to that of Zerumbona (0.8) and ZZHE presented three spots developed in iodine and UV 254nm. In acute toxicity and Subchronic, one may assert that ZZGE and ZZHE extracts do not produce any toxicity or mortality in mice and rats, yet some dependent doses caused some significant. ZZGE and ZZHE presented the LC50 of 117.49 and 181.9??g???mL on the assays with Artemia salina. The extracts had a strong inhibitory effect against gastric lesions have gastroprotective effect. Assessment of abdominal writhing test demonstrated that ZZGE and ZZHE in the doses of 250 and 500 mg/kg produced a significant anti-nociceptive effect. In the hot plate test, the extracts showed to hold analgesic activity. The anti-inflammatory activity assessment showed it to be significant in physiological action in determined doses in models of anti-inflammatory pain brought about by chemical stimulus. According to their preliminary outcomes both ZZGE and ZZHE presented anti-nociceptive, anti-inflammatory and anti-ulcerous effects, and held no toxicity. Probably, these extracts may be encapsulated as functional food, thus introducing a differentiated product into the Market and contributing to the scientific studies on this valuable, medicinal plant.

Effects of Eugenia punicifolia (Khunt ) DC related to metabolic syndrome in experimental models . Manaus, 2014. 133p. Thesis (Ph.D.) - Graduate Program Multi - Institutional Biotechnology, Federal University of Amazonas. The Eugenia punicifolia species is a plant popularly known as "pedra-ume-caa", widely distributed in the Amazon region. In the state of Amazonas, this species has been widely used in folk medicine to treat inflammatory and hyperglycemic disorders. However, studies involving other parameters found in the metabolic syndrome (MS) have not yet been elucidated. The MS is characterized by a complex disorder, represented by a number of cardiovascular risk factors involving central adiposity, hypertension, endothelial dysfunction and insulin resistance. Studies have shown that nutritional supplementation with plants that effectively address the pathogenic mechanisms of this syndrome represents an effective improvement for the problem. This project aim was to analyze biological activities in vitro and in vivo of the aqueous extract of the leaves of E. punicifolia (EEP), emphasizing the inhibitory activity digestive enzymes, angiotensin converting enzyme (ACE) inhibitor, antioxidants, hypolipemiant, cytotoxic and anti antihypertensive effects. Enzyme inhibition assays, as well as the determination of the ACE activity in Wistars rats serum were performed by the fluorometric method and the indirect tests of cell viability, antioxidants, determining the presence of flavonoids and phenolic compounds were performed by colorimetric assays. The evaluation of the EEP on blood pressure parameters was performed by using five experimental groups, which were randomly divided into: normotensive group (2R), the hypertensive group (2R1C), enalapril hypertensive group (ENAL), extract hypertensive group 150 mg/kg/day (EEP150) and extract hypertensive group 300 mg/kg/dia (EEP300). The effects of EEP in systolic pressure were acquired indirectly by tail plethysmography in conscious rats. Biochemical parameters were measured in rat???s plasma through appropriate kits. The study has shown, for the first time, that EEP exhibited scavenging activity on ABTS??+, DPPH??, and O2??- (IC50=10.5 ?? 1.2; 28.84 ?? 0.54 and 38.12 ?? 2.6 ??g/mL, respectively). The EEP also exhibited cellular antioxidant activity in vitro concentration-dependent manner, and did not show cell cytotoxic effects. Furthermore, the EEP inhibited ACE activity (70.8%) using the concentration of 0.3 mg/mL (IC50 of 24.3 ?? 0.7 mg/mL), and inhibited the activities of the enzymes ??-amylase (IC50 = 122.8 ?? 6.3 mg/mL), ??-glucosidase (IC50 = 2.9 ?? 0.1 mg/mL) and xanthine oxidase (IC50 = 23.5 ?? 2.6 mg/mL). Both the EEP150 (21.7%) and the EEP300 (16.8%) reduced systolic blood pressure. The hypotensive effect of EEP correlated with the reduction of plasma ACE activity and also reduced total cholesterol (48%), triglycerides (52%), VLDL (52%) and LDL (55%). In addition, the EEP wasn???t cause renal, hepatic or neurotoxic acute or chronic toxicity. The EEP showed antioxidants, hypotensive, hypolipidemic effects and digestive enzymes and ACE were inhibited. These activities are probably related to the presence of antioxidant such as phenols and flavonoids compounds present in the extract, which act by inhibiting key enzymes of carbohydrate metabolism and angiotensin converting enzyme. These findings may support further clinical studies to further evaluate the potential of EEP on the SM. Galeno, D.M.L. O EEP mostrou atividades antioxidantes, hipotensores, hipolipemiantes e inibiu as enzimas digest??rias e a ECA.

The objective of this study was to investigate the determinants of LBW. Cross-sectional study of descriptive population-based involved 384 mothers attended in a public hospital. Forms with closed questions were used on the socioeconomic, demographic condition of pregnancy and nutritional standard features, and anthropometric measurements of prenatal card. The results showed an incidence of LBW infants 6.5%, not registering this association influence of socioeconomic variables (p> 0.05). In relation to maternal characteristics, the number of consultations in prenatal care, the previous history of abortion and the type of delivery showed a statistically significant relationship in determining the BPN (p 0.05), and zero the occurrence of this event in mothers with older age (??? 35 years). Analysis of these results shows a low occurrence of LBW in the universe studied, however it reinforces the need to implement a program of assistance and guidance to more efficient prenatal care for pregnant women teens. Oxidative stress caused by the action of free radicals is involved in the pathogenesis of many chronic diseases such as CVD. Flavonoids and vitamin C are antioxidant nutrients found in fruits and vegetables, and its regular consumption is associated with decreased risk of developing chronic diseases. Camu-camu (Myrciaria dubia [Kunth] McVaugh) is a fruit from the Amazon region that has significant levels of antioxidants, especially vitamin C and flavonoids. This study aimed to evaluate the effect of dust from the pulp of camu-camu on glucose and lipid profile in young adults. The pulp of camu-camu was lyophilized and its nutritional composition was analyzed using the methods indicated by Instituto Adolpho Lutz for the centesimal composition and HPLC method for vitamin C. For the intervention, was conducted a not randomized, double-blind, controlled clinical trial with 18 volunteers of both genders, aged between 21 and 35 years. The volunteers were divided into two groups: 1) intervention group that received capsules of lyophilized camu-camu containing 320 mg of vitamin C; 2) control group that received capsules containing 320 mg of synthetic vitamin C. The ingestion of capsules was daily for 15 days and blood samples were collected and analyzed before and after the intervention. Statistical differences in the levels of vitamin C in plasma, fasting glucose and lipid profile were verified by means of Student's ???t??? test. The lyophilized camu-camu presented 20,310 mg of vitamin C/100g and 12.89 mg of flavonoids/100g. At the end of the intervention was reported a significant decrease in fasting glucose, total cholesterol and HDL-cholesterol (p 0.05). It was concluded that the capsules of camu-camu were more efficient, presenting hypolipidemic and hypoglycemic action in volunteers studied. These results demonstrate the potential benefit of vitamin C and camu-camu health, presenting the fruit as an excellent source of this vitamin.

Pesquisa do Estado do Amazonas Dyslipidemia is a modification in blood levels of fat, one of the higher incidence of the pathologies in the world population, currently regarded as a public health concern. Some factors contribute greatly to increase cardiovascular diseases; among them, the lipid metabolism is one of most important. This factor is related to ingestion of lipid but the consumption of monounsaturated fatty acid (oleic) show neutral effect over cholesterolemy. However, it has been noticed that high oleic acid diets increase High Density Lipoprotein level (HDL-c) and may reduce the Low Density Lipoprotein level (LDL-c). The objective was to evaluate the Dyslipidemia Reduction Factor and compare the lipid profile and nutrition of the animals treated with the commercial diet supplemented and compare physicochemical fat and oil indexes using extra virgin olive oil and cupua??u fatty seed. In this comparative study was used experimentally fat of cupua??u seeds (Theobroma grandiflorum, Schum.) and olive oil (Olea europaea) extra virgin as a food supplementing in Wistar rats male youths. In the experiment were used six groups of eight animals and five of them were induced to dyslipidemia by use of monosodium glutamate. The applicability was 28 days which was provided usual feeding with supplemented of (0.1ml to 0.3ml) of one type of lipid in each group and gas chromatography and physicochemical analysis such as saponification, acidity, peroxide, and free fatty acid levels. So according to the procedures of gavage and oral route there was no difference, however between the lipidogram tests performed, Treatment with fat cupua??u seed was what achieved the best action of Dyslipidemia Reduction Factor, except for HDL cholesterol that there was no difference in any treatment. Results show that extra virgin olive oil and cupua??u fatty seed have same fatty acids; although, different amounts for these constituents. These results indicate that different quantities of same fatty acids on different levels may interfere on hipodislipidemic factor for cupua??u fatty seed diets.

In Brazil, studies on the use of scarce anabolizantes are e, in this study, searched considera????es siginificativas to compose theoretical referencial on the alert subject and to the users of these products. It was examined perception of risk to the health, associated with the consumption of anabolizantes between practitioners of physical activities in the academies of the city of Manaus-AM. For in such a way, the collection proceeded from qualitative data and quantitative, such as etnografia, half-structuralized interviews and focal group with users of anabolizantes, using themselves applied analytical methods in the description of each stage of the experiments, had understood, beyond the laboratorias piss analyses, the analysis of chromatography and spectrometry, by means of statistical methods, analyzing the qui-square of the result for its validation. The produced data had described main used substances and the standards of use, between practitioners of physical activities in the city of Manaus pointing preponderant evidences to the masculine sex, as bigger substantial food consumer to the organism, amongst them: Calcium, Grain, Copper, Zinc, Iron, Manganese, B1 Vitamins, and the C, and Liquids, it is fallen again substance of Carboidratos, Fruits and Oil and Fat more for the consumption of the feminine sex. In the analyses physicist-chemistry of the urinas we find eighteen positive samples for forbidden substances, of these, twelve met in the results of the masculine sex. Amongst substances we have of the bigger evidence of pH Metiltestosterona and THC, Testosterone and DHEA, causers of pathological effect in the human metabolism. The results make in them to analyze that it has absence of real cares to the organic body, therefore the interviewed ones are cliente of males that such substances can cause. One reveals, thus, the desire of the ideal physical body to the standards taxes in the society, to this overlap to the risk of harmful collateral effect how much to the bad use of these substances. It is considered that more must be promoted the prevention and the orientation with regard to the anabolizantes use of esteroides next to this population. This study it intended to well more aim at the education of the anab??licos users of esteroides and the users in potential, looking for to inform involved with the practical sport and the porting one, therefore exactly with studies related to this still it is lacked of prevention.

Pesquisa do Estado do Amazonas Obesity is a disease that occurs due to the unbalance between the ingested energy and the spent energy to maintain the vital processes. Obesity has incresingly wrong and reached, nowadays, and epidemic level. It has also been considered the main public health issues in modern society. It is asociated with the rising risk of developing Diabetes Melliteus, cardiovascular diseases, dislipodemia, and several other chronic diseases. The diet based on fat maybe crucial reason to develop such diseases. The fishing also provides a large sort of food to thousands of local inhabitants nutrional development. Among the most well-known and consumed Amazon fish there is Tambaqui (Colossoma macropomum), Matrinx?? (Brycon cephalus) and Mapar?? (Hypophthalmus edentatus), which are all considered very fat species. And so, the main purpose of this essay is to analyse the effects of a diet based on Amazon fat fish (Mapar??, Matrinx?? and Tambaqui) on male mice lipydic metabolism from wistar blood line. They were feeded with Adlibitum for 14 days. To make it happen doses of total cholesterol and triglycerids, glycoses, proteins and plasma lipydic fraction were made. The rate of lypogenesis again in vivo has been measured on RET, EPI, FIG and CARC, by incorporating 3 H2O in lipy. Our results show that the enrichment of a diet based on Amazon fat fish has altered the mice lipydic metabolism, dicreased effectively the lipid plasmatic concentration (Col. TG, e LDL) and increased the HDL besides the lipolitic activity on fatty tissues has grown after 4 weeks of feeding. Admiting that the ingestion of hiperlipydic diets may have caused an aterogenic effect, mainly because they have reduced certain lipydic fractions that have been claimed to cause serious vascular problems.
