Fault slip is a complex natural phenomenon involving multiple spatiotemporal scales from seconds to days to weeks. To understand the physical and chemical processes responsible for the full fault slip spectrum, a multidisciplinary approach is highly recommended. The Near Fault Observatories (NFOs) aim at providing high-precision and spatiotemporally dense multidisciplinary near-fault data, enabling the generation of new original observations and innovative scientific products. The Alto Tiberina Near Fault Observatory is a permanent monitoring infrastructure established around the Alto Tiberina fault (ATF), a 60 km long low-angle normal fault (mean dip 20°), located along a sector of the Northern Apennines (central Italy) undergoing an extension at a rate of about 3 mm yr−1. The presence of repeating earthquakes on the ATF and a steep gradient in crustal velocities measured across the ATF by GNSS stations suggest large and deep (5–12 km) portions of the ATF undergoing aseismic creep. Both laboratory and theoretical studies indicate that any given patch of a fault can creep, nucleate slow earthquakes, and host large earthquakes, as also documented in nature for certain ruptures (e.g., Iquique in 2014, Tōhoku in 2011, and Parkfield in 2004). Nonetheless, how a fault patch switches from one mode of slip to another, as well as the interaction between creep, slow slip, and regular earthquakes, is still poorly documented by near-field observation. With the strainmeter array along the Alto Tiberina fault system (STAR) project, we build a series of six geophysical observatory sites consisting of 80–160 m deep vertical boreholes instrumented with strainmeters and seismometers as well as meteorological and GNSS antennas and additional seismometers at the surface. By covering the portions of the ATF that exhibits repeated earthquakes at shallow depth (above 4 km) with these new observatory sites, we aim to collect unique open-access data to answer fundamental questions about the relationship between creep, slow slip, dynamic earthquake rupture, and tectonic faulting.

Sharks may occupy both intermediate and upper levels of marine food webs. They are overfished worldwide and constitute one of the largest portions of the bycatch. The removal of top-predator species has negative cascading effects on the entire food web, causing the “mesopredator release” phenomenon, which leads to an increase in mesopredators with consequent changes in the ecosystem’s energetic balance. Despite their important ecological role, information on their trophic ecology is limited. This essential information can be obtained through the analysis of stomach contents and, more recently, by using less invasive techniques, such as the stable isotope analysis of muscle tissue, obtained through biopsies. Here, we analysed the trophic ecology of the pyjama shark or striped catshark Poroderma africanum, an endemic species of South Africa, by means of SIA. The results obtained from SIA were analysed using the R SIMMR and SIBER packages to estimate the contribution of potential food sources to the diet and to evaluate the extent of the trophic niches. The SIMMR outputs showed that adults select more benthic prey than juveniles, which consume more planktonic species, with juveniles being more generalist than adults, according to SIBER outputs. As assessed by IUCN, the population of P. africanum is increasing, and given its role as mesopredator, future monitoring efforts could be crucial to elucidating their potential effects in marine food webs.

IntroductionTrisomy 21 (T21), which causes Down syndrome (DS), is the most common chromosomal aneuploidy in humankind and includes different clinical comorbidities, among which the alteration of the immune system has a heavy impact on patient’s lives. A molecule with an important role in immune response is zinc and it is known that its concentration is significantly lower in children with T21. Different hypotheses were made about this metabolic alteration and one of the reasons might be the overexpression of superoxide dismutase 1 (SOD1) gene, as zinc is part of the SOD1 active enzymatic center.MethodsThe aim of our work is to explore if there is a linear correlation between zinc level and immune cell levels measured in a total of 217 blood samples from subjects with T21. Furthermore, transcriptome map analyses were performed using Transcriptome Mapper (TRAM) software to investigate whether a difference in gene expression is detectable between subjects with T21 and euploid control group in tissues and cells involved in the immune response such as lymphoblastoid cells, thymus and white blood cells.ResultsOur results have confirmed the literature data stating that the blood zinc level in subjects with T21 is lower compared to the general population; in addition, we report that the T21/control zinc concentration ratio is 2:3, consistent with a chromosomal dosage effect due to the presence of three copies of chromosome 21. The transcriptome map analyses showed an alteration of some gene’s expression which might explain low levels of zinc in the blood.DiscussionOur data suggest that zinc level is not associated with the levels of immunity cells or proteins analyzed themselves and rather the main role of this ion might be played in altering immune cell function.

The redox state of the Earth's upper mantle (i.e., oxygen fugacity, fO2) is a key variable that influences numerous processes occurring at depth like the mobility of volatile species, partial melting, and metasomatism. It is linked to the oxidation state of peridotite rocks, which is normally determined through the available oxythermobarometers after measuring the chemical composition of equilibrated rock-forming minerals and the Fe3+ in redox-sensitive minerals like spinel or garnet. To date, accurate measurements of Fe3+ / ∑Fe in peridotites have been limited to those peridotites (e.g., harzburgites and lherzolites) for which an oxythermobarometer exists and where spinel (or garnet) crystals can be easily separated and measured by conventional 57Fe Mössbauer spectroscopy. Wehrlitic rocks have been generally formed by the interaction of a lherzolite with carbonatitic melts and, therefore, have recorded the passage of (metasomatic) fluids at mantle conditions. However, no oxythermobarometer exists to determine their equilibrium fO2. The aim of this study was to retrieve the fO2 of the mantle beneath Mt. Vulture volcano (Italy) through the study of a wehrlitic lapillus emitted during the last eruption (∼ 140 kyr ago) that contain olivines with multiple tiny spinel inclusions with sizes < 40 µm. To our knowledge, the Fe oxidation state of these inclusions has been never determined with the Mössbauer technique due to their small sizes. Here, we present measurements of the Fe3+ / ∑Fe using in situ synchrotron Mössbauer spectroscopy coupled with chemical and spectroscopic analysis of both host olivine and spinel inclusions. The results show Fe3+ / ∑Fe ratios of 0.03–0.05 for olivine and 0.40–0.45 for the included spinels, the latter of which appear higher than those reported in literature for mantle spinel harzburgites and lherzolites. Given the evidence of the mantle origin of the trapped spinels, we propose that the high fO2 (between 0.81 and 1.00 log above the fayalite–magnetite–quartz buffer; FMQ) likely results from the interaction between the pristine spinel lherzolite and a CO2-rich metasomatic agent prior to the spinel entrapment in olivines at mantle depths.

Down syndrome (DS) or trisomy 21 is the most common genetic cause of intellectual disability (ID), but a pathogenic mechanism has not been identified yet. Studying a complex and not monogenic condition such as DS, a clear correlation between cause and effect might be difficult to find through classical analysis methods, thus different approaches need to be used. The increased availability of big data has made the use of artificial intelligence (AI) and in particular machine learning (ML) in the medical field possible.The purpose of this work is the application of ML techniques to provide an analysis of clinical records obtained from subjects with DS and study their association with ID.We have applied two tree-based ML models (random forest and gradient boosting machine) to the research question: how to identify key features likely associated with ID in DS. We analyzed 109 features (or variables) in 106 DS subjects. The outcome of the analysis was the age equivalent (AE) score as indicator of intellectual functioning, impaired in ID. We applied several methods to configure the models: feature selection through Boruta framework to minimize random correlation; data augmentation to overcome the issue of a small dataset; age effect mitigation to take into account the chronological age of the subjects.The results show that ML algorithms can be applied with good accuracy to identify variables likely involved in cognitive impairment in DS. Abstract Background Down syndrome (DS) is caused by the presence of an extra copy of full or partial human chromosome 21 (Hsa21). Partial (segmental) trisomy 21 (PT21) is the duplication of only a delimited region of Hsa21 and can be associated or not to DS: the study of PT21 cases is an invaluable model for addressing genotype–phenotype correlation in DS. Previous works reported systematic reanalyses of 132 subjects with PT21 and allowed the identification of a 34-kb highly restricted DS critical region (HR-DSCR) as the minimal region whose duplication is shared by all PT21 subjects diagnosed with DS. Methods We report clinical data and cytogenetic analysis of two children with PT21, one with DS and the other without DS. Moreover, we performed a systematic bibliographic search for any new PT21 report. Results Clinical and cytogenetic analyses of the two PT21 children have been reported: in Case 1 the duplication involves the whole long arm of Hsa21, except for the last 2.7 Mb, which are deleted as a consequence of an isodicentric 21: the HR-DSCR is within the duplicated regions and the child is diagnosed with DS. In Case 2 the duplication involves 7.1 Mb of distal 21q22, with a deletion of 2.1 Mb of proximal 20p, as a consequence of an unbalanced translocation: the HR-DSCR is not duplicated and the child presents with psychomotor development delay but no clinical signs of DS. Furthermore, two PT21 reports recently published (named Case 3 and 4) have been discussed: Case 3 has DS diagnosis, nearly full trisomy for Hsa21 and a monosomy for the 21q22.3 region. Case 4 is a baby without DS and a 0.56-Mb duplication of 21q22.3. Genotype–phenotype correlation confirmed the presence of three copies of the HR-DSCR in all DS subjects and two copies in all non-DS individuals. Conclusions The results presented here are fully consistent with the hypothesis that the HR-DSCR is critically associated with DS diagnosis. No exception to this pathogenetic model was found. Further studies are needed to detect genetic determinants likely located in the HR-DSCR and possibly responsible for core DS features, in particular intellectual disability.

Abstract Deception Island is one of the most active volcanoes in Antarctica with more than twenty explosive eruptions in the past two centuries. Any future volcanic eruption(s) is a serious concern for scientists and tourists, will be detrimental to marine ecosystems and could have an impact to global oceanographic processes. Currently, it is not possible to carry-out low and high frequency volcanic gas monitoring at Deception Island because of the arduous climatic conditions and its remote location. Helium, neon and argon isotopes measured in olivine samples of the main eruptive events (pre-, syn- and post caldera) offer insights into the processes governing its volcanic history. Our results show that: (i) ascending primitive magmas outgassed volatiles with a MORB-like helium isotopic signature (3He/4He ratio); and (ii) variations in the He isotope ratio, as well as intensive degassing evidenced by fractionated 4He/40Ar* values, occurred before the beginning of the main eruptive episodes. Our results show how the pre-eruptive noble gas signals of volcanic activity is an important step toward a better understanding of the magmatic dynamics and has the potential to improve eruption forecasting.

Crustal degassing of 4He from fault zones occurs throughout the seismic cycle which suggests variations in this flux could be a valuable seismic monitoring tool, according to an analysis of He flux calculations and seismic data from the Irpinia Fault Zone, Italy.

Static and dynamic stress, along with earthquakes, can trigger the emission and migration of crustal fluids, as frequently observed on the surface and within the upper crust of tectonically active areas such as the northern Apennines of Italy. To investigate the origin of these fluids and their interconnection with the seismogenic process, we complemented The Alto Tiberina Near Fault Observatory (TABOO-NFO), a multidisciplinary monitoring infrastructure composed of a dense array of seismic, geodetic, strain, and radon sensors, with a proper geochemical network grounded on four soil CO2 flux monitoring stations and weather sensors, placed near the main vents of the superficial manifestations. Data about Palaeolithic peopling, settlement dynamics and techno-economy of the south-western margin of the Alpine region are sketchy. In this area, the lack of systematic research and the scarcity of lithic raw materials, spread the idea that Piedmont was not inhabited during Palaeolithic. In 2009, the re-starting of the excavations at the Ciota Ciara cave, gave rise to new questions and to the development of research projects at a regional scale. The Ciota Ciara cave is the only Middle Palaeolithic site object of multidisciplinary and systematic investigations. Its lithic assemblage, analysed through a techno-economic approach, allows to understand in detail the technological choices and the land mobility of the Neanderthal groups on a local and sub-regional scale. Other Middle Palaeolithic assemblages are known in the region and are all issued from surface collections. They come from the northern part of the region, from Vaude Canavesane, Trino, Baragge biellesi and Colline Novaresi. The technological study of these assemblages led to the identification of strong similarities in the technological choices of the Middle Palaeolithic human groups: they based their technology on the exploitation of vein quartz, a rock diffused all over the regional territory, from time to time accompanied by other local (spongolite, rhyolite, metamorphic rocks, jasper) and allochthonous (radiolarite and flint) lithic resources, with technological adaptation to their quality and mechanical properties both when it comes to predetermined methods (Levallois and discoid) and when expedient reduction sequences are used. Concerning Upper Palaeolithic, the only lithic assemblage issued from an archaeological excavation (and therefore with a clear stratigraphic context) is that from the Epigravettian site of Castelletto Ticino. Other lithic artefacts referable on a techno-typological basis to Upper Palaeolithic are from Trino and Colline Novaresi. As for Middle Palaeolithic, the techno-economic approach used in the analysis of these lithic assemblages, allow to have, for the first time, reliable data at a regional scale. In this work we present the data obtained after about ten years of research in Piedmont: they outline a scenario where, even in the limits of analysis mostly based on materials issued from surface collections, we can see both clear differences between the Middle and the Upper Palaeolithic technological behaviours and hypothesise the land mobility of the hunter-gatherers’ groups that inhabited the region.

Simple Summary: Sharks, as important members of marine food webs, are often targeted by commercial and sport fishing, and moreover they constitute a significant part of the bycatch along with marine mammals and turtles. Overfishing, especially of top-predator species, disrupts the entire food web through a process called "mesopredator release", where the removal of top predators leads to an increase in mid-level predators, altering the ecosystem balance. Abstract The Down syndrome (DS) phenotype is usually characterized by relative strengths in non-verbal skills and deficits in verbal processing, but high interindividual variability has been registered in the syndrome. The goal of this study was to explore the cognitive profile, considering verbal and non-verbal intelligence, of children and adolescents with DS, also taking into account interindividual variability. We particularly aimed to investigate whether this variability means that we should envisage more than one cognitive profile in this population. The correlation between cognitive profile and medical conditions, parents’ education levels and developmental milestones was also explored. Seventy-two children/adolescents with DS, aged 7–16 years, were assessed with the Wechsler Preschool and Primary Scale of Intelligence-III. Age-equivalent scores were adopted, and Verbal and Non-Verbal indices were obtained for each individual. The cognitive profile of the group as a whole was characterized by similar scores in the verbal and non-verbal domain. Cluster analysis revealed three different profiles, however: one group, with the lowest scores, had the typical profile associated with DS (with higher non-verbal than verbal intelligence); one, with intermediate scores, had greater verbal than non-verbal intelligence; and one, with the highest scores, fared equally well in the verbal and non-verbal domain. Three cognitive profiles emerged, suggesting that educational support for children and adolescents with DS may need to be more specific.

Abstract Down syndrome (DS) is characterised by several clinical features including intellectual disability (ID) and craniofacial dysmorphisms. In 1976, Jackson and coll. identified a checklist of signs for clinical diagnosis of DS; the utility of these checklists in improving the accuracy of clinical diagnosis has been recently reaffirmed, but they have rarely been revised. The purpose of this work is to reassess the characteristic phenotypic signs and their frequencies in 233 DS subjects, following Jackson's checklist. 63.77% of the subjects showed more than 12 signs while none showed less than 5, confirming the effectiveness of Jackson's checklist for the clinical diagnosis of DS. An association between three phenotypic signs emerged, allowing us to distinguish two sub-phenotypes: Brachycephaly, short and broad Hands, short Neck (BHN), which is more frequent, and "non-BHN". The strong association of these signs might be interpreted in the context of the growth defects observed in DS children suggesting decreased cell proliferation. Lastly, cognitive assessments were investigated for 114 subjects. The lack of association between the presence of a physical sign or the number of signs present in a subject and cognitive skills disproves the stereotype that physical characteristics are predictive of degree of ID.

IntroductionDown syndrome (DS) is the most common chromosomal disorder and it is caused by trisomy of chromosome 21 (Hsa21). Subjects with DS show a large heterogeneity of phenotypes and the most constant clinical features present are typical facies and intellectual disability (ID). Several studies demonstrated that trisomy 21 causes an alteration in the metabolic profile, involving among all the one-carbon cycle.MethodsWe performed enzyme-linked immunosorbent assays (ELISAs) to identify the concentration of 5 different intermediates of the one-carbon cycle in plasma samples obtained from a total of 164 subjects with DS compared to 54 euploid subjects. We investigated: tetrahydrofolate (THF; DS n = 108, control n = 41), 5-methyltetrahydrofolate (5-methyl-THF; DS n = 140, control n = 34), 5-formyltetrahydrofolate (5-formyl-THF; DS n = 80, control n = 21), S-adenosyl-homocysteine (SAH; DS n = 94, control n = 20) and S-adenosyl-methionine (SAM; DS n = 24, control n = 15).ResultsResults highlight specific alterations of THF with a median concentration ratio DS/control of 2:3, a decrease of a necessary molecule perfectly consistent with a chromosomal dosage effect. Moreover, SAM and SAH show a ratio DS/control of 1.82:1 and 3.6:1, respectively.DiscussionThe relevance of these results for the biology of intelligence and its impairment in trisomy 21 is discussed, leading to the final proposal of 5-methyl-THF as the best candidate for a clinical trial aimed at restoring the dysregulation of one-carbon cycle in trisomy 21, possibly improving cognitive skills of subjects with DS.

Abstract Post‐orogenic back‐arc magmatism is accompanied by hydrothermal ore deposits and mineralizations derived from mantle and crustal sources. We investigate Zannone Island (ZI), back‐arc Tyrrhenian basin, Italy, to define the source(s) of mineralizing hydrothermal fluids and their relationships with the regional petrological‐tectonic setting. On ZI, early Miocene thrusting was overprinted by late Miocene post‐orogenic extension and related hydrothermal alteration. Since active submarine hydrothermal outflow is reported close to the island, Zannone provides an ideal site to determine the P‐T‐X evolution of the long‐lived hydrothermal system. We combined field work with microstructural analyses on syn‐tectonic quartz veins and carbonate mineralizations, X‐ray diffraction analysis, microthermometry and element mapping of fluid inclusions (FIs), C, O, and clumped isotopes, and analyses of noble gases (He‐Ne‐Ar) and CO2 content in FIs. Our results document the evolution of a fluid system of magmatic origin with increasing mixing of meteoric fluids. Magmatic fluids were responsible for quartz veins precipitation at ∼125 to 150 MPa and ∼300°C–350°C. With the onset of extensional faulting, magmatic fluids progressively interacted with carbonate rocks and mixed with meteoric fluids, leading to (a) host rock alteration with associated carbonate and minor ore mineral precipitation, (b) progressive fluid neutralization, (c) cooling of the hydrothermal system (from ∼320°C to ∼86°C), and (d) embrittlement and fracturing of the host rocks. Both quartz and carbonate mineralizations show noble gases values lower than those from the adjacent active volcanic areas and submarine hydrothermal systems, indicating that the fossil‐to‐active hydrothermal history is associated with the emplacement of multiple magmatic intrusions.

Carbon dioxide (CO2) is released from the Earth’s interior into the atmosphere through both volcanic and non-volcanic sources in a variety of tectonic settings. A quantitative understanding of CO2 outgassing fluxes in different geological settings is thus critical for decoding the link between the global carbon budget and different natural processes (e.g., volcanic eruption and earthquake nucleation) and the effects on the climate evolution over geological time. It has recently been proposed that CO2 degassing from non-volcanic areas is a major component of the natural CO2 emission budget, but available data are still sparse and incomplete. Here, we report the results of a geochemical survey aimed at quantifying CO2 emissions through cold and thermal springs of the tectonically active Pollino Massif and Calabrian arc (Southern Italy). The chemical ad isotopic (He and C) composition of fifty-five dissolved gas samples allows to identify two different domains: 1) a shallow system dominated by gas components of atmospheric signature (helium, hereafter He) and biogenic origin (C), and 2) a deeper system in which crustal/deep fluids (CO2 and He) are dominant. The measured He isotope ratios range from 0.03 to 1.1 Ra (where Ra is the He isotopic ratio in the atmosphere) revealing a variable atmospheric contamination. Furthermore, the He isotopic data indicate the presence of traces of mantle He contributions (2%–3%) in the thermal groundwater. The prevailing low R/Ra values reflect the addition of crustal radiogenic 4He during groundwater circulation. Using helium and carbon isotope data, we explore the possible sources of fluids and the secondary processes (dissolution/precipitation) that act to modify the chemistry of pristine volatiles. For the thermal springs, we estimate a deep C output of 2.3 x 107 to 6.1 x 108 mol year−1. These values correspond to deep CO2 fluxes per square km comparable with those estimated in several active and inactive volcanic areas and in continental regions affected by metamorphic CO2 degassing (e.g., the southern margin of the Tibetan Plateau).

Abstract Background Trisomy 21 (T21) is a genetic alteration characterised by the presence of an extra full or partial human chromosome 21 (Hsa21) leading to Down syndrome (DS), the most common form of intellectual disability (ID). It is broadly agreed that the presence of extra genetic material in T21 gives origin to an altered expression of genes located on Hsa21 leading to DS phenotype. The aim of this study was to analyse T21 and normal control blood cell gene expression profiles obtained by total RNA sequencing (RNA-Seq). Results The results were elaborated by the TRAM (Transcriptome Mapper) software which generated a differential transcriptome map between human T21 and normal control blood cells providing the gene expression ratios for 17,867 loci. The obtained gene expression profiles were validated through real-time reverse transcription polymerase chain reaction (RT-PCR) assay and compared with previously published data. A post-analysis through transcriptome mapping allowed the identification of the segmental (regional) variation of the expression level across the whole genome (segment-based analysis of expression). Interestingly, the most over-expressed genes encode for interferon-induced proteins, two of them (MX1 and MX2 genes) mapping on Hsa21 (21q22.3). The altered expression of genes involved in mitochondrial translation and energy production also emerged, followed by the altered expression of genes encoding for the folate cycle enzyme, GART, and the folate transporter, SLC19A1. Conclusions The alteration of these pathways might be linked and involved in the manifestation of ID in DS.

ABSTRACT This study provides new mineral chemistry data together with micro‐thermometric measurements on fluid inclusions hosted in ultramafic xenoliths (lherzolite, wehrlite, and dunite) brought to the surface by the last Mt. Vulture volcano activity (140 ka; southern Italy), and fed by melilitite‐carbonatite magmas. Petrographic evidence and mineralogical compositions of Mt. Vulture xenoliths are consistent with an origin in the upper mantle. Fluid inclusions in rock‐forming minerals of lherzolite and wehrlite xenoliths are CO2‐dominated. The equilibrium temperature calculated by geothermometric estimates ranges from 1039 C (±36°C) to 1142°C (±15°C), and entrapment pressures of fluid inclusions with post‐trapping re‐equilibration correspond to the local crust–mantle boundary (32 km depth), and to a shallow reservoir located at 12–14 km depth.
