Abstract

A mild and transition-metal-free method for the alpha-arylation of aliphatic nitriles with activated heteroaryl halides that were developed at ambient temperature was described. Generation of the nitrile anion in the presence of the heteroaryl halide was the success of the process.

Abstract

2,2-Bis{2-{4(S)-tert-butyl-1,3-oxazolinyl}}propane {(S,S)-tert-butylbis(oxazoline)} was synthesized through a three-step procedure that required less purification. The method involved catalyzing the reaction of dihydroxy malonodiamide with p-toluenesulfonyl chloride and triethylamine with 4-(dimethylamino)pyridine. Catalysis led to the formation of bis(tosylate), which further cyclizated to form bis(oxazoline). The compound was produced in 82% yield after extractive isolation, followed by recrystallization from pentane.

Combination of s-BuLi and a (+)-sparteine surrogate is used to recognize the high yielding asymmetric deprotonation trapping of N-Boc piperidine. The lithiation by in situ react IR is used to facilitate the direct observation of a prelithiation complex.

Abstract

A general, enantioselective, palladium-catalyzed arylation of N-Boc-pyrrolidine that proceeds via in situ generated 2-pyrrolidinozinc reagent is reported. This sequence has offered a number of advantages over existing methods and has represented the most convenient and practical synthesis of enantiomerically enriched 2-arylpyrrolidines and 2,5-diarylpyrrolidines.

The stereogenic center was set using asymmetric allylic alkylation chemistry, and the core of the structure was constructed via Pd-catalyzed N-cyclization or Heck methodology.

The use of chiral mixed phosphorus/sulfur ligands 1-3 is shown to be effective in enantioselective Rh-catalyzed dehydroamino acid hydrogenation and ketone hydrosilylation reactions. A model for asymmetric enantioselectivities hydrogenation reaction is discussed.

Abstract

An asymmetric synthesis of HCV NS5B nucleoside polymerase inhibitor (1) is described. This novel route features several remarkably diastereoselective and high-yielding transformations, including construction of the all-carbon quaternary stereogenic center at C-2 via a thermodynamic aldol reaction. A subsequent glycosylation reaction with activated uracil via C-1 phosphate and installation of the cyclic phosphate group using an achiral phosphorus(III) reagent followed by oxidation provides 1.

Abstract

A novel class of mixed phosphorus/sulfur ligands that incorporates a metal-bound thioether as a chiral control element in asymmetric catalysis is reported. These three ligands are comprised of three subunits which include the Ar2P- and RS-heteroatom fragments and the interconnecting skeletal backbone. The palladium-catalyzed allylic alkylation with enol-malonate and amine nucleophiles demonstrates the utility of these ligands. The three ligands are also shown to promote allylic amination with benzylamine in 95%-99% enantiomeric excess.

Abstract

Research was conducted to to test whether ligand architecture can play a major role in the design of Sn(II)-based chiral Lewis acidic complexes. It was assumed that chelate organization between the catalyst and carbonyl-containing reactant may provide a stereoselective process with related substrates. Results showed that the ligand-metal complexes exhibited efficient turnover in the catalytic cycle and may be utilized in amounts as low as 2 mol%.
