The cultivation of the olive tree is of great economic importance worldwide. For the 2022-2023 harvest, were produced around 3,010,000 Mg of olive oil and 2,955,500 Mg of table olives of selected varieties, managing to establish itself in approximately 56 countries with different environments. In most of the cultivated areas only monovarietals were used, but mixtures of varieties have been recommended, which act as pollinators, avoiding reproduction problems. Furthermore, global warming also affects production efficiency and the quality of derived products. The olive grove underwent important changes due to genetic improvement. However, despite modern molecular tools, most of the work continues to be based on agronomic classical methods such as crossing and selection in progeny, delaying the development of new varieties. Models with high resolution climate projections are being used, allowing predicting the future evolution of the olive tree. Unfortunately, Brazil produces only 0.2% of its internal consumption and therefore has become the world's second largest importer after the United States to cover the growing demand. Based on this information, this review aims to discuss the adjustments in olive growing, which has been gaining more and more areas dedicated to its cultivation in Brazil.

ABSTRACT Rice (Oryza sativa) is among the most important crops worldwide; however, rice crops demand high water consumption. Future projections indicate reduced water availability and severe drought events, which may affect rice crops as the cereal is highly sensitive to drought stress. Thus, cultivars with lower water demand for irrigation or drought escape capacities are among the strategies to address this issue. This study aimed to assess five mutant rice genotypes in terms of growth cycle length, other agronomic interest traits, and drought response in the reproductive stage. The mutant genotypes evaluated showed a shortening of the growth cycle compared to the original cultivar, BRS Pampeira, representing a drought escape strategy. In addition, mutations did not negatively affect plant height and crop yield. However, similar to the original cultivar, mutants are not tolerant to water deficit in the reproductive stage. The genotypes evaluated have potential to be released as early cycle cultivars, which can reduce water demand during the harvest season while presenting a drought escape strategy.

Abstract The objective of this work was to map the chromosomal regions responsible for iron accumulation in rice grains, in Southern Brazil. Eighty-one rice accessions were genotyped and phenotyped for Fe accumulation. Single nucleotide polymorphisms were mapped in the whole grain on chromosomes 1, 5, 6, and 10, from which 13 candidate genes were identified. Some of the genes, such as O s10g040680 0, seem to have a relationship with Fe homeostasis, while others are related to other metabolic processes or have an unknown function.

Abstract Effective strategies for disease control are crucial for sustaining world food production and ensuring food security for the population. Wheat blast, a disease caused by the pathogen Magnaporthe oryzae pathotype Triticum, has been a concern for cereal producers and researchers due to its aggressiveness and rapid expansion. To solve this problem, the development of resistant varieties with durable resistance is an effective, economical and sustainable way to control the disease. Conventional breeding can be aided by several molecular tools to facilitate the mining of many sources of resistance, such as R genes and QTLs. The identification of new sources of resistance, whether in the wheat crop or in other cereals are an opportunity for efficient wheat breeding through the application of different techniques. Since this disease is still poorly studied in wheat, knowledge of the rice Magnaporthe pathotype may be adapted to control wheat blast. Thus, genetic mapping, molecular markers, transgenic approaches, and genomic editing are valuable technologies to fight wheat blast. This review aimed to compile the biotechnological alternatives available to accelerate the development of improved cultivars for resistance to wheat blast.

The genus Vitis belongs to the Vitaceae family and is divided into two subgenera: Muscadinia and Vitis, the main difference between these subgenera being the number of chromosomes. There are many hypotheses about the origin of the genus, which have been formed with archaeological studies and lately with molecular analyses. Even though there is no consensus on the place of origin, these studies have shown that grapes have been used by man since ancient times, starting later on its domestication. Most studies point to the Near East and Greece as the beginning of domestication, current research suggests it took place in parallel in different sites, but in all cases Vitis vinifera (L.) subsp. sylvestris [Vitis vinifera (L.) subsp. sylvestris (Gmelin) Hagi] seems to be the species chosen by our ancestors to give rise to the now known Vitis vinifera (L.) subsp. vinifera [=sativa (Hegi)= caucasica (Vavilov)]. Its evolution and expansion into other territories followed the formation of new empires and their expansion, and this is where the historical importance of this crop lies. In this process, plants with hermaphrodite flowers were preferentially selected, with firmer, sweeter, larger fruits of different colors, thus favoring the selection of genes associated with these traits, also resulting in a change in seed morphology. Currently, genetic improvement programs have made use of wild species for the introgression of disease resistance genes and tolerance to diverse soil and climate environments. In addition, the mapping of genes of interest, both linked to agronomic and fruit quality traits, has allowed the use of molecular markers for assisted selection. Information on the domestication process and genetic resources help to understand the gene pool available for the development of cultivars that respond to producer and consumer requirements.

WRKY transcription factor genes compose an important family of transcriptional regulators that are present in several plant species. According to previous studies, these genes can also perform important roles in bilberry (Vaccinium myrtillus L.) metabolism, making it essential to deepen our understanding of fruit ripening regulation and anthocyanin biosynthesis. In this context, the detailed characterization of these proteins will provide a comprehensive view of the functional features of VmWRKY genes in different plant organs and in response to different intensities of light. In this study, the investigation of the complete genome of the bilberry identified 76 VmWRKY genes that were evaluated and distributed in all twelve chromosomes. The proteins encoded by these genes were classified into four groups (I, II, III, and IV) based on their conserved domains and zinc finger domain types. Fifteen pairs of VmWRKY genes in segmental duplication and four pairs in tandem duplication were detected. A cis element analysis showed that all promoters of the VmWRKY genes contain at least one potential cis stress-response element. Differential expression analysis of RNA-seq data revealed that VmWRKY genes from bilberry show preferential or specific expression in samples. These findings provide an overview of the functional characterization of these proteins in bilberry.

In Rio Grande do Sul, the main rice producer State in Brazil, low temperatures can occur during germination and seedling establishment, and in some cases, during the reproductive stage. When low temperatures occur in the early developmental stages cause delay in germination, resulting in a non-homogeneous growing. In reproductive stage, low temperatures cause spikelet sterility, directly interfering with plant yield. Researchers have shown that some phenolic compounds such as proanthocyanidins and anthocyanin are associated with low temperature tolerance in plants due to their antioxidant capacity. The red and black color in the seeds of some rice genotypes is conferred by the phenolic compounds proanthocyanidins and anthocyanin, respectively. Therefore, tis study aimed to verify whether rice genotypes with red or black seeds are more tolerant to low temperatures during germination. In this study, five rice genotypes were tested, two present seeds without pigmentation and with contrasting response to low temperature tolerance (BRS Bojuru - tolerant and BRS Pampeira - sensitive), two genotypes with red seeds (BRS 902, SCS 119 Rubi) and one black seed genotype (SCS 120 Ônix). As expected, the genotypes with pigmented seeds had a greater total phenolic compounds content. However, under low temperature conditions, the genotypes with pigmented seed showed a similar response to the sensitive genotype. Therefore, the presence of proanthocyanidins and anthocyanin in the seed of the studied genotypes does not provide tolerance to low temperatures during germination.

Rice production (Oryza sativa L.) is among the most economically important activities in the world. However, soil and salinity coming from irrigation water reduce rice yield. Therefore, the identification and/or development of salt-tolerant rice genotypes is a strategy to minimize this problem. The development of new genotypes depends on the presence of genetic diversity, and understanding the heritability of a desired trait can help in the selection process. Thus, this study aimed to identify superior genotypes, analyze the genetic diversity and estimate the heritability for salinity tolerance at the seedling stage in rice genotypes used in Brazil. For this, seedlings of 69 genotypes were kept in hydroponic solution with 40 mM NaCl (4 dSm-1) for seven days. Shoot length, root length, shoot dry weight, and root dry weight) were evaluated and the results were converted into relative performance. Tolerant and moderately salt-tolerant genotypes were identified at the seedling stage, which can be used in breeding programs and can be cultivated in high salinity areas. Principal component analysis showed the presence of genetic diversity for salinity response. Finally, it was shown that most of the observed variation is of genetic origin, which can make the breeding process less difficult.

The olive crop has a large diversity in cultivars around the world, whose identification have been based on leaf, stem or fruit traits. However, these traits may be influenced by the environment, which has led to the misidentification of cultivars. The variability of the pistil and pollen structure under electron scanning and light microscopy were studied using the cultivars 'Arbequina', 'Arbosana', 'Frantoio', 'Koroneiki', 'Manzanilla de Sevilla' and 'Picual', cultivated in Southern Brazil. The aim was to find unique patterns for their identification. This type of study has not been carried out in the conditions of Southern Brazil, where the identification of commercial cultivars sometimes is complex and confusing. There were significant differences in the variables, showing uniformity within each cultivar in different locations. The principal component analysis indicated a separation of cultivars according to similarities or micromorphological differences in the pollen wall, area of the lumen and the structural elements of the tectum. For the analysis of the pistil, the separation was even clearer, each cultivar individually forming a cluster, using as characters the length of the style in contrast to the length, width and area of the stigma. This evidenced little variation among individuals within the same cultivar. The information provided can contribute to the individual identification of very close olive tree cultivars in the conditions of Southern Brazil.

Canola is an important oilseed crop and its oil is used in human diet and biodiesel production. Abiotic stressors severely constrain canola growing causing economic losses. Under adverse conditions, plant cells coordinate their metabolic pathways to produce anti-stress agents. The understanding of the metabolic profile can be used to improve target traits since there is a direct relationship with the phenotype. Here we take advantage of the non-targeted metabolite profiling analysis GC–MS technique to assess the differences in metabolite accumulation in canola seedling tissues subjected to heat, waterlogging, drought and salt stresses. Phenotypically, abiotic stresses affected canola root development, mainly in waterlogging, salt and drought conditions, in which root length was increased. We assayed the metabolic profile in each tissue and abiotic stress and organic acids, free amino acid and sugars were detected. Accumulation of these metabolites were variable between shoots and roots under abiotic stresses. Heat stress affected mainly shoots increasing some essential and non-essential amino acids and sucrose while decreasing glucose and fructose contents. Waterlogging also affected mainly shoots decreasing malic and citric acids, some essential and non-essential amino acids and xylose content. Salt stress leads to a reduction of most of the metabolic compounds from organic acids, free amino acid and sugar groups in shoots and roots. Controversially, drought stress resulted in an increasing in most of the metabolite groups in shoots and roots. Shoots presented greater variation in metabolite content in response to abiotic stress. Our data is useful as basic knowledge to improve canola specific traits.

In soils under acidic conditions, Aluminum (Al) is solubilized to its ionic form, which is toxic to plants. Al rapidly inhibits root elongation, water and nutrient uptake, resulting in crop yield reduction. Members of the MATE family are responsible for citrate transport and Al detoxification in different species. In rice, the OsFRDL1 gene (MATE family) is homologous to the HvAACT1 and SbMATE, which are involved in Al tolerance in barley and sorghum, respectively. Silencing OsFRDL1 showed that it is not involved in Al tolerance in rice. However, the OsFRDL1 expression was not accessed in rice genotypes contrasting for Al tolerance. Thus, in this study, four Brazilian rice genotypes were evaluated in response to Al treatment in different time of exposition and OsFRDL1 expression was analyzed. The analyzed cultivars displayed different responses to Al dose x time. Al affected root growth in all analyzed genotypes, however, a minor negative effect that only occurred after 72 and 48 hours of exposure was detected in Farroupilha and BRS Curinga cultivars, respectively. In contrast, BR-IRGA 410 and IAS 12-9 showed a negative effect in root growth from the first hours of exposure to Al. Two cultivars differing in Al tolerance were used for gene expression analysis. The expression of OsFRDL1 was highly increased in Al-tolerant cultivar Farroupilha than in the Al-sensitive cultivar BR IRGA 410. This results indicates that OsFRDL1 is regulated by Al. This finding suggests that OsFRDL1 is involved in Al stress response, however seems to be insufficient in controlling Al tolerance.

The high selection pressure applied in rice breeding since its domestication thousands of years ago has caused a narrowing in its genetic variability. Obtaining new rice cultivars therefore becomes a major challenge for breeders and developing strategies to increase the genetic variability has demanded the attention of several research groups. Understanding mutations and their applications have paved the way for advances in the elucidation of a genetic, physiological, and biochemical basis of rice traits. Creating variability through mutations has therefore grown to be among the most important tools to improve rice. The small genome size of rice has enabled a faster release of higher quality sequence drafts as compared to other crops. The move from structural to functional genomics is possible due to an array of mutant databases, highlighting mutagenesis as an important player in this progress. Furthermore, due to the synteny among the Poaceae, other grasses can also benefit from these findings. Successful gene modifications have been obtained by random and targeted mutations. Furthermore, following mutation induction pathways, techniques have been applied to identify mutations and the molecular control of DNA damage repair mechanisms in the rice genome. This review highlights findings in generating rice genome resources showing strategies applied for variability increasing, detection and genetic mechanisms of DNA damage repair.

Abstract: The objective of this work was to compare uni- and multivariate biometric methods to evaluate the adaptability and stability of an important group of white oat (Avena sativa) cultivars grown in Southern Brazil. The used experimental design was a randomized complete block, in a factorial arrangement of 12 environments x 7 cultivars, with three replicates. The analysis of variance and the methods of Eberhart & Russel, Annicchiarico, and the harmonic mean of the relative performance of predicted genetic values (MHPRVG) were assessed. In the general comparison of the methods, the 'UPFA Gaudéria' and 'URS Guapa' genotypes were more stable regarding grain yield. The 'UPFA Gaudéria' and 'URS-21' genotypes stood out for hectoliter weight, presenting the best performances by the methods of Annicchiarico and the MHPRVG. For thousand-grain weight, all methods showed similar results, indicating that the 'UPFA Gaudéria' genotype presented the best results. The 'URS Guapa' genotype was superior when using the methods of Eberhart & Russel, Annicchiarico, and the MHPRVG. The uni- and multivariate methods evaluated are suitable to estimate with high confidence the adaptability and stability of cultivars for each targeted grain production, yield, and quality.

Abstract Iron is a well-known metal. Used by humankind since ancient times in many different ways, this element is present in all living organisms, where, unfortunately, it represents a two-way problem. Being an essential block in the composition of different proteins and metabolic pathways, iron is a vital component for animals and plants. That is why iron deficiency has a severe impact on the lives of different organisms, including humans, becoming a major concern, especially in developing countries where access to adequate nutrition is still difficult. On the other hand, this metal is also capable of causing damage when present in excess, becoming toxic to cells and affecting the whole organism. Because of its importance, iron absorption, transport and storage mechanisms have been extensively investigated in order to design alternatives that may solve this problem. As the understanding of the strategies that plants use to control iron homeostasis is an important step in the generation of improved plants that meet both human agricultural and nutritional needs, here we discuss some of the most important points about this topic.

Canola (Brassica napus L. var. oleifera) is cultivated in the Southern Brazil as an alternative winter crop in the rotation system. No-tillage and hydromorphic soils favor the formation of aliphatic organic acids with short chain and low molecular weight, as acetic, butyric, and propionic acids. These acids may cause negative effects on the canola production. Thus, this study aimed to evaluate the response of five canola hybrid genotypes under the effect of acetic, butyric and propionic acids on the germination and initial phase of seedling growth. The results showed the characters evaluated respond differently to treatments applied within the same genotype. Therefore, the selection of a genotype with a resilient performance for the characters evaluated under stress is complex. Nevertheless, a sensitive profile response was observed in Hyola 433 genotype under different organic acids. Furthermore, acetic acid showed no negative impact on germination and initial growth of the genotypes.

Background: Fusarium Head Blight (FHB) is a worldwide devastating disease of bread wheat (Triticum aestivum L.). Genetic resistance is the most effective way to control FHB and many QTL related to this trait have been mapped on the wheat genetic map. This information, however, must be refined to be more efficiently used in breeding programs and for the advance of the basic research. The objective of the present study was to in-depth analyze the QTLome of FHB resistance in bread wheat, further integrating genetic, genomic, and transcriptomic data, aiming to find candidate genes.Methods: An exhaustive bibliographic review on 76 scientific papers was carried out collecting information about QTL related to FHB resistance mapped on bread wheat. A dense genetic consensus map with 572,862 loci was generated for QTL projection. Meta-analysis could be performed on 323 QTL. Candidate gene mining was carried out within the most refined loci, containing genes that were cross-validated with publicly available transcriptional expression data of wheat under Fusarium infection. Most highlighted genes were investigated for protein evidence.Results: A total of 556 QTL were found in the literature, distributed on all sub-genomes and chromosomes of wheat. Meta-analysis generated 65 meta-QTL, and this refinement allows one to find markers more tightly linked to these regions. Candidate gene mining within the most refined meta-QTL, meta-QTL 1/chr. 3B, harvested 324 genes and transcriptional data cross-validated 10 of these genes, as responsive to FHB. One is of these genes encodes a Glycosiltransferase and the other encodes for a Cytochrome P450, and these such proteins have already been verified as being responsible for FHB resistance, but the remaining eight genes still have to be further studied, as promising loci for breeding.Conclusions: The QTLome of FHB resistance in wheat was successfully assembled and a refinement in terms of number and length of loci was obtained. The integration of the QTLome with genomic and transcriptomic data has allowed for the discovery of promising candidate genes for use in breeding programs.

O objetivo deste trabalho foi descrever a influência da variação de temperatura na germinação de quatro híbridos de canola indicados para cultivo no Brasil. O estudo foi realizado no ano de 2016, na Universidade Federal de Pelotas, – RS. O delineamento experimental utilizado foi o DIC, dispostos em esquema fatorial, 4 Híbridos x 3 temperaturas de geminação, com 4 repetições. Os híbridos utilizados foram o Hyola 50, Hyola 61, Hyola 571CL e Hyola 575CL e as temperaturas testadas foram 10, 20 e 30°C. A temperatura influenciou diretamente a germinação dos híbridos de canola. As temperaturas de 20° a 30°C foram as que promoveram maior incremento nas variáveis: primeira contagem, percentual de sementes germinadas, índice de velocidade de germinação, comprimento de radícula, comprimento de parte aérea, massa seca de plântula e redução no tempo médio de germinação. Os híbridos Hyola 61 e Hyola 571CL apresentaram melhor germinação quando expostos a temperatura de 10°C.

O objetivo deste trabalho foi determinar as relações genéticas de 21 cultivares de marmeleiro com base no marcador "amplified fragment length polymorphism" (AFLP), para melhoramento e conservação de recursos genéticos da espécie na região sul do Rio Grande do Sul. O DNA das cultivares foi extraído pelo método CTAB, e as reações de AFLP foram realizadas com os iniciadores EcoRI/MseI. Foram identificados dois grupos entre as 21 cultivares de marmeleiro, um com quatro e outro com sete cultivares geneticamente mais relacionadas. As cultivares de marmeleiro apresentam alta variabilidade genética, com máximo de 43% de similaridade.

Resumo O objetivo deste trabalho foi determinar os efeitos de diferentes tecnologias de armazenamento na qualidade físico-química de kiwis da cultivar Tewi. Após a colheita, parte dos frutos foi submetida ao tratamento com 1-metilciclopropeno (1-MCP) e, posteriormente, frutos tratados e não tratados foram submetidos a diferentes condições de armazenamento, conforme segue: armazenamento em temperatura ambiente (20 °C ± 0,5 °C) durante 12 dias; armazenamento refrigerado (0°C ± 0,5°C) com ou sem controle da atmosfera (3 KPa O2 e 5 KPa CO2), e com ou sem absorção de etileno durante dois e quatro meses. Foram avaliadas a firmeza de polpa, o teor de sólidos solúveis e a acidez titulável. Verificou-se que, independentemente da tecnologia de conservação, houve uma diminuição na firmeza de polpa já aos dois meses de armazenamento, sendo a condição atmosfera controlada associada ao uso do 1-MCP a mais eficaz na retenção desse atributo. Frutos mantidos em temperatura ambiente apresentaram relação direta entre a redução da firmeza de polpa e o aumento do índice de sólidos solúveis; no entanto, após o armazenamento, essa relação nem sempre ocorreu. A maior acidez titulável foi verificada em frutos que receberam 1-MCP. Kiwis ‘Tewi’ apresentam bom potencial de conservação, podendo permanecer armazenados por até quatro meses em atmosfera controlada associada ao controle do etileno pelo uso de 1-MCP ou por absorção do etileno.

Existem poucos trabalhos que avaliem a influência do manejo na qualidade do leite e, ainda, com resultados pouco conclusivos. Este trabalho teve como objetivo avaliar o efeito do manejo aversivo no perfil eletroforético qualitativo das caseínas do leite. Foi determinado o perfil eletroforético das caseínas do leite de vacas, da raça Holandês , submetidas aos tratamentos de manejo aversivo e manejo não aversivo, por um período de 24 dias. Ao final deste período de manejo, foram coletadas quatro amostras de leite de cada tratamento, com posterior centrifugação e liofilização. A técnica de dodecil-sulfato de sódio de poliacrilamida (SDS-PAGE) foi utilizada para migração eletroforética e, devido ao melhor resultado na visualização do gel apresentado por amostras submetidas ao processo de diálise, adotou-se este como procedimento padrão nas análises. Concluiu-se que o leite de vacas submetidas ao manejo aversivo não apresenta alteração do perfil eletroforético qualitativo das caseínas.

Due to the known difficulties on managing the design process of built environments, researchers and entrepreneurs have continuously developed and implemented best practices from areas such as Project Management (PM). However, there are still gaps to be filled, such as difficulties on monitoring the stakeholder’s requirements and their dynamics. In attempt to eliminate this gap, this paper aims to analyze if requirements management (RM) can be integrated to PM and which are the main advantages and disadvantages of this integration. The research includes a literature review which presents PM and RM concepts, the last one based on software engineering approach. Moreover, a case study was performed in a construction company, using semi-structured interviews. The research showed that the use of RM is advantageous, even not solving all the projects difficulties, and that the proper structuring of the projects is a condition for the success of RM implementation. The main contribution is a generic and systematic proposal for integrating the stages of the RM to the projects.

Proposal: As new demands from sustainable development, environmental requirements arise as another challenge to design process management. It is already known that companies which design buildings are usually exposed to many managerial difficulties. Faced to the environmental demands, these companies require new facilities to align environmental requirements to the business goals and to include them properly in design process. This paper is based on a case study in a construction company, which was developed through interviews and document analysis. It is intended to present a procedure for the project environmental requirements elicitation, organization and analysis, which is based on the requirements engineering (ER) concepts. As results it was concluded that the ER concepts are useful for the environmental requirements integration into the design process and that strategic planning should give directions for the effective environmental requirements adherence. Flesh softening, skin browning and rotting are chief problems during cold storage (CS) of 'Fuyu' Persimmon. We studied the effects of boron (B) and calcium (Ca) sprayed on the trees during three consecutive years, on the development of skin cracks, grooves and browning in persimmon fruit under CS in Farroupilha, RS, Brazil (29°31' south, 51°21' west, about 750 m altitude). A homogeneous orchard area of 0.5 ha was delimitated and three sets of five plants for each treatment were randomly selected. The persimmon trees were sprayed at a 20 day interval, from 15th January until harvest, for three consecutive years, with: T1) water; T2) calcium nitrate at 0.5% (m/v); T3) calcium chloride at 0.5% (m/v); and T4) boron at 0.3% (m/v). The fruit were harvest with orange-reddish colour; 18-20°Brix, pulp firmness of 45 to 60N, and kept under CS at 0±1°C for 45 days. The fruits were evaluated immediately before CS, after six hours at 23±2°C after removal from CS, and after four days at 23±2°C after removal from CS. Equally boron and calcium sprayed on the trees prevented skin cracks, skin grooves and skin browning. Besides, when boron was sprayed on the trees, the mentioned effects were additive in the following year.As principais alterações indesejáveis observadas no período pós-colheita de caquis em armazenamento refrigerado (AR) são a perda de firmeza de polpa, e a ocorrência de escurecimento epidérmico e de podridões. Este trabalho teve por objetivo principal estudar as respostas da aplicação de boro (B) e cálcio (Ca), em três safras sucessivas, na prevenção da ocorrência de fissuras, estrias e escurecimento epidérmico de caquis "Fuyu". No pomar (Farroupilha-RS-Brasil, 29°31' Sul, 51°21' Oeste, aproximadamente 750m altitude) delimitou-se uma área homogênea de 0,5ha., marcando-se, ao acaso, três repetições de cinco plantas para cada tratamento: T1 - controle, caquizeiros não-pulverizados com B e Ca; T2 - pulverizações com Nitrato de Ca 0,5% (m/v); T3 - pulverizações com Cloreto de Ca 0,5% (m/v); e T4 - pulverizações com B 0,3% (m/v). As aplicações foram realizadas em três anos sucessivos a partir de 15 de janeiro a intervalos de 20 dias. Foram colhidos os caquis na coloração alaranjado-avermelhada, com 18-20°Brix e 45 a 60N, que posteriormente foram armazenados em AR a 0±1°C por 45 dias. As avaliações foram realizadas no dia do armazenamento, após seis horas a 23±2°C a partir da retirada AR, e após quatro dias a 23±2°C a partir da retirada do AR. Foi avaliada a ocorrência de frutas com fissuras, estrias e escurecimento epidérmico. Comprovou-se o efeito benéfico da aplicação de B ou Ca na prevenção de fissuras, estrias e escurecimento epidérmico. Adicionalmente, foi detectado o incremento da resposta à aplicação de B, que, além de prevenir (à semelhança das aplicações com Ca), apresentou efeito aditivo no segundo ano. O resultado sugere que as respostas às aplicações com B são progressivas.

Reverse Transcription quantitative PCR (RT-qPCR) is one of the most important techniques for gene expression profiling due to its high sensibility and reproducibility. However, the reliability of the results is highly dependent on data normalization, performed by comparisons between the expression profiles of the genes of interest against those of constitutively expressed, reference genes. Although the technique is widely used in fruit postharvest experiments, the transcription stability of reference genes has not been thoroughly investigated under these experimental conditions. Thus, we have determined the transcriptional profile, under these conditions, of three genes commonly used as reference--ACTIN (MdACT), PROTEIN DISULPHIDE ISOMERASE (MdPDI) and UBIQUITIN-CONJUGATING ENZYME E2 (MdUBC)--along with two novel candidates--HISTONE 1 (MdH1) and NUCLEOSSOME ASSEMBLY 1 PROTEIN (MdNAP1). The expression profile of the genes was investigated throughout five experiments, with three of them encompassing the postharvest period and the other two, consisting of developmental and spatial phases. The transcriptional stability was comparatively investigated using four distinct software packages: BestKeeper, NormFinder, geNorm and DataAssist. Gene ranking results for transcriptional stability were similar for the investigated software packages, with the exception of BestKeeper. The classic reference gene MdUBC ranked among the most stably transcribed in all investigated experimental conditions. Transcript accumulation profiles for the novel reference candidate gene MdH1 were stable throughout the tested conditions, especially in experiments encompassing the postharvest period. Thus, our results present a novel reference gene for postharvest experiments in apple and reinforce the importance of checking the transcription profile of reference genes under the experimental conditions of interest.

Esse trabalho objetivou verificar a conservação de tangerinas ‘Clemenules’, ‘Marisol’ e ‘Nova’ minimamente processadas. As frutas foram selecionadas, descascadas e seus gomos separados individualmente. No armazenamento, foram embaladas em bandejas,recobertas com filme de polietileno (60 mm), por 6 dias a 5±1°C e 85±5% de umidade relativa (UR), mantidas para comercialização simulada (CS) de um ou de dois dias a 10±1°C e 90±5% de UR. Realizaram-se análises no armazenamento e no 6º dia e também noperíodo de CS, através dos parâmetros: perda de peso (PP), sólidos solúveis totais (SST), acidez total titulável (ATT), relação ATT/SST, pH, ácido ascórbico e ocorrência de podridões. A PP foi maior aos seis mais dois dias de CS. O pH, os SST e o ácido ascórbico apresentaram decréscimo progressivo com a evolução do armazenamento. As tangerinas ‘Clemenules’, ‘Marisol’ e ‘Nova’ minimamente processadas mantiveram-se com qualidade adequada para o consumo, por 6 dias de armazenamento, mais dois dias de CS.This work was objective to verify the conservation of minimally processed ‘Clemenules’, ‘Marisol', and ‘Nova’ mandarins’ fruit. The fruits were selected, peeled and the buds had been broken individually. At the stored were packed in trays, covered with polyethylene film (60 mm), for six days at 5±1°C, and relative humidity (RH) 85±5%, plus simulated commercialization (SC) of one or two days at 10±1°C and RH 90±5%. The analysis has been developed during the storage and on the six days and also in SC period, considering the parameters: weight loss (WL), total soluble solids (TSS), total titratable acidity (TTA), TTA/TSS, pH, acid ascorbic, and rottenness. The WL was higher on the six more two days of SC. The pH, TSS, and the acid ascorbic presented gradual decrease with the storage period. The minimally processed ‘Clemenules’, ‘Marisol', and ‘Nova’ mandarins fruit maintained adequate quality to the consumption, after six days of storage plus two days of SC.
