We introduce an NMR method to help in the analysis of complex mixtures. The spectra of molecular fragments are obtained as the traces of a correlation spectrum of the regular [sup.1]H NMR spectrum on one dimension with the one of the highest possible [sup.1]H multiple-quantum (MaxQ) order. As this latter is a function of the number of distinguishable protons in a given molecular fragment, the analysis of a series of multiple-quantum spectra is required to achieve a complete assignment. This MaxQ NMR approach is likely to perform best in the case of signals concentrated in a very narrow frequency range, which is a challenging situation commonly encountered in many relevant analytical problems such as the characterization of extraction fractions (oil, plants, tissues), biological fluids, or environmentally relevant samples. As a demonstration, we apply the MaxQ NMR analysis to a mixture of 11 poly- and monocyclic aromatic hydrocarbons. 10.1021/ac100009y

The diffusional properties of molecules in solution vary dramatically upon addition of a solid chromatographic phase. This effect can be monitored via pulsed-gradient spin-echo NMR used in conjunction with moderately fast rotation of the sample (high-resolution magic angle spinning) to produce exploitable spectra. The molecular diffusion coefficients observed in this condition are averages reflecting the equilibrium population distribution among the different phases. It is thus possible to use this information for investigating a crucial step of reversed-phase chromatography, namely, the partitioning of the analyte between different phases. In this work, we describe the evolution of the apparent diffusion coefficient of typical solutes for water/acetonitrile solvent mixtures of varying proportions.

Research is done on single-file diffusion behavior, which is expected for atoms and molecules in one-dimensional gas phases of nanochannels with transverse dimensions that do not allow for the particles to bypass each other. In conclusion, using continuous-flow laser-polarized 129Xe NMR spectroscopy, an intriguing correlation is described between the observed NMR line shape of xenon within the nanochannels and the gas transport in the channels.

A study was conducted to determine the feasibility of using deuterium-carbon nuclear magnetic resonance correlation spectroscopy to measure the deuterium quadrupolar couplings in liquid crystals. A fully deuterated nematic 4-n-pentyl-4'-cyanobiphenyl was used since it is a well-characterized model of thermotropic liquid crystal. Results indicated that the method is effective since it can overcome overlap problems.

A novel approach to selective correlation spectroscopy which offers increased sensitivity is presented. It is applied to a small molecule with nonlinear optical properties. It is likewise demonstrated by the determination of scalar coupling constants in the peptide gramicidin and in a 57 amino acid fragment of the nucleocapsid of the hepatitis C virus. Improvements in sensitivity between a factor of 2 and 6 were noted in these samples.

International audience; We illustrate here as the combination of high-order maximum-quantum (MaxQ) and Diffusion-Ordered SpectroscopY (DOSY) NMR experiments in a 3D layout allows superior resolution for crowded NMR spectra. Non-uniform sampling (NUS) allows compressing the experimental time effectively to reasonable durations. Because diffusion effects were encoded within multiple-quantum co-herences, increased sensitivity to magnetic field gradients is observed, requiring compensation for convection effects. The experiment was demonstrated on the spectra of a mix of small polyaromatic molecules. Specifically, in the case analyzed, the experiment provided an extreme simplification through the MaxQDOSY-MaxQ projection plane that presents one peak per molecule.

A systematic study is carried out on cationic and anionic substitution in hydroxyapatite structures in order to understand the impact of the ion exchange on the crystalline structure and properties of these materials. The existence of a wide variety of environments for OH in substituted apatites is shown by [super 1]H NMR and the mobility of the water molecules has changed upon introduction of lead into the structure.

Experimental and theoretical aspects of carbon-13 line shapes in static solids are described for on-resonance spin decoupling conditions. A relatively simple theoretical approach is provided for describing line shapes in static solids based on an operator representation of static second-order perturbation theory and theoretical line shapes in I2S and InS systems are calculated. The line shapes are predicted to comprise a single center line and ‘‘decoupling sidebands’’ on each side of the center line which move outward and diminish in amplitude as the decoupling field is increased. The predicted behavior is confirmed by experiments on an isolated seven spin system, where the decoupling sidebands are observed directly, and some organic solids in which the decoupling sidebands are not observed directly but in which their presence can be deduced from the behavior of the center line. A comparison is made between the theoretical predictions based on a complete quantum mechanical treatment and the predictions made using classical approximations in the model for the line shape. We conclude, based on our experimental results, that the line shape has a character which reflects the quantum nature of the spin system, even in organic solids, and that on-resonance terms appear to dominate experimental line shapes. © 1996 American Institute of Physics. The triclinic form of AlPO4-34, a microporous aluminophosphate with the chabazite (CHA) topology, adopts a rhombohedral symmetry upon calcination. Following these structural changes in situ by X-ray diffraction revealed that there are actually two stable rehydrated phases, which differ from each other by one water molecule in the channel.

OBJECTIVE: To evaluate the usefulness of a nested polymerase chain reaction (PCR) for Mycobacterium tuberculosis complex on routinely stained cytologic samples from patients with extrapulmonary tuberculosis. STUDY DESIGN: Nested PCR for the detection of a fragment of the IS6110 insertion sequence of M tuberculosis complex was applied to Ziehl-Neelsen-negative archival cytologic slides of serous effusions (pleural [n = 7], peritoneal [n = 1] and pericardial [n = 1]) and a lymph node fine needle aspirate (n = 1) from nine human immunodeficiency virus (HIV)-positive patients with autopsy-proven active extrapulmonary tuberculosis. Malignant effusions and aspirates from nine HIV-positive patients with non-Hodgkin's lymphoma and pleural effusions from seven HIV-negative patients with heart failure were used as controls. DNA was extracted after removing the coverslip and gently scraping the cytologic sample from the slides. RESULTS: In all cases, enough DNA was obtained for PCR without any significant loss of integrity, as demonstrated by PCR positive for HLA-Dq. PCR for M tuberculosis was positive in 8 of the 10 samples (80%) from patients with tuberculosis but also in three samples (30%) from HIV-positive patients in the control group. None of the samples from the HIV-negative patients was positive. CONCLUSION: PCR for M tuberculosis can be reliably performed on archival cytologic slides from extrapulmonary samples, but although it is highly sensitive, it may lead to positive results in immunocompromised patients without any sign of active tubercular disease.

Pleomorphic xanthoastrocytoma (PXA) is a rare cerebral tumor of young adults with a slow growth and a good prognosis. Due to its peculiar histopathological findings, the tumor resemble to the lytic phase of progressive multifocal leukoencephalopathy (PML), a JC Virus (JCV) induced disease. For these reasons, the presence of JCV genoma and viral particles were searched for by means of nested polymerase chain reaction (nPCR) and electron microscopy (EM) in a 9-year-old child with PXA. Although EM did not reveal any viral particles, nPCR did reveal genomic sequences of the LT, R, and VP1 regions of JCV. Sequence analysis showed that the R region was mutated with respect to the archetypal form thus yielding the Mad 4 variant of JCV previously reported as being oncogenic in animals. We suggest that JCV may have played a role in the development of this tumor.
