Per- and polyfluoroalkyl substances (PFAS) are a large group of persistent industrial chemicals that can harm reproductive health. PFAS levels were analysed to determine the current sources of exposure and possible associations between prenatal PFAS exposure and adverse pregnancy outcome. Samples from 136 mother-newborn pairs recruited between 2017 and 2019 were analysed for the presence of 31 target PFAS in maternal serum, umbilical cord serum, and placental tissue by high-performance liquid chromatography coupled to a tandem mass spectrometer. Questionnaires and medical records were used to survey sources of exposure and pregnancy outcome, including small for gestational age (SGA), fetal growth restriction (FGR), preeclampsia (PE), preterm birth, large for gestational age (LGA) and gestational diabetes mellitus (GDM). Data were analysed for individual PFAS and sum4PFAS (sum of perfluorooctanoic acid (PFOA), perfluorononanoic acid (PFNA), perfluorohexane sulfonate (PFHxS), and perfluorooctane sulfonate (PFOS) serum levels) in logistic regression analyses and categorical regression analyses. Compared to data from a previous Viennese study in 2010–12, sum4PFAS levels were generally lower. Sum4PFAS serum levels of three women (2.2%) exceeded 6.9 µg/L, a level that corresponds to the recently established tolerable weekly intake (TWI) of EFSA for nursing mothers aged 35 years; in the 2010/2012 study it was 13.6%. The large contribution of unidentified extractable organofluorine (EOF) fractions to total PFAS exposure is a concern. Study site, mean maternal corpuscular hemoglobin (MCH), use of facial lotion, and owning upholstered furniture were significantly influencing maternal exposure. While no effect of sum4PFAS on pregnancy outcome could be detected, we found highest placental PFDA levels in SGA births. PFHxS levels in umbilical cord and placenta were highest in preterm births. Further studies are needed to elucidate the relationship of prenatal PFAS exposure and pregnancy outcome, in particular to confirm whether and how placental PFDA levels may contribute to an increased risk for SGA.

Dioxins and furans (PCDD/Fs) are anthropogenic pollutants that persist in the environment for long years, bioaccumulating in food & contaminating humans. In pregnancy, they can transfer through the placenta and reach the fetus, which negatively affects fetal growth. They can also reach newborns through breastfeeding. In this study, we focused on this critical subpopulation and identified the presence of PCDD/Fs among pregnant women in breast milk (n = 41) and cord serum (n = 49); we assessed the correlation between different matrices, evaluated the predictors and associations with newborn anthropometric measurements. Over 70.7 % of PCDD/Fs were detected in breast milk and 46.9–55.1 % in cord serum. Cord/maternal serum and breast milk to maternal serum ratios were > 1 with a significant positive Spearman correlation (0.669–0.729). Breast milk & maternal serum PCDD/Fs were associated inversely with age and positively with red meat intake. Cord serum PCDD/Fs were inversely associated with pre-pregnancy weight loss and passive smoking. Parity and gestational weight gain showed positive associations with Z-scores at birth. Z -score differences showed negative and positive associations with passive smoking and pre-pregnancy BMI respectively. Aims/hypothesis: Both arachidonic acid (AA, 20:4 n-6) and docosahexaenoic acid (DHA,22:6 n-3), long-chain polyunsaturated fatty acids (LCPUFA), are involved in fetal development and, based on their percentage compositions, appear to be specifically accumulated in fetal circulation in a proposed phenomenon known as biomagnification. Discrepancies exist in the literature concerning the effect of gestational diabetes mellitus (GDM) on circulating fatty acids. Our objective was to analyse individual fatty acid concentrations in a large cohort of maternal and cord paired serum samples from pregnant women with and without GDM. Methods: Overnight fasted maternal and cord blood paired samples from 84 women with GDM and well controlled blood glucose levels and 90 healthy pregnant women (controls) were drawn at term. Individual fatty acids within total serum lipids were analysed by gas chromatography and expressed both as concentrations of fatty acid (mmol/l) and as a percentage of total fatty acids. Results: In the serum of overnight fasted pregnant women with GDM, the concentrations of most fatty acids were lower than in control women, except for AA and DHA, which remained the same. The concentrations of most fatty acids in cord serum were also lower in the GDM group than in the control group, except for α-linolenic acid (ALA,18:3 n-3), which was higher in the GDM group. In both groups, the concentrations of all fatty acids were lower in cord serum than in maternal serum. In GDM participants only, a positive and significant correlation between cord and maternal serum concentration of AA and DHA was observed. Conclusions/interpretation: The expression of fatty acids in molar concentrations reveals that GDM decreases the concentration of most fatty acids in both maternal and cord serum. Abstract As endocrine disrupting chemicals, hexachlorocyclohexane (HCH) isomers were reported to impair the intrauterine growth. Although the findings of HCHs with preterm birth were well established, the associations with gestational age were limited. In the present study, we examined whether exposure to HCHs would influence gestational age. The study population included 1028 pregnant women and their offspring who were born in 2014–2015 from a birth cohort in Wuhan, China. Associations of the cord serum HCH levels with gestational age were estimated using generalized linear models. We found higher HCH levels in pregnant women, who were elder, had higher body mass index (BMI) before pregnancy, received higher education, or were exposed to smoking passively. For term birth, the 3rd tertiles of α-HCH and γ-HCH were significantly associated with shorter gestational age [crude β = −1.017, confidence interval (CI): − 2.017, − 0.018 for α-HCH, crude β = −1.068, CI: − 2.067, − 0.070 for γ-HCH], and relationships were similar after adjusted by covariates. Stratified analysis showed positive associations between α-HCH and gestational age for mothers younger than 25 years old (adjusted β = 0.610, CI: 0.061, 1.158), while showing negative relationships for mothers elder than 35 years old (adjusted β = −1.365, CI: −2.414, −0.317). In summary, our results indicated cord serum levels of HCHs were associated with gestational age at birth. Abstract There has been substantial evidence showing the adverse effects of organochlorine pesticide (OCP) exposure on human, but studies focused on the prenatal exposure effects at low OCP levels on infant birth size were scarce and controversial. In this study, cord serum samples were collected at the delivery from 1028 pairs of mothers and newborns in Wuhan, China and investigated the associations of prenatal exposure to OCPs and birth size. The prenatal exposure of hexachlorocyclohexane isomers (HCHs), p,p′ -dichlorodiphenyltrichloroethane (p,p′ -DDT) and its metabolites were analyzed. The associations between birth size and prenatal OCP exposure were examined by multiple linear regressions. A sex-specific relationship between the OCP exposure and birth size was observed. β-HCH was negatively associated with birth weight and ponderal index for boys [adjusted β = −28.61; 95% confidence interval (CI): −54.84, −4.37 and adjusted β = −0.17; 95% CI: −0.32, −0.01, respectively], whilst no significant associations with prenatal exposure of OCPs were found among girls. The inverse association of prenatal exposure to low levels of β-HCH was shown sex-specific difference, which was only observed significantly in boys. The findings strengthened the evidence that the fetal development was influenced by prenatal exposure to certain OCPs and the effects might be different in the newborn sex. Organochlorine pesticides (OCPs) had been widely used in China before they were banned decades ago due to the concern of the toxicology to human. Previous reports showed that OCPs were still often detectable in human bodies. However, there is limited study concerning the body burden of OCPs in infants and fetus in China. In this study, the prenatal exposure to OCPs was evaluated by measuring OCPs in cord serum. A total of 1046 cord serum samples were collected in Wuhan during 2014 and 2015, and analyzed for the concentrations of hexachlorocyclohexanes (HCHs), dichlorodiphenyltrichloroethanes (DDTs), dichlorodiphenyldichloroethanes (DDDs) and dichlorodiphenyldichloroethylenes (DDEs) by using gas chromatography-tandem mass spectrometry (GC–MS/MS). Perfluoroalkyl substances (PFASs) are a class of widely used chemicals that have been detected in the environment and general population. However, the isomer patterns in human are poorly characterized. Previous studies observed the isomer-specific maternal-fetal transfer of PFASs in human. In our current study, we first examined the profile of PFASs and isomers, including 17 linear PFASs and 10 branched PFOS/PFOA isomers by using isotopic internal standards in umbilical cord serum samples from Guangzhou, China. We collected a total of 321 of cord blood serum samples from July to October in 2013, and analyzed the PFASs concentration with isomer-specific PFASs analysis method. The results showed that 9 out of 17 PFASs (linear PFASs) were detected (>50% detection rate). Perfluorohexane sulfonate (PFHxS, median 3.87 ng/mL) was the predominant, followed by total PFOS (median 2.99 ng/mL) and total PFOA (median 1.23 ng/mL) in cord serum. In addition, 1m-, iso-, ∑3+4+5m-PFOS and iso-PFOA were the branched PFASs detected in the current study. The proportion of linear PFOS (n-PFOS) was 75.16% of ∑PFOS which was similar to the proportion of electrochemical fluorination that produces ca. 70% linear PFOS and 30% branched. On the contrary, linear PFOA (n-PFOA) accounted for 98.69% ∑PFOA in cord serum samples. Our finding indicates distinct PFASs and PFOS/PFOA isomer profile in cord serum, suggesting there might be a different exposure pathway and metabolism of PFASs. [ABSTRACT FROM AUTHOR]

Previous in vivo studies have suggested that prenatal exposure to dichlorodiphenyltrichloroethane (DDT) is endocrine disrupting, and may cause adverse health risks to newborns. In this cross-sectional study, non-invasive cord serum samples were collected from maternal-neonate pairs of an island population. Concentrations of DDT and its metabolites were analyzed to provide insights into the environmental exposure patterns and to elucidate their effects on birth outcomes. The average concentrations of p , p ’-DDE, o , p ’-DDD, p , p ’-DDD, o , p ’-DDT, p , p ’-DDT and total DDTs were 0.463, 0.293, 0.089, 0.098, 0.441 and 1.384 μg g − 1 lipid weight, respectively. Several maternal characteristics were identified as influencing factors on the environmental exposure distribution. The isomer ratios of individual components and the enantiomeric patterns of o , p ’-DDD and o , p ’-DDT indicated that historical technical DDT remains the predominant exposure source, though new sources of dicofol-type of DDT pollution must not be ignored. Using multivariable linear regression, increasing p , p ’-DDD and p , p ’-DDT levels were found to be significantly associated with an increase in neonatal birth weight, which deserves additional attention to obesity risks. No other birth outcome was found to be significant regarding DDT exposure. These findings raise the awareness of the prenatal risk of DDT and its metabolites among infants in contaminated areas. Perfluoroalkyl substances (PFASs), a group of environmental pollutants, persistently exist in the environment. To investigate the associations between PFASs levels in cord serum and birth weight, birth length and ponderal index, we measured PFASs in cord serum samples from 170 infants from Feb. 2012 to Jun. 2012 in Beijing, China. The mean concentrations in cord serum samples for perfluorooctanoic acid (PFOA), perfluorooctane sulfonic acid (PFOS), perfluorohexane sulfonic acid (PFHxS), perfluorononanoic acid (PFNA), perfluorodecanoic acid (PFDA) and perfluoroundecanoic acid (PFUnA) were 1.285 ng/mL, 1.228 ng/mL, 0.230 ng/mL, 0.224 ng/mL, 0.100 ng/mL and 0.085 ng/mL, respectively. First-born children had slightly higher exposure levels of PFHxS (p < 0.001) and PFOA (p = 0.03) than second-born or third-born children. The spearman correlation coefficients with gestation time were individually 0.160 (p = 0.038) for PFHxS and 0.202 (p = 0.008) for PFOA. Both univariate and multivariate linear regression analysis showed that the exposure levels of PFASs had no statistically significant associations with birth weight, birth length or ponderal index in the present population. The demonstrated developmental and reproductive toxicity of perfluorooctanoic acid (PFOA) and perfluorooctane sulfonate (PFOS), coupled with the increasing production and use of emerging per- and polyfluoroalkyl substances (PFASs) has resulted in progressively higher human exposure levels. This has raised concerns about PFAS exposure levels in the fetus, which is highly susceptible to the potential effects of hazardous environmental chemicals. However, in utero exposure to PFASs and health implications have not been fully characterized in China. To fill this gap, we analyzed 19 PFASs in umbilical cord serum samples (n = 66). Information about the mothers and newborns was obtained through questionnaires. Associations between maternal characteristics and neonatal birth weight and PFAS concentrations were analyzed using nonparametric tests. As results, PFOA was detected in all serum samples. The highest median concentration of PFOS in umbilical serum was 1.092 ng·mL-1, followed by perfluoropentanoic acid (median: 0.633 ng·mL-1). Trifluroacetic acid and perfluoropropanoic acid were detected in cord serum for the first time, and their median concentrations were 0.229 and 0.266 ng·mL-1, respectively. Neonatal birth weight was negatively correlated with long-chain PFOS (r = −0.319, P < 0.05), and the concentrations of perfluoroundecanoic acid and perfluorododecanoic acid were significantly different between the birth weight groups. Maternal age, maternal education, diet, and nutritional supplementation during pregnancy can all affect umbilical serum exposure to PFASs. These results demonstrate that legacy PFASs remain major contributors to the composition of human PFASs, while the concentration levels of emerging short-chain alternatives have increased significantly. Modifying the mother's diet may reduce the risk of intrauterine PFAS exposure. Special attention to exposure to highly novel PFASs and confirmation of potential determinants should be taken as a priority in the plan for risk management and actions in this area. Per- and polyfluoroalkyl substances (PFAS), a kind of emerging environmental endocrine disruptors, may interfere with the secretion of adipokines and affect fetal metabolic function and intrauterine development. However, the epidemiological evidence is limited and inconsistent. We examined the associations of single and multiple PFAS exposures in utero with adipocytokine concentrations in umbilical cord serum. This study included 1111 mother-infant pairs from Sheyang Mini Birth Cohort Study (SMBCS), and quantified 12 PFAS and two adipokine in umbilical cord serum. Generalized linear models (GLMs) and Bayesian Kernel Machine Regression (BKMR) models were applied to estimate the associations of single- and mixed- PFAS exposure with adipokines, respectively. Furthermore, sex-stratification was done in each model to assess the sexually dimorphic effects of PFAS. 10 PFAS were detected with median concentrations (μg/L) ranging from 0.04 to 3.97, (except 2.7% for PFOSA and 1.7% for PFDS, which were excluded). In GLMs, for each doubling increase in PFBS, PFHpA, PFHxS, PFHpS, PFUnDA and PFDoDA, leptin decreased between 14.04% for PFBS and 22.69% for PFHpS (P < 0.05). PFAS, except for PFNA, were positively associated with adiponectin, and for each doubling of PFAS, adiponectin increased between 3.27% for PFBS and 12.28% for PFHxS (P < 0.05). In addition, infant gender modified the associations of PFAS with adipokines, especially the associations of PFBS, PFOA and PFHxS with adiponectin. Similarly, significant associations of PFAS mixtures with leptin and adiponectin were observed in the BKMR models. PFDA, PFOS, PFNA and PFHpS were identified as important contributors. In the sex-stratified analysis of BKMR models, the associations between PFAS mixtures and adipokines were more pronounced in males. PFAS levels were significantly associated with adipokines in cord serum, suggesting that intrauterine mixture of PFAS exposure may be related to decreased fetal leptin level but increased fetal adiponectin level and the associations may be sex-specific. The effects of exposure to the perfluoroalkyl and polyfluoroalkyl substances (PFASs) perfluorooctane sulfonate (PFOS) and perfluorooctanoic acid (PFOA) on birth weight have been examined in several studies, but other PFASs have not been considered. We conducted a cross-sectional survey of newborns in Seoul, South Korea, collecting 118 serum samples, for 85 of which we had a full range of information. We conducted multiple regression analyses to examine the association between nine PFAS concentrations in cord serum and birth weight. Seven PFASs were found in cord serum, PFOA and PFOS being dominant, with mean concentrations of 1.11 and 0.87 ng/mL, respectively. The adjusted birth weight changes (natural log) were −0.14 (95% confidence interval −0.33–0.03) for PFOS and −0.03 (95% confidence interval −0.25–0.18) for PFOA. A study of iron, zinc, copper and selenium concentration levels was carried out in three compartments namely, maternal serum (MS), colostrums and cord blood serum (CS) of healthy Indian mothers (n = 42) who delivered healthy normal neonates without any congenital anomalies at Bhabha Atomic Research Centre hospital, Mumbai. Fe, Zn, Cu in maternal serum, cord blood and colostrums were estimated by flame atomic absorption spectrometry while Se was determined by graphite furnace absorption spectrometry. It was seen that there was a significant difference in the level of trace elements in the three compartments. The average levels of Fe in the three compartments were 1,132 ± 519, 2,312 ± 789 and 1,183 ± 602 μg/L while Zn was 514 ± 149, 819 ± 224 and 7,148 ± 2,316 μg/L respectively. Mean Cu values were 1,614 ± 295, 301 ± 77 and 392 ± 174 μg/L respectively while Se values were 70 ± 15, 36 ± 10 and 23 ± 8 μg/L respectively. The results indicated a positive correlation of Fe and Zn concentrations in MS versus CS which were (r = 0.386), (r = 0.572) respectively and Fe levels in MS and colostrums (r = 0.235). A few inter element correlations were found within compartments. Zn and Se showed a negative correlation in both MS (r = −0.489) and colostrums (r = −0.258) while a positive inter correlation of Fe and Zn was seen in MS (r = 0.44) and in CS (r = 0.54). Abstract: The process of the growth of the fetus begins in the uterus and gets further accelerated following the birth, especially during initial few months. The role of the growth factors in the physiology of the cellular growth is already well established. Vascular endothelial growth factor (VEGF) and hepatocyte growth factor (HGF) seem to be imperative for angiogenesis, cell development and proliferation as well as maintenance of the tissues. The levels of these factors in the maternal serum during pregnancy as well as during postpartum period are insignificant. Consequently, we hypothesized that the fetus receives moderate supply of these growth factors from the placenta during its stay in the uterus. This supply gets further augmented during the postpartum period through the different source, i.e. mother’s milk. To study this physiological transition of the source of the growth factors from the placenta to the breast milk, the concentrations of VEGF and HGF in the cord serum of full term neonates and that in the breast milk of the corresponding mothers were analyzed during ELISA. The human milk, especially the colostrum revealed significantly higher levels of VEGF and HGF (1541.759±119.349pg/ml and 7129.249±273.472pg/ml) than cord serum (16.632±0.773pg/ml and 2581.6±108.275pg/ml) respectively. The multifold higher levels of VEGF observed in colostrum probably correlates with its high neonatal requirement for the maturation of the gastrointestinal epithelium following birth. The higher levels of both the growth factors in the breast milk than those observed in the cord serum probably explain their higher needs by the neonates for immunological protection, protein synthesis and neurocognitive development. The observations of the present study strengthen the policy of the colostrum feeding, which is promoted by organizations like World Health Organization (WHO). Abstract: Previous studies of in utero exposure to dichlorodiphenyltrichloroethane (DDT) have shown mixed results for the harmful effects on fetal growth and development. This cross-sectional study was designed to: (1) examine the extent of DDT exposure in 1578 women, aged 28.5±6.0 who delivered between June 2005 and 2006 in the area of Al-Kharj, Saudi Arabia; and (2) assess its influence on neonatal anthropometric measurement of newly born babies. DDT and its metabolites, namely 1,1-dichloro-2,2-bis (p-chlorophenyl) ethylene (p,p′-DDE), 1,1-dichloro-2,2-bis (p-chlorophenyl) ethane (p,p′-DDD) and 1,1,1-trichloro-2,2′ bis (p-chlorophenyl) ethane (p,p′-DDT) were measured in cord and maternal serum as well as placenta by Gas Chromatography coupled with an Electron Capture Detector (GC/ECD). p,p′-DDE was detected in 28.3% of cord and 54.4% of maternal serum, reflecting past exposure, whereas the p,p′-DDT was only found in 0.46% cord and 1.2% maternal samples. As expected the p,p′-DDE cord levels (0.197±0.961μg/L) were 2.8 times lower than the maternal levels (0.551±1.778μg/L), and both were significantly correlated (r =0.517) suggesting its transplacental transfer. The picture was different in placental tissues. p,p′-DDE and p,p′-DDT were detected in 84% and 99% of placental tissues, with the highest p,p′-DDT in placental tissues (29.62±158.282µg/kg dry wt.) compare to p,p′-DDE (10.167±18.851μg/kg dry wt.). In general, the presence of DDT metabolites in our participants indicates that these chemicals are still relevant despite the fact that they have been banned or restricted in the study area for many years. Our results support the view for an association between low in utero exposure to DDT and the anthropometric development of the fetus leading to a reduction in its head circumference, crown–heel length, birth weight and birth height. Since the reduction in these measures was independent of gestational age and/or preterm births, our findings suggest a detrimental effect of the DDT exposure on fetal growth. Neonatal anthropometric measures are considered as an important tool to detect newborns at higher risk of morbidity and impairment of growth. Efforts should be made to decrease exposure of women of reproductive age and to examine maternal DDT exposure in relation to long-term impact on health. [Copyright &y& Elsevier]

Background/Aims: Retinol-binding protein 4 (RBP4) has been shown to be associated with insulin resistance and fatty acid metabolism. We hypothesize that RBP4 might play a role in fetal growth and that cord serum RBP4 may act as a marker of fetal growth, independent of fetal insulin levels. Methods: Twenty-one women having fetuses in the top quartile (>75th percentile) of birth weights for gestational age were enrolled into the trial, along with 21 women having fetuses in the bottom quartile (<25th percentile) of birth weights for gestational age. Serum RBP4 and insulin levels were analyzed. Results: Cord serum RBP4 and insulin concentrations were significantly higher in the top quartile group (14.3 ± 3.7 ng/ml, 3.8 ± 5.2 μIU/ml) than in the bottom quartile group (11.3 ± 2.6 ng/ml, 0.9 ± 1.4 μIU/ml; p = 0.004, p = 0.017). Cord serum RBP4 and insulin as well as gestational age (r = 0.744, r2 = 0.553, p < 0.001) were significantly correlated with fetal birth weights. Conclusion: Cord serum RBP4 concentrations were higher in subjects with top quartile fetuses than in those with bottom quartile fetuses. Cord serum RBP4 concentrations were significantly correlated with fetal birth weight. These findings may indicate that cord serum RBP4 plays a regulatory role in fetal growth. Copyright © 2011 S. Abstract: Polybromodiphenyl ethers (PBDEs) were analyzed in blood serum of pregnant women and in cord blood serum of their newborns from a general population cohort (n=174; Valencia, Spain). The most abundant PBDE congeners identified were BDE 47, BDE 99, BDE 153, BDE 154 and BDE 209. Their cord blood serum concentrations were about 45% of those in maternal serum but after lipid normalization about the same concentrations were observed in both types of samples. Thus, median of total PBDEs was 9.6ng/g lipid in cord serum (range between not detected and 140ng/g lipid) and 9.6ng/g lipid in maternal serum (range between not detected and 120ng/g lipid). The distributions of these compounds were dominated by BDE 47 in both cases. In cord blood serum the decreasing order of abundance was BDE 47>BDE 99>BDE 209>BDE 153>BDE 154. The congener composition in maternal serum followed a similar trend: BDE 47>BDE 153>BDE 154>BDE 209>BDE 99. The congener concentrations exhibited a higher degree of correlation in cord blood than in maternal serum. Use of the maternal determinants for categorization of the observed maternal and fetal PBDE concentrations only showed significant associations for the levels in umbilical cord. Neonates from rural areas exhibited statistically significantly lower concentrations than those from urban, semi-urban or metropolitan sites. Maternal serum also showed this difference but the higher dispersion of the concentrations in maternal serum did not afford its recognition with statistical significance. The lower qualitative and quantitative variability in the PBDE concentrations of cord blood serum than maternal serum suggest that the latter is reflecting PBDE contributions from a wider diversity of sources than the former whereas cord blood sera seem to represent the long term standing stock of these compounds accumulated in the maternal tissues. Abstract: This study is one of the very few investigating the body burdens of persistent organic pollutants in residents of Hong Kong. Twenty-nine human milk samples and 21 human blood (and cord blood) samples collected from 2005 were analyzed for PAHs, OCPs and PCBs levels. Higher levels of PAHs, DDTs and PCBs were detected in human milk samples when compared to maternal serum and cord serum (PAHs: milk: 1981ngg−1 fat, maternal serum: 1461, cord serum: 1158; DDTs: 3099, 1934, 1556; PCBs: 49, 41, 40). Among the 16 PAHs, naphthalene (human milk: 786ngg−1 fat, maternal serum: 331, cord serum: 348), phenanthrene (361, 144, 193), pyrene (187, 154, 98) and fluoranthene (158, 128, 89) were the major PAHs detected in three human tissues. p,p''-DDE and p,p''-DDT were the only metabolites of DDT detected in the three types of human tissues. High detection rate of the p,p''-DDE and p,p''-DDT (>90%) were noted in the three types of human samples. On the contrary, low detection frequency of PCBs in human milk (10.7%), maternal serum (1.7%) and cord serum (0.8%) were observed. The correlation coefficients of the PAHs, DDTs and PCBs levels in the three types of human tissues together with fish consumption, maternal age and tissue fat were analyzed. The following significant correlations were observed: Σ DDTs and p,p''-DDE in human milk with consumption of freshwater and marine fish, and maternal age; Σ PCBs in human milk with marine fish consumption, and maternal age; Σ PAHs in human milk with maternal age, respectively. The estimated daily intakes of DDTs by infants indicated that 7 out of 29 of the human milk samples exceeded 20ngg−1 day−1, the tolerable daily intake (TDI) proposed by the Health Canada Guideline in terms of DDTs levels. The high intake of DDTs by infants may be of concern as infants are more susceptible to the adverse effects imposed by various environmental contaminants. Human milk is a reliable and comparatively non-invasive tool for monitoring body loading of POPs, which also allows health risk assessment of residual chemicals on our next generation. Objective: To evaluate the predictive value of a cord serum screen test and possible subsequent development of allergic disease in infants. Design: Cohort study. Setting: 100 pregnant women were randomly recruited for the study. Methods: The maternal serum and the cord serum of their matched newborn were analyzed for total serum immunoglobulin E (IgE), gamma interferon (γ IFN), house dust mite- Dermatophagoides pteronyssinus allergen (Der p1) and Blomia tropicalis allergen (Blo t5) using immunoassay methods. All infants were followed up for one year. Results: Infants who had allergic diseases in the one year follow-up (n=45) had significantly (P<0.001) elevated IgE, Der p1, Blo t5, and significantly low γ IFN levels in cord serum as compared to the same parameters of infants who did not develop allergic disease in the one year follow- up (n=43). Conclusion: In utero exposure to HDM allergens Der p1 and Blo t5 is prevalent. We have successfully established a cord serum screening test for predicting allergic diseases in infancy with 93% specificity and sensitivity. [ABSTRACT FROM AUTHOR]

The present study objective was to investigate ratios and correlation coefficients between dichlorodiphenyltrichloroethane (DDT) compounds in cord and maternal sera of mother-infant pairs from northern Thailand. The study site was located in Chiang Dao District of Chiang Mai Province which was an agricultural and former malaria endemic area. DDT compounds were analyzed in 88 cord and maternal serum samples using gas chromatography-electron capture detection (GC-ECD). p,p'-DDE (1,1-dichloro-2,2-bis(p-chlorophenyl)ethylene) was the major component and detected in every cord and maternal serum samples with geometric means of 1,255 and 1,793 n g-1 lipids, respectively. p,p'-DDT (1,1,1-trichloro-2,2-bis(p-chlorophenyl)ethane) was detected at 89.8 and 100% of cord and maternal serum samples, respectively. The second and third highest levels detected were p,p'-DDD (1,1-dichloro-2,2-bis(p-chlorophenyl)ethane) and p,p'-DDT, respectively. The ratios between cord and maternal sera for p,p'-DDE, p,p'-DDT, and p,p'-DDD that were less than 1 had high correlation coefficients (ratio = 0.70, r = 0.82 for p,p'-DDE, ratio = 0.62, r = 0.66 for p,p'-DDT, and ratio = 0.79, r = 0.78 for p,p'-DDD). The high correlation coefficients indicate that cord serum levels of DDT compounds could be accurately estimated from maternal serum levels. It can be concluded that cord serum levels of p,p'-DDE, p,p'-DDT, and p,p'-DDD were approximately 70%, 62%, and 79% of maternal serum levels, respectively. BACKGROUND: Hexachlorobenzene (HCB) is an organochlorine chemical that has been used in agriculture and industrial processes. Behavioral impairment after HCB exposure has been described in animal models, but little information is available in humans. OBJECTIVES: Our goal was to study the association of prenatal exposure to HCB with the social behavior of preschool children. METHODS: Two birth cohorts in Ribera d'Ebre and Menorca (Spain) were set up between 1997 and 1999 (n = 475). The California Preschool Social Competence Scale and the Attention-Deficit Hyperactivity Disorder (ADHD) were scored by each 4-year-old child's teacher. Organochlorine compounds were measured in cord serum. Children's diet and parental sociodemographic information were obtained through questionnaire. RESULTS: Children with concentrations of HCB > 1.5 ng/mL at birth had a statistically significant increased risk of having poor Social Competence [relative risk (RR) = 4.04; 95% confidence interval (CI), 1.76-9.58] and ADHD (RR = 2.71; 95% CI, 1.05-6.96) scores. No association was found between HCB and the cognitive and psychomotor performance of these children. CONCLUSIONS: Prenatal exposure to current concentrations of HCB in Spain is associated with a decrease in the behavioral competence at preschool ages.
